Abstract:The effect and mechanism of action of lanthanide ions on sarcoplasmic reticulum (SR) (Ca2++Mg2+)-ATPase activity are investigated. Low concentrations of Gd3+ stimulate the SR ATPase activity and the activity is inhibited with the increasing of the concentrations of Gd3+. Gd3+ inhibits the purified ATPase activity. When the purified ATPase is reconstituted with phosphatidic acid (PA) or cardiolipin (CL), low concentrations of Gd3+ activate its activity, whereas the activity will be inhibited if the enzyme is reconstituted with phosphatidylcholine (PC), phosphatidylserine (PS) and the mixture of phosphatidylcholine and phosphatidylethanolamine (PC/PE). Furthermore, different actions of Tb3+ on Ca2+-binding sites of the SR and purified ATPase have been observed.
Parathyroid hormone selectively inhibits L-type calcium channels in single vascular smooth muscle cells of the rat.

Abstract:1. The active synthetic N-terminal fragment of bovine parathyroid hormone, bPTH-(1-34) at a concentration of 1-mu-M, decreased the peak amplitude of the long-lasting (L-type) calcium channel current by 37% (n = 14, P < 0.01) in rat tail artery smooth muscle cells. By contrast, this fragment of parathyroid hormone (PTH) (1-mu-M) had no effect on the transient (T-type) calcium channel current in the same cell preparation. 2. The inhibitory effect of bPTH-(1-34) on L-channel currents was reversible and could be antagonized by the L-channel agonist, Bay K 8644. In contrast, bPTH-(1-34) inhibited Bay K 8644-induced amplification of L-channel currents. 3. The inhibitory effect of bPTH-(1-34) on L-Channel currents was dose dependent with a threshold concentration of less than 10(-7) M, and voltage dependent with increased inhibition at more positive holding potentials. However, this effect of bPTH-(1-34) was not dependent on different pulse lengths or interpulse intervals. 4. The kinetics of deactivation of L-channel currents were not changed although the instantaneous amplitude of the L-channel tail current was reduced by bPTH-(1-34). 5. Application of bPTH-(1-34) antagonists (10(-6) m-bPTH-(3-34) and 10(-5) M-bPTH-(7-34)) did not result in any significant change in the magnitude of L-channel currents (n = 15 and n = 7, respectively). 6. Pre-incubation of cells with bPTH-(3-34) for more than 15 min abolished the inhibitory effect of bPTH-(1-34) on L-channel currents. 7. The present study provides direct evidence to demonstrate that PTH, an endogenous circulating hormone, is a selective inhibitor of L-channel currents in vascular smooth muscle cells.
Specific Inhibition of Long-Lasting, L-type Calcium Channels by Synthetic Parathyroid Hormone.

Abstract:The effect of an active synthetic N-terminal fragment of bovine parathyroid hormone (bPTH), bPTH-(1-34), on Ca2+ channels was studied in mouse neuroblastoma cells (N1E-115). With the whole-cell variation of the patch-clamp technique, T (transient) and L (long-lasting) types of Ca2+ currents were identified. Pharmacological characterization showed that the L current was amplified by the Ca2+ channel stimulator BAY K-8644, but the T current was unaffected. The administration of bPTH-(1-34) produced dose-related inhibition of the L current, which could be reversed by BAY K-8644. The peptide had no effect on the T current. In addition, use of the fluorescent indicator fura-2 showed that bPTH-(1-34) inhibited the KCl-stimulated increase in intracellular free Ca2+ in neuroblastoma cells with L channels but not in cells with T channels. An inactivated (oxidized) preparation of bPTH-(1-34) failed to affect the L current. High-affinity binding of labeled PTH analog to these neuroblastoma cells was also demonstrated. In addition, bPTH-(1-34) inhibited the L current in cultured vascular smooth muscle cells from rat tail artery. These data indicate that, in some tissues, PTH can act as an endogenous blocker of Ca2+ entry.

Abstract:The present studies demonstrate a stimulatory effect of intact parathyroid hormone on hepatic glucose production in vivo. This increased glucose production was accompanied by augmented alanine uptake by the liver. In addition, PTH increased the conversion of 14C-alanine to 14C-glucose. These results suggest that the increased hepatic release of glucose produced by PTH is mediated in part by increased gluconeogenesis. On the other hand, the synthetic 1-34 amino terminal fragment of PTH, which is not extracted by the liver, did not stimulate hepatic glucose production.

