Abstract:This study investigated the changes in the structure of wheat starch after synergistic fermentation of Lactobacillus plantarum and Saccharomyces cerevisiae at different wheat bran dietary fiber (WBDF) levels. The results showed that WBDF was slightly resistant to the decrease in acidity within the fermentation system. The amylose content decreased from 32.12% to 19.92% (P < 0.05), amylose/amylopectin ratio decreased from 0.47 to 0.25 (P < 0.05), and relative crystallinity decreased from 12.17% to 9.40% (P < 0.05) in the samples containing WBDF compared with the control. Scanning electron microscopy showed more eroded starch as the WBDF level increased. Fourier-transform infrared spectroscopy revealed a decrease in the starch-hydrogen binding absorbance in the 3600-3000 cm-1 wavemumber; and the 1047/1022 and 995/1022 cm-1 data indicated an increase in the degree of order and degree of double helix of the samples containing WBDF. The results of the study might help understand the interaction between dietary fibers and starch during fermentation and guide the production of fermented high-fiber flour products.

Abstract:The influences of various gelatin emulsion additions on gelatinization, rheology, retrogradation and structural properties of waxy rice starch (WRS) were investigated. The gelatin emulsion was uniformly dispersed in the starch pastes, which increased the particle size through agglomerating around the starch powder particles. As gelatin emulsion content increased, the pasting viscosity of WRS pastes significantly increased (P < 0.05), whereas its setback value, gelatinization and retrogradation enthalpy decreased. Rheological measurements indicated that the addition of gelatin emulsion did not change the shear thinning behavior of WRS pastes blends, and the storage modulus and loss modulus were positively correlated with gelatin emulsion content. During the heating stage of the pasting process, the interfacial organization consisting of gelatin undergoes disintegration and melting, reconfiguring the emulsion system. Gelatin dispersed in the interfacial and continuous phases interacts with starch to form hydrogen bonds and inhibit starch rearrangement. The starch-lipid complex formed by the interaction between lipids released from emulsion and starch is capable of destroying the crystalline region. Therefore, after 14 d of storage at 4 C-degrees, the relative crystallinity of WRS decreased by 72.4 % as the gelatin emulsion addition was increased from 0 % to 14 %. The addition of gelatin emulsion induced the formation of steric hindrance effects, which impeded the mobility of water molecules and improved freeze-thaw stability. These results may contribute to the creation of high-quality and innovative water-oil-starch mixture food products that remain fresh for a longer period.

Abstract:A synergistic fermentation system was constructed using single strains of Lactobacillus plantarum and Saccharomyces cerevisiae cultured separately; wheat starches containing different wheat bran dietary fiber (WBDF) levels (0, 3, 6, 9 & 12%) were fermented in this system. The thermal properties of materials were measured by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and rapid viscosity analysis (RVA). The results showed that WBDF may alter the thermal behavior of starch by forming hydrogen bonds with the leached starch chains and limit the available water of starch. The viscosity properties (peak, trough, and final viscosity) and setback decreased, and they were negatively correlated with the WBDF levels. In addition, dynamic rheological measurements showed that the addition of WBDF significantly enhanced the elasticity of fermented starch gels while slightly improving the mechanical strength, and 6% level of WBDF had the largest contribution. This study provides some data for the production of high dietary fiber fermented flour products, both common and gluten-free.

Abstract:Insoluble dietary fiber (IDF) were isolated from wheat bran (WB) after microbial fermentation with single or mixed strain [ Lactobacillus plantarum, Lactobacillus acidophilus, Bacillus subtilis or mixed lactic acid bacteria ( L. plantarum and L. acidophilus with ration of 1:1)]. Structure, physicochemical, functional properties, and antioxidant activity of the wheat bran insoluble dietary fiber (W-IDF) modified by fermentation were studied. Fourier transformed infrared spectroscopy (FT-IR), and scanning electron microscopy (SEM) analysis suggested the successful modification of W-IDF. After fermentation with L. plantarum and mixed lactic acid bacteria, the water retention capacity (WRC), oil retention capacity (ORC), and water swelling capacity (WSC) of W-IDF were improved. The sodium cholate adsorption capacity (SCAC), and cation exchange capacity (CEC) of W-IDF modified with L. acidophilus fermentation were significantly increased. Although the cholesterol adsorption capacity (CAC) of W-IDF decreased after modification with probiotic fermentation, nitrite ion adsorption capacity (NIAC), and total phenolic content (TPC) were enhanced. Additionally, W-IDF modified by fermentation with B. subtilis or mixed lactic acid bacteria exhibited superior antioxidant capacity verified by DPPH, ABTS and total reducing power assays. Results manifested that microbial fermentation is a promising methods to modify the W-IDF to provide high-quality functional IDF for food processing and human health management.

