Abstract:Concerns about environmental and food contamination caused by chlortetracycline (CTC) residues have prompted people to explore efficient and convenient CTC monitoring platforms. However, the reported fluorescent probes generally fail to selectively detect CTC due to the structural similarity of tetracycline antibiotics. Herein, an intrinsic dual-emission carbon dots (D-CDs) ratiometric fluorescence sensor was prepared for highly sensitive and selective determination of CTC over other tetracyclines by one-step synthesis. The sensor exhibited a significant fluorescence enhancement at 425 nm after introducing CTC. The fluorescence "turn on" of the sensing system is due to aggregation-induced emission (AIE) phenomenon formed by hydrogen bonds and π conjugation promoting the specific recognition of CTC by D-CDs. The linear detection varied from 0.98 to 143.67 ng mL −1 with a low limit of detection (LOD) of 1.29 ng mL −1 ( R 2 = 0.998), which was lower than most reported in the literature. The D-CDs sensor was applied to detect CTC in spiked milk, blocked normal human serum, and fish samples with recoveries of 95.5–104.2% and relative standard deviations (RSDs) of 2.6%. Particularly, D-CDs based test papers with a smartphone were prepared for portable and visual detection of CTC by analyzing the various color changes of RGB of fluorescence color, with an LOD of 7.18 ng mL −1 ( R 2 = 0.9909). The fluorescence sensor designed in this work could be used as a rapid tool with high performance and selectivity for monitoring control in foods.

Abstract:The residue of chlortetracycline is potentially hazardous to human health; it is meaningful to exploit a portable, rapid, sensitive, and selective method for detection of chlortetracycline (CTC). In this study, a novel fluorescence bionic sensing probe (NH2-MIL-53&N,P-CDs@MIP) was successfully prepared based on the nitrogen and phosphorus codoped carbon dots decorated iron-based metal-organic frameworks combining with molecular imprinted polymer for the detection of CTC. A fluorescence intensity-responsive "on-off" detection of CTC on account of the inner-filter effect (IFE) was achieved by NH2-MIL-53&N,P-CDs@MIP. Under the optimal conditions, the fluorescence quenching degree of NH2-MIL-53&N,P-CDs@MIP presented a good linear relationship with the CTC concentration in the range 0.06-30 μg mL-1 and the limit of detection (LOD) was 0.019 μg mL-1. The fluorescent probe was applied to detect CTC in milk samples, and experimental results showed a good recovery rate (88.73%-96.28%). Additionally, a smartphone-integrated fluorescence sensing device based on NH2-MIL-53&N,P-CDs@MIP was exploited to replace the expensive and bulky fluorescence spectrophotometer for quantitative determination of CTC with the LOD of 0.033 μg mL-1. The sensing system showed high selectivity, strong stability, high specificity, and portability, which provide a great strategy for the quantitative detection of antibiotic residue.
A dual-response ratiometric fluorescent sensor by europium-doped silicon nanoparticles for fluorescent and smartphone imaging detection of tetracycline

Abstract:Given the threat to human health posed by the abuse of tetracycline (TC), the development of a portable, on-site methods for highly sensitive and rapid TC detection is crucial. In this work, we initially synthesized europium-doped silicon nanoparticles (SiEuNPs) through a facile one-pot microwave-assisted method. Due to its blue-red dual fluorescence emission (465 nm/621 nm), which was respectively attributed to the silicon nanoparticles and Eu3+, SiEuNPs were designed as a ratiometric fluorescent sensor for TC detection. For the dual-signal reverse response mechanism: TC quenched the blue emission from silicon nanoparticles through inner filter effect (IFE), and enhanced the red emission through "antenna effect" (AE) between TC and Eu3+, the nanoprobe was able to detect TC within a range of 0.2-10 μM with a limit of detection (LOD) of 10.7 nM. Notably, the equilibrium detection time was only 1 min, achieving rapid TC detection. Furthermore, TC was also measured in real samples (tap water, milk and honey) with recoveries ranging from 95.7 % to 117.0 %. More importantly, a portable smartphone-assisted on-site detection platform was developed, enabling real-time qualitative identification and semi-quantitative analysis of TC based on fluorescence color changes. This work not only provided a novel doped silicon nanoparticles strategy, but also constructed a ratiometric sensing platform with dual-signal reverse response for intuitive and real-time TC detection.
Smartphones and Test Paper-Assisted Ratiometric Fluorescent Sensors for Semi-Quantitative and Visual Assay of Tetracycline Based on the Target-Induced Synergistic Effect of Antenna Effect and Inner Filter Effect

