Abstract:This study aimed to investigate the antioxidant effect of rosiglitazone (ROG) and pioglitazone (POG) on oxidative damage and dysfunction of hepatic tissue in hypothyroid rats. The male rats were classified into six groups: (1) Control; (2) Hypothyroid, (3) Hypothyroid-POG 10, (4) Hypothyroid-POG 20, (5) Hypothyroid-ROG 2, and (6) Hypothyroid-ROG 4. To induction hypothyroidism in rats, propylthiouracil (PTU) (0.05 %w/v) was added to drinking water. In groups 2–6, besides PTU, the rats were also intraperitoneal administrated with 10 or 20 mg/kg POG or 2 or 4 mg/kg ROG for six weeks. Finally, after deep anesthesia, the blood was collected to measure the serum biochemical markers and hepatic tissue was separated for biochemical oxidative stress markers. Administration of PTU significantly reduced serum thyroxin concentration, total thiol levels, activity of superoxide dismutase (SOD) and catalase (CAT) enzymes, and increased serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (Alk-P) and malondialdehyde (MDA) in the liver. Additionally, our results showed that prescription of POG or ROG for six weeks to hypothyroid rats resulted in an improvement in liver dysfunction (decrease in serum levels of AST, ALT, and ALK-P) through reducing oxidative damage in hepatic tissue (increase in CAT, SOD, or total thiols and decrease in MDA levels). The findings of the present study presented that the IP administration of POG and ROG for six weeks improves liver dysfunction induced by hypothyroidism in juvenile rats by reducing oxidative damage.
Effects of Rosiglitazone on Serum Levels of Lipids and Inflammatory Factors in Atherosclerotic Rats

Abstract:OBJECTIVE To explore the effects of rosiglitazone on serum levels of lipids and inflammatory factors in atherosclerotic rats.METHODS 55 male Wistar rats were randomly divided into 3 groups,the normal group(N,n=20) with ordinary diet for 12 weeks,the model group(M,n=20) and the treatment group(RSG,n=15) with intra-abdominal injection of a single dose of vitamin D3(400,000 IU·kg-1) and atherogenic diet for 6 weeks,then be administered with normal saline(10 mg·kg-1·d-1) and rosiglitazone(3 mg·kg-1·d-1),respectively for 6 weeks.Before and after treatment the serum levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein-cholesterol(LDL-C),high density lipoprotein-cholesterol(HDL-C) and pathomorphology in heart tissue were detected.The levels of serum C-reactive protein(CRP),monocyte chemoattractant protein-1(MCP-1) and intercellular adhesion molecule-1(ICAM-1) were determined by ELISA. RESULTS 6 weeks later the model group rats showed high serum levels of TC,TG,LDL-C,CRP,MCP-1,ICAM-1 and low serum level of HDL-C than that of the normal group(P0.05).Coronary atherosclerotic plaques had been observed.After treatment for 6 weeks,compared with the model group rats,rosiglitazone could significantly reduce the levels of serum TC,TG,LDL-C,CRP and MCP-1(P0.05 or P0.01),which were still higher than that of the normal group(P0.05).However,the changes of the serum HDL-C and ICAM-1 levels has no difference(P0.05).The pathomorphology degree of coronary atherosclerosis has decreased significantly.CONCLUSION Rosiglitazone can reduce the serum levels of TC,TG,LDL-C and inhibit the production of CRP and MCP-1,which may account for the anti-arterioisclerosis effects.
Rosiglitazone Via Upregulation of Akt/Enos Pathways Attenuates Dysfunction of Endothelial Progenitor Cells, Induced by Advanced Glycation End Products

