Abstract:Egg ovalbumin (OVA) as a prevalent dietary protein and has the potential to serve as a carrier for unstable bioactive compounds, however, understanding their interaction mechanism is the preliminary step. In this work, the interactions between cyanidin-3-O-glucoside (C3G) and OVA in both acidic and neutral pH environment were investigated by spectroscopic methods and molecular docking analysis. The results revealed that fluorescence quenching mechanism of OVA-C3G was predominantly static. The main acting forces were hydrogen bonds and van der Waals forces under varying pH conditions. However, the binding affinity of C3G to OVA was higher in neutral environment than that in acidic condition. The binding of C3G slightly increased the diameter of the complex, resulting in increase of α-helix, decrease of β-turn, random coil, and total main secondary structure. Moreover, the thermostability of C3G was significantly improved after OVA addition, suggesting its promising application in functional foods.
Formation of Tannic Acid-Binding Ovalbumin Amyloid Fibril Hydrogels: Enhanced Antibacterial and Antioxidant Properties

Abstract:The protein-induced amyloid fibrils (AFs) are limited in food industry due to their poor antioxidant and antibacterial activities. However, polyphenol shows the potential functional modifications of AFs, and extend their application in food processing. In present study, tannic acid-binding ovalbumin-induced AFs (TA-OAFs) hydrogels were fabricated via different concentrations of TA and ovalbumin amyloid fibrils, and the rheological properties, thermal stability, and morphology of the hydrogels were investigated. Then, the physiochemical, antioxidant, and antibacterial of TA-OAFs hydrogels were further analyzed. The obtained results indicated that the addition of TA could increase the average diameter of nanofibrils with increased β-sheet contents, and showed a TA concentration-dependent tendency. Moreover, TA-OAFs hydrogels also possessed better thermal stability, antioxidant activity, and antibacterial capacity for both Gram-negative and Gram-positive bacteria. The present study might contribute to a better understanding of antimicrobial materials designing of protein-induced amyloid fibrils in the food field with beneficial properties.

Abstract:Studying the interactions of bioactive phenolic acids [ferulic acid (FA), caffeic acid (CA), and chlorogenic acid (CGA)] with the carrier protein bovine hemoglobin (BHb) helps to understand their modes of binding and transport, thereby affecting their bioavailability. The protein-ligand complexes exhibited a binding constant (Kb) in the range of 104 M-1, indicating a reversible mode of association. BHb-CA complexation attenuated the inherent fluorescence of BHb (β2-Trp37 dominant fluorophore) through a static quenching mechanism. However, a combined (static and dynamic) quenching mechanism was observed for BHb-FA/CGA complex. In this study, the thermodynamic parameter, ΔG0, determined from fluorescence spectroscopy, was negative, suggesting the complexation of BHb with phenolic acids was spontaneous. The negative change in enthalpy [ΔH0 =– (8.539±1.331), –(10.620±1.028), and –(8.637±0.232) for BHb-FA, BHb-CA, BHb-CGA complexes, respectively] and positive entropy change [ΔS0 =+(63.862±1.218), +(67.108±1.681), and +(62.540±1.058) for BHb-FA, BHb-CA, BHb-CGA complexes, respectively] indicated that hydrogen bonding and hydrophobic forces stabilized the complexes. Computational studies also illustrated similar interactive forces in BHb-ligand complexes. Changes around the microenvironment of the Trp residues in the binding process were verified from various spectral measurements. Circular dichroism and Fourier infrared spectroscopy illustrated a decrease in the α-helical content of BHb during complexation. Fibrillar inhibition properties of the ligands were verified using ThT assay, ANS binding studies, congo red assay, turbidity assay, Soret absorption measurements, and CD spectral measurements. A structure-activity relationship, binding affinity, and interactive forces between the protein and phenolic acids could explain the order of anti-fibrillation properties.

