Abstract:Forty Holstein dairy cows were allocated equally into a control and an experimental group according to matched-pair grouping design. At the beginning of parturition until 90 days after parturition, cows in the experimental group were fed basic diet supplemented with 400 g rumen-undegraded fat. Milk yield was detected. Blood samples were collected from vena caudalis for detection of plasma total protein, albumin, goblin, glucose, urea nitrogen and nonesterified fatty acids (NEFA) on days 0, 15, 30, 45, 60, 75 and 90 after parturition, respectively. It was found that supplemental rumen-passed fat increased milk yield without altering the concentration of plasma total protein, albumin, goblin or glucose. However, supplementation of fat significantly increased plasma NEFA concentration during the first 30 days after parturition. In conclusion, supplemental fat increased milk yield and enhanced lipolysis in a short term; however, it had no effect on plasma protein and glucose concentration.

Abstract:Forty multiparous Holstein dairy cows with similar parity and milk yielding of the last year were allocated equally into a control and an experimental group according to matched-pair grouping design. Beginning at the day of parturition until 90 days after parturition, cows in the experimental group were fed basic diet supplemented with 400 g rumen-passed fat. It was found that rumen-passed fat did not alter dry matter intake (DIM)(P0.05). However, it increased milk yielding(P0.05). Milk fat percentage tended to be increased by supplemental fat, although the differences between the two groups were not pronounced(P0.05). There were no differences in milk protein percentage between the two groups(P0.05). Supplemental fat had minor effect on total number of peripheral leukocytes and the number of lymphocyte(P0.05); however, it tended to reduce somatic cell counts (SCC) in milk (P0.05). It was concluded that supplemental fat increased milk yielding, and improved immune function to a certain extent.

Abstract:This study aimed to evaluate the effects of parity, body condition score (BCS) at calving, and milk yield on the metabolic profile of Gyr (Zebu) cows. Healthy cows in late pregnancy were grouped according to parity (primiparous, biparous, and multiparous); to BCS scale at calving (high—HBCS and normal—NBCS); and to milk yield (high—HP and moderate—MP production). BCS was assessed, and blood samples were collected on −21, −7, 0, 7, 21, and 42 days relative to parturition. The concentrations of non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), cholesterol, glucose, total protein (TP), albumin, total calcium (Ca), phosphorus (P), and magnesium (Mg); and activities of aspartate aminotransferase and gamma-glutamyltransferase were measured. Data were analyzed by two-way repeated measures ANOVA. The frequencies of high lipomobilization, subclinical ketosis, subclinical hypocalcemia (SCH), and the occurrence of diseases during early lactation were established. Regardless of grouping, NEFA, BHB, and cholesterol increased during early lactation; glucose showed higher values at calving; TP and albumin were higher at 21 and 42 DIM; and Ca, P, and Mg were lower at calving. Parity had little effect on the metabolic profile, HBCS did not differ from NBCS cows, and HP did not differ from MP cows in most metabolites. High lipomobilization in early lactation and SCH at calving were the most common imbalances but were not related to postpartum diseases. High-yielding Gyr cows have a balanced metabolic profile during the transition period, with few biologically relevant effects of parity, BCS at parturition, or milk yielded.

