Abstract:Probiotic supplementation in fish feed is becoming popular as a viable alternative to curb the indiscriminate use of antibiotics in aquaculture. The effect of inactivated Lactobacillus plantarum on growth performance and health related indices of black sea bream (Acanthopagrus schlegelii) fingerlings was studied, using different concentrations of a commercial inactivated L. plantarum product (Immuno-LP20): D1 (control, 0 mg/kg), D2 (100 mg/kg), D3 (200 mg/kg), D4 (400 mg/kg), D5 (800 mg/kg), and D6 (1600 mg/kg). After an eight-week feeding trial during which the diets were fed to triplicate groups of fish (initial weight: 1.58 ± 0.05 g), D2, D3, and D4 produced fish with significantly higher growth parameters than the control fish (P < 0.05), whilst all fish fed the diets with L. plantarum had significantly improved feed utilization parameters than the control group. Serum total protein content, and lysozyme activity significantly increased in fish fed D4 than the control fish, whilst complement 4 content was higher in fish fed almost all the inactivated probiotic diets (except D5) than the control fish. Serum superoxide dismutase activity was significantly higher in fish fed all the Immuno-LP20 diets except D2, and liver glutathione peroxidase activity in D2, D3, and D4 fish were significantly higher than in the control fish. Intestinal structure was also significantly improved in fish fed the Immuno-LP20 diets. According to these results, using inactivated L. plantarum as dietary supplement could be beneficial in the production of black sea bream at an optimum inclusion rate similar to 100 mg/kg ≤ 400 mg/kg Immuno-LP20.

Abstract:The interactive effect of dietary inactivated Lactobacillus plantarum and berberine on black sea bream, Acanthopagrus schlegelii, was investigated with three diets designated as D1 (Con: basal diet), D2 (LP: basal diet + 400 mg/kg L. plantarum), and D3 (LPBB: basal diet + 400 mg/kg L. plantarum + 50 mg/kg berberine) and fed to juvenile black sea bream ( 5.67 ± 0.05 g) for 56 days. The growth performance and feed utilization parameters, as well as intestinal trypsin activity, were significantly improved in the LP and LPBB groups ( P < 0.05 ). Fish fed the LPBB diet showed better serum and hepatic antioxidant capacity, whilst the LP group had better hepatic antioxidant capacity, than the control fish ( P < 0.05 ). Intestinal IgM and C3 levels significantly increased in the LPBB fish than the rest of the groups ( P < 0.05 ). NF-κB was significantly upregulated in the LP group ( P < 0.05 ). Nrf2 and IL-10 were significantly upregulated, whilst Keap1b and NF-κB were significantly downregulated in the LPBB group ( P < 0.05 ). These findings show that a combination of inactivated L. plantarum and berberine in the diet of black sea bream can improve the immune response and antioxidant capacity than a diet with only the inactivated probiotic, whilst both diets can equally improve growth performance.

Abstract:This study aimed to investigate the effects of graded butyrate glyceride (BG) supplementation levels in high soybean meal diet on juvenile black sea bream. Three hundred and sixty fish were fed diets containing 199 g/kg fishmeal and 450 g/kg soybean meal, with increasing levels of BG at 0, 2, 4, 6, 8 or 16 g/kg for eight weeks. The growth performance of the fish increased with increasing dietary BG supplementation up to 6 g BG/kg, beyond which growth rate reduced significantly (p < .05). Dietary BG supplementation increased the intestinal protease activity, but had no significant (p > .05) effect on lipase and amylase activities. Fish fed the basal diet exhibited villus shortening with a decrease in the number of goblet cells and a reduction in absorptive and digestive epithelial surface, while fish fed >= 4 g BG/kg diets showed a well-integrated gut, with large absorptive and digestive epithelial surface. Dietary BG supplementation also improved antioxidative capacity by increasing superoxide dismutase and glutathione peroxidase activities while decreasing malondialdehyde content. The inclusion of BG in high soybean meal diets can improve growth performance, maintain healthy gut morphology and enhance antioxidative capacity of black sea bream.
Multiple effects of dietary supplementation with Lactobacillus reuteri and Bacillus subtilis on the growth, immunity, and metabolism of largemouth bass (Micropterus salmoides)

