Abstract:As a new type of transdermal drug delivery system (TDDS), microneedles have garnered significant attention for their noninvasiveness, portability, controllable drug administration, and wide-ranging loading capacity. While initially designed for transdermal administration via the stratum corneum for minimizing adverse reactions associated with systemic administration and enhancing patient compliance, microneedles have recently found applications in a multitude of medical contexts. Hence, their potential in medical applications transcends mere drug delivery. Recent reviews on microneedles have extensively covered various aspects, including needle types, materials, and manufacturing methods, as well as their applications in treating skin diseases. This review aims to provide a comprehensive overview of the diverse designs and applications of microneedles from different new perspectives, which encompass traditional integrated microneedles as well as more intelligent and structurally optimized ones in nowadays. Additionally, this review offers a comprehensive analysis of the material properties utilized in microneedles for different applications, including nondegradable materials and degradable materials. Furthermore, the review presents the wide-ranging applications of these microneedles in different medical scenarios such as regeneration and repair, disease treatment, monitoring, and prevention. Moreover, the discussion of the article highlights the future clinical applications of microneedles and the necessary efforts required to achieve their widespread adoption. This review serves as a valuable reference for bridging the gap between the laboratory setting and the clinical practice in terms of using microneedles in medical scenarios.

Abstract:The microneedles have attracted great interests for a wide range of transdermal biomedical applications, such as biosensing and drug delivery, due to the advantages of being painless, semi-invasive, and sustainable. The ongoing challenges are the materials and fabrication methods of the microneedles in order to obtain a specific shape, configuration and function of the microneedles to achieve a target biomedical application. Here, this review would introduce the types of materials of the microneedles firstly. The hardness, Young's modulus, geometric structure, processability, biocompatibility and degradability of the microneedles are explored as well. Then, the fabrication methods for the solid and hollow microneedles in recent years are reviewed in detail, and the advantages and disadvantages of each process are analyzed and compared. Finally, the biomedical applications of the microneedles are reviewed, including biosensing, drug delivery, body fluid extraction, and nerve stimulation. It is expected that this work provides the fundamental knowledge for developing new microneedle devices, as well as the applications in a variety of biomedical fields.

Abstract:<h2>Abstract</h2><p>The microneedle (MN), a highly efficient and versatile device, has attracted extensive scientific and industrial interests in the past decades due to prominent properties including painless penetration, low cost, excellent therapeutic efficacy, and relative safety. The robust microneedle enabling transdermal delivery has a paramount potential to create advanced functional devices with superior nature for biomedical applications. In this review, a great effort has been made to summarize the advance of microneedles including their materials and latest fabrication method, such as three-dimensional printing (3DP). Importantly, a variety of representative biomedical applications of microneedles such as disease treatment, immunobiological administration, disease diagnosis and cosmetic field, are highlighted in detail. At last, conclusions and future perspectives for development of advanced microneedles in biomedical fields have been discussed systematically. Taken together, as an emerging tool, microneedles have showed profound promise for biomedical applications.</p>

Abstract:Microneedles (MNs) are micro-scale needles used for drug delivery and other targets. Micro-scale size endows them with many advantages over hypodermic needles, including painlessness, minimal invasiveness and convenient operation, but it may also lead to risk of mechanical failures, which should be prevented in the clinical applications of MNs. The objective of this review is mainly to introduce studies on the mechanics problems with respect to MNs. Firstly, the basic knowledge of MNs is introduced in brief, so that readers can understand the basic characteristics of MNs. Secondly, researches on inserting behavior and mechanical performances of MNs are discussed. Thirdly, literatures on the drug delivery and the pain resulted from the insertion of MNs are overviewed. Finally, some bio-microneedles and bio-inspired MNs are introduced.

