Abstract:Triclosan (TCS), an antimicrobial agent extensively incorporated into pharmaceuticals and personal care products, poses significant environmental risks because of its persistence and ecotoxicity. So far, a few microorganisms were suggested to degrade TCS, but the microbial degradation mechanism remains elusive. Here, a two-component angular dioxygenase (TcsAaAb) responsible for the initial TCS degradation was characterized in Sphingomonas sp. strain YL-JM2C. Whole-cell biotransformation and crude enzyme assays demonstrated that TcsAaAb catalyzed the conversion of TCS to 4-chlorocatechol and 3,5-dichlorocatechol rather than the commonly suggested product 2,4-dichlorophenol. Then two intermediates were catabolized by tcsCDEF cluster via an ortho -cleavage pathway. Critical residues (N262, F279, and F391) for substrate binding were identified via molecular docking and mutagenesis. Further, TcsAaAb showed activity toward methyl triclosan and nitrofen, suggesting its versatile potential for bioremediation. In addition, TCS-degrading genes were also present in diverse bacterial genomes in wastewater, ocean and soil, and a relatively high gene abundance was observed in marine metagenomes, revealing the transformation fate of TCS in environments and the microbial potential in pollutant removal. These findings extend the understanding of the microbe-mediated TCS degradation and contribute to the mining of TCS-degrading strains and enzymes, as well as their application in the bioremediation of contaminated environments.

Abstract:Dehalogenation is one of the most important reactions for eliminating trace organic pollutants in natural and engineering systems. This study investigated the dehalogenation of a model organohalogen compound, triclosan (TCS), by aqueous biochars (a-BCs) (<450 nm). We found that TCS can be anaerobically degraded by reduced a-BCs with a pseudo first-order degradation rate constant of 0.0011–0.011 h–1. The 288 h degradation fraction of TCS correlated significantly with the amount of a-BC-bound electrons (0.055 ± 0.00024 to 0.11 ± 0.0016 mol e–/mol C) available for donation after 24 h of pre-reduction by Shewanella putrefaciens CN32. Within the reduction period, the recovery of chlorine based on residual TCS and generated Cl– ranged from 73.6 to 85.2%, implying that a major fraction of TCS was fully dechlorinated, together with mass spectroscopic analysis of possible degradation byproducts. Least-squares numerical fitting, accounting for the reactions of hydroquinones/semiquinones in a-BCs with TCS and byproducts, can simulate the reaction kinetics well (R2 > 0.76) and suggest the first-step dechlorination as the rate-limiting step among the possible pathways. These results showcased that the reduced a-BCs can reductively degrade organohalogens with potential applications for wastewater treatment and groundwater remediation. Abstract:Triclosan (TCS) is a broad-spectrum antimicrobial agent that is broadly used in personal care products. It has been shown to cause the contamination of a variety of aquatic environments. Since algae has been the primary producers of aquatic ecosystems, understanding the toxicological mechanisms and the metabolic fate of TCS is vital for assessing its risk in an aquatic environment. In our study, 0.5–4 mg L−1 TCS treatments for 72 h in a culture of Chlamydomonas reinhardtii (C. reinhardtii) showed progressive inhibition of cell growth and reduced the chlorophyll content. The EC50 value of C. reinhardtii after 72 h was 1.637 mg L−1, which showed its higher level of resistance to TCS in comparison with other algal species. The exposure to TCS led to oxidative injuries of algae in relation to the increment of malonaldehyde content, cell membrane permeability, and H2O2 levels. Furthermore, the oxidative stress from TCS stimulated a series of antioxidant enzyme activities and their gene expressions. Simultaneously, the accumulated TCS in C. reinhardtii arouses the detoxification/degradation-related enzymes and related gene transcriptions. In the medium, approximately 82% of TCS was removed by C. reinhardtii. Importantly, eight TCS metabolites were identified by ultra-performance liquid chromatography-high-resolution mass spectrometry and their relative abundances were measured in a time-course experiment. Six of these metabolites are reported here for the first time. The metabolic pathways of triclosan via C. reinhardtii including reductive dechlorination, hydroxylation, sulfhydrylation, and binding with thiol/cysteine/GSH/glycosyl were manifested to broaden our understanding of the environmental fate of TCS.Graphical