Abstract:Probiotics play an essential role in the largemouth bass (Micropterus salmoides) aquaculture sector. They aid the fish in sickness prevention, intestinal structure improvement, food absorption, and immune system strengthening. In this experiment, Bacillus subtilis (BS, 107 CFU/g) and Lactobacillus reuteri (LR, 107 CFU/g) were added to the feed and then fed to M. salmoides for 35 days. The effects of two probiotics on the growth, immunity, and metabolism of M. salmoides organisms were studied. The results revealed that the BS group significantly increased the growth rate and specific growth rate of M. salmoides, while both the BS and LR groups significantly increase the length of villi M. salmoides intestines. The BS group significantly increased the levels of AKP, T-AOC, and CAT in the blood of M. salmoides, as well as AKP levels in the intestine. Furthermore, the BS group significantly increased the expression of intestinal genes Nrf2, SOD1, GPX, and CAT, while significantly decreasing the expression of the keap1 gene. M. salmoides gut microbial analysis showed that the abundance of Planctomycetota was significantly different in both control and experimental groups. Analyzed at the genus level, the abundance of Citrobacter, Paracoccus, Luedemannella， Sphingomonas, Streptomyces and Xanthomonas in the both control and experimental groups were significantly different. The BS group's differentially expressed genes were predominantly enriched in oxidative phosphorylation pathways in the intestine, indicating that they had a good influence on intestinal metabolism and inflammation suppression. In contrast, differentially expressed genes in the LR group were primarily enriched in the insulin signaling and linoleic acid metabolism pathways, indicating improved intestine metabolic performance. In conclusion, B. subtilis and L. reuteri improve the growth and health of M. salmoides, indicating tremendous potential for enhancing intestinal metabolism and providing significant application value.
Functional Properties of Protein Hydrolysates on Growth, Digestive Enzyme Activities, Protein Metabolism, and Intestinal Health of Larval Largemouth Bass (<i>Micropterus salmoides</i>)

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.
Antagonistic Effects and the Underlying Mechanisms of Bacillus velezensis and its Antibacterial Peptide LCI Against Aeromonas hydrophila Infection in Largemouth Bass

Abstract:Aeromonas hydrophila is one of the most prevalent pathogenic bacteria in largemouth bass. The use of antibiotics to inhibit A. hydrophila poses a significant threat to fish and environmental safety. Bacillus velezensis, a safe bacterium with probiotic and antibacterial characteristics, is an ideal candidate for antagonizing A. hydrophila. This study explored the antagonistic effects of B. velezensis FLU-1 on A. hydrophila in vivo and in vitro. In addition, we explored the antimicrobial peptides (AMPs) produced by strain FLU-1 and clarified the underlying antibacterial mechanisms. The results showed that strain FLU-1 could inhibit a variety of fish pathogens, including A. hydrophila. The challenge test showed that dietary supplementation with B. velezensis FLU-1 significantly improved the survival rate of largemouth bass and reduced the bacterial load in liver. Subsequently, the AMP LCI was isolated from B. velezensis FLU-1 and was found to be effective against A. hydrophila in vitro and in vivo. Transcriptomic analysis revealed that LCI downregulated the genes associated with flagellar assembly and peptidoglycan synthesis in A. hydrophila. Phenotypic test results showed that LCI disrupted the membrane integrity, markedly reduced the biofilm biomass and diminished the swimming motility of A. hydrophila. Furthermore, the results showed that LCI bound to the genomic DNA of A. hydrophila and destroyed the DNA structures. Overall, these findings elucidated the mechanism of action of LCI against A. hydrophila at the phenotypic and physiological levels. This study suggests that B. velezensis FLU-1 and its AMP LCI could serve as antibiotic alternatives for controlling pathogens in aquaculture.
Modulation of growth performance, antioxidant capacity, non-specific immunity and disease resistance in largemouth bass (Micropterus salmoides) upon compound probiotic cultures inclusion

