Abstract:RATIONALE Porous graphic carbon chromatography (PGC) has different mechanism in retention of tryptic peptides compared with reversed phase chromatography and in this study it was shown that coupling PGC with tandem mass spectrometry offer advantages for the quantitation of phosphorylation stoichiometry and characterization of site-specific glycosylation. METHODS Digests of protein standards (horse myoglobin, bovine fetuin and β-casein) were analyzed with a capillary LC-MS/MS system by coupling Agilent 1100 HPLC to Synapt G2-Si HDMS (Waters). Peptides were separated using a HyperCarb PGC column (300 μm i.d.× 100 mm) packing with 3 μm particles. MS/MS data were collected in data-dependent mode and 3 MS/MS scans were acquired after full MS scan. RAW data were transformed to.mgf by PLGS (Waters) and searched against Swissprot database by Mascot. Chromatogram and MS/MS spectra of identified were extracted with Masslynx (Waters) and imported to Origin for analysis. Glycan composition and peptide sequence was manually annotated. RESULTS PGC-MS/MS enabled accurate quantitation of the stoichiometry of specific phosphorylation sites from β-casein by efficient separation of the phosphopeptide and its non-phosphorylated counterpart, which cannot be achieved by reversed phase chromatography. PGC-MS/MS also enabled comprehensive characterization of protein sialoglycosylation as isomeric glycopeptides with different combination of α2-3 and α2-6 linked sialic acids can be separated and the ratio of each combination were verified by exoglycosidase digestion. CONCLUSION PGC has demonstrated superior separation of peptides with phosphorylation and glycosylation and can be used as alternative in proteomic characterization of PTMs by polar groups.

Abstract:In order to use thermoelectric energy harvesters (TEHs) as a truly autonomous energy source for size-limited sensing applications, it is essential to improve the power conversion efficiency and energy density. This study presents a thin-film, array-based TEH with a surface area of 0.83 cm2. The TEH autonomously supplies a power management IC fabricated in a 65-nm CMOS technology. The IC utilizes a single-inductor topology with integrated analog maximum power point tracking (MPPT), resulting in a 68% peak end-to-end efficiency (92% converter efficiency) and less than 20-ms MPPT. In an in-vivo test, a 645-μW regulated output power (effective 3.5 K of temperature gradient) was harvested from a rat implanted with our TEH, demonstrating true energy independence in a real environment while showing a 7.9 × improvement in regulated power density compared to the state-of-the-art. The system showed autonomous operation down to 65-mV TEH input.

Abstract:Abstract Factors which determine the line shape in the electron paramagnetic resonance (EPR) spectra of Mn(II) bound to proteins are considered. The conclusion that a reduction in the amplitude of the Mn(II) derivative spectrum in some protein complexes may be caused by restriction of motional freedom of the bound ion rather than to line broadening caused by rapid electron spin relaxation is supported by frequency and temperature dependencies of the EPR spectra for the Mn(II)-concanavalin A complex in solution. The characteristics of the solution EPR spectra show that the environment of the bound Mn(II) is not highly distorted from cubic symmetry (zero field splitting is less than 20 gauss) and that the rotational motion of the Mn(II) is highly restricted. EPR spectra at 35.0 GHz (K band) are greatly simplified relative to spectra at 9.1 GHz (X band) thus permitting more quantitative evaluation of spectral parameters.

