Abstract:Diabetic ulcers are one of the common complications in diabetic patients. Delayed wound healing is associated with persistent pro-inflammatory M1 polarization, reduced angiogenesis and increased reactive oxygen species (ROS) in the microenvironment. Wound healing consists of multiple phases and therefore requires treatment tailored to each phase. In this study, a biphasic drug-releasing microneedle (MN) was fabricated to achieve early ROS scavenging and late accelerated angiogenesis to promote wound healing. Vascular endothelial growth factor (VEGF) was first encapsulated in methacryloylated sulfonated chitosan (SCSMA) microspheres (V@MP), and then V@MP was loaded into hyaluronic acid (HA) microneedles along with cerium dioxide nanoparticles (CONPs). Rapid dissolution of HA rapidly releases the CONPs to clear ROS, whereas the V@MP stays in the wound. SCSMA slow degradation prolongs the release of VEGF, thereby promoting angiogenesis. In vitro and in vivo studies have shown that this biphasic drug-releasing smart microneedle improves cell proliferation and migration, effectively scavenges ROS, promotes angiogenesis and tissue regeneration, and synergistically promotes M2 macrophage polarization. It provides a new delivery mode for nano-enzymes and growth factors that could be multifunctional and synergistic in the treatment of diabetic ulcers. STATEMENT OF SIGNIFICANCE: In our study, we present a microneedle (V@MP/C@MN) that can release drugs biphasically, which showed good repair ability in diabetic ulcer model. Large amounts of CONPs were rapidly released to alleviate oxidative stress during the inflammation of the wound, and V@MP stayed in the wound for a long period of time to release VEGF and promote angiogenesis in the late stage of wound healing. The results indicated that V@MP/C@MN could promote cell proliferation and migration, effectively scavenge ROS, promote angiogenesis and tissue regeneration, and synergistically promote M2 macrophage polarization, which could play a multifunctional and synergistic role in the treatment of diabetic ulcers.

Abstract:Chronic non‐healing diabetic wounds are a serious complication of diabetes, with a high morbidity rate that can cause disability or death. The long period of inflammation and dysfunctional angiogenesis are the main reasons for wound‐healing difficulty in diabetes. In this study, we constructed a multifunctional double‐layer microneedle (DMN) to control infection and promote angiogenesis, meeting the multiple demands of the healing process of a diabetic wound. The double‐layer microneedle consisted in a hyaluronic acid (HA) substrate and a mixture of carboxymethyl chitosan and gelatin (Gel‐CMC) as the tip. The antibacterial drug tetracycline hydrochloride (TH) was loaded into the substrate of the microneedle to achieve rapid sterilization and promote resistance to external bacterial infections. The microneedle tip loaded with recombinant human epidermal growth factor (rh‐EGF) was inserted into the skin, in response to gelatinase produced by resident microbe and disassociate to achieve the enzymatic response release. The double‐layer drug‐loaded microneedles (DMN@TH/rh‐EGF) have antibacterial and antioxidant effects, and promote cell migration and angiogenesis in vitro. In an in vivo diabetic wound model, using rats, the DMN@TH/rh‐EGF patch was able to inhibit inflammation, promote angiogenesis, collagen deposition, and tissue regeneration during the wound healing process, promoting its healing. Abstract:High glucose level, bacterial infection, and persistent inflammation within the microenvironment are key factors contributing to the delay of diabetic ulcers healing, while traditional therapeutic methods generally fail to address these issues simultaneously. Here, we present a spatiotemporally responsive cascade bilayer microneedle (MN) patch for accelerating diabetic wound healing via local glucose depletion and sustained nitric oxide (NO) release for long-term antibacterial and anti-inflammatory effects. The MN patch (G/AZ-MNs) possesses a degradable tip layer loading glucose oxidase (GOx), as well as a dissolvable base layer encapsulating L-arginine (Arg)-loaded nanoparticles (NPs). After wound administration, the base part rapidly dissolved, resulting in prompt separation of the MN tip within the wound tissue, which subsequently responded to the overexpressed matrix metalloproteinase-9 (MMP-9) in diabetic lesions, leading to the responsive release of GOx. The released enzyme catalyzed glucose into gluconic acid and hydrogen peroxide (H 2 O 2 ), which not only reduced glucose level within the diabetic wound, but also initiated the cascade reaction between H 2 O 2 with the Arg that was released from NPs, thereby achieving continuous production of NO for 7 days. Our findings demonstrate that a single administration of the MN patch could effectively heal non-infected or biofilm-infected diabetic wounds with the multifunctional properties.