Abstract:An 8-week feeding trial was conducted to evaluate the effects of dietary supplementation of compound probiotic cultures (CPC; Bacillus subtilis, Lactobacillus plantarum and Saccharomyces cerevisiae) on the growth performance, antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass. Triplicate groups of largemouth bass (average weight 42.05 ± 0.02 g), with a destiny of 30 individuals per tank, were fed diets supplemented with different concentration of compound probiotic cultures (CPC) (0%, CPC (0.0); 0.5%, CPC (0.5); 1.0%, CPC (1.0); 2.0%, CPC (2.0)). After the feeding trial, tissue samples of largemouth bass were collected and the challenge test with Aeromonas hydrophila was performed. Results indicated that the CPC supplementation produced no significant difference on the growth performance, feed utilization and body composition of largemouth bass, while significantly increased the cumulative survival rate in the Aeromonas hydrophila challenge test. Meanwhile, the inclusion of CPC elevated the hepatic antioxidant capacity, and the highest activity of antioxidant enzymes, including T-AOC, CAT, GPx and T-SOD, was observed in the CPC (2.0) group. Meanwhile, the transcription of Nrf2/keap1 and antioxidant related genes, including CAT, GPx, GST, SOD1 and SOD2, was significantly elevated with the inclusion of CPC. In addition, the inclusion of CPC improved the non-specific immunity of largemouth bass. The activity of serum lysozyme was significantly elevated in the CPC (2.0) group, while the transcription of RelA and pro-inflammatory factors, including TNF-α and IL-1β, was inhibited with the inclusion of CPC. Meanwhile, related genes potentially linked to RelA, including TLR2 and p38 MAPK, were detected that their relative expression was significantly inhibited with the inclusion of CPC. The current findings indicated that the inclusion of 2% CPC improved the antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass, and suggested that 2% CPC as a functional additive could be applied to the diet of juvenile largemouth bass in aquaculture practice.
Effects of Co-Fermented Feed Using Lactobacillus acidophilus, Limosilactobacillus reuteri and Lactiplantibacillus plantarum on Growth, Antioxidant Capacity, Fatty Acids and Gut Microbiota of Largemouth Bass (Micropterus salmoides)

Abstract:The effects of diets fermented with compound probiotics, namely Lactobacillus acidophilus, Limosilactobacillus reuteri and Lactiplantibacillus plantarum, on the growth performance, physiological and biochemical indexes, fatty acid composition and intestinal health of juvenile largemouth bass (Micropterus salmoides) were investigated. Three hundred healthy juvenile M. salmoides (5.29 ± 0.02 g) were selected and randomly divided into two groups with triplicates for each. The basic diet was set as the control group (CON), and fermentation of the basic diet with a mixed bacterial solution (1.8 × 109 cfu/mL, L. acidophilus:L. reuteri:L. plantarum = 1:1:1) was set as the fermentation group (FER). Fish were hand fed to satiation for 56 days and two-thirds of the culture water was renewed every 3 days. The results showed that feed intake of fish in the FER group was significantly lowered, thereby increasing feed efficiency (FE) and protein efficiency (PER) (p < 0.05). Serum alanine aminotransferase (ALT) activity was significantly decreased, and catalase (CAT) activity was significantly increased in the FER group (p < 0.05). The liver superoxide dismutase activity (SOD) was significantly enhanced, and intestinal trypsin was significantly increased in the FER group (p < 0.05). Being fed with the fermentation diet significantly increased the content of n-3 polyunsaturated fatty acids (PUFA), docosahexaenoic acid (DHA) and the n-3/n-6 PUFAs ratios in the liver (p < 0.05). Intestinal histology showed that villus height and width of the intestine and the number of goblet cells were significantly increased in the FER group (p < 0.05). Those fed with fermentation diets had limited diversity of gut microbiota. Compared to the CON group, the relative abundance of Aeromonas decreased significantly (p < 0.05), while the relative abundance of Fusobacteria, Cetobacteria and Lactobacillusis in FER increased greatly in the gut microbiota of the FER group. In conclusion, fermented feed with the three probiotics effectively improved the feed utilization and antioxidant capacity, promoted digestion and absorption of dietary protein, improved the ability of synthesize DHA and n-3 PUFAs in the liver and reduced the abundance of pathogenic bacteria in the gut. Therefore, the present research provided a new way of co-fermented feed with three probiotics for the aquaculture of M. salmoides.

