Abstract:Plateau lakes characterized by salinization and eutrophication are essential aquatic ecosystems. A myriad of microorganisms serve as crucial biological resources in plateau lakes and drive the elemental cycles of these ecosystems. Currently, there is a paucity of knowledge regarding the impacts of salinization and eutrophication dynamics on the microbiota in plateau lakes. Here, high-throughput sequencing of the 16S ribosomal RNA genes (V4 region) was used to characterize microbial community structure and assembly in plateau lakes with different salinities and trophic levels. Water samples were collected at 191 sites across 24 lakes on the Qinghai-Tibet and Inner Mongolia Plateaus in northern China. The results showed that high salinity considerably reduced microbial alpha-diversity and niche breadth while increasing within-group similarity among various lake types. High salinity additionally decreased the complexity of microbial networks and enhanced network robustness. The assembly of microbial communities was primarily governed by deterministic processes in high-salinity and eutrophic low-salinity lakes. At decreased salinity, trophic level played a leading role in shaping microbial community structure, and the ecological processes shifted from deterministic processes driven by high salinity to eutrophication-driven deterministic processes. The biomarkers also varied from taxa adapted to high-salinity environments (e.g., Nanoarchaeaeota , Rhodothermia ) to those suited for living in freshwater and low-salinity habitats (e.g., Alphaproteobacteria , Actinobacteria ). In the case of eutrophication, Actinobacteria , Chloroflexi , and Cyanobacteria became the dominant taxa. Our findings indicate that decreased salinity enables trophic level to play an enhanced role in shaping microbial community structure and assembly in plateau lakes. This study enriches our knowledge about the ecological impacts of salinization and eutrophication in plateau lakes.

Abstract:Microorganisms in lakeshore zones are essential for pollution interception and biodiversity maintenance. However, the biogeographic patterns of bacterioplankton communities in lakeshore zones and the mechanisms that driving them are poorly understood. We analyzed the 16 S rRNA gene sequences of particle-associated (PA) and free-living (FL) bacterioplankton communities in the lakeshore zones of 14 alpine lakes in two seasons on Qinghai-Tibet Plateau to investigate the bacterial diversity, composition and assembly processes. Our results revealed that PA and FL bacterioplankton communities were driven by both seasonality and salinity in the lakeshores on Qinghai-Tibet Plateau. Compared to FL bacterioplankton, PA bacterioplankton communities were more susceptible to seasonality than spatial salinity. FL bacterioplankton communities were more salinity constrained than the PA counterpart. Besides, the Stegen null model analyses have validated a quantitative bias on stochastic processes at different spatial scales. At a regional scale, stochasticity was the predominant assembly process in both PA and FL bacterioplankton. While at a subregional scale, dispersal limitation was the main contributor of stochastic processes for PA bacterioplankton in summer and heterogeneous selection was the dominant deterministic processes in winter, whereas the community assembly of FL bacterioplankton was more stochastic processes (i.e., dispersal limitation) dominated in the freshwater type but deterministic process (i.e., heterogeneous selection) increased with increasing salinity. Our study provides new insights into both significant spatiotemporal patterns and distinct assembly processes of PA and FL bacterioplankton in alpine lakeshores on the northeastern Qinghai-Tibet Plateau.

Abstract:Abstract Apocytochrome P-450cam in a reconstitutable form was prepared from the purified native holoenzyme by acid-acetone treatment at -20°. The prosthetic group is ferriprotoporphyrin IX, identified by the pyridine hemochromogen spectrum and by co-chromatography of authentic and isolated samples. The apocytochrome is not protected by the substrate and is less stable at -20°. The catalytically active holocytochrome P-450cam has been prepared from the apoenzyme in greater than 66% yield via P-420 on addition of equimolar heme in 4% methanol-phosphate buffer, pH 7.0, followed by a cysteine-glycerol treatment.
Bacterial and Microeukaryotic Community Compositions and Their Assembly Processes in Lakes on the Eastern Qinghai-Tibet Plateau.