Abstract:1.1. A second hormone of the parathyroid glands, calcitonin, that lowers blood calcium has been postulated by Copp and Cameron on the basis of experiments in intact dogs. The reported hypocalcemic effects are characterized by rapid onset, small magnitude, and short duration.2.2. The classic parathyroid hormone, a calcium-mobilizing polypeptide, has been most effectively purified from a phenol extract of bovine parathyroid glands by the use of countercurrent distribution or gel filtration. The molecular weight has been estimated at approximately 9000, with 76 to 83 amino acid residues in a single chain. Purified polypeptide preparations of lower molecular weight and lower specific biological activity have been obtained from extracts made with dilute hydrochloric acid and with acetic acid.3.3. One plausible but unproved explanation of the loss, recovery, or enhancement of biological activity of the hormone when treated with various reagents is the reversible oxidation and reduction of the methionine residue (with a subsequent conformational change in the molecule about its active site(s)).4.4. Biological assay methods in current use for following the purification of parathyroid hormone and for standardization are not sensitive enough for satisfactory quantitation of the hormone in blood or urine. However, new in vitro and immunochemical methods may satisfy this need and in addition provide the means for investigating possible species differences.

Abstract:Objective To observe the levels of PTH secretion in primary cultured parathyroid cells and par-athyroid tissues stimulated by high phosphate.Methods The parathyroid glands of 28 Wistar female rats were success-fully resected by surgical operation under dissection microscope and were prepared for primary parathyroid cell culture and tissue culture.The cells and tissues were identified by histomorphology and PTH measurement and then cultured.Parathyroid tissues and cells were divided into 2 groups respectively as below: tissue groups:control group(TN,Pi 1.0mml/l,Ca 1.25mmol/l),experimental group(TH,Pi 3.5mmol/l,Ca 1.25mmol/l);cell groups:control group(CN,Pi 1.0mml/l,Ca 1.25mmol/l)and experimental group(CH,Pi 3.5mmol/l,Ca 1.25mmol/l).The medium was changed every two days.And the parathyroid hormone obtained from the supernatant for 0h,4h,12h,24h,48,72h,5d,7d,9d was measured.Results Parathyroid glands were identified by histomorphology and PTH measured.The PTH secretion curves of parathyroid cells and tissues incubated in normal phosphate and normal calcium medium were similar,and the peak time was 48h.Varing medium phosphate(1mM or 3.5mM) have no effect on PTH secretion(P > 0.05) in cell groups.But when PTG tissues were incubated in high phosphate medium,PTH levels could increase to 10-fold higher than baseline at 48h.There was a significant difference between group TH and group TN at the same time except H0(P < 0.01).Conclusion High phosphate had a direct effect on PTH release from parathyroid tissue but not from dispersed parathyroid cells.Parathyroid tissues should be chosen to study the effect of phosphate on parathy-roid function in vitro,and the time selected should be within 48h.

Abstract:The parathyroid hormone receptor-1 (PTH1R) is a class B G protein-coupled receptor central to calcium homeostasis and a therapeutic target for osteoporosis and hypoparathyroidism. Here we report the cryo-electron microscopy structure of human PTH1R bound to a long-acting PTH analog and the stimulatory G protein. The bound peptide adopts an extended helix with its amino terminus inserted deeply into the receptor transmembrane domain (TMD), which leads to partial unwinding of the carboxyl terminus of transmembrane helix 6 and induces a sharp kink at the middle of this helix to allow the receptor to couple with G protein. In contrast to a single TMD structure state, the extracellular domain adopts multiple conformations. These results provide insights into the structural basis and dynamics of PTH binding and receptor activation.
Short carboxyl terminal parathyroid hormone peptides modulate human parathyroid hormone signaling in mouse osteoblasts