Abstract:With the increasing demand for functional foods, the study on binding of active molecules and ovalbumin (OVA) via weak interaction has attracted widespread attention. In this work, the interaction mechanism of OVA and caffeic acid (CA) was revealed using fluorescence spectroscopy and dynamics simulation. The CA-induced fluorescence decrease of OVA was static quenching. Their binding complex had about 1 binding site and a 3.39 × 10<sup>5</sup> L·mol<sup>-1</sup> affinity ability. Based on thermodynamic calculations and molecular dynamics simulation, the complex structure of OVA and CA were stable using hydrophobic interactions as the main force, where CA preferred to interact with a stable binding pocket consisting of E256, E25, and V200 with N24 amino acid residues. In the binding process of CA and OVA, the conformation of OVA was altered with a slight reduction of α-helix and β-sheet. The reduced molecular volume and more compact structure of the protein indicated that CA is beneficial to the structural stability of OVA. The research provides some new insights into the interaction between dietary proteins and polyphenols, expanding the application prospects of OVA as a carrier.

Infidelity and behavioral couple therapy: optimism in the face of betrayal.

Abstract:Infidelity is a common issue with which distressed couples and their therapists grapple. However, there are no data on the efficacy of commonly used therapies to treat couples in which there has been an affair. In the present exploratory study, the authors examined the therapy outcomes of a sample of infidelity couples (n=19) who had participated in a randomized clinical trial of marital therapy (N=134). Results show that infidelity couples began treatment more distressed than noninfidelity couples; however, evidence suggests that couples who had an affair and who revealed this affair prior to or during therapy showed greater improvement in satisfaction than noninfidelity couples. Implications for therapy with infidelity couples are discussed.

Abstract:The use of bioactive flavonoids as drugs has long mesmerized the scientific world. Their small size and planar structure enables them to interact with limitless substrates especially biomolecules. Taxifolin is a flavonoid well known for its anti-oxidizing and metal chelating properties. Its interaction with a few biomolecules has been studied so far to exploit its pharmacological activities. Hemoglobin, an iron containing macromolecule acts as a major carrier protein and is also associated with the occurrence of many diseases. Our present study lays emphasis on the interaction of flavanonol taxifolin with bovine hemoglobin at physiological pH. This was achieved by monitoring the changes in the absorbance, fluorescence, anisotropic, lifetime and circular dichroic spectra. Benesi-Hildebrand plot determined a binding constant value of 20.0 × 103 M-1 at 25 °C. Stern-Volmer quenching studies reveal that the binding is associated with a static mode of quenching. The complexation is thermodynamically favored as indicated by the negative value of enthalpy and positive value of entropy changes seen from the van't Hoff plot. Theoretical DFT calculations were used to find out an optimized geometry and HOMO-LUMO energy gap for taxifolin. Molecular docking studies revealed the location of taxifolin inside the hemoglobin moiety.

Abstract:The solvation behavior of protein is an important factor in protein-based food products. In the present study, the xylitol (XY) - ovalbumin (OVN) interaction in an aqueous solution of different pH conditions is analyzed in two methods. In one method, the thermodynamic parameters Gibbs free energy, free volume, and internal pressure are calculated by using ultrasonic velocity, density, and viscosity in addition the refractive index is also measured. The second method is a theoretical method in which using the Laplace transform technique the diffused amount of protein have been calculated for OVN with and without XY in different pH environment. The addition of XY with OVN makes the system with more free energy and free volume as the internal pressure decreases. This trend shows that preferential interaction occurs between solvent-solute molecules. The diffusivity of OVN is reduced after the addition of XY representing the strength of protein-protein interaction. The effect of pH changes is well reflected in both experimental and theoretical results. The results confirm that acidic pH extremity offers more solvation of OVN compared to alkaline pH extremity.