Abstract:In recent years, transdermal drug delivery based on microneedles (MNs) technology has received extensive attention, which offers a safer and painless alternative to hypodermic needle injections. They can pierce the stratum corneum and deliver drugs to the epidermis and dermis-structures of skin, showing prominent properties such as minimally invasiveness, bypassing first-pass metabolism, and can be self-administered. A range of materials has been used to fabricate MNs, such as silicon, metal, glass, and polymers. Among them, polymer MNs have gained increasing attention from pharmaceutical and cosmetic companies as one of the promising drug delivery methods. MN products have recently become available on the market, and some of them are under evaluation for efficacy and safety. This paper focuses on the current state of polymer MNs in drug transdermal delivery. The materials and methods for the fabrication of polymer MNs and their drug administration are described. The recent progress of polymer MNs for treatment of cancer, vaccine delivery, blood glucose regulation, androgenetic alopecia, obesity, tissue healing, myocardial infarction, and gout are reviewed. The challenges of MNs technology are summarized and the future development trend of MNs is also prospected.

Abstract:Microneedles (MNs) have been used to deliver drugs for over two decades. These platforms have been proven to increase transdermal drug delivery efficiency dramatically by penetrating restrictive tissue barriers in a minimally invasive manner. While much of the early development of MNs focused on transdermal drug delivery, this technology can be applied to a variety of other non-transdermal biomedical applications. Several variations, such as multi-layer or hollow MNs, have been developed to cater to the needs of specific applications. The heterogeneity in the design of MNs has demanded similar variety in their fabrication methods; the most common methods include micromolding and drawing lithography. Numerous materials have been explored for MN fabrication which range from biocompatible ceramics and metals to natural and synthetic biodegradable polymers. Recent advances in MN engineering have diversified MNs to include unique shapes, materials, and mechanical properties that can be tailored for organ-specific applications. In this review, we discuss the design and creation of modern MNs that aim to surpass the biological barriers of non-transdermal drug delivery in ocular, vascular, oral, and mucosal tissue. Abstract:The polymer‐based microneedles are reinforced with layer‐by‐layer films of silica nanoparticles and a polycation followed by hydrothermal calcination. The mechanical strength is significantly improved and shows lower threshold pressure to penetrate skin. Moreover, the drug loading and releasing properties are significantly enhanced due to an increase in the surface area and interfacial interaction. Microneedle (MN) technology offers a powerful approach for transdermal delivery enabling painless injection and facilitating self‐administration without the need for professional assistance. However, the weak mechanical strength of MNs can lead to inefficient drug delivery and serious skin irritation if the MNs fracture during administration and leave fragments under the skin. Thus, the MNs need to be mechanically robust to avoid fracture during penetration through the skin while maintaining efficient drug delivery. Herein, the polymer‐based MNs with layer‐by‐layer (LbL) films of silica (SiO2) nanoparticles (NPs) and a polycation (poly(diallyldimethylammonium chloride) (PDADMAC)) followed by hydrothermal calcination are reinforced. The mechanical strength of the MNs is significantly improved after LbL assembly and shows lower threshold pressure to penetrate skins. Moreover, their drug loading and releasing properties are significantly enhanced due to an increase in the surface area and interfacial interaction. These SiO2 nanoparticle‐containing LbL thin films have great potential for the surface modification of 3D microstructured devices such as MNs, as evidenced by their enhanced mechanical strength and drug coating efficiency that result in a promising MN drug delivery model.

Abstract:<p>Microneedles (MNs) are a self-administrable and painless alternative to hypodermic needles for bolus drug delivery. Sustained drug release is the preferable drug delivery method because it reduces the chemical burden in patients and can deliver therapeutics that require long-term exposure. Here, we review recent advances in long-acting MNs, summarizing polymers used to fabricate long-acting MNs and mechanisms controlling drug release. We also introduce novel designs of separable needles to achieve implantable tips and bioinspired needles for enhanced skin adhesion, as well as strategies using the back layer as a drug reservoir.</p>

Abstract:Microneedles are an efficient and minimally invasive approach to transdermal drug delivery and extraction of skin interstitial fluid. Compared to solid microneedles made of silicon, metals and ceramics, polymeric microneedles have attracted extensive attention due to their excellent biocompatibility, biodegradability and nontoxicity. They are easy to fabricate in large scale and can load drugs in high amounts. More importantly, polymers with different degradation profiles, swelling properties, and responses to biological/physical stimuli can be employed to fabricate polymeric microneedles with different mechanical properties and performance. This review provides a guideline for the selection of polymers and the corresponding fabrication methods for polymeric microneedles while summarizing their recent application in drug delivery and fluid extraction. It should be noted that although polymeric microneedles can achieve efficient transdermal delivery of drugs, their wide applications were limited by their unsatisfactory transdermal therapeutic efficiency. Delivery of nanomedicines that incorporate drugs into functional nanoparticles/capsules can address this problem and thus may be an interesting direction in the future.

