Abstract:Soil organic matter (SOM) stocks contain nearly three times as much carbon (C) as the atmosphere and changes in soil C stocks may have a major impact on future atmospheric carbon dioxide concentrations and climate. Over the past two decades, much research has been devoted to examining the influence of warming on SOM decomposition in topsoil. Most SOM, however, is old and stored in subsoil. The fate of subsoil SOM under future warming remains highly uncertain. Here, by combining a long-term field warming experiment and a meta-analysis study, we showed that warming significantly increased SOM decomposition in subsoil. We also showed that a decade of warming promoted decomposition of subsoil SOM with turnover times of decades to millennia in a tall grass prairie and this effect was largely associated with shifts in the functional gene structure of microbial communities. By coupling stable isotope probing with metagenomics, we found that microbial communities in warmed soils possessed a higher relative abundance of key functional genes involved in the degradation of organic materials with varying recalcitrance than those in control soils. These findings suggest warming may considerably alter the stability of the vast pool of old SOM in subsoil, contributing to the long-term positive feedback between the C cycle and climate.
Subsoil SOC Increased by High C:N Ratio Straw Application with Optimized Nitrogen Supplementation

Abstract:The application of straw and nutrients to agricultural subsoils is of significant interest to enhance carbon (C) sequestration and soil fertility. However, little research has explored the effect straw application on microbial stoichiometry, soil organic carbon (SOC) mineralization and accumulation and their relationships, in subsoil. In order to address these knowledge gaps, we examined the soil characteristics (organic carbon mineralization, available nutrient contents and microbial stoichiometric ratio) under straw addition (maize leaf and stem) with and without nitrogen (N) supplementation (no addition, 30 mg N kg(-1) dry soil(-1), 60 mg N kg(-1) dry soil(-1)) during an 80-day incubation experiment. The microbial stoichiometric ratio imbalance (C:N and C:Phosphorus (P)) and extracellular enzyme stoichiometry were measured as indicators of the systematic relationship between soil resource availability and the mineralization of organic carbon. Our study demonstrated that the addition of straw significantly enhanced CO2 emissions and led to an increase in the C:N imbalance, while simultaneously decreasing microbial carbon use efficiency (CUE). In addition, stem addition showed 5.6% lower CUE, but 8.2% higher SOC compared with leaf addition. We also found that nitrogen addition to subsoil alleviated microbial nitrogen limitation. 60 mg N kg(-1) dry soil rates of nitrogen application had a positive effect on reducing C:N imbalance and promoting the accumulation of SOC. Extracellular enzyme activity and microbial stoichiometric ratio were the most important controlling factors of SOC mineralization and microbial CUE, respectively. In conclusion, the application of straw alongside N to balance stoichiometric ratios can significantly increase SOC content, indicating the potential for carbon sequestration in agricultural subsoils.

Abstract:Chemical fertilization in excess and warming disrupt the soil microbes and alter resource stoichiometry, particularly in intensive vegetable soils, while the effects of these variables on the temperature sensitivity of soil organic carbon (SOC) decomposition (Q10) and SOC stability remain elusive. Thus, we collected six long-term vegetable soils along a climatic gradient to examine the microbial mechanisms and resource stoichiometry effects on fluctuations in Q10 and SOC stability induced by warming and fertilization from vegetable soils. Our results showed that the SOC decomposition was dominated by microbes and regulated by stoichiometry. Compared to cold sites, higher Q10 of SOC decomposition was observed in warm sites, accompanied by lower enzyme activities, microbial CUE, and C:N ratio. In this context, warming reduced SOC stability as evidenced by up to 31.8% greater Q10 (1.45) at warm sites than at cold sites (1.10) owing to less richness of microbial communities and lower microbial CUE. The relatively lower pH and labile organic C value restricted the development of microbial richness, and decreased C- and N-related enzyme activities and a lower C:N ratio resulted in microbial CUE reduction. Additionally, N fertilization altered the C:N imbalance and enhanced SOC stability in vegetable soils, exhibiting an increase of Q10 values, particularly of great importance in warm sites. Collectively, our findings emphasize the importance of the microbial mechanism and resource stoichiometry in predicting variations in Q10 and fluctuations in SOC stability, and provide theoretical advice on improving management policies in the context of warming and fertilization from vegetable soils.

