Abstract:A near-infrared fluorescent probe (IC-V) for detecting viscosity is constructed. The probe has a large Stokes shift (170 nm) and an about 180-fold increase in fluorescence intensity at 700 nm. In addition, IC-V can not only distinguish cancer cells from normal cells, but also monitor viscosity in normal mice and tumor-bearing mice.

Abstract:A universal and non-invasive approach for cancer diagnosis is desired to lessen the burden of cancer mortality. In this work, we exploited viscosity in the cancer microenvironment as a diagnosis indicator and designed a universal probe, the CBT-Q (4-(2-(5-(9-(4-aminophenyl)-9H-carbazol-3-yl) thiophen-2-yl)vinyl)-1-ethylquinolin-1-ium) nanoprobe, for detecting viscosity variation in the cells, plasma, and solid tumor-mice to obtain a cancer diagnosis. The near infrared (NIR) fluorescence of CBT-Q was specifically triggered due to a change in its structure from a flexible to a rigid one in the presence of a higher viscosity agent, such as a cancer microenvironment or cancer patient plasma. The Stokes shift of CBT-Q nanoprobe (CBT-Q-N) could be up to 178 nm, enabling the diagnosis of cancer with a low biological fluorescent background. Not only could the CBT-Q-N remain in cells for 72 h, but detect the changes in cell viscosity induced by various models. Also, the CBT-Q-N differentiated early and terminal tumor mice via the fluorescent intensity of plasma viscosity. The CBT-Q-N enables non-invasive recognition of tumors in vivo by fluorescence imaging. Hence, this work is beneficial to increase the understanding of tumor-related viscosity, which will facilitate a new universal strategy for present cancer diagnostics.

Abstract:Multifunctional nanoprobe was drawing increased attention in tumor diagnosis and therapy. The simple and effective establishment of the theranostic nanoplatforms was still under urgent need. Meanwhile, the targeting ability and morphology of nanoprobe were essential for the effective endocytosis, which could further affect the diagnosis. In this work, two morphologies of nanoprobes were fabricated using gold nanorods (AuNRs) and gold nanospheres (AuNSs). Thiolated-hyaluronic acid labeled with nile blue (HS-HA-NB), a near-infrared (NIR) fluorescence dye, was coated on the surface of the gold nanoparticles to form stable nanoprobes (AuNR@HS-HA-NB, AuNS@HS-HA-NB). The fluorescence of NB molecules quenched outside cells due to the fluorescence resonance energy transfer (FRET), and recovered after the HA degradation inside the cells. HA also could enhance cellular uptake in CD44 receptor highly expressed human breast carcinoma cells (MCF-7). In this way, bioprobes realized the MCF-7 cell images through intracellular fluorescent light-up. Comparing with the sphere bioprobe, the rod-shaped bioprobe dramatically promoted endocytosis to achieve a better diagnosis effect in a short time. After NIR light irradiation, severe MCF-7 apoptosis was observed with AuNR@HS-HA-NB existed. Our studies suggested that the AuNR@HS-HA-NB nanoparticles were the excellent candidates of versatile bioprobes to realize rapid, precise image and photothermal therapy to MCF-7 cells. Abstract:In recent years, targeting drugs made by physical loading or chemical bonding of drugs on small molecular carriers have shown a very wide application prospect in the field of tumor and cancer treatment. How to achieve the release of drugs in cancer cells has become the core of this research. One of the most important bases for drug localization is to use the difference of small molecular biothiol concentration between cancer cells and normal cells. Details of the changes of biothiol levels in the growth and reproduction of cancer cells are still poorly understood, and the main reason is the lack of sensitive real-time imaging tools for biothiols in cancer cells. In this work, we reasonably designed and synthesized the combination of 4-hydroxy-1,8-naphthalimide and NBD-Cl as a concise fluorescent probe HN-NBD for imaging biothiols in live cells and zebrafish. In addition, due to the advantages of HN-NBD design, it is sufficiently sensitive to biothiols, and further imaging can distinguish cancer cells from normal cells. Probe HN-NBD would be of great significance to biomedical researchers for the study of biothiol-related diseases, the screening of new anticancer drugs, and the early diagnosis and treatment of cancers.