Abstract:Development of selective and sensitive methods for on-site assay of tetracycline (TC) is of great significance for public health and food safety. Herein, a valid ratiometric fluorescence strategy using g-C3N4 nanosheets coupled with Eu3+ is designed for the assay of TC. In this strategy, both Eu3+ and g-C3N4 nanosheets serve as the recognition units of TC. The blue fluorescence of g-C3N4 nanosheets can be quenched by TC via the inner filter effect (IFE); meanwhile, the red fluorescence of Eu3+ can be enhanced by TC through the antenna effect (AE). The synergistic effect of AE and IFE caused by TC makes the developed ratiometric fluorescent sensor display a wide linear range for TC from 0.25 to 80 μM with a detection limit of 6.5 nM and a significant fluorescence color evolution from blue to red. Given its simplicity, free-label, excellent selectivity, high sensitivity, and recognizable color change, point-of-care testing systems, including smartphones and test paper-based assays, are developed for the visual sensing of TC. The integration of smartphones and test paper on a ratiometric fluorescent sensor greatly reduces the detection cost and time, providing a promising method for the qualitative discernment and semi-quantitative assay of TC on-site. Abstract:The widespread presence of tetracyclines in the environment has raised concerns about the potential risks to ecosystems and human health. The ratiometric fluorescence sensor for detecting tetracyclines was developed using europium-doped carbon dots (Eu-CDs) as probes under alkaline conditions. The sensing mechanism of sensor for tetracyclines was considered as inner filter effect (IFE), antenna effect (AE), and self-quenching effect (SQE). The sensor had a wide linear detection range than the reported europium ions-based tetracyclines sensors. The linear detection ranges of oxytetracycline (OTC), tetracycline (TC), doxycycline (DC) and chlorotetracycline (CTC) were respectively 0.00–603.75 μM, 0.00–623.82 μM, 0.00–594.61 μM and 0.00–601.54 μM, and the corresponding detection limits were respectively 9.50 nM, 15.80 nM, 10.4 nM and 90.3 nM. The smartphone with RGB Color Picker was further employed to analyze the concentration of tetracyclines, which provided a new method for visual tetracyclines detection. The application of Eu-CDs test paper was also explored, and the results showed that the Eu-CDs test paper has great potential application in the visual detection of tetracyclines. In addition, the accuracy of the established tetracyclines sensor was compared with that of the China national standard method by high-performance liquid chromatography (HPLC), and the results showed that the established method in this work has similar accuracy to the China national standard method. The sensor has been employed to detect tetracyclines in the actual samples with satisfactory results, which indicated that this method has promising applications in the real-time monitoring tetracyclines of food and environment.

Abstract:As widely used antibiotics, tetracycline residues exist in food and environmental media, which pose certain hidden dangers and negative effects on public health. Therefore, the sensing and discrimination of tetracycline analogs (TCs) have great significance for improving food safety and preventing environmental pollution. Herein, a 7-hydroxycoumarin-3-carboxylic acid-embedded Eu-MOF (HC@Eu-MOF) material was constructed and then developed for the detection of TCs. Upon addition of TCs, the synthesized sensor displays opposite fluorescence changes at two different wavelengths due to the simultaneous presence of the inner filter effect (IFE) and the antenna effect (AE), and achieves a stable ratio signal response within 90 s. In addition, six important tetracycline analogs, including chlortetracycline (CTC), oxytetracycline (OTC), tetracycline (TC), metacycline (MC), doxycycline (DC) and demeclocycline (DMC) can be discriminated with 100 % accuracy through the principal component analysis even in extremely complicated mixtures. Further, a smartphone-assisted portable device was applied for visual sensing of TCs. The as-developed platform possessed the characteristics of simple synthesis, fast response, high sensitivity, and high stability, which further lays a further foundation for the on-site visual detection and discrimination of TCs in real samples.