Abstract:<p class="a-plus-plus">Transdermal drug delivery is an emerging field in the pharmaceutical remit compared with conventional methods (oral and parenteral). Microneedle (MN)-based devices have gained significant interest as a strategy to overcome the skin's formidable barrier: the stratum corneum. This approach provides a less invasive, more efficient, patient friendly method of drug delivery with the ability to incorporate various therapeutic agents including macromolecules (proteins and peptides), anti-cancer agents and other hydrophilic and hydrophobic compounds. This short review attempts to assess the various materials involved in the fabrication of MNs as well as incorporation of other excipients to improve drug delivery for novel medical devices. The focus will be on polymers, metals and other inorganic materials utilised for MN drug delivery, as well as their application, limitations and future work to be carried out.</p>

Abstract:Microneedles (MNs) prepared from polymeric materials are painless and minimally invasive, safe and efficient, but they hindered by low mechanical strength and single diverse drug release pattern. Due to the distinctive mechanical strength and dimensions of poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs), the integration of nano-technology with microneedles can effectively improve penetration and delivery efficiency through the stratum corneum. We herein designed a simple paroxetine (PAX)-loaded PLGA nanoparticles-integrated dissolving microneedles system (PAX-NPs-DMNs), aiming to improve the bioavailability of PAX through the synergistic permeation-enhancing effect of dissolving microneedles (DMNs) and NPs. PAX-NPs-DMNs had a complete tips molding rate ( N eff ) of (94.06 ± 2.16) %, a 15×15 quadrangular-conical microneedle array and an overall fracture force of 301.10 N, which were improved nearly 0.50 times compared with the blank microneedles (HA-DMNs) and PAX microneedles (PAX-DMNs). PAX-NPs-DMNs could extend the release duration of PAX from 1 h to 24 h and the cumulative permeability per unit area (Q n ) was 47.66 times and 7.37 times higher than the PAX and the PAX-DMNs groups. PAX-NPs-DMNs could be rapidly dissolved within 10 min without hindering skin healing or causing adverse reactions. This study confirmed that PAX-NPs-DMNs can effectively improve the bioavailability of PAX and the mechanical strength of DMNs, which can easily penetrate the skin to provide sustained and painless delivery without causing adverse effects, thus offering a more convenient and effective method for central nervous diseases.

Abstract:Our comprehensive review plunges into the cutting-edge advancements of polymeric microneedle drug delivery systems, underscoring their transformative potential in the realm of transdermal drug administration. Our scrutiny centers on the substrate materials pivotal for microneedle construction and the core properties that dictate their efficacy. We delve into the distinctive interplay between microneedles and dermal layers, underscoring the mechanisms by which this synergy enhances drug absorption and precision targeting. Moreover, we examine the acupoint–target organ–ganglion nexus, an innovative strategy that steers drug concentration to specific targets, offering a paradigm for precision medicine. A thorough analysis of the clinical applications of polymeric microneedle systems is presented, highlighting their adaptability and impact across a spectrum of therapeutic domains. This review also accentuates the systems' promise to bolster patient compliance, attributed to their minimally invasive and painless mode of drug delivery. We present forward-looking strategies aimed at optimizing stimulation sites to amplify therapeutic benefits. The anticipation is set for the introduction of superior biocompatible materials with advanced mechanical properties, customizing microneedles to cater to specialized clinical demands. In parallel, we deliberate on safety strategies aimed at boosting drug loading capacities and solidifying the efficacy of microneedle-based therapeutics. Abstract:Microneedle (MN) delivery system has been greatly developed to deliver drugs into the skin painlessly, noninvasively, and safety. In the past several decades, various types of MNs have been developed by the newer producing techniques. Briefly, as for the morphologically, MNs can be classified into solid, coated, dissolved, and hollow MN, based on the transdermal drug delivery methods of “poke and patch,” “coat and poke,” “poke and release,” and “poke and flow,” respectively. Microneedles also have other characteristics based on the materials and structures. In addition, various manufacturing techniques have been well-developed based on the materials. In this review, the materials, structures, morphologies, and fabricating methods of MNs are summarized. A separate part of the review is used to illustrate the application of MNs to deliver vaccine, insulin, lidocaine, aspirin, and other drugs. Finally, the review ends up with a perspective on the challenges in research and development of MNs, envisioning the future development of MNs as the next generation of drug delivery system.