Abstract:As a basic parameter of the intracellular microenvironment, viscosity is closely related to the development of cancer. Thus, it is necessary to utilize a sensitive tool to visualize the viscosity in tumor cells and mice, which is helpful for the diagnosis of cancer. Herein, a novel dual-modal probe (IX-V) that has a near-infrared fluorescence (NIRF) and photoacoustic (PA) response to viscosity is synthesized. In low viscosity media, the probe has no fluorescence. With the increase of viscosity, the fluorescence is produced in the near-infrared region due to the inhibition of the TICT process. At the same time, the probe shows different photoacoustic (PA) signals in different viscosity media. Most notably, the viscosity in tumor cells has been imaged successfully by the application of IX-V, and the probe can effectively distinguish cancer cells from normal cells co-cultured in one dish by the difference of fluorescence intensity. In addition, the probe has been used for dual-modal imaging (NIRF and PA) of viscosity in tumor mice, which provides a tool for exploring the relationship between viscosity and diseases. That is to say, IX-V can achieve complementary imaging effects and has great application prospects in the tumor diagnosis.

Abstract:The higher viscosity and lower pH in lysosomes of cancer cells highlight their potential as biomarkers for cancer. Therefore, the development of acid-activated viscosity fluorescent probes is significant for the early diagnosis and treatment of cancer. Based on this, we have designed and synthesized a near-infrared fluorescent probe based on the 2-(2-hydroxyphenyl)benzothiazole (HBT) group, namely HBTH , to monitor the viscosity changes within lysosomes. It has been demonstrated that HBTH was extremely sensitive to viscosity, with a strong linear relationship between fluorescence intensity and log(viscosity) within the range of (logη) = 0–3.06 (a correlation coefficient of 0.98), proving its capability for quantitative viscosity measurement. In particular, the most obvious fluorescence enhancement of HBTH was only efficiently triggered by the combined effect of low pH and high viscosity. Furthermore, HBTH can rapidly localize to lysosomes by wash-free procedure at a low concentration (100 nM) and achieve high-fidelity imaging within 20 s. It can also monitor the dynamic processes of lysosomes in cells, viscosity changes under drug stimuli, and lysosomal behavior during mitophagy. Importantly, HBTH is capable of identifying tumors in tumor-bearing nude mice through in vivo imaging. These features make HBTH a powerful tool for the early diagnosis and treatment of cancer.

Abstract:Although the cure rate of cancer is very low, the survival rate of patients can be improved if it is diagnosed early and treated in time. Polarity, as a significant parameter of tumor microenvironment, is bound up with the development of cancer. Unfortunately, there is currently no report about tumor-targeted and polarity-specific NIR fluorescent probe for accurate cancer diagnosis. Here, a polarity-specific near-infrared (NIR) fluorescent probe named CX-P based on coumarin-xanthene dye is reported. The probe has the advantages of long emission wavelength (>650 nm) and outstanding tumor targeting ability. CX-P is highly sensitive to polarity, and has a linear range from Delta integral = 0.284 (30% water) to Delta integral = 0.321 (100% water). Moreover, the probe can actively target cancer cells and display strong red fluorescence, while the fluorescence in normal cells is negligible. CX-P is also used to real-time monitor the polarity changes after the treatment of cancer cells with LPS, sucrose and taxol. Based on its NIR characteristics, CX-P is applied to real-time imaging of tumor-bearing mice and the significant tumor targeting ability is further confirmed. As far as we know, this is the first tumor-targeting and polarity -specific NIR fluorescent probe, which will offer a powerful means for accurate cancer diagnosis in the future.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.

