Effects of Exercise Training on Myocardial Fatty Acid Metabolism in Rats with Depressed Cardiac Function Induced by Transient Ischemia

Abstract:The effects of exercise training on metabolic and functional recovery after myocardial transient ischemia were investigated in a rat model. Male Wistar Kyoto rats were subjected either to a 30-min left coronary artery occlusion followed by reperfusion or to a sham operation. At 4 weeks after operation, the rats were randomly assigned either to sedentary conditions or to exercise training for 6 weeks. In the ischemic rats, pinhole SPECT (single photon emission computed tomography) imaging with thallium-201 (201Tl) and 123I-(rho-iodophenyl)-3-R,S-methylpentadecanoic acid (BMIPP) showed a reduction of both myocardial perfusion and fatty acid metabolism in the risk zone of the left ventricle (LV). The LV was dilated and the ejection fraction was decreased after ischemic injury. The severity score showed a significant decrease on both 201Tl and BMIPP (201Tl, from 19.9+/-2.7 to 17.0+/-2.2, p<0.05; BMIPP, from 21.5+/-2.4 to 18.6+/-1.9, p<0.05) after exercise training in the ischemic trained rats, but did not change significantly in their sedentary counterparts. Plasma levels of free fatty acids normalized in the ischemic trained rats, but elevated in the ischemic sedentary rats (0.53+/-0.05 vs 0.73+/-0.06 mmol/L, p<0.05). Furthermore, the trained rats had a significant increase in LV stroke volume (0.25+/-0.02 vs 0.21+/-0.01 ml/beat, p<0.05) and adaptive cardiac hypertrophy. These findings demonstrate that adaptive improvements in myocardial perfusion, fatty-acid metabolism and LV function were induced by exercise training after transient ischemia.
Effect of farnesyltransferase inhibition on cardiac remodeling in spontaneously hypertensive rats

Abstract:Background Farnesyltransferase (FT), an essential enzyme at the downstream of mevalonate pathway, was reported to be upregulated in hypertrophic cardiomyocytes of spontaneously hypertensive rats (SHRs) compared with myocardium of Wistar-Kyoto rats (WKYs). This upregulation was accompanied with cardiac remodeling. This study was designed to determine whether FT inhibition can alter cardiac remodeling in SHRs. Methods Twelve-week-old SHRs were randomized to receive infusion of either NS or FTI-276 (307μg/kg/d i.v. each n=10). WKY rats served as normal controls (n=6). Echocardiography was performed before and after intervention. SHR hearts were perfused ex vivo for the evaluation of cardiac performance, collagen deposition and biochemical changes (activation of Ras, extracellular-signal regulated kinases/ERK1/2, procollagen type І/Ш, TGF-β1, connective tissue growth factor/CTGF, and bone morphogenetic protein-7/BMP-7 expression). Results FTI-276 intervention decreased interventricular septum wall thickness at end- diastole (IVSd) and relative wall thickness (RWT) of SHRs (P<0.05). Three week intervention with FTI-276 attenuated hydroxyproline content (P<0.05), collagen deposition (P<0.01), Ras activation, ERK1/2 phosphorylation (P<0.01) and mRNA expression of procollagen type I, TGF-β1 and CTGF and elevated mRNA expression of BMP-7 (P<0.05) in left ventricle of SHRs. Conclusion The present study indicated that FT inhibition could attenuate myocardial fibrosis and partly improve cardiac remodeling in SHRs. The beneficial effects might be mediated through suppression of the activation of Ras and ERK1/2 phosphorylation pathway. The enhanced mRNA expression of BMP-7 with inhibition of TGF-β1 and CTGF mRNA expression might be an important mechanism.
Effects of moderate-continuous and high-intensity interval aerobic training on cardiac function of spontaneously hypertensive rats

