Abstract:Milk is not only a composite of nutrients but emerged as a source of exosomes acting as a promising drug delivery vehicle for small interfering RNA (siRNA). siRNA is known for its immense therapeutic potential but has various physiological limitations, including stable delivery. To investigate the suitability of siRNA for physiological stability and oral delivery, we encapsulated scrambled Alexa Fluor (AF)-488 siRNA in milk whey exosomes using lipofection and evaluated stability against the digestive processes along with its uptake and transepithelial transport by intestinal epithelial cells. Milk exosomal siRNA were found resistant to different digestive juices, including saliva, gastric, bile, and pancreatic juices, in vitro and were internalized by Caco-2 cells. The stable delivery of exosomal AF-488 siRNA along with its transepithelial transport was confirmed by fluorescence microscopy and fluorescence intensity measurements. In summary, the encapsulation of siRNA in milk exosomes resists harsh digestive processes, improving intestinal permeability and payload protection.
Co-exposure of parabens, benzophenones, triclosan, and triclocarban in human urine from children and adults in South China

Abstract:Endocrine-disrupting chemicals (EDCs) are pervasive in the environment, prompting significant public concern regarding human exposure to these pollutants. In this study, we analyzed the levels of various endocrine-disrupting compounds, including parabens (PBs), benzophenones (BzPs), triclocarban (TCC) and triclosan (TCS), across 565 urine samples collected from residents of South China. All 11 target chemicals were detected at relatively high frequencies (41-100%), with the most prevalent ones being 3,4-dihydroxybenzoic acid (5.39 ng/mL), methyl-paraben (5.12 ng/mL), ethyl-paraben (3.11 ng/mL) and triclosan (0.978 ng/mL). PBs emerged as the most predominant group with a median concentration of 32.2 ng/mL, followed by TCs (sum of TCC and TCS, 0.998 ng/mL) and BzPs (0.211 ng/mL). Notably, urinary concentrations of PBs in adults were significantly higher (p < 0.01) compared to children, while BzPs and TCs were elevated in children (p < 0.001). The increased presence of BzPs and TCs in children is a cause for concern, given their heightened sensitivity and vulnerability to chemicals. Significant correlations were found between urinary target compounds and demographic factors, including gender, age and body mass index. Specifically, females, younger adults (18 ≤ age ≤ 35) and individuals with under/normal weight (16 ≤ BMI ≤ 23.9) were found to have higher exposure levels to EDCs, as indicated by the median values of their estimated daily intakes. Despite these higher levels still being lower than the acceptable daily intake thresholds, the health risks stemming from simultaneous exposure to these EDCs must not be overlooked.
Exposure of Chinese adult females to parabens from personal care products: Estimation of intake via dermal contact and health risks

Abstract:Parabens are added into foodstuffs, pharmaceuticals and personal care products (PCPs) as additives extensively due to their excellent antiseptic and antibacterial effects. In the past decade, parabens have raised great concerns on their potential harm to humans. Existing studies have suggested positive correlations between PCP application and urinary paraben concentrations in females, but little is known about paraben exposure levels and health risks arising from PCP use. In this study, 150 PCP samples covering eleven categories were collected from South China and measured for the concentrations of five parabens, including methyl paraben (MeP), ethyl paraben (EtP), propyl paraben (PrP), butyl paraben (BuP) and benzyl paraben (BeP). Parabens were widely detected in PCPs, with a detection frequency of 100%, 99.3%, 80.0%, 74.0% and 13.3%, for MeP, EtP, PrP, BuP and BeP, respectively. The median concentration of Σ5parabens was 126 μg/g with a range of 6.38-424 μg/g across all PCP samples. The contents of MeP, EtP and PrP measured in leave-on PCPs were obviously higher than those in the rinse-off ones (p < 0.05). MeP and PrP were the main paraben analogues, together accounting for 93.6% of Σ5parabens in all PCPs. The daily intakes of parabens through dermal absorption by Chinese adult females estimated by measurements obtained in the present study were 0.15 and 83.2 μg/kg-bw/day on basis of the application of rinse-off and leave-on PCPs, respectively. Among the eleven categories, sunscreen, body lotion and mask constituted the main exposure sources of parabens to females. The hazard quotients of parabens were far less than 1, indicating no considerable health risk for Chinese adult females.