Abstract:Bacterial and microeukaryotic community compositions and their assembly processes have remained challenging and remained unclear in lake ecosystems on the Qinghai-Tibet Plateau (QTP). We revealed the diversity and community compositions, driving factors, ecological assembly processes, and co-occurrence networks of bacterial and microeukaryotic communities in water bodies of the eight lake ecosystems across the Eastern QTP. The results demonstrated that the predominant bacteria in most samples were Proteobacteria, with an average relative abundance of 41.78%, whereas the most abundant of microeukaryotes differed among the sample sites. The redundancy analysis revealed that latitude and pH were the most important driving factors in shaping the bacterial and microeukaryotic community compositions. Homogeneous selection (56.40%) was the dominant process in assembling the bacterial communities, whereas dispersal limitation (67.24%) was the major process in governing the microeukaryotic communities. Furthermore, dissolved organic carbon and salinity were the major factors mediating the balance of deterministic and stochastic assembly processes in the bacterial and microeukaryotic communities. Both the bacterial and microeukaryotic community co-occurrence networks exhibited topological features of modularity and non-random topological features. The results offer insights into the mechanisms underpinning bacterial and microeukaryotic diversities and communities in the lake ecosystems on the QTP.

Abstract:Microbes serve as important components of ecosystem services and biogeochemical processes in plateau lakes. However, the features of microbiota assembly, abundance and diversity in plateau lakes remain unclear. The microbial communities in surface water from nine plateau lakes in Yunnan Plateau, southwestern China, in the dry and rainy seasons were explored using 16S rRNA gene and 18S rRNA gene sequencing. The results showed that the bacterial community compositions were homogeneous while those of micro-eukaryotes were heterogeneous in plateau lakes. In both seasons, the bacterial phyla of Proteobacteria and Actinobacteriota predominated in oligotrophic lakes. The mesotrophic lakes were dominated by Proteobacteria, Actinobacteriota, Bacteroidota and Cyanobacteria. The eutrophic lakes were mainly dominated by Proteobacteria, Actinobacteriota and Cyanobacteria. The phylum SAR_k_norank had the major micro-eukaryotes in these plateau lakes. The alpha-diversity of bacteria declined in the rainy season, while that of micro-eukaryotes varied from lake to lake. The drivers of microbiotic community assembly in the dry season were identified as nutrient level-related factors. In the rainy season, however, the microbiota in oligotrophic lakes were related to nutrient levels. Microbial communities were driven by precipitation in mesotrophic and eutrophic lakes with large water volumes, while those in lakes with small water volumes were regulated by nutrient level-related factors. Our findings pose first and unique insights into the microbiota of the nine plateau lakes in Yunnan Plateau, providing important ecological knowledge for these lakes with different characteristics.

Abstract:Oligosaline lakes in arid and semi-arid regions play a crucial role in providing essential water resources for local populations. However, limited research exists on the impact of the environment on bacterial community structure in these lakes, co-occurrence patterns and the mechanisms governing bacterial community assembly. This study aims to address this knowledge gap by examining samples collected from five areas of Lake Bosten over four seasons. Using the 16S rRNA gene sequencing method, we identified a total of 510 to 1,005 operational taxonomic units (OTUs) belonging to 37 phyla and 359 genera in Lake Bosten. The major bacterial phyla were Proteobacteria (46.5%), Actinobacteria (25.9%), Bacteroidetes (13.2%), and Cyanobacteria (5.7%), while the major genera were hgcI_clade (12.9%), Limnohabitans (6.2%), and Polynucleobacter (4.7%). Water temperature emerged as the primary driver of these community structure variations on global level. However, when considering only seasonal variations, pH and nitrate were identified as key factors influencing bacterial community structures. Summer differed from other seasons in aspects of seasonal symbiotic patterns of bacterial communities, community assembly and function are different from other seasons. There were notable variations in bacterial community structures between winter and summer. Deterministic processes dominated community assembly, but there was an increase in the proportion of stochastic processes during summer. In summer, the functions related to photosynthesis, nitrogen fixation, and decomposition of organic matter showed higher abundance. Our findings shed light on the response of bacterial communities to environmental changes and the underlying mechanisms of community assembly in oligosaline lakes in arid regions.