Effects of galactomannans of varied structural features on the functional characteristics and in vitro digestibility of wheat starch

Abstract:This study explores the effects of four natural galactomannans (GMs) with varying degrees of branching (fenugreek gum, guar gum, tara gum and locust bean gum) on the functional properties and in vitro digestibility of wheat starch (WS). Results from rapid viscosity analysis (RVA) and low-field nuclear magnetic resonance (LF-NMR) analysis revealed that GMs with lower branching degrees were correlated with higher paste viscosity, peak viscosity, and greater water-holding capacity in the WS-GM mixtures. Additionally, these lower branching GMs more effectively inhibited amylose leaching during starch gelatinization, leading to a softer gel texture and increased transparency of the mixtures. X-ray diffraction (XRD) and differential scanning calorimetry (DSC) analysis demonstrated that starch mixtures containing lower branching GMs exhibited reduced relative crystallinity and enthalpy values during aging. Furthermore, the incorporation of lower branching GMs resulted in decreased starch digestibility in vitro, thereby enhanced resistant starch content. These findings highlight the potential of selectively branched GMs to modulate the functional properties and nutritional profile of WS, providing a promising approach for the development of starch-based products with improved health benefits.

Abstract:The effects of insoluble dietary fiber (IDF) on heat-induced gelation properties, such as microstructural changes and the molecular conformation of myofibrillar proteins (MP), were analyzed by image analyses and Raman spectroscopy. Scanning electron microscopy revealed that pure MP gelation contained a loose and dispersed network with interconnected water channels. With IDF addition, the lacunarity and the particle size of water pores both significantly decreased (P < 0.05) and fractal dimension significantly increased (P < 0.05), which indicated the formation of a homogenous and compact three-dimensional network. Through Raman spectra, IDF addition resulted in modification of amide I and III regions, by a significant decrease in α-helix content, accompanied by an increase of β-sheets, β-turns, and random coil content (P < 0.05); the intensity of CU+005CH stretching vibrations bending vibrations bands were significantly decreased (P < 0.05). Principal component analyses showed significant correlations between textual property, microstructure and molecular conformations. Cluster analyses indicated IDF improved the functionality of MP gel by 1): IDF stable the moisture phase 2) "concentrated" MP promoted structural integrity and compact gelation.

Abstract:This study aimed to evaluate the effects of insoluble dietary fiber (IDF) and ferulic acid (FA) on the properties of rice starch (RS), including gelatinization, thermodynamic, rheological parameters, and freeze-thaw stability. Rapid viscosity analysis (RVA), differential scanning calorimetry (DSC), rheological analysis, and freeze-thaw stability analysis were performed. The results showed that the presence of IDF and FA could significantly delay the short-term retrogradation of RS, especially at high FA concentrations. Rheological tests showed that IDF was not conducive for the elasticity, viscosity enhancement, and system stability of the starch gels. However, FA could offset the deterioration of the system caused by IDF and further improve the gel properties. The presence of IDF and FA weakened the freeze-thaw stability of the starch gel, unlike their single action on the starch gel, correspondingly. The results show that FA could alleviate the degradation of RS gel performance caused by IDF in the ternary system. The findings provide potential possibilities for improvements in the quality of rice starch gel-based products.

Improvement of kefir fermentation on rheological and microstructural properties of soy protein isolate gels

Abstract:Soy protein isolate (SPI) has become a promising plant-based material as an animal protein products alternative. However, its application was limited due to the weak gelling properties. To investigate the effect of kefir fermentation on SPI gels properties, SPI-polysaccharide gels was produced by unfermented and kefir-fermented SPI using different concentration of KGM, chitosan, and calcium chloride in this study. Characterization of fermented SPI gels showed that fermentation by kefir grains can be applied to improve the textural strength, mechanical structure, and thermal characteristics of SPI gels. Compared to unfermented SPI gels, the water-holding capacity was remarkably enhanced to 63.11% and 65.71% in fermented SPI-chitosan gels. Moreover, the hardness of fermented SPI-KGM gels were significantly increased to 13.43 g and 27.11 g. And the cohesiveness and resilience of fermented-KGM gels were also improved than unfermented samples. Results of rheological characterization and thermogravimetric analysis revealed the strengthened mechanical features and higher thermal stability of fermented SPI gels. Additionally, the main role of hydrophobic interactions and secondary structure variations of SPI gels were demonstrated by intermolecular force measurements, Fourier-transform infrared spectroscopy, and X-ray diffraction. Moreover, the network structure was observed more compact and homogeneous performed by microstructural images in fermented SPI gels. Therefore, this research provided a novel approach combining multi-species fermentation with protein gelation to prepare SPI gel materials with improved nutrition and structural properties.