Abstract:Fifty Holstein cows (2.6 ± 1.0 lactations) in early lactation (52.6 ± 16.4 days in milk) calved in autumn and producing 34.3 (±4.4) kg milk per cow ·day -1 were randomly assigned for 27 weeks to one of two treatments (feeding systems) in 10 groups (pens) of 5 animals each (5 groups/treatment). The confined treatment was a total mixed ration (TMR_100) whereas the supplemented grazing system was defined by a partially mixed ration (PMR_75) with 75% TMR and 25% oat pasture ( Avena sativa L.) during the autum-winter (Period I) and alfalfa pasture ( Medicago sativa L.) plus concentrate (7.0 kg per cow ·day -1) (P + C) during the next spring (Period II). Milk production was daily and individually recorded throughout the trial while the chemical composition of milk was measured on individual samples every 2 weeks. Live weight (LW) and body condition score (BCS) using a scale of 1 to 5 were recorded every 3 weeks after the morning milking. Simultaneously, blood samples were taken for determinations of glucose, urea, non-esterified fatty acids (NEFA), insulin, somatotrophin (GH) and somatomedin C (IGF-I). Total DM intake was measured using the difference method four times per repetition in the months of July, August, September and October. The experimental herd was inseminated at fixed time at the start of the service (May-July) and on the return with estrus detection. The diagnosis of pregnancy was made by transrectal ultrasonography at 30 and 60 days after the start of the service. The productive data were analyzed according to a model with repeated observations in time adjusted by covariate with 2 periods and using the animal as the experimental unit. Data of DM intake were analyzed using ANOVA for 2 treatments with 5 repetitions. Cows from the TMR_100 group produced more milk (kg ·cow -1 ·day -1) during Period I (33.7 vs. 32.3) and Period II (28.8 vs. 27.8) (P < 0.01). Yields were also higher (P < 0.05) in TMR_100 for milk fat (kg ·cow -1 ·day -1) in Periods I (1.33 vs. 1.24) and II (1.10 vs. 0.92) and for milk protein (1.19 vs. 1.13 and 0.99 vs. 0.93). The lower energy intake of grazing cows (45.0 vs. 43.1 and 40.9 vs. 38.9 Mcal per cow · day -1 for TMR_100 vs. PMR_75 and TMR_100 vs. P + C) coupled to the extra energy cost associated with grazing activity would explain the difference in milk production. Milk protein content (%) resulted higher (P < 0.05) in TMR_100 over the two experimental periods (3.53 vs. 3.47 and 3.42 vs. 3.31) while milk fat content (%) was similar between TMR_100 vs. PMR_75 (3.90) but lower in P + C (3.30) compared to TMR_100 (3.87). The greatest LW gain ( kg · cow -1 · day -1) in the cows of the TMR_100 group in both periods (0.69 vs. 0.34 and 0.49 vs. -0.22) was consistent with the increase in the plasmatic levels of glucose and IGF-I and the reduction in the circulating levels of GH, but not with the lack of increases in plasma insulin concentration or decreases in circulating levels of NEFA, parameters that were not affected by treatments. Despite the positive effects of TMR_100 on parameters linked to energy balance, no significant differences were detected in any of the reproductive parameters evaluated and the final rate of pregnancy was 80.0 and 91.3% for the confined and grazing system with supplementation respectively. Free milk liters over feeding costs were higher in the grazing system with supplementation compared to the confined system (18.5 vs. 15.5 l, respectively). The results indicate that the grazing system with supplementation can be economically competitive compared to the confined system of milk production.

Abstract:The objectives of this study were to evaluate the effects of dietary supplementation of ruminally protected lysine (RPL), or methionine (RPM), and their combination (RPML) on the production efficiency of transition cows. A total of 120 pre-partum multiparous Holstein cows were assigned to four treatments based on previous lactation milk production, days (d) of pregnancy, lactation, and body condition score (BCS). Cows were fed a basal diet [pre-calving: 1.53 Mcal/kg dry matter (DM) and post-calving: 1.70 Mcal/kg DM] with or without supplemental ruminally protected amino acids (RPAA). Treatments were the basal diets without supplemental amino acids (CONTROL, n = 30), with supplemental methionine (RPM, pre-calving at 0.16% of DM and post-calving at 0.12% of DM, n = 30), with supplemental lysine (RPL, pre-calving at 0.33% of DM and post-calving at 0.24% DM, n = 30), and the combination (RPML, pre-calving at 0.16% RPM + 0.33% RPL of DM and post-calving at 0.12% RPM + 0.24 % RPL DM, n = 30). The dietary content of lysine was balanced to be within 6.157.2% metabolizable protein (MP)–lysine and that of methionine was balanced within 2.1–2.35% MP-methionine. Dry matter intake (DMI) was measured daily. Milk samples were taken on d 7, 14, and 21 days relative to calving (DRC), and milk yields were measured daily. Blood samples were taken on d −21, −14, −7 before expected calving and d 0, 7, 14, and 21 DRC. Data were analyzed using SAS software. There were significant Trt × time interactions ( P < 0.01) for DMI pre- and post-calving period. The CON cows had lower DMI than RPM, RPL, and RPML, both pre-calving ( P < 0.01) and post-calving periods ( P < 0.01). Energy-corrected milk ( P < 0.01), milk fat ( P < 0.01), protein ( P = 0.02), and lactose ( P < 0.01) percentage levels were greater for RPM, RPL, and RPML cows compared to CON. Supplementing RPAA assisted in maintaining BCS post-calving than CON ( P < 0.01). Blood concentrations of β-hydroxybutyrate decreased with RPM or RPL or the combination pre-calving ( P < 0.01) and tended to decrease post-calving ( P = 0.10). These results demonstrated that feeding RPL and RPM improved DMI and milk production efficiency, maintained BCS, and reduced β-hydroxybutyrate concentrations of transition cows.