Abstract:Background and purpose:Advanced glycation end products (AGEs) and endothelial progenitor cells (EPCs) play key roles in pathogenesis of diabetes-related vascular complications. AGEs can induce dysfunction in EPCs. The peroxisome proliferator-activated receptor-gamma (PPAR gamma) agonists are widely used in the treatment of type 2 diabetes, and it remains unknown if they could attenuate EPC dysfunction induced by AGEs.Experimental approach:EPCs isolated from healthy adults were cultured with various concentrations of AGEs (0, 50, 100 and 200 mg center dot L-1) with or without rosiglitazone (10 nM), antibody for the receptors for AGE-human serum albumin (anti-receptor for advanced glycation end products (RAGE); 50 mu g center dot mL-1), phosphatidylinositol-3-kinase (PI3K) inhibitor (LY294002, 5 mu M), nitric oxide (NO) synthase inhibitor (L-NG-nitro-arginine methyl ester (L-NAME), 100 mu M) or sodium nitroprusside (SNP, 25 mu M). Proliferation, apoptosis, cell adhesion, migration and NO production in EPCs were assessed, and expressions of endothelial NO synthase (eNOS) and Akt were determined.Key results:Number, proliferation/migration capacities, eNOS and Akt phosphorylation as well as NO synthesized by EPCs were increased by rosiglitazone and reduced by AGEs. AGEs promoted while rosiglitazone reduced EPC apoptosis. The AGE-induced effects were significantly ameliorated by pre-incubation with rosiglitazone, RAGE antibody and SNP. The beneficial effects of rosiglitazone could be blocked by pretreatment with L-NAME and LY294002.Conclusions and implications:The PPAR gamma agonist rosiglitazone increased EPC function and attenuated EPC dysfunction induced by AGEs via upregulating the Akt-eNOS signal pathways of EPCs.

Abstract:Objective To investigate whether peroxisome proliferator-activated receptor-gamma(PPARγ)agonists rosiglitazone can ameliorate the dysfunction of endothelial progenitor cells(EPCs)induced by advanced glycation end products(AGEs).Methods EPCs isolated from healthy adults were cultured with various concentrations of advanced glycation end product-human serum albumin(AGE-HSA)(0,50,100 and 200 mg/L)for 24 h in the absence or presence of rosiglitazone(10 nmol/L),anti-RAGE antibody(50 mg/L).We also included a blank control group.The proliferation,apoptosis,cell adhesion,migration,NO production and vascular cell adhesion molecule(VCAM)-1 secretion by EPCs were assessed.Results AGE-HSA at 200 mg/L significantly impaired the proliferation(reduced by [37.7±6.3]% compared with control,P<0.05),adherent([35.6±1.5]/HPF vs.[20.2±1.3]/HPF,P<0.05),and migration([28.6±2.8]/HPF vs.[15.0±2.1]/HPF,P<0.05)of EPCs,and aggravated the apoptosis([4.6±0.8]% vs.[15.2±1.0]%,P<0.05)of EPCs.Rosiglitazone significantly reversed the impairment effect of AGE-HSA on the proliferation(increased to [89.3±5.9]%,P<0.05),adherent(increased to [31.2±2.6]/HPF,P<0.05)and migration(increased to [21.6±3.1]/HPF,P<0.05)of EPCs,and ameliorated the apoptosis(reduced to [6.1±0.9]%,P<0.05)of EPCs.Rosiglitazone also increased the production of NO([16.2±1.0]μmol/L vs.[10.2±0.5] μmol/L,P<0.05)and reduced the secretion of VCAM-1([5.2±0.04] μg/L vs.[6.2±0.1] μg/L,P<0.05)of EPCs.Conclusion PPARγ agonist rosiglitazone ameliorates the dysfunction of endothelial progenitor cells induced by advanced glycation end products.

Abstract:Thyroid hormone is an important factor for proper development of the mammalian brain. Perinatal hypothyroidism leads to long-term behavior and neuromotor competence alterations in humans and animals. Our study aimed to investigate the effects of perinatal hypothyroidism on behavior changes of rat pups and its relation with the apoptosis of hippocampus neurons. Behavior tests were taken to evaluate the effects caused by perinatal hypothyroidism. TUNEL staining was used to analyze the apoptosis of neurons on CA3 region of hippocampus. The study suggested that perinatal hypothyroidism affects behavior development, as well as leading to the decrease in spatial learning and memory capability. This condition can be improved with hormone substitute treatment. Furthermore, the changes of learning and memory capability are closely related to the increasing number of apoptotic neurons in the hippocampus.
Effects of PI3K/Akt signaling pathway on learning and memory abilities in neonatal rats with hypoxic-ischemic brain damage