Higher Contribution of Generalists Than Specialists to the Bacterial Biogeography Across Lakes in Northern China

Abstract:Aim Given that microbial communities normally comprise generalist and specialist species, there have been limited efforts to estimate the contribution of these sub‐communities to bacterial biogeography regarding ecological assembly processes and nestedness structure. To resolve this gap, we studied the spatial distribution of bacterioplankton communities and their generalist and specialist sub‐communities, as well as the underlying assembly processes. Location Six lakes span three climate zones in northern China. Time Period September 2021. Methods By separately analysing the generalist and specialist sub‐communities from the whole communities, we investigated the relationships between sub‐communities and the entire communities in terms of distance‐decay pattern, nestedness, and assembly processes. As such, we assessed the contribution of generalist and specialist sub‐communities to the biogeographic patterns of the whole communities. Major Taxa Studies Bacterial community. Results Generalist sub‐communities were more diverse but less abundant than specialist sub‐communities. In line with the whole communities, both types of sub‐communities exhibited distance‐decay relationships at the regional scale, with the slope of specialists being steeper than that of generalists. Additionally, generalist sub‐communities and whole communities also exhibited similar nestedness patterns, in which assemblages from saline water were the subset of those from freshwater. In terms of assembly processes, generalist sub‐communities and whole communities were strongly regulated by homogeneous selection, whereas specialist sub‐communities were mainly shaped by dispersal limitation. More importantly, there was a transition from stochastic to deterministic assembly processes for generalist sub‐communities and whole communities and generalist sub‐communities with increasing total dissolved solids. Main Conclusions Relative to the specialists, the assembly processes and nestedness pattern were more altered if the generalists were absent, which implies the higher contribution of generalist sub‐communities on the biogeography of whole communities at the regional scale.

Similar Community Assembly Mechanisms Underlie Similar Biogeography of Rare and Abundant Bacteria in Lakes on Yungui Plateau, China

Abstract:The revolution of molecular techniques in the field of microbial ecology not only allows an in‐depth exploration of abundant bacteria, but also confirms the existence of “rare biosphere.” However, the mechanisms underlying the assembly process of abundant and rare bacteria in plateau lakes are still poorly understood. In the present study, distribution of abundant and rare bacteria in 21 lakes on Yungui Plateau, China and the relative importance of deterministic and neutral factors to their biogeographic patterns were examined. Results surprisingly showed that similar biogeographic patterns of rare and abundant taxa were generally shaped via similar assembly mechanisms. For both bacterial groups, deterministic process strongly influenced the distribution, while neutral process was not at play. In addition, they both exhibited a weak distance–decay relationship. Nevertheless, environmental drivers of rare and abundant taxa and their importance were not exactly the same. Water physicochemical property was the most dominant driver shaping the biogeographic pattern of rare taxa, followed by lake morphology and watershed land‐use, while for abundant taxa, watershed land‐use was the most dominant driver and lake morphology did not play a significant role. This study suggested that rare bacteria in high‐altitude aquatic ecosystems might experience equally low extinction risk and respond to environmental changes in a similar manner to abundant ones, but their ecological niches and functions were not identical. To fully understand the assembly mechanism of bacterial community, it is necessary to differentiate the community by traits of taxa.
Metagenomics reveals the diversity and role of surface-water microbes in biogeochemical cycles in lakes at different terrain ladders

Abstract:As the most important component of ecosystems, microbial communities play a significant role in global biogeochemical cycles. Geographical barriers created by topographic differences are proposed as one of the main factors to shape microbial diversity, functional composition and their evolution across aquatic ecosystem. There are few studies that compare the microbial community structure and functional potential of lakes with different terrain ladders (terrains of contrasting elevation levels), especially those involving Qinghai-Tibet Plateau. This study compared microbial 16S rRNA data from 51 lakes with different terrain ladders, showing that the composition of the dominant microbial community was similar, but the microbial abundance was quite different in different terrain ladders. Actinobacteria, Proteobacteria, Cyanobacteria, Planctomycete, Verrucomicrobia and Bacteroidetes were the dominant taxa. Through the correlation analysis between environmental factors and the microbial community structure, it was found that two environmental factors (elevation and salinity) had significant contributions to the microbial composition. Metagenomics of six representative lakes revealed the relationship between microbial composition and metabolic processes related to carbon, nitrogen and sulfur cycles. The comparative analysis of genes related to metabolism cycles showed that functional processes such as aerobic respiration, nitrogen assimilation, the mineralization of nitrogen and sulfur had highest metabolic potential in the ecosystems. The analysis of indicator species based on the metabolic process in the carbon, nitrogen and sulfur cycles showed that the microbial community structure is related to the dominant metabolic functions within biogeochemical cycles. Different dominant species play an important and distinct role in lakes with different gradients. Actinobacteria, Cyanobacteria and Proteobacteria were the most important indicator species on the Qinghai-Tibet Plateau, Yunnan-Guizhou Plateau and the middle and lower reaches of the Yangtze River Plain, respectively. In general, this study explored the functional distribution patterns of the dominant microbial communities in the lakes in biogeocycles.