Impact of postpartum milking frequency on the immune system and the blood metabolite concentration of dairy cows

Abstract:The transition from pregnancy to lactation is marked by metabolic, hormonal, and immunological changes that have an impact on the incidence of infectious and metabolic diseases. The aim of this study was to evaluate the effect on immune function and blood metabolite concentration of limiting milk production in early lactation to reduce negative energy balance. Twenty-two multiparous Holstein cows were milked either once a day (1x) or twice a day (2x) for the first week postpartum. All cows were milked twice daily for the rest of lactation. Blood concentrations of nonesterified fatty acids (NEFA), beta-hydroxybutyric acid (BHBA), calcium, bilirubin, urea, phosphorus, glucose, leptin, stanniocalcin-1, and 17beta-estradiol were determined in samples collected from 5 wk before scheduled calving to 5 wk after calving. Polymorphonuclear leukocytes (PMNL) were isolated from blood to conduct assays for chemotaxis, phagocytosis, and respiratory burst. Peripheral blood mononuclear cells (PBMC) were isolated to evaluate lymphocyte proliferation and cytokine production (tumor necrosis factor-alpha, IL-4, and interferon-gamma). Cows milked 1x produced 31% less milk than cows milked 2x during the first week of lactation. Over the following 13 wk of lactation, the milk production of cows milked 1x during the first week was 8.1% lower than for cows milked 2x. However, because the percentages of fat and protein were greater in the milk from 1x cows, the yields of milk components and energy-corrected milk were similar. Calving induced an increase in the concentrations of NEFA, BHBA, urea, and bilirubin. The increases in levels of NEFA and BHBA were greater in cows milked 2x than in cows milked 1x. During the same period, the serum glucose concentration decreased but remained greater in cows milked 1x. Serum calcium on d 4 and serum phosphorus on d 4 and 5 were greater in cows milked 1x. The differences between the 2 groups persisted beyond treatment until postpartum d 24 for NEFA and glucose and until postpartum d 14 for BHBA. After calving, the concentrations of leptin and stanniocalcin-1 decreased. During the first week postpartum, the decrease of leptin was less marked in cows milked 1x. The immune functions of PBMC and PMNL isolated from experimental cows and incubated using a standard medium did not show clear-cut peripartum immunosuppression. These variables were not significantly affected by the treatments, with the exception of interferon-gamma secretion, which was greater on d 5 and 14 in cows milked 1x. In conclusion, limiting milk production in early lactation had positive effects on metabolite concentration, but larger studies are necessary to establish if this could reduce disease incidence.

Effect of peripartum dietary energy supplementation of dairy cows on metabolites, liver function and reproductive variables