Abstract:The interaction of atorvastatin with bovine serum albumin (BSA) was investigated using multi-spectroscopic methods and molecular docking technique for providing important insight into further elucidating the store and transport process of atorvastatin in the body and the mechanism of action and pharmacokinetics. The experimental results revealed that the fluorescence quenching mechanism of BSA induced atorvastatin was a combined dynamic and static quenching. The binding constant and number of binding site of atorvastatin with BSA under simulated physiological conditions (pH=7.4) were 1.41 × 10(5) M(-1) and about 1 at 310K, respectively. The values of the enthalpic change (ΔH(0)), entropic change (ΔS(0)) and Gibbs free energy (ΔG(0)) in the binding process of atorvastatin with BSA at 310K were negative, suggesting that the binding process of atorvastatin and BSA was spontaneous and the main interaction forces were van der Waals force and hydrogen bonding interaction. Moreover, atorvastatin was bound into the subdomain IIA (site I) of BSA, resulting in a slight change of the conformation of BSA.
Insufficient self-care is an independent risk factor for adverse clinical outcomes in Japanese patients with heart failure.

Abstract:Self-care is a cornerstone for the successful management of heart failure (HF). The purpose of this study was to examine the impacts of HF self-care on prognosis in Japanese patients with HF. A total of 283 HF outpatients (age 64 ± 14, 70% male, 52% HFrEF) were enrolled. We asked patients to answer about their adhevence to 5 self-care behaviors (medication, eating a low-sodium diet, regular exercise, daily weight check, and treatment seeking behavior). On the basis of the results, we classified patients into a good self-care group and a poor self-care group. The primary outcome was HF hospitalization and/or cardiac death. In total, 65% of patients were classified into the poor self-care group. During a median follow-up of 2 years, cardiac events occurred more frequently in the poor self-care group (22% versus 9.6%, P = 0.013). Poor self-care was an independent risk factor for cardiac events in Cox regression analysis adjusted for clinical parameters (hazard ratio = 2.86, P = 0.005). Poor self-care was also associated with an increased number of HF hospitalizations as well as an extended length of hospital stay for HF. Poor knowledge about HF was an independent determinant for poor self-care in multivariate logistic regression analysis (odds ratio = 0.92, P = 0.019). Insufficient self-care is an independent risk factor for cardiac events in Japanese patients with HF.
Binding and displacement study of gentamicin, 5-fluorouracil, oxytetracycline and rolitetracycline with (BSA: Drug2) complex using spectroscopic and calorimetric techniques: Biophysical approach

Abstract:Understanding of energetics of interactions between drug and protein is essential in pharmacokinetics and pharmacodynamics study. The binding affinity (K) helps in investigating how tightly or loosely drug is bound to protein. The binding, displacement, conformational change and stability study of drugs- gentamicin (GM), 5-fluorouracil (5FU), oxytetracycline (OTC) and rolitetracycline (RTC) with bovine serum albumin (BSA) has been carried out in presence of each other drug by fluorescence, UV-visible spectroscopy, molecular docking, circular dichroism techniques and thermal denaturation method. The site marker study and docking methods have confirmed that 5FU and GM are able to bind at site 1 and OTC and RTC at site II of BSA. The order of their binding affinities with BSA for the binary system were as GM <5FU < OTC < RTC with the order of 102 < 103 < 105 < 105-6 M-1. The displacement study has shown that higher affinity drug decreases the equilibrium constant of another drug already in bound state with BSA if both these drugs are having the same binding site. Therefore 5FU, GM (binding site 1) drugs were not able to displace OTC and RTC (binding site 2) and vice-versa as they are binding at two different sites. The binding constant values were found to be decreasing with increasing temperature for all the systems involved which suggests static or mixed type of quenching, however can only confirmed with the help of TCSPC technique. The ΔG0 (binding energy) obtained from docking method were in accordance with the ITC method. From molecular docking we have determined the amino acid residues involved in binding process for binary and ternary systems by considering first rank minimum binding energy confirmation. From CD it has been observed that RTC causes most conformational change in secondary and tertiary structure of BSA due to the presence of pyrrole ring. OTC-RTC with higher affinity showed highest melting temperature Tm values while low affinity drugs in (5FU-GM) combination showed lowest Tm value. 5FU showed large endothermic denaturation enthalpy ΔHd0 due to the presence of highly electronegative fluorine atom in the pyridine analogue.