Abstract:The massive residues of tetracycline (TC) have caused serious harm to the human body and the environment, so it is urgent to explore a portable, on-site, sensitive and selective method to detect TC. In this study, a dual response ratiometric fluorescent probe (S,O-C 3 N 4 QDs-CitNa-Eu 3+ ) for detecting TC was constructed by mixing sodium citrate (CitNa), europium ion (Eu 3+ ) and sulfur and oxygen co-doped graphite phase carbon nitride quantum dots (S,O-C 3 N 4 QDs) solution in a specific proportion. The S,O-C 3 N 4 QDs was initially synthesized by using citric acid and thiourea as the precursors via a hydrothermal route. The blue fluorescence of S,O-C 3 N 4 QDs ( λ em = 445 nm) was decreased while the red fluorescence of Eu 3+ was increased ( λ em = 620 nm) own to the synergistic effect of internal filter effect, static quenching and antenna effect when TC was added to the mixed system containing CitNa and Eu 3+ . As a result, the ultra-sensitive detection of TC was realized from 0.25 to 120 μM with a detection limit of 1.3 nM. An obvious color change of the solution from blue to purple to red with continuous adding TC was observed such that the semi-quantitative detection of TC can be realized. Moreover, a smartphone APP monitoring platform based on test paper was constructed by using color scanning application, demonstrating that the ratiometric fluorescent probe constructed in this work has great potential in medical and environmental fields.

Abstract:The detection of tetracycline antibiotics (TCs) in food holds great significance in minimizing their absorption within the human body. Hence, this study aims to develop a rapid, convenient, real-time, and accurate detection method for detecting antibiotics in an authentic market setting. A colorimetric fluorescence sensor was devised for tetracycline detection utilizing PVA aerogels as the substrate. Its operating principle is based on the IFE effect and antenna effect. A detection device is designed to capture fluorescence images while deep learning was employed to aid in the detection process. The sensor exhibits high responsiveness with a mere 60-s requirement for detection and demonstrates substantial color changes(blue to red), achieving 99% accuracy within the range of 10-100 μM with the assistance of deep learning (Resnet18). Real sample simulation tests yielded recovery rates between 95% and 130%. Overall, the proposed strategy proved to be a simple, portable, reliable, and responsive solution for rapid real-time TCs detection in food samples.

Abstract:Exploring high-performance fluorescence strategies for onsite monitoring of oxytetracycline (OTC) is of great significance for protecting human health. Herein, an innovative ratiometric fluorescence biomimetic sensor featured with high sensitivity and selectivity was constructed based on magnesium and nitrogen co-doped carbon dots embedded europium metal–organic framework and incorporating molecularly imprinted polymer (Mg,N-CDs/Eu-MOFs@MIP). In this protocol, MIP layer functioning as recognition units endowed Mg,N-CDs/Eu-MOFs@MIP with satisfactory binding efficiency and anti-interference ability. Mg,N-CDs/Eu-MOFs as both supporter and signal tag enabled the sensor to not only demonstrate a fast fluorescence response, but also yield reverse response signals depending on the synergistic effect of inner filter effect (IFE) and antenna effect (AE), achieving signal amplification. Benefiting from the efficient dual-channel strategy, Mg,N-CDs/Eu-MOFs@MIP exhibited a wide linear range (0.02–50 μg mL−1) with a sensitive detection limit (6.6 ng mL−1) and a marked color transformation (blue-to-red), which was highly desirable for monitoring of oxytetracycline. Simultaneously, aiming at attaining precise onsite analysis, a portable intelligent sensing platform was devised by integrating Mg,N-CDs/Eu-MOFs@MIP and a smartphone-assisted device. The smartphone with an installed color recognizer application as signal reader and analyzer realized oxytetracycline visual sensing by capturing and digitizing fluorescence images. More significantly, the portable platform was successfully applied to the onsite monitoring of OTC in water samples and milk samples with acceptable results. Accordingly, the as-developed platform demonstrated low-cost, portability, easy implementation, desirable specificity and sensitivity, presenting great potential for point-of-care testing.

