Abstract:Monofloral and multifloral honey produced in different regions may have different bioactive compounds and antioxidant capacities, resulting in changes in the antimicrobial activity of honey. However, many of these compounds degrade due to the extreme digestion conditions, which may inhibit the antimicrobial activity. Given this context, this study aimed to describe the bioactive compounds of honey produced in Brazil and verify if honey samples from different botanical and geographical origins differ in bioactive compounds, and if honey maintains its antimicrobial activity after digestion simulation. Multivariate analysis was used to identify characteristics that differentiated the honey samples according to the botanical and geographical origin criteria. The amount of the bioactive compounds varied significantly: the total phenolic compound content varied from 20.49 to 101.44 mg GAE per 100 g, flavonoids varied from 1.41 to 13.52 mg QE per 100 g, phenolic acids varied from 13.61 to 56.41 mg CAE per 100 g, and carotenoids varied from 0.66 to 4.27 mg β-carotene per g. Multifloral honey (H22) produced in the dry season of northeastern Brazil presented the highest bioactive compound concentration except for the carotenoid content. HPLC-MS analysis showed the presence of six hydroxybenzoic acids, four hydroxycinnamic acids, eight flavonols, three flavanones, two flavones and two isoflavonoids; Pterodon pubescens monofloral honey (H14) from midwestern Brazil stood out in terms of the carotenoid content. All analyzed honey samples exhibited antimicrobial activity against Staphylococcus aureus and Escherichia coli bacteria before digestive process simulation, and bacteria were inhibited during in vitro digestion; this activity decreased during the simulation of the oral phase, remained in the gastric phase, and disappeared in the intestinal phase.

Abstract:During in vitro myogenesis, a portion of myoblasts undergo apoptosis, whereas others continue with their differentiation program and form myotubes that are resistant to cell death. Previous work has shown that the expression of the Cdk inhibitor p21 correlates with enhanced resistance to apoptosis and that forced expression of p21 will confer this phenotype on differentiating myocytes. Here we examine the role of the retinoblastoma gene (Rb) in myocyte survival. Compared with wild-type myocytes, CC42 (Rb-/-) myocytes undergo higher frequencies of apoptosis during mitogen deprivation-induced myogenesis. Despite these features, Rb-/- myocytes display normal up-regulation of p21 and down-regulation of Cdk activities upon differentiation. Adenoviral constructs expressing the Cdk inhibitors p21 or p16 inhibit apoptosis in wild-type but not Rb-/- myocyte cultures. On the other hand, a Rb-expressing adenoviral construct inhibited apoptosis in both cell types. These data demonstrate that Rb functions downstream from the Cdk inhibitors to coordinate cell cycle withdrawal with programmed cell death during myocyte differentiation.

Abstract:In the present study, chemical characterisation, thermal analysis and antibacterial activity of honeys from Melipona spp. with occurrence in Caatinga biome of Brazil. The honeys presented pH from 4.07 to 4.14, density of 1.41 g/cm 3 and °Brix value of 79.90. The thermogravimetry (TG) analysis presented six-seven events and differential thermal analysis (DTA) presented three-four endothermic peaks. HPLC fingerprint revealed a predominant presence of gallic acid and vanillin. Antioxidant activity evaluated using in vitro 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) radical scavenging with IC 50 values in the range of 14.5404 to 15.2454 mg/mL. The honeys also showed antimicrobial activities against Staphylococcus aureus and Pseudomonas aeruginosa using a modified agar diffusion and microdilution method. The results of the present study demonstrate that the honey from stingless bees by Caatinga biome indicate polyphenol compounds, antioxidant activity and in vitro antimicrobial potential. The analytical methos permitted of fingerprint of samples.

