Abstract:Surface-enhanced Raman scattering (SERS) offers a route for trace detection of mercury ions (Hg2+) owing to its great enhancement near the surface of the substrate. However, most of the methods are inconvenient due to the long reaction time and the strict measurement conditions. Based on a reaction between silver nanoparticles (AgNPs) and mercury ions, a simple but effective method is proposed in this paper. In the experimental research, a tapered optical fiber (TOF) modified with AgNPs is employed as a sensing probe and Rhodamine 6G (R6G) is taken as the Raman reporter. The SERS substrate is consumed by the Hg2+, causing a spectrum decrease. In the Hg2+ concentration range of 10-12 M to 10-4 M, the spectra evolution is detected using a mobile Raman spectrometer. By analyzing the decrease of the Raman peak after a 5-minutes interval, the concentration of Hg2+ can be quantified. A limit of detection of 5.15 x 10-13 M is achieved, which is much below the maximum Hg2+ content of drinkable water specified by USEPA (10-8 M). Verification tests were performed with the inductively coupled plasma-mass spectrometry (ICP-MS), the results showed that the proposed SERS method is effective for real samples detection. In addition, the reproductivity and the selectivity are experimentally confirmed. With this reliable, rapid, ultrasensitive and accurate method, trace Hg2+ could be detected in many applications.

Abstract:A technique for encapsulating fluorescent organic probes in a micelle system offers an important alternative method to manufacture watersoluble organic nanoparticles (ONPs) for use in sensing Hg2+. This article reports on a study of a surfactant-free micelle-like ONPs based on a 3,6-di(2-thieny1)-2,5-dihydropyrrolo[3,4-c]pyrrole-1,4-dione (TDPP) amphiphile, (2(2-(2-methoxyethoxy)ethyl)-3,6-di(2-thiophyl)-2,5-dihydropyrrolo [3,4-c] pyrrole-1,4-dione (NDPP) fabricated to monitor Hg2+ in water. NDPP was synthesized through a simple one-step modification of a commercially available dye TDPP with a flexible and hydrophilic alkoxy. This study reports, for the first time, that TDPP dyes can respond reversibly, sensitively, and selectively to Hg2+ through TDPP-Hg-TDPP complexation, similar to the well-known thymine(T) Hg thymine(T) model and the accompanying molecular aggregation. Interestingly, transmission electron microscopy (TEM) and dynamic light scattering (DLS) confirmed that, in water, NDPP forms loose micelle-like fluorescent ONPs with a hydrohobic TDPP portion encapsulated inside. These micelle-like nanoparticles offer an ideal location for TDPP Hg complexation with a modest molecular aggregation, thereby providing both clear visual and spectroscopic signals for Hg2+ sensing. An estimated detection limit of 11 nM for Hg2+ sensing with this NDPP nanoparticle was obtained. In addition, NDPP ONPs show good water solubility and high selectivity to Hg2+ in neutral or alkalescent water. It was superior to most micelle based nanosensors, which require a complicated process in the selection or synthesis of suitable surfactants. The determinations in real samples (river water) were made and satisfactory results were achieved. This study provides a low-cost strategy for fabricating small molecule-based fluorescent nanomaterials for use in sensing Hg2+. Moreover, the NDPP nanoparticles show potential ability in Hg2+ ion adsorption and recognization of cysteine using NDPP-Hg composite particle.

Abstract:As a kind of heavy metal ions, mercury ions (Hg2+) have important effects on ecological environment and human health. Since Hg2+ interact with mercaptan molecules in the living body, they gather in the living body and endanger health or even life, so the detection of Hg2+ is indispensable. In this research, taking advantage of thymine moiety as a specific ligand for chelating with Hg2+, a new type of "T-Hg-T" fluorescent probe BC-Hg was designed and synthesized to specially detect Hg2+ in real water samples and living systems. The barbituric acid unit in the probe can react with Hg2+ and then deprotonate to form polymer and precipitate. Probe BC-Hg has a rapid response to the detection of Hg2+ and strong anti-interference ability. It has a good linear detection of Hg2+ in the range of 0-1.5 mu M, so it can quantitatively detect Hg2+ within a certain concentration range. Probe BC-Hg has obvious "naked eye" colorimetric effect and can be used to determine the generation of Hg2+ polymer by the Tyndall effect. In addition, probe BC-Hg can also be used to detect Hg2+ in real water samples, cells, and zebrafish. Therefore, the probe can serve as an effective tool for detecting Hg2+ in environment and organisms.

