Promotion of Biogas Production of Wheat Straw by Controling Composting Pretreatment Temperature

Abstract:Composting pretreatment has been widely used in biogas plant of agricultural straw for improving biotransformation rate of lignocellulosic materials. However, the mechanisms on composting pretreatment of agricultural straw for biogas production need more research. In this study, two experiments were conducted to evaluate the impacts of composting pretreatment on biogas production of wheat straw. In the first experiment, the wheat straw was directly used for composting and then composted straw was used in the biomethane potential (BMP) test. For the second one, wheat straw was sterilized by gamma ray radiation pretreatment and treated under different temperature with different time. Then the treated straw was used for the BMP test. The results of experiments indicated that large percentage of organic matter in wheat straw was degraded during composting process. The total solid (TS) loss rate of wheat straw was only 4.06%when composting pile temperature was less than 55℃. When composting pile temperature was set up at 55℃, the TS loss rate of wheat straw increased 22.45%after 10 days’ composting treatment. Gas data showed that biogas production rate of wheat straw was not improved obviously. The TS biogas yield of wheat straw increased with composting pile temperature and then decreased. The highest TS biogas yield of wheat straw of 349.92 mL/g was observed at 55℃of composting pile temperature, which is 7.56% higher than that of uncomposted wheat straw. There was no significantly improvement in biogas production between composted and uncomposted wheat straw during composting process for organic matter loss of wheat straw. However, longer composting time led to lower biogas yield of wheat straw. When composting pile temperature was kept at 55℃for 9 days, The TS biogas yield of composted wheat straw was only 66.58%. When the content of organic matter and material composition of wheat straw were changed, content of hemicellulose of wheat straw was decreased by 28.10%. Results of gas data of simulated composting experiment showed the same trend as the first experiment. The highest TS biogas yield of wheat straw of 342.36 mL/g was obtained at 55℃ with 8.35%, which is higher than that of control. Therefore the high temperature from composting process is an important factor of destruction of lignocellulose structure and improving biotransformation rate of wheat straw. When the composting pile temperature was set up at 55℃, anaerobic digested inoculums was added into straw pile and can be used for biogas production.
Effect of Ensiling Density and Storage Temperature on Fermentation Quality, Bacterial Community, and Nitrate Concentration of Sorghum-Sudangrass Silage

Abstract:This study aimed to evaluate the fermentation quality, bacterial community, and nitrate content of sorghum-sudangrass silage with two ensiling densities [550 kg fresh weight (FW)/m 3 (low density, LD) and 650 kg FW/m 3 (high density, HD)] stored at two temperatures [10°C (low temperature, LT) and 25°C (normal temperature, NT)] for 60 days. The fermentation parameters, microbial counts, bacterial community, nutritional composition, and nitrate and nitrite levels were assessed. The pH and ammonia nitrogen (N) in all silages were below 4.0 and 80 g/kg total N, respectively. Compared with LT treatments, NT treatments had lower pH and lactic acid (LA) bacteria and yeasts counts and contained higher LA and LA/acetic acid (LA/AA) ( p < 0.05). The LT-LD contained more ammonia–N than LT-HD ( p < 0.05) and had higher nitrate and lower nitrate degradation than other treatments ( p < 0.05). Lactobacillus was the most dominant genus with all treatments (57.2–66.9%). The LA, LA/AA, and abundances of Pantoea , Pseudomonas , and Enterobacter in the silage negatively correlated with nitrate concentration and positively correlated with nitrate degradation ( p < 0.05). Moreover, pH and ammonia–N were positively correlated with nitrate concentration and negatively correlated with nitrate degradation ( p < 0.05). Overall, all silage had satisfactory fermentation quality, and the silage with HD and NT had better fermentation quality and higher nitrate degradation. The bacterial communities in all silages were dominated by Lactobacillus . The nitrate degradation during the fermentation process might be related to the fermentation quality and the activity of Pantoea , Pseudomonas , and Enterobacter in silage.