Abstract:The biodiversity of Brazil provides an excellent climate and favorable pollination conditions for Apis mellifera L., especially in the Eastern Amazon region, which boasts vast floral wealth, including an abundance of açaí (Euterpe oleracea) flowers and fruits. In the present study, seven types of honey were evaluated: three containing floral nectar from açaí (Açaí honey) collected in the Eastern Amazon region (Açaí honey from Breu Branco (AH1 and AH2) and Açaí honey from Santa Maria (AH3), both from the state of Pará, Brazil) and four honeys from different regions of Brazil (Aroeira honey from Minas Gerais, Cipó-Uva honey from Distrito Federal, Mangue honey from Pará, and Timbó honey from Rio Grande do Sul). The characteristics of these honeys were evaluated by examining their physicochemical properties, melissopalynological aspects, color, antioxidant potential, and their constituent compounds, which were confirmed through GC-MS analysis. Açaí floral nectar honeys presented physicochemical results similar to those of other honeys, aligning with Brazilian legislation norms, but differed in their high values of free acidity, apparent sugars, and lower reducing sugars, which are directly related to their botanical origin. These differences correlate with unique flavor and distinct aroma characteristics. Melissopalynological analysis confirmed the botanical origin of the honeys containing açaí floral nectar, which had a color range from amber to dark amber. The three açaí honeys demonstrated high antioxidant capacity and superior flavonoid and polyphenol content compared to other honeys, particularly the açaí honey from Breu Branco (AH1), which had four times the content to combat free radicals compared to the honey with the highest potential (Aroeira honey). GC-MS analysis confirmed the presence of antioxidant properties as well as potential anti-inflammatory, antibacterial, antimicrobial, and antitumor capabilities in açaí honeys, which have not yet been fully studied.

Abstract:Stingless bees belong to the Meliponini tribe and are widely distributed in the tropics and subtropics, where they perform important ecological services. Among the best distributed groups of stingless bees is the genus Scaptotrigona, which includes 22 species distributed throughout the neotropical region, including the area from Mexico to Argentina. Bees of this genus are responsible for the production of products such as honey, propolis, geopropolis and fermented pollen ("saburá"). This review aimed to provide an overview of the chemical composition and biological activities associated with derived products from stingless bees of the genus Scaptotrigona. The bibliographic review was carried out through searches in the Scopus, Web of Science, ScienceDirect and PubMed databases, including publications from 2003 to January 2023. The study of the chemodiversity of products derived from Scaptotrigona demonstrated the mainly presence of flavonoids, phenolic acids, terpenoids and alkaloids. It was also demonstrated that products derived from bees of the genus Scaptotrigona exhibit a wide range of biological effects, such as antibacterial, antioxidant, anti‐inflammatory and antifungal activities, among other bioactivities. This review provides an overview of phytochemical and pharmacological investigations of the genus Scaptotrigona. However, it is essential to clarify the toxicity and food safety of these products.

Abstract:Abstract The antioxidant activity of Tetragonisca angustula honey (TAH) and its ethanolic extract (TAEE) were investigated. The total levels of phenolic (TPC) and flavonoids (TFC) were also evaluated. The results for TPC were 19.91 ± 0.38 and 29.37 ± 1.82 mg GAE g-1 and for TFC 0.20 ± 0.02 and 0.14 ± 0.01 mg QE g-1 of TAH and TAEE, respectively. Antioxidant activities were 73.29 ± 0.49% and 93.36 ± 0.27% in the DPPH● assay and 71.73 ± 4.07% and 97.86 ± 0.35% in ABTS●+ for TAH and TAEE, respectively. The total reducing activity was determined by the method of reducing power (PR) and iron ion (Fe III) and the results varied in PR from 151.7 ± 25.7 and 230.7 ± 25.2 mg GAE L-1, for TAH and TAEE respectively and for (Fe III) in EC50 0.284 in TAEE and 0.687 in TAH. Chemical analysis by HPLC-DAD of the ethanolic extract (TAEE) revealed the presence of ferulic acid as majority phenolic component in the extract. The 1H NMR analysis confirmed this structure and showed the also presence of glucose, citric acid, succinic acid, proline and hydrocarbon derivatives. In addition, the botanical origin was also investigated and showed a multifloral characteristic, having found 19 pollen types with a botanical predominance of the Anacardiaceae family, with Tapirira pollen occurring as predominant (42.6%) and Schinus as secondary (25.7%). The results showed that T. angustula honey is an interesting source of antioxidant phenolic compounds due to its floral origin and can act as a protector of human health when consumed.