Suppression of Immunological Activities by Mouse Amniotic Fluid

Abstract:Mouse amniotic fluid (MAF) was shown to be capable of suppressing those antibody responses observed in euthymic or athymic mouse spleen cell cultures to the T‐independent antigens dinitrophenylated Ficoll (DNP‐Ficoll) and trini‐trophenylated lipopolysaccharide (TNP‐LPS) and to the polyclonal B‐cell activators LPS and purified protein derivative of tuberculin (PPD). Titration experiments demonstrated that the suppressive capacity of MAF for either LPS or DNP‐Ficoll responses was maintained up to a MAF dilution of 1:120. Pre‐incubation of spleen cells obtained from athymic mice with MAF for 8 h significantly suppressed polyclonal B‐cell activation of such cells induced by LPS, although suppression was greater when MAF was present during the entire culture period. In addition, the suppressive activity that MAF demonstrated for antibody production induced by DNP‐Ficoll or LPS was not lost as a result of dialysis. MAF also suppressed the secondary in vitro prolixferative responses of lymph node cells sensitized to the T‐dependent antigen human gamma globulin (HGG). HGG‐induced proliferation of such cells appeared to be more susceptible to suppression effected by MAF than concanavalin‐A‐induced proliferation.
Dietary L-Valine Improved Growth, Antioxidant Capacity, and Alleviated Endoplasmic Reticulum Stress-Mediated Apoptosis in the Intestine of Juvenile Largemouth Bass Micropterus Punctulatus After Infection With Aeromonas Veronii

Abstract:Abstract Background: The intestine is the first line of defense against bacteria, toxins and antigens presents in the lumen of the instine. Aeromonas veronii is an opportunistic pathogen that has been related to mortality in fish. However, the protective mechanisms of intestinal damage caused by Aeromonas veronii are poorly defined. Thus, the objetive of this study was to evaluate the influence of dietary valine (Val) on growth, antioxidant capacity, and endoplasmic reticulum stress-mediated apoptosis in intestines of the juvenile largemouth bass Micropterus punctulatus after infection with Aeromonas veronii (A. veronii). Methods: A total of 720 fish (37.99 ± 0.01 g) were randomly allocated to six groups with three replicates each group, with different levels of Val (11.0, 14.0, 17.0, 20.1, 23.1, and 26.1 g/kg diet, respectively) for 77 d, and then challenged with A. veronii. Results: In the present study, we demonstrated that dietary Val: (1) increased the percent weight gain (PWG), specific growth rate (SGR), feed intake (FI), feed efficiency (FE), protein efficiency ratio (PER), protein production value (PPV), lipid production value (LPV), and ash production value (APV) (P< 0.05); (2) improved survival rate (SR) and alleviated intestinal injury; (3) reduced the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and protein carbonyl (PC) (P< 0.05), and improved the intestinal antioxidant capacity by enhancing the mRNA levels of Cu/Zn superoxide dismutase (SOD1), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR), and γ-glutamyl cysteine ligase catalytic subunit (GCLC) (P< 0.05); (4) increased nuclear NFE2-related factor 2 (Nrf2) mRNA and protein levels (P< 0.05); (5) reduced intestinal DNA fragments, and down-regulated the mRNA levels of 78 KD glucose regulated protein (GRP78), activating transcription factor 6 (ATF6), inositol requiring enzyme-1 (IRE1), protein kinase RNA-dependent-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α (eIF2α), activating transcription factor 4 (ATF4), C/EBP homologous protein (CHOP), cysteine-aspartic protease 9 (caspase 9), and cysteine-aspartic protease 3 (caspase 3), while up-regulating that of B-cell lymphoma-2 (Bcl2) (P< 0.05); (6) decreased phosphorylation of PERK (p-PERK) and phosphorylation of eIF2α (p-eIF2α), caspase 9, and caspase 3 protein levels. Conclusion: Dietary optimal Val level enhanced the growth performance and the intestinal antioxidant capacity via the Nrf2 signaling pathway after infection with A. veronii. Furthermore, this study is the first to demonstrate the role of Val in reducing ERS-mediated apoptosis through modulation of the PERK/eIF2α signaling pathway. Finally, Quadratic regression analysis of the SGR indicated that the dietary Val requirement of largemouth bass (37.99-162.92 g) was 21.50 g/kg diet, corresponding to 46.64 g/kg protein.
Effects of tributyrin and alanyl-glutamine dipeptide on intestinal health of largemouth bass (Micropterus salmoides) fed with high soybean meal diet

