Abstract:Targeting Glutathione peroxidase 4 (GPX4) has become a promising strategy for drug-resistant cancer therapy via ferroptosis induction. It was found that the GPX4 inhibitors such as RSL3 have GPX4 degradation ability via not only autophagy-lysosome pathway but also ubiquitin-proteasome system (UPS). Proteolysis targeting chimeras (PROTACs) using small molecule with both inhibition and degradation ability as the ligand of protein of interest (POI) have not been reported. To obtain better compounds with effective disturbance of GPX4 activity, and compare the difference between GPX4 inhibitors with degradation ability and their related PROTACs, we designed and synthesized a series of GPX4 degraders using PROTAC technology in terms of its excellent characteristics such as high efficiency and selectivity and the capacity of overcoming resistance. Hence, 8e was discovered as a potent and highly efficacious GPX4 degrader based upon the inhibitor RSL3. It was 2–3 times more potent than RSL3 in all the in vitro anti-tumor assays, indicating the importance of the PROTAC ternary complex of GPX4, 8e and E3 ligase ligand. 8e revealed better potency in resistant tumor cells than in wide type cells. Furthermore, we discovered for the first time that degrader 8e exhibit GPX4 degradation activity via ubiquitin-proteasome system (UPS) and autophagy-lysosome pathway with UPS plays the major role in the process. Our data also suggested that 8e and RSL3 could potently induce ferroptosis of HT1080 cells via GPX4 inhibition and degradation. In summary, our data revealed that the GPX4 degrader 8e achieves better degradation and anti-tumor effects compared to its related GPX4 inhibitor RSL3. Thus, an efficient strategy to induce GPX4 degradation and subsequent ferroptosis was established in this study for malignant cancer treatment in the future.

Abstract:Glutathione peroxidase 4 (GPX4) is the most promising target for inducing ferroptosis. GPX4-targeting strategies primarily focus on inhibiting its activity or adjusting its cellular level. However, small inhibitors have limitations due to the covalent reactive alkyl chloride moiety, which could lead to poor selectivity and suboptimal pharmacokinetic properties. Herein, we designed and synthesized a series of proteolysis targeting chimeras (PROTACs) by connecting RSL3, a small molecule inhibitor of GPX4, with six different ubiquitin ligase ligands. As a highly effective degrader, compound 18a is a potent degrader (DC 50, 48h = 1.68 μM, D max, 48h = 85 %). It also showed an obvious anti-proliferative effect with the IC 50 value of 2.37 ± 0.17 μM in HT1080. Mechanism research showed that compound 18a formed a ternary complex with GPX4 and cIAP and induced the degradation of GPX4 through the ubiquitin-proteasome system pathway. Furthermore, compound 18a also induced the accumulation of lipid peroxides and mitochondrial depolarization, subsequently triggering ferroptosis. Our work demonstrated the practicality and efficiency of the PROTAC strategy and offered a promising avenue for designing degraders to induce ferroptosis in cancer cells.

Abstract:Ferroptosis is an iron-dependent, non-apoptotic form of cell death involving in various disease processes. Mechanistically, glutathione peroxidase 4 (GPX4) which belongs to the redox enzyme can convert lipid hydroperoxides into innocuous lipid alcohol to protect cells from ferroptosis. Therefore, targeting manipulation of GPX4 may represent a promising strategy for regulating cell redox homeostasis and ferroptosis. In this work, we designed, synthesized and evaluated a series of RSL3-based GPX4 degraders using PROTAC strategy. The structure-activity relationship of these compounds with different E3 ligase ligands, linker lengths and chemical compositions was systematically studied. Compound R17 with carbon chain linker and lenalidomide E3 ligand was selected as the most potent GPX4 degrader for degrading GPX4 protein in nanomolar level either in wild tumor cells or in drug-resistant tumor cells. We also optimized the POI ligand of R17 with chloracetylamine replaced to propionamide to construct noncovalent GPX4 degrader NC-R17 . Such noncovalent modification led to a moderate GPX4 degradation activity and represents a promising strategy for the development of noncovalent GPX4 PROTACs. In general, we screened a set of GPX4 degraders to give the compound R17 with excellent protein degradation activity, and further optimization gave the noncovalent degrader NC-R17 with moderate efficacy. These results lay a firm foundation for the discovery of novel anti-tumor drugs targeting GPX4 and offer the proof of concept for the design of noncovalent GPX4 PROTACs.

Abstract:Compounds that inhibit glutathione peroxidase 4 (GPX4) hold promise as cancer therapeutics in their ability to induce a form of nonapoptotic cell death called ferroptosis. Our research identified 24, a structural analog of the potent GPX4 inhibitor RSL3, that has much better plasma stability (t1/2 > 5 h in mouse plasma). The bioavailability of 24 provided efficacious plasma drug concentrations with IP dosing, thus enabling in vivo studies to assess tolerability and efficacy. An efficacy study in mouse using a GPX4-sensitive tumor model found that doses of 24 up to 50 mg/kg were tolerated for 20 days but had no effect on tumor growth, although partial target engagement was observed in tumor homogenate.