Abstract:Background: Cirrhosis represents the terminal stage of liver disease progression and timely intervention in a diseased liver can enhance the likelihood of recovery. Viscosity, a crucial parameter of the cellular microenvironment, is intricately linked to the advancement of cirrhosis. However, viscosity monitoring still faces significant challenges in achieving non-invasive and rapid early diagnosis of cirrhosis. Near-infrared (NIR) fluorescence imaging has the advantages of high sensitivity, non-destructive detection, and ignoring background fluorescence interference, plays an important role in diagnosing and treating various biological diseases. Hence, monitoring cellular viscosity changes with NIR fluorescence probe holds great significance in the early diagnosis of cirrhosis. Results: In this study, the NIR fluorescence probe based on the intramolecular charge transfer (TICT) mechanism was developed for imaging applications in mouse model of liver cirrhosis. A molecular rotor-type viscosity-responsive probe was synthesized by linking dioxanthracene groups via carbon-carbon double bonds. The probe demonstrated remarkable sensitivity, high selectivity and photostability, with its responsiveness to viscosity largely unaffected by factors such as polarity, pH, and interfering ions. The probe could effectively detect various drug-induced changes in cellular viscosity, enabling the differentiation between normal cells and cancerous cells. Furthermore, the enhanced tissue penetration capabilities of probe facilitated its successful application in mouse model of liver cirrhosis, allowing for the assessment of liver disease severity based on fluorescence intensity and providing a powerful tool for early diagnosis of cirrhosis. Significance: A NIR viscosity-sensitive fluorescent probe was specifically designed to effectively monitor alterations in cellular and organ viscosity, which could advance the understanding of the biological characteristics of cancer and provide theoretical support for the early diagnosis of cirrhosis. Overall, this probe held immense potential in monitoring viscosity-related conditions, expanding the range of biomedical tools available.

Abstract:Abnormal viscosity is closely related to the occurrence of many diseases, such as cancer. Therefore, real-time detection of changes in viscosity in living cells is of great importance. Fluorescent molecular rotors play a critical role in detecting changes in cellular viscosity. Developing red emission viscosity probes with large Stokes shifts and high sensitivity and specificity remains an urgent and important topic. Herein, a novel viscosity-sensitive fluorescent probe (TCF-VIS1) with a large stokes shift and red emission was prepared based on the 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) skeleton. Due to intramolecular rotation, the probe itself does not fluorescence at low viscosity. With the increase in viscosity, the rotation of TCF-VIS1 is limited, and its fluorescence is obviously enhanced. The probe has the advantages of simple preparation, large Stokes shift, good sensitivity and selectivity, and low cytotoxicity, which make it successfully used for viscosity detection in living cells. Moreover, TCF-VIS1 showed its potential for cancer diagnosis at the cell level and in tumor-bearing mice by detecting viscosity. Therefore, the probe is expected to enrich strategies for the detection of viscosity in biological systems and offer a potential tool for cancer diagnosis.

Abstract:Mitochondria, as an important organelle in cells, play an important role in the production of adenosine triphosphate (ATP) and cell signal transduction, as well as control of the cell cycle and growth. Abnormal viscosity is a pivotal factor for indicating mitochondrial dysfunction. In addition, abnormal changes in mitochondrial viscosity have been confirmed to be closely related to many diseases and malfunctions, such as cell malignancy, atherosclerosis, diabetes, and Alzheimer's disease. Therefore, the development of mitochondrial viscosity response probe and its application in mitochondrial viscosity measurement are considered as a new tool to understand diseases. So far, most of the fluorescent probes used to detect mitochondrial viscosity have emission wavelengths in the visible region, making them less penetrable to tissue. In this work, we report a novel near-infrared fluorescent probe (MN-V) for the detection of mitochondrial viscosity in live cells, which exhibited near infrared emission (795 nm) and high sensitivity towards viscosity changes. The fluorescence intensity of the probe at 795 nm has a 37 times increase from pure PBS solution to glycerol. There was a linear relationship between the fluorescence intensity and the media viscosity. The probe MN-V was applicable to detection of the viscosity changes in living cells and zebrafish. Most importantly, with the probe we found that during cancer cells ferroptosis process was accompanied by an increase in mitochondrial viscosity.