Abstract:Humans are constantly exposed to parabens (PBs), triclosan (TCS), benzophenones (BPs), and phthalate esters (PAEs) due to the widespread existence of these chemicals in personal care products (PCPs), and the high frequency of usage for humans. Previous studies indicated each class of the above-mentioned chemicals can exhibit potential adverse effects on humans, in particular DNA oxidative damage. However, the health risk assessment of combined exposures to multiple PCPs is limited, especially the overall dose-effect of mixtures of these chemicals on DNA oxidative damage. In this study, we measured the urinary levels of 6 PBs, TCS, 8 BPs, 15 metabolites of PAEs (mono-PAEs), and 8-hydroxy-2′-deoxyguanosine (8-OHdG) from 299 adults simultaneously. PBs, TCS, BPs, and mono-PAEs were frequently detected in urinary samples with median concentrations of 52.888, 0.737, 1.305, and 141.381 ng/ml, suggesting a broad, low-level exposure among participants. Risk assessments indicated approximately 22% and 15% of participants suffered health risks (Hazard index >1) from exposure to TCS and PAEs. The relationship between 8-OHdG levels and chemical exposure was estimated by Bayesian kernel machine regression (BKMR) models. It indicated an overall positive correlation between the mixture of these chemicals and 8-OHdG, with methylparaben and mono-benzyl phthalate contributing the most to this association. Of note, sex-related differences were observed, in which exposure to PCPs led to higher health risks and more pronounced dose-effect on DNA damage in the female population. Our novel findings reveal the health risks of exposure to low-level PCPs mixtures and further point out the overall dose-response relationship between DNA oxidative damage and PCP mixtures.

Abstract:Parabens, triclosan (TCS) and triclocarban (TCC) are emerging endocrine disrupting chemicals, which are commonly used in personal care products and household applications in daily life. Due to their adverse health effects, human exposure to these chemicals has been a public concern. Despite evidence showing different exposure pathways of these chemicals, few studies have examined contribution of certain exposure to total human exposure. In this study, we measured six parabens, TCS and TCC in 129 indoor dust samples and these chemicals plus four paraben metabolites in 203 urine samples from two different cities in China (Suizhou, a typical small city in central China and Beijing, the capital of China). The median concentrations of ∑6Parabens (1050 ng/g) and ∑TCS + TCC (565 ng/g) in dusts from Beijing were 1.9–3.3 times higher than those from Suizhou (∑6Parabens: 314, ∑TCS + TCC: 294 ng/g). The ∑6Parabens in urines from Suizhou and Beijing were in the range of 0.208–645 and 0.455–2300 μg/g Creatinine (Cr), respectively. The ∑TCS + TCC concentrations in urine were 1–2 orders of magnitude lower than those found for ∑6Parabens. Comparatively, women had relatively higher body burden of parabens than men, which was more noticeable when considering the use of skin care products. In this regard, relatively higher levels of parabens were also found in women in couples. Positive correlations were found between concentrations of methyl-paraben (MeP) and ethyl-paraben (PrP) in paired dust and urine samples (p < 0.05). Human exposure to such chemicals was evaluated. Based on the measured concentrations in paired human urine and indoor dust samples, our results provided direct evidence that the contribution of indoor dust ingestion to the total exposure was minor. These findings are essential for clarifying the sources and potential exposure routes of these chemicals in humans.