Abstract:Experimental Biology and Medicine, Ahead of Print. The aim of this study was to verify the effects of moderate-intensity continuous (MICT) and high-intensity interval (HIIT) aerobic training on cardiac morphology and function and the mechanical properties of single cardiomyocytes in spontaneously hypertensive rats (SHR) in the compensated phase of hypertension. Sixteen-week-old male SHR and normotensive Wistar (WIS) rats were allocated to six groups of six animals each: SHR CONT or WIS CONT (control); SHR MICT or WIS MICT (underwent MICT, 30 min/day, five days per week for eight weeks); and SHR HIIT or WIS HIIT (underwent HIIT, 30 min/day, five days per week for eight weeks). Total exercise time until fatigue and maximum running speed were determined using a maximal running test before and after the experimental period. Systolic (SAP), diastolic (DAP), and mean (MAP) blood pressures were measured using tail plethysmography before and after the experimental period. Echocardiographic evaluations were performed at the end of the experimental period. The rats were euthanized after in vivo assessments, and left ventricular myocytes were isolated to evaluate global intracellular Ca2+ transient ([Ca2+]i) and contractile function. Cellular measurements were performed at basal temperature (~37°C) at 3, 5, and 7 Hz. The results showed that both training programs increased total exercise time until fatigue and, consequently, maximum running speed. In hypertensive rats, MICT decreased SAP, DAP, MAP, interventricular septal thickness during systole and diastole, and the contraction amplitude at 5 Hz. HIIT increased heart weight and left ventricular wall thickness during systole and diastole and reduced SAP, MAP, and the time to peak [Ca2+]i at all pacing frequencies. In conclusion, both aerobic training protocols promoted beneficial adaptations to cardiac morphology, function, and mechanical properties of single cardiomyocytes in SHR.

Abstract:We aimed to study the effects and underlying mechanism of different intensities of continuous training (CT) on vascular inflammation and oxidative stress in spontaneously hypertensive rats (SHR). Rats were divided into five groups (n = 12): Wistar-Kyoto rats sedentary group (WKY-S), sedentary group (SHR-S), low-intensity CT group (SHR-L), medium-intensity CT group (SHR-M) and high-intensity CT group (SHR-H). Changes in body mass, heart rate and blood pressure were recorded. The rats were euthanized after 14 weeks, and blood and vascular tissue samples were collected. Haematoxylin and Eosin staining was used to observe the aortic morphology, and Western blot was used to detect the expression of mesenteric artery proteins. After CT, the mean arterial pressures improved in SHR-L and SHR-M and increased in SHR-H compared with those in SHR-S. Vascular inflammation and oxidative stress levels significantly subsided in SHR-L and SHR-M (p < 0.05), whereas in SHR-H, only vascular inflammation significantly subsided (p < 0.05), and oxidative stress remained unchanged (p > 0.05). AMPK and SIRT1/3 expressions in SHR-L and SHR-M were significantly up-regulated than those in SHR-S (p < 0.05). These results indicated that low- and medium-intensity CT can effectively reduce the inflammatory response and oxidative stress of SHR vascular tissue, and high-intensity CT can improve vascular tissue inflammation but not oxidative stress.
Low‐intensity aerobic exercise improves cardiac remodelling of adult spontaneously hypertensive rats

Abstract:<p>We evaluated the influence of aerobic training on cardiac remodeling in untreated spontaneously hypertensive rats (SHR). Four experimental groups were used: sedentary (W‐SED, n=27) and trained (WEX, n=31) normotensive Wistar rats, and sedentary (SHR‐SED, n=27) and exercised (SHR‐EX, n=32) hypertensive rats. At 13 months old, trained groups underwent treadmill exercise five days a week for four months. Statistical analysis: ANOVA or Kruskal‐Wallis. Exercised groups had higher physical capacity. Hypertensive groups presented left ventricular (LV) concentric hypertrophy with impaired function. Left atrium diameter, LV posterior wall thickness and relative thickness, and isovolumetric relaxation time were lower in SHR‐EX than SHR‐SED. Interstitial collagen fraction and Type I‐Type III collagen ratio were higher in SHR‐SED than W‐SED. In SHR‐EX these parameters had intermediate values between W‐EX and SHRSED with no differences between either group. Myocardial matrix metalloproteinase‐2 activity, evaluated by zymography, was higher in SHR‐SED than W‐SED and SHR‐EX. TIMP‐2 was higher in hypertensive than normotensive groups. In conclusion, low intensity aerobic exercise reduces left atrium dimension and LV posterior wall thickness, and improves functional capacity, diastolic function, and metalloproteinase‐2 activity in adult SHR.</p>