Abstract:The effect and mechanism of soybean insoluble dietary fiber (SIDF) (0~4%) and CaCl2 (0~0.005 M) on the properties of soybean protein isolate (SPI)–wheat gluten (WG) composite gel were studied. It was revealed that the addition of insoluble dietary fiber (1~2%) increased the strength and water-holding capacity (WHC) of the composite gel (p < 0.05) and enhanced the gel network structure compared with the control. WHC and LF-NMR showed that the water-binding ability of the gel system with only 2%SIDF was the strongest. The addition of excessive SIDF increased the distance between protein molecules, impeded the cross-linking of protein, and formed a three-dimensional network with low gel strength. The infrared spectrum and intermolecular force indicated that the interaction between SIDF and SPI were mainly physical, and the hydrophobic interaction and disulfide bond were the main forces in the gel system. The addition of CaCl2 can increase the critical content of gel texture destruction caused by SIDF, and the gel strength attained its peak at 3% SIDF, indicating that appropriate CaCl2 improved gel structure weakening caused by excessive SIDF. This study provides insights in enhancing the production of multi-component composite gel systems.

Abstract:In this study, the roles of kiwifruit soluble/insoluble dietary fiber (SDF/IDF, respectively) in the pasting characteristics and in vitro digestibility of wheat starch were explored. According to RVA and rheological tests, the IDF enhanced the wheat starch viscosity, decreased the gelatinization degree of the starch granules, and exacerbated starch retrogradation. The addition of SDF in high quantities could reduce the starch gelatinization level, lower the system viscosity, and exacerbate starch retrogradation. Through determining the leached amylose content and conducing scanning electron microscopy, the IDF and SDF added in high quantities was combined with the leached amylose wrapped around the starch granules, which reduced the leached amylose content and decreased the gelatinization degree of the starch granules. The Fourier transform infrared results showed that the addition of both the IDF and SDF resulted in an enhancement in hydrogen bonding formed by the hydroxyl groups of the system. The in vitro digestion results strongly suggested that both the IDF and SDF reduced the wheat starch digestibility. The above findings are instructive for the application of both IDF and SDF in starchy functional foods.

Fructooligosaccharides Impact on the Hydration and Retro-Gradation of Wheat Starch and Gel

Abstract:In the present work, the hydration and retro-gradation of mixtures of wheat starch and fructooligosaccharides in diluted hydrated matrixes were discussed. Rapid Visco-Analyzer, differential scanning calorimeter, Fourier transform-infrared spectroscopy, scanning electron microscopy, and x-ray diffraction were used to determine the properties of the mixture of wheat starch and fructooligosaccharides. Swelling of the wheat starch showed significant changes when the addition of fructooligosaccharides surpassed 0.1 g/g starch. Water absorption index showed obvious change but there was no regular variation. The water solubility index of starch increased after adding the fructooligosaccharides and the highest value was up to 19.85 as compared with the control (3.61). There was no significantly change for the pasting parameters of the mixture. Fructooligosaccharides significantly affected the enthalpy of wheat starch from the result of differential scanning calorimeter analysis, and enlarged the freezing scope of the mixture gel (from -23.09 +/- 0.25oC to -38.85 +/- 0.32oC). The melting enthalpy of sample with fructooligosaccharides decreased by 9.9 J/g as compared with the control. Fourier transform-infrared spectroscopy results showed that there were no obvious differences between the samples with or without fructooligosaccharides except that the O-H stretching vibration of hydrogen bond of the mixture gel strengthened after storage for 2 or 4 days. Scanning electron microscopy results indicated fructooligosaccharides should be conductive for starch to form fine gel structures and could inhibit the retrogradation of wheat starch. X-ray diffraction spectrum further illustrated the fructooligosaccharides could interfere with the retrogradation of wheat starch during storage.