Abstract:The emergence of “superbugs”, which can cause great panic, has been proved to be related to the pollution of antibiotics in the environment water and food. It is of great research value to develop a portable, on-site and simple method for sensitive and selective detection of antibiotics. In this work, a laponite-based dual-channel fluorescent nanoprobe for the detection of tetracycline (TC) was manufactured through a simple assembly process. The nanoprobe can detect TC within two concentration channel from as low as 30 nM up to 54 μM with a detection limit of 9.5 nM, which met the needs in various analytical samples and showed excellent selectivity over various interferers. More significantly, the paper-based and glove-based visual probes were devised for the detection of TC by multi-color fluorescence change. And the tetracycline was also measured in milk, honey and lake after post-treatment of the samples. Combined with smartphone chromaticity analysis application (APP), the portable and disposable detection of TC can be realized. In addition, according to the fluorescence color change of the probe corresponding to the concentration of TC, two logic gates were also designed, which provided the possibility for the application of the probe in the field of smart devices.

Abstract:Environmental pollution caused by tetracycline antibiotics is a major concern of global public health. Here, a novel and portable molecularly imprinted electrochemiluminescence (MIECL) sensor based on smartphones for highly sensitive detection of chlortetracycline (CTC) has been successfully established. The high-performance ECL emitter of biomass carbon (BC) encapsulated CdZnTeS (CdZnTeS@BC) was successfully synthesized by hydrothermal. The enhanced ECL performance was ascribed to the introduction of the BC and increased the overall electrical conductivity of the nanoemitter, as well as increased the number of sulfur vacancies and doping on the surface of the emitter based on density functional theory calculations. An aniline-CTC molecular imprinted polymer was synthesized on the surface of the CdZnTeS@BC modified electrode by in-situ electropolymerization. The decrease in MIECL signal was attributed to the increase in impedance effect. The MIECL nanoplatform enabled a wide linear relationship in the range of 0.05-100 μmol/L with a detection limit of 0.029 μmol/L for spectrometer sensors. Interestingly, the light emitted during the MIECL reaction can be captured by a smartphone. Thus, machine learning was used to screen the photos that were taken, and color analysis was carried out on the screened photos by self-developed software, thus achieving a portable, convenient, and intelligent sensing mode. Finally, the sensor obtains satisfactory results in the detection of actual samples, with no significant differences from those of liquid chromatography.

Abstract:A new fluorescent probe based on graphitic carbon nitride (g-C3N4) combined with molecularly imprinted silica was successfully fabricated and used to selectively recognize chlortetracycline (CTC). The g-C3N4 used in this study has the characteristics of low toxicity and high chemical stability. This synthetic composite was characterized by transmission electron microscopy, Fourier transform infrared spectroscopy, UV spectroscopy, X-ray diffraction, and fluorescence spectroscopy. The material was used to detect CTC by the fluorescence quenching technique. The fluorescence quenching was due to g-C3N4 and the benzene ring of CTC through π–π electron donor–acceptor interaction and electrostatic force. Hydrogen bonds formed between CTC and 3-aminopropyltriethoxysilane during the polymerization process. Eventually, a considerable amount of selective recognition holes were formed in the composite material and could specifically recognize the template molecule CTC. In addition, the probe strategy was successfully applied to milk analysis, and the recoveries ranged from 90.1% to 95.7%, with relative standard deviations of 1.8–2.8%; the detection limit for CTC was 8 ng mL-1. The results indicate that this method combined the sensitivity of fluorescence detection with the excellent selectivity of a molecularly imprinted polymer. Abstract:Oxytetracycline (OTC) residues have serious adverse effects on human health. Thus, selective and sensitive field detection methods are needed. Here, we successfully synthesized nitrogen-doped carbon quantum dots (N-CQDs) with good water-solubility, prominent fluorescence (FL), and favorable stability via a one-step hydrothermal method using banana peel as a carbon source and diethylenetriamine as a nitrogen source. We used the N-CQDs to create a ratiometric FL probe by self-assembling them with europium ions (Eu 3+ ), enabling OTC detection. The FL intensity of N-CQDs at 445 nm was quenched due to the inner filter effect, while the FL intensity of Eu 3+ at 621 nm markedly increased via the antenna effect upon OTC introduction. The ratiometric FL sensor demonstrated two linear relationships with OTC concentrations, ranging from 0.01 to 5 μg mL −1 and 5 to 25 μg mL −1 , with a limit-of-detection of 4.9 ng mL −1 for OTC. Additionally, this platform depicted an apparent color change from blue to purple under a 365 nm ultraviolet–visible lamp, and a visual semi-quantitative analysis of OTC was achieved using a mobile phone application. We successfully implemented this method to detect OTC in pork muscle and swine urine, achieving remarkable recovery results (96.89–103.20%). Therefore, our findings demonstrate the potential of this approach for real-time and semi-quantitative OTC detection.