Abstract:AIMS: Regular exercise as an effective non-pharmacological antihypertensive therapy is beneficial for prevention and control of hypertension, but the central mechanisms are unclear. In this study, we hypothesized that chronic exercise training (ExT) delays the progression of hypertension and attenuates cardiac hypertrophy by up-regulating anti-inflammatory cytokines, reducing pro-inflammatory cytokines (PICs) and restoring the neurotransmitters balance in the hypothalamic paraventricular nucleus (PVN) in young spontaneously hypertensive rats (SHR). In addition, we also investigated the involvement of nuclear factor-κB (NF-κB) p65 and NAD(P)H oxidase in exercise-induced effects.METHODS AND RESULTS: Moderate-intensity ExT was administrated to young normotensive Wistar-Kyoto (WKY) and SHR rats for 16 weeks. SHR rats had a significant increase in mean arterial pressure and cardiac hypertrophy. SHR rats also had higher levels of glutamate, norepinephrine (NE), phosphorylated IKKβ, NF-κB p65 activity, NAD(P)H oxidase subunit gp91(phox), PICs and the monocyte chemokine protein-1 (MCP-1), and lower levels of gamma-aminobutyric acid (GABA) and interleukin-10 (IL-10) in the PVN. These SHR rats also exhibited higher renal sympathetic nerve activity (RSNA), and higher plasma levels of PICs, and lower plasma IL-10. However, ExT ameliorates all these changes in SHR rats.CONCLUSION: These findings suggest that there are the imbalances between excitatory and inhibitory neurotransmitters and between pro- and anti-inflammatory cytokines in the PVN of SHR rats, which at least partly contributing to sympathoexcitation, hypertension and cardiac hypertrophy; chronic exercise training attenuates hypertension and cardiac hypertrophy by restoring the balances between excitatory and inhibitory neurotransmitters and between pro- and anti-inflammatory cytokines in the PVN; NF-κB and oxidative stress in the PVN may be involved in these exercise-induced effects.

Prevention of myocardial fibrosis by using nitrendipine to the spontaneously hypertensive rats

Abstract:Aim: To determine the effects of nitrendipine on blood pressure, left ventricular hypertrophy (LVH) and myocardial fibrosis of both ventricles of spontaneously hypertensive rats (SHR). Method: Antihypertensive treatment with calcium channel blocker nitrendipine (30 mg �� kg-1/d) was given to 14-week old male SHRs (n = 8) for 12 weeks with age matched SHRs and Wistar-Kyoto rats (WKY) (n = 8) received no treatment as controls. Systolic blood pressure was measured by tail-cuff method. LVH was defined according to the ratio of left to right ventricular weights or to body weights. Myocardial fibrosis was evaluated by collagen content and concentration mg/mg dry wt. in both ventricles. Hydroxyprolin content was measured spectrophotometrically by its reaction with Ehrich's reagent (Stegemann method). And collagen content was calculate by multiplying the hydroxyprolin content with the constant 8.2. Results: Nitrendipine significantly decreased systolic blood pressure in SHRs (P < 0.05) although the value remained significantly higher than that of WKY controls. The ratio of left ventricular weight to body weight was significantly decreased in nitrendipine treated compared with untreated SHRs (P < 0.05). Right ventricular weight of the experimental group showed no significant change versus that of the controls. Collagen content and concentration of the left ventricle and right ventricle were lower in the nitrendipine treated group compared with the untreated groups. Conclusions: Nitrendipine could attenuate hypertension and decrease collagen content and concentration of both ventricles as well as left ventricular mass in SHRs. So nitrendipine might have the potential to improve the myocardial pathology in essential hypertension.
Chronic antioxidant treatment improves sympathetic functions and β-adrenergic pathway in the spontaneously hypertensive rats