Abstract:Two lactic acid bacteria (LAB) strains with different ferulic acid esterase (FAE) activities were isolated: Lactobacillus farciminis (LF18) and Lactobacillus plantarum (LP23). The effects of these strains on the fermentation quality, in vitro digestibility and phenolic acid extraction yields of sorghum (Sorghum bicolor L.) silage were studied at 20, 30 and 40 °C. Sorghum was ensiled with no additive (control), LF18 or LP23 for 45 days. At 40 °C, the lactic acid content decreased, whereas the ammonia nitrogen (NH3-N) content significantly increased (p < 0.05). At all three temperatures, the inoculants significantly improved the lactic acid contents and reduced the NH3-N contents (p 0.05). The LP23 group exhibited higher phenolic acid extraction yields at 30 °C (p < 0.05), and the corresponding yields of the LF18 and control groups were improved at 40 °C (p < 0.05). FAE-producing LABs might partially ameliorate the negative effects of high temperature and improve the fermentation quality of sorghum silage. The screened FAE-producing LABs could be candidate strains for preserving sorghum silage at high temperature, and some further insights into the relationship between FAE-producing LABs and ensiling temperatures were obtained.

Abstract:OBJECTIVE: In tropical regions, as in temperate regions where seasonality of forage production occurs, well-preserved forage is necessary for animal production during periods of forage shortage. However, the unique climate conditions (hot and humid) and forage characteristics (high moisture content and low soluble carbohydrate) in the tropics make forage preservation more difficult. The current study used natural ensiling of tropical forage as a model to evaluate silage characteristics under different temperatures (28°C and 40°C).METHODS: Four tropical forages (king grass, paspalum, white popinac, and stylo) were ensiled under different temperatures (28°C and 40°C). After ensiling for 30 and 60 days, samples were collected to examine the fermentation quality, chemical composition and microbial community.RESULTS: High concentrations of acetic acid (ranging from 7.8 to 38.5 g/kg dry matter [DM]) were detected in silages of king grass, paspalum and stylo with relatively low DM (ranging from 23.9% to 30.8% fresh material [FM]) content, acetic acid production was promoted with increased temperature and prolonged ensiling. Small concentrations of organic acid (ranging from 0.3 to 3.1 g/kg DM) were detected in silage of white popinac with high DM content (50.8% FM). The microbial diversity analysis indicated that Cyanobacteria originally dominated the bacterial community for these four tropical forages and was replaced by Lactobacillus and Enterobacter after ensiling.CONCLUSION: The results suggested that forage silages under tropical climate conditions showed enhanced acetate fermentation, while high DM materials showed limited fermentation. Lactobacillus and Enterobacter were the most probable genera responsible for tropical silage fermentation.

Abstract:Abstract In practical of silage production, variations in the fermentation parameters of silage might be attributable to changes in weather data, especially for ambient temperature. The objective of this study was to investigate how the changed temperature of outside weather condition affects distributions of pH and fermentation products in large round bale silage. The bale silages (1.22 m × 1.25 m ø; density, about 168.9 kg dry matter [DM]/m 3 ) were produced by BR6090 Combi from New Holland Agriculture, wrapped with six layers of stretch film, stored at ambient temperature (2–20°C) and constant temperature (10°C; 15°C; 25°C) for 3, 9, 15, 45 and 90 days. Compared with other silages, silage stored at 25°C had higher ( p < 0.05) lactic acid content, lower ( p < 0.05) pH, butyric acid content and ammonia‐nitrogen (N) ratio of total N. Fluctuant storage temperature limited lactic acid fermentation process with delayed pH decline and continuous butyric production and crude protein (CP) degradation. The differences of pH, lactic acid, propionic acid and ammonia‐N were considerably less evident in the centre of silages stored at different temperatures, and they increased towards the boundaries. The data shows that fluctuant storage temperature had undesirable effect on the fermentation quality of silage, especially for the external part of silo; relatively high and stable storage temperature for initial stage of ensiling process was necessary, but did not promote a positive response to the homogeneous distributions of pH and fermentation products in large round bale silage.