Abstract:Phthalates are used in a wide variety of personal care products (PCPs) as solubilizer, denaturant or color fixative. However, there are few studies referring the correlation between phthalate exposure and PCPs use among general population in China. In this study, ten metabolites of phthalates in spot urine samples (N = 500) were quantified using ultra-performance liquid chromatography tandem mass spectrometry. The frequency and duration of 12 types of PCPs were collected by questionnaire survey. The associations of phthalate metabolites and PCPs use were assessed by multivariable liner regression models. Median concentration of mono-benzylphthalate (MBzP) was significantly higher among frequent users of almost all PCPs. Low molecular weight phthalates (monomethyl phthalate (MMP), mono-n-butylphthalate (MnBP) and monoisobutylphthalate (MiBP)) were significantly lower among frequent user of some rinse-off PCPs (e.g., shampoo, facial cleanser, and body wash). Meanwhile, monoethylphthalate (MEP) was positive correlated with frequently use of facial moisturizer. Mono-2-ethylhexylphthalate (MEHP) and MBzP showed a significant positive association with frequently use of body lotion. Univariate linear analyses indicated a significant decreasing trend between urinary concentrations of MMP, MnBP, and the number of rinse-off PCPs being frequently used, and a significant increasing trend between urinary MBzP and the number of leave-on products being frequently used. These findings indicated that the use of some PCPs, especially leave-on PCPs, could be a potential source of exposure to some phthalates.

Abstract:Parabens and triclocarban are widely applied as antimicrobial preservatives in foodstuffs, pharmaceuticals, cosmetics, and personal care products. However, few studies have been conducted on large-scale biomonitoring of parabens and triclocarban in the Chinese general population. In the present study, there were 1157 urine samples collected from 26 Chinese provincial capitals for parabens and triclocarban measurement to evaluate the exposure levels, spatial distribution, and influencing factors, as well as associated health risks in the Chinese population. The median concentrations of Σparabens and triclocarban were 14.0 and 0.03 μg/L, respectively. Methyl paraben was the predominant compound. Subjects in western China were more exposed to parabens, possibly due to climate differences resulting in higher consumption of personal care products. Subjects who were female, aged 18-44 years, or had a higher education level were found to have higher paraben concentrations. The frequency of drinking bottled water was positively associated with paraben exposure. The assessment of health risk based on urinary paraben concentrations indicated that 0.8 % of the subjects had a hazard index exceeding one unit, while Monte Carlo analysis suggested that 3.6 % of the Chinese population exposure to parabens had a potential non-carcinogenic risk. This large-scale biomonitoring study will help to understand the exposure levels of parabens and triclocarban in the Chinese general population and provide supporting information for government decision-making.

Abstract:Personal care products, such as additives, have raised widespread concerns about the potential threat to male reproductive health. The spermatogenesis in humans lasts for approximately 90 days, the average levels of these chemicals remain unclear during spermatogenesis. In our study, we pooled urine samples from each man during the days of 1–15, 16–31, 32–63, and ≥64, and examined exposure to 48 typical additive chemicals. By principal component analysis (PCA), k-means clustering, and Spearman's rank correlations, we then identified 6 PC scores and 4 clusters based on profiles of these chemicals. Some industrial, commercial or structural similar chemicals (e.g., phthalates) were significantly correlated compared to unrelated chemicals (e.g., benzophenone). PCA scores were associated with individual lifestyles (e.g., household income, tea consumption, and drinking tap water). Distinct exposure components and exposure patterns of personal care products may help the reproductive health assessment of men. We suggested more concerns for widespread exposure to these chemicals for men.