Abstract:BACKGROUND: Correlations have been found between oxidative stress and hypertension.OBJECTIVE: To determine whether antioxidants can normalize sympathetic dysfunction at pre- or postsynaptic levels in spontaneously hypertensive rats (SHRs).METHODS AND RESULTS: Untreated SHRs and Wistar-Kyoto (WKY) rats were compared with rats treated with melatonin (30 mg/kg per day) or N-acetylcysteine (NAC) (4 g/kg per day) given in drinking water for 4 weeks. At the presynaptic level, SHRs had greater plasma noradrenaline concentrations (P <0.01) and an enhanced release of [3H]noradrenaline from isolated atria (P <0.001). At the postsynaptic level, they exhibited an increased proportion of beta2-adrenoceptors in the heart (P <0.001)and a decrease in the chronotropic and mean arterial pressure (MAP) responses to isoproterenol (P <0.001). Melatonin and NAC decreased MAP (P <0.001) and heart rate (P <0.05), and restored the plasma noradrenaline concentrations (P<0.01 and P <0.001, respectively), the chronotropic response to isoproterenol (P <0.05) and the proportions of beta(1)/beta(2)-adrenoceptors in the heart (P <0.05) in SHRs to the levels found in WKY rats. The same treatments decreased the release of [3H]noradrenaline from isolated atria (P <0.05), and melatonin slightly improved the relaxation in the aorta in SHRs only (P <0.05). Plasma concentrations of adrenaline, the isoproterenol-induced relaxation in mesenteric arteries, the total density and affinity of beta-adrenoceptors in the heart, and the adenylate cyclase reactivity of cardiac membranes to isoproterenol, forskolin, sodium fluoride and guanylylimidophosphate were not altered by the treatments.CONCLUSION: These results suggest that NAC and melatonin decreased the MAP and heart rate and improved the chronotropic response to isoproterenol in SHRs, in association with an inhibition of sympathetic activity and the restoration of cardiac beta-adrenoceptor function.

Abstract:The activity of the ubiquitin proteasome system (UPS) and the level of oxidative stress contribute to the transition from compensated cardiac hypertrophy to heart failure in hypertension. Moreover, aerobic exercise training (AET) is an important therapy for the treatment of hypertension, but its effects on the UPS are not completely known. The aim of this study was to evaluate the effect of AET on UPS’s activity and oxidative stress level in heart of spontaneously hypertensive rats (SHR). A total of 53 Wistar and SHR rats were randomly divided into sedentary and trained groups. The AET protocol was 5×/week in treadmill for 13 weeks. Exercise tolerance test, non-invasive blood pressure measurement, echocardiographic analyses, and left ventricle hemodynamics were performed during experimental period. The expression of ubiquitinated proteins, 4-hydroxynonenal (4-HNE), Akt, phospho-Aktser473, GSK3β, and phospho-GSK3βser9 were analyzed by western blotting. The evaluation of lipid hydroperoxide concentration was performed using the xylenol orange method, and the proteasomal chymotrypsin-like activity was measured by fluorimetric assay. Sedentary hypertensive group presented cardiac hypertrophy, unaltered expression of total Akt, phospho-Akt, total GSK3β and phospho-GSK3β, UPS hyperactivity, increased lipid hydroperoxidation as well as elevated expression of 4-HNE but normal cardiac function. In contrast, AET significantly increased exercise tolerance, decreased resting systolic blood pressure and heart rate in hypertensive animals. In addition, the AET increased phospho-Akt expression, decreased phospho-GSK3β, and did not alter the expression of total Akt, total GSK3β, and ubiquitinated proteins, however, significantly attenuated 4-HNE levels, lipid hydroperoxidation, and UPS’s activity toward normotensive group levels. Our results provide evidence for the main effect of AET on attenuating cardiac ubiquitin proteasome hyperactivity and oxidative stress in SHR rats.

Ceramic Dust as Construction Material in Rigid Pavement

Abstract:Ceramic dust is produced as waste from ceramic bricks, roof and floor tiles and stoneware waste industries. Concrete (M35) was made by replacing % (up to 30%) of cement (OPC 53) grade with ceramic dust (passing 75µm) shows good workability, compressive strength, split-tensile strength, flexural strength and elastic modulus. In this experimental investigation, concrete specimens were tested at different age for different mechanical properties. The results show that with water – cement ratio (0.46), core compressive strength increase by 3.9% to 5.6% by replacing 20% cement content with ceramic dust. It was observed that no significant change in flexural strength and split-tensile strength when compared to the conventional concrete.
Simultaneous targeting of oxidative stress and fibrosis reverses cardiomyopathy‐induced ventricular remodelling and dysfunction