Preliminary evaluation of the operation variables and degradation pathway in the alkali-catalyzed hydrothermal oxidation of triclosan

Abstract:Due to the lack of green treatment technologies for chlorinated organic contaminant detoxification, in this study, the effect of non-hazardous alkali-catalyzed hydrothermal oxidation (HTO) on triclosan (TCS) was investigated. The optimal parameters determined by single-factor experiments were: a H2O2/TCS (H/T) ratio of 2.5 mL/mg, a temperature of 180 °C, a residence time of 120 min. Using these parameters, the final dechlorination efficiency (DE) of 99.69% was obtained. Response surface methodology (RSM) was employed to further determine the optimal conditions under multi-factor interactions. The optimal parameters were an H/T ratio of 2.5 mL/mg, a temperature of 187 °C, and a residence time of 150 min. The determined DE was 99.47%. The results of three-dimensional excitation-emission matrix (3D-EEM) fluorescence spectroscopy showed that the degradation of TCS generated humic acid-like organics with aromatic ring and phenolic hydroxyl functional groups. Furthermore, fluorescence spectroscopy analysis showed that the residence time increased from 30 to 120 min, and the fluorescence intensity difference increased from 100.5 to 218 au, which confirmed that •OH participated in the alkali-catalyzed HTO process. There were two different reaction pathways: (1) Cleavage of the C–O bond between the two benzene rings, followed by the opening and dechlorination of the benzene ring via oxidation; (2) The benzene rings were directly hydroxylated, opened, and dechlorinated through oxidation. The intermediate products were further oxidized to non-Cl-containing organics, achieving the harmless degradation of TCS. Further analysis showed that the total organic carbon (TOC) removal efficiency was about 80% at a residence time of 140 min, indicating that TCS was almost completely mineralized. This study provides a green and environmentally benign approach for the detoxification of chlorinated organic contaminants.

Abstract:The emerging contaminant triclosan (TCS) is widely distributed both in surface water and in wastewater and poses a threat to aquatic organisms and human health due to its resistance to degradation. The dioxygenase enzyme TcsAB has been speculated to perform the initial degradation of TCS, but its precise catalytic mechanism remains unclear. In this study, the function of TcsAB was elucidated using multiple biochemical and molecular biology methods. Escherichia coli BL21(DE3) heterologously expressing tcsAB from Sphingomonas sp. RD1 converted TCS to 2,4-dichlorophenol. TcsAB belongs to the group IA family of two-component Rieske nonheme iron ring-hydroxylating dioxygenases. The highest amino acid identity of TcsA and the large subunits of other dioxygenases in the same family was only 35.50%, indicating that TcsAB is a novel dioxygenase. Mutagenesis of residues near the substrate binding pocket decreased the TCS-degrading activity and narrowed the substrate spectrum, except for the TcsAF343A mutant. A meta-analysis of 1492 samples from wastewater treatment systems worldwide revealed that tcsA genes are widely distributed. This study is the first to report that the TCS-specific dioxygenase TcsAB is responsible for the initial degradation of TCS. Studying the microbial degradation mechanism of TCS is crucial for removing this pollutant from the environment.