Colorimetric and SERS Dual-Mode Sensing of Mercury (II) Based on Controllable Etching of Au@Ag Core/shell Nanoparticles

Abstract:Mercury ion (Hg2+) is a bio-accumulative and toxic heavy metal, which has caused significant damage to our health. It is vital to establish a method for the determination of Hg2+. Here, a new strategy for colorimetric/SERS dual-mode determination of Hg2+ was proposed based on controllable etching Au@Ag NPs. The silver shells could be etched by S2O32- causing the orange colloidal solution to turn into brick red. However, after Hg2+ is added, the etching effect is suppressed due to the reaction between Hg2+ and S2O32- to form an insoluble substance HgS. As a result, the UV-vis absorption at 394 nm and Raman intensity of R6 G increase with a silver shellless etched by S2O32-. About the colorimetric method, the limit of detection was 2 mu M and 0.2 mu M through the naked eye and UV-vis spectroscopy, respectively. Moreover, the SERS method possesses a linear regression for the concentration of Hg2+ (0.1 nM(-1) mu M), and the LOD was 0.1 nM. Furthermore, the sensing strategy has been used to determine Hg2+ in traditional Chinese medicine and real water samples. The selectivity and practicability studies indicate that our method has excellent specificity and sensitivity. Thus, this strategy is suitable for on-site fast detecting of Hg2+.
Naphthalimide-porphyrin Hybrid Based Ratiometric Bioimaging Probe for Hg2+: Well-Resolved Emission Spectra and Unique Specificity.

Abstract:In this paper, we unveil a novel naphthalimide-porphyrin hybrid based fluorescence probe (1) for ratiometric detection of Hg2+ in aqueous solution and living cells. The ratiometric signal change of the probe is based on a carefully predesigned molecule containing two independent Hg2+-sensitive fluorophores with their maximal excitation wavelengths located at the same range, which shows reversibly specific ratiometric fluorescence responses induced by Hg2+. In the new developed sensing system, the emissions of the two fluorophores are well-resolved with a 125 nm difference between two emission maxima, which can avoid the emission spectra overlap problem generally met by spectra-shift type probes and is especially favorable for ratiometric imaging intracellular Hg2+. It also benefits from a large range of emission ratios and thereby a high sensitivity for Hg2+ detection. Under optimized experimental conditions, the probe exhibits a stable response for Hg2+ over a concentration range from 1.0 x 10(-7) to 5.0 x 10(-5) M, with a detection limit of 2.0 x 10(-8) M. The response of the probe toward Hg2+ is reversible and fast (response time less than 2 min). Most importantly, the ratiometric fluorescence changes of the probe are remarkably specific for Hg2+ in the presence of other abundant cellular metal ions (i.e., Na+, K+, Mg2+, and Ca2+), essential transition metal ions in celsl (such as Zn2+, Fe3+, Fe2+, Cu2+, Mn2+, Co2+, and Ni2+) and environmentally relevant heavy metal ions (Ag+, Pb2+, Cr3+, and Cd2+), which meets the selective requirements for biomedical and environmental monitoring application. The recovery test of Hg2+ in real water samples demonstrates the feasibility of the designed sensing system for Hg2+ assay in practical samples. It has also been used for ratiometric imaging of Hg2+ in living cells with satisfying resolution, which indicates that our novel designed probe has effectively avoided the general emission spectra overlap problem of other ratiometric probes.

