Abstract:Laccases hold great potential for biotechnological applications, particularly in environmental pollutant remediation. Laccase activity is governed by the solvent environment, and ionic liquids (ILs) emerge as a versatile solvent for activation or stabilization of enzymes. Herein, effects of cholinium-based ILs formulated with carboxylic acids, inorganic acid, and amino acids as anionic species, on the catalytic activity of laccase from Trametes versicolor were investigated by experimental and computational approaches. Experimental results showed that laccase activity was enhanced by 21.39 % in 0.5 M cholinium dihydrogen citrate ([Cho][DHC]), in relation to the laccase activity in phosphate buffer medium. However, cholinium aminoate ILs negatively affected laccase activity, as evidenced by the partial deactivation of laccase in both cholinium glycinate and cholinium phenylalaninate, at concentrations of 0.1 M and 0.5 M, respectively. Molecular dynamics studies revealed that the enhancement of laccase activity in [Cho][DHC] might be attributed to the highly stabilized and compact structure of laccase, facilitating a better internal electron transfer during the laccase-substrate interactions. Enhanced catalytic performance of laccase in [Cho][DHC] was postulated to be driven by the high accumulation level of dihydrogen citrate anions around laccase's surface. [Cho][DHC] holds great promise as a cosolvent in laccase-catalyzed biochemical reactions.
Screening of newly synthesized Ionic Liquids and Deep eutectic solvent for enhancing the activity and stability of fungal laccase: empowering kinetic parameters and long-term conservation

Abstract:Ionic Liquids (ILs) and Deep Eutectic Solvents (DES) are currently with great interest due to their highly interesting physicochemical properties, it was studied throughout many chemical and biological approaches and it was used intensively in biocatalysis and biomedicine. In the present study, 10 newly synthesized ILs and 04 newly synthesized DES has been used to study their effect on laccase enzyme. Hence ILs and DES have been screened in conventional reaction buffered medium (25°C and pH 7.0) and using ABTS and DMP as substrates to study the catalytic ability and the stability of the enzyme. From 14 newly synthetized items, only three water-miscible ILs ([BMIM][Ac], [Chol][Ac], and [Chol][Prop]) and one DES ([Chol][Ac.Ox]) have shown interest for improving the enzyme activity and stability. Among them, the [BMIM][Ac] has shown impressive empowering power for the laccase activity (almost 20 folds enhancement versus control) however it was not efficient to protect the enzyme against high temperature or long conservation. The two Choline based ILs, [Chol][Ac] and [Chol][Prop], have allowed the increase of laccase activity with a rate of 1.7 folds and 7.8 folds, respectively and it was shown interesting properties to protect the enzyme against temperature and storage. Regarding the DES [Chol][Ac.Ox], it was improved the laccase at about 8.9 folds but this rate has not been stable along the experiment due to the high hydrophobic nature of the solution, it could be more useful when applied in high temperature. The four screened items in general improved the activity of the enzyme especially at pH7.0 and shown a great sensitivity against temperature. By kinetic studies versus [BMIM][Ac], it was demonstrated that the IL can improve the laccase activity by increasing the catalytic markers (Vm and Kcat), however a decrease of the affinity has been notified by Km decreasing, which testify that the interaction of the enzyme with ILs can affect the proteinic structure in aqueous medium.

Abstract:The ability of laccases to work effectively in properly selected aqueous-organic solutions significantly expands the scope of their application, making them commercially attractive and more competitive. However, it must be considered that molecules of organic solvent are very aggressive towards enzymes and destroy weak non-covalent interactions that maintain the spatial structure of the protein. In this work, we quantitatively described the influence of the nature of water-miscible organic solvents and their concentration on the activity and stability of Trametes versicolor laccase during the oxidation of ABTS (2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) through the maximum rates and the Michaelis constants K M . The obtained kinetic parameters make it possible to compare the effect of various organic solvents on laccase and select the most optimal one, which minimizes the decrease in enzyme activity, maintains its stability and allows repeated use. Among the solvents studied, polyhydroxy compounds have the least negative effect on laccase activity; however, in all cases, a high concentration of organic solvent (≥30% (v/v)) leads to suppression of enzymatic activity. It has been shown that the longer the alkyl chain (hydrophobic fragment of the structure), the stronger the negative effect of alcohols on enzymes. The most toxic solvent for Trametes versicolor laccase is isopropanol. In addition, numerous examples of various uses of Trametes versicolor in aquatic-organic environments are discussed here.