Abstract:Background: Novel human parathyroid hormone (hPTH) peptides of unknown biological activity have recently been identified in the serum of subjects with normal renal function, chronic renal failure, and end-stage renal disease through the application of liquid chromatography-high resolution mass spectrometry. Purpose: of experiments: To determine the bioactivity of these peptides, we synthesized hPTH28-84, hPTH38-84, and hPTH45-84 peptides by solid phase peptide synthesis and tested their bioactivity in MC3T3-E1 mouse osteoblasts, either individually or together with the native hormone, hPTH1-84, by assessing the accumulation of 3´,5´-cyclic adenosine monophosphate (cAMP) and the induction of alkaline phosphatase activity. Results: Increasing doses of hPTH1-84 (1-100 nM) increased the accumulation of cAMP and alkaline phosphatase activity in osteoblasts. hPTH28-84, hPTH38-84, and hPTH45-84 in concentrations of 1-100 nM were biologically inert. Surprisingly, 100 nM hPTH38-84 and hPTH45-84 increased the accumulation of cAMP in osteoblasts treated with increasing amounts of hPTH1-84. Human PTH28-84 had no effects on cAMP activity alone or in combination with hPTH1-84. Conversely, 100 nM hPTH38-84, hPTH45-84, and hPTH28-84 blocked the activation of alkaline phosphatase activity by hPTH1-84. Conclusions: The data show that the short carboxyl-terminal hPTH peptides, hPTH38-84 and hPTH45-84, increase the amount of cellular cAMP generated in cultured osteoblasts in response to treatment with full-length hPTH1-84 when compared to full-length hPTH1-84 alone. Human PTH28-84 had no effect on cAMP activity alone or in combination with hPTH1-84. Human PTH28-84, hPTH38-84 and hPTH45-84 reduced the effects of hPTH1-84 in osteoblasts with respect to the induction of alkaline phosphatase activity compared to hPTH1-84 alone. Short carboxyl peptides of human PTH are biologically inert but when administered together with full-length hPTH1-84 modulate the bioactivity of hPTH1-84 in osteoblasts.

Abstract:Central to the maintenance of calcium homeostasis is the regulated reabsorption of calcium along the nephron. To this end, parathyroid hormone (PTH) is released from the parathyroid gland in response to lowered plasma calcium levels. This hormone acts through the PTH 1 receptor along the nephron to increase urinary phosphate excretion and decrease urinary calcium excretion. In the proximal tubule, PTH inhibits phosphate reabsorption by reducing the abundance of sodium phosphate cotransporters in the apical membrane. PTH likely decreases calcium reabsorption from the proximal tubule, by reducing the reabsorption of sodium, an event necessary for the paracellular movement of calcium across this segment. In the thick ascending limb (TAL), PTH increases calcium permeability and may increase the electrical driving force thereby increasing calcium reabsorption in the TAL. Finally, in the distal convolution, PTH acts to increase transcellular calcium reabsorption by increasing the activity and abundance of the apically expressed calcium channel TRPV5.

Abstract:We study a dynamic approach to the problem of scheduling real time requests in a video on demand server. Most of the previous VOD scheduling approaches use limited run time information, and thus cannot exploit the potential capacity fully. Our approach improves throughput by making use of run time information to relax admission control, and to reduce admission to start delay by providing earliest initial service. We establish conditions for greedy admission, dynamic control of the sequence and sizes of disk accesses, and fast initial service. We conduct thorough simulations over a wide range of buffer capacities, load settings, and over varying playout rates to demonstrate the significant improvements in throughput and admission re start delay of our approach relative to a typical static approach which does not use run time information.

Abstract:As a continuation of our studies on mineralization in epiphyseal growth plate (GP) chondrocyte cultures, the effects of tri-iodothyronine (T3) in both beta-glycerophosphate-containing, serum-free (HL-1) and beta-glycerophosphate-free, serum-containing medium (DATP5) were studied. The GP cells responded to T3 in a serum-, stage-, and dosage-dependent manner. Added at graded levels (0.1-10.0 nM) to preconfluent cultures (from day 7) in both HL-1 and DATP5, T3 caused progressive decreases in protein, collagen, and DNA synthesis but increased mineral deposition. In postconfluent cultures, these effects of T3 were generally muted. In preconfluent cultures, proteoglycan (PG) levels were not significantly affected in DATP5, although in HL-1 they were decreased by approximately 50%. In postconfluent cultures, T3 increased PG levels in DATP5 but had no effect in HL-1. In HL-1, alkaline phosphatase (ALP) activity was progressively increased by 200-500% in both pre- and postconfluent cultures. In DATP5 in preconfluent cultures, T3 initially stimulated but later suppressed ALP; in postconfluent cultures, T3 also transiently increased ALP but did not suppress activity upon longer exposure. The inhibitory effects of T3 on protein, PG, and DNA levels of GP chondrocytes suggest that in vivo its effects on bone growth must occur primarily after cellular proliferation. Apparently by binding to the 50 kDa thyroxine-binding globulin, which cannot penetrate the PG barrier, accessibility of T3 to GP chondrocytes is limited until the time of vascular penetration when its stimulatory effects on ALP and mineral deposition become critical for continued bone development.