Abstract:Understanding the physicochemical properties of pharmacy and the concept of pharmacophore is essential to devise guidelines for the synthesis of target-oriented drugs. In the present study, thermal denaturation, circular dichroism, intrinsic fluorescence, and docking were employed for studying the effects of the non-steroidal anti-inflammatory drug, diclofenac sodium, on the stability and structure of bovine pancreatic ribonuclease (RNase A) taken as a model of an enzyme. Since drug-protein interaction is an important pharmacokinetic parameter of a drug, it was deemed significant to study the effects of diclofenac on protein structure and stability. When the thermodynamic parameters were assessed, it was shown that RNase-A was destabilized in the presence of diclofenac sodium in terms of Tm and ΔGD°, while the addition of osmolytes stabilized the protein. In the presence of diclofenac sodium and osmolyte, there was a change in a tertiary structure of the protein but not in its secondary structure. Spectral results revealed a decrease in fluorescence intensity confirming fluorescence quenching of RNase-A by diclofenac sodium and osmolytes. Molecular docking analysis suggested that hydrogen bonds and Van der Waals are involved in the binding of RNase-A to diclofenac sodium. The RNAse-A activity is decreased in the presence of diclofenac sodium while osmolyte can restore the RNase-A activity and the indices kcat increase and Km values decrease. This study presents new combination of drug therapy comprising diclofenac sodium and osmolytes to reduce the risk of adverse effects of diclofenac sodium.
In-vitro Interactions of Esculin and Esculetin with Bovine Hemoglobin Alter its Structure and Inhibit Aggregation: Insights from Spectroscopic and Computational Studies

Abstract:Hemoglobin serves a multitude of biological functions whose activity and stability are influenced by structural alterations. Fibrillation of this protein is one such complication that can hinder its physiological levels resulting in numerous health disorders. This study focuses on providing a comprehensive elucidation on the binding interaction of bioactive esculin and esculetin with bovine hemoglobin (BHb) and ultimately determining the anti-fibrillation properties of these compounds. UV/vis spectroscopy and fluorescence measurements verified the presence of a static form of quenching mechanism for BHb-Esculin complex and a combined quenching mechanism for BHb-Esculetin complex. The binding affinity (Kb) was determined to be higher for BHb-Esculetin complex than BHb-Esculin complex. This could be due to a lower binding distance observed between β2-Trp37 residue and esculetin, as observed through Förster's theory, molecular docking, and molecular dynamic (MD) simulation. The calculated thermodynamic parameters indicated the presence of hydrophobic forces and hydrogen bonding stabilizing the complexes. Synchronous, 3D fluorescence, REES, and anisotropy measurements revealed microenvironmental alterations around the Trp residue induced by esculin/esculetin binding. Secondary structural changes of BHb in presence of esculin/esculetin were observed through UV/vis absorption, CD, and FTIR studies. BHb was subjected to fibrillation in presence of fructose, and ThT fluorescence, fluorescence microscopy, Soret absorption, ANS measurements, and Congo red assay provided evidence of the anti-fibrillation effects of esculin and esculetin.

Abstract:The binding interaction between quinapril (QNPL) and bovine serum albumin (BSA) in vitro has been investigated using UV absorption spectroscopy, steady-state fluorescence spectroscopic, synchronous fluorescence spectroscopy, 3D fluorescence spectroscopy, Fourier transform infrared spectroscopy, circular dichroism, and molecular docking methods for obtaining the binding information of QNPL with BSA. The experimental results confirm that the quenching mechanism of the intrinsic fluorescence of BSA induced by QNPL is static quenching based on the decrease in the quenching constants of BSA in the presence of QNPL with the increase in temperature and the quenching rates of BSA larger than 10(10)Lmol(-1)s(-1), indicating forming QNPL-BSA complex through the intermolecular binding interaction. The binding constant for the QNPL-BSA complex is in the order of 10(5)M(-1), indicating there is stronger binding interaction of QNPL with BSA. The analysis of thermodynamic parameters together with molecular docking study reveal that the main binding forces in the binding process of QNPL with BSA are van der Waal's forces and hydrogen bonding interaction. And, the binding interaction of BSA with QNPL is an enthalpy-driven process. Based on Forster resonance energy transfer, the binding distance between QNPL and BSA is calculated to be 2.76nm. The results of the competitive binding experiments and molecular docking confirm that QNPL binds to sub-domain IIA (site I) of BSA. It is confirmed there is a slight change in the conformation of BSA after binding QNPL, but BSA still retains its secondary structure -helicity.