Abstract:Human milk oligosaccharides (HMOs) exhibit various biological activities for infants, such as serving as prebiotics, blocking pathogens, and aiding in brain development. HMOs are a complex mixture of hetero-oligosaccharides that are generally highly branched, containing multiple structural isomers and no intrinsic chromophores, presenting a challenge to both their resolution and quantitative detection. While liquid chromatography-mass spectrometry (LC-MS) has become the primary strategy for analysis of various compounds, the very polar and chromophore-free properties of native glycans hinder their separation in LC and ionization in MS. Various labeling approaches have been developed to achieve separation of glycans with higher resolution and greater sensitivity of detection. Here, we compared five commonly used labeling techniques [by 2-aminobenzamide, 2-aminopyridine, 2-aminobenzoic acid (2-AA), 2,6-diaminopyridine, and 1-phenyl-3-methyl-5-pyrazolone] for analyzing HMOs specifically under hydrophilic-interaction chromatography-mass spectrometry (HILIC-MS) conditions. The 2-AA labeling showed the most consistent deprotonated molecular ions, the enhanced sensitivity with the least structural selectivity, and the sequencing-informative tandem MS fragmentation spectra for the widest range of HMOs; therefore, this labeling technique was selected for further optimization under the porous graphitized carbon chromatography-mass spectrometry (PGC-MS) conditions. The combination strategy of 2-AA labeling and PGC-MS techniques provided online decontamination (removal of excess 2-AA, salts, and lactose) and resolute detection of many HMOs, enabling us to characterize the profiles of complicated HMO mixtures comprehensively in a simple protocol.

Comparison of PGSE NMR and ESI-MS Measurements on Methylaluminoxane

Abstract:PGSE NMR and ESI-MS studies of different grades of hydrolytic MAO demonstrate that the average dimensions and anion distribution of MAO are correlated with one another, as revealed through studies of aging and gelation of a commercial formulation of MAO. Formulations featuring an anion distribution skewed to higher m/z ratios have significantly higher activator contents as measured by 1H NMR spectroscopy in otherwise very similar formulations. PSGE NMR studies in toluene vs. chlorobenzene media demonstrate that the average dimensions of MAO are largely unaffected by solvent polarity, though aggregation behavior is more pronounced in the more polar solvent. As for catalyst activation and ion-pair speciation, the situation in polar solvents is complicated by the formation of solvated cations (and anions) in chlorobenzene which dramatically lower dimensions. DFT studies of model aluminoxane structures in the size range for MAO featuring a variety of architectures, molar mass and Me3Al content reveal that a linear correlation between Dt, as measured by PGSE NMR, can be related to molar mass using simple relationships and estimates of molecular volume if suitable high molecular weight standards are available. There is a reasonable agreement in molar mass with available ESI-MS data, recognizing that MAO is not monodisperse.

Abstract:ABSTRACT Porous Graphitized Carbon nano-liquid chromatography tandem mass spectrometry (PGC-nLC-MS/MS) is a glycomics technique with the unique capacity to differentiate isobaric glycans. The lack of suitable software tools integrating chromatography and MS-information delivered by PGC-nLC-MS/MS has been limiting fast and robust glycan identification and quantitation. We report a LC-system-independent strategy called GlycoRRT that combines relative retention time (RRT) and negative ion fragment spectra analyses for isobaric structure-specific glycomics of PGC-nLC-MS/MS data. The GlycoRRT toolset is fully customizable and easily adaptable enabling semi-automated high-throughput structural assignments. The current library contains over 200 entries and their individual meta-data (MS instrumentation, experimental conditions, retention times, fragmentation profiles and glycan structural diagnostic ion features) relevant for reliable data analyses. The GlycoRRT workflow was employed to map the N - and O - glycome in blood group matched human plasma and urine as well as decipher Immunoglobulin (IgG) glycosylation features from 13 different animal species. We have also developed visualization tools to enable a consistent, reliable, and reproducible analysis of large sets of multidimensional PGC-nLC-MS/MS glycomics data. This comprehensive glycan resource provides the glycan map of human and animal species, will serve as a reference in dissecting the role of glycans in host pathogen interaction and zoonotic disease transmission.