Abstract:Growth hormone pituitary adenoma (GHPA) is a major subtype of pituitary adenoma (PA), with tumor enlargement and abnormal secretion of growth hormone (GH) often causing complications. Rac GTPase-activating protein 1 (RACGAP1), a member of the guanine triphosphatase-activating protein family, is highly overexpressed in multiple tumors and promotes tumor growth. However, the role of RACGAP1 in GHPA remains unelucidated. Besides, specific inhibitors targeting RACGAP1 have not yet been developed. In this study, we aimed to determine the expression and function of RACGAP1 in GHPA and identify effective inhibitors against RACGAP1. Immunohistochemistry was used to detect the expression of RACGAP1 in GHPA and normal pituitary tissues. The effect of RACGAP1 on cell proliferation, apoptosis, and cell cycle was evaluated by knockdown of RACGAP1 in GH3 cells in vitro and xenograft models of GHPA in vivo. The downstream mechanism of RACGAP1 was explored by RNA sequencing, bioinformatic analysis, and Western blot. Inhibitors targeting RACGAP1 were screened and verified through a structure-based virtual docking method, cell viability assays, and surface plasmon resonance (SPR) experiments. RACGAP1 expression was increased in GHPA compared with normal pituitary tissues. Knocking down RACGAP1 suppressed cell growth in vitro and in vivo. Preliminary mechanism studies indicated that inhibition of RACGAP1 led to the upregulation of p21 and the downregulation of several genes involved in the cell cycle signaling pathway, such as Cyclin A, CDK1, and CDK2. Moreover, DB07268 was identified for the first time as an effective RACGAP1 inhibitor that could prominently restrain the proliferation of GH3 cells. This study demonstrates that RACGAP1 plays a critical role in GHPA, highlighting the novel inhibitor DB07268 as a promising therapeutic approach.
Effects of Calcitonin Gene-Related Peptide on Bone Resorption Mediated by Interleukin-1.

Abstract:We examined the effects of calcitonin gene-related peptide CGRP on the bone resorption stimulated by IL-1 alpha after neonatal mouse calvaria were cultured in vitro and bone resorption was determined by measuring the release of calcium into the medium after alpha 48-hour incubation. CGRP (10(-9)-10(-8) M) blocked completely the IL1 alpha-mediated increase in bone resorption and inhibited the basal bone resorption. In accordance with the effects of calcitonin on osteoclasts, CGRP caused cessation of lamellipodia activity and gradual retraction of lamellipodia when added into the culture medium. Quantitative analysis showed that CGRP (10(-6) M) caused a gradual reduction of osteoclastic surface area and convex perineum. These data show that CGRP inhibits IL1 alpha mediated bone resorption because of its direct regulation of osteoclast activity.
Comparison of the Pharmacological Effects of Paricalcitol and Doxercalciferol on the Factors Involved in Mineral Homeostasis

Abstract:Vitamin D receptor agonists (VDRAs) directly suppress parathyroid hormone (PTH) mRNA expression. Different VDRAs are known to have differential effects on serum calcium (Ca), which may also affect serum PTH levels since serum Ca regulates PTH secretion mediated by the Ca-sensing receptor (CaSR). In this study, we compared the effects of paricalcitol and doxercalciferol on regulating serum Ca and PTH, and also the expression of PTH, VDR, and CaSR mRNA. The 5/6 nephrectomized (NX) Sprague-Dawley rats on a normal or hyperphosphatemia-inducing diet were treated with vehicle, paricalcitol, or doxercalciferol for two weeks. Both drugs at the tested doses (0.042–0.33 g/kg) suppressed PTH mRNA expression and serum PTH effectively in the 5/6 NX rats, but paricalcitol was less potent in raising serum Ca than doxercalciferol. In pig parathyroid cells, paricalcitol and the active form of doxercalciferol induced VDR translocation from the cytoplasm into the nucleus, suppressed PTH mRNA expression and inhibited cell proliferation in a similar manner, although paricalcitol induced the expression of CaSR mRNA more effectively. The multiple effects of VDRAs on modulating serum Ca, parathyroid cell proliferation, and the expression of CaSR and PTH mRNA reflect the complex involvement of the vitamin D axis in regulating the mineral homeostasis system.