Abstract:Many environmental phenols, such as bisphenols, benzophenones and parabens, are known as endocrine dis-ruptors and can adversely affect human health. However, the knowledge of human exposure to common envi-ronmental phenols in Chinese rural areas is insufficient. In this context, 181 urine samples were collected from participants in a rural area in Northwest China and were analyzed for nine bisphenols, three benzophenones and four parabens. Bisphenol A (BPA), bisphenol S, benzophenone-1 (BP-1), benzophenone-3 (BP-3), 4-hydroxyben-zophenone, methylparaben (MeP), ethylparaben and propylparaben (PrP) were detected in more than 50% of the urine samples, with median concentrations of 0.938 ng/mL, 0.0111 ng/mL, 0.191 ng/mL, 1.30 ng/mL, 0.0320 ng/mL, 25.9 ng/mL, 4.31 ng/mL and 1.94 ng/mL, respectively. A significant positive correlation was observed between BP-1 and BP-3, as well as between MeP and PrP, indicating metabolic transformation and combined use, respectively. The concentrations of MeP and PrP in females were significantly higher than those in males, sug-gesting that females were exposed to more MeP and PrP than males. Urinary concentrations of BPA, BP-3, MeP and PrP could be influenced by age. Other demographic information, such as annual household income, edu-cation and occupation was not associated with the exposure level of the targeted phenols in adults. The estimated daily intakes of the analytes except BPA were all below their respective tolerable/acceptable daily intake levels. This study profiles the demographic differences in the exposure to environmental phenols in general populations from rural areas and provides information on risk assessments.
Cumulative risk assessment and exposure characteristics of parabens in the general Taiwanese using multiple hazard indices approaches

Abstract:Parabens, a group of endocrine disrupting chemicals (EDCs), are well known preservatives in pharmaceuticals and personal care products (PPCPs). However, studies on parabens exposure and their cumulative effects in Asian population are limited. This study aimed to identify the exposure characteristics and estimate the cumulative risk of four parabens in the general Taiwanese. We have collected urine samples including 271 adults (18-97 yrs old) and 95 minors (7-17 yrs old), from Taiwan Environmental Survey for Toxicants 2013, and analyzed for four urinary parabens including methyl (MeP)-, ethyl (EtP)-, propyl (PrP)-, and butylparaben (BuP) by using ultraperformance liquid chromatography-tandem mass spectrometry. The health-based guidance value (HBGV) and the antiandrogenic properties of parabens were used to calculate the hazard index (HI) for cumulative risk. MeP and PrP were most abundant compounds and startlingly higher than those in other countries. Adults had a higher geometric mean level of four parabens than minors (adults: MeP, 381.7; PrP, 108.6; EtP, 39.6 and BuP 6.3 ng/mL; minors: MeP, 65.7; PrP, 7.9, EtP, 2.6 and BuP 2.2 ng/mL). Participants who used a higher number of personal care products had a significantly higher risk with higher concentrations of PrP (above 75th %tile) [adjusted odds ratio (aOR): 1.79, 95 % CI: 1.01-3.15] and BuP [aOR: 1.78, 95 % CI: 1.03-3.07]. The median and 95th %tile HI (the sum of the HQs of each paraben) was as 1.10 and 4.39-fold higher than acceptable cumulative threshold (HI <1) and PrP accounted for 90 % of the HI. Our results indicate omnipresent exposure to parabens among the Taiwanese population, which might cause certain level of concerns. These significant increasing trends of HI with age dependence were observed, which mainly driven by PPCPS used. Routine survey of parabens in PPCPs and continued biomonitoring needs to be urgently addressed.

Abstract:Phthalate esters (PE) are synthetic chemicals widely used in industry, and have been detected in personal care products (PCP). Recent findings of human reports demonstrated endocrine-disrupting action associated with phthalate exposures. The aims of this study were to (1) measure levels of 11 PE in 198 PCP collected from retail markets in Shanghai and (2) assess daily dermal exposure in adult females and infants. The health risk of cumulative exposure to eight PE on reproductive system function derived from dermal PCP use was further assessed by utilizing the hazard index (HI) approach. Diethyl phthalate (DEP) was the most frequently detected compound (29.8%), followed by diisobutyl phthalate (DiBP) (6.6%). The geometric mean (GM) concentrations of daily exposure to DEP, bis(2-methoxyethyl) phthalate (DMEP), DiBP, dibutyl phthalate (DBP), diphenyl phthalate (DPP), and bis(2-ethylhexyl) phthalate (DEHP) in female adults were 0.018, 0.012, 0.002, 0.001, 0.003, and 0.002 mu g/kg body weight (bw)/d, respectively. The GM daily exposure levels to PE in infants and adult females were similar except for DEHP, which was higher in infants. DEP exposure was highest in both subpopulations at either GM or maximal level. All HI of 8 PE were far less than 1, ranging from 0.0002 to 0.005, indicating no cumulative reproductive risks to these populations. DBP, DMEP, and DEHP were three major contributors to the cumulative HI. In summary, the level of phthalate in PCP from Shanghai retail markets posed no apparent cumulative risk to adult females and infants in China.