Abstract:Characterization of glyco- and phosphoproteins as well as their modification sites poses many challenges, the greatest being loss of their signals during mass spectrometric detection due to substoichiometric amounts and the ion suppression effect caused by peptides of high abundance. We report here an optimized protocol using electrostatic repulsion hydrophilic interaction chromatography for the simultaneous enrichment of glyco- and phosphopeptides from mouse brain membrane protein digest. With this protocol, we successfully identified 544 unique glycoproteins and 922 glycosylation sites, which were significantly higher than those from the commonly used hydrazide chemistry method (192 glycoproteins and 345 glycosylation sites). Moreover, a total of 383 phosphoproteins and 915 phosphorylation sites were recovered from the sample, suggesting that this protocol has the potential to enrich both glycopeptides and phosphopeptides simultaneously. Of the total 995 glycosylation sites identified from both methods, 96% were considered new as they were either annotated as putative or not documented in the newly released Swiss-Prot database. Thus, this study could be of significant value in complementing the current glycoprotein database and provides a unique opportunity to study the complex interaction of two different post-translational modifications in health and disease without being affected by interexperimental variations.
Preparation of Carbon-Functionalized Magnetic Graphene/mesoporous Silica Composites for Selective Extraction of Miglitol and Voglibose in Rat Plasma.

Abstract:In this work, magnetic graphene/mesoporous silica composites with carbon-functionalized pore-walls (denoted as MG@mSiO(2)-C composites) were synthesized and applied as restricted access matrix solid phase extraction (RAM-SPE) adsorbents for the determination of miglitol and voglibose in rat plasma by LC-MS/MS. The MG@mSiO(2)-C composites were synthesized by using the template (Cetyltrimethyl Ammonium Bromide, CTAB) as carbon source with sulfuric acid pretreated. The obtained nano-composites were proven to have many unique properties such as large specific surface area of 277.1 cm(2) g(-1), uniform mesopores with average pore size of 3.35 nm, and carbon-functionalized pore-walls. Taking advantage of the hydrophilic interaction between carbon and glycans, a-glucosidase inhibitors (miglitol and voglibose) could be directly extracted from rat plasma with no need of other pre-treatment procedures. The SPE conditions such as the adsorbent amount, elution solvent type, adsorption time and elution time were optimized. For both miglitol and voglibose, good linearities of 10-2000 ng mL(-1) were obtained with determination coefficients (R-2) > 0.99. The intra-day and inter-day RSDs were 3.3-6.9% (n = 6) and 6.0-8.0% (n = 6), respectively. The recoveries were in the range of 99.9-100.4% and the sensitivities were as low as 2-2.5 ng mL(-1) (LOD). This MG@mSiO(2)-C composites-based RAM-SPE method offers high extraction efficiency for the determination of a-glucosidase inhibitor in plasma.

Abstract:Polyethylene glycol (PEG) has found tremendous applications in pharmaceutical products and has played a critical role in PEGylated drug modalities to improve pharmacokinetic properties and biological efficacy. The characterization and quantitation of PEGs are essential to control manufacture process and drug product quality. However, the assay value of PEG could change dramatically depending on the structures of the PEG and the detection techniques used. In this study, we developed a size exclusion chromatographic (SEC) method for quantitative PEG analysis, and we systematically evaluated the performance of three online detectors with different operating principles: a charged aerosol detector (CAD), a differential refractive index (dRI) detector, and a multi-angle light scattering detector (MALS). Fourteen PEG compounds covering a wide range of molecular weight (MW, 1 - 40 kDa) and molecular architectures (linear, branched, Y-shaped and multi-arm geometries) were evaluated by these three detection techniques. Our study revealed that the dRI showed the most universal responses among all the PEGs regardless of their molecular weight or geometries. In the contrast, CAD and MALS detector showed MW-dependent and semi-universal geometry-dependent responses. Another key finding is that the relative response factor for each multi-arm PEG in the CAD and the MALS were inversely correlated, suggesting both can be applied to qualitatively assess polymers of different architectures, including the ones with subtle differences in their core structures. The comparison of the three detectors not only provides the fundamental and comprehensive understanding of PEG quantitation but also enables the process development and control of high-quality PEGs in producing PEGylated therapeutics in the pharmaceutical industry.