Abstract:Based on OR logic gate, we proposed a smart near-infrared (NIR) fluorescent probe, named VPCPP, for simultaneously monitoring local microviscosity, micropolarity, and carboxylesterases (CEs) in living cells through blue and red channels. This proposed probe was capable of distinguishing cancer cells from normal cells and had good potential for identifying living liver cell lines. Furthermore, the fluctuations of the three analytes of interest in different cell status was successfully explored. Particularly, facilitated with high-content analysis (HCA) and VPCPP, a simple and efficient high-throughput screening (HTS) platform was first constructed for screening antitumor drugs and studying their effect on the analytes. For the first time, we found that sorafenib-induced ferroptosis led to an increase in the microviscosity and up-regulation of CEs at the same time. Additionally, the procedure that aristolochic acid (AA) induced the overexpression of CEs was verified. Besides, VPCPP was utilized for imaging the variations of the two microenvironment parameters and CEs in the inflammation model. Abstract:A viscosity-sensitive, lysosome-targeted near-infrared fluorescent probe (PYATT) was reported in this paper. The fluorescent spectra of PYATT are strongly dependent on viscosity, resulting in a Stokes shift of about 190 nm. Given its photostability, low cytotoxicity, and high fluorescence quantum yield, PYATT is expected to be used in cell imaging. Due to the higher viscosity of tumor cells than normal cells, the fluorescence intensity of PYATT in tumor cells is higher than normal cells, which can realize the visualization of tumors. The near-infrared probe (PYATT) is viscosity-dependent in lysosomes, which is valuable in early diagnosis and treatment of tumor.

Abstract:Developing novel small-molecule-based probes with both deep tissue imaging and therapeutic functions is highly significant in cancer diagnosis and treatment. Herein, we report a novel second near-infrared (NIR-II) fluorescent probe QT-RGD constructed with a NIR-II emissive organic fluorophore and two cyclic-(arginine-glycine-aspartic acid) (cRGD) peptides that can specifically bind to the tumor-associated αvβ3 integrin for accurate tumor diagnosis and targeting therapy. The isotopic 125I-labeled probe exhibited great tumor targeting ability and emitted intensive NIR-II/photoacoustic (PA)/single-photon emission computed tomography (SPECT) signals, which allows specific and sensitive multimodal visualization of tumors in vivo. More notably, this probe could also be applied for effective imaging-guided photothermal therapy (PTT) of tumors in mouse models owing to its prominent photothermal conversion efficiency and excellent photothermal stability. We thus envision that our work which unveils a combination of NIR-II/PA/SPECT imaging and PTT would offer a valuable means of improving tumor diagnostic accuracy as well as therapeutic efficacy.
GSH Activated Biotin-tagged Near-Infrared Probe for Efficient Cancer Imaging.

Abstract:Tumor imaging tools with high specificity and sensitivity are needed to aid the boundary recognition in solid tumor diagnosis and surgical resection. In this study, we developed a near infra-red (NIR) probe (P6) for in vitro/in vivo tumor imaging on the basis of the dual strategy of cancer cell targeting and stimulus-dependent activation. The selective imaging capacity towards cancer cells of P6 was thoroughly investigated, and the potential mechanisms of endocytosis were preliminary explored. Methods: GSH-activated biotin labelled NIR probe (P6) was designed, synthesized and characterized. The GSH responsive properties were systematically illustrated through UV-vis, fluorescent tests and LC-MS analysis. In vitro fluorescent imaging of probe P6 was collected in various living cancer cell lines (i.e. SW480, HGC-27, H460, BxPC-3, KHOS) and normal cell lines (i.e. BEAS-2B, HLF-1, THP1) under confocal laser scanning microscopy. Probe P6 was further applied to image primary human cancer cells which were freshly isolated from the peritoneal carcinoma and rectal cancer patients. Serial sections of human tumor tissues were collected and sent for H&E (hematoxylin-eosin) staining and P6 imaging. Live fluorescent and photoacoustic imaging were used to investigate the in vivo imaging of P6 in both tumor and normal tissues in HGC 27 and KHOS xenograft model. Results: Probe P6 could be recognized and transported into cancer cells by tumor specific biotin receptors and efficiently be triggered by GSH to release fluorophore 4. In fact, the cellular uptake of P6 could be partially blocked by the addition of free biotin. Furthermore, probe P6 could image various cancer cell lines, as well as primary cancer cells, exhibiting a ten-fold increase in fluorescence intensity over normal cells. In freshly dissected cancer tissues, P6 fluorescent imaging distinguished the cancerous area under confocal laser scanning microscopy, which was exact the same area as indicated by H&E staining. We also found that P6 exhibited superior selectivity against cancer tissues by local injection. Conclusion: In this study, we developed a dual-modal NIR probe P6 with enhanced cellular uptake into cancer cells and environmental stimulus triggered fluorescence. Our strategy provided a novel insight into the development of imaging tools that could be potentially used for fluorescent image-guided cancer boundary recognition and possibly cancer diagnosis.