Abstract:The usage of parabens, bisphenol A and triclosan in diverse consumer products is in widespread. Nevertheless, there are limited data concerning exposure to these chemicals in human being, especially in primiparas. Biomonitoring of chemicals in primiparas is useful for the estimation of chemical exposure risks for both primiparas and their offspring. This study aims to investigate urinary levels of parabens, bisphenol A and triclosan of 84 primiparas from Shenzhen, China and to evaluate their potential health risks. Methyl, ethyl, and n-propyl parabens bisphenol A and triclosan exhibited high detection rates (DRs) (> 97%) in urine samples, suggesting that primiparas are exposed to them widely. The median concentrations of methyl, ethyl, and n-propyl parabens, bispenol A and triclosan in urine were 2.14, 4.10, 0.46, 1.30 and 3.00 µg/L, respectively. Ethyl paraben was the predominant paraben accounting for nearly half of Σ3parabens (The sum concentrations of methyl, ethyl, n-propyl parabens). Positive associations with significance (p < 0.05) were found between the usage of plastic containers and urinary concentrations of ethyl paraben or BPA, indicating plastic containers might be an important factor influencing primipara exposure to these two chemicals. Urinary concentrations of methyl paraben were positively associated (p < 0.05) with the time of computer use by participant, suggesting that indoor dust might constitute an important source of parabens. The estimated daily intakes of parabens, bisphenol A and triclosan contrasted with the acceptable daily intakes in a comparatively low level. The hazard quotients (HQs) of these chemicals were all less than 1, suggesting no health risks for primiparas from South China.

Abstract:Maternal exposure to benzene and related compounds, trichloroethylene, and polycyclic aromatic hydrocarbons has been suggested as risk factors for reproductive and developmental problem. In different countries and groups, there are differences in socioeconomic circumstances, consumer products, dietary habits, lifestyles, and so on, resulting in different exposure risks from these chemicals. This study investigated the correlation between their metabolite concentrations and socioeconomic, demographic, and dietary factors to explore the possible exposure source of the concerned pollutants in pregnant women. We conducted biological monitoring to assess the exposure of these chemicals using urine samples from 590 to 639 pregnant women during 2nd or 3rd trimester of pregnancy in six cities of China. Socioeconomic and demographic characteristics and dietary habits were collected from questionnaires. The detection rate was over 74% of the urine samples for all metabolites. Compared with the Fourth National Report on Human Exposure to Environmental Chemical concentrations for females (FNRHEEC, the US Centers for Disease Control and Prevention), the metabolite concentrations of benzene compounds and trichloroethylene, excluding MA reported here, were higher. Principal component analysis results showed that SBMA and MHA could be proxy of the principal sources of metabolites for benzene compounds. The concentrations of SBMA and MA were higher in Fuzhou and Wuhan, respectively. The concentration of DCVMA was higher in Shenzhen, Xi’an, and Nanning, and the concentration of PGA was higher in Fuzhou, Wuhan, and Xi’an. Also, the 1-OHPG concentration in Wuhan is higher than that in Fuzhou and Shenzhen. Unhealthy dietary habits, using cosmetics and indoor exposure, contacting chemical solvent during pregnancy were associated with increased benzene compounds, trichloroethylene, and PAH exposure. There were significant positive associations between 1-OHPG level and maternal BMI, low education status, and cooking without a range hood. Pregnant women in China may be at a greater risk of exposure to most of the target compounds than US females, and their exposure levels varied in different regions. Some adverse environmental and behavioral factors may increase the exposure of environmental toxins, which can urge people to take measures to reduce the health risk to pregnant women during pregnancy.