Abstract:Parathyroid hormone (PTH) receptor (PTHR) is a medically important family B G Protein Coupled Receptor (GPCR) that primarily couples to Gs/cAMP and Gq/Ca2+ signaling pathways and has a central role in regulating Ca2+ homeostasis and bone turnover. PTHR is activated by two endogenous hormones, endocrine PTH and paracrine PTH related protein (PTHrP), but only PTH mediates prolonged activation of the receptor and sustained cAMP responses after receptor internalization to the endosomes. To investigate the structural basis of prolonged activation of PTHR, we employed cryo‐EM and solved the high resolution (3Å) structure of PTHR coupled to Gs and activated by long‐acting PTH (LA‐PTH), an analog of PTH that remarkably sustains endosomal PTHR signaling and induces significantly prolonged elevation in blood calcium in mice and monkeys. LA‐PTH binds the receptor as a single extended α‐helix with its N‐terminus inserted deep into the transmembrane domain (TMD) of the receptor, while positioning the extracellular domain (ECD) of PTHR perpendicular to the cell membrane. The receptor activating N‐terminal half of LA‐PTH forms a dense network of hydrophobic and polar interactions with receptor's TMD resulting in a tight binding and giving structural basis for remarkably sustained activation of PTHR by LA‐PTH. Binding of LA‐PTH activates the receptor by inducing partial unwinding of the C‐terminus of transmembrane helix 6 (TM6) and a sharp kink at the middle of this helix, a hallmark of family B GPCR activation. This results in a pronounced outward movement of TM6, opening the cytosolic cavity of the receptor, which allows subsequent G protein coupling. Additionally, our cryo‐EM data revealed structural dynamics of PTHR demonstrated by the ability of receptor ECD to adopt multiple conformational states. TRPC3 Is Downregulated in Primary Hyperparathyroidism

Abstract:Transient receptor potential canonical sub-family channel 3 (TRPC3) is considered to play a critical role in calcium homeostasis. However, there are no established findings in this respect with regard to TRPC6. Although the parathyroid gland is a crucial organ in calcium household regulation, little is known about the protein distribution of TRPC channels—especially TRPC3 and TRPC6—in this organ. Our aim was therefore to investigate the protein expression profile of TRPC3 and TRPC6 in healthy and diseased human parathyroid glands. Surgery samples from patients with healthy parathyroid glands and from patients suffering from primary hyperparathyroidism (pHPT) were investigated by immunohistochemistry using knockout-validated antibodies against TRPC3 and TRPC6. A software-based analysis similar to an H-score was performed. For the first time, to our knowledge, TRPC3 and TRPC6 protein expression is described here in the parathyroid glands. It is found in both chief and oxyphilic cells. Furthermore, the TRPC3 staining score in diseased tissue (pHPT) was statistically significantly lower than that in healthy tissue. In conclusion, TRPC3 and TRPC6 proteins are expressed in the human parathyroid gland. Furthermore, there is strong evidence indicating that TRPC3 plays a role in pHPT and subsequently in parathyroid hormone secretion regulation. These findings ultimately require further research in order to not only confirm our results but also to further investigate the relevance of these channels and, in particular, that of TRPC3 in the aforementioned physiological functions and pathophysiological conditions.

Abstract:Parathyroid hypertensive factor (PHF) has been implicated in regulation of vascular smooth muscle tone and pathogenesis of several forms of hypertension. Earlier studies have suggested that PHF enhances the actions of other vasoconstrictors, while it has no in vitro vasoconstrictor property of its own. PHF was previously found to enhance the L-type Ca channel currents and intracellular Ca responses to depolarization in vascular smooth muscle cells (VSMCs). The present study examined whether PHF might act on K channels in the plasma membrane of VSMCs. Primary cultured VSMCs from rat tail artery were used. The whole-cell version of the patch-clamp technique was used under conditions in which there was no contribution of Ca-activated K channels to the outward current. Both purified and semipurified PHF inhibited the delayed rectifier type potassium current in a dose-dependent manner. The effect was time dependent and was first significantly different from the control current after 30 min. The inhibition of the delayed rectifier K channel was associated with a time-dependent decrease in the resting membrane potential. Therefore, PHF may alter VSMC cellular Ca responses by reducing the membrane potential to a level closer to the activation potential of Ca channels.
[Effect of Parathyroid Hormone and 1,25-(OH)2-D3 on the Expression of the Mrna of Calbindin D9k in Caco-2 Cells].