Fermentative products and bacterial community structure of C4 forage silage in response to epiphytic microbiota from C3 forages

Abstract:Objective: The observation that temperate C3 and tropical C4 forage silages easily produce large amounts of ethanol or acetic acid has puzzled researchers for many years. Hence, this study aimed to assess the effects of epiphytic microbiota from C3 forages (Italian ryegrass and oat) on fermentative products and bacterial community structure in C4 forage (sorghum) silage. Methods: Through microbiota transplantation and γ-ray irradiation sterilization, the irradiated sorghum was treated: i) sterile distilled water (STSG); ii) epiphytic microbiota from sorghum (SGSG); iii) epiphytic microbiota from Italian ryegrass (SGIR); iv) epiphytic microbiota from oat (SGOT). Results: After 60 days, all the treated groups had high lactic acid (>63.0 g/kg dry matter [DM]) contents and low pH values (<3.70), acetic acid (<14.0 g/kg DM) and ammonia nitrogen (<80.0 g/kg total nitrogen) contents. Notably, SGIR (59.8 g/kg DM) and SGOT (77.6 g/kg DM) had significantly (p<0.05) higher ethanol concentrations than SGSG (14.2 g/kg DM) on day 60. After 60 days, Lactobacillus were predominant genus in three treated groups. Higher proportions of Chishuiella (12.9%) and Chryseobacterium (7.33%) were first found in silages. The ethanol contents had a positive correlation (p<0.05) with the abundances of Chishuiella, Acinetobacter, Stenotrophomonas, Chryseobacterium, and Sphingobacterium. Conclusion: The epiphytic bacteria on raw materials played important roles in influencing the silage fermentation products between temperate C3 and tropical C4 forages. The quantity and activity of hetero-fermentative Lactobacillus, Chishuiella, Acinetobacter, Stenotrophomonas, Chryseobacterium, and Sphingobacterium may be the key factors for the higher ethanol contents and DM loss in silages.
Effect of Different Regions and Ensiling Periods on Fermentation Quality and the Bacterial Community of Whole-Plant Maize Silage

Abstract:This study aimed to explore the changes in the microbial community on the silage material surface and during the ensiling process of whole-plant maize in different regions. Whole-plant maize silages were sampled in Ziyun, Guanling, and Weinning counties within warm and humid climate areas in southern China. Silages were sampled at 0, 2, 5, 10, 20, and 45 days during ensiling. The nutritional components, fermentation properties, and microbiomes were examined to evaluate the influence of sampling area and fermentation time on the quality of silage. The results showed that the pH values of all silages significantly decreased (<4.2 at ensiling day 2) during fermentation and all silages achieved satisfactory fermentation at 45 days. Butyric acid was not detected during ensiling, and the contents of acetic acid and ammonia nitrogen in the final silages were below 6 g/kg DM and 50 g/kg total nitrogen, respectively. Weissella was the dominant epiphytic bacteria of raw material in Ziyun and Weinning, while Lactobacillus was prevalent in Guanling. Lactobacillus dominated the ensiling process, and its abundance significantly increased with increasing fermentation time in the three groups. Lactobacillus was negatively correlated with pH of all silages ( p < 0.05) and positively correlated with lactic acid, propionic acid and acetic acid ( p < 0.05). Furthermore, the bacterial community was significantly correlated with environmental factors. Altitude had a highly positive correlation with the abundance of Stenotrophomonas , Chryseobacterium , and Massilia ( p < 0.01), while precipitation was negatively correlated with these bacteria. The humidity and average temperature significantly influenced the Lactobacillus and Weissella abundances of fresh whole-plant maize. During the ensiling process, the silages from three regions had similar bacterial dynamic changes, and the Lactobacillus formed and maintained good fermentation characteristics in whole-plant maize silage.