Abstract:To investigate the effects of dietary tributyrin (TB) and alanyl-glutamine (AGn) on the intestinal health of largemouth bass ( Micropterus salmoides ) fed with high-level soybean meal (SM) diet, six isonitrogenous (41.36%) and isolipidic (10.25%) diets were formulated and fed to largemouth bass (initial body weight 25.5 ± 0.5g) for 8 weeks. The two control diets contained 34.8% peanut meal (PM) and 41.3% SM, while the other four experimental diets supplemented TB at 0.1% (TB0.1), 0.2% (TB0.2) and AGn at 1% (AGn1), 2% (AGn2) in SM, respectively. The results showed that there were no significant differences in weight gain, survival rate, and hepatosomatic index among all groups ( P >0.05), while feed coefficient rate in AGn1, AGn2 and TB0.2 groups was significantly lower than that in SM group ( P < 0.05). Compared with the PM group, the intestinal inflammation of largemouth bass in SM group were obvious, accompanied by the damage of intestinal structure, the decrease of digestive enzyme activity, and the up-regulation of proinflammatory cytokines. Compared with the SM group, the activities of intestinal trypsin, lipase and foregut amylase in TB and AGn groups increased significantly ( P <0.05), and the gene expression levels of acetyl-CoA carboxylase (ACC), caspase-3, caspase-8, caspase-9, tumor necrosis factor alpha (TNF-α), and interleukin-1 beta (IL-1β) were down-regulated, while the gene expression levels of target of rapamycin (TOR) and eIF4E-binding protein (4E-BP) were up-regulated in all experimental groups ( P <0.05). It can be concluded that supplementation of 1%-2% AGn and 0.1%-0.2% TB can alleviate enteritis caused by high-level soybean meal, and the recommend level is 2% AGn and 0.2% TB.

We investigated the effects of AMP supplementation on the antioxidant capacity, non-specific immunity, and gut microbiota of tsinling lenok trout. 240 fish were fed diets (CT, A120, A240 and A480) containing different amounts of AMP peptides (0, 120 mg kg −1 , 240 mg kg −1 , 480 mg kg −1 ) for 8 weeks. Our results showed that the activity of total antioxidant capacity (T-SOD) and glutathione peroxidase (GSH-Px), lysozyme (LZM), catalase (CAT) and acid phosphatase (ACP) in the A240 and A480 group were higher than that in the CT group ( P < 0.05). The content of malondialdehyde (MDA) in AMP group was significantly lower than that in CT group ( P < 0.05). Furthermore, we harvested the mid-gut and applied next-generation sequencing of 16S rDNA. The results showed that the abundance of Halomonas in AMP group was significantly lower than that in CT group. Functional analysis showed that the abundance of chloroalkane and chloroalkene degradation pathway increased significantly in AMP group. In conclusion, AMP enhanced the antioxidant capacity, non-specific immunity, and intestinal health of tsinling lenok trout.