Abstract:The turnover of soil organic carbon (SOC) in cropland plays an important role in terrestrial carbon cycling, but little is known about the temperature sensitivity ( Q 10 ) of SOC decomposition below the topsoil layer of arable soil. Here, samples of topsoil (0–20 cm) and subsoil (20–40 cm) layers were obtained from paddy fields and upland croplands in two regions of China. Using a sequential temperature changing method, soil respiration rates were calculated at different temperatures (8 °C to 28 °C) and fitted to an exponential equation to estimate Q 10 values. The average SOC decomposition rate was 59% to 282% higher in the topsoil than in the subsoil layer because of higher labile carbon levels in the topsoil. However, Q 10 values in the topsoil layer (5.29 ± 1.47) were significantly lower than those in the subsoil layer (7.52 ± 1.84). The pattern of Q 10 values between the topsoil and subsoil was significantly negative to labile carbon content, which is consistent with the carbon quality-temperature hypothesis. These results suggest that the high temperature sensitivity of SOC decomposition in the subsoil layer needs to be considered in soil C models to better predict the responses of agricultural SOC pools to global warming.

Abstract:The decomposition of soil organic carbon (SOC) is intrinsically sensitive to temperature. However, the degree to which the temperature sensitivity of SOC decomposition (as often measured in Q 10 value) varies with soil depth and labile substrate availability remain unclear. This study explores (1) how the Q 10 of SOC decomposition changes with increasing soil depth, and (2) how increasing labile substrate availability affects the Q 10 at different soil depths. We measured soil CO 2 production at four temperatures (6, 14, 22 and 30 °C) using an infrared CO 2 analyzer. Treatments included four soil depths (0–20, 20–40, 40–60 and 60–80 cm), four sites (farm, redwood forest, ungrazed and grazed grassland), and two levels of labile substrate availability (ambient and saturated by adding glucose solution). We found that Q 10 values at ambient substrate availability decreased with increasing soil depth, from 2.0–2.4 in 0–20 cm to 1.5–1.8 in 60–80 cm. Moreover, saturated labile substrate availability led to higher Q 10 in most soil layers, and the increase in Q 10 due to labile substrate addition was larger in subsurface soils (20–80 cm) than in surface soils (0–20 cm). Further analysis showed that microbial biomass carbon (MBC) and SOC best explained the variation in Q 10 at ambient substrate availability across ecosystems and depths (R 2 = 0.37, P < 0.001), and MBC best explained the variation in the change of Q 10 between control and glucose addition treatment (R 2 = 0.14, P = 0.003). Overall, these results indicate that labile substrate limitation of the temperature sensitivity of SOC decomposition, as previously shown in surface soils, is even stronger for subsoils. Understanding processes controlling the labile substrate availability (e.g., with rising atmospheric CO 2 concentration and land use change) should advance our prediction of the fate of subsoil SOC in a warmer world.