Abstract:Aim The geographical distribution of bacteria is an important, but poorly understood, topic in microbial ecology. A major question is how broadly distributed generalist taxa, and limitedly distributed specialist taxa, vary across a latitudinal gradient in freshwater ecosystems. We predict that: (a) generalists and specialists exhibit latitudinal diversity gradient with different patterns; (b) their community assemblies are mainly driven by stochastic processes; and (c) generalists coexist through niche differences, while specialists coexist through fitness differences. Location Sixty lakes and reservoirs in China, ranging from 18° N to 50° N. Time Period July–August in 2012 and 2018. Taxa Studied Bacterioplankton. Methods We clustered zero‐radius operational taxonomic unit (zOTU) (subspecies‐like level) into operational taxonomic unit (OTU) (species‐like level) and calculated the intra‐specific variation and spatial asynchrony of OTU. We examined the relative effects of environment versus space (latitude) on bacterial community and quantified the community assembly processes of generalists and specialists, respectively. The binary‐state speciation and extinction (BiSSE) model has been used to estimate the evolutionary characteristics. The abundances of the top zOTUs within each OTU were identified to reveal their niche preferences (distribution patterns at different sites). Results We found that bacterial generalists had a higher intra‐specific variation, greater asynchrony and wider niche width than the specialists. With increasing latitude, bacterial diversity decreased with abundance variations in generalists and occurrence variations in specialists, respectively. Diversification in stochastic processes was the main driving factor in both groups, though deterministic processes showed greater effects on specialists than generalists in local communities. Bacterial generalists coexisted through niche differences, whereas specialists coexisted through average fitness differences. Main Conclusions We demonstrated distinct latitudinal distribution patterns of bacterial generalists and specialists. These results highlight differences in biogeography of microbial generalist and specialist taxa mediated by intra‐specific variation and emphasize their distinctly vital roles mediated by differing mechanisms in freshwater ecosystems.

Abstract:Due to the recent decades of climate change and intensive human activities, endorheic lakes are threatened by both salinization and eutrophication. However, knowledge of the aquatic bacterial community’s response to simultaneous increasing salinity and trophic status is still poor. To address this knowledge gap, we collected 40 surface water samples from five lakes and six rivers on the semi-arid Inner Mongolia Plateau, and investigated their bacterial communities using 16S rRNA gene-targeted amplicon sequencing. We found that bacterial species diversity significantly decreased from the mesotrophic freshwater river habitat to the eutrophic high-brackish lake habitat; salinity was more important than trophic status in explaining this decreased diversity. Salinity was the most important environmental factor in shaping community composition, while increased nitrogen loading was more important in structuring predicted functional composition. Within the lake habitats, the impact of environmental filtering on bacterial community assembly increased with the increasing salinity. The results suggested that the elevated salinity and nutrients have combined effects on the aquatic bacterial community, resulting in dramatic declines in species diversity, and promoted the importance of deterministic processes in community assembly. Our findings provide new insights into bacterial communities’ responses to the intensified climate-driven and anthropogenic environmental changes in aquatic ecosystems.

Differences of Bacterial Community Co-Occurrence Network and Assembly Processes Be-Tween Sediment and Water in Lakes on the Qinghai-Tibet Plateau

Abstract:It is of great scientific significance to study feature differences of bacterial community structure between sediment and water in the plateau lakes for water ecological management and protection.We selected 19 objective lakes in the central and eastern of the Qinghai-Tibet Plateau,collected the samples of sediment and water,and analyzed differences of bacterial community structures,spatial distributions,co-occurrence network patterns,community assembly processes between sediment and water in lakes based on high-throughput sequencing and statistical analysis.The results showed that alpha diversity indices of bacterial community in sediment,including Chaol,Shannon diversity,Faiths phylogenetic diversity and Pielou ’s evenness indices,were significantly higher than those in water.Dominant phylum in both sediment and water was Proteobacteria with the relative abundances of 45.33 %and 41.77%,respectively,while bacterial community structures in water presented stronger spatial heterogeneity than in sediment.The results of principal coordinate analysis and similarity analysis showed that there were significant differences in bacterial community composition between sediment and water.Altitude and Latitude were the most important environmental drivers in shaping bacterial community compositions in sediment and water.Both sediment and water bacterial community had distance-decay patterns,while bacterial spatial turnover rate in sediment was lower than that in water.The co-occurrence network complexity and stability of bacterial community in sediment were higher than those in water.The mean value of normalized stochasticity ratios in sediment and water bacterial were 0.24 and 0.72,indicating deterministic process played a key role in sediment bacterial community assembly processes,whereas water bacterial community assembly processes was predominant by stochastic process.These findings about bacterial community features,spatial patterns,stability of co-occurrence networks,assembly processes,and their differences of bacterial between lake sediment and water in the Qinghai-Tibet Plateau would provide the scientific support for in-depth understanding mechanisms of bacterial diversity and composition formation in plateau lakes.
Salinity-triggered homogeneous selection constrains the microbial function and stability in lakes