Abstract:Mass spectrometry imaging using matrix-assisted laser desorption/ionization and desorption electrospray ionization has recently been employed to investigate the distribution of neurotransmitters, including biogenic amines and amino acids, directly in brain tissue sections. Ionization is facilitated by charge-tagging through pyrylium derivati-zation of primary amine containing neurotransmitters directly in tissue sections, significantly improving the limit of detection. Since the derivatization adds carbon and hydrogen to the target compounds, the resulting isotopic pat-terns of the products are not distinctive from those of the non-derivatized species. Here, we describe an approach for chemically modifying the reactive pyrylium ion to introduce the distinct isotopic signature of bromine in mass spectra of chemically derivatized substances in tissue sections. The method enables monoamine compounds to be distinguished directly in tissue sections, facilitating their identification.

Abstract:Determination of pegylated proteins' intact mass by mass spectrometry is challenging due to the molecules' large size, excessive charges, and instrument limitations. Previous efforts have been reported. However, signal variability, ion coalescence, and a generally low degree of robustness have been observed. In this work, we have explored the capabilities of post-column infusion of dimethyl sulfoxide (DMSO) following reversed-phase liquid chromatography-mass spectrometry (RP-LCMS) to determine PEG-filgrastim' intact mass, and to characterize its PEG moiety. The method was optimized around reproducibility (six preparations, and three injection replicates) with an in-house prepared PEG-filgrastim standard. The method showed a mass accuracy of ≤1.2 Da. The average molecular weight (MW EO=483 ) was 40147.9 Da. The number average molecular weight (Mn) and the weight average molecular weight (Mw) were observed to be 40101.1 and 40113.9 Da, respectively, both with an RSD of 0.03%. The molecular weight distribution of ethylene oxide (EO), the polydispersity index (PDI), was 1.0003 for all preparations with a minimum and maximum number of EO units of 448±2 and 516±2, respectively. The method was finally applied to commercially available Neulasta® lots where the Mn and Mw were 39995.8 and 40008.8 Da, respectively, both with an RSD of 0.1%. The minimum and maximum EO units across the lots were observed to be 444.5±1.5 and 514±3, respectively. The PDI for all Neulasta® lots was 1.0003. This study provides an insightful characterization of Neulasta® and describes a robust LC-MS methodology for the characterization of the PEGylated proteins.

Abstract:Human milk oligosaccharides (HMOs), the third most abundant components in breastmilk, show high structural diversity with many isomers. Here, we proposed a trapped ion mobility spectrometry-mass spectrometry (TIMS-MS) strategy for the differentiating and isomer-specific quantification of HMOs combined with Girard reagent P-d(0)/d(5) (GP-d(0)/d(5)) derivatization. Three functional HMO isomer pairs including LNT/LNnT, 3'-SL/6'-SL, and 2'-FL/3'-FL were investigated by the proposed method. The GP-derivatized HMO isomers showed enhanced ionization efficiency (8-fold) and achieved mobility separation with Delta CCS % >= 3.7% in TIMS-MS. With the GP-d(5) derivatized HMOs as an internal standard, HMOs in real samples could be quantified by derivatization with GP-d(0), and each isomeric species could be further determined based on their corresponding mobility peaks. The recoveries of spiked HMOs are obtained at 72.4 - 109%. Thus, an isomer-specific quantification strategy for HMO isomers was developed based on the GP-d(0)/d(5) derivatization and TIMS-MS.