Collisional depolarization of OH(A) with Ar: Experiment and theory.

Abstract:Zeeman quantum beat spectroscopy has been used to measure the 300 K rate constants for the angular momentum depolarization of OH(A (2)Sigma(+)) in the presence of Ar. We show that the beat amplitude at short times, in the absence of collisions, is well described by previously developed line strength theory for (1+1) laser induced fluorescence. The subsequent pressure dependent decay of the beat amplitude is used to extract depolarization rate constants and estimates of collisional depolarization cross sections. Depolarization accompanies both inelastic collisions, giving rise to rotational energy transfer, and elastic collisions, which change m(j) but conserve j. Previous experimental studies, as well as classical theory, suggest that elastic scattering contributes around 20% to the observed total depolarization rate at low j. Simulation of the experimental beat amplitudes, using theoretical calculations presented in the preceding paper, reveals that depolarization of OH(A) by Ar has a rate constant comparable to, if not larger than, that for energy transfer. This is consistent with a significant tilting or realignment of j(') away from j on collision. The experimental data are used to provide a detailed test of quantum mechanical and quasiclassical trajectory scattering calculations performed on a recently developed ab initio potential energy surface of Kłos et al. [J. Chem. Phys. 129, 054301 (2008)]. The calculations and simulations account well for the observed cross sections at high N, but underestimate the experimental results by between 10% and 20% at low N, possibly due to remaining inaccuracies in the potential energy surface or perhaps to limitations in the dynamical approximations made, particularly the freezing of the OH(A) bond.

Abstract:Oxygen plays an important role in diabetic chronic wound healing by regulating various life activities such as cell proliferation, migration, and angiogenesis. Therefore, oxygen-delivering systems have drawn much attention and evolved continuously. Here, we propose that an active Chlorella vulgaris (Cv)-loaded separable microneedle (MN) can be used to control oxygen delivery, which then promotes wound healing. The Cv-loaded microneedles (CvMN) consist of a polyvinyl acetate (PVA) substrate and gelatin methacryloyl (GelMA) tips with encapsulated Cv. Once CvMN is applied to diabetic wound, the PVA basal layer is rapidly dissolved in a short time, while the noncytotoxic and biocompatible GelMA tips remain in the skin. By taking advantage of the photosynthesis of Cv, oxygen would be continuously produced in a green way and released from CvMN in a controlled manner. Both in vitro and in vivo results showed that CvMN could promote cell proliferation, migration, and angiogenesis and enhance wound healing in diabetic mice effectively. The remarkable therapeutic effect is mainly attributed to the continuous generation of dissolved oxygen in CvMN and the presence of antioxidant vitamins, gamma-linolenic acid, and linoleic acid in Cv. Thus, CvMN provides a promising strategy for diabetic wound healing with more possibility of clinical transformations.
A one-stop integrated natural antimicrobial microneedles with anti-inflammatory, pro-angiogenic and long-term moisturizing properties to accelerate diabetic wound healing

Abstract:Diabetic ulcers present a formidable obstacle in diabetes management, typically leading to high mortality and amputation rates. To overcome traditional monotherapy drawbacks, We developed a novel microneedle strategy for combined antimicrobial action: ingeniously integrating quercetin with Platelet-derived Growth Factor-BB(PDGF-BB) and Sucrose Octasulfate(SOS) into the microneedle system(QPS MN). This method allows to penetrate through biofilms, administering quercetin nanocrystals and PDGF-BB deep into the tissue to combat microbial infection, mitigate inflammation, and promote angiogenesis. The accompanying backing material contains SOS, which absorbs wound exudate and forms a dressing that provides a moist environment for wound healing In an in vitro wound-scratch assay demonstrated that co-cultivating Human Umbilical Vein Endothelial Cells(HUVEC) with QPS MN for 48 h (90.3 ± 2.51 %) significantly enhanced cell migration compared to the control group (20.2 ± 1.41 %). Moreover, treatment of streptozotocin-induced diabetic wounds in rats with QPS MN for 14 days resulted in a wound healing rate of 96.56 ± 3.44 %, far surpassing the healing rate of only 40.34 ± 7.26 % observed in the untreated control group. Furthermore, the QPS MN treated wounds exhibited a notable increase in skin appendages and neovascularisation, indicating promising potential for achieving complete wound healing. These results suggest that QPS MN may offer substantial therapeutic benefits for addressing diabetic wounds.