Abstract:Objective: To study the effects of PI3K/Akt signaling pathway inhibitor wortmannin on long-term learning and memory abilities in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods: Forty-eight neonatal rats were randomly assigned to blank control (n = 8), sham-operated (n = 8), HIBD model (n = 10), HIBD + DMSO (dimethyl sulfoxide, n = 8) and HIBD + wortmannin groups (n = 8). Wortmannin (2 μL) was injected to the left hippocampus 30 minutes before HIBD inducement in the HIBD + wortmannin group. The Morris water maze test was used to examine the long-term learning and memory abilities at the age of 28 days. Results: With the increased number of swimming, the escape latency was shortened in various groups. From the second day, the escape latency in the HIBD + wortmannin group was significantly longer than that in the sham-operated and the blank control groups (P < 0.05), and the differences increased with the time. On the fourth day, there were significant differences in the escape latency between the HIBD + wortmannin group and the HIBD + DMSO group as well as the HIBD model group (P <0.05). On the eighth day (retention trial) , there were the most obvious differences in the escape latency between the HIBD + wortmannin group with the other four groups. In the space exploration test, the number of times crossing the former platform location within 120 seconds after removing the platform in the HIBD + DMSO and the HIBD model group was lower than the sham-operated and the blank control groups (P <0.05). The HIBD + wortmannin group showed lower number of times crossing the former platform location compared with the HIBD + DMSO and the HIBD model groups (P < 0.05), as well as the sham-operated and the blank control groups (P <0.01). Conclusions: P13K/Akt signaling pathway inhibitor wortmannin can aggravate the cognitive impairments, thus affecting adversely long-term learning and memory abilities in neonatal rats with HIBD.

Abstract:The aim of our studies is to determine the possible therapeutic benefit of using thiazolidinediones (TZD), selective agonists of Peroxisome Proliferator Activated Receptor isoform gamma (PPAR-c) in the treatment of brain tumors. In view of our findings that TZDs induce substantial metabolic changes in astrocytes and PPAR-c agonists have been shown to induce apoptosis in some tumor cells, we tested the effect of two TZDs, troglitazone (TRO) and pioglitazone (PIO) on the survival of primary rat astrocytes and glioma cell lines C6 and GL261. Treatment with TRO significantly inhibited cell proliferation in glioma cells in a dose and time-dependent manner. Measurement of LDH release showed the growth arrest was associated with cell death. A reduced cytotoxicity was observed in PIO-treated compared to TRO-treated glioma cells suggesting a PPAR-c independent mechanism, since PIO is more effective than TRO to activate the receptor. In contrast, in nontransformed primary rat astrocytes both TZDs did not show cytotoxic effects. We characterized TRO-induced cell death as apoptotic since the presence of a pan-caspase inhibitor blocked TRO-induced LDH release. As further proof we found TRO caused caspase 3 activation. We extended these studies in vivo by implanting GL261 cells intracerebrally in adult C57BL/6 mice and providing themwith control or pioglitazone diet. The survival ratewith PIO treatmentwas improved vs. controls. Our results highlight the potential of TZDs as novel chemotherapeutic agents for the treatment of gliomas. Abstract:AIM:To investigate the expression of renal peroxisome proliferator-activated receptor gamma(PPARγ)in rats with adrimycine nephrosis(ADR),and the effect of rosiglitazone on the activation of NF-κB p65 in renal tissue rats with ADR.METHODS:The rats were randomly assigned to following groups:control(CTR)group,adrimycine nephrosis(ADR)group,and ADR treated with rosiglitazone(5 mg·kg-1·d-1)group(RGL).The levels of urinary protein,albumin,total cholesterol,triglyceride and renal function change in rats were measured after 12 weeks.The nuclear-translocation of cortical NF-κB p65 was detected by immunohistochemistry.The activity of cortical NF-κB p65 was measured by sandwich ELISA.The mRNA levels of cortical PPARγ and TGF-β1 were detected by RT-PCR.The protein expressions of PPARγ and TGF-β1 in the rat kidney tissues were detected by Western blotting.RESULTS:As compared to ADR group,the urinary protein excretion in RGL treatment group was decreased and the serum albumin levels were increased,but the serum total cholesterol and triglyceride were decreased and the renal pathological lesion was ameliorated.The activity of NF-κB p65 and the expressions of TGF-β1 mRNA and protein were significantly decreased in rosiglitazone group,while the expression of PPARγ mRNA and protein was increased in RGL group(P<0.01).The correlation analysis was manifested:in ADR and RGL group,a negative correlation between the activity of NF-κB p65 and the expression of PPARγ in renal tissue(r=-0.8305,P<0.01)was observed.There was a negative correlation between the expression of TGF-β1 and PPARγ in renal tissues(r=-0.7938,P<0.01).CONCLUSION:The expression of renal cortical PPARγ is up-regulated in rats with adrimycine nephrosis by rosiglitazone.Rosiglitazone inhibits the activation of renal cortical NF-κB p65 in part,so it inhibits the gene expression of renal TGF-β1 and relieves the renal pathological lesion.