Abstract:The micellization of choline-based anionic surface-active ionic liquids (SAILs) having lauroyl sarcosinate [Sar]-, dodecylsulfate [DS]- and deoxycholate [Doc]- as counter ion has been investigated in the aqueous medium. The density functional theory (DFT) has been employed to investigate net interactional energy (Enet), extent of non-covalent interactions and band gap of choline-based SAILs. The critical micelle concentration (cmc) along with various parameters related to surface adsorption, counter-ion binding (β), and polarity of the core of micelles has been deduced employing surface tension measurements, conductometric titrations and fluorescence spectroscopy, respectively. Dynamic light scattering (DLS) equipped with zeta-potential measurement set-up and small angle neutron scattering (SANS) have been utilized to predict size, zeta-potential and morphology, respectively of the formed micelles. The thermodynamic parameters such as standard Gibb's free energy (∆G_mic^o) and standard enthalpy (∆H_mic^o) change of micellization have been calculated using isothermal titration calorimetry (ITC). On comparing with the sodium salt analogues, it has been established that the micellization is predominantly governed by the extent of hydration of [Cho]+, head groups of respective anions and degree of counter-ion binding (β). Considering the concentration dependence of enzyme-SAIL interactions, the aqueous solutions of synthesized SAILs at two different concentrations (below and above cmc) have been utilized as medium for testing the enzymatic activity of cellulase. The activity of cellulase has been found to ~7- to ~13-fold higher as compared to that observed in buffer in monomeric solution of SAILs and follows the order, [Cho][Sar] > [Cho][DS] > [Cho][Doc]. In micellar solution, ~ 4- to 5-fold increase in enzymatic activity has been observed.

Abstract:Enzymatic polymerization of lignosulfonate (LS) has a high potential for various applications. Here, the effect of different ions in low concentrations on enzymatic polymerization of LS was investigated, including salt solutions consisting of mono‐ and dicarboxylic acids, sulfate, phosphate and chloride with sodium as counter ion. LS polymerization was followed by viscometry and size exclusion (SEC) chromatography. Interestingly, there was only a small effect of ions on the activity of the laccase on standard substrate ABTS, while the effect on polymerization of LS was substantially different. The presence of acetate led to a 39% higher degree of polymerization (DP) for LS. Small angle X‐ray scattering (SAXS) revealed that the structure of the enzyme was largely unaffected by the ions, while the determination of the zeta potential showed that those ions conveying higher negative surface charges onto LS particles showed lower DPs, than those not affecting the surface charge. Further, electron paramagnetic resonance (EPR) spectroscopy showed 5‐times higher intensity in phenoxyl radicals for the monovalent ions compared to the divalent ones. It was concluded that the DPs of LS could be tuned in the presence of certain ions, by facilitating the interaction between the laccase substrate‐binding site and the LS molecules.

Molecular dynamics investigation of the ionic liquid/enzyme interface: Application to engineering enzyme surface charge

Abstract:Molecular simulations of the enzymes Candida rugosa lipase and Bos taurus α-chymotrypsin in aqueous ionic liquids 1-butyl-3-methylimidazolium chloride and 1-ethyl-3-methylimidazolium ethyl sulfate were used to study the change in enzyme-solvent interactions induced by modification of the enzyme surface charge. The enzymes were altered by randomly mutating lysine surface residues to glutamate, effectively decreasing the net surface charge by two for each mutation. These mutations resemble succinylation of the enzyme by chemical modification, which has been shown to enhance the stability of both enzymes in ILs. After establishing that the enzymes were stable on the simulated time scales, we focused the analysis on the organization of the ionic liquid substituents about the enzyme surface. Calculated solvent charge densities show that for both enzymes and in both solvents that changing positively charged residues to negative charge does indeed increase the charge density of the solvent near the enzyme surface. The radial distribution of IL constituents with respect to the enzyme reveals decreased interactions with the anion are prevalent in the modified systems when compared to the wild type, which is largely accompanied by an increase in cation contact. Additionally, the radial dependence of the charge density and ion distribution indicates that the effect of altering enzyme charge is confined to short range (≤1 nm) ordering of the IL. Ultimately, these results, which are consistent with that from prior experiments, provide molecular insight into the effect of enzyme surface charge on enzyme stability in ILs.