Abstract:Triclosan (TCS), an extensively used broad-spectrum antimicrobial agent, has raised significant environmental concerns regarding its widespread occurrence in waters. In this study, the removal of TCS in aqueous solution via peroxymonosulfate (PMS) activated by an extremely low-level Co2+ (0.02 mu M) was systematically investigated. During preliminary test, TCS (10 mu M) was totally degraded in 30 min by using 0.1 mu M Co2+ and 40 mu M PMS at pH 7.0 with a degradation rate constant of 0.1219 min(-1). A first-order apparent degradation rate of TCS was found with respect to the PMS concentrations. At extremely low dosage of Co2+ (0.02 mu M), the presence of NO3-, HCO3-, PLFA, and SRHA within test concentrations significantly inhibited TCS removal, while a dual effect of Cl- on the degradation rate of TCS was observed. The quenching experiments verified that SO4- was the dominant reactive oxygen species (ROS) rather than %OH. Six major intermediates were identified using TOF-LC-MS, based on which we proposed three associated reaction pathways including hydroxylation, ether bond breakage, and dechlorination. Toxicity predictions by ECOSAR software exhibited aquatic toxicity reduction of TCS after Co2+/PMS treatment. We outlook these findings to advance the feasibility of organic contaminants removal via Co2+/PMS system with Co2+ at extremely low levels.

Abstract:Triclocarban (TCC) and its metabolite, 3,4-dichloroaniline (DCA), are classified as emerging organic contaminants (EOCs). Significant concerns arise from water and soil contamination with TCC and its metabolites. These concerns are especially pronounced at high concentrations of up to approximately 20 mg/kg dry weight, as observed in wastewater treatment plants (WWTPs). Here, a TCC-degrading co-culture system comprising Rhodococcus rhodochrous BX2 and Pseudomonas sp. LY-1 was utilized to degrade TCC (14.5 mg/L) by 85.9% in 7 days, showing improved degradation efficiency compared with monocultures. A combination of high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), genome sequencing, transcriptomic analysis, and quantitative reverse transcription-PCR (qRT-PCR) was performed. Meanwhile, through the combination of further experiments involving heterologous expression and gene knockout, we proposed three TCC metabolic pathways and identified four key genes (tccG, tccS, phB, phL) involved in the TCC degradation process. Moreover, we revealed the internal labor division patterns and connections in the co-culture system, indicating that TCC hydrolysis products were exchanged between co-cultured strains. Additionally, mutualistic cooperation between BX2 and LY-1 enhances TCC degradation efficiency. Finally, phytotoxicity assays confirmed a significant reduction in the plant toxicity of TCC following synergistic degradation by two strains. The in-depth understanding of the TCC biotransformation mechanisms and microbial interactions provides useful information for elucidating the mechanism of the collaborative biodegradation of various contaminants.
Alkali-catalyzed hydrothermal oxidation treatment of triclosan in soil: Mechanism, degradation pathway and toxicity evaluation

Abstract:The continuous accumulation of chlorinated organic pollutants in soil poses a potential threat to ecosystems and human health alike. Alkali-catalyzed hydrothermal oxidation (HTO) can successfully remove chlorinated organic pollutants from water, but it is rarely applied to soil remediation. In this work, we assessed this technique to degrade and detoxify triclosan (TCS) in soil and we determined the underlying mechanisms. The results showed a dechlorination efficiency of TCS (100 mg per kg soil) of 49.03 % after 120 min reaction (H 2 O 2 /soil ratio 25 mL·g −1 , reaction temperature 180 °C in presence of 1 g·L −1 NaOH). It was found that soil organic constituents (humic acid, HA) and inorganic minerals (SiO 2 , Al 2 O 3 , and CaCO 3 ) suppressed the dechlorination degradation of TCS, with HA having the strongest inhibitory effect. During alkali-catalyzed HTO, the TCS molecules were effectively destroyed and humic acid-like or fulvic acid-like organics with oxygen functional groups were generated. Fluorescence spectroscopy analysis showed that hydroxyl radicals (OH) were the dominant reactive species of TCS degradation in soil. On the basis of the Fukui function and the degradation intermediates, two degradation pathways were proposed. One started with cleavage of the ether bond between the benzene rings of TCS, followed by dechlorination and the opening of benzene via oxidation. The other pathway started with direct hydroxylation of the benzene rings of TCS, after which they were opened and dechlorinated through oxidation. Analysis of the soil structure before and after treatment revealed that the soil surface changed from rough to smooth without affecting soil surface elements. Finally, biotoxicity tests proved that alkali-catalyzed HTO effectively reduced the toxicity of TCS-contaminated soil. This study suggests that alkali-catalyzed hydrothermal oxidation provides an environmentally friendly approach for the treatment of soil contaminated with chlorinated organics such as TCS.