Abstract:Chronic hard-healing diabetic wounds constitute an immense health and economic burden on global health. Great efforts have been put to develop multifunctional drug delivery systems for improving wound healing. Here, inspired by the physiologically immunogenic response processes, we presented a novel hierarchically structured delivery system with macrophage-like nanovesicle (NV) encapsulation for promoted diabetic wound healing. The NVs were assembled by the fusion membranes of macrophage and synthetic lipid; while the hierarchically structured delivery system was constructed by encapsulating these NVs inside hydrogel microspheres (NV-MPs) using microfluidics. The hydrogel MPs could protect and release the encapsulated NVs in the oxidative stress environment of the diabetic wound. As the NVs possessed the ability of pro-inflammatory cytokine neutralization and intracellular delivery of their loaded simvastatin (SIM), the bioinspired hierarchically structured delivery system could greatly enhance the migration ability and tube formation of human umbilical vein endothelial cells (HUVECs). Based on these features, the practical performances of the resultant delivery system were demon-strated in full-thickness cutaneous wounds in type I diabetic rats, which exhibited desirable wound healing conditions by effectively reducing inflammation levels and facilitating blood vessel formation and collagen deposition in the wound tissues. Thus, we believe that the proposed bioinspired hierarchical delivery system would be an ideal carrier for treating wound healing and other tissue regeneration-related applications.
Multifunctional Bioactive Nanozyme Systems for Enhanced Diabetic Wound Healing

Abstract:The protracted transition from inflammation to proliferation in diabetic wound healing poses significant challenges, exacerbated by persistent inflammatory responses and inadequate vascularization. To address these issues, a novel nanozymatic therapeutic approach utilizing asymmetrically structured MnO₂-Au-mSiO₂@aFGF Janus nanoparticles is engineered. Nanozymes featuring a mSiO₂ head and MnO₂ extensions, into which acidic fibroblast growth factor (aFGF) is encapsulated, resulting in MnO₂-Au-mSiO₂@aFGF Janus nanoparticles (mSAM@aFGF), are synthesized. This nanozyme system effectively emulates enzymatic activities of catalase (CAT) and superoxide dismutase (SOD), catalyzing degradation of reactive oxygen species (ROS) and generating oxygen. In addition, controlled release of aFGF fosters tissue regeneration and vascularization. In vitro studies demonstrate that mSAM@aFGF significantly alleviates oxidative stress in cells, and enhances cell proliferation, migration, and angiogenesis. An injectable hydrogel based on photocrosslinked hyaluronic acid (HAMA), incorporating the nanozymatic ROS-scavenging and growth factor-releasing system, is developed. The HAMA-mSAM@aFGF hydrogel exhibits multifaceted benefits in a diabetic wound model, including injectability, wound adhesion, hemostasis, anti-inflammatory effects, macrophage polarization from M1 to M2 phenotype, and promotion of vascularization. These attributes underscore the potential of this system to facilitate transition from chronic inflammation to the proliferative phase of wound repair, offering a promising therapeutic strategy for diabetic wound management.
Using Hybrid MnO2-Au Nanoflowers to Accelerate ROS Scavenging and Wound Healing in Diabetes