Abstract:Laccase is a delignifying enzyme that belongs to the oxidoreductase family, and it has long been investigated as a pretreatment agent in biofuel production. In this study, amino acid sequences of five bacterial laccases from Bifidobacterium breve , Klebsiella pneumonia , Pseudodesulfovibrio hydrargyri , Pseudomonas aeruginosa and Veillonella rodentium have been retrieved from UniProtKB for sequence alignment, phylogenetic analysis using MEGA 7.0 and 3 D structure prediction by homology modeling in SWISS-MODEL. Multiple sequence alignment between all the bacterial laccase sequences revealed a similar structural fold, although the overall protein sequence varied greatly with the substrate binding sites. Further molecular docking in AutoDock Vina and MD stimulation (MDS) in GROMACS for those modelled enzymes were performed considering both apo and ligand bound structures considering both apo and its ligand bound form. Investigation of molecular interaction utilizing docking of five bacterial laccases with three substrates (ABTS, DMP and Guaiacol) revealed that ABTS with K. pneumoniae laccase had the highest binding energy of −7.00 kcal/mol. In the current MDS investigation, bacterial laccases demonstrated greater binding and substrate energy in the ligand bound complex than in the apo form for ABTS, DMP and Guaiacol. In most cases of bacterial laccase, MDS revealed that DMP bound complex was more stable within an average RMSD value lower than 0.5 nm throughout 100 ns time scale. Thus, in silico studies undertaken in this work will be useful in determining the stable enzyme-substrate complex which further might improve the enzymatic catalysis of bacterial laccases for lignin breakdown and biofuel generation.
Recent environmental applications of and development prospects for immobilized laccase: a review

Abstract:Laccases have enormous potential as promising 'green' biocatalysts in environmental applications including wastewater treatment and polluted soil bioremediation. The catalytic oxidation reaction they perform uses only molecular oxygen without other cofactors, and the only product after the reaction is water. The immobilization of laccase offers several improvements such as protected activity and enhanced stability over free laccase. In addition, the reusability of immobilized laccase is adistinct advantage for future applications. Laccase-Catalyzed Reactions in Ionic Liquids for Green Sustainable Chemistry

Abstract:Biocatalysis is an important and enabling tool in organic synthesis, degradation of chemical compounds, and biosensors. Laccases are copper ions containing oxidase and catalyze the conversion of polyphenol or amine derivatives using molecular oxygen. Recently, significant interests have gained attention for the development of a biochemical degradation method of lignocellulosic materials from the standpoint of green sustainable chemistry. Since these compounds are highly oxidized compounds, it is difficult to convert them stereo- or chemo-selectively to high-value small molecules. Chemical oxidations generally require hazardous reagents and harsh reaction conditions; hence, the resulting produced compounds are generally low-value molecules or complicated mixtures. On the contrary, laccases allow hazardous-reagent-free highly selective oxidative chemical conversions. The reactivity of the enzymes is influenced by a set of reaction conditions, in particular, the solvent system. Ionic liquids (ILs) have now been acknowledged as useful reaction media for biotransformations. Laccase-mediated reactions in ILs have been reviewed and focused on four topics: the first is "laccase-catalyzed reactions in ILs". The second is "biosensor system using laccase-mediated reaction in ILs". The third is "improved stability of laccase by immobilization or IL type additives", and the last chapter is "improved activity of laccase in ILs by protein engineering". Our opinion toward future perspectives of laccase-mediated reactions has also been discussed in "conclusions and future perspectives".This article has not yet been cited by other publications.

Abstract:This study investigates the enhancement of enzymatic catalytic performance by immobilizing laccase on various nanostructured mesoporous silica materials (SBA-15, MCF, and MSU-F). The activity of immobilized laccase was evaluated under different hydrothermal, pH, and solvent conditions, with laccase@MSU-F showing a three-fold increase in stability. Laccase immobilized on these materials demonstrated stability in a pH range of 4.5 to 10.0, while free laccase was inactivated at pH higher than 7. Molecular dynamics simulations revealed that electrostatic interactions and protective confinement effects contribute to the improved stability of immobilized laccase. Overall, the findings suggest that nanomaterials can enhance the operational stability and recovery of enzymes. Communicated by Ramaswamy H. Sarma