Abstract:Multiparous Holstein cows (n=58) were used to study the effects of peripartum dietary supplementation on metabolic status, liver function and reproduction variables. Diets for cows were as follows: (a) no supplementation (CTL), (b) prilled fatty acids as 1.9% of DM (PrFA), (c) calcium salts of long chain n-6 fatty acids as 2.24% of DM (CaLFA) or (d) daily topdressing with 769 g of 65% propylene glycol (PGLY). Supplements were fed during the last 21 days before expected calving except for PGLY that continued until 21 days after parturition. Ovarian activity was monitored by transrectal ultrasonography and days to first ovulation were recorded. Liver biopsies were obtained on day 8 and 21 postpartum and analyzed for triglyceride content and mRNA expression of pyruvate carboxylase, cytosolic phosphoenolpyruvate carboxykinase, carnitine palmytoyltransferase 1A, and peroxisome proliferator-activated receptor-alpha. At 71 days following parturition, stage of ovarian cycles was synchronized and at day15 of the cycle oxytocin was injected i.v., blood samples were obtained at frequent intervals, and analyzed for 13,14 dihydro, 15-keto PGF(2alpha) (PGFM). Milk production and milk components were not different among treatment groups. Cows in PGLY gained body condition score (BCS) prepartum and net energy balance prepartum tended to be greater, but was not different postpartum from other groups. PGLY supplementation increased plasma insulin concentration prepartum, but not during the postpartum period. No significant differences were observed in plasma concentrations of glucose, NEFA, and insulin-like growth factor or hepatic triglyceride content, but all supplements tended to decrease beta hydroxybutyrate postpartum compared to CTL cows. Abundance of mRNA of gluconeogenic and lipid oxidation genes was not different among treatment groups. Days to first ovulation and uterine PGF(2alpha) production in response to an oxytocin treatment were not significantly different among treatment groups. Peripartum supplementation did not result in the substantial improvement of metabolic profile in early lactation nor significantly affect days to first ovulation and PGFM response to an oxytocin treatment.
Maternal pre- and postpartum supplementation of a Bacillus-based DFM enhanced cow and calf performance

Abstract:This study evaluated the effects of maternal supplementation of a Bacillus-based direct-fed microbial (DFM) on the physiology and growth performance of Bos indicus-influenced cow-calf pairs. On day 0 (~139 d before expected calving date), 72 fall-calving, Brangus crossbred beef heifers (20 to 22 mo of age) pregnant with first offspring were stratified by their initial body weight (BW; 431 ± 31 kg) and body condition score (BCS; 6.0 ± 0.36; scale 1 to 9), and randomly allocated into 1 of 12 bahiagrass pastures (1 ha and six heifers per pasture). Treatments were randomly assigned to pastures (six pastures per treatment) and consisted of heifers supplemented with 1 kg/d of soybean hulls (dry matter, DM) that was added (BAC) or not (CON) with DFM containing Bacillus subtilis and B. licheniformis (Bovacillus; Chr. Hansen A/S, Hørsholm, Denmark). Treatments were provided from days 0 to 242 (139 ± 4 d prepartum to 104 ± 4 d postpartum). Calves were weaned on day 242 (96 ± 30 d of age) and then allocated into 1 of 16 drylot pens and fed the same concentrate at 3.25% of BW (DM) until day 319. Maternal treatment effects were not detected (P ≥ 0.29) for herbage allowance and forage chemical composition. Heifer BCS on days 39 and 63 tended (P ≤ 0.09) to be greater for BAC vs. CON heifers, whereas heifer BCS on day 91 was greater (P = 0.01) for BAC vs. CON heifers. Heifer BCS did not differ (P ≥ 0.20) between treatments on days 179 and 242. Plasma glucose concentration did not differ from days 0 to 63 (P ≥ 0.14) but were greater (P < 0.01) on day 179 and tended (P = 0.09) to be greater on day 242 for BAC vs. CON heifers. Calf BW at birth, ADG from birth to weaning, and BW at weaning did not differ (P ≥ 0.19) between treatments, but calf BW at drylot exit (day 319) was greater (P = 0.05) for BAC vs. CON calves. Maternal treatment effects were not detected (P ≥ 0.42) for calf serum concentration of IgG at birth and postvaccination plasma concentrations of glucose, cortisol, and haptoglobin. Serum titers against bovine respiratory syncytial virus (BRSV) were greater (P = 0.04) for BAC vs. CON calves on day 287, whereas seroconversion against parainfluenza-3 virus (PI-3) was greater (P < 0.01) for BAC vs. CON calves on day 271. Thus, maternal supplementation of a Bacillus-based DFM increased prepartum BCS gain and postpartum plasma glucose concentration of heifers and led to positive carryover effects on postweaning BW gain and humoral immune response in their offspring.