Abstract:Information about the temperature dependence of soil organic carbon (SOC) mineralization is important to predict the possible carbon (C) release from the terrestrial ecosystem facing global warming. However, the way in which SOC quality and mineral protection of SOC affects mineralization rates and their response to increasing temperature remains poorly understood. Here, we focused on investigating thermal stability and chemical composition of soils which differed greatly with the type and extent of organo-mineral associations, and revealing their implications for SOC mineralization. Chemical composition and thermal stability of SOC were analyzed with 13C nuclear magnetic resonance spectroscopy (13C NMR) and simultaneous thermogravimeteric analysis and differential scanning calorimetry (TG-DSC), respectively. Soil mineralization and its temperature sensitivity (Q10) were assessed by incubating soils at 15 and 25 degrees C for 90 days.Our results showed that SOC of subsoil was more biologically stable, but exhibited higher temperature sensitivity than those of topsoil. Significant differences in chemical composition and thermal stability of SOC were detected across soils developed from four parent materials but not between two soil layers, and indicators obtained from the two techniques showed apparent associations with depth-specific SOC mineralization rates. Increase in SOC mineralization rates at two incubation temperatures and two soil layers were observed when the soils were characterized by higher percentage of labile SOC fractions (i.e., O-alkyl C and organic carbon oxidized during exothermic reactions). The Q10 values of topsoil were positively and mostly related to the percentage of Alkyl C. In addition to notable relationships with chemical composition, the Q10 values of subsoil were positively correlated with Exo1/Exo2 ratios and Energy density values, and negatively correlated with TG-50. It was interesting to note that thermal stability was possibly as a function of soil texture and total oxides. Results from VPA analysis further revealed that SOC mineralization rates at two soil layers and Q10 of topsoil were mainly driven by SOC quality, while it was related to both SOC quality and chemical protection for subsoil, implying that factors controlling the temperature sensitivity of SOC mineralization were depth-specific. Altogether, substrate accessibility as a function of chemical composition and chemical protection would play vital role in determining the feedbacks of the subsoil SOC pool to the future climate warming.
Microbial community mediated response of organic carbon mineralization to labile carbon and nitrogen addition in topsoil and subsoil

Abstract:Over 50 % of soil carbon (C) is stored in subsoil (below 30 cm). Inputs of labile C and nutrients can stimulate soil organic carbon (SOC) mineralization priming effect (PE) and subsequently affect subsoil C dynamics. However, little is known about the magnitude and mechanism of the PE occurring in subsoil, which complicates the prediction of subsoil C dynamics. Using a lab incubation experiment, the effects of glucose and nitrogen (N) addition on SOC mineralization were studied for three soil layers (0–10, 10–30, and 30–60 cm) from a subtropical forest. Five glucose (5.16 atom % 13C) levels were applied according to soil microbial biomass in each soil layer. Meanwhile, community-level physiological profiling was conducted to reflect microbial functional diversity and activity. We found positive PE for all soil layers. The PE magnitude in subsoil was about two times higher than that in topsoil with stronger increase in the microbial activity in mining components of SOC. N addition led to a reduction of 45 % in the PE magnitude in topsoil with relatively lower microbial activity in mining N-containing substrates (amino acids and amines) but caused an increase of 18 % in subsoil PE. Soil C and N availability were associated with microbial functional activity, the shifts of which then mediated the SOC mineralization in the presence of labile C and nutrient. These results suggested that mineralization of subsoil C was more sensitive to labile C and N addition. Any future changes in environmental conditions that affect the input and distribution of labile C and N in soil profiles could affect C dynamics in deep soil.

Abstract:Decadally cycling soil carbon (dSOC) is the main component of the terrestrial soil carbon (C) pool. The response of dSOC to warming largely determines the feedback between climate warming and the C cycle. However, there is a lack of investigations about the effect of warming on the relative change in turnover rate (RCT) of dSOC and annually cycling SOC (aSOC) in dissolved organic carbon (DOC), microbial biomass carbon (MBC) and CO 2 . We clarified this issue by incubating two C 3 -C 4 vegetation switch soils (23 years switch, HA soil and 55 years switch, GG soil) at 20°C and 30°C in the recently improved continuous airflow CO 2 trapping system for 1 year. Warming increased the contribution of dSOC (C 3 -C) by 21c/o (soil HA) and 8c/o (soil GG) in MBC, and 38c/o (soil HA) and 15c/o (soil GG) in CO 2 , while only 2%–3c/o increase in DOC at the final stage of the incubation. Furthermore, warming increased the RCT in MBC and CO 2 by 5.3- and 4.1-fold, respectively, but had no significant influence on the RCT in DOC, indicating that soil microbes may be an important engine to accelerate dSOC-derived CO 2 emission in a warming world.