Abstract:Over the past decades, the use of microneedles, a unique transdermal drug delivery technology, to deliver drugs to the skin for biological effects has been very active, and the interest in dissolving microneedles in this field has also increased rapidly. However, ensuring the effectiveness of microneedles requires attention to several key steps. The microneedle must pierce the stratum corneum first, and then continue to penetrate to a certain depth under the action of applied force, without bending or breaking during the puncture process, to ensure the accurate and controllable delivery of the microneedles. In the process of microneedle penetration, the skin is easily deformed due to its elasticity, and the microneedle often cannot penetrate the skin completely, which reduces the delivery efficiency of the microneedle and limits the application of the microneedle in drug delivery, especially for dissolving microneedles whose mechanical strength is weaker than that of metal and monocrystalline silicon microneedles. The delivery efficiency of the microneedle is mainly related to the penetration depth of the microneedle, which in turn is related to factors such as the mechanical properties of the skin, the structure of the microneedle, and the way of administration. Clearly, there are still many technical challenges to make the effectiveness of dissolving microneedles in line with requirements of clinical medication. Therefore, it is necessary to systematically review the influencing factors of the penetration depth of the dissolving microneedles and the methods to improve the delivery efficiency. Technological improvement strategies suitable for enhancing the drug delivery efficiency of dissolving microneedles will certainly contribute to more accurate delivery of bioactive ingredients to the body and a wider range of medical applications.
Dividing Lines : Racial Redistricting and Substantive Representation ∗

Abstract:We present a model of optimal redistricting schemes to promote minority interests, incorporating both electoral and legislative stages. In the model, after districting, candidates from majority and minority groups run for office in each district. The election winners then form a legislature, which in turn passes a redistributive policy. The results show that minorities with relatively little political power prefer to concentrate their voters in a few districts, while more powerful minorities do best by distributing their voters more evenly across districts. Furthermore, declining majority racism has two effects on minorities: it helps them by making it easier to elected minorities to office, but it may also hurt them by making majority voters more pivotal and therefore increasing their relative power at minorities’ expense. In addition, the impact of adding more minority voters to a given district is non-monotonic, and in some cases can have the perverse effect of electing a candidate less favored by the minority community.

Abstract:Highlights • A review focusing on microneedle-assisted wound management and tissue regeneration is given. • Drug & material selection, microneedle designs are summarized to give a guideline to construct microneedle systems. • Recent advances in application of microneedles in healing of different tissues and organs are reviewed. Microneedle, as a novel drug delivery system, has attracted widespread attention due to its non-invasiveness, painless and simple administration, controllable drug delivery, and diverse cargo loading capacity. Although microneedles are initially designed to penetrate stratum corneum of skin for transdermal drug delivery, they, recently, have been used to promote wound healing and regeneration of diverse tissues and organs and the results are promising. Despite there are reviews about microneedles, few of them focus on wound healing and tissue regeneration. Here, we review the recent advances of microneedles in this field. We first give an overview of microneedle system in terms of its potential cargos (e.g., small molecules, macromolecules, nucleic acids, nanoparticles, extracellular vesicle, cells), structural designs (e.g., multidrug structures, adhesive structures), material selection, and drug release mechanisms. Then we briefly summarize different microneedle fabrication methods, including their advantages and limitations. We finally summarize the recent progress of microneedle-assisted wound healing and tissue regeneration (e.g., skin, cardiac, bone, tendon, ocular, vascular, oral, hair, spinal cord, and uterine tissues). We expect that our article would serve as a guideline for readers to design their microneedle systems according to different applications, including material selection, drug selection, and structure design, for achieving better healing and regeneration efficacy. Abstract:Abstract Transdermal microneedle (MN) patches are a promising tool used to transport a wide variety of active compounds into the skin. To serve as a substitute for common hypodermic needles, MNs must pierce the human stratum corneum (~ 10 to 20 μm), without rupturing or bending during penetration. This ensures that the cargo is released at the predetermined place and time. Therefore, the ability of MN patches to sufficiently pierce the skin is a crucial requirement. In the current review, the pain signal and its management during application of MNs and typical hypodermic needles are presented and compared. This is followed by a discussion on mechanical analysis and skin models used for insertion tests before application to clinical practice. Factors that affect insertion (e.g., geometry, material composition and cross-linking of MNs), along with recent advancements in developed strategies (e.g., insertion responsive patches and 3D printed biomimetic MNs using two-photon lithography) to improve the skin penetration are highlighted to provide a backdrop for future research.