Abstract:Thirty-five crossbred Angus heifers (initial BW = 359.5 ± 7.1 kg) were randomly assigned to a 2 × 2 factorial design to evaluate effects of vitamin and mineral supplementation [VMSUP; supplemented (VTM) vs. unsupplemented (NoVTM)] and different rates of gain [GAIN; low gain (LG), 0.28 kg/d, vs. moderate gain (MG), 0.79 kg/d] during the first 83 d of gestation on dam hormone and metabolic status, fetal tissue and organ mass, and concentration of glucose and fructose in fetal fluids. The VMSUP was initiated 71 to 148 d before artificial insemination (AI), allowing time for mineral status of heifers to be altered in advance of breeding. At AI heifers were assigned their GAIN treatment. Heifers received treatments until the time of ovariohysterectomy (d 83 ± 0.27 after AI). Throughout the experiment, serum samples were collected and analyzed for non-esterified fatty acids (NEFA), progesterone (P4), insulin, and insulin-like growth factor 1 (IGF-1). At ovariohysterectomy, gravid reproductive tracts were collected, measurements were taken, samples of allantoic (ALF) and amniotic (AMF) fluids were collected, and fetuses were dissected. By design, MG had greater ADG compared to LG (0.85 vs. 0.34 ± 0.04 kg/d, respectively; p < 0.01). Concentrations of NEFA were greater for LG than MG (p = 0.04) and were affected by a VMSUP × day interaction (p < 0.01), with greater concentrations for NoVTM on d 83. Insulin was greater for NoVTM than VTM (p = 0.01). A GAIN × day interaction (p < 0.01) was observed for IGF-1, with greater concentrations for MG on d 83. At d 83, P4 concentrations were greater for MG than LG (GAIN × day, p < 0.01), and MG had greater (p < 0.01) corpus luteum weights versus LG. Even though fetal BW was not affected (p ≥ 0.27), MG fetuses had heavier (p = 0.01) femurs than LG, and VTM fetuses had heavier (p = 0.05) livers than those from NoVTM. Additionally, fetal liver as a percentage of BW was greater in fetuses from VTM (P = 0.05; 3.96 ± 0.06% BW) than NoVTM (3.79 ± 0.06% BW), and from LG (p = 0.04; 3.96 ± 0.06% BW) than MG (3.78 ± 0.06% BW). A VMSUP × GAIN interaction was observed for fetal small intestinal weight (p = 0.03), with VTM-MG being heavier than VTM-LG. Therefore, replacement heifer nutrition during early gestation can alter the development of organs that are relevant for future offspring performance. These data imply that compensatory mechanisms are in place in the developing conceptus that can alter the growth rate of key metabolic organs possibly in an attempt to increase or decrease energy utilization.

Abstract:Reduced liver function, increased oxidative stress, and inflammation in early lactation negatively impact lactation performance and health of fresh cows. Previous findings from our group demonstrated that branched-chain amino acids (BCAA) infusion improved lactation performance and branched-chain ketoacids (BCKA) infusion decreased liver triglyceride (TG) in fresh cows. The objectives of this study were to determine the effect of BCAA and BCKA on blood and liver biomarkers of liver function, oxidative stress, and inflammation as well as expression of genes regulating inflammation and antioxidant metabolism in the liver. Thirty multiparous Holstein cows were used in a randomized block design receiving continuous abomasal infusion for 21 d after parturition. Treatments (10 cows each) were control (CON), cows abomasally infused with 0.9% saline; BCA, cows abomasally infused with BCAA (67 g valine, 50 g leucine, and 34 g isoleucine); and BCK, cows abomasally infused with BCKA (77 g ketovaline, 57 g ketoleucine, and 39 g ketoisoleucine). All cows were randomly assigned to treatments after parturition and received the same diet throughout the experimental period. Blood was collected at 3, 7, 14, and 21 d postpartum for liver function, oxidative stress, and inflammation biomarker profiling. Liver was also harvested on 7, 14, and 21 d postpartum for quantification of glutathione, protein carbonylation, and expression of genes. ANOVA was conducted for all data using PROC GLIMMIX in SAS. No treatment differences were observed for liver function biomarkers (bilirubin, gamma-glutamyl transferase, and aspartate aminotransferase). Cows receiving BCAA had lower blood NO2- and NO3- concentrations compared with CON. A tendency for lower advanced oxidized protein products was also observed in BCA cows compared with CON. Additionally, on d 7, BCA cows had lower protein carbonylation in the liver compared with CON. In contrast, BCK cows had higher plasma thiol and albumin, as well as liver reduced and total glutathione compared with CON cows. Compared with CON, BCK cows had higher expression glutathione reductase in the liver. Overall, these results suggest favorable alterations in oxidative stress and inflammation status in fresh cows receiving BCAA or BCKA infusion during the first 3 weeks of lactation, which likely contributed to previously-observed changes in lactation performance and liver TG concentrations. Future work is required to evaluate the interrelated metabolism of BCAA and BCKA to better understand their effects on oxidative and immune metabolism.
Effect of close-up metabolizable protein supply on colostrum yield, composition, and immunoglobulin G concentration and associations with prepartum metabolic indicators of Holstein cows