Abstract:Numerous studies have investigated the impact of nitrogen (N) addition on ecosystem carbon (C) storage and cycling. However, how N addition regulates the dynamics of different soil organic carbon (SOC) fractions and the underlying microbial mechanisms remain unclear. In this study, we assessed microbial controls (through biomass, residues and enzymes) of different SOC fractions (particulate organic carbon, POC and mineral-associated organic carbon, MAOC) in response to six years of N addition (50 kg N ha(-1) yr(-1)) in two temperate forests (Betula platyphylla vs. Quercus wutaishanica) in Northern China. Plant inputs (root biomass and leaf litterfall) and soil chemistry (pH, extractable inorganic N, and exchangeable cations) were unaltered by N addition in both forests. In the Q. wutaishanica forest, microbial biomass, residues, and enzymes were not sensitive to N addition, which may explain the lack of response in SOC and two fractions (POC and MAOC). However, in the B. platyphylla forest, although microbial biomass and enzymes as well as SOC and POC did not significantly change after N addition, both microbial residues (amino sugars) and MAOC significantly increased after N addition. Moreover, there was a strong positive correlation between microbial residues and MAOC pool within or across the Iwo forests. Collectively, these results suggest that the dynamics of microbial residues play a crucial role in controlling the response of mineral-associated SOC to N addition in these two forests. Separating bulk soil into distinct functional pools and considering microbial residues should help reveal the nuanced response of soil C dynamics under N addition. (C) 2020 Elsevier B.V. Enhanced Decomposition of Stable Soil Organic Carbon and Microbial Catabolic Potentials by Long-Term Field Warming

Abstract:Quantifying soil organic carbon (SOC) decomposition under warming is critical to predict carbon-climate feedbacks. According to the substrate regulating principle, SOC decomposition would decrease as labile SOC declines under field warming, but observations of SOC decomposition under warming do not always support this prediction. This discrepancy could result from varying changes in SOC components and soil microbial communities under warming. This study aimed to determine the decomposition of SOC components with different turnover times after subjected to long-term field warming and/or root exclusion to limit C input, and to test whether SOC decomposition is driven by substrate lability under warming. Taking advantage of a 12-year field warming experiment in a prairie, we assessed the decomposition of SOC components by incubating soils from control and warmed plots, with and without root exclusion for 3years. We assayed SOC decomposition from these incubations by combining inverse modeling and microbial functional genes during decomposition with a metagenomic technique (GeoChip). The decomposition of SOC components with turnover times of years and decades, which contributed to 95% of total cumulative CO2 respiration, was greater in soils from warmed plots. But the decomposition of labile SOC was similar in warmed plots compared to the control. The diversity of C-degradation microbial genes generally declined with time during the incubation in all treatments, suggesting shifts of microbial functional groups as substrate composition was changing. Compared to the control, soils from warmed plots showed significant increase in the signal intensities of microbial genes involved in degrading complex organic compounds, implying enhanced potential abilities of microbial catabolism. These are likely responsible for accelerated decomposition of SOC components with slow turnover rates. Overall, the shifted microbial community induced by long-term warming accelerates the decomposition of SOC components with slow turnover rates and thus amplify the positive feedback to climate change.
Warming Reduces Priming Effect of Soil Organic Carbon Decomposition Along a Subtropical Elevation Gradient

Abstract:The priming effects (PEs) of soil organic carbon (SOC) decomposition is a crucial process affecting the C balance of terrestrial ecosystems. However, there is uncertainty about how PEs will respond to climate warming. In this study, we sampled soils along a subtropical elevation gradient in China and conducted a 126‐day lab‐incubation experiment with and without the addition of 13C‐labeled high‐bioavailability glucose or low‐bioavailability lignin. Based on the mean annual temperature (MAT) of each elevation (9.3–16.4°C), a temperature increase of 4°C was used to explore how PEs mediate the decomposition of SOC in response to warming. Our results showed that the magnitude of glucose‐induced PEs (PEglucose) was higher than lignin‐induced PEs (PElignin), with both PEs linearly increasing with MAT. Across the MAT (i.e., elevation) gradient, short‐term warming had a constant magnitude of negative effects on PEglucose, whereas rising MAT exacerbated the negative effects of short‐term warming on PElignin. Moreover, the temperature sensitivity of SOC decomposition decreased after adding glucose and lignin across the MAT gradient, suggesting that fresh C inputs may prime the microbial breakdown of labile SOC under warming. Taken together, warming alleviated SOC loss due to PEs through varying mechanisms depending on substrate bioavailability. Warming mediated the PEglucose by increasing available nitrogen and weakening microbial nitrogen‐mining but inhibited the PElignin by shifting from microbial nitrogen‐mining to microbial co‐metabolization. Our findings highlight the role of warming in regulating the PEs and suggest that incorporating the suppression effect of warming on PEs can contribute to the accurate prediction of soil C dynamics in a warming world.