Abstract:Climate change and anthropogenic exploitation have led to the gradual salinization of inland waters worldwide. However, the impacts of this process on the prokaryotic plankton communities and their role in biogeochemical cycles in the inland lake are poorly known. Here, we take a space-for-time substitution approach, using 16S rRNA gene amplicon sequencing and metagenomic sequencing. We analyzed the prokaryotic plankton communities of 11 lakes in northwest China, with average water salinities ranging from 0.002 to 14.370%. The results demonstrated that, among the various environmental parameters, salinity was the most important driver of prokaryotic plankton β-diversity (Mantel test, r = 0.53, P < 0.001). (1) Under low salinity, prokaryotic planktons were assembled by stochastic processes and employed diverse halotolerant strategies, including the synthesis and uptake of compatible solutes and extrusion of Na + or Li + in exchange for H + . Under elevated salinity pressure, strong homogeneous selection meant that only planktonic prokaryotes showing an energetically favorable halotolerant strategy employing an Mnh-type Na + /H + antiporter remained. (2) The decreasing taxonomic diversity caused by intense environmental filtering in high-salinity lakes impaired functional diversity related to substance metabolism. The prokaryotes enhanced the TCA cycle, carbon fixation, and low-energy-consumption amino acid biosynthesis in high-salinity lakes. (3) Elevated salinity pressure decreased the negative:positive cohesion and the modularity of the molecular ecology networks for the planktonic prokaryotes, indicating a precarious microbial network. Our findings provide new insights into plankton ecology and are helpful for the protecting of the biodiversity and function of inland lakes against the background of salinization.

Distribution of Bacterial Communities Across Plateau Freshwater Lake and Upslope Soils.

Abstract:Microorganisms are involved in a variety of biogeochemical processes in natural environments. The differences between bacterial communities in freshwaters and upslope soils remain unclear. The present study investigated the bacterial distribution in a plateau freshwater lake, Erhai Lake (southwestern China), and its upslope soils. Illumina MiSeq sequencing illustrated high bacterial diversity in lake sediments and soils. Sediment and soil bacterial communities were mainly composed of Proteobacteria, Acidobacteria, Actinobacteria, Bacteroidetes, Chloroflexi and Planctomycetes. However, a distinctive difference in bacterial community structure was found between soil and sediment ecosystems. Water content, nitrogen and pH affected the distribution of the bacterial community across Erhai Lake and its upslope soils. Moreover, the soil bacterial community might also be shaped by plant types. This work could provide some new insights into plateau aquatic and terrestrial microbial ecology.
A meta-analysis reveals that geographical factors drive the bacterial community variation in Chinese lakes

Abstract:The biogeographical distribution of plants and animals has been extensively studied, however, the biogeographical patterns and the factors that influence bacterial communities in lakes over large scales are yet to be fully understood, even though they play critical roles in biogeochemical cycles. Here, bacterial community compositional data, geographic information, and environmental factors were integrated for 326 Chinese lakes based on previously published studies to determine the underlying factors that shape bacterial diversity among Chinese lakes. The composition of bacterial communities significantly varied among the three primary climatic regions of China (Northern China, NC; Southern China, SC; and the Tibetan Plateau, TIP), and across two different lake habitats (waters and sediments). Sediment bacterial communities exhibited significantly higher alpha-diversity and distance-decay relationships compared to water communities. The results indicate that the "scale-dependent patterns" of controlling factors, primarily influenced by geographical factors, become increasingly pronounced as the spatial scale increases. At a national scale, geographical factors exerted a dominant influence on both the water and sediment communities across all lakes, as geographical barriers restrict the dispersal of individuals. At smaller spatial scales, temperature-driven selection effects played a greater role in shaping water bacterial community variation in the NC, SC, and TIP, while geographical factors had a stronger association with sediment bacterial community variation in the lakes of the three regions. This synthesis offers novel insights into the ecological factors that determine the distribution of bacteria in Chinese lakes.