Abstract:A hybrid material, M@C-Br@GMA@Glu, consisting of porphyrin-based metal-organic framework-covalent organic framework (MOF@COF), has been meticulously synthesized using a post-synthetic modification approach. This advanced material has demonstrated exceptional effectiveness in glycopeptide enrichment, characterized by an impressively low detection limit (0.2 fmol·μL−1), high selectivity (1:2000), and loading capacity for glycopeptides (100 mg·g−1). In practical applications with complex biological samples, 210 glycopeptides associated with 87 glycoproteins from the serum of the healthy group, and 156 glycopeptides related to 85 glycoproteins from the serum of cervical cancer were identified after enrichment with M@C-Br@GMA@Glu. Subsequent genetic ontology analysis has elucidated the relationship between cervical cancer and glycosylation, focusing on biological processes such as complement activation, innate immune response mechanisms, and the structural dynamics of the extracellular matrix. The collective findings not only validate the material's proficiency in the sensitive and selective enrichment of glycopeptides, but also underscore its potential in biomarker discovery applications.

Abstract:Glioblastoma (GBM) is a highly aggressive primary brain tumor encompassing both histological and molecular subtypes. Metabolites within the tumor microenvironment play critical roles in regulating tumor behavior and response to therapy. Understanding their spatial distribution provides valuable insights for diagnosis and treatment. We investigated metabolite patterns of histological GBMs (hist-GBMs) and molecular GBMs (mol-GBMs) by developing a novel MOF/MXene platform for enhanced surface-assisted laser desorption/ionization mass spectrometry. Utilizing this platform, we analyzed the metabolomes of three hist-GBMs and three mol-GBMs. Our platform exhibited exceptional sensitivity, repeatability, and stability in detecting metabolite molecules. Analysis of the metabolomes revealed distinct spatial distributions and concentrations of various metabolites, including carbohydrates, amino acids, organic acids, nucleotides, and their precursors. Notably, certain metabolites such as TG(15:0/20:3n6/20:5(5Z,8Z,11Z,14Z,17Z)), PG(20:3(6,8,11)-OH(5)/i-21:0), and SM(d20:1/20:3(6,8,11)-OH(5)) were significantly enriched in hist-GBMs compared to mol-GBMs, while others, including lithocholic acid glycine conjugate, PE(P-16:0/14:1(9Z)), and CE(11:1D5), exhibited higher peak intensity in mol-GBMs. Overall, our findings underscore the existence of unique metabolomic signatures distinguishing hist-GBMs from mol-GBMs. In conclusion, heterogeneous metabolite distributions among hist-GBMs and mol-GBMs were revealed by our novel MOF/MXene assisted LDI-MSI platform, and unique metabolic signatures were identified. Advanced metabolomics analysis holds promise for precise biomedical diagnostics and individualized therapeutic strategies in GBMs.

Abstract:Glutathione (γ-L-glutamyl-L-cysteinyl-glycine, γ-Glu-Cys-Gly) is the most abundant intra-cellular dicarboxylic tripeptide with multiple physiological roles. In biological samples, glutathione exists in its reduced form GSH and in two stable oxidized forms, i.e., in its symmetric disulfide form GSSG and as S-glutathionyl residue in proteins. S-Glutathionylation is a post-translational modification, which is involved in several pathophysiological processes, including oxidative stress. The GSH-to-GSSG molar ratio is widely used as a measure of oxidative stress. γ-Glutamyl is the most characteristic structural moiety of GSH. We performed gas chromatography-mass spectrometry (GC-MS) studies for the development of a highly specific qualitative and quantitative method for γ-glutamyl peptides. We discovered intra-molecular conversion of GSH, GSSG, γ-Glu-Cys and of ophthalmic acid (OPH; γ-glutamyl-α-amino-n-butyryl-glycine) to pyroglutamate (pGlu; 5-oxo-proline, also known as pidolic acid) during their derivatization with 2 M HCl/CH3OH (60 min, 80 °C). For GC-MS analysis, the methyl esters (Me) were further derivatized with pentafluoropropionic (PFP) anhydride in ethyl acetate (1:4, v/v; 30 min, 65 °C) to their PFP derivatives. At longer reaction times, pGlu is hydrolyzed to Glu. Internal standards were prepared by derivatizing GSH, GSSG, γ-Glu-Cys and OPH in 2 M HCl/CD3OD. Quantification of the Me-PFP derivative of pGlu was performed in the electron-capture negative-ion chemical ionization (ECNICI) mode by selected-ion monitoring (SIM) of the mass-to-charge (m/z) ions 269 for unlabeled pGlu (d0Me-PFP-pGlu) and m/z 272 for the in situ prepared deuterium-labeled pGlu (d3Me-PFP-pGlu). Although not inherent to the analysis of small peptides, the present GC-MS method is useful to study several biochemical aspects of GSH. Using pentafluorobenzyl bromide (PFB-Br) as the derivatization reagent, we found that synthetic pGlu is converted in aqueous acetone (60 min, 50 °C) into its pentafluorobenzyl (PFB) ester (PFB-pGlu). This derivatization procedure is useful for the GC-MS analysis of free pGlu in the ECNICI mode. Quantitative analysis of PFB-pGlu by GC-MS requires the use of stable-isotope labeled analogs of pGlu as an internal standard.