Abstract:Objectives: Excessive reactive oxygen species (ROS) in diabetic wounds are major contributors to chronic wounds and impaired healing, posing significant challenges in regenerative medicine. Developing innovative drug delivery systems is crucial to address these issues by modifying the adverse microenvironment and promoting effective wound healing. Methods: Herein, we designed a novel drug delivery platform using manganese dioxide nanoflower hybridized gold nanoparticle composites (MnO2-Au) synthesized via a hydrothermal reaction, and investigated the potential of MnO2-Au nanoflowers to relieve the high oxidative stress microenvironment and regulate diabetic wound tissue healing. Results: This hybrid material demonstrated superior catalytic activity compared to MnO2 alone, enabling the rapid decomposition of hydrogen peroxide and a substantial reduction in ROS levels within dermal fibroblasts. The MnO2-Au nanoflowers also facilitated enhanced dermal fibroblast migration and Col-I expression, which are critical for tissue regeneration. Additionally, a hydrogel-based wound dressing incorporating MnO2-Au nanoflowers was developed, showing its potential as an intelligent drug delivery system. This dressing significantly reduced oxidative stress, accelerated wound closure, and improved the quality of neonatal epithelial tissue regeneration in a diabetic rat skin defect model. Conclusions: Our findings underscore the potential of MnO2-Au nanoflower-based drug delivery systems as a promising therapeutic approach for chronic wound healing, particularly in regenerative medicine.

Biofilm Microenvironment Triggered Self-Enhancing Photodynamic Immunomodulatory Microneedle for Diabetic Wound Therapy.

Abstract:The treatment of diabetic wounds faces enormous challenges due to complex wound environments, such as infected biofilms, excessive inflammation, and impaired angiogenesis. The critical role of the microenvironment in the chronic diabetic wounds has not been addressed for therapeutic development. Herein, we develop a microneedle (MN) bandage functionalized with dopamine-coated hybrid nanoparticles containing selenium and chlorin e6 (SeC@PA), which is capable of the dual-directional regulation of reactive species (RS) generation, including reactive oxygen species (ROS) and reactive nitrogen species (RNS), in response to the wound microenvironment. The SeC@PA MN bandage can disrupt barriers in wound coverings for efficient SeC@PA delivery. SeC@PA not only depletes endogenous glutathione (GSH) to enhance the anti-biofilm effect of RS, but also degrades GSH in biofilms through cascade reactions to generate more lethal RS for biofilm eradication. SeC@PA acts as an RS scavenger in wound beds with low GSH levels, exerting an anti-inflammatory effect. SeC@PA also promotes the M2-phenotype polarization of macrophages, accelerating wound healing. This self-enhanced, catabolic and dynamic therapy, activated by the wound microenvironment, provides an approach for treating chronic wounds.

Abstract:The treatment of diabetic wounds remains a major challenge in clinical practice, with chronic wounds characterized by multiple drug-resistant bacterial infections, angiopathy, and oxidative damage to the microenvironment. Herein, a novel in situ injectable HA@MnO2 /FGF-2/Exos hydrogel is introduced for improving diabetic wound healing. Through a simple local injection, this hydrogel is able to form a protective barrier covering the wound, providing rapid hemostasis and long-term antibacterial protection. The MnO2 /ε-PL nanosheet is able to catalyze the excess H2 O2 produced in the wound, converting it to O2 , thus not only eliminating the harmful effects of H2 O2 but also providing O2 for wound healing. Moreover, the release of M2-derived Exosomes (M2 Exos) and FGF-2 growth factor stimulates angiogenesis and epithelization, respectively. These in vivo and in vitro results demonstrate accelerated healing of diabetic wounds with the use of the HA@MnO2 /FGF-2/Exos hydrogel, presenting a viable strategy for chronic diabetic wound repair.

Synergistically Detachable Microneedle Dressing for Programmed Treatment of Chronic Wounds

Abstract:Chronic wounds such as diabetic feet undergo a lifetime risk of developing into incurable ulcers. Current treatments for chronic wounds remain unsatisfactory due to the lack of ideal wound dressings that integrate facile dressing change, long-acting treatment, and high therapeutic efficacy into one system. Herein, a synergistically detachable microneedle (MN) dressing with a dual-layer structure is presented to enable programmed treatment via one-time dressing application. Such a dual-layer dressing MN system (DDMNS) is composed of chitosan (CS) hydrogel dressing (CSHD) on top of a detachable MN patch with a CS tip and a polyvinyl pyrrolidone (PVP) backing substrate incorporated with magnesium (Mg). The synergistic detachment is achieved with the backing Mg/PVP substrate dissolving within minutes due to the local moist environment of the CSHD enhancing the reaction between Mg and inflammation microenvironment. The combined treatment of Mg and panax notoginseng saponins (PNS) loaded in DDMNS achieves antibacterial, neovascularization, and activating a benign immune response so that the three overlapping periods of the inflammation, tissue proliferation, and tissue remodeling of wound healing reach a dynamic balance. This advanced DDMNS provides a facile approach for the programmed treatment of chronic wound management indicating potential value in wound healing and other related biomedical fields.