Abstract:This study aims to investigate the effects of adding Lactobacillus buchneri (LB), Lactobacillus brevis (LBR) and Bacillus subtilis (BS) on the fermentation quality, nitrate degradation and bacterial community of sorghum-sudangrass silage. The results showed that the addition of LB significantly increased the pH and acetic acid content ( p < 0.05), but high-quality silage was obtained. The addition of LBR and BS improved the fermentation quality of sorghum-sudangrass silage. The use of additives reduced the nitrate content in sorghum-sudangrass silage. The LB group increased the release of N 2 O at 3–7 days of ensiling ( p < 0.05), and LBR and BS increased the release of N 2 O at 1–40 days of ensiling ( p < 0.05). On the first day of ensiling, all silages were dominated by Weisslla , over 3 days of ensiling all silages were dominated by Lactobacillus . Acinetobacter , Serratia , Aquabacterium, and unclassified_f_enterobacteriaceae showed significant negative correlations with nitrate degradation during sorghum-sudangrass ensiling ( p < 0.05). The BS and LBR groups increased the metabolic abundance of denitrification, dissimilatory nitrate reduction, and assimilatory nitrate reduction ( p < 0.05). Overall, the additive ensures the fermentation quality of sorghum-sudangrass silage and promotes the degradation of nitrate by altering the bacterial community.

Insight of CO2 and ethanol emission from maize silage: A case study with real-time identification of aerobic and anaerobic microbial respiration using a multi-sensor-fusion method

Abstract:Silage is produced worldwide for both livestock feeding and biogas production. Sustainable silage production requires characterization and mitigation of potential effects on environmental quality, particularly from greenhouse gas emissions during the production cycle. Ex-situ sampling has demonstrated that major emissions are carbon dioxide (CO2) and ethanol (EtOH). In-situ gas measurements from farm silo and bale silage are rare and may be important to improve our knowledge of the physical and biochemical causes, and constraints on these gas emissions. This study focused on tracking the kinetics of CO2 and EtOH emissions from bale maize silage, with real-time identification, quantification and separation of aerobic and anaerobic respiratory components in the period following opening of the silage. For this, an automatic multi-sensor gas-flux chamber (AMGC) was developed. Three bales (mean weight: 890 kg) of maize silage were tested (n = 3). Oxygen (O2) and temperature (Tsi) sensors were co-located at 10- and 20-cm behind the open face of the bales. Over the two weeks of the experiment we observed: (i) significant initial discharge of CO2 across the open face (1.68-2.55 mol m-2 h-1) and EtOH (0.027-0.034 mol m-2 h-1); (ii) peak CO2 emission occurred when O2 concentration (10 cm depth) was 3∼8% vol., while peak EtOH emission occurred below 2% vol. O2, (iii) dynamic conversion of O2 to CO2 from aerobic respiration; and (iv) the cumulative emission of EtOH during the anaerobic period was 4-6 times greater than that during aerobic plus semi-aerobic periods. These novel measurements provide mechanistic understanding, and may facilitate improved management of silage production to minimize environmental impact and aerobic loss of silage.
Impacts of Low Temperature and Ensiling Period on the Bacterial Community of Oat Silage by SMRT

Abstract:The objective of this study was to investigate how storage temperatures influence the bacterial community of oat silage during the ensiling process via PacBio single molecule, real-time sequencing technology (SMRT). Forage oat was ensiled at four different temperatures (5 °C, 10 °C, 15 °C, and 25 °C) and ensiling days (7, 14, 30, and 60 days). With the rise in storage temperature, the lactic acid content showed an increased trend. Acetic acid production was observed highest in silage fermented at 5 °C compared with other treatments, and Enterococcus mundtii was also the dominant bacterial species. Lactiplantibacillus pentosus and Loigolactobacillus rennini were exclusively detected in silages at 10 °C, 15 °C, and 25 °C, and dominated the fermentation after 60 days of ensiling at 10 °C and 25 °C, respectively. In addition, L. pentosus, L. rennini, and E. mundtii may be related to changes in the fermentation products due to the differences in ensiling temperature. In conclusion, results of this study improve our understanding of the complicated microbial composition underlying silage fermentation at low temperatures, which might contribute to target-based regulation methods for enhancing silage quality and developing new inoculants.