Abstract:Peroxisome proliferator-activated receptor gamma (PPAR-γ) agonists have been suggested to have some effects of central nervous system. In the present study, the effects of PPAR-γ agonist pioglitazone (pio) on anxiety and depression-like behaviors were investigated in lipopolysaccharide (LPS) injected rats. The animal groups include (n = 10 in each): (1) control, (2) LPS (1 mg/kg, two hours before behavioral tests), (3–5) LPS + Pio 10, LPS + Pio 20, and LPS + Pio 30 groups which received 10, 20, and 30 mg/kg of pio respectively and 30 min later were injected by LPS. Pretreatment by pio improved the behaviors of the LPS injected rats in open field, elevated plus maze and forced swimming tests protected from brain tissues oxidative damage and reduced in total white blood cell (WBC). It was indicated that pio had anti-anxiety and anti-depressant effect in LPS injected rats.

Abstract:Aim:To investigate the protective effect of pioglitazone on intracerebral hemorrhage in rats and its possible mechanism.Methods: A total of 72 male SD rats were divided randomly into six groups.Intervention group 1(n=12): Pioglitazone peroxisome proliferator-activated receptor gamma(PPARγ activator) were gastrically administrated 6 h before modeling,and the original dosages were maintained after modeling.Intervention group2(n=12): Pioglitazone were gastrically administrated 6 h before modeling,and were withdrawn after modeling.Intervention group 3(n=12): The drugs were not given before modeling,and were gastrically administrated aftermodeling.Model group 4(n=12): The drugs were not given before and after modeling.Sham operation group(n=12): The rats were performed with needling insertion without injection.Normal group(n=12) was set up.Thedose of gastrically administrated pioglitazone was 15 mg.kg-1.d-1.Rats in each group were sacrificed at 72 h afteroperation.Biochemical detection of the superoxide dismutase(SOD) and malondialdehyde(MDA) concentration of brain tissue were performed.The Western blot was used to determine the PPARγ and caspase-3 proteinexpression in brain tissue.HE and TUNEL staining were used to observe the brain edema and apoptosis respectively.Results: PPARγ activator pioglitazone could significantly reduce the extent of brain edema and inflammatory cellinfiltration of the brain tissue in rats at 72 h after cerebral hemorrhage,as well as reduce the number of apoptoticcells in brain tissue,and enhance the SOD activity and increase the expression of PPARγ,and caspase-3 proteinin brain tissue,and inhibit the MDA levels.The effect of prophylactic administration of pioglitazone beforecerebral hemorrhage was better than that of the treatment after hemorrhage.Conclusion: PPARγ activatorpioglitazone can relieve the hemorrhagic brain injury in the rat brain,and protect the brain,possibly by increasingthe PPARγ protein expression in brain tissue and by enhancing the SOD activity,inhibiting the free radicals levelsin brain tissue and the apoptosis of brain cells.
Pioglitazone Ameliorates Lipopolysaccharide-Induced Behavioral Impairment, Brain Inflammation, White Matter Injury and Mitochondrial Dysfunction in Neonatal Rats