Abstract:Triclocarban (TCC), an emerging organic contaminant, poses a potential threat to human health with long-term exposure. Here, Rhodococcus rhodochrous BX2 and Pseudomonas sp. LY-1 were utilized to degrade TCC at environmental related concentrations for enhancing TCC biodegradation and investigating whether the toxicity of intermediate metabolites is lower than that of the parent compound. The results demonstrated that the bacterial consortium could degrade TCC by 82.0% within 7 days. The calculated 96 h LC50 for TCC, as well as its main degradation product 3,4-Dichloroaniline (DCA) were 0.134 mg/L and 1.318 mg/L respectively. Biodegradation also alleviated histopathological lesions induced by TCC in zebrafish liver and gut tissues. Liver transcriptome analysis revealed that biodegradation weakened differential expression of genes involved in disrupted immune regulation and lipid metabolism caused by TCC, verified through RT-qPCR analysis and measurement of related enzyme activities and protein contents. 16 S rRNA sequencing indicated that exposure to TCC led to gut microbial dysbiosis, which was efficiently improved through TCC biodegradation, resulting in decreased relative abundances of major pathogens. Overall, this study evaluated potential environmental risks associated with biodegradation of TCC and explored possible biodetoxification mechanisms, providing a theoretical foundation for efficient and harmless bioremediation of environmental pollutants.
Enrichment of Tetracycline-Degrading Bacterial Consortia: Microbial Community Succession and Degradation Characteristics and Mechanism

Abstract:Tetracycline (TC) is an antibiotic that is recently found as an emerging pollutant with low biodegradability. Biodegradation shows great potential for TC dissipation. In this study, two TC-degrading microbial consortia (named SL and SI) were respectively enriched from activated sludge and soil. Bacterial diversity decreased in these finally enriched consortia compared with the original microbiota. Moreover, most ARGs quantified during the acclimation process became less abundant in the finally enriched microbial consortia. Microbial compositions of the two consortia as revealed by 16 S rRNA sequencing were similar to some extent, and the dominant genera Pseudomonas, Sphingobacterium, and Achromobacter were identified as the potential TC degraders. In addition, consortia SL and SI were capable of biodegrading TC (initial 50 mg/L) by 82.92% and 86.83% within 7 days, respectively. They could retain high degradation capabilities under a wide pH range (4−10) and at moderate/high temperatures (25–40 °C). Peptone with concentrations of 4–10 g/L could serve as a desirable primary growth substrate for consortia to remove TC through co-metabolism. A total of 16 possible intermediates including a novel biodegradation product TP245 were detected during TC degradation. Peroxidase genes, tetX-like genes and the enriched genes related to aromatic compound degradation as revealed by metagenomic sequencing were likely responsible for TC biodegradation.

Abstract:Triclosan (TCS) is an emerging contaminant that threatens the environment and human health. This study was conducted to investigate TCS abatement by a novel electro-oxidation (EO) process, which used a Ti-based nickel and antimony doped tin oxide (NATO/Ti) anode and a carbon nanotubes (CNTs) doped carbon/PTFE (CNTs-C/PTFE) gas diffusion electrode (GDE) for oxygen reduction reaction (ORR). A comparative study was also performed for TCS degradation by using a traditional EO with a nickel foam cathode, termed as HER-EO. The optimal initial TCS concentration, current density and solution pH for TCS degradation during the ORR-EO and HER-EO were investigated. Results showed that ORR-EO removed more than 98% of TCS in 10–60 min under the concentration of 5–50 mg/L. The TCS degradation followed pseudo-first-order kinetics and its main intermediates were observed during the ORR-EO and HER-EO using liquid chromatography combined mass (LC-MS). The results of FED analysis and toxicity prediction by ECOSAR software showed that less intermediates accumulated during the ORR-EO and the residues were less harmful. The ORR-EO degradation mechanism for TCS was attacking on the ether bond and the benzene ring by •OH. This novel ORR-EO process exhibits a great merit in the field of emerging contaminants abatement.