Physico-chemical Protection, Rather Than Biochemical Composition, Governs the Responses of Soil Organic Carbon Decomposition to Nitrogen Addition in a Temperate Agroecosystem

Abstract:The heterogeneous responses of soil organic carbon (SOC) decomposition in different soil fractions to nitrogen (N) addition remain elusive. In this study, turnover rates of SOC in different aggregate fractions were quantified based on changes in δ13C following the conversion of C3 to C4 vegetation in a temperate agroecosystem. The turnover of both total organic matter and specific organic compound classes within each aggregate fraction was inhibited by N addition. Moreover, the intensity of inhibition increases with decreasing aggregate size and increasing N addition level, but does not vary among chemical compound classes within each aggregate fraction. Overall, the response of SOC decomposition to N addition is dependent on the physico-chemical protection of SOC by aggregates and minerals, rather than the biochemical composition of organic substrates. The results of this study could help to understand the fate of SOC in the context of increasing N deposition.
Inactive and inefficient: Warming and drought effect on microbial carbon processing in alpine grassland at depth

Abstract:Subsoils contain > 50% of soil organic carbon (SOC) globally yet remain under‐investigated in terms of their response to climate changes. Recent evidence suggests that warmer, drier conditions in alpine grasslands induce divergent responses in SOC decomposition and carbon accrual in top‐ vs. subsoils. However, longer‐term effects on microbial activity (i.e. catabolic respiration versus anabolic growth) and belowground carbon cycling are not well understood. Here we utilized a field manipulation experiment on the Qinghai‐Tibetan Plateau and conducted a 110‐day soil incubation with and without 13C‐labeled grass litter to assess microbes' role as both SOC 'decomposers' and 'contributors' in the top‐ (0–10 cm) versus subsoils (30−40 cm) after five years of warming and drought treatments. Microbial mineralization of both SOC and added litter was examined in tandem with potential extracellular enzyme activities, while microbial biomass synthesis and necromass accumulation were analyzed using phospholipid fatty acids and amino sugars coupled with 13C analysis, respectively. We found that warming and, to a lesser extent, drought decreased the ratio of inorganic nitrogen (N) to water extractable organic carbon in the subsoil, intensifying N limitation at depth. Both SOC and litter mineralization was reduced in the subsoil, which may also be related to N limitation, as evidenced by lower hydrolase activity (especially leucine‐aminopeptidase) and reduced microbial efficiency (lower biomass synthesis and necromass accumulation relative to respiration). However, none of these effects were observed in the topsoil, suggesting that soil microbes became inactive and inefficient in subsoil but not topsoil environments. Given increasing belowground productivity in this alpine grassland under warming, both elevated root deposits and diminished microbial activity may contribute to new carbon accrual in the subsoil. However, the sustainability of plant growth and persistence of subsoil SOC pools deserve further investigation on a longer term, given the aggravated N limitation at depth.