Abstract:In this work, a simple dichlororesorufin-based colorimetric and fluorescent probe R-Hg with the recognition receptor of carbon- othioate was designed, synthesized, and exploited to determine mercury ions in aqueous solution and living cells as well as in zebrafish. After a series of analytical tests, probe R-Hg showed some excellent characteristics, such as ultrasensitivity, high selectivity, and good water solubility. The probe solution exhibited a very large fluorescence enhancement (about 150-fold) upon addition of He2+. Additionally, probe R-Hg exhibited excellent sensitivity, which could quantificationally detect Hg2+ at the nanomolar level, and the detection limit was 0.49 nM. Moreover, probe R-Hg could be used as a "naked-eye" probe for monitoring Hg2+, because the color of its solution transformed remarkably from yellow into pink with the participation of Hg2+. Importantly, probe R-Hg has potential application value and has been successfully applied to the determination of He2+ in practical water samples and in living RAW 264.7 cells as well as in zebrafish as a bioimaging reagent.

Abstract:Mercury ion (Hg 2+ ) detection plays a crucial role in safeguarding the environment and protecting human health. In this study, we developed a microfluidic biosensing platform that utilizes a phosphorothioate-RNA (PS-RNA) probe and gold-coated magnetic nanoparticles to quantify Hg 2+ visually. The PS-RNA probe was linked to gold-coated magnetic nanoparticles via Au–S interaction and biotin–avidin interaction and then combined with polystyrene microspheres to form a self-assembled structure called a magnetic bead-AuNP-probe-polystyrene (MAPP) bead. We designed and fabricated a microfluidic chip that contained only a magnetic separator and a particle trap. The presence of Hg 2+ cut the PS-RNA probe, leading to the separation of magnetic beads and polystyrene microspheres. The MAPP was removed by the magnetic separator, while the cut polystyrene microspheres flowed along the channel and accumulated in the particle trap, forming a long visible strip. The length of the polystyrene microsphere strip was proportional to the Hg 2+ concentration. The microfluidic biosensing platform exhibited high sensitivity and specificity for Hg 2+ detection, with a detection limit of 21.9 nM. By integrating the microfluidic magnetic manipulation system with a sensitive nucleic acid probe, we developed an automated and visual biosensor that is a valuable reference for the on-site visual detection of heavy metal ions.

Abstract:A novel ratiometric fluorescent Hg(2+) detecting system was rationally developed based on the typical excited state intramolecular proton transfer (ESIPT) characteristic of the latent fluorophore, 2-(1-(p-tolyl)-1H-phenanthro[9,10-d]imidazol-2-yl)phenol (Pol) and the Hg(2+)-mediated cleavage of the vinyl group. The probe responds selectively to Hg(2+) over various other metal ions with a larger bathochromic shift (∼100 nm). The sensing mechanism was investigated in detail by fluorescence spectroscopy, NMR spectra and mass spectrometry. Taking advantage of the enhancement effect of dichloromethane on the ESIPT efficiency, a facile dichloromethane extraction was introduced in the process of detection of Hg(2+), which affords a high sensitivity for the probe with a detection limit of 7.8 × 10(-9) M for Hg(2+). By using the new strategy, the novel probe can be used for the detection of Hg(2+) in practical water samples with good recovery. Moreover, the probe was successfully applied to the fluorescence image of Hg(2+) in living cells. These results indicated that the probe and the proposed method have promising applications for Hg(2+) sensing in biological and environmental sciences.