Abstract:In this study, different methods of electron beam (EB) irradiation and potassium ferrocyanide (K4Fe(CN)(6)) activated by simulated sunlight irradiation (lambda >= 290 nm) were adopted to generate hydrated electron (e(aq)(-)), respectively. Steady-state and laser flash photolysis experiments were conducted to evaluate the feasibility of using e(aq)(-) reduction to enhance the dechlorination of TCS in aqueous solution. Under the EB irradiation, e(aq)(-) was proved to be an effective radical, and significant degradation of TCS was observed. Under simulated sunlight irradiation, TCS degradation efficiency within 60 min increased from 70.3% (without K4Fe(CN)(6)) to 100% [TCS](0)/[K4Fe(CN)(6)](0) = 1/5), and the average dechlorination percentage after 2 h increased from 62.7% to 90.7%, respectively. The degradation of TCS in the presence of e(aq)(-) adhered to the apparent first-order kinetics, and the optimal rate constant (k(p)) was (1.54 +/- 0.02) x 10(-3) s(-1). Abstract:Chlorinated phenolic compounds are a class of toxic and refractory organic pollutants. The pollution caused by chlorophenols poses serious ecological and environmental problems. A stable bacterial consortium capable of reductively dechlorinating trichlorophenol was isolated using chlorophenol as the sole source of carbon and energy. The physiological characteristics of the mixed cultures were studied and the results show that the consortium could use pyruvate as the carbon and energy source. The fermentation of pyruvate, sulfate reduction and dechlohnation process proceeded strictly in succession within this consortium. The effect of specific inhibitors on the dechlorinating activity of the consortium was investigated, and the results indicate that sulfate and molybdate (1 mmol/L) have a strong inhibitive influence on the dechlohnation activity. Fluorescence in situ hybridization (FISH) technique was applied to analyzing the composition of the consortium and the results reveal that one major subpopulation within the consortium was phylogenetically affiliated to gamma and delta subclass of Proteobacteria.

Abstract:Triclosan (TCS) is an antimicrobial compound ubiquitously found in surface waters throughout the world. Although several studies have focused on the photochemical degradation of TCS, there is still limited knowledge about its environmental fate. In this study, we got molecular-level insights into the photochemical degradation of TCS. Significant stable carbon isotope fractionation was observed during photodegradation; different bond-cleavage reaction pathways under different photolytic conditions were characterized, using compound specific isotope analysis (CSIA). Photochemical modeling of TCS photodegradation showed that direct photolysis would be the main transformation pathway if pH > 7, even in presence of dissolved organic matter. Moreover, by use of ultrahigh resolution mass spectrometry, FT-ICR-MS, a broad and complex spectrum of organic by-products (some of which potentially toxic, as assessed by a quantitative structure-activity relationship approach) were identified. A detailed reaction mechanism was developed on the basis of the detected compounds. A possible sequence of steps leading to some of the detected product compounds in aqueous solution is suggested.

Abstract:Triclocarban (TCC) is an emerging and intractable environmental contaminant due to its hydrophobicity and chemical stability. However, the antibacterial property of TCC limits its biodegradation, and only the functional enzyme TccA involved in TCC degradation has been characterized to date. In this study, we report a highly efficient TCC-degrading bacterium, Rhodococcus rhodochrous BX2, that could degrade and mineralize TCC (10 mg/L) by 76.8% and 56.5%, respectively, within 5 days. Subsequently, the TCC biodegradation pathway was predicted based on the detection of metabolites using modern mass spectrometry techniques. Furthermore, an amidase (TccS) and a novel phenol hydroxylase (PHIND) encoded by the tccS and PHIND genes, respectively, were identified by genomic and transcriptomic analyses of strain BX2, and these enzymes were further unequivocally proven to be the key enzymes responsible for the metabolism of TCC and its intermediate 4-chloroaniline (4-CA) by using a combination of heterologous expression and gene knockout. Our results shed new light on the mechanism of TCC biodegradation and better utilization of microbes to remediate TCC contamination.
Microbial Communities Associated with Sustained Anaerobic Reductive Dechlorination of -, -, -, and -Hexachlorocyclohexane Isomers to Monochlorobenzene and Benzene