Abstract:Soil organic carbon (SOC) is the largest carbon sink in terrestrial ecosystems and plays a critical role in mitigating climate change. Increasing reactive nitrogen (N) in ecosystems caused by anthropogenic N input substantially affects SOC dynamics. However, uncertainties remain concerning the effects of N addition on SOC in both organic and mineral soil layers over time at the global scale. Here, we analyzed a large empirical data set spanning 60 years across 369 sites worldwide to explore the temporal dynamics of SOC to N addition. We found that N addition significantly increased SOC across the globe by 4.2% (2.7–5.8%). SOC increases were amplified from short‐ to long‐term N addition durations in both organic and mineral soil layers. The positive effects of N addition on SOC were independent of ecosystem types, mean annual temperature and precipitation. Our findings suggest that SOC increases largely resulted from the enhanced plant C input to soils coupled with reduced C loss from decomposition and amplification was associated with reduced microbial biomass and respiration under long‐term N addition. Our study suggests that N addition will enhance SOC sequestration over time and contribute to future climate change mitigation.

Abstract:Accurately predicting the feedback mechanisms between forest ecosystem carbon cycling and climate change is crucial for effective climate mitigation. Understanding soil organic carbon (SOC) responses to the combined impacts of plant biomass, litter, and nitrogen deposition, especially regarding temperature sensitivity, is essential but remains poorly understood. We conducted incubation experiments using buried black soil from a cool temperate deciduous broad-leaved forest in Japan, which has high C content and a highly stable molecular structure. The stepwise addition of glucose and a temperature increase from 15 to 35 °C accelerated SOC mineralization by 74.0 mg C kg−1 with a positive priming effect (PE) during the 49-day incubation period, while the simultaneous addition of nitrogen had no significant effect on this phenomenon, with SOC mineralization measured at 75.5 mg C kg−1. Conversely, glucose mineralization was significantly accelerated by 10%, from 241.0 to 261.3 mg C kg−1, by stepwise nitrogen addition and temperature increase. Under the combined impacts, the Q10 value of the soil increased significantly from 1.6 to 2.0 compared to that in the unmodified conditions, primarily due to the stepwise addition of glucose. We also found a strong positive correlation between activation energy (Ea) and Q10. This result strongly supports the carbon quality–temperature (CQT) hypothesis. These results likely stem from interactions between SOC quality and carbon availability, suggesting that, in the future, climate change is likely to have a positive feedback effect, especially on buried black soils.

Abstract:The temperature sensitivity (Q10) of soil organic carbon (SOC) decomposition is an important parameter in the accurate projections of SOC dynamics and its feedback to climate change in terrestrial ecosystems. However, how Q10 responds to N deposition across environmental gradients and the underlying mechanism remain largely unresolved. Here, we conducted a novel incubation experiment with periodically varying temperature based on the mean annual temperature of soil origin sites to elucidate the responses of Q10 to N addition across China. Our results demonstrated that N addition effects (NAEs) on Q10 were negatively related to latitude and strong site‐dependent. Bioclimatic, edaphic, and microbial variables together explained 50.1% of the total variation in NAEs on Q10, but bioclimate (16.0%) had the greater unique explanation than edaphic (11.8%) and microbial properties (6.3%). The response of soil exchangeable Ca2+ to N addition was a predictive power for NAEs on Q10, contributing 7.2% relative importance of regulating this variation. Furthermore, arbuscular mycorrhizal fungi indicated by Glomeromycota were the best microbial predictor and contributed 10.9% relative importance of the variation regulating NAEs on Q10. Overall, our results suggest that increasing N addition will increase the sensitivity of SOC decomposition to global warming and highlight the importance of bioclimate, exchangeable Ca2+ and arbuscular mycorrhizal fungi in predicting the response of Q10 to N deposition in natural terrestrial ecosystems. The biogeographic variation in response of Q10 to N deposition should be considered in carbon‐climate models to decrease the prediction uncertainties of SOC dynamics and its feedback to global warming.