Abstract:Diabetes chronic wound is a severe and frequently occurring medical issue in patients with diabetes that often leads to more serious complications. Microneedles (MNs) can be used for wound healing as they can effectively pierce the epidermis and inject drugs into the wound tissue. However, common MN patches cannot provide sufficient skin adhesion to prevent detachment from the wound area. Inspired by the barb hangnail microstructure of porcupine quills, a porcupine quill-like multilayer MN patch with an adhesive back patching for tissue adhesion and diabetic wound healing was designed. Sodium hyaluronate-modified CaO2 nanoparticles and metformin (hypoglycemic agent) were loaded into the polycaprolactone tips of MNs, endowing them with exceptional antibacterial ability and hypoglycemic effect. A flexible and adhesive back patching was formed by polyacrylamide-polydopamine/Cu2+ composite hydrogel, which ensures that the MN patches do not peel off from the application sites and reduce bacterial infection. The bioinspired multilayer structure of MN patches exhibits satisfactory mechanical and antibacterial properties, which is a potential multifunctional dressing platform for promoting wound healing.Statement of significanceThe porcupine quill-like microneedles (MNs) with PAM-PDA/Cu2+ (PPC) composite hydrogel back patching have been fabricated, which can enhance the adhesion property of MNs to the skin through a physical interlock of multilayer MNs and chemical bonding of hydrogel patching. CaO2-HA NPs and metformin were loaded into the polycaprolactone tips of MNs, endowing them with the exceptional antibacterial ability and hypoglycemic effect, which could accelerate diabetic wound healing.As a safe and effective strategy in transdermal delivery of drugs, the as-fabricated flexible multilayer MN patch with good antibacterial, hypoglycemic, and biocompatibility has been used to promote the healing of diabetic wound by releasing oxygen and inhibiting inflammation at the wound site.

Abstract:Deoxyribonucleic acid hydrogel microneedles combined with double bionics and Chinese paper‐cutting technology have excellent stability, skin penetration, clamping, large deformation, tissue adhesion, and antibacterial properties. The loaded extracellular vesicle endows the system with immune regulation and activation pathways, thereby reducing pathological pain and promoting angiogenesis, neurogenesis, and tissue regeneration. Due to the high blood glucose and low oxygen environment in diabetes wounds, complex pathological problems arise, such as failure to upregulate healing factors, biological barriers generation, microvascular lesions, and skin neuropathy. Inspired by nature and traditional Chinese paper‐cutting technology, polypeptide deoxyribonucleic acid (DNA) hydrogel microneedles (MNs) (P‐DNA gel MNs) are prepared. The obtained P‐DNA gel MNs possess optimized structure and large deformability, providing excellent clamping, stability, skin penetration, large deformation, tissue adhesion, and antibacterial properties. By incorporating extracellular vesicles (EVs) extracted under hypoxia (H‐EVs‐hypoxia), P‐DNA gel MNs can regulate tissue microenvironment, scavenge free radicals, and reduce inflammation. Notably, P‐DNA gel MNs integration system activates immune regulation and beneficial pathways, promoting neurogenesis and angiogenesis, reducing pathological pain, and achieving high‐quality healing. Overall, P‐DNA gel MNs system presents a fundamental mechanism for clinical transformation in treating diabetes wounds.

Multifunctional Hyaluronic Acid Microneedle Patch Embedded by Cerium/Zinc‐based Composites for Accelerating Diabetes Wound Healing