Abstract:Communication with the biliary tree is the most frequent complication of hepatic hydatid disease. This may result in fistula formation after surgical management of liver hydatidosis. Although frank ruptures are usually diagnosed preoperatively and managed surgically, simple openings may result in biliary fistula formation. It is very difficult to accurately diagnose a small hole in the cyst during surgery in most patients. We describe an easy and reliable technique for finding the point of communication via direct visualization using a telescope during conservative surgery for hepatic hydatidosis.

Abstract:The effects of sugarcane insoluble dietary fiber (SIDF) on water holding capacity (WHC), gel strength, microstructure, and secondary structures of myofibrillar protein (MP) gels were studied. The gel strengths and WHC were improved with the increase of SIDF content. Paraffin section showed that SIDF did not have direct contact with the MP and was simply trapped in the gel network. The SEM micrograph indicated that water channels appeared through the pure MP gel network, so the integrity of gel network and the gelation quality would de deteriorated. The SIDF acted as active dehydrating agent and change the water distribution. Hence, the water channels disappeared in the internal of heat-induced gelation, which promote to aggregate compact and well-linked gel structure. The analysis of T2 relaxation revealed that T21 relaxation time of MP gel decreased significantly with SIDF addition, which indirectly suggested that the gel with SIDF had better three-dimension network and could bind the water more firmly. Raman spectroscopic study showed the SIDF addition resulted in the left shift and right shift of characteristic peak in Amide I and Amide III of the gelation, respectively. Moreover, the intensity of characteristic peak in aliphatic residues band also had a significant decrease. The changes of Raman spectroscopy reflected the SIDF addition have the positive effect on forming firm and compact MP gel.

Abstract:Modified starch was better suitable for food processing, and fermentation was one of the effective methods to modify starch. This study investigated the separate and synergistic fermentation of Lactiplantibacillus plantarum and Saccharomyces cerevisiae could whether affect the multi-scale structure and physicochemical properties of highland barley starch (HBS). The results of multi-scale structure determination of HBS showed that compared with unfermented HBS (32.27%), fermentation showed a significant increase in relative crystallinity (35.49–39.81%), while the crystal type of HBS as observed from X-ray diffraction (A-type crystalline pattern) did not change. The small angle X-ray scattering revealed that fermentation reduced the amorphous region and increased the crystalline layer thickness of the HBS. Consequently, the changes in the physicochemical properties showed that the peak viscosity, trough viscosity, and final viscosity of HBS were reduced after fermentation, and the aging of starch gel was delayed significantly. Our results showed that Lactiplantibacillus plantarum and Saccharomyces cerevisi ae fermentations improve the physicochemical properties of HBS by modifying the multi-scale structure of starch, especially the synergistic fermentation effect was more effective.

Abstract:Gelatinization, retrogradation and gel properties of wheat starch–wheat bran arabinoxylan (WS–WBAX) complexes have been evaluated. The results of rapid viscosity analyzer (RVA), differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FTIR) confirmed that WBAX samples with larger Mw and branching degree (HWBAX) significantly impeded gelatinization process of starch by effectively reducing the amount of water available for starch gelatinization. DSC analysis showed that both molecular characteristics and additive amount of WBAX samples have an effect on the long-term retrogradation behavior of starch. For the rheological studies of WS–WBAX mixed gels, the elastic moduli (G’) and shear viscosity of WS–WBAX mixed gels increased with the increase in additive amount of WBAX. WS–HWBAX mixed gels exhibited the lower G’ compared with starch gels containing WBAX with lower Mw and branching degree (LWBAX) at the same amount. The scanning electron micrographs (SEM) revealed that the microstructures of WS–WBAX mixed gels were mainly affected by the amount of WBAX, but hardly by the molecular characteristics of WBAX. Texture profile analysis (TPA) showed that the cohesiveness of fresh WS–WBAX mixed gels became larger with an increase in the WBAX addition amount. The hardness of WS–WBAX mixed gels tended to increase over the 14-day storage.