Abstract:Intensive historical and worldwide use of pesticide formulations containing hexachlorocyclohexane (HCH) has led to widespread contamination. We derived four anaerobic enrichment cultures from HCH-contaminated soil capable of sustainably dechlorinating each of alpha-, beta-, gamma-, and delta-HCH isomers stoichiometrically to benzene and monochlorobenzene (MCB). For each isomer, the dechlorination rates, inferred from production rates of the dechlorinated products, MCB and benzene, increased progressively from <3 to similar to 12 mu M/day over 2 years. The molar ratio of benzene to MCB produced was a function of the substrate isomer and ranged from beta (0.77 +/- 0.15), alpha (0.55 +/- 0.09), gamma (0.13 +/- 0.02), to delta (0.06 +/- 0.02) in accordance with pathway predictions based on prevalence of antiperiplanar geometry. Data from 16S rRNA gene amplicon sequencing and quantitative PCR revealed significant increases in the absolute abundances of Pelobacter and Dehalobacter, most notably in the alpha-HCH and delta-HCH cultures. Cultivation with a different HCH isomer resulted in distinct bacterial communities, but similar archaeal communities. This study provides the first direct comparison of shifts in anaerobic microbial communities induced by the dechlorination of distinct HCH isomers. It also uncovers candidate microorganisms responsible for the dechlorination of alpha-, beta-, gamma-, and delta-HCH, a key step toward better understanding and monitoring of natural attenuation processes and improving bioremediation technologies for HCH-contaminated sites.

Abstract:Triclosan (TCS), a kind of pharmaceuticals and personal care products (PPCPs), is widely used and has had a large production over years. It is an emerging pollutant in the water environment that has attracted global attention due to its toxic effects on organisms and aquatic ecosystems, and its concentrations in the water environment are expected to increase since the COVID-19 pandemic outbreak. Some researchers found that microbial degradation of TCS is an environmentally sustainable technique that results in the mineralization of large amounts of organic pollutants without toxic by-products. In this review, we focus on the fate of TCS in the water environment, the diversity of TCS-degrading microorganisms, biodegradation pathways and molecular mechanisms, in order to provide a reference for the efficient degradation of TCS and other PPCPs by microorganisms.

Abstract:Photodegradation is an essential process for the in-stream elimination of triclosan (TCS). There are still some knowledge gaps concerning reaction kinetics, pathways, and transformation products, which are important for understanding the environmental fate of TCS. For example, the effects of ionic strength and day-night alternation on TCS photodegradation have been barely considered in previous studies. To fill these gaps, this study assesses the effect of sulfate ions (SO 4 2− ) on the photodegradation of TCS under day-night shifts. Compound-specific carbon and chlorine isotope analysis were applied to characterize different bond-cleavage pathways. Moreover, variations in TCS reaction products distribution were analyzed by using Fourier transform ion cyclotron resonance mass spectrometry. A numerical model was developed to describe concentration and stable carbon isotopic evolution of TCS under different conditions. Based on the observed experimental data, we show that SO 4 2− can affect the kinetics and product distribution during the photodegradation of TCS. In particular, sulfate ions (100 mM) inhibited the yield of the toxic product, 2,8-dichlorodibenzo-p-dioxin (2,8-DCDD). Furthermore, day-night alternations lead to different predominant bond-cleavage pathways depending on sulfate concentration. By so doing, we propose a realistic framework for the direct photodegradation of TCS in sunlit natural surface waters.