Abstract:Ferroptosis has recently emerged as a non-apoptotic form of programmed cell death and promising target for anticancer treatment. However, it is challenging to discover ferroptosis inducers with both highly selective tumour targeting and low cytotoxicity to normal cells. Here, we report an Ir(iii) complex, Ir1, that contains a novel chiral pyridine RAS-selective lethal ligand (Py-RSL). This complex effectively inhibits glutathione peroxidase 4 (GPX4) and ferroptosis suppressor protein 1 (FSP1) to induce ferroptosis in human fibrosarcoma (HT-1080) cells. Notably, metal coordination not only endows Ir1 with fluorescent properties for convenient cellular real-time tracking but also efficiently reduces the off-target toxicity of the Py-RSL ligand. Furthermore, label-free quantitative proteomic profiling revealed that Ir1 simultaneously inhibits the ErbB signalling pathway to enhance tumour suppression. Our work is the first to report a ferroptosis-inducing iridium complex with dual mechanisms of inhibition and provides a highly selective and efficient route to develop new ferroptosis-inducing metallodrugs.

Abstract:Ferroptosis is a novel form of oxidative cell death triggered by iron-dependent lipid peroxidation. The induction of ferroptosis presents an attractive therapeutic strategy for human diseases, such as prostate cancer and breast cancer. Herein, we describe our design, synthesis, and biological evaluation of endogenous glutathione peroxidase 4 (GPX4) degraders using the proteolysis targeting chimera (PROTAC) approach with the aim of inducing ferroptosis in cancer cells. Our efforts led to the discovery of compound 5i (ZX703), which significantly degraded GPX4 through the ubiquitin–proteasome and the autophagy–lysosome pathways in a dose- and time-dependent manner. Moreover, 5i was found to induce the accumulation of lipid reactive oxygen species (ROS) in HT1080 cells, thereby inducing ferroptosis. This study provides an attractive intervention strategy for ferroptosis-related diseases.

Abstract:Ferroptosis is an iron-dependent cell death caused by the accumulation of lipid peroxidation. The glutathione peroxidase 4 (GPX4) is an antioxidative enzyme and a major regulator of ferroptosis. Targeting GPX4 has become a promising strategy for cancer therapy. Here in this article, we designed and synthesized a series of GPX4 degraders using ML210 as a warhead. DC-2 among them has been found to have the best degradation activity with the DC<sub>50</sub> value of 0.03 μM in HT1080 cells. It also showed an obvious cell growth inhibition effect with the IC<sub>50</sub> value of 0.1 μM in HT1080 cells. Mechanism research showed that DC-2 induced GPX4 degradation via the ubiquitin-proteasome pathway and autophagy-lysosome pathway. GPX4 degradation induced by DC-2 could result in the accumulation of ROS and subsequent ferroptosis. The pharmacodynamics study showed that DC-2 could reduce the GPX4 level in HT1080 tumor tissue in mice and has a good safety profile. Above all, a potent and safe compound DC-2 has been found to induce GPX4 degradation and subsequent ferroptosis. This study may lay the foundation for a highly efficient and safe drug with a new mechanism for cancer therapy.

Abstract:Glutathione peroxidase 4 (GPX4) plays a crucial role in the ferroptosis pathway, emerging as a potential drug target in the treatment of refractory tumors. Unfortunately, the development of GPX4-targeted treatment has been very limited due to the poor selectivity and drug-like properties of current GPX4 inhibitors. Here, we report our proof-of-concept study of potent anti-GPX4 nanobodies, successfully identified by multiple biophysical techniques, especially various Förster resonance energy transfer strategies. Utilizing a cell-penetrating peptide fusion strategy and a newly reported thiol-related CPP additive, these high-affinity anti-GPX4 nanobodies efficiently internalized in cells and formed the basis for further application. In particular, 12E was demonstrated to selectively bind to the active site of GPX4 and significantly inhibited cellular GPX4 activity, and consequently induced remarkable ferroptosis in cancer cells. Furthermore, 12E could impair zebrafish dorsal organizer formation in vivo evidenced by a phenotype comparable to that observed in zebrafish with the gpx4b gene knocked out. The new GPX4-inhibiting nanobody described here exhibits superior proteome-wide selectivity and a vastly improved safety profile compared to existing GPX4 inhibitors. These incredible features of 12E, as an anti-GPX4 nanobody, may not only contribute to ferroptosis-related anticancer treatment but also establish a new paradigm of nanobody in drug development for traditionally undruggable targets.