Abstract:Glycosylation is one of the most ubiquitous post-translational modifications (PTM) of proteins. Although the ionization efficiency of native glycans is fairly low, with the assistance of chemical derivation strategies, mass spectrometry (MS) has been extensively used in glycomics because of its high sensitivity, accuracy, and speed. In this study, a novel glycan labelling reagent, (4-hydrazidebutyl) triphenylphosphonium bromide (P4HZD), with a permanent positive charge was developed. The comprehensive capabilities of P4HZD for MS analysis of oligosaccharides were evaluated in detail using maltodextrin as a standard. This labelling reagent can be used in common biological MS techniques such as matrix-assisted laser desorption ionization (MALDI) and electrospray ionization (ESI) mass spectrometry. The MS signal intensity of maltodextrin species could be enhanced up to 96-fold in MALDI-MS by labelling with P4HZD, making P4HZD favorable for MALDI-MS-based high-throughput screening of oligosaccharides. Moreover, P4HZD-labelled oligosaccharides with a degree of polymerization (DP) from 1 to 18 could be separated and analysed by capillary electrophoresis (CE) combined with positive ion mode ESI-MS. In comparison with a commercialized oligosaccharide tag, Girard's reagent P (GirP), P4HZD was more effective for enhancing the signal of oligosaccharides in the middle or higher mass range using both ESI and MALDI ion sources. Two biologics, immunoglobulin G 2 (IgG 2) and fusion protein (FP), were chosen as model complex biological samples to test the efficacy of detection and separation of oligosaccharides by MALDI-MS and CE-ESI-MS analysis with P4HZD labelling. The results indicated that P4HZD is a promising labelling reagent for the detection of oligosaccharides in complex biological samples. The tandem workflow combines the strengths of MALDI-MS and CE-ESI-MS to fulfil the analytical demands of high-throughput screening, while affording good separation.

Abstract:The visualization of small metabolites by MALDI mass spectrometry imaging in brain tissue sections is challenging due to low detection sensitivity and high background interference. We present an on-tissue chemical derivatization MALDI mass spectrometry imaging approach for the comprehensive mapping of carboxyls and aldehydes in brain tissue sections. In this approach, the AMPP (1-(4-(aminomethyl)phenyl)pyridin-1-ium chloride) derivatization reagent is used for the covalent charge-tagging of molecules containing carboxylic acid (in the presence of peptide coupling reagents) and aldehydes. This includes free fatty acids and the associated metabolites, fatty aldehydes, dipeptides, neurotoxic reactive aldehydes, amino acids, neurotransmitters and associated metabolites, as well as tricarboxylic acid cycle metabolites. We performed sensitive ultrahigh mass resolution MALDI-MS detection and imaging of various carboxyl- and aldehyde-containing endogenous metabolites simultaneously in rodent brain tissue sections. We verified the AMPP-derivatized metabolites by tandem MS for structural elucidation. This approach allowed us to image numerous aldehydes and carboxyls, including certain metabolites which had been undetectable in brain tissue sections. We also demonstrated the application of on-tissue derivatization to carboxyls and aldehydes in coronal brain tissue sections of a nonhuman primate Parkinson’s disease model. Our methodology provides a powerful tool for the sensitive, simultaneous spatial molecular imaging of numerous aldehydes and carboxylic acids during pathological states, including neurodegeneration, in brain tissue.