Abstract:Protein-ligand interactions are crucial for developing and identifying novel therapeutic targets. In this study, we investigate the interaction of the alkali induced state of human serum albumin (pH 11.2) with three biologically important antioxidants known as hydroxycinnamic acid derivatives (HCDs): ferulic acid (FA), sinapic acid (SA) and trans-o-coumaric acid and compare the outcomes with the results obtained at physiological pH = 7.4. This study aims to explore the interaction of altered protein conformation with small molecules. Spectroscopic characterizations show conformation of HSA and the ionic properties of HCDs are pH-dependent. Fluorescence, FRET analysis and lifetime measurements reveal the binding of HCDs with HSA is different at both pH 7.4 and 11.2. Despite the moderate binding of HCDs to HSA, circular dichroism and thermal denaturation studies report no conformational change in HSA in the presence of HCDs. Isothermal titration calorimetry is employed to assess their binding based on structure and energetics using thermodynamic parameters. Standard molar enthalpy change (ΔH_m^O) and standard molar entropy change (ΔS_m^O) values from pH 7.4 to 11.2 vary accordingly with the contributions from exothermicity and hydrophobicity of functional and aromatic groups of HCDs. Ferulic acid (FA) and sinapic acid (SA) binding to HSA is entropically driven whereas trans-o-coumaric acid (CA) acid binding is enthalpically favourable. Our ITC studies also reveal that the involvement of -OH functional groups present in CA with HSA is greater than that of FA and SA at pH 11.2. Overall, this experimental study shows the comparable binding strength of HCDs to both the alkali-induced state of HSA and native HSA (pH 7.4). However, the mechanism of their binding is different.

Abstract:155 patients with 210 ovarian tumours were examined pre-operatively by CT from December 1984 to February 1988. 27 patients had dermoid cysts, unilateral in 25 cases and bilateral in 2 cases. These cystic teratomas were benign in 26 patients. In one case, malignant transformation occurred. CT detected 29/29 tumours (100%). Presence of fat (26/29, 90%), calcification (20/29, 69%), Rokitansky protuberance (25/29, 86%), tufts of hair (20/29, 69%), or fat fluid level (4/29, 14%), allowed to make a definite diagnosis in 28/29 cases (96%). These findings and especially demonstration of fat, appeared to be specific for dermoid cysts in our series. The diagnosis of torsion of the cyst associated with torsion of the adnexa was made in 2 patients. No abdomino-pelvic extension occurred in 26/26 patients presenting with a benign dermoid cyst and CT correctly diagnosed the lymph node and hepatic spread in the single case of cancer.

Abstract:Lactoferrin, a member of the transferrin family, is one of the promoter proteins for calcium oxalate-type kidney stone formation. It exhibits a remarkable ability to interact with metals and oxalate ions. The prevalence of calcium oxalate in kidney stones was confirmed by the Fourier transform infrared spectra. The quantum chemical properties of calcium oxalate and dolichin A calculated by density functional theory and time-dependent density functional theory indicate their potential for hydrogen bonding and nonbonding interactions with the receptor proteins. From molecular docking analysis, the binding free energy of dolichin A was −7.78 kcal/mol, which was the best of twenty-four phytochemicals from Macrotyloma uniflorum , and that of calcium oxalate was −3.86 kcal/mol to lactoferrin. Furthermore, dolichin A having favorable physicochemical and pharmacokinetic properties offers post molecular dynamics molecular mechanics generalized Born surface area free energy of −17.61 ± 4.03 kcal/mol, indicating the strong binding interactions, and, therefore, it acts as a potential inhibitor of the lactoferrin.