Abstract:The prepartum diet as well as individual metabolic status of the cow influences colostrum parameters. The objectives of this study were to 1) investigate the effect of increasing prepartum dietary MP supply on colostrum yield, composition, and immunoglobulin G (IgG) concentration, and 2) identify prepartum metabolic indicators associated with these outcomes. Multiparous Holstein cows (n = 96) were blocked by expected calving date and randomly assigned to 1 of 2 prepartum diets formulated to contain a control (CON; 85 g of MP/kg DM; 1,175 g of MP/d) or high (HI; 113 g of MP/kg DM; 1,603 g of MP/d) level of MP starting at 28 d before expected calving. Both prepartum diets were formulated to supply Met and Lys at an equal amount of 1.24 and 3.84 g/Mcal of metabolizable energy (ME), respectively. Metabolic indicators were determined in serum (albumin, glutamate dehydrogenase, cholesterol, aspartate transaminase, total protein, total bilirubin, and IgG) or plasma (Ca, glucose, fatty acids, BHB, and urea nitrogen) twice weekly in a subset of cows (n = 60). Colostrum was harvested at 3.6 ± 2.4 h from calving and yield as well as concentrations of IgG, fat, protein, and Ca were determined. Cows were retrospectively grouped based on the typical volume of colostrum needed for 2 colostrum meals (<6 or ≥ 6 kg), IgG concentration (<100 or ≥ 100 g/L), as well as the median concentrations of fat (<4.4 or ≥ 4.4%), protein (<16.5 or ≥ 16.5%), Ca (<0.21 or ≥ 0.21%), and total colostrum ME (<8.65 or ≥ 8.65 Mcal). Data were analyzed using mixed effects ANOVA, with repeated measures where applicable. Feeding HI tended to increase colostrum yield in cows entering parity 2 (9.4 vs. 7.2 ± 0.9 kg), but treatment did not affect yield from cows entering parity ≥3 (5.1 vs. 6.4 ± 1.0 kg). Supply of MP did not affect concentrations of IgG, fat, protein, or Ca. Cows that produced ≥ 6 kg vs. those producing <6 kg of colostrum had lower plasma concentrations of glucose. Metabolic indicators were not associated with IgG group. Colostrum fat ≥4.4% was associated with cows having lower prepartum concentrations of glucose, total protein, albumin, and aspartate transaminase activity. Colostrum protein ≥ 16.5% was associated with lower circulating serum IgG and elevated cholesterol. Elevated glucose as well as lower cholesterol and BHB concentrations were associated with colostrum Ca ≥ 0.21%. Further, higher albumin and fatty acids as well as lower glucose concentrations were associated with a greater colostrum energy output. In conclusion, increasing prepartum MP supply tended to increase colostrum yield in cows entering parity 2, but did not affect the composition or IgG concentration. The observed associations between metabolic indicators and colostrum parameters suggest that slight adjustment in metabolism during late gestation might be necessary to support colostrogenesis, but the causality of these relationships should be considered.