Abstract:It is generally accepted that taxa exhibit genetic variation in phenotypic plasticity, but many questions remain unan- swered about how divergent plastic responses evolve under dissimilar ecological conditions. Hormones are signaling molecules that act as proximate mediators of phenotype expression by regulating a variety of cellular, physiological, and behavioral re- sponses. Hormones not only change cellular and physiological states but also influence gene expression directly or indirectly, thereby linking environmental conditions to phenotypic development. Studying how hormonal pathways respond to environ- mental variation and how those responses differ between individuals, populations, and species can expand our understanding of the evolution of phenotypic plasticity. Here, we explore the ways that the study of hormone signaling is providing new insights into the underlying proximate bases for individual, population or species variation in plasticity. Using several studies as exem- plars, we examine how a 'norm of reaction' approach can be used in investigations of hormone-mediated plasticity to inform the following: 1) how environmental cues affect the component hormones, receptors and enzymes that comprise any endocrine sig- naling pathway, 2) how genetic and epigenetic variation in endocrine-associated genes can generate variation in plasticity among these diverse components, and 3) how phenotypes mediated by the same hormone can be coupled and decoupled via independent plastic responses of signaling components across target tissues. Future studies that apply approaches such as reaction norms and network modeling to questions concerning how hormones link environmental stimuli to ecologically-relevant phenotypic re- sponses should help unravel how phenotypic plasticity evolves (Current Zoology 59 (4): 506-525, 2013). Abstract:When braking on split-μ road, yaw moment and steering force of vehicle will be produced because the braking force is different for two side wheels. In order to deal with this problem, the modified independent control mode is widely applied, however, there also exits some defects. Because the gap of the adhensive coefficient of both sides is uncertain while braking, if the fixed modified coefficient is applied, it is difficult to keep vehicle direction stability and shorten the braking distance simultaneously under different gap former mentioned. This paper proposes a method named split-μ road identification. Compare the difference of slip integral of front wheels to threshold values during a certain pressure state in order to identify whether the vehicle brakes on split-μ road. According to the integral of slip from both side wheels and vehicle deceleration, the control mode dynamically changes the modified coefficients of pressure. Simulation results show that the algorithm can identify whether the vehicle is braking on split-μ road timely and also can identify the friction difference between the two sides accurately. The pressure regulation mode can regulate brake pressure appropriately and keep vehicle directional stability and shorten braking distance simultaneously.

Abstract:Recently, the honey and propolis of stingless bees have been attracting growing attention because of their health-promoting properties. However, studies on these products of African Meliponini are still very scarce. In this preliminary study, we analyzed the chemical composition of honey, two cerumen, and two resin deposits (propolis) samples of Meliponula ferruginea from Tanzania. The honey of M. ferruginea was profiled by NMR and indicated different long-term stability from Apis mellifera European (Bulgarian) honey. It differed significantly in sugar and organic acids content and had a very high amount of the disaccharide trehalulose, known for its bioactivities. We suggested trehalulose to be a potential marker for African stingless bee honey analogously to the recent proposal for Meliponini honey from Asia, South America, and Australia and demonstrated its easy discrimination by 13C NMR. Propolis and cerumen were studied by GC-MS (gas chromatography-mass spectometry). The samples contained mainly terpenoids (di-and triterpenes) but demonstrated qualitative and quantitative differences. This fact was an indication that possibly M. ferruginea has no strict preferences for resins used to construct and protect their nests. The antimicrobial and anti-quorum sensing properties of the two materials were also tested. These first results demonstrated that the honey, cerumen, and propolis of African stingless bees were rich in biologically active substances and deserved further research.