Abstract:This research addresses the crucial necessity for a deeper understanding of the binding interactions between surfactants and proteins, with a specific focus on ovalbumin . Considering ovalbumin's role in diverse biochemical processes, it remains a subject of significant interest for drug discovery and design. To fill existing knowledge gaps, we investigated the binding interaction between dicloxacillin and cetyltrimethylammonium bromide (CTAB) on ovalbumin, employing a comprehensive approach that combines computational modeling with experimental validations. Using the ezPocket tool, the computational phase predicted ten relevant binding sites on ovalbumin's surface. The isobologram combination index (CI) heatmap strongly suggested a complex interplay of antagonistic and synergistic effects. Besides, a conformational drug-drug interaction network was proposed to explore the stability of the surfactant mixture within specific binding sites of ovalbumin, revealing a dynamic landscape of suggested antagonist effects. Experimental validations through UV–vis, Fluorescence, and circular dichroism (CD) spectroscopy further corroborated the computational findings, confirming the formation of stable complexes. Finally, this study not only advances our comprehension of ovalbumin's interactions with surfactants but also offers a multidimensional perspective and an advanced methodological framework for efficient therapeutic strategies, opening new avenues for future applications in drug development and applied biochemistry.

Interaction Mechanism between OVA and Flavonoids with Different Hydroxyl Groups on B-Ring and Effect on Antioxidant Activity

Abstract:Ovalbumin (OVA) is a common carrier with high efficiency to deliver flavonoids. The aim of this study was to investigate the interaction mechanism of OVA and four flavonoids (quercetin (Que), myricetin (Myri), isorhamnetin (Ish), and kaempferol (Kaem)) with similar structures by fluorescence spectra, SDS-PAGE, FT-IR, and molecular docking analysis, and the effect on the antioxidant abilities of flavonoids was also evaluated. Results indicated that the antioxidant activity of flavonoids was positively correlated to the number of phenolic hydroxyl groups of on the B-ring, and weakened when the C-3' position was replaced by a methoxy group. The addition of OVA enhanced the antioxidant activity of Que/Kaem, while it masked the antioxidant activity of Myri. The formation of Que/Myri/Ish/Kaem-OVA complexes was a spontaneous exothermic process driven mainly by hydrogen bond and van der Waals force, which could result in the change in OVA conformation and induce the transformation of α-helix to β-sheet. Among these, Kaem exhibited the strongest binding ability with OVA, and showed the greatest impact on the secondary and conformational structure of OVA, followed by Que. The hydroxylation of C-3' and methoxylation of C-5' weaken the interaction of Kaem with OVA. Molecular docking analysis suggested that Que, Myri, Ish, and Kaem formed six, three, five, and four hydrogen bonds with OVA, and the number of hydrogen bonds was not positively correlated with their binding constants. Our findings can provide a theoretical basis for the application of OVA on improving the antioxidant activity of flavonoids, and may help to explain the delivery efficiency of OVA on different bioactive constituents.
Epilactose production by 2 cellobiose 2-epimerases in natural milk.

Abstract:It was reported recently that cellobiose 2-epimerases (CE) from various aerobic microorganisms convert lactose to epilactose in defined buffer systems. In this study, we showed that CE from 2 mesophilic microorganisms, Flavobacterium johnsoniae and Pedobacter heparinus, were capable of converting lactose to prebiotic epilactose not only in buffer but also in a complex milk system. First, the 2 enzymes were separately cloned, recombinantly expressed in Escherichia coli, and purified by column chromatography. The production of F. johnsoniae CE was carried out in a stirred-tank reactor, indicating that future upscaling is possible. The bioconversions of milk lactose were carried out at an industrially relevant low temperature of 8°C to avoid undesired microbial contaminations or chemical side reactions. Both enzymes were reasonably active at this low temperature, because of their origin from mesophilic organisms. The enzymes showed different operational stabilities over a 24-h time-course. A conversion yield of about 30 to 33% epilactose was achieved with both enzymes. No side products were detected other than epilactose. Therefore, CE may introduce an added value for particular dairy products by in situ production of the prebiotic sugar epilactose.