Porous Graphitic Carbon-Based Imprint Mass Spectrometry Imaging with an Ambient Liquid Extraction Technique for Enhancing Coverage of Glycerolipids and Sphingolipids in Brain Tissue

Abstract:Localization of lipidomes and tracking their spatial changes by mass spectrometry imaging (MSI) is critical for the mechanism studies on living process, disease, and therapeutic treatment. However, due to the strong ion suppression in complex biotissue, the imaging coverage for lipids with low polarity or low abundances, such as glycerolipids and sphingolipids, is usually limited. To address this issue, we utilized a porous graphitic carbon (PGC) material to imprint brain tissue sections for selective enrichment of neutral lipids with polar phospholipids removed. Then, the tissue imprint was scanned for desorption by the ambient liquid extraction MSI system. It was found that on the PGC surface, hydrophobic interaction dominates in protic solvents, and polar interaction dominates in aprotic solvents. Accordingly, methanol was selected as the spray solvent for tissue imprinting, and 75% acetonitrile-methanol was selected as the desorption solvent for the ambient liquid extraction MSI system. The results showed that glycerides had high recoveries after the imprinting-desorption process (recovery ∼ 70%) with phospholipids eliminated (recovery < 7%). To increase the transferring efficiencies of lipids from tissue onto PGC, electrospray was used for solvent application during imprinting, and the signals of diglycerides (DGs) in the imprint MSI of brain tissue increased by 2-3 times as compared to those via air spray. Finally, the new imprint MSI approach was applied to the imaging of the rat cerebellum and was compared with direct tissue MSI. The results showed that with imprint MSI, the coverage of DGs, sphingomyelins (SMs), and ceramides was enhanced by 4-5-fold (32 vs 6, 4 vs 1, and 5 vs 0). The ion images showed that with imprint MSI, higher signal intensities and clearer spatial distribution of DGs and SMs were obtained without interference from phosphatidylcholine signals compared with tissue MSI. This new method provides a complementary approach for traditional MSI to address the issues in imaging poorly ionizable or low-abundance lipids.

Abstract:Enol-keto tautomers of methyl 3-oxo pentanoate were separated with a HP-5 capillary column by GC-MS method.The two chromatographic peaks were identified by analyzing the characteristic ions and with the aid of deuterium-labeled technology.It is found that both enol and keto tautomers are also deauterized.

Abstract:Polyethylenglycole (PEG) is a widespread linear polymer which can be utilized as a solute digestive and intestinal permeability marker in nutritional physiology studies depending on chain length/molecular mass. PEG 400 is proposed to be an ideal permeability marker. Due to its molecular mass (238-590 g/mol) and characteristics, PEG 400 is suggested to be used as a surrogate for studying the paracellular permeability of small hydrophilic molecules. For this purpose, a liquid chromatographic-tandem mass spectrometric method has been developed for the determination of the major oligomers of PEG 400 in porcine plasma. The analysis included a simple and rapid clean-up step where proteins were precipitated. The most intense ions corresponding to seven PEG 400 oligomers were separated within 7 min. Validation of the optimized method was performed in the range of 500-18,000 ng/mL. Mean recoveries between 93 and 105% were achieved using spiked plasma samples in three different concentration levels. The limit of quantification ranged between 11 and 244 ng/mL. The applicability of the method was demonstrated by the analysis of porcine plasma samples obtained from an animal experiment with barrows. The kinetic course of administrated PEG 400 was shown based on the dataset of two barrows selected from the control group, and it was figured out that relative proportion of each PEG oligomer in portal plasma decreased with increasing molecular mass.