Abstract:Macrophages play an important role during the inflammatory process. These cells can adopt either the pro- or anti-inflammatory phenotypes. While stingless bee honeys have demonstrated evidence of anti-inflammatory potential, their capacity to induce a shift from a pro-inflammatory state to an inflammation-resolution state has not been thoroughly investigated. In this study, the anti-inflammatory activity of two stingless bees ( Scaptotrigona bicunctata —honey A and Melipona quadriasciata —honey G) honeys in J774 macrophages induced by LPS was evaluated. Both honeys exhibited non-cytotoxic effects and reduced nitrite and IL-4 levels. However, only honey G increased the levels of the anti-inflammatory cytokine IL-13, by 163.1 ± 14.8% ( p < 0.05) and was further investigated for its immunomodulatory effect. This honey reduced the expression of TLR4 by 59.3 ± 3.5% ( p < 0.001) and increased the mannose receptor levels by 67.3 ± 2.4% ( p < 0.001). Moreover, it increased the phagocytic activity by 25.0 ± 7.7% ( p < 0.01) and decreased the death of the macrophages by 32.1 ± 1.7% ( p < 0.001). Collectively, these findings highlight stingless bee honey from Melipona quadriasciata bee has an important immunomodulatory effect, as it reduces the markers of the pro-inflammatory state of J774 cells and increases the markers of resolution or anti-inflammatory responses.

Abstract:Stingless bee honey (SBH) is an astounding ‘miracle liquid’ with countless medicinal properties for various diseases such as gastroenteritis, cataracts, as well as for wound-healing. However, knowledge regarding it is still rather scarce. Henceforth, it is intriguing for us to contemplate on the less-studied stingless bee and its honey in particular. First and foremost, the antimicrobial ability of honey from eight different stingless bee species was tested to further proven its health benefit. Homotrigona fimbriata honey showed the highest antimicrobial activity with inhibition against five bacteria; Serratia marcescens, Escherichia coli, Bacillus subtilis, Alcaligenes faecalis and Staphylococcus aureus. The next aim of our study is to characterize their honey bacterial community via the use of 16S rRNA amplicon sequencing technology. A total of eight bacterial phyla, 71 families, 155 genera and 70 species were identified from our study and two of the stingless bee species honey were determined to have the highest bacterial diversity compared to other six stingless bee species, namely Heterotrigona erythrogastra and Tetrigona melanoleuca. Furthermost, Lactobacillus malefermentans was thought to be the native dominant bacteria of SBH due to its predominant presence throughout all studied species. The aforementioned SBH’s antimicrobial results and characterization study of its bacterial diversity are hoped to carve the pathway towards extending its probiotic ability into our everyday lives.

Abstract:Bee bread, a valuable bee product that has recently attracted significant public interest as a nutritional supplement. The aim of this study was to evaluate the presence of phenolic compounds in bee bread samples from the Aegean Region and assess their bioaccessibility using a simulated human digestion model. Various extraction techniques, such as maceration, ultrasound‐assisted extraction, and supercritical fluid extraction were employed to obtain extracts of bee bread. The antioxidant capabilities of these extracts were carried out using assays like DPPH•, ABTS•+, CUPRAC, and β‐carotene linoleic acid bleaching, and their effectiveness was quantified through IC50 values. The bioaccessibility of phenolic compounds was analysed by using LC‐HRMS in a simulated human digestive system using ethanol extracts obtained from bee bread samples of each season by ultrasound‐assisted extraction, which has the highest antioxidant activity. In the Aegean bee bread, a total of 25 phenolic compounds which were major phenolics including quercetin, ascorbic acid, isorhamnetin, kaempferol, and hyperoside were identified and quantified. Also, ascorbic acid was the one of the most bioaccessible compounds with the bioaccessibility index 35.38% for 2021, 16.79% for 2022. These findings underscore the substantial transformation of the phenolic profile of bee bread as it traverses the human digestive system.

Abstract:Honey exhibits antibacterial and antioxidant activities that are ascribed to its diverse secondary metabolites. In the Philippines, the antibacterial and antioxidant activities, as well as the bioactive metabolite contents of the honey, have not been thoroughly described. In this report, we investigated the in vitro antibacterial and antioxidant activities of honey from Apis mellifera and Tetragonula biroi, identified the compound responsible for the antibacterial activity, and compared the observed bioactivities and metabolite profiles to that of Manuka honey, which is recognized for its antibacterial and antioxidant properties. The secondary metabolite contents of honey were extracted using a nonionic polymeric resin followed by antibacterial and antioxidant assays, and then spectroscopic analyses of the phenolic and flavonoid contents. Results showed that honey extracts produced by T. biroi exhibits antibiotic activity against Staphylococcal pathogens as well as high antioxidant activity, which are correlated to its high flavonoid and phenolic content as compared to honey produced by A. mellifera. The bioassay-guided fractionation paired with Liquid Chromatography Mass Spectrometry (LCMS) and tandem MS analyses found the presence of the flavonoid isorhamnetin (3-methylquercetin) in T. biroi honey extract, which was demonstrated as one of the compounds with inhibitory activity against multidrug-resistant Staphylococcus aureus ATCC BAA-44. Our findings suggest that Philippine honey produced by T. biroi is a potential nutraceutical that possesses antibiotic and antioxidant activities.