Abstract:Raman spectroscopy has been applied to a model biomembrane structure in order to obtain information about phospholipid hydrocarbon chain ordering. The intensity of the 1130-cm-1 Raman line obtained from a dipalmitoylphosphatidylcholine (DPPC) coarse aqueous dispersion has been measured as a function of temperature. The intensities of this line anther with the pretransition was observed. A theory of chain conformations as a function of temperature and rules for the assignment of Raman scattering intensities for this line have been constructed. Good agreement with the DPPC experimental data has been obtained. Predictions for the intensity of this line as a function of temperature from dimyristoyl- and distearoylphosphatidylcholine dispersions have also been made.

Abstract:Lanthanide coordination polymer nanoparticles (LML-Eu3+-GMP CPNPs) were fabricated as a ratiometric fluorescence sensor for real-time and visual assays of tetracycline (TC). The LML-Eu3+-GMP CPNPs were prepared by spontaneous self-assembly of Eu3+, luminol (LML) and guanosine 5'-monophosphate (GMP) at room temperature, which only emitted blue fluorescence dominated by LML, and no red characteristic fluorescence of Eu3+ was observed. In the presence of TC, TC can trigger the antenna effect (AE) and inner filter effect (IFE), the blue fluorescence of LML-Eu3+-GMP CPNPs is quenched by TC via IFE, and the red characteristic fluorescence of Eu3+ can be turned on through AE. When sodium citrate (Cit) is added to the sensing system, it can further enhance the red fluorescence of Eu3+. The dual-ligand CPNP sensor has a rapid fluorescence response, noticeable fluorescence color change, good selectivity, and high sensitivity with a detection limit of 3.4 nM. It was successfully applied to detect TC in honey, milk, lake water and tap water. Meanwhile, the portable smartphone and test paper were applied to develop POCT devices for real-time and visual detection of TC, which could offer a promising application for real-time and semiquantitative assays of TC.
A Molecularly Imprinted Electrochemical Sensor Based on TiO2@Ti3C2Tx for Highly Sensitive and Selective Detection of Chlortetracycline

Abstract:In view of the serious side effects of chlortetracycline (CTC) on the human body, it is particularly important to develop rapid, sensitive, and selective technologies for the detection of CTC in food. In this work, a molecularly imprinted electrochemical sensor with [Fe(CN)6]3−/4− as signal probe was proposed for the highly sensitive and selective detection of CTC. For this purpose, TiO2, which acts as an interlayer scaffold, was uniformly grown on the surface of Ti3C2Tx sheets through a simple two-step calcination process using Ti3C2Tx as the precursor to effectively avoid the stacking of Ti3C2Tx layers due to hydrogen bonding and van der Waals forces. This endowed TiO2@Ti3C2Tx with large specific surface, abundant functional sites, and rapid mass transfer. Then, polypyrrole molecularly imprinted polymers (MIPs) with outstanding electrical conductivity were modified on the surface of TiO2@Ti3C2Tx via simple electro-polymerization, where the pyrrole was employed as a polymeric monomer and the CTC provided a source of template molecules. This will not only provide specific recognition sites for CTC, but also facilitate electron transport on the electrode surface. The synergistic effects between TiO2@Ti3C2Tx and polypyrrole MIPs afforded the TiO2@Ti3C2Tx/MIP-based electrochemical sensor excellent detection properties toward CTC, including ultra-low limits of detection (LOD) (0.027 nM), a wide linear range (0.06–1000 nM), and outstanding stability, reproducibility, selectivity, and feasibility in real samples. The results indicate that this strategy is feasible and will broaden the horizon for highly sensitive and selective detection of CTC.