Abstract:In this paper, an electrochemical aptamer sensor with high sensitivity and selectivity was proposed for the detection of mercury ion (Hg 2+ ). The orderly assembled gold nanorods-modified screen-printed electrode (AuNRs/SPE) was used to improve the sensor with higher sensitivity and data reproducibility. The sensor was designed based on the specific recognition of Hg 2+ with aptamer to form T-Hg 2+ -T binding structure. Two aptamer chains were applied in the designed sensor, aptamer S1 with terminal thiolate group was self-assembled on the AuNRs/SPE via Au-S bond, and complementary S2 labeled with ferrocene (Fc) would generate a strong redox electrical signal. The addition of Hg 2+ would mediate the folding of the T-base of S1 to form a hairpin structure, resulting in the dissociation of the Fc-labelled S2 from the sensing system and a significant reduction in the redox current. The electrochemical properties of the sensor were further investigated using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). The effects of aptamer reaction time, concentration, and target incubation time on the sensor response were also investigated. The results showed that the sensor had a sensitive response to Hg 2+ at concentrations ranging from 1 pM to 1 μM, with a detection limit of 0.3 pM. Furthermore, the proposed sensor was applied in the determination of Hg 2+ in real samples, demonstrating its potential practical value.

Abstract:We report a highly sensitive electrochemical sensor for the detection of Hg(2+) ions in aqueous solution by using a thymine (T)-rich, mercury-specific oligonucleotide (MSO) probe and gold nanoparticles (Au NPs)-based signal amplification. The MSO probe contains seven thymine bases at both ends and a "mute" spacer in the middle, which, in the presence of Hg(2+), forms a hairpin structure via the Hg(2+)-mediated coordination of T-Hg(2+)-T base pairs. The thiolated MSO probe is immobilized on Au electrodes to capture free Hg(2+) in aqueous media, and the MSO-bound Hg(2+) can be electrochemically reduced to Hg(+), which provides a readout signal for quantitative detection of Hg(2+). This direct immobilization strategy leads to a detection limit of 1 microM. In order to improve the sensitivity, MSO probe-modified Au NPs are employed to amplify the electrochemical signals. Au NPs are comodified with the MSO probe and a linking probe that is complementary to a capture DNA probe immobilized on gold electrodes. We demonstrated that this Au NPs-based sensing strategy brings about an amplification factor of more than 3 orders of magnitude, leading to a limit of detection of 0.5 nM (100 ppt), which satisfactorily meets the sensitivity requirement of U.S. Environmental Protection Agency (EPA). This Au NPs-based Hg(2+) sensor also exhibits excellent selectivity over a spectrum of interference metal ions. Considering the high sensitivity and selectivity of this sensor, as well as the cost-effective and portable features of electrochemical techniques, we expect this Au NPs amplified electrochemical sensor will be a promising candidate for field detection of environmentally toxic mercury.
A novel fluorescent probe based on rhodamine B derivative for highly selective and sensitive detection of mercury(II) ion in aqueous solution

Abstract:In the present work, a new rhodamine B Schiff base 3′,6′-bis(diethylamino)-2-(2-oxoethylideneamino)-spiro[isoindoline-1,9′-xanithen]-3-one (RHO) was designed and easily prepared as a Hg2+ fluorescent and colorimetric probe, which could selectively and sensitively detect Hg2+ in 7:3 (v/v) ethanol–water solution by showing enhanced fluorescence emission. Meanwhile distinct color change from colorless to pink also provided “naked eye” detection of Hg2+ owing to the ring opening of spirolactam of the rhodamine derivative. Other ions including alkali and alkaline earth metal ions (K+, Mg2+, and Ca2+), and some transition metal ions (Ni2+, Co2+, Fe2+, Cu2+, Zn2+, Cd2+, Ag+, Pb2+, Cr3+, and Fe3+) could hardly induce any interference. For further use in practical applications in optical ion sensing devices, we immobilized the probe onto silica spheres to explore its functionality in inorganic and organic hybrid materials. As a result, excellent discrimination of Hg2+ from other cations could also be obtained.