Abstract:Both planktonic and sediment bacterial assemblages are the important components of freshwater lake ecosystems. However, their spatiotemporal shift and the driving forces remain still elusive. Eutrotrophic Dianchi Lake and mesotrophic Erhai Lake are the largest two freshwater lakes on the Yunnan Plateau (southwestern China). The present study investigated the spatiotemporal shift in both planktonic and sediment bacterial populations in these two plateau freshwater lakes at different trophic status. For either lake, both water and sediment samples were collected from six sampling locations in spring and summer. Bacterioplankton community abundance in Dianchi Lake generally far outnumbered that in Erhai Lake. Sediment bacterial communities in Erhai Lake were found to have higher richness and diversity than those in Dianchi Lake. Sediments had higher bacterial community richness and diversity than waters. The change patterns for both planktonic and sediment bacterial communities were lake-specific and season-specific. Either planktonic or sediment bacterial community structure showed a distinct difference between in Dianchi Lake and in Erhai Lake, and an evident structure difference was also found between planktonic and sediment bacterial communities in either of these two lakes. Planktonic bacterial communities in both Dianchi Lake and Erhai Lake mainly included Proteobacteria (mainly Alpha- , Beta- , and Gammaproteobacteria ), Bacteroidetes , Actinobacteria , Cyanobacteria , and Firmicutes , while sediment bacterial communities were mainly represented by Proteobacteria (mainly Beta- and Deltaproteobacteria ), Bacteroidetes , Chlorobi , Nitrospirae , Acidobacteria , and Chloroflexi . Trophic status could play important roles in shaping both planktonic and sediment bacterial communities in freshwater lakes.
Variations in bacterial diversity and community structure in the sediments of an alkaline lake in Inner Mongolia plateau, China

Abstract:Alkaline lakes are a special aquatic ecosystem that act as important water and alkali resource in the arid-semiarid regions. The primary aim of the study is to explore how environmental factors affect community diversity and structure, and to find whether there are key microbes that can indicate changes in environmental factors in alkaline lakes. Therefore, four sediment samples (S1, S2, S3, and S4) were collected from Hamatai Lake which is an important alkali resource in Ordos' desert plateau of Inner Mongolia. Samples were collected along the salinity and alkalinity gradients and bacterial community compositions were investigated by Illumina Miseq sequencing. The results revealed that the diversity and richness of bacterial community decreased with increasing alkalinity (pH) and salinity, and bacterial community structure was obviously different for the relatively light alkaline and hyposaline samples (LAHO; pH < 8.5; salinity < 20‰) and high alkaline and hypersaline samples (HAHR; pH > 8.5; salinity > 20‰). Firmicutes, Proteobacteria and Bacteriodetes were observed to be the dominant phyla. Furthermore, Acidobacteria, Actinobacteria, and low salt-tolerant alkaliphilic nitrifying taxa were mainly distributed in S1 with LAHO characteristic. Firmicutes, Clostridia, Gammaproteobacteria, salt-tolerant alkaliphilic denitrifying taxa, haloalkaliphilic sulfur cycling taxa were mainly distributed in S2, S3 and S4, and were well adapted to haloalkaline conditions. Correlation analysis revealed that the community diversity (operational taxonomic unit numbers and/or Shannon index) and richness (Chao1) were significantly positively correlated with ammonium nitrogen (r = 0.654, p < 0.05; r = 0.680, p < 0.05) and negatively correlated with pH (r = -0.924, p < 0.01; r = -0.800, p < 0.01; r = -0.933, p < 0.01) and salinity (r = -0.615, p < 0.05; r = -0.647, p < 0.05). A redundancy analysis and variation partitioning analysis revealed that pH (explanation degrees of 53.5%, pseudo-F = 11.5, p < 0.01), TOC/TN (24.8%, pseudo-F = 10.3, p < 0.05) and salinity (9.2%, pseudo-F = 9.5, p < 0.05) were the most significant factors that caused the variations in bacterial community structure. The results suggested that alkalinity, nutrient salt and salinity jointly affect bacterial diversity and community structure, in which one taxon (Acidobacteria), six taxa (Cyanobacteria, Nitrosomonadaceae, Nitrospira, Bacillus, Lactococcus and Halomonas) and five taxa (Desulfonatronobacter, Dethiobacter, Desulfurivibrio, Thioalkalivibrio and Halorhodospira) are related to carbon, nitrogen and sulfur cycles, respectively. Classes Clostridia and Gammaproteobacteria might indicate changes of saline-alkali conditions in the sediments of alkaline lakes in desert plateau.