Abstract:The cultivation of largemouth bass ( Micropterus salmoides ), a species of significant economic value in aquaculture, has experienced notable growth recently. However, the deterioration of water quality seriously affects the metabolic responses of M. salmoides . While compound microbial agent (CMA) is widely utilized for ecological rehabilitation and water filtration, its application in M. salmoides has not been reported. Here, based on physio-biochemical tests and 16S rRNA sequencing, we investigated the effects of CMA (yeast, Bacillus subtilis , and lactic acid bacteria) on the water quality within the recirculating aquaculture system, along with physiological indices and gut microbiota of M. salmoides . Compared to the control and single microbial agent (yeast), CMA treatment improved the water quality by improving the dissolved oxygen and delaying the increase of pH, total nitrogen, total phosphorus, ammonia nitrogen, and nitrite. The 16s rRNA gene sequencing revealed that the water treated with CMA exhibited elevated levels of chao1, Shannon, Pd, and a larger population of dominant bacterial. Besides, higher values of ACE, chao1, Shannon, and OTU level, and lower Simpson index were found in CMA treated M. salmoides samples, suggesting that CMA treatment enhanced the species richness and diversity of gut microbiota of M. salmoides . Furthermore, CMA treatment hindered the generation and proliferation of harmful bacteria, such as the Mycoplasma mobile 163K species and the Erysipelotrichaceae family, which was associated with enhanced antioxidant enzymatic activity and decreased MDA level in both the serum and liver. These findings shed light on the essential roles of CMA in M. salmoides culturing and introduce an innovative approach to enhance the aquatic environment.

Abstract:Introduction As the aquaculture industry intensifies to enhance production efficiency and capacity, the risk of disease outbreaks in high-density systems, such as those for largemouth bass, has escalated. This necessitates the exploration of novel strategies for disease prevention and control. Studies have shown that Atractylodes macrocephala polysaccharide (AMP) possesses the functions of promoting growth and enhancing immune capacity, making it a potential feed additive in animal production. Methods This study aimed to assess the impact of AMP on the growth performance, serum biochemical indices, and intestinal flora structure of largemouth bass. A total of 360 healthy largemouth bass (mean weight: 15.25 ± 1.29 g) were randomly assigned to four groups, each with three replicates. The groups were fed diets supplemented with 0% (Group D, control group), 0.4% (Group A), 0.8% (Group B), or 1.2% (Group C) AMP for 42 days. Results The results showed that the addition of an appropriate amount of AMP in the feed significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of largemouth bass, reduced the feed conversion ratio (FCR), and enhanced growth performance. AMP reduced the levels of AST and ALT, indicating a hepatoprotective effect on largemouth bass, with significant differences from the control group (P < 0.05). AMP also improved the intestinal microbiota composition of largemouth bass, positively affecting intestinal health. The analysis of intestinal microbiota revealed 1,288 amplicon sequence variants (ASVs) in the intestine of largemouth bass in Group B, dominated by Firmicutes and Bacteroidota. In contrast, Group C (1.2%) had 920 ASVs, with Fusobacteriota and Firmicutes as the major components, while the relative abundance of Firmicutes was lower, showing significant differences from the control Group D. The functional analysis of intestinal microbiota based on KEGG showed significant differences (P < 0.05) among the four groups in Carbon metabolism, Biosynthesis of amino acids, and Metabolic pathways. Discussion The study concludes that the inclusion of AMP at 0.8% – 1.2% in the feed can enhance the growth performance and intestinal health of largemouth bass, offering a promising strategy for disease prevention and control in intensive aquaculture settings.