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> Background and purpose</h3><p>Oxidative stress and fibrosis are hallmarks of cardiomyopathy‐induced heart failure, yet are not effectively targeted by current frontline therapies. In this study, the therapeutic effects of the anti‐oxidant, N‐Acetylcysteine (NAC) were compared, and combined, with the acute heart failure drug with established anti‐fibrotic effects, serelaxin (RLX), in a murine model of cardiomyopathy.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Experimental approach</h3><p>Adult male 129sv mice were subjected to repeated isoproterenol (ISO; 25mg·kg<sup>‐1</sup>)‐induced cardiac injury for five consecutive days, then left to undergo fibrotic healing until day‐14. Sub‐groups of ISO‐injured mice (n=5‐7/group) were treated with either RLX (0.5 mg·kg<sup>‐1</sup> ·day<sup>‐1</sup>), NAC (25 mg·kg<sup>‐1</sup> ·day<sup>‐1</sup>) or both combined; all given subcutaneously via osmotic minipumps from days‐7‐14. Control mice received repeated saline instead of ISO. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Key results</h3><p>ISO‐injured mice underwent significantly increased LV inflammation (by ~5‐fold), oxidative stress (~1‐2.5‐fold), cardiomyocyte hypertrophy (~25%), cardiac remodelling, fibrosis (~2‐2.5‐fold) and dysfunction by day‐14 post‐injury. NAC alone abrogated the cardiomyopathy‐induced increase in LV superoxide levels to a greater extent than RLX. Additionally, either treatment alone only partially reduced several measures of LV inflammation, remodelling and fibrosis. In comparison, the combined effects of RLX and NAC completely ameliorated the cardiomyopathy‐induced LV macrophage infiltration, remodelling, fibrosis and cardiomyocyte size, to a greater extent than either treatment alone after 7 days. The combination therapy also significantly restored the ISO‐induced reduction in LV function, without affecting systolic blood pressure.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Conclusions and implications</h3><p>These findings demonstrated that the simultaneous targeting of oxidative stress and fibrosis is key to treating the pathophysiology and dysfunction induced by cardiomyopathy.</p></section>

Abstract:Background: Cardiovascular Diseases are one of the main global public health problems, with increasing mortality due to population aging and Hypertension (HTN). HTN is a risk factor for other diseases, such as aortic stenosis, renal dysfunction, and cardiac hypertrophy (CH). The hemodynamic overload caused by HTN affects the heart and kidneys, but the exact mechanisms of these alterations are not fully understood. Experimental models help study these mechanisms; however, a well-established model of CH with cardiorenal dysfunction does not yet exist. Objective: to evaluate whether the combination of cardiac hypertrophy, induced by transverse aortic constriction (TAC), and renal injury, induced by soluble sodium oxalate (OXA), can be a new model of CH with cardiorenal dysfunction. Methods: Male C57BL mice were randomized as follows: SHAM, receiving saline or OXA solution, and TAC, receiving saline or OXA solution. Echocardiography, direct blood pressure (BP) recording, metabolic cage, histology were performed to evaluate cardiac and renal function, hemodynamics, autonomic modulation, and baroreflex. Results: The TAC+OXA group showed the highest systolic BP(SBP) (SBP: SHAM: 122±5.24 vs. TAC+OXA: 160±5.24 mmHg, p<0.01), increased double product (DP) (DP: SHAM: 64.65±4.76 mmHg/bpm vs. TAC: 89.53±5.85 mmHg/bpm, p=0.02), significant reduction in autonomic modulation (RMSSD: OXA: 12.5±1.8 vs. TAC+OXA: 2.8±0.2 ms, p<0.01) and reflex tachycardia, eccentric hypertrophy, and worse renal function. The TAC group showed eccentric hypertrophy, increased BP variability (SD-SBP: SHAM: 2.97±0.07 mmHg vs. TAC: 5.63±0.57 mmHg, p≤0.01), diastolic dysfunction, and dilated left ventricle in diastole. The OXA group showed increased autonomic modulation and heart rate variability, renal damage characterized by inflammation, proximal tubule damage, and interstitial fibrosis when associated with TAC. All treated groups showed a significant reduction in reflex tachycardia. Conclusion: Transverse aortic constriction combined with OXA gavage is a model of CH with renal dysfunction, as the combination caused more significant renal damage, systemic hypertension, decreased autonomic modulation and baroreflex sensitivity, and eccentric hypertrophy with subsequent LV diastolic dysfunction.