Abstract:Ferroptosis is a recently discovered emerging regulated cell death pattern characterized by iron-dependent lipid peroxidation and has enormous potential in tumor therapy. However, the antioxidant defense system during tumor cell metabolism hampers the ferroptosis-mediated anticancer effect, especially the GSH/GPX4 pathway, which is regarded as the classic means of scavenging excessive lipid peroxidation production. Here, we developed an iron-based nanodrug (ipFFR) with efficient Fenton catalytic activity and encapsulated the ferroptosis inducer RSL3. This formulation consists of Fe@Fe 3 O 4 nanoparticles and RSL3, encapsulated in nanoliposomes containing DSPE-PEG-iRGD for precise tumor-targeted delivery of ipFFR. The ultrasmall Fe@Fe 3 O 4 metal nanoparticles were enabled to catalyze endogenous H 2 O 2 and produce toxic •OH under weakly acidic condition and were then quickly excreted from the body. Furthermore, the released ferroptosis inducer RSL3 specifically inhibited the activity of glutathione peroxidase 4 (GPX4) to block the antioxidant pathway of ferroptosis, synergizing and thereby enhancing ferroptosis therapy. This study may open up new avenues in designing nanomedicines with synergistic effects to enhance ferroptosis therapy.

Abstract:Ferroptosis is a regulated cell death pathway which depends on iron. Ferroptosis can be induced by limiting intracellular glutathione (GSH) synthesis, or inhibiting the activity of GPX4, or increasing intracellular accumulation of PE-AA-OOH, all of which involve NADPH. Therefore, NADPH depletion, excessive PE-AA-OOH, and GPX4 deficiency are generally considered to be the main characteristics of ferroptosis. In this research, the novel self-assembly nanomicelles modified by maltose ligand (Malt-PEG-Abz@RSL3) with superior nano characteristics were designed and fabricated. Malt-PEG-Abz@RSL3 micelles achieved active targeted drug delivery due to the high expression of glucose transporter (GLUT) and high uptake by HepG2 cells. Maltose-polyethylene glycol broke to release RSL3 for inhibiting GPX4 activity when Malt-PEG-Abz@RSL3 micelles entered the cells. Meanwhile, key coenzyme NADPH that participated in synthesis of GSH and Trx(SH)2 was depleted by azobenzene moiety, resulting in decreasing GSH and Trx(SH)2, which dually induced ferroptosis in tumour cells and promoted cell apoptosis.

Abstract:Targeted degradation of proteins, especially those regarded as undruggable or difficult to drug, attracts wide attention to develop novel therapeutic strategies. Glutathione peroxidase 4 (GPX4), the key enzyme regulating ferroptosis, is currently a target with just covalent inhibitors. Here, we developed a targeted photolysis approach and achieved efficient degradation of GPX4. The photodegradation-targeting chimeras (PDTACs) were synthesized by conjugating a clinically approved photosensitizer (verteporfin) to noninhibitory GPX4-targeting peptides. These chimeras selectively degraded the target protein in both cell lysates and living cells upon red-light irradiation. The targeted photolysis of GPX4 resulted in dominant ferroptotic cell death in malignant cancer cells. Moreover, the dying cells resulting from the PDTACs exhibited potent immunogenicity in vitro and efficiently elicited antitumor immunity in vivo. Our approach therefore provides a novel method to induce GPX4 dysfunction based on noncovalent binding and specifically trigger immunogenic ferroptosis, which may boost the application of ferroptosis in cancer immunotherapy.

Abstract:The hazards of exposure to formaldehyde have been widely discussed in recent years on account of the health complaints of exposed persons. Both domestic and occupational exposure to formaldehyde is common. In the present study the effects of formaldehyde alone and in combination with wood dust, another nasal irritant, on the upper and lower respiratory tract have been investigated. The effects were correlated with the duration and degree of exposure. Discomfort from both the upper and lower airways was more frequent in the exposed group than in the unexposed group. Nasal obstruction among the exposed subjects in general could not be detected by rhinomanometric measurements, but, for the group exposed to formaldehyde alone and with a history of nasal blocking, there was significant nasal mucosal swelling. The nasal mucociliary clearance was significantly delayed, and the sense of smell was significantly reduced in the exposed groups when they were compared with the referents. Spirometry showed a significantly decreased forced vital capacity in the exposed groups. There were no signs in this investigation that duration of exposure or level of exposure to formaldehyde would have any influence on the severity of symptoms or the impairment of physiological conditions.

In this paper we have designed and implemented a research prototype web service based e-ATM and medicine searching services in .Net and Java technology respectively. We have performed a relative evaluation on availability and reliability under massive loads for each service. Web service testing predicts the correct and robustness of services before deployment. In this perspective we present a framework of testing the services, statistical analysis over the service metrics for estimation and prediction of availability and reliability of the proposed services.