Abstract:The development of probes for selective detection of mercury ions (Hg2+) is an important mission to accomplish because of the toxicity and ubiquity of Hg2+. Herein, we designed and synthesized a novel fluorescent probe O-(N-butyl-1,8-naphthalimide)-4-yl-O-phenyl carbonothioate (CBONT) for selective and sensitive detection of Hg2+ by turn-on fluorescence spectroscopy. Probe CBONT exhibited a fast response for Hg2+ with excellent sensitivity (limit of detection = 1.9 nM, 3 sigma/slope), and it might be attributed to the adoption of a new recognition receptor of carbonothioate moiety. Additionally, probe CBONT could serve as a "naked-eye" indicator for Hg2+. Finally, probe CBONT could be successfully applied to detect the concentrations of Hg2+ in real water samples. Our proposed recognition receptor would open up new, exciting opportunities for designing highly selective and ultrasensitive fluorescent probes for the determination of Hg2+ in real water samples.

Abstract:Mercury ions are highly toxic to living organisms and environment, hence Monitoring and quantification of mercury ions remain great significance. Herein, we have designed and synthesized a novel reaction-based fluorescence probe RANS for detection of Hg2+ by using a xanthene derivate as fluorophore and a thiosemicarbazide moiety as the recognition site. By applying mercury-promoted desulfurization reactions, probe RANS exhibited excellent capability of detecting Hg2+, such as rapid response (120 s), high sensitivity (0.019 μM) and good selectivity. Upon reaction with Hg2+, the thiosemicarbazide group of probe RANS formed 1,3,4-oxadiazole along with a significant color change from colorless to light green. Importantly, RANS has been successfully applied to selectively imaging Hg2+ in living cells with low cytotoxicity and quantitative detection of Hg2+ in real water samples.

Multicolor Aptasensor Based on DNA-Induced Au–Ag Nanorods for Simultaneous and Visual Detection of Inorganic and Organic Mercury

Abstract:Compared to inorganic mercury (Hg2+), methyl-mercury (CH3Hg+) and ethyl-mercury (C2H5Hg+) (organic mercury) not only have a much stronger toxicity but also are more easily accumulated by marine organisms to produce bioamplification. Therefore, the simultaneously onsite detection of Hg2+ and organic mercury is of great significance to ensure the safety of seafood, and it is also a hard challenge. We designed a T-rich aptamer, HT7, for specifically recognizing Hg2+ and organic mercury and developed a multicolor aptasensor for simultaneous discrimination and detection of Hg2+ and organic mercury with only bare-eye observation using HT7 as a recognition probe and gold nanorods (AuNRs) as a signal. In the presence of Hg2+ and Ag+, Hg2+ preferentially and specifically bind with HT7 immobilized on AuNRs surface and induce the formation of a monolayer Ag/Hg amalgam on the AuNRs surface after reduction, resulting in a change in color from orange to faint purple and a corresponding shift in the absorption peak from 820 to 730 nm in the solution. However, in the presence of CH3Hg+ or C2H5Hg+ and Ag+, CH3Hg+ or C2H5Hg+ preferentially bind with HT7 immobilized on the AuNRs surface and induce the formation of a monolayer Ag0 on the AuNRs surface after reduction, which results in the change in color from orange to atrovirens and the corresponding shift in the absorption peak shift from 820 to 670 nm in the solution. Thus, the inorganic and organic mercury (total of CH3Hg+ and C2H5Hg+) can be specifically discriminated and detected by only bare-eye observation. The method can be used to simultaneously detect inorganic and organic mercury in seawater by the bare-eye observation with a visual detection limit of 2.0 ppm for Hg2+ and 10.0 ppm for organic mercury. The success of this study is a useful enlightenment to develop an instrument-free method for an onsite detection of trace inorganic and organic mercury in environment by a bare-eye observation, although the sensitivity of the method is relatively low.