Abstract:Abstract The objective was to compare the effect of applying silage additives to male sterile sorghum hybrids on temperature production during ensiling time (30 d) and at feed-out period after 30 or 90 d of ensiling period. Two male-sterile sorghum hybrids; F430 and F465, with and without BMR12 trait, respectively (Richardson Seed Ltd.); were randomly allocated in plots blocked by irrigation section in a dairy farm in West Texas. During harvest, duplicate samples from each plot were obtained, placed on plastic trays, and transported to one of three stations to be treated with chemical silage preservative (SG, SiloGuard), heterofermentative bacteria inoculant with mold inhibitor solution (SM, Silage Maker 250) or distilled water (DW, control). Treatments were applied with spray bottles following a protocol that consisted of two sets of 5 sprays (1.1 mL each and 11 mL total) followed by mixing of the forage. Personnel applying the treatment were blind as to which treatment they were applying and all spray bottles, trays and tables from each station were color coded to prevent cross contamination. After treated, 3.63 kg of forage was packed in three consecutive steps using a manual press on 4.7L buckets (Uline) and a temperature data logger was placed in the center of each of the 30 d mini silos. The amount of material packed was estimated to achieve a 230 kg of DM/m3 compaction at 30% DM of the forage. At opening, the material from each silo was moved to a tray and mixed to be homogenized and exposed to oxygen. Then, the material was added back to its correspondent bucket until it filled up to the border without compressing. Heat production during the feed-out period and air-stability were assessed for both ensiling periods, whereas fermentation heat production during ensiling period was only assessed for 30 d silos. Statistical analysis was performed using mixed linear regression models (GLIMMIX procedure of SAS) with ensiling period, hybrid, treatment and the interaction of day and treatment included in the model as independent variables. Fourth root and natural log transformation were performed to obtain a normal distribution of residuals, back transformed LSM and 95% CI are reported in Table 1. Treatment effected the silage temperature during the feed-out period (P < 0.0001) and air-stability test (P = 0.02). Whereas a tendency was observed for daily heat production during the ensiled period (P = 0.07). Male-sterile sorghum silage treated with SG had the least temperature during the ensiling period and longer air-stability than SM.

Abstract:To better understand the ensiling characteristics of sorghum stalk, the dynamic changes of fermentation parameters, nutrient components and bacterial community of sorghum stalk silage were analyzed by intermittently sampling on day 0, 1, 3, 7, 14, 28, and 56 of ensiling duration. The results showed that high-moisture sorghum stalk was well preserved during ensiling fermentation, with the DM loss of 4.10% and the little difference between the nutrients of sorghum stalk before and after ensiling. The pH value of silage declined to its lowest value of 4.32 by Day 7 of ensiling, and other fermentation parameters kept steady since Day 28 of ensiling. The amplicon sequencing analysis revealed that the alpha diversity parameters of silage bacterial community including Shannon index, observed features, Pielou evenness and Faith PD gradually declined ( P < 0.01) with ensiling duration. Principal coordinate analysis (PCoA) revealed that bacterial profiles of raw material would experience a succession becoming a quite different community during ensiling fermentation. Taxonomic classification revealed a total of 10 and 173 bacterial taxa at the phylum and genus level, respectively, as being detected with relative abundances higher than 0.01% and in at least half samples. LEfSe analysis revealed that 26 bacterial taxa were affected by sampling timepoint ( P < 0.05 and LDA score > 4). When focusing on the dynamic trend of silage bacterial taxa, lactic acid bacteria successfully dominated in the bacterial community on Day 1 of ensiling, and the bacterial community almost came to a plateau by Day 28 of ensiling, with Lactobacillus and Leuconostoc as the dominant genera. In a word, the succession of fermentation parameters, nutrient components and bacterial community indicate a successful dominance establishment of LAB and a fast advent of fermentation plateau, suggesting that high-moisture sorghum stalk can be ensiled directly, but the pH of mature silage is a little high.