Abstract:Knowledge about the fate of beebread bioactive compounds throughout the human gastrointestinal tract are scarce. The present study aimed at assessing the effects of gastrointestinal digestion followed by intestinal permeability in vitro on phenolic profile and anti-inflammatory activity of Baccharis beebread. Palynological analysis confirmed the beebread is predominantly composed by pollen grains from Baccharis species, which are endemic in south and southeast Brazil. Flavonols and phenylamides were found in beebread hydroalcoholic extract by HPLC-ESI-QTOF-MS analysis. Moreover, simulated digestion lead to compounds' breakage, releasing both aglycones from glycosylated flavonols and p-coumaric acid, but not caffeic acid from phenylamides. Only spermidines crossed the Caco-2 cell monolayer, possibly due to spermine oxidation. Free p-coumaric acid was released after digestion, and epithelial transport. Concomitantly, NF-κΒ activation and TNF-α level was decreased by beebread even after Caco-2 transport, which indicates spermidines conjugated with p-coumaric acid may be bioavailable compounds with anti-inflammatory activity.

Abstract:To evaluate the antimicrobial activity of Brazilian honeys against oral microorganisms. Design: Organic honeys (OH-1 to OH-8) were diluted (%-w/v) and sterilized by filtration. Antimicrobial activity was defined by determining MIC and CBM against oral Streptococcus . The component responsible for the antimicrobial action was defined by a catalase assay. Antibiofilm activity was evaluated against the monospecies biofilm of Streptococcus mutans (ATCC 700610).

Abstract:Natural products are historically regarded as the main sources of bioactive compounds to fight the most diverse diseases; among them, propolis deserves to be highlighted due to several biological activities, such as antioxidant, anti-inflammatory, healing, antibacterial and antileishmanial activity. In this study, the antioxidant and antipromastigote activities and the cytotoxicity of propolis of the Scaptotrigona aff. postica stingless bee were evaluated. Propolis extract was analyzed by infrared spectroscopy, thermogravimetry and spectrophotometry for total polyphenols and flavonoid contents. Antioxidant activity was assessed spectrophotometrically by the DPPH, ABTS and FRAP methods, while antipromastigote activity and cytotoxicity were assessed by the MTT assay. The infrared spectra showed the characteristic bands of both phenols and alcohols. The thermogravimetry study revealed two events and thermal stability around 370 °C, while differential scanning calorimetry showed two endothermic events and an exothermic event. Total polyphenol and flavonoid contents were 21.29 ± 0.003 mg/g and 8.17 ± 0.00 mg/g, respectively, and the antioxidant activities identified by the DPPH, ABTS and FRAP methods were 910.09 ± 0.0264 μM Trolox (IC50 18.9 ± 0.01 μg/mL), 859.97 ± 0.0854 μM Trolox (IC50 692 ± 0.01 μg/mL) and 1613.67 ± 0.2610 μM Trolox (IC50 80 ± 0.1 μg/mL), respectively. The satisfactory antioxidant activity of propolis can be related to the high content of phenolic compounds, which adds value to this product and can contribute to the development of meliponiculture in the Brazilian state of Pará. The propolis extract caused a toxic effect on promastigote forms of Leishmania amazonensis at all concentrations tested, with an IC50 value of 1.50 μg/mL and a statistically significant difference compared to the negative control (p < 0.001). These results show that the propolis extract from S. postica bees may be a promising alternative for treatment against promastigote forms of L. amazonensis.