Abstract:This study demonstrates a facile yet effective electrochemical method to investigate the conformational flexibility of the active sites of Trametes versicolor (Tv) laccase based on sensitive determination of copper ions (Cu(2+)) dissociated from the enzyme with the cysteine-modified Au electrodes. In the native state, the multicopper active sites are deeply buried in the polypeptide of Tv laccase and are thus not electrochemically detectable even at the cysteine-modified Au electrodes. Upon the unfolding of Tv laccase induced by guanidine hydrochloride (GdnHCl), copper ions dissociate from the peptide chain and, as a consequence, are electrochemically reduced and thus detected at the cysteine-modified Au electrodes. Such a property could be used to investigate the conformational flexibility of multicopper active sites of Tv laccase in a simple way. We find that both the conformation of the multicopper active sites in Tv laccase and the enzyme activity change with the presence of a low concentration of GdnHCl denaturant (midpoint, where 50% of the enzyme is unfolded, at 0.7 M). This concentration is lower than that required to induce the conformational changes of Tv laccase molecule as a whole (midpoint at 3.4 M), as investigated by the intrinsic fluorescence of Tv laccase. This observation suggests that the multicopper active sites are formed by relatively weak interactions and hence may be conformationally more flexible than the intact enzyme. The electrochemical method demonstrated in this study is technically simple yet effective and could be potentially useful for investigation on the thermodynamics and kinetics of the conformational changes of multicopper oxidases induced by different denaturants.
The Modification of Dissolution Kinetics and Solubility of Lysozyme Crystals by Ionic Liquids

Abstract:Knowing about the dissolution kinetics and solubility is necessary for controlling crystallization separation process of lysozyme. In this study, the impact of four ionic liquids (ILs), namely, 1-butyl-3-methylimidazolium tetrafluoroborate [C 4 mim]BF 4 , 1-butyl-3-methylimidazole chloride [C 4 mim]Cl, 1-butyl-3-methylimidazole bromide [C 4 mim]Br, and 1,3-dimethylimidazolium iodide [dmim]I, on the dissolution rate of lysozyme was determined in an aqueous solution at 20 °C, pH 5.01, under 101.3 kPa. The results revealed that the dissolution rate of lysozyme increased with increasing concentrations of [C 4 mim]BF 4 and [C 4 mim]Cl, while it remained stable with increasing concentrations of [C 4 mim]Br. In contrast, the dissolution rate gradually decreased with increasing concentrations of [dmim]I. This suggests that the interaction between lysozyme molecules is influenced by the ILs, leading to variations in the dissolution rates. Additionally, the effect of anions and cations on the equilibrium solubility was analyzed. The results indicated that the order of anionic and cationic effects on equilibrium solubility is as follows: BF 4− < Cl − < Br − = [C 4 mim] + < [dmim] + < I − . Furthermore, dissolution kinetic models were established, which could be used to predict the dissolution behavior of large molecules like lysozyme in ILs aqueous solution. These findings have significant implications for the design of crystallization process and optimization of parameters during lysozyme recovery.

Abstract:A new laccase gene from newly isolated Bacillus licheniformis TCCC 111219 was actively expressed in Escherichia coli. This recombinant laccase (rLAC) exhibited a high stability towards a wide pH range and high temperatures. 170% of the initial activity was detected at pH 10.0 after 10-d incubation, and 60% of the initial activity was even kept after 2-h incubation at 70 °C. It indicated that only single type of extreme environment, such as strong alkaline environment (300 K, pH 12) or high temperature (370 K, pH 7), did not show obvious impact on the structural stability of rLAC during molecular dynamics simulation process. But the four loop regions of rLAC where the active site is situated were seriously destroyed when strong alkaline and high temperature environment existed simultaneously (370 K, pH 12) because of the damage of hydrogen bonds and salt bridges. Moreover, this thermo- and alkaline-stable enzyme could efficiently decolorize the structurally differing azo, triphenylmethane, and anthraquinone dyes with appropriate mediator at pH 3.0, 7.0, and 9.0 at 60 °C. These rare characteristics suggested its high potential in industrial applications to decolorize textile dyeing effluent.