Abstract:Image-guided surgery is crucial for achieving complete tumor resection, reducing postoperative recurrence, and improving patient survival. However, current clinical near-infrared fluorescent probes, such as indocyanine green (ICG), face two main limitations: 1) lack of active tumor targeting, and 2) short retention time in tumors, which restricts real-time imaging during surgery. To address these issues, we developed a near-infrared fluorescent probe capable of in situ nanofiber formation within tumor lesions. This probe actively targets the integrin α v β 3 receptors overexpressed on breast cancer cells and exhibits assembly/aggregation-induced retention effects at the tumor site, significantly extending the imaging time window. Additionally, we found that the probe's fluorescence intensity can be enhanced under receptor induction. Due to its excellent tumor specificity and sensitivity, 1FCG-FFGRGD not only identifies primary breast cancer but also precisely locates smaller lymph node metastases and detects sub-millimeter peritoneal metastases. In summary, this near-infrared probe, leveraging assembly/aggregation-induced retention effects, holds substantial potential for various biomedical applications.

Abstract:Purpose: Tumor-free surgical margin is crucial but challenging in breast-conserving surgery (BCS). Fluorescence imaging is a promising strategy for surgical navigation that can reliably assist the surgeon with visualization Of the tumor in real-time. Notably, finding an optimized fluorescent probe has been a challenging research topic. Herein, we developed a novel near-infrared (NIR) fluorescent probe based on tailored Hepatitis B Core virus-like protein (HBc VLP) and presented the preclinical imaging-guided surgery.Methods: The RGD-HBc160 VLP was synthesized by genetic engineering followed encapsulation of ICG via disassembly-reassembly. The applicability of the probe was tested for cell and tissue binding capacities through cell-based plate assays, xenograft mice model, and MMTV-PyVT mammary tumor transgenic mice. Subsequently, the efficacy of RGD-HBc160/ICG-guided surgery was evaluated in an infiltrative tumor-bearing mouse model. The protein-induced body's immune response was further assessed.Results: The prepared RGD-HBc160/ICG showed outstanding integrin αvβ3 targeting ability in vitro and in vivo. After intravenous administration of probe, the fluorescence guidance facilitated more complete tumor resection and improved overall survival Of the infiltrative tumor-bearing mice. The probe also showed the excellent capability to differentiate between benign and malignant breast tissues in the mammary tumor transgenic mice. Interestingly, the ingenious tailoring of HBc VLP could not only endow its tumor-targeting ability towards integrin αvβ3 but also significantly reduce the humoral and cellular immune response.Conclusion: The RGD-HBc160/ICG holds promise as an effective tool to delineate tumor margin. These results have translational potential to achieve margin-negative resection and improve the stratification of patients for a potentially curative.