Abstract:Previous studies have demonstrated that pioglitazone, a peroxisome proliferator-activated receptor gamma (PPARγ) agonist, inhibits ischemia-induced brain injury. The present study was conducted to examine whether pioglitazone can reduce impairment of behavioral deficits mediated by inflammatory-induced brain white matter injury in neonatal rats. Intraperitoneal (i.p.) injection of lipopolysaccharide (LPS, 2 mg/kg) was administered to Sprague-Dawley rat pups on postnatal day 5 (P5), and i.p. administration of pioglitazone (20 mg/kg) or vehicle was performed 5 min after LPS injection. Sensorimotor behavioral tests were performed 24 h after LPS exposure, and changes in biochemistry of the brain was examined after these tests. The results show that systemic LPS exposure resulted in impaired sensorimotor behavioral performance, reduction of oligodendrocytes and mitochondrial activity, and increases in lipid peroxidation and brain inflammation, as indicated by the increment of interleukin-1β (IL-1β) levels and number of activated microglia in the neonatal rat brain. Pioglitazone treatment significantly improved LPS-induced neurobehavioral and physiological disturbances including the loss of body weight, hypothermia, righting reflex, wire-hanging maneuver, negative geotaxis, and hind-limb suspension in neonatal rats. The neuroprotective effect of pioglitazone against the loss of oligodendrocytes and mitochondrial activity was associated with attenuation of LPS-induced increment of thiobarbituric acid reactive substances (TBARS) content, IL-1β levels and number of activated microglia in neonatal rats. Our results show that pioglitazone prevents neurobehavioral disturbances induced by systemic LPS exposure in neonatal rats, and its neuroprotective effects are associated with its impact on microglial activation, IL-1β induction, lipid peroxidation, oligodendrocyte production and mitochondrial activity.
Effects of Low Doses of Pioglitazone on Resting-State Functional Connectivity in Conscious Rat Brain

Abstract:Pioglitazone (PIO) is a peroxisome proliferator-activated receptor-γ (PPARγ) agonist in clinical use for treatment of type 2 diabetes (T2DM). Accumulating evidence suggests PPARγ agonists may be useful for treating or delaying the onset of Alzheimer's disease (AD), possibly via actions on mitochondria, and that dose strengths lower than those clinically used for T2DM may be efficacious. Our major objective was to determine if low doses of pioglitazone, administered orally, impacted brain activity. We measured blood-oxygenation-level dependent (BOLD) low-frequency fluctuations in conscious rats to map changes in brain resting-state functional connectivity due to daily, oral dosing with low-dose PIO. The connectivity in two neural circuits exhibited significant changes compared with vehicle after two days of treatment with PIO at 0.08 mg/kg/day. After 7 days of treatment with a range of PIO dose-strengths, connections between 17 pairs of brain regions were significantly affected. Functional connectivity with the CA1 region of the hippocampus, a region that is involved in memory and is affected early in the progression of AD, was specifically investigated in a seed-based analysis. This approach revealed that the spatial pattern of CA1 connectivity was consistent among all dose groups at baseline, prior to treatment with PIO, and in the control group imaged on day 7. Compared to baseline and controls, increased connectivity to CA1 was observed regionally in the hypothalamus and ventral thalamus in all PIO-treated groups, but was least pronounced in the group treated with the highest dose of PIO. These data support our hypothesis that PIO modulates neuronal and/or cerebrovascular function at dose strengths significantly lower than those used to treat T2DM and therefore may be a useful therapy for neurodegenerative diseases including AD.

Abstract:The objective was to evaluate the efficacy and safety of recombinant activated factor VII in patients with massive bleeding. Forty-five patients with severe massive hemorrhage requiring= 14 transfusion units of packed red blood cells received recombinant activated factor VII. Postdrug blood loss and transfusion requirements were assessed, and mortality was compared with predicted outcomes. Blood loss was markedly reduced in 40 of 43 (93.0%) patients, and transfusion requirements decreased after recombinant activated factor VII administration. Mortality rate in trauma patients who had massive hemorrhage was significantly reduced compared with predictions using scoring systems. This may be associated with the use of recombinant activated factor VII. This study failed to demonstrate an improvement in surgical patients. The absence of concurrent controls prevents definitive conclusions regarding actual safety or efficacy of recombinant activated factor VII.
Effects of long-term treatment with pioglitazone on cognition and glucose metabolism of PS1-KI, 3xTg-AD, and wild-type mice