Abstract:Abstract BACKGROUND Laccases belong to the family of blue multicopper oxidases, these enzymes are efficient in many applications, including in biofuel cells to produce electricity through chemical reactions. Several laccase immobilization methods have been elucidated, such as covalent link and entrapment. RESULTS The present work describes how, for the first time, laccase was successfully immobilized by entrapment into polymer inclusion ionic liquid membranes (PILMs) based on ionic liquids and polyvinylchloride. The immobilization rate was very high in all cases (99.2 ± 0.6%), so that the enzyme entrapment strategy described can be considered as successful and one of the best methods for laccases immobilization that have been reported to date. The assayed membranes were evaluated as performing carrier for laccase by means of the ABTS oxidation. Four screened ionic liquids were used for the first time for this purpose: [OMIM][NTf 2 ], [EMIM][NTf 2 ], [Epy][NTf 2 ] and [Chol][NTf 2 ]. This work demonstrated the efficiency in terms of the activity rate and stability of the enzyme versus IL concentrations. The most suitable IL as regards activity [Chol][NTf 2 ]. However, the reuse assays showed that the most stable PILM was the one constituted by 50% [Epy][NTf 2 ]. For physical resistance and water insolubility, the [OMIM][NTf 2 ] based PILM was selected for microbial fuel cell (MFC) application. CONCLUSION This is the first report describing this original entrapment strategy for laccase immobilization into PILM. The application of this laccase‐membrane in MFC showed high stability during almost one week of use and good efficiency in producing bio‐energy and removing COD from industrial wastewater. © 2017 Society of Chemical Industry

Theoretical Insights into the Catalytic Oxidation of Phenols and Arylamines by Laccases via the Proton-Coupled Electron Transfer Mechanism

Abstract:Laccases play a vital role in the degradation of toxic phenolic and aromatic amine compounds, generating considerable attention in ecological pollution remediation. However, the distinct mechanism of the laccase-catalyzed oxidation of phenols and arylamines remains unclear. Here, we examined the catalytic oxidation mechanisms of phenols and arylamines by Trametes versicolor (TvL) and Melanocarpus albomyces (MaL) laccases using molecular docking, quantum mechanics (QM), and QM/molecular mechanics (QM/MM) calculations. We docked four phenolic substrates, including 1,2-benzenediol, 2-propenylphenol, 2-methoxyhydroquinone, and 2-aminophenol, to TvL and identified their favorable reaction conformations, in which Asp206 of TvL plays an important role in binding substrates to promote the catalytic reactions. Based on the docking conformations, the QM and QM/MM calculations revealed that the oxidation reactions take place via a proton-coupled electron transfer mechanism, with proton transfer (PT) from the hydroxyl groups of substrates to the side chain of Asp206 and synchronous electron hopping from the aromatic ring of substrates to the type one copper (T1Cu) of TvL. For the MaL and 2,6-dimethoxyphenol interacting system, the oxidation reactions occur through a concerted double-proton-coupled electron transfer mechanism with a water-mediated indirect PT from the hydroxyl group of substrates to the conserved Glu235 and electron hopping from the substrate to T1Cu at the same time. The corresponding energy barriers change from 0.7 to 18.4 kcal/mol, indicating the different degradation rates of the phenols and arylamines by laccases. These findings provide insights into the oxidation mechanism of phenols and arylamines by laccases and may extend the applications of laccases.
Increase in incidence of infectious syphilis in Auckland, New Zealand: results from an enhanced surveillance survey.

Abstract:We instigated an enhanced voluntary surveillance system to determine the epidemiology of infectious syphilis in Auckland because it is not a notifiable condition. The study took place over a 12-month period from July 2006 to July 2007. We identified 92 cases of infectious syphilis, of which 71 were male (77%). Forty-four (48%) cases were in men who have sex with men, who made up the biggest proportion. Forty-eight cases (52%) acquired their infection in New Zealand and only 48 of all identified cases were symptomatic (52%). Our results confirm that infectious syphilis incidence is currently under-reported.
Purification, characterization and three-dimensional structure prediction of multicopper oxidase Laccases from Trichoderma lixii FLU1 and Talaromyces pinophilus FLU12