Abstract:A novel thionocarbonate-coumarin-thiourea triad-based probe with dual recognition sites for sensing mercury (Hg2+) ion was developed. The synthesized probe possessed both fluorogenic ("off-on") and chromogenic (from colorless to blackish brown) sensing performance towards Hg2+ ions. The fluorescence intensity was increased by 70 fold after the addition of Hg2+. As expected, the probe exhibited excellent selectivity and sensitivity for Hg2+ compared to other common competitive metal ions. The fluorescence intensity of the probe improved linearly with the increase of the concentration of Hg2+ (0-40 μM). Also, the minimum limit of detection (LOD) of the synthesized probe was 0.12 μM. Considering the importance of test feasibility in the harsh environment, the developed probe was applicable for detecting Hg2+ ions over a broad working pH range of 3-11. It is reliable and qualifies for the quantitative determination of Hg2+ concentrations in actual water samples. Finally, the probe achieved the bioimaging performance of Hg2+ in living cells and plants with good biocompatibility.

Abstract:The detection of heavy metals such as Hg2+ and Ag+ is important and urgent. In this work, – NO2/– NH2/C=S boron dipyrromethene small molecular derivatives were synthesized at first. Then they were incorporated into polymer chains. The macromolecular fluorescent probes were obtained via Sonogashira reaction using the small molecular probes as building blocks. The as-prepared small-molecule fluorescent probe BO3 exhibits high sensing performance for Hg2+. By introducing it into macromolecules, the sensing ability still remains, and even more, the recognition performance is improved. The macromolecular fluorescent probes P1, P2, and P3 also have high recognition ability for Ag+ with a binding ratio of 2:1 (metal ion to probe ratio). Through the study of the sensing mechanism and the recycling experiments, it is found that the probes responded by the photo-induced electron transfer mechanism and can be recycled and reused. At the same time, BO3, P2, and P3 show excellent recognition performance for Hg2+ in living cells and zebrafish. Living cell imaging experiments indicated that these fluorescent probes had good cell membrane permeability and low cytotoxicity, and could realize bioimaging of Hg2+. Therefore, the application value of these fluorescent probes could be enlarged.

Abstract:Mercury ions as high toxic pollutants have received wide-spread attention because of their poisonousness, persistence and enrichment. To better understand the distribution of mercury species and supplement more detailed toxicological research, it is necessary to develop some methods for monitoring mercury ions with high sensitivity and selectivity. Therefore, a simple rhodol-based highly selective fluorescent probe, RH-Hg , has been developed for monitoring Hg^2+ with thiocarbamate as the recognition receptor. The probe RH-Hg can quantificationally detect mercury ions in aqueous solution assisted by hydrogen peroxide (H_2O_2), and it can discriminate Hg^2+ through “naked-eye” observation of the color changes from light orange to dark pink. Finally, the practical applications of the probe RH-Hg in the river water further demonstrated that it will be an effective and economical tool for monitoring the distribution of Hg^2+ in the environment.
Surface-enhanced Raman scattering in nanoliter droplets: towards high-sensitivity detection of mercury (II) ions

Abstract:We report a new method for the trace analysis of mercury (II) ions in water. The approach involves the use of droplet-based microfluidics combined with surface-enhanced Raman scattering (SERS) detection. This novel combination provides both fast and sensitive detection of mercury (II) ions in water. Specifically, mercury (II) ion detection is performed by using the strong affinity between gold nanoparticles and mercury (II) ions. This interaction causes a change in the SERS signal of the reporter molecule rhodamine B that is a function of mercury (II) ion concentration. To allow both reproducible and quantitative analysis, aqueous samples are encapsulated within nanoliter-sized droplets. Manipulation of such droplets through winding microchannels affords rapid and efficient mixing of the contents. Additionally, memory effects, caused by the precipitation of nanoparticle aggregates on channel walls, are removed since the aqueous droplets are completely isolated by a continuous oil phase. Quantitative analysis of mercury (II) ions was performed by calculating spectral peak area of rhodamine B at 1,647 cm(-1). Using this approach, the calculated concentration limit of detection was estimated to be between 100 and 500 ppt. Compared with fluorescence-based methods for the trace analysis of mercury (II) ions, the detection sensitivities were enhanced by approximately one order of magnitude. The proposed analytical method offers a rapid and reproducible trace detection capability for mercury (II) ions in water.