Abstract:In this study, we investigated the effects of long-term (9-month) treatment with pioglitazone (PIO; 20 mg/kg/d) in two animal models of Alzheimer’s disease (AD)-related neural dysfunction and pathology: the PS1-KIM146V (human presenilin-1 M146V knock-in mouse) and 3xTg-AD (triple transgenic mouse carrying AD-linked mutations) mice. We also investigated the effects on wild-type (WT) mice. Mice were monitored for body mass changes, fasting glycemia, glucose tolerance, and studied for changes in brain mitochondrial enzyme activity (complexes I and IV) as well as energy metabolism (lactate dehydrogenase (LDH)). Cognitive effects were investigated with the Morris water maze (MWM) test and the object recognition task (ORT). Behavioral analysis revealed that PIO treatment promoted positive cognitive effects in PS1-KI female mice. These effects were associated with normalization of peripheral gluco-regulatory abnormalities that were found in untreated PS1-KI females. PIO-treated PS1-KI females also showed no statistically significant alterations in brain mitochondrial enzyme activity but significantly increased reverse LDH activity.PIO treatment produced no effects on cognition, glucose metabolism, or mitochondrial functioning in 3xTg-AD mice. Finally, PIO treatment promoted enhanced short-term memory performance in WT male mice, a group that did not show deregulation of glucose metabolism but that showed decreased activity of complex I in hippocampal and cortical mitochondria. Overall, these results indicate metabolically driven cognitive-enhancing effects of PIO that are differentially gender-related among specific genotypes.

Abstract:There is increasing interest in the association between depression and the development of metabolic diseases. Rosiglitazone, a therapeutic drug used to treat type 2 diabetes mellitus, has shown neuroprotective effects in patients with stroke and Alzheimer's disease. The present study was performed to evaluate the possible roles of rosiglitazone in in vivo (unpredictable chronic mild stress-induced depressive mouse model) and in vitro (corticosterone-induced cellular model) depressive models. The results showed that rosiglitazone reversed depressive behaviors in mice, as indicated by the forced swimming test and open field test. Rosiglitazone was also found to inhibit the inflammatory response, decrease corticosterone levels, and promote astrocyte proliferation and neuronal axon plasticity in the prefrontal cortex of mice. This series of in vivo and in vitro experiments showed that autophagy among neurons was inhibited in depressive models and that rosiglitazone promoted autophagy by upregulating LKB1, which exerted neuroprotective effects. Rosiglitazone was also found to activate the Akt/CREB pathway by increasing IGF-1R expression and IGF-1 protein levels, thereby playing an anti-apoptotic role in astrocytes. Rosiglitazone's autophagy promotion and neuroprotective effects were found to be reversed by the PPARγ antagonist T0070907 in primary neurons and by PPARγ knockdown in an N2a cell line. In conclusion, we found that rosiglitazone protects both neurons and astrocytes in in vivo and in vitro depressive models, thereby playing an anti-depressive role. These findings suggest that PPARγ could be a new target in the development of anti-depressive drugs.

Abstract:In this study, we determined how rosiglitazone (RSG) differentially affected hippocampal neurogenesis in mice fed a low-fat diet (LFD) or high-fat diet (HFD; 60% fat). LFD and HFD were given to the mice for 8 weeks. Four weeks after initiating the LFD and HFD feeding, vehicle or RSG was administered orally once a day to both groups of mice. We measured cell proliferation and neuroblast differentiation in the subgranular zone of the dentate gyrus using Ki67 and doublecortin (DCX), respectively, as markers. In addition, we monitored the effects of RSG on the levels of DCX and brain-derived neurotrophic factor (BDNF) in hippocampal homogenates. At 8 weeks after the LFD feeding, the numbers of Ki67- and DCX-positive cells as well as hippocampal levels of DCX and BDNF were significantly decreased in the RSG-treated group compared to the vehicle-treated animals. In contrast, the numbers of Ki67- and DCX-positive cells along with hippocampal levels of DCX and BDNF in the HFD fed mice were significantly increased in the RSG-treated mice compared to the vehicle-treated group. Our data demonstrate that RSG can modulate the levels of BDNF, which could play a pivotal role in cell proliferation and neuroblast differentiation in the hippocampal dentate gyrus.