Abstract:Ferroptosis is a novel type of regulated cell death characterized by the accumulation of lipid peroxides to lethal levels. Most tumor cells are extremely vulnerable to ferroptosis due to the high levels of reactive oxygen species (ROS) produced by their active metabolism. Therefore, tumor cells rely on glutathione (GSH) to reduce lipid peroxides catalyzed by glutathione peroxidase 4 (GPX4), and this pathway is also an important target for a variety of drugs that promote tumor cell ferroptosis. Herein, RSL3@PCA was designed to simultaneously deplete intracellular GSH and inhibit the activity of GPX4, thereby significantly promoting tumor cell ferroptosis. RSL3@PCA was successfully prepared by encapsulating a selective inhibitor of GPX4 into acid-responsive nanoparticle PCA. After being taken up by tumor cells, the acid-responsive nanoparticle gradually degraded to release cinnamaldehyde (CA) and the encapsulated RSL3. CA and RSL3 block the reduction of lipid peroxides in cells, thereby inducing ferroptosis. By a cytotoxicity assay and 4T1 cell xenotransplantation model, we confirmed that RSL3@PCA has excellent inhibition of tumor growth without significant toxicity to normal cells and tissues and still has a good therapeutic effect on tumor cells that are resistant to conventional chemotherapy drugs. This work provides new drug combinations for promoting ferroptosis in tumor cells without severe side effects in normal organs.

Abstract:Ferroptosis, as a uniquely regulated form of cell death, has garnered significant interest in the field of oncology. Most ferroptosis inducers facilitate the intracellular buildup of lipid peroxidation by inhibiting glutathione peroxidase 4 (GPX4). However, the high saturation level of membrane lipids can shield cancer cells from reactive oxygen species-induced damage, preventing lipid peroxidation triggered by GPX4 inhibitors. Herein, we precisely constructed a lipid nanoregulator of RSL3 and Orlistat through a molecular engineering strategy. In this nanosystem, Orlistat functions as a “dual enhancer”, significantly enhancing both the assembly capability and the anti-tumor effectiveness of RSL3. Orlistat can significantly enhance the synthesis of polyunsaturated fatty acids and reduce the saturation level of lipid membranes by inhibiting fatty acid synthase (FASN). Consequently, Orlistat successfully dismantles the final defense (lipid membrane protection), synergistically amplifying ferroptosis with RSL3-induced GPX4 depletion. As expected, the lipid nanoregulator demonstrates a potent antitumor activity in 4T1 tumor-bearing mice. This study represents an effective paradigm for ferroptosis-driven antitumor nanotherapeutics.
Novel Allosteric Activators for Ferroptosis Regulator Glutathione Peroxidase 4.

Abstract:Glutathione peroxidase 4 (GPX4) is essential for cell membrane repair, inflammation suppression, and ferroptosis inhibition. GPX4 upregulation provides unique drug discovery opportunities for inflammation and ferroptosis-related diseases. However, rational design of protein activators is challenging. Until now, no compound has been reported to activate the enzyme activity of GPX4. Here, we identified a potential allosteric site in GPX4 and successfully found eight GPX4 activators using a novel computational strategy and experimental studies. Compound 1 from the virtual screen increased GPX4 activity, suppressed ferroptosis, reduced pro-inflammatory lipid mediator production, and inhibited NF-kappa B pathway activation. Further chemical synthesis and structure activity relationship studies revealed seven more activators. The strongest compound, 1d4, increased GPX4 activity to 150% at 20 mu M in the cell-free assay and 61 mu M in cell extracts. Therefore, we demonstrated that GPX4 can be directly activated using chemical compounds to suppress ferroptosis and inflammation. Meanwhile, the discovery of GPX4 activators verified the possibility of rational design of allosteric activators.
Discovery of Novel Potent Covalent Glutathione Peroxidase 4 Inhibitors as Highly Selective Ferroptosis Inducers for the Treatment of Triple-Negative Breast Cancer

Abstract:Glutathione peroxidase 4 (GPX4) is a promising target to induce ferroptosis for the treatment of triple-negative breast cancer (TNBC). We designed and synthesized a novel series of covalent GPX4 inhibitors based on RSL3 and ML162 by structural integration and simplification strategies. Among them, compound C18 revealed a remarkable inhibitory activity against TNBC cells and significantly inhibited the activity of GPX4 compared to RSL3 and ML162. Moreover, it was identified that C18 could notably induce ferroptosis with high selectivity by increasing the accumulation of lipid peroxides (LPOs) in cells. Further study demonstrated that C18 covalently bound to the Sec46 of GPX4. Surprisingly, C18 exhibited an outstanding potency of tumor growth inhibition in the MDA-MB-231 xenograft model with a TGI value of 81.0%@20 mg/kg without obvious toxicity. Overall, C18 could be a promising GPX4 covalent inhibitor to induce ferroptosis for the treatment of TNBC.