Abstract:Both Bacteria and Archaea might be involved in various biogeochemical processes in lacustrine sediment ecosystems. However, the factors governing the intra-lake distribution of sediment bacterial and archaeal communities in various freshwater lakes remain unclear. The present study investigated the sediment bacterial and archaeal communities in 13 freshwater lakes on the Yunnan Plateau. Quantitative PCR assay showed a large variation in bacterial and archaeal abundances. Illumina MiSeq sequencing illustrated high bacterial and archaeal diversities. Bacterial abundance was regulated by sediment total organic carbon and total nitrogen, and water depth, while nitrate nitrogen was an important determinant of bacterial diversity. Proteobacteria, Acidobacteria, Actinobacteria, Bacteroidetes, Chlorobi, Chloroflexi, Cyanobacteria, Firmicutes, Gemmatimonadetes, Nitrospirae, Planctomycetes, and Verrucomicrobia were the major components of sediment bacterial communities. Proteobacteria was the largest phylum, but its major classes and their proportions varied greatly among different lakes, affected by sediment nitrate nitrogen. In addition, both Euryarchaeota and Crenarchaeota were important members in sediment archaeal communities, while unclassified Archaea usually showed the dominance.
Distinct Multitrophic Biodiversity Composition and Community Organization in a Freshwater Lake and a Hypersaline Lake on the Tibetan Plateau

Abstract:Alpine lakes play pivotal roles in plateau hydrological processes but are highly sensitive to climate change, yet we lack comprehensive knowledge of their multitrophic biodiversity patterns. Here, we compared the biodiversity characteristics of diverse taxonomic groups across water depths and in surface sediments from a freshwater lake and a hypersaline lake on the northwestern Tibetan Plateau. Using multi-marker environmental DNA metabarcoding, we detected 134 cyanobacteria, 443 diatom, 1,519 invertebrate, and 28 vertebrate taxa. Each group had a substantially different community composition in the two lakes, and differences were also found between water and sediments within each lake. Cooccurrence network analysis revealed higher network complexity, lower modularity, and fewer negative cohesions in the hypersaline lake, suggesting that high salinity may destabilize ecological networks. Our results provide the first holistic view of Tibetan lake biodiversity under contrasting salinity levels and reveal structural differences in the ecological networks that may impact ecosystem resilience.
Heterogeneous bacterial communities affected by phytoplankton community turnover and microcystins in plateau lakes of Southwestern China

Abstract:Both phytoplankton and bacteria are fundamental organisms with key ecological functions in lake ecosystems. However, the mechanistic interactions through which phytoplankton community change and bacterial communities interact remain poorly understood. Here, the responses of bacterial communities to the community structure, resource-use efficiency (RUE), and community turnover of phytoplankton and microcystins (MCs) were investigated in Lake Dianchi, Lake Xingyun, and Lake Erhai of Southwestern China across two seasons (May and October 2020). Among phytoplankton, Cyanobacteria was the dominant species in all three lakes and attained greater dominance in October than in May due to variation in the RUE of nitrogen and phosphorus and environmental changes. The production of MCs, including MC_LR, MC_RR and MC_YR, was the result of the massive Cyanobacteria. Decreases in diversity and increases in heterogeneity were observed in the bacterial community structure. Nutrient levels, environmental factors and MCs (especially MC_YR) jointly affected the bacterial community in lakes, namely its diversity and community assembly. The cascading effects in lakes mediated by environmental conditions, phytoplankton community composition, RUE, community turnover, and MCs on bacterial communities were revealed in this study. These findings underscore the importance of relating phytoplankton community change and MCs to the bacterial community, which is fundamental for better understanding the lake ecosystem functioning and potential risks of MCs.