Abstract:Plant residue input plays a vital role in cropland carbon (C) balance, which can be greatly affected by climate change. Our knowledge of the effect of plant residue on soil respiration is crucial for evaluating C exchange between the atmosphere and terrestrial ecosystems, but large uncertainties remain in the effect of plant input on the temperature sensitivity (Q 10 ) of soil organic C (SOC) decomposition in cropland ecosystems. Here, soils were sampled from two bare fallow plots (including a Mollisol and an Alfisol with different climate and land-use history) and their adjacent old field plots, and were incubated at 10 °C and 20 °C for 815 days. The 'equal-time', 'equal-C', 'one-pool model' and 'two-pool model' methods were used to evaluate the Q 10 of SOC decomposition. Results indicated that in the early stage of incubation, exclusion of plant input increased the Q 10 (Alfisol) or had minor effect on the Q 10 (Mollisol), which may be related to the interactions between substrate quality decrease (increase of chemical stability) and clay mineral suppression on microbial decomposition. However, in the later stage of incubation, plant residue removal decreased the Q 10 values in both soils possibly due to the limitation of substrate availability on microbial decomposition. Overall, the role of plant input in the temperature sensitivity of SOC decomposition should be considered when predicting SOC stocks in a future warmer world.

Abstract:The Loess Plateau soil in northwest China originated from wind sediments and is characterized by deep soil profiles and large organic carbon (C) content. Severe soil erosion constantly exposes deep soils to the surface, making the organic C vulnerable to microbial decomposition. Few, however, have so far examined how soil microbial activity and community composition in the deep loess soil respond to perturbations. We examined microbial responses in three layers of a clay‐loam loess (topsoil, 0–20 cm; midsoil, 40–60 cm; subsoil, 80–100 cm) to substrate additions (0.8 g glucose‐C kg −1 soil) under two temperature regimes (25 and 35°C). Soil C:N ratio was significantly larger in the subsoil (20.3) than topsoil (7.4). Glucose addition significantly increased CO 2 efflux during a 30‐day incubation period and the relative magnitude of the increase was four times larger in the subsoil than topsoil. The temperature sensitivity ( Q 10 ) of soil CO 2 efflux increased significantly with soil depth in the absence of glucose addition (i.e., ambient soil), but it decreased under glucose addition. Also, glucose addition significantly increased phenol oxidase and peroxidase activities in the subsoil, which might contribute to the stimulation of microbial CO 2 efflux. Composition of the microbial community was more affected by temperature increase in the topsoil, but more responsive to labile C addition in the subsoil. Together, these results indicated that the composition of soil communities and microbial activities in the topsoil and deep soil responded differently to warming and labile C input. Our findings suggest that organic C in deep loess soils can be highly sensitive to environmental changes, emphasizing the need for more long‐term monitoring and quantitative assessment of organic C release from this important C pool. Highlights Microbial responses to labile C and warming were examined along a Loess Plateau soil profile. Microbial respiration was more responsive to C addition and warming in deep soil than topsoil. Microbial composition and activity were sensitive to temperature in the topsoil but to labile C in the subsoil. Climate change may facilitate CO 2 efflux from deep Loess Plateau soils.

Abstract:Primary forest occupies 34 % of the global forest area and contributes to the mitigation of atmospheric carbon (C) dioxide through C sequestration in vegetation and soil. Environmental changes, including climate warming and increased nitrogen (N) deposition, have been found to affect soil C dynamics. However, there is a major gap in knowledge regarding the response and potential mechanisms of primary forest C cycling to climate warming and increased N deposition. We conducted a three year (2018–2020) field-based soil warming (+3 °C above ambient soil temperature), N addition (50 kg N ha−1 year−1) and their interaction experiment in an over 300-year-old primary forest in subtropical China, and measured soil respiration monthly. In addition, soil enzyme activities and physico-chemical properties were also analyzed. Three years of W treatment did not significantly increase soil respiration, while N treatment significantly decreased it by 24.29 %. Cumulative soil respiration in N and WN treatments decreased by 2.17 and 2.14 Mg C ha−1 year−1, while W treatment increased annual cumulative soil respiration by 0.95 Mg C ha−1 year−1. W treatment indirectly affected soil respiration through its negative impact on C-degrading enzymatic activities and soil moisture. N treatment directly impacted soil respiration and indirectly by effect on soil organic C. These findings demonstrate that soil respiration in the old-growth subtropical forest would cause a minor soil C-climate feedback to 3 °C warming within the first three years, and N deposition may have not strong interaction with warming.