Abstract:Sweet sorghum is an important forage in arid and semi-arid climatic regions. This study aimed to reveal the fermentation weight loss (FWL), fermentation quality, and bacterial community of ensiling of sweet sorghum with lactic acid bacteria LAB; ( Lactiplantibacillus plantarum and Lentilactobacillus buchneri ) at different silo densities. For this study, sweet sorghum was harvested at the first spikelet of inflorescence stage and ensiled without or with LAB (CK or L) in polyethylene laboratory-scale silos (diameter, 20 cm; height, 30 cm) at densities of 650 (CK_650 and L_650), 700 (CK_700 and L_700), and 750 kg/m 3 (CK_750 and L_750), respectively. The FWL, fermentation quality, microbial counts, and bacterial community of the silage were assessed after 100 days of ensiling. L_750 had a lower FWL than CK_650, _700, and _750 after 100 days of ensiling ( P < 0.005), and the FWL was affected by silo density and inoculating LAB ( P < 0.005). All silages had low pH (<4.0) and ammonia nitrogen content (<50 g/kg total nitrogen) and did not contain propionic and butyric acids; moreover, inoculating LAB increased lactic and acetic acids ( P < 0.005). Bacterial communities in inoculated and uninoculated silages were clustered together, respectively, and clearly separated from each other. The total abundance of Lactiplantibacillus and Lentilactobacillus in fresh forage was <1%. Lactiplantibacillus had the highest abundance in all silages (from 71.39 to 93.27%), followed by Lentilactobacillus (from 3.59 to 27.63%). Inoculating LAB increased the abundance of Lentilactobacillus in each silo density ( P < 0.005) and decreased Lactiplantibacillus in the silage in densities of 700 and 750 kg/m 3 ( P < 0.005); moreover, increasing silo density decreased Lactiplantibacillus abundance and increased Lentilactobacillus abundance in inoculated silages ( P < 0.005). Overall, sweet sorghum silage showed satisfactory fermentation quality, with a density of no <650 kg/m 3 , and inoculating LAB improved fermentation quality and reduced FWL. Lactiplantibacillus and Lentilactobacillus presented as minor taxa in fresh sweet sorghum and dominated the bacterial community of all silages. Inoculating LAB was the main factor affecting the bacterial community of sweet sorghum silage. Moreover, inoculating LAB and increasing silo density can contribute to the decreasing Lactiplantibacillus abundance and increasing Lentilactobacillus abundance.

Abstract:Temperature is known to have a clear influence on the formation of biogenic amines during fermentation. To improve the quality of oat silage, the impact of ensiling temperature on the fermentation, microbiological and chemical characteristics, as well as biogenic amines (BAs) was investigated. Vacuum bag mini silos of oat forage were incubated at four different temperature levels (10, 20, 30 and 37 °C) and opened on day 0, 1, 3, 7, 15 and 60. All oat silages were sampled to evaluate the fermentation quality and biogenic amine production. Results showed that putrescine, cadaverine and tyramine were the most prevalent biogenic amines in oat silage, representing approximately about 90% of the total biogenic amines (TBAs) investigated. Ensiling increased the β–phenylethylamine, putrescine, cadaverine, histamine and tyramine accumulation in oat silage at the four incubation temperatures. On day 60, the β–phenylethylamine, cadaverine, histamine, tyramine and TBAs levels at a high temperature (37 °C) were significantly higher than those at a lower temperature (10, 20 and 30 °C); 10 °C fermentation increased the putrescine content in oat silage. A closed relationship between fermentation properties and BAs showed that the silages containing higher lactic acid, propionic acid and ammonia nitrogen and lower pH value had more BA content in oat silage. In conclusion, the ensiling process caused a significant increase in the amounts of BAs, except spermidine and spermine. The oat silage made in elevated temperature (30 and 37 °C) environments may accumulate more BAs than at a low temperature (10 °C), but low temperature (10 °C) fermentation may increase the putrescine levels in silage. The results suggested that ensiling at the proper temperature could retard BA formation and enhance the quality of oat silage.