Abstract:A large class of generic stochastic processes which are not necessarily analytically solvable but are still numerically tractable can be described by giving transition probability kernels over a contiguous set of time intervals. From the numerical viewpoint, this procedure is highly eff ective on current microchip architectures as kernels can be conveniently evaluated using GPU co-processors and then used for scenario generation while storing them in CPU caches. This paper describes the pricing methodology and a mathematical framework for Finance based on direct kernel manipulations, i.e. operator methods. We also discuss a number of techniques based on measure changes to accomplish tasks such as variance reduction and sensitivity calculations. Numerical experiments are included along with performance benchmarks. Source code is distributed separately online under GPL license in a library named OPLib.
Rosiglitazone attenuates hypoxia-induced renal cell apoptosis by inhibiting NF-κB signaling pathway in a PPARγ-dependent manner

Abstract:In recent years, peroxisome proliferator-activated receptor γ (PPARγ) has been found to be closely associated with hypoxia renal disease. The aim of this study was to investigate the relationship between rosiglitazone and mitochondrial apoptosis in renal tissue and its associated mechanisms. Twenty-four male Sprague-Dawley rats were randomly divided into three groups ( n = 8 in each): normal control group, hypoxia injury group (equal volume of 0.9% saline), and PPARγ agonist group (Rosiglitazone, 10 mg/kg · d, intraperitoneally). The hypoxia injury group and PPARγ agonist group were placed in a hypoxia chamber and the simulated altitude was set at 7,000 m for 7 days. Blood and kidney samples were collected after 7 days. The quantitative real-time polymerase chain reaction and Western blot methods were used to determine the expression of PPARγ, nuclear factor kappa-B (NF-κB), B-cell lymphoma-2 (Bcl-2), and Bax. The results showed that compared with the normal control group, the renal tissue of rats after hypoxia was severely damaged, as shown by massive renal tubular epithelial cell degeneration and detachment, and renal tubular dilation. The NF-κB protein expression significantly increased, the Bcl-2 protein and mRNA expression significantly decreased, and Bax protein and mRNA expression significantly increased ( p < .05 for all). Renal injury was much less severe in the PPARγ agonist group compared to the hypoxia injury group. Rosiglitazone can alleviate hypoxia renal injury, with the possible mechanism involving attenuation of apoptosis by inhibiting the activation of the NF-κB signaling pathway in a PPARγ-dependent manner and increasing Bcl-2 and decreasing Bax expression.

Abstract:Background and Purpose-Oligodendrogenesis is essential for white matter repair after stroke. Although agonists of peroxisome proliferator-activated receptors. confer neuroprotection in models of cerebral ischemia, it is not known whether this effect extends to white matter protection. This study tested the hypothesis that the peroxisome proliferator-activated receptors. agonist rosiglitazone enhances oligodendrogenesis and improves long-term white matter integrity after ischemia/reperfusion.Methods-Male adult C57/BL6 mice (25-30 g) were subjected to 60-minute middle cerebral artery occlusion and reperfusion. Rosiglitazone (3 mg/kg) was injected intraperitoneally once daily for 14 days beginning 2 hours after reperfusion. Sensorimotor and cognitive functions were evaluated <= 21 days after middle cerebral artery occlusion. Immunostaining was used to assess infarct volume, myelin loss, and microglial activation. Bromodeoxyuridine (BrdU) was injected for measurements of proliferating NG2(+) oligodendrocyte precursor cells (OPCs) and newly generated adenomatous polyposis coli(+) oligodendrocytes. Mixed glial cultures were used to confirm the effect of rosiglitazone on oligodendrocyte differentiation and microglial polarization.Results-Rosiglitazone significantly reduced brain tissue loss, ameliorated white matter injury, and improved sensorimotor and cognitive functions for at least 21 days after middle cerebral artery occlusion. Rosiglitazone enhanced OPC proliferation and increased the numbers of newly generated mature oligodendrocytes after middle cerebral artery occlusion. Rosiglitazone treatment also reduced the numbers of Iba1(+)/CD16(+) M1 microglia and increased the numbers of Iba1(+)/CD206(+) M2 microglia after stroke. Glial culture experiments confirmed that rosiglitazone promoted oligodendrocyte differentiation, perhaps by promoting microglial M2 polarization.Conclusions-Rosiglitazone treatment improves long-term white matter integrity after cerebral ischemia, at least, in part, by promoting oligodendrogenesis and facilitating microglial polarization toward the beneficial M2 phenotype.