Abstract:In this study, a micro-molding technology was used to prepare the microneedles (MNs), while a texture analyzer was used to measure its Young’s modulus, Poisson’s ratio and compression breaking force, to evaluate whether the MNs can penetrate the skin. The effects of different materials were characterized by their ability to withstand stresses using the Structural Mechanics Module of COMSOL Multiphysics. Carboxymethylcellulose (CMC) was chosen as the needle formulation material with varying quantities of polyvinyl pyrrolidone (PVP), polyvinyl alcohol (PVA) and hyaluronic acid (HA) to adjust the viscosity, brittleness, hardness and solubility of the material. The results of both the experimental tests and the predictions indicated that the hardest tip material had a solids content of 15% (w/w ) with a 1:2 (w/w) CMC: HA ratio. Furthermore, it was shown that a solid content of 10% (w/w) with a 1:5 (w/w) CMC: PVA ratio is suitable for making patches. The correlation between the mechanical properties and the different materials was found using the simulation analysis as well as the force required for different dissolving microneedles (DMNs) to penetrate the skin, which significantly promoted the research progress of microneedle transdermal drug delivery.

Abstract:Abstract In the past two decades, microneedles (MNs), as a painless and simple drug delivery system, have received increasing attention for various biomedical applications such as transdermal drug delivery, interstitial fluid (ISF) extraction, and biosensing. Among the various types of MNs, porous MNs have been recently researched owing to their distinctive and unique characteristics, where porous structures inside MNs with continuous nano- or micro-sized pores can transport drugs or biofluids by capillary action. In addition, a wide range of materials, including non-polymers and polymers, were researched and used to form the porous structures of porous MNs. Adjustable porosity by different fabrication methods enables the achievement of sufficient mechanical strength by optimising fluid flows inside MNs. Moreover, biocompatible porous MNs integrated with biosensors can offer portable detection and rapid measurement of biomarkers in a minimally invasive manner. This review focuses on several aspects of current porous MN technology, including material selection, fabrication processes, biomedical applications, primarily covering transdermal drug delivery, ISF extraction, and biosensing, along with future prospects as well as challenges. Abstract:The applications of microneedles (MNs) are becoming popular with the promise of efficient and advanced drug delivery. MNs were developed to overcome the limitations of conventional drug delivery systems and bypass biological barriers. While most MN applications in the past decades focused on transdermal biomedical applications, recent advancements in engineering and technology have enabled MNs to be used in a wide range of non-transdermal applications. Compared with the other types of MNs, polymer-based MN composites have attracted more attention for non-transdermal drug delivery because they exhibit excellent biological properties, including being nontoxic, biocompatible, and biodegradable, making them ideal biomaterials for drug delivery applications that overcome the metabolic constraints of drug delivery for macromolecular payloads across a variety of tissues and organs other than the skin. This review provides an overview of recent advancements in polymer-based MN composite carriers that aim to overcome the delivery challenges for non-transdermal drug delivery, specifically in the vascular, ocular, gastrointestinal tract, buccal transmucosal, periodontal, cardiovascular, and vaginal tissue. Furthermore, this review will discuss future perspectives and challenges for polymeric MN composites in non-transdermal drug delivery that must be resolved.