Abstract:We compared the influence of aerobic and resistance exercise on cardiac remodelling, physical capacity and skeletal muscle oxidative stress in rats with MI‐induced heart failure. Three months after MI induction, Wistar rats were divided into four groups: Sham; sedentary MI (S‐MI); aerobic exercised MI (A‐MI); and resistance exercised MI (R‐MI). Exercised rats trained three times a week for 12 weeks on a treadmill or ladder. Statistical analysis was performed by ANOVA or Kruskal‐Wallis test. Functional aerobic capacity was greater in A‐MI and strength gain higher in R‐MI. Echocardiographic parameters did not differ between infarct groups. Reactive oxygen species production, evaluated by fluorescence, was higher in S‐MI than Sham, and lipid hydroperoxide concentration was lower in A‐MI than the other groups. Glutathione peroxidase activity was higher in A‐MI than S‐MI and R‐MI. Superoxide dismutase was lower in S‐MI than Sham and R‐MI. Gastrocnemius cross‐sectional area, satellite cell activation and expression of the ubiquitin‐proteasome system proteins did not differ between groups. In conclusion, aerobic exercise and resistance exercise improve functional capacity and maximum load carrying, respectively, without changing cardiac remodelling in infarcted rats. In the gastrocnemius, infarction increases oxidative stress and changes antioxidant enzyme activities. Aerobic exercise reduces oxidative stress and attenuates superoxide dismutase and glutathione peroxidase changes.

Abstract:AIM The cardio-ankle vascular index (CAVI) reflects overall arterial stiffness from the aorta to the ankle, independent of blood pressure. Cerebral small vessel diseases (SVDs), including white matter lesions (WML), silent lacunar infarction (SLI) and cerebral microbleeds (CMB), are considered to increase the risk of stroke and cognitive impairment further. We aimed to investigate the association of cerebral SVD with CAVI in asymptomatic young and middle-aged subjects. METHODS Asymptomatic Korean individuals aged 30 to 59 years old without neurologic disease who had CAVI and brain magnetic resonance imaging (MRI) were evaluated retrospectively. RESULTS Among 484 subjects, cerebral SVDs (advanced WML, SLI and CMB) were found in 20 (4.1%). Subjects with SVDs tended to be older and to have higher systolic blood pressure (SBP) and higher CAVI. From multivariate regression analysis, including pulse pressure (PP) or SBP, CAVI showed a significant association with SVD [adjusted OR (95% CI): 1.889 (1.094-3.263), p= 0.002 and 1.793 (1.020-3.153), p= 0.043, for PP and SBP, respectively]. When CAVI was assessed by quartiles, the highest quartile of CAVI (CAVI >7.65) showed a significant association with SVD, after adjustment for PP [adjusted OR (95% CI): 2.766 (1.115-6.866), p= 0.028]. CONCLUSION In young and middle-aged subjects, cerebral SVD was significantly associated with arterial stiffness measured by CAVI after adjusting for PP or SBP.
Research on Improvement of Left Ventricular Structural Remodeling after Myocardial Infarction in Rats by N-Acetylcysteine