Abstract:OBJECTIVE:To study the effect of parathyroid hormone (PTH) on the expression of the mRNA of calbindin D9k(CaBP-D9k) in Caco-2 Cells and whether PTH influences the effect of 1,25-(OH)2-D3 on the expression of calbindin mRNA in Caco-2 Cells.METHODS:Caco-2 cells were used as the model of human small intestinal enterocytes. Caco-2 cells were treated 5, 10, 20, 40 and 80 min at the dose of 10(-8), 10(-9) and 10(-12) mol/L PTH, another groups of Caco-2 cells were treated for 2, 4, 8, 16 and 24h at the dose of 10(-8) mol/L 1,25-(OH)2-D3 alone. The other groups of Caco-2 cells were treated for 2, 4, 8, 16 and 24h at the dose of 10(-8) mol/L 1,25-(OH)2-D3 plus at the dose of 10(-8), 10(-9) and 10(-12) mol/L of PTH respectively. 0.1% ethanol solutions were used as the vehicle for the later two groups. The expression of calbindin D9k mRNA was determined by RT-PCR (the level of GAPDH was taken as internal control).RESULTS:The expressions of calbindin D9k mRNA were more higher than those of blank controls when Caco-2 cells were treated at the dose of 10(-8) mol/L 1,25-(OH)2-D3 for 2, 4, 8, 16 and 24h, the expressions of CaBP-D9k mRNA were higher than those of vehicle. The increases of CaBP-D9k mRNA levels caused by 10(-8) mol/L 1,25-(OH)2-D3 plus PTH were more lower than those caused by 1,25-(OH)2-D3 alone.CONCLUSION:10(-8) and 10(-12) mol/L PTH probably caused increase on the expression of CaBP-D9k mRNA in short period (1 - 20min). 10(-8) mol/L 1,25-(OH)2-D3 increases CaBP-D9k mRNA levels in Caco-2 cells. PTH may inhibit or counteract the increase in CaBP-D9k mRNA caused by 1,25-(OH)2-D3 in Caco-2 cells.

Abstract:Our previous studies have shown PTH to be an effective relaxant of smooth muscle throughout the mammalian tract. Recently, we found PTHrP to be equally as potent and effective on the gut as PTH, and we hypothesized that PTHrP, rather than PTH, might be the natural ligand for the gut receptors which mediate GI smooth muscle relaxation. To approach this question, we asked whether rat GI tissue expresses mRNA for PTHrP. Using selective reverse transcription and PCR we have found PTHrP mRNA in smooth muscle throughout the rat GI tract and in gastric and colonic mucosa as well. Our findings support the idea that PTHrP can be produced by GI tissues and that it may function there as an autocrine or paracrine factor. One of its actions may involve regulation of GI muscle tone.

Abstract:The modulation of [Ca2+](i) by parathyroid hormone (PTH) has been extensively studied in vertebrates. The present study examined the effects of PTH on [Ca2+](i) in isolated invertebrate neurons B5 from buccal ganglia of the pond snail, Helisoma trivolvis, utilizing the Fura-2 fluorescence technique. Bovine PTH, bPTH-(1-84), induced a slow and sustained increase in [Ca2+](i) in neurons B5. In contrast, the elevation of extracellular K+ concentration from 1.7 mM to 15 mM induced a rapid and transient increase in [Ca2+](i). Simultaneous application of 15 mM KCl and bPTH-(1-84), or application of 15 mM KCl in the presence of bPTH-(1-84) additively increased [Ca2+](i) in neurons B5. An increase in [Ca2+](i) in neurons B5 was also induced by a PTH agonist [bPTH-(1-34)], but not by a PTH antagonist [bPTH-(3-34)]. The absence of calcium, or the presence of lanthanum (2 mM) or omega-conotoxin (10 mu M), in the bath solution abolished the effect of bPTH-(1-84) on [Ca2+](i). Furthermore, our results demonstrated that the effect of PTH on [Ca2+](i) in neurons B5 was not due to the hormonal modulation of voltage-dependent Na+ or K+ channels or a Na+/Ca2+ exchanger. The results from this study show that PTH can modulate [Ca2+](i) in an identified invertebrate neuron mainly by promoting extracellular calcium influx via the N-like voltage-dependent calcium channel.