Abstract:In this study, the effects of epiphytic microbiota from different forages on the fermentation characteristics and microbial community structure of forage sorghum silage were investigated. The gamma irradiated sterilized forage sorghum was treated through sterile water, epiphytic microbiota of forage sorghum (FSm), Sudan grass (SDm), Napier grass (NPm), and maize (MZm). NPm and SDm inoculated silages showed similar pH value and lactic acid (LA) and acetic acid (AA) contents at day 3 and 60 of ensiling. The final silage of FSm and MZm showed lower (p < 0.05) pH and AA content and a higher LA content compared to the NPm and SDm silages. Bacterial species from the Weisella genus were predominantly present in FSm, NPm, and SDm, while Lactococcus dominated the MZm silage during early ensiling. Lactobacillus was predominant in all inoculated terminal silages. Overall, the four inoculated microbiota decreased the pH value of silage and were dominated by lactic acid bacteria (LAB); however, the NPm and SDm treatments resulted in comparatively higher AA contents which could have an inhibitory effect on the secondary fermentation developed by the yeast and enhanced the aerobic stability of forage sorghum silage.
Exploring the Effects of Different Bacteria Additives on Fermentation Quality, Microbial Community and In Vitro Gas Production of Forage Oat Silage

Abstract:Bacterial inoculants are considered as a good choice for successful ensiling, playing a key role in improving the silage quality. However, the potential of different bacteria, especially the propionic acid bacteria, in forage oat ensiling is yet to be explored. Therefore, the purpose of this study was to investigate the regulation effects of different bacterial additives on the fermentation quality of forage oat silage. Four additives (Lactiplantibacillus plantarum F1, LP; Lacticaseibacillus 0rhamnosus XJJ01, LR; Lacticaseibacillus paracasei XJJ02, LC; and Propionibacterium acidipropionici 1.1161, PP; without additives, CK) were inoculated in forage oat silage, and the fermentation quality and organic compounds were determined after 60 days of ensiling. Notably, LR showed higher dry matter preservation compared to other additives and CK. In addition, LP and LR showed strong lactic acid synthesis capacity, resulting in lower pH compared to other additives and CK. The treatments of PP and LC increased the bacterial diversity in silage, while the bacterial community in the LR group was different from that in other groups. In addition, the PP- and LC-treated oat silage showed significantly lower total in vitro gas production and a lower methane content. These results suggest that LP is more favorable for producing high-quality oat silage than LR, LC, or PP. Both the PP- and LC- treated oat silage may reduce rumen greenhouse gas emissions.

Abstract:Carbon dioxide (CO 2 ) is a primary greenhouse gas and the main cause of global warming. Respiration from plant cells and microorganisms enables CO 2 to be produced during ensiling, a method of moist forage preservation applied worldwide. However, limited information is available regarding CO 2 emissions and mitigation during ensiling. Pyroligneous acid, a by-product of plant biomass pyrolysis, has a strong antibacterial capacity. To investigate CO 2 production and the influence of pyroligneous acid, fresh stylo, and rice straw were ensiled with or without 1% or 2% pyroligneous acid. Dynamics of the fermentation characteristics, CO 2 production, and bacterial communities during ensiling were analyzed. Pyroligneous acid increased the lactic acid content and decreased the weight losses, pH, ammonia-N content, butyric acid content, and coliform bacterial numbers (all P < 0.05). It also increased the relative abundance of Lactobacillus and decreased the relative abundances of harmful bacteria such as Enterobacter and Lachnoclostridium . Adding pyrolytic acids reduced the gas production, especially of CO 2 . It also increased the relative abundances of CO 2 -producing bacterial genera and of genera with the potential for CO 2 fixation. In conclusion, adding pyroligneous acid improved the fermentation quality of the two silages. During ensiling, CO 2 production was correlated with bacterial community alterations. Using pyroligneous acid altered the bacterial community to reduce CO 2 production during ensiling. Given the large production and demand for silage worldwide, application of pyroligneous acid may be an effective method of mitigating global warming via CO 2 emissions.