Abstract:Objective: To investigate the potential effects and possible mechanisms of N-acetylcysteine(NAC) on left ventricular structural remodeling after myocardial infarction in rats.Methods: Total 96 Sprague-Dawley were randomly divided into blank control group(BC group,n=6),the rest 90 underwent myocardial infarction model were randomly divided into NAC group(n=34) and NS group(n=32) 24 h after survival.Rats in NAC group were intraperitoneal injected with NAC(0.2 mg.kg-1.2d-1) for 4 weeks,while rats in NS group were given the same volume of normal saline as control.After 4 weeks,the systolic left ventricular posterior wall thickness(LVPW;s),the diastolic left ventricular posterior wall thickness(LVPW;d),systolic Interventricular septal thickness(IVS;s),diastolic Interventricular septal thickness(IVS;d),LV Mass Corrected and LV Mass were measured by Ultrasound Cardiography,the level of GSH was measured by GSH/GSSG kit,the morality was also calculated.Results: In comparison with BC group,GSH level in NS group was significantly lower(P0.05),LVPW;d,LVPW;s,IVS;d,IVS;s,LV Mass Corrected and LV Mass appeared much thinner(P0.05),the survival rate decreased in both NS group and NAC group.In comparison with NS group,GSH level was significantly higher in NAC group(P0.05),LVPW;d,IVS;d,IVS;s,LV Mass Corrected and LV Mass appear much thicker(P0.05),but no significant difference was observed on the survival rate(P0.05).Conclusions: NAC was capable to inhibit the left ventricle remodeling from myocardial infarction in rats at least in part through up-regulation of GSH,but it didn't improved the survival after myocardial infarction.

Abstract:Although exercise derived activation of Nrf2 signaling augments myocardial antioxidant signaling, the molecular mechanisms underlying the benefits of moderate exercise training (MET) in the heart remain elusive. Here we hypothesized that exercise training stabilizes Nrf2-dependent antioxidant signaling, which then protects the myocardium from isoproterenol-induced damage. The present study assessed the effects of 6 weeks of MET on the Nrf2/antioxidant function, glutathione redox state, and injury in the myocardium of C57/BL6J mice that received isoproterenol (ISO; 50 mg/kg/day for 7 days). ISO administration significantly reduced the Nrf2 promoter activity (<i>p</i> < 0.05) and downregulated the expression of cardiac antioxidant genes (<i>Gclc, Nqo1, Cat, Gsr, and Gst-</i>μ) in the untrained (UNT) mice. Furthermore, increased oxidative stress with severe myocardial injury was evident in UNT+ISO when compared to UNT mice receiving PBS under basal condition. Of note, MET stabilized the Nrf2-promoter activity and upheld the expression of Nrf2-dependent antioxidant genes in animals receiving ISO, and attenuated the oxidative stress-induced myocardial damage. Echocardiography analysis revealed impaired diastolic ventricular function in UNT+ISO mice, but this was partially normalized in the MET animals. Interestingly, while there was a marginal reduction in ubiquitinated proteins in MET mice that received ISO, the pathological signs were attenuated along with near normal cardiac function in response to exercise training. Thus, moderate intensity exercise training conferred protection against ISO-induced myocardial injury by augmentation of Nrf2-antioxidant signaling and attenuation of isoproterenol-induced oxidative stress.

Abstract:Objective: The purpose of this study was to explore the effect of blood flow-restricted low resistance training on microvascular rarefaction in the myocardium of spontaneously hypertensive rats (SHRs). Methods: Four-week-old male SHRs were randomly divided into the following groups: Wistar-Kyoto (WKY), SHR control (SHR-SED), high-intensity resistance training (HIRT), low-intensity resistance training (LIRT), and blood flow-restricted low resistance training (BFRT). The exercise groups began to receive exercise intervention for 8 weeks at the age of 7 weeks. Blood pressure (BP), heart rate (HR), cardiac function, capillary density, and Vascular endothelial growth factor -Phosphatidylinositol 3-kinase-Protein kinase B-Endothelial nitric oxide synthetase (VEGF-Pi3k-Akt-eNOS) were assessed. Results: 1) BP and HR of BFRT decreased significantly, Ejection fraction (EF) and Fraction shortening (FS) increased, and the effect of BFRT on lowering BP and HR was better than that of other groups ( p < 0.05); 2) The expression of VEGF, VEGFR2, p-VEGFR2, Pi3k, Akt, p-Akt, eNOS and p-eNOS in the myocardium of the BFRT was significantly upregulated, and eNOS expression was significantly higher than other groups ( p < 0 05); 3) the expression of VEGF in the blood of the BFRT was significantly upregulated, higher than SHR-SED, lower than HIRT ( p < 0.05), and there was no significant difference between BFRT and LIRT( p > 0.05); 4) the capillary density in the myocardium of BFRT was significantly higher than other exercise groups ( p < 0 05). Conclusion: Blood flow-restricted low resistance training can activate the VEGF-Pi3k-Akt-eNOS pathway, upregulate the expression of VEGF in blood, improve microvascular rarefaction, and promote myocardial microvascular circulation, thereby improving cardiac function and lowering blood pressure, achieving the preventive effect of early hypertension.