Abstract:The adverse effects of parabens raise concerns about their extensive use as preservatives in consumer products, especially in cosmetics. Until now, their distribution and excretion in humans have attracted little attention. Here, we quantified various agents including, for the first time, methyl-; ethyl-; n-propyl-; n-butyl-, and i-butylparaben (MeP, EtP, PrP, n-BuP, i-BuP); methyl- and ethyl-protocatechuate (OH-MeP and OH-EtP); hydroxybenzoic acid (4-HB); and 3,4-dihydroxybenzoic acid (3,4-DHB) in urine, serum, and seminal plasma samples from 50 healthy Chinese men in Beijing, China. Urine paraben concentrations were 1–2 orders of magnitudes higher than those in serum and seminal plasma. MeP and PrP were predominant and correlated with each other in the urine, serum, and seminal plasma. In urine, we observed a significant correlation between MeP and OH-MeP; EtP and OH-EtP; and 4-HB and 3,4-DHB concentrations. All these results provide new information on parabens as biomarkers for the assessment of exposure.Graphical abstract

Abstract:Phthalate esters (PAEs) are widely used in consumer and industrial products and may thus pose significant health risks. Urine analysis, which has usually been applied to assess the health risks of PAEs, has the drawback of small sample sizes and insufficient representativeness. Wastewater-based epidemiology (WBE) collects wastewater samples containing urine from the entire community and thus is more representative than urine samples. In this work, exposure levels and health risks of PAEs were estimated on a national scale for the first time through the WBE approach. Wastewater samples were collected from 54 wastewater treatment plants in 27 major cities that cover all of the geographic regions of China. The estimated ∑5PAEs exposure levels ranged from 290 μg/inh/d (Lhasa) to 3642 ± 467 μg/inh/d (Zhengzhou) with a mean level of 2184 ± 1173 μg/inh/d. Di-n-butyl phthalate accounted for the highest proportion (65%) in the total exposure level. The ∑5PAEs exposure levels in Southwest China were significantly lower than those in other regions due to the low production and consumption of plastics in the region. The health risks of PAEs were assessed by comparing the estimated daily exposure levels to the acceptable daily exposure levels. For adults, the hazard index that represents cumulative risk of PAEs was above or below 1 depending on a particular reference dose (total daily intake values or the reference dose for anti-androgenicity) that was used for risk calculation. In contrast, the hazard index was above 1 for a significant number of cities for children, regardless of which reference dose was used. The results indicate that health risks of PAE exposure in China cannot be overlooked. Children in China are under considerably greater risks than adults, which warrants further research or proper regulation of PAE use in China.

Abstract:Parabens are widely applied as preservatives in cosmetics, drugs and food. Previous studies suggested that parabens could exhibit potential risks to human health. However, data on human exposure levels and health effects of parabens remain limited, especially in the potential effects on DNA oxidative stress. This study aimed to investigate urinary levels of parabens in adults from South China and explore the relationships between urinary parabens and DNA oxidative stress. Five short chain parabens, including methyl paraben (MeP), ethyl paraben (EtP), n-propyl paraben (PrP), butyl paraben (BuP) and benzyl paraben (BzP), were determined in urine from 319 adults in Shenzhen, China. MeP, EtP and PrP were frequently detected in urine samples (detection frequencies >66.5%), suggesting broad exposure in South China adults. Median concentrations of MeP, EtP, PrP, BuP and BzP were 5.78, 0.39, 0.35, 0.01 and 0.02 μg/L, respectively. A significantly positive correlation was observed between the urinary concentrations of MeP and PrP (p < 0.01), suggesting similar sources for these two chemicals. In addition, participants with alcohol consumption exhibited significantly lower paraben concentrations in urine than those without alcohol drinking (p < 0.05). Significant association was observed between urinary concentrations of parabens and 8-hydroxy-2′-deoxyguanosine (8-OHdG) levels (p < 0.01), while no significant dose-response relationship was found (p > 0.05). A potential risk from PrP exposure was found in South China adults.