Abstract:Development of efficient and portable biosensor to achieve the accurate onsite detection of food contaminants is of very great significance for safeguarding food safety and human health. With the popularity of smartphone, the digital imaging feature makes it to be an outstanding analytical platform to develop the point-of-care testing sensors. Compared with single-emission fluorescent assay, the ratiometric fluorescent assay possesses the built-in self-calibration function to avoid the interferences caused by various target-independent factors, circumventing/reducing false positive result output, thus improving the detection accuracy and expanding the dynamic detection range in complicated food matrixes. Based on these, vast efforts focus on integrating smartphone with ratiometric fluorescent strategies to develop portable sensors for realizing the onsite visual detection of food contaminants in a rapid, low cost, reliable, and even easy to implement in resource-limited areas way. In this review, we summarize the recent development of ratiometric fluorescent signals-driven smartphone-based sensors for onsite visual detection of food contaminants (e.g., antibiotics, pesticides, & harmful ions), discuss the confronting challenges, and propose an outlook on the future of this advanced analytical technology to help better achieve the precisely onsite detection of food contaminants, ensuring food safety and human health.

Abstract:The molecular imprinting ratiometric fluorescence sensor has exhibited great potential in biological and environmental detection because of the combination between the excellent selectivity of molecular imprinting technique and outstanding robustness of ratiometric measurement methods On this basis, two kinds of different well-resolved wavelengths and high-fluorescent carbon dots (CDs) were prepared by various solvent extraction using osmanthus fragrans leaves as carbon source. Then the ratiometric fluorescence sensor (MIPs@rCDs/bCDs@SiO2) has been constructed by the obtained CDs which the red-emission CDs (rCDs) were used to responsive signal and the blue-emission CDs (bCDs) was served as reference signal and applied for highly selective and excellent sensitive detection of tetracycline (TC). Under optimum conditions, the ratiometric fluorescence MIPs@rCDs/bCDs@SiO2 sensor has also exhibited lower detection limit of 1.19 nM and the linear range of 0-50 nM. The ratiometric fluorescence sensor was further successfully employed to the determination of TC in water samples gathered from local river water and tap water, indicating its great worth toward water sample analysis in complicated environments. Integrated Eu3+ loaded covalent organic framework with smartphone for ratiometric fluorescence detection of tetracycline

Abstract:Developing rapid tetracycline sensing system is of great significance to monitor the illegal addition to drugs and pollution to food and ecosystem. By loading covalent organic frameworks (COFs) with Eu3+, a new hybridized material (COF@Eu3+) was prepared for tetracycline determination. Based on the Schiff base reaction, the COFs were by synthesized through solvent evaporation in 30 min at room temperature. Thereafter, Eu3+ was modified into COFs to develop the COF@Eu3+ sensing platform by adsorption and coordination. In presence of tetracycline, tetracycline can displace water molecules and coordinate with Eu3+ through the antenna effect. As a result, the red fluorescence of Eu3+ was enhanced by tetracycline with green fluorescence of COF as a reference. The developed ratiometric fluorescence sensor exhibits a linear range of 0.1-20 μM for detecting tetracycline with a detection limit of 30 nM. Integrated with a smartphone, the rapid tetracycline detection can be realized in situ, which is potential for high-throughput screening of tetracycline contaminated samples. Furthermore, the COF@Eu3+ fluorescence sensor has been successfully applied to the detection of tetracycline in traditional Chinese medicine compound preparation with satisfied recoveries. Therefore, a smartphone-assisted device was successfully developed based on Eu3+-functionalized COF, which is an attractive candidate for further applications of fluorescence sensing and visual detection.