Abstract:This study investigated the effects of developmental stage and store time on the chemical composition, microbial community, co-occurrence networks and fermentation characteristics of sweet sorghum silage. Sweet sorghum harvested at two developmental stages (heading stage, S 1 ; hard dough stage, S 2 ) was treated as follows: (I) natural fermentation of S 1 (NS 1 ); (II) natural fermentation of S 2 (NS 2 ) and ensiled for 1, 3, 7, 15, 30 and 60 days. After 60 days of ensiling, NS 2 silage had higher lactic acid concentration and the ratio of lactic to acetic acid, and lower pH value and acetic acid concentration than NS 1 silage. Meanwhile, NS 2 silage also had higher ammonia nitrogen (NH 3 -N) content than NS 1 silage, but the NH 3 -N content of both treatments was lower than 100 g/kg TN. Leuconostoc and Lactococcus were respectively dominant in both 7-day NS 1 and NS 2 silages, while Lactobacillus was the most abundant genus in 30-day NS 1 and NS 2 silages. The developmental stage altered the bacterial co-occurrence networks of fresh and ensiled sweet sorghum. Spearman's correlation heatmap showed that the higher lactic acid content and ratio of lactic to acetic acid in NS 2 silage could be associated with the higher water-soluble carbohydrate content in S 2 material, the higher abundance of Lactococcus in the initial phase of ensiling and the higher abundance of Lactobacillus in the late phase of ensiling. These results concluded that the sweet sorghum harvested at the hard dough stage is preferred for silage production over the sweet sorghum harvested at the heading stage. Microbial succession pattern was studied on fresh and ensiled samples. Growth stage affected the chemical and microbial parameters of sweet sorghum. Epiphytic microbiota was correlated with the chemical composition of forage. Metagenomic sequencing showed Lactobacillus was dominated in all silages. Growth stage accelerated the fermentation of sweet sorghum silage.

Abstract:Dried grass silage (GS) was pre-treated at different NaOH loading rates (1%, 2.5%, 5% and 7.5% by volatile solids (VS) mass in grass silage) and temperatures (20 °C, 60 °C, 100 °C and 150 °C) to determine effects on its bio-degradability in terms of the hydrolysis yield and degradation of ligno-cellulosic materials for biogas production. At 100 °C and the four NaOH loadings, up to 45% of the total COD was solubilised and up to 65.6%, 36.1% and 21.2% of lignin, hemicellulose and cellulose were removed, respectively; biological methane production potentials obtained were 359.5, 401.8, 449.5 and 452.5 ml CH₄/g VS added, respectively, being improved by 10-38.9% in comparison with untreated GS. VS removals following anaerobic digestion were 67.6%, 76.9%, 85.3%, 95.2% and 96.7% for untreated GS and GS treated at the four NaOH loadings, respectively. 100 °C and the NaOH loading rate of 5% is recommended as a proper GS pre-treatment condition.
The greenhouse gas emission potential and phytotoxicity of biogas slurry in static storage under different temperatures

Abstract:With the booming development of biogas industry to treat organic waste in China, the by-product of biogas slurry was accompanied with a huge amount. Static storage process of biogas slurry was normally operated under different seasons before application to land which would cause nutrition decomposition and greenhouse gas emission. Thus, the aim of this study was to investigate the nutrition decomposition, greenhouse gas emission (CH4 and N2O), and phytotoxicity of biogas slurry under different static temperatures, furthermore to illuminate the network among them and functional microorganism. According to the results, higher temperature at 30 °C contributed to fast and complete degradation of COD. In addition, more quantity of NH4+ conversion and NO3- formation appeared at 30 °C. These factors resulted in relatively less crop toxicity together. CH4 was the dominant greenhouse gas emission than N2O and was highest in 30 °C treatment with total emission of 273.7 L/(m3·d) and greenhouse gas emission of 20.01 kg CO2e (carbon dioxide equivalent). Lower temperature was conductive to N reservation and reduction of greenhouse gas emission, but making against with stabilization of organic matter and crop safety. At the same dilution times (≤3) of biogas slurry with deionized water, higher temperature at 30 °C could reduce 30 days of storage time, but 10 °C was still unsafe for crop. Structural equation model was further illustrated the positive effect of temperature on NO3-, CH4, GI, and N2O and negative on COD and NH4+. These results could help to monitor the environmental risk, evaluate the maturity, guide the irrigation scheme, and regulate the static storage condition of biogas slurry under different seasons.