Abstract:Broad-spectrum biocatalysts enzymes, Laccases, have been implicated in the complete degradation of harmful pollutants into less-toxic compounds. In this study, two extracellularly produced Laccases were purified to homogeneity from two different Ascomycetes spp. Trichoderma lixii FLU1 (TlFLU1) and Talaromyces pinophilus FLU12 (TpFLU12). The purified enzymes are monomeric units, with a molecular mass of 44 kDa and 68.7 kDa for TlFLU1 and TpFLU12, respectively, on SDS-PAGE and zymogram. It reveals distinct properties beyond classic protein absorption at 270-280 nm, with TlFLU1's peak at 270 nm aligning with this typical range of type II Cu site (white Laccase), while TpFLU12's unique 600 nm peak signifies a type I Cu2+ site (blue Laccase), highlighting the diverse spectral fingerprints within the Laccase family. The Km and kcat values revealed that ABTS is the most suitable substrate as compared to 2,6-dimethoxyphenol, caffeic acid and guaiacol for both Laccases. The bioinformatics analysis revealed critical His, Ile, and Arg residues for copper binding at active sites, deviating from the traditional two His and a Cys motif in some Laccases. The predicted biological functions of the Laccases include oxidation-reduction, lignin metabolism, cellular metal ion homeostasis, phenylpropanoid catabolism, aromatic compound metabolism, cellulose metabolism, and biological adhesion. Additionally, investigation of degradation of polycyclic aromatic hydrocarbons (PAHs) by purified Laccases show significant reductions in residual concentrations of fluoranthene and anthracene after a 96-h incubation period. TlFLU1 Laccase achieved 39.0% and 44.9% transformation of fluoranthene and anthracene, respectively, while TpFLU12 Laccase achieved 47.2% and 50.0% transformation, respectively. The enzyme structure-function relationship study provided insights into the catalytic mechanism of these Laccases for possible biotechnological and industrial applications.

Abstract:Laccases are attractive biocatalysts for industry due to their broad substrate spectrum, the use of oxygen as final electron acceptor, and water as the sole byproduct. Increasing efforts have been devoted to the engineering of laccases to improve their properties. The droplet-based microfluidic screening (DMFS) technology can accelerate the screening procedure and probe the large sequence space. In this study, a DMFS system including a heating step and picoinjection was used to sort large laccase libraries, yielding 12 variants with enhanced thermotolerance. All the obtained amino acid substitutions are distributed on the surface of the laccase. Interestingly, recombination of three identified substitutions of Asp to Asn on the surface resulted in the best variant M20, exhibiting 24.0-fold higher remaining activity at 58.8 °C and 1.9-3.4-fold higher remaining activity after incubation in organic solvents solution (20% (v/v) methanol and ethanol) and ionic liquid solution (20% (v/v) 1-ethyl-3-methylimidazolium ethyl sulfate) for 12 h. Furthermore, molecular dynamic simulations revealed that the recombination of the three beneficial substitutions, Asp98Asn, Asp474Asn, and Asp340Asn on the surface introduced more hydrogen bonds compared to the wild type, which made M20 more thermostable. This study highlighted the importance of the DMFS system for an efficient identification of beneficial long-distance amino acid substitutions.

Ion-combination specific effects driving the enzymatic activity of halophilic alcohol dehydrogenase 2 from Haloferax volcanii in aqueous ionic liquid solvent mixtures

Abstract:Biocatalysis in ionic liquids enables novel routes for bioprocessing. Enzymes derived from extremophiles promise greater stability and activity under ionic liquid (IL) influence. Here, we probe the enzyme alcohol dehydrogenase 2 from the halophilic archaeon Haloferax volcanii in thirteen different ion combinations for relative activity and analyse the results against molecular dynamics (MD) simulations of the same IL systems. We probe the ionic liquid property space based on ion polarizability and molecular electrostatic potential. Using the radial distribution functions, survival probabilities and spatial distribution functions of ions, we show that cooperative ion-ion interactions determine ion-protein interactions, and specifically, strong ion-ion interactions equate to higher enzymatic activity if neither of the ions interact strongly with the protein surface. We further demonstrate a tendency for cations interacting with the protein surface to be least detrimental to enzymatic activity if they show a low polarizability when combined with small hydrophilic anions. We also find that the IL ion influence is not mitigated by the surplus of negatively charged residues of the halophilic enzyme. This is shown by free energy landscape analysis in root mean square deviation and distance variation plots of active site gating residues (Trp43 and His273) demonstrating no protection of specific structural elements relevant to preserving enzymatic activity. On the other hand, we observe a general effect across all IL systems that a tight binding of water at acidic residues is preferentially interrupted at these residues through the increased presence of potassium ions. Overall, this study demonstrates a co-ion interaction dependent influence on allosteric surface residues controlling the active/inactive conformation of halophilic alcohol dehydrogenase 2 and the necessity to engineer ionic liquid systems for enzymes that rely on the integrity of functional surface residues regardless of their halophilicity or thermophilicity for use in bioprocessing.