Abstract:Chronic nonhealing diabetic wounds are becoming increasingly severe, with high rates of mortality and disability, owing to the difficulty in wound healing caused by hyperglycemia, blocked angiogenesis, biofilm infection, and excessive oxidative stress. W e report a multicomponent enzyme‐responsive natural polymer, a hyaluronic acid (HA) microneedle, embedded in a cerium/zinc‐based nanomaterial (ZCO) for the treatment of diabetic wounds. ZCO‐HA can destroy the oxidation balance of bacteria, kill bacteria, and scavenge reactive oxygen species (ROS) to alleviate oxidative stress via the adjustable release of Zn2+ and Ce3+/4+. Additionally, ZCO‐HA exhibited good anti‐inflammatory activity through the nuclear factor kappa‐B (NF‐κB) pathway, which reduces the inflammatory state of macrophages and promotes cell proliferation, migration, and angiogenesis. In vitro experiments shows that ZCO‐HA accompanies mouse fibroblast migration, promoting human umbilical vein endothelial cell tube formation. In vivo studies in mice with streptozotocin‐induced (STZ)‐induced diabetes revealed that this microneedle accelerated wound healing without systemic toxicity. RNA transcriptome sequencing illustrated that the multicomponent HA microneedle accelerated wound healing in diabetes through cell migration and inhibited inflammatory reactions and oxidative damage in mice via the NF‐κB signaling pathway. Abstract:Chronic wounds affect healthcare systems and increase the risk of infection. Despite the clinical demand for improving healing, an effective and safe therapeutic approach is still lacking. Here, we present a novel spatio-temporal-varying adhesive microneedle (MN) patch encapsulated with mesenchymal stem cell-derived exosomes (MSC-exos) and antibacterial Ag nanoparticles (AgNPs) for improving wound healing. This MN patch is fabricated by replicating a predesigned flexible template. Consisting of porous methacrylate gelatin hydrogels, the MN tips can continuously deliver anti-inflammatory and pro-angiogenic MSC-exos to wound beds, which can accelerate healing by improving cell functions, remodeling blood vessels and restoring immune systems. Besides, the silk-fibroin-composed MN back enables the patch to stick to the skin well and fix at the wounded site, thus avoiding secondary bandage. By incorporating AgNPs in the MN back, the MN patch is endowed with the ability to efficiently resist microbial contamination and further benefits wound repair. Such a composite MN patch performs well in relieving inflammation, promoting angiogenesis, and suppressing wound-infecting bacteria during wound injury in diabetic rats, with no adverse effects observed. All the features make this multifunctional MN patch potentially valuable for clinical applications.

Abstract:A novel strategy is developed for fabricating a multifunctional silk fibroin methacryloyl hydrogel multifunctional microneedle (MN) patch, encapsulating Prussian blue nanozymes (PBNs) and vascular endothelial growth factor (VEGF) in the tips of MN patches, and encapsulating polymyxin in the base layer of the MN patch, which exhibits excellent biocompatibility, sustained drug‐release, pro‐angiogenesis, antioxidant, and antibacterial properties. Diabetic wound is one of the common complications in diabetic patients, which exhibits chronic, hard‐to‐heal characteristics. The healing process of wounds is impaired by several factors, including excessive oxidative stress, blocked angiogenesis, and bacterial infection. The therapeutic effects of traditional microneedle patches remain not satisfactory, due to their difficulty simultaneously targeting multiple targets to treat diabetic wounds. As such, there is an urgent need to develop a multifunctional microneedle (MN) patch for promoting the healing of diabetic wounds. A multifunctional MN patch with antioxidant, proangiogenesis, and antibacterial capacities was fabricated to target the pathogenesis of diabetic wounds. Silk fibroin methacryloyl, which has excellent biocompatibility, stable mechanical properties, and well processability, and is selected as the base material for multifunctional MN patches. Prussian blue nanozymes (PBNs) and vascular endothelial growth factor (VEGF) are encapsulated in tips of MN patches, Polymyxin is encapsulated in base layers of MN patches. Based on synergic properties of these components, multifunctional MN patches exhibit excellent biocompatibility, drug‐sustained release, proangiogenesis, antioxidant, and antibacterial properties. The developed multifunctional MN patches accelerate diabetic wound healing, providing a potential therapeutic approach.