Abstract:Noninvasive fluorescence detection of tumor-associated biomarker dynamics provides immediate insights into tumor biology, which are essential for assessing the efficacy of therapeutic interventions, adapting treatment strategies, and achieving personalized diagnosis and therapy evaluation. However, due to the absence of a single biomarker that effectively reflects tumor development and progression, the currently available optical diagnostic agents that rely on "always-on" or single pathological activation frequently show nonspecific fluorescence responses and limited tumor accumulation, which inevitably compromises the accuracy and reliability of tumor imaging. Herein, based on intramolecular charge transfer (ICT) and twisted intramolecular charge-transfer (TICT) hybrid mechanisms, we report a tandem-locked probe, NTVI-Biotin, for simultaneously specific imaging-guided tumor resection and ferroptosis-mediated tumor ablation evaluation under the coactivation of nitro reductase (NTR)/viscosity. The dual-stimulus-responsive design strategy ensures that NTVI-Biotin exclusively activates near-infrared (NIR) fluorescence signals upon interaction with both NTR and elevated viscosity levels through triggering ICT on while inhibiting the TICT process. Meanwhile, functionalization with a tumor-targeting hydrophilic biotin-poly(ethylene glycol) moiety enhances tumor accumulation. The probe's dual-response and tumor-targeting design minimizes nonspecific tissue activation, allowing for precise tumor identification and lesion removal with a superior tumor-to-normal tissue (T/N > 6) ratio. More importantly, NTVI-Biotin was capable of evaluating ferroptosis-mediated chemotherapeutics by real-time monitoring of the alternations of NTR/viscosity levels. The results reveal that the increased tumor signals of NTVI-Biotin following the combination of ferroptosis and chemotherapy correlate well with the tumor growth inhibition, demonstrating the potential of NTVI-Biotin to assess therapeutic efficacy.
Targeted Delivery of a Γ-Glutamyl Transpeptidase Activatable Near-Infrared-Fluorescent Probe for Selective Cancer Imaging

Abstract:The noninvasive and specific detection of cancer cells in living subjects has been essential for the success of cancer diagnoses and treatments. Herein, we report a strategy of combining an αvβ3-integrin-receptor-targetable ligand, c-RGD, with the γ-glutamyl transpeptidase (GGT)-recognizable substrate, γ-glutamate (γ-Glu), to develop a tumor-targeting and GGT-activatable near-infrared (NIR)-fluorescent probe for the noninvasive imaging of tumors in living mice. We demonstrated that the probe's fluorescence was off initially, but when the γ-Glu in the probe was specifically cleaved by GGT, the fluorescent product was released and could be selectively taken up by U87MG-tumor cells via αvβ3-receptor-mediated endocytosis. Remarkably, enhanced intracellular NIR fluorescence distributed mainly in the lysosomes was observed in the tumor cells only, showing that the probe was capable of differentiating the tumor cells from the GGT-positive, αvβ3-deficient normal cells. Moreover, the probe also showed a high selectivity for the real-time and noninvasive detection of GGT activity in xenograft U87MG tumors following iv administration. This study reveals the advantage of using a combination of receptor-mediated cell uptake and molecular-target-triggered activation to design molecular probes for improved cancer imaging, which could facilitate effective cancer diagnoses.

Abstract:Fluorescence imaging with tumor-specific fluorescent probe has emerged as a tool to aid surgeons in the identification and removal of tumor tissue. We report here a new lysosome-targeting fluorescent probe (NBOH) with BODIPY fluorephore to distinguish tumor tissue out of normal tissue based on different pH environment. The probe exhibited remarkable pH-dependent fluorescence behavior in a wide pH range from 3.0 to 11.0, especially a sensitive pH-dependent fluorescence change at pH range between 3.5 and 5.5, corresponding well to the acidic microenvironment of tumor cells, in aqueous solution. The response time of NBOH was extremely short and the photostability was proved to be good. Toxicity test and fluorescence cell imaging together with a sub-cellular localization study were carried out revealing its low biotoxicity and good cell membrane permeability. And NBOH was successfully applied to the imaging of tumor tissue in tumor-bearing mice suggesting potential application to surgery as a tumor-specific probe.