Elevated PrPC Expression Predisposes to Increased HSV-1 Pathogenicity

Abstract:PrPC is a ubiquitously expressed glycophos-phatidylinositol-linked cell-surface glycoprotein found primarily in neural tissue. Although its normal function has not been established, there is evidence suggesting that PrPC is involved in cell signalling and cellular homeostasis. This suggests that variation in neuronal expression levels of this protein contributes towards pathogenicity induced by neurotropic agents. We have investigated the pathological response to infection with herpes simplex virus type-1 (HSV-1) in strains of mice that express different levels of PrPC. Prnp−/− mice fail to express PrPC due to an interruption in the open reading frame of the Prnp gene, whilst tg19 and tga20 mice express approximately 5 and 10 times more PrPC protein, respectively, than wild-type animals. Mice that express normal or increased levels of PrPC protein were more susceptible to acute HSV-1 infection than Prnp−/− mice. Following ear pinna inoculation with HSV-1 SC16, the order of susceptibility was tga20>tg19>wild-type>Prnp−/−. This trend was reversed when latent virus was assessed. Prnp−/− mice expressed significantly higher levels of latency-associated transcript-positive neurons in various tissues when compared with wild-type, tg19 and tga20 mice. Collectively, our data show that acute HSV-1 replication proceeds more efficiently in neuronal tissue that expresses PrPC protein and lends support to the view that this protein is involved in regulation of neurotropic viral pathogenesis. This suggests that interference of PrPC expression, or possible biochemical pathways associated with its function, may serve as an effective means of limiting the pathogenesis of acute HSV-1 infection.

Abstract:The purpose of this study was to investigate the molecular interactions in debranched waxy starch and their effects on digestibility and hydrogel properties. Molecular weight distribution, molecular entanglements, hydrogel properties, water mobility and digestibility were investigated. The results showed short chain amylose released from amylopectin possessed great mobility required for molecule alignment and aggregation. Effective molecular entanglements were established in debranched waxy starch owing to the strong interactions among short chain amylose molecules. Higher storage modulus (G′) and loss modulus (G″) values indicated denser and more solid-like gels were formed in debranched waxy starch. Additionally, amylase hydrolysis of debranched waxy starch was retarded leading to the decrease of rapidly digestible starch (RDS) and the increase of slowly digestible starch (SDS). These results confirmed debranched starch shows great potential in food industry due to its digestibility and hydrogel properties.
Gel properties and formation mechanism of soy protein isolate gels improved by wheat bran cellulose

Abstract:The effect and mechanism of wheat bran cellulose (WBC) on the gelling characteristics of soy protein isolate (SPI) were evaluated. It was found that the water holding capacity, gel strength, and viscoelasticity of SPI gel were improved with the increase of WBC concentration. The addition of WBC (0.5-2.0%, w/v) stabilized the moisture phase and induced the construction of the regular and homogenous three-dimensional gel network. The Raman spectroscopy revealed that WBC addition caused a significant reduction in α-helix percentage (28.92-63.08%) (p < 0.05) with a concomitant increase in β-sheet (16.92-34.37%) (p < 0.05) and β-turn (8.09-13.54%) (p > 0.05) percentages of the pure SPI gel. Additionally, hydrogen-bonding interaction between SPI and WBC and the enhanced thermal stability were proposed in the composite gels. Overall, WBC is effective in improving the gel properties of SPI, suggesting its potential application as novel gel modifier in the food industry.
Effect of curdlan addition and thermal sterilization on the structural and properties of rice starch gel

Abstract:This study delves into the effects of curdlan integration and thermal sterilization on the rheological properties, structure, and quality attributes of concentrated rice starch gel. Acting as a heat-set polysaccharide, curdlan established a dual-network gel structure with rice starch gel, displaying strong interactions with rice starch, as confirmed by confocal laser scanning microscopy and Fourier-transform infrared spectroscopy. The addition of curdlan expedited the gel formation of rice starch, yielding a denser gel structure. Consequently, this enhanced G', solid-like behavior, textural properties, and cooking quality while reducing frequency-dependence. Given the cooling-induced gelation behavior of pure rice starch, thermal treatment disrupted inter-chain hydrogen bonding, compromising the structural integrity of the gel. This disruption manifested in a softer texture and diminished mechanical properties and cooking quality. Notably, this decline in mechanical properties and cooking quality of rice starch gel was markedly ameliorated with the incorporation of curdlan, particularly at a content of ≥1.0 %. Compared with pure RS, 1.0 % CD inclusion showed a reduction in cooking breakage rate by 30.69 % and an increase in hardness by 38.04 %. This work provides valuable insights for the advancement of fresh starch gel-based foods that exhibit exceptional quality and an extended shelf life.