Abstract:Directing protein and energy sources towards lactation is crucial to optimise milk production in sows but how this influences colostrum yield (CY) remains unknown. The aim of this study was to identify associations between CY and the sow's use of nutrient resources. We included 37 sows in the study that were all housed, fed and managed similarly. Parity, back fat change (ΔBF), CY and performance parameters were measured. We obtained sow serum samples 3 to 4 days before farrowing and at D1 of lactation following overnight fasting. These were analysed for non-esterified fatty acids (NEFA), urea, creatinine, (iso)butyrylcarnitine (C4) and immunoglobulins G (IgG) and A (IgA). The colostrum samples collected 3, 6 and 24 h after the birth of the first piglet were analysed for their nutrient and immunoglobulins content. The technical parameters associated with CY were parity group (a; parities 1 to 3=value 0 v. parities 4 to 7=value 1) and ΔBF D85-D109 of gestation (mm) (b): CY (g)=4290-842a-113b. (R 2=0.41, P<0.001). The gestation length (P<0.001) and the ΔBF between D109 and D1 of lactation (P=0.050) were identified as possible underlying factors of the parity group. The metabolic parameters associated with CY were C4 at 3 to 4 days before farrowing (a), and 10logC4 (b) and 10logNEFA (c) at D1 of lactation: CY (g)=3582-1604a+1007b-922c (R 2=0.39, P=0.001). The colostrum composition was independent of CY. The negative association between CY and ΔBF D85-D109 of gestation could not be further explained based on our data. Sows that were catabolic 1 week prior to farrowing seemed unable to produce colostrum to their full potential. This was especially the case for sows with parities 4 to 7, although they had a similar feed intake, litter birth weight and colostrum composition compared with parities 1 to 3 sows. In conclusion, this study showed that parity and the use of body fat and protein reserves during late gestation were associated with CY, indicating that proper management of the sow's body condition during late gestation could optimise the intrinsic capacity of the sow's CY.

Combining rhinoplasty with septal perforation repair.

Abstract:A combined septal perforation repair and rhinoplasty was performed in 80 patients presenting with septal perforations (size 1 to 5 cm) and external nasal deformities. The external rhinoplasty approach was used for all cases and the perforation was repaired using bilateral intranasal mucosal advancement flaps with a connective tissue interposition graft in between. Complete closure of the perforation was achieved in 90% of perforations of size up to 3.5 cm and in only 70% of perforations that were larger than 3.5 cm. Cosmetically, 95% were very satisfied with their aesthetic result. The external rhinoplasty approach proved to be very helpful in the process of septal perforation repair especially in large and posteriorly located perforations and in cases where the caudal septal cartilage was previously resected. Our results show that septal perforation repair can be safely combined with rhinoplasty and that some of the routine rhinoplasty maneuvers, such as medial osteotomies and dorsal lowering, could even facilitate the process of septal perforation repair.
Metabolic and hormonal profiles and productive performance in primiparous and multiparous cows grazing different forage allowance in late gestation

Abstract:The aim of this study was to evaluate two levels of forage allowance (FA) during the prepartum period on metabolic, hormonal, productive, and reproductive variables in primiparous (P) and multiparous (M) beef cows. Six weeks before calving, 40 P and 42 M cows were assigned to two FA (native pastures) treatments until calving (day 0): High (15 kg DM/100 BW; H) and Low (5 kg DM/100 BW; L). After calving all cows were managed together. High FA cows presented greater intake than L cows, while M tended to have greater intake than P cows. Increased herbage allowance in late gestation in beef cows successfully improved forage intake and energy balance reflected in body condition score and metabolic and endocrine markers. Also, high forage allowance increased milk yield (5.4 vs 4.6 kg/d, P < 0.05) and calves daily gain rate during the first two months of age (0.88 vs 0.82 kg/d, P < 0.05) in H respect to L cows. Pregnancy rate was greater in H vs L cows (40 vs 28 %, P < 0,05). The endocrine metabolic response to higher forage allowance depended on parity, as primiparous cows showed a greater imbalance around calving. Moreover, most primiparous cows were not pregnant at the end of the breeding season, consistent with their prolonged anestrous. Multiparous cows had better productive and reproductive performance than primiparous cows, showing that growing cows have a pronounced negative energy balance reflected in most of the evaluated parameters.