Abstract:The aim of the present study was to evaluate the effect of exercise training (ET) and l-arginine on oxidative stress and ventricular function in rat with myocardial infarction (MI). Four weeks after the surgical procedures, 40 Wistar male rats were randomized to the following groups: MI-sedentary (Sed); MI-exercise (Ex); MI-sedentary + l-arginine (Sed + LA); and MI-exercise + l-arginine (Ex + LA); the rats were subjected to aerobic training in the form of treadmill running. Rats in the l-arginine-treated groups drank water containing 4 % l-arginine. Before and after the training program, all subjects underwent resting echocardiography. Catalase (CAT) glutathione peroxidase (GPx), malondialdehyde (MDA) and myeloperoxidase (MPO) were measured. Cardiac output, stroke volume and fractional shortening in Ex and Ex + LA groups significantly increased in comparison with the Sed group. Cardiac systolic function indices in Ex + LA group were significantly greater than Ex group. Also, GPx activity and MDA, respectively, increased and decreased in response to ET, but no change was observed in MPO and CAT. These results suggest that ET increased LV function by decreasing oxidative stress and increasing antioxidant defense system in rats with MI. In addition in response to training, l-arginine appears to have additive effect on cardiac function, but have no effect on oxidative stress indices.

Towards Adaptive and Composable Services

Abstract:In complex distributed environments, it is important to achieve high degree of machine-to-machine interaction, not only on a functional, but on a semantic level. Therefore, we propose the NOMADS (Networks of Mobile Adaptive Dependable Systems) Republic, a vision of the future community open to all interested hardware and software entities, and based on adaptive and, if necessary, composable services. One needs very little to become citizen of this Republic: one needs to know how to communicate and to discover, provide and/or use NOMADS services, in an infrastructure that guarantees three basic properties: mobility, adaptivity and dependability. We concentrate on NOMADS services which could potentially provide an unprecedented level of highly adaptive and semantic collaboration. The idea is to hide the complexity of underlying infrastructure from the users, showing only the services they can invoke. This approach promises seamless interoperation. We provide a way to discover not only what services can collaborate with each other on the functional basis, but rather on the semantic basis, meaning which service compositions can produce useful results. This way we are able to offer new services not originally programmed.
Protective effects of the R-(+)-thioctic acid treatment: possible anti-inflammatory activity on heart of hypertensive rats

Abstract:Background: In cardiovascular disease, high blood pressure is associated with oxidative stress, promoting endothelial dysfunction, vascular remodeling, and inflammation. Clinical trials are discordant that the most effective treatment in the management of hypertension seems to be the administration of anti-hypertensive drugs with antioxidant properties. The study aims to evaluate the effects of the eutomer of thioctic acid on oxidative stress and inflammation in the heart of spontaneously hypertensive rats compared to normotensive Wistar Kyoto rats. Methods: To study the oxidative status, the malondialdehyde and 4-hydroxynonenal concentration, protein oxidation were measured in the heart. Morphological analysis were performed. Immunohistochemistry and Western blot were done for alpha-smooth muscle actin and transforming growth factor beta to assess fibrosis; cytokines and nuclear factor kappaB to assess inflammatory processes. Results: Spontaneously hypertensive rats were characterized by hypertension with increased malondialdehyde levels in the heart. OxyBlot in the heart of spontaneously hypertensive rats showed an increase in proteins' oxidative status. Cardiomyocyte hypertrophy and fibrosis in the ventricles were associated with an increased expression of alpha-smooth muscle actin and pro-inflammatory cytokines, reduced by the eutomer of thioctic acid supplementation. Conclusions: Based on this evidence, eutomer of thioctic acid could represent an appropriate antioxidant molecule to reduce oxidative stress and prevent inflammatory processes on the cardiomyocytes and cardiac vascular endothelium.