Abstract:Diabetic wounds tend to develop into nonhealing wounds associated with the complex inflammatory microenvironment of uncontrollable bacterial infection, reactive oxygen species (ROS) accumulation, and chronic hypoxia. Damaged blood vessels hinder metabolic circulation, aggravating hypoxia, and ROS accumulation and further exacerbating the diabetic wound microenvironment. However, existing treatments with a single functionality have difficulty healing complicated diabetic wounds. Therefore, developing an integrative strategy to improve the hostility of the diabetic wound microenvironment is urgently needed. Herein, multifunctional genipin (GP)-crosslinked chitosan (CS)-based hydrogels decorated with the biomimetic metal-organic framework (MOF)-nanozymes and the natural antibacterial agent chlorogenic acid (CGA), which is named MOF/CGA@GP-CS (MCGC), are prepared. With catalase (CAT)-like activity, these dual-metal MOF-nanozymes are promising bioreactors for simultaneously alleviating ROS accumulation and hypoxia by converting elevated endogenous H2O2 into dissolved oxygen in diabetic wounds. In addition, the other component of natural polyphenolic CGA acts as a mild antibacterial agent, efficiently inhibiting wound infection and avoiding antibiotic resistance. Impressively, the MCGC hydrogels accelerate infected diabetic wound healing by eliminating oxidative stress, increasing oxygenation, and reversing bacterial infection in vivo. In this work, an effective strategy based on multifunctional hydrogel wound dressings is successfully developed and applied in diabetic wound management.

Abstract:Diabetic wounds pose a clinical challenge due to persistent inflammation, severe bacterial infections, inadequate vascularization, and pronounced oxidative stress. Current therapeutic modalities fail to provide satisfactory outcomes in managing these conditions, resulting in considerable patient distress. Two-dimensional nanomaterials (2DNMs), characterized by their unique nanosheet structures, expansive surface areas, and remarkable physicochemical properties, have garnered considerable attention for their potential in therapeutic applications. Emerging 2DNMs can be loaded with various pharmacological agents, including small molecules, metal ions, and liposomes. Moreover, they can be integrated with various biomaterials such as hydrogels, microneedles, and microspheres, thus demonstrating unprecedented advantages in expediting the healing process of diabetic wounds. Moreover, 2DNMs exhibit exceptional performance characteristics, including high biocompatibility, effective antimicrobial properties, optimal phototherapeutic effects, and enhanced electrostimulation capabilities. These properties enable them to modulate the wound microenvironment, leading to widespread application in tissue repair with remarkable outcomes. This review delineates several emerging 2DNMs, such as graphene and its derivatives, black phosphorus, MXenes, and transition metal dichalcogenides, in the context of diabetic wound repair. Furthermore, it elucidates the translational challenges and future perspectives of 2DNMs in wound healing treatments. Overall, 2DNMs present a highly promising strategy for ameliorating diabetic wounds, thus providing novel avenues for diagnostic and therapeutic strategies in diabetic wound management.
Guiding the Path to Healing: CuO2‐Laden Nanocomposite Membrane for Diabetic Wound Treatment

Abstract:Electrospun fibers with the n‐CuO2 NPs loaded PVP/PCL as the sheath and PCL as the core. When exposed to the wound's moist environment, PVP disintegrates to release the embedded n‐CuO2, to trigger the H2O2 self‐supplied chemodynamic therapy (CDT) for antibacterial, antiinflammation, and angiogenesis properties. Simultaneously, the dissolution of PVP also exposes nano‐grooved patterns for cell‐guiding function to accelerate skin regeneration. Diabetic chronic wounds pose significant clinical challenges due to their characteristic features of impaired extracellular matrix (ECM) function, diminished angiogenesis, chronic inflammation, and increased susceptibility to infection. To tackle these challenges and provide a comprehensive therapeutic approach for diabetic wounds, the first coaxial electrospun nanocomposite membrane is developed that incorporates multifunctional copper peroxide nanoparticles (n‐CuO2). The membrane's nanofiber possesses a unique "core/sheath" structure consisting of n‐CuO2+PVP (Polyvinylpyrrolidone)/PCL (Polycaprolactone) composite sheath and a PCL core. When exposed to the wound's moist environment, PVP within the sheath gradually disintegrates, releasing the embedded n‐CuO2. Under a weakly acidic microenvironment (typically diabetic and infected wounds), n‐CuO2 decomposes to release H2O2 and Cu2+ ions and subsequently produce ·OH through chemodynamic reactions. This enables the anti‐bacterial activity mediated by reactive oxygen species (ROS), suppressing the inflammation while enhancing angiogenesis. At the same time, the dissolution of PVP unveils unique nano‐grooved surface patterns on the nanofibers, providing desirable cell‐guiding function required for accelerated skin regeneration. Through meticulous material selection and design, this study pioneers the development of functional nanocomposites for multi‐modal wound therapy, which holds great promise in guiding the path to healing for diabetic wounds.