Abstract:Automated milking systems (AMS) are increasingly adopted for dairy cow production, promoting individualized cow management dependent on factors like lactation stage, age, and productivity. The study objective was to investigate the effects of early lactation milking frequency on cows milked via AMS. Multiparous Holstein cows blocked by parity and due date were randomly assigned to treatments (n = 8 per treatment): three (3X) or six (6X) milkings per day (MPD). The experimental phase (EXP) was defined as 4 to 29 days in milk (DIM). The AMS settings were programed so 3X cows were limited to three MPD while 6X cows were allowed six MPD. Afterwards was the carry over phase (CO) ranging from 30 to 90 DIM; all cows were allowed up to six MPD. Measurements by the AMS included bodyweight, milk yield (MY), and pellet intake. Weekly composite milk samples were analyzed for macronutrient composition and fatty acid (FA) profile. Coccygeal blood was sampled at 3, 8 ± 1, and 13 ± 1 DIM; concentrations of blood plasma analytes were quantified. Greater MPD was achieved for 6X cows versus 3X cows during EXP, but similar during the CO. Daily MY was non-separable during the EXP while 6X cows in their third or greater lactation group (3 + LG) had greater MY than 3X cows of the same LG during the CO. Milk fat content and 4% fat-corrected MY were both greater for 6X, 3 + LG cows during the EXP compared to 3X, 3 + LG cows. Milk FA methyl esters (FAME) proportions were different between MPD groups, with 6X, 3 + LG cows having the lowest short, even-chain FA from de novo or post-absorptive origin. Differences in analytes indicated that 6X, 3 + LG cows experienced metabolic stress and incorporated greater FA from adipose tissue. Greater early lactation MPD in AMS may shift cow nutrient partitioning to support greater production in 3+ parity cows.

Abstract:Background: Bovine mastitis causes major economic losses for milk producers by reducing the quantity and the quality of the milk or even leading to the complete loss of the mammary gland secretory capacity. During the transition period, dairy cows are susceptible to infectious diseases; therefore, markers that allow early identification of cows in higher risk of developing diseases are especially useful at this time. Therefore, the aim of this study was to evaluate serum markers in the pre and postpartum of multiparous dairy cows with clinical mastitis and with health condition in the postpartum period in a semi-extensive management system.Materials, Methods & Results: Thirty-Six Holstein cows were monitored daily during milking until 59 days postpartum and were categorized according to the pre-milking strip cup test into clinical mastitis (mastitis group (MG)) and absence of symptoms (control group (CG)) that were negative to the test, representing the health cows. All cows were reared as one group and maintained in a semi-extensive pasture-based system. Blood samples were collected weekly after morning milking via venipuncture of the coccinea vein into tubes without anticoagulant and grouped for prepartum (-21 to 0 days from calving), early postpartum (0 to 30 days from calving), and late postpartum (30 to 59 days from calving) periods. Milk production was recorded daily. The serum markers albumin, aspartate aminotransferase (AST), phosphorus, gammaglutamyltransferase (GGT) and non-esterified fatty acids (NEFA) were measured. Statistical analyses were performed using SAS®. The cases of clinical mastitis occurred on average at 37.2 ± 4.9 days postpartum. Health cows (CG) had higher milk production compared to the mastitis group (MG) only in the late postpartum period (P 0.05). In the early postpartum period the AST activity was higher in CG than in MG (P = 0.02). The GGT enzyme tended to be more concentrated in the CG than in the MG during the early (P = 0.06) and late (P = 0.08) postpartum periods. Late postpartum phosphorus concentration was lower for MG than CG (P = 0.04). In the prepartum and early postpartum periods, there was no difference among groups for phosphorus concentration (P > 0.05).Discussion: A decrease in milk production in MG compared to CG observed in late postpartum period was due to the incidence of mastitis observed around 37 days postpartum. Cows that presented clinical mastitis in the postpartum period did not differ in the blood concentration of NEFA in the prepartum period. In the late postpartum period higher concentration of phosphorus was observed in the CG than in MG, indicating that animals affected by mastitis may be in the weakest energy status. Regarding liver health, the concentration of AST was higher in the recent postpartum period for CG, in disagreement with previous studies that related AST to tissue injury caused by mastitis. The GGT enzyme tended to had higher concentrations in CG than MG during the whole postpartum period and may be related to increased hepatic metabolism due to higher production. There were no changes in albumin levels among healthy and mastitis cows, indicating that this marker can not be used to predict clinical mastitis. There were no metabolic alterations in the prepartum period related to the occurrence of postpartum mastitis in multiparous cows in a semi-extensive management system.