Probiotics and Antimicrobial Proteins

Abstract:This study was conducted to evaluate different doses of two species of Bacillus (Bacillus licheniformis and Bacillus subtilis), on growth parameters, chemical composition of fish, activity of liver, and digestive enzymes of Asian sea bass. During 8 weeks, juvenile Asian sea bass received diets supplemented with 1 × 103, 1 × 106, and 1 × 109 CFU g−1 probiotic in addition to a control diet without added microorganisms. At the end of the trial, growth indices (total weight, total length, specific growth rate, total weight gain, food conversion ratio, and condition factor), body composition (crude protein, crude lipid, ash, and dry matter), digestive enzymes (protease, lipase, and amylase), liver enzymes [aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP)], immunologic indicators (lysozyme), and hematological parameters [hematocrit (Hct), hemoglobin (Hb), red blood cells (RBCs), white blood cells (WBCs)] were assessed. Asian sea bass receiving diets supplemented with probiotic Bacillus (Bacillus licheniformis and Bacillus subtilis) showed significantly better growth than those fed the basal diet (control). Regarding body composition, total protein levels and dry matter were higher and lipid levels were lower in fish fed the diet containing 1 × 106 CFU g−1 probiotic compared with the control group (P < 0.05). Digestive enzymes (protease, lipase, and amylase) and hematological parameters (RBC, WBC, and Hb) were all highest in fish fed diet supplemented with 1 × 106 CFU g−1 probiotic Bacillus. Also, liver enzymes (AST, ALT, ALP) were lower in fish fed diet supplemented with 1 × 106 CFU g−1 probiotic Bacillus. Being that supplementation of 1 × 106 CFU g−1 of Bacillus in the diet is the dose which delivers the best results.

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-β and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:This study aimed to identify potential probiotic strains of Bacillus subtilis from healthy fish gut microbiota for application in aquaculture. The effects of dietary B. subtilis administration on growth performance, serum enzyme activity, immune gene expression, and disease resistance in darkbarbel catfish (Pelteobagrus fulvidraco) were investigated. The isolate, identified through gene sequencing and biochemical tests, demonstrated resilience to pH 3.0% and 6.0% bile, and exhibited extracellular protease, cellulose, lipase, and amylase production. Darkbarbel catfish were fed diets with varying B. subtilis concentrations (0 CFU/kg [T0], 107 CFU/kg [T1], 108 CFU/kg [T2], and 109 CFU/kg [T3]). After 8 weeks, significant increases (p < 0.05) were observed in final body weight, weight gain rate, specific growth rate, serum lysozyme, serum superoxide dismutase, alkaline phosphatase, and total antioxidant capacity, whereas malondialdehyde levels significantly decreased. Feeding darkbarbel catfish with B. subtilis diets increased immunoglobulin M (IgM) and C3 gene expression (p < 0.05), indicating a positive impact on the fish's immune system. The strain upregulated interleukin 10 (IL-10) and transforming growth factor-β (TGF-β) expression and downregulated TNF-α and IL-1β, suggesting potential anti-inflammatory effects. Following a 7-day challenge with Aeromonas hydrophila, fish fed with B. subtilis exhibited lower mortality, with higher survival rates in the T2 and T3 groups. In conclusion, supplementing darkbarbel catfish diets with B. subtilis effectively enhances growth performance, immune response, and disease resistance.

Abstract:To investigate the activities of intestinal digestive enzymes, liver antioxidant enzymes, immunological enzymes, and glucometabolic enzymes in largemouth bass (Micropterus salmoides) under the biofloc model, an experiment was conducted in 300-liter glass tanks. The experiment comprised a control group, which was fed a basal diet, and a biofloc group, where glucose was added to maintain a C/N ratio of 15. Each group had three parallel setups, with a stocking density of 20 fish per tank. The experiment ran for 60 days, employing a zero-water exchange aquaculture model. The results showed that at the end of the culture period, there were no significant differences between the initial weight, final weight, WGR, SGR, and SR of the biofloc group and the control group of largemouth bass (p > 0.05), whereas the lower FCR and the higher PER in the biofloc group were significant (p < 0.05); intestinal α-amylase, trypsin, and lipase activities of largemouth bass in the biofloc group were significantly increased by 37.20%, 64.11%, and 51.69%, respectively, compared with the control group (p < 0.05); liver superoxide dismutase and catalase activities, and total antioxidant capacity of largemouth bass in the biofloc group were significantly increased by 49.26%, 46.87%, and 98.94% (p < 0.05), while the malondialdehyde content was significantly reduced by 19.91% (p < 0.05); liver lysozyme, alkaline phosphatase, and acid phosphatase activities of largemouth bass in the biofloc group were significantly increased by 62.66%, 41.22%, and 29.66%, respectively (p < 0.05); liver glucokinase, pyruvate kinase, glucose-6-phosphate kinase, pyruvate kinase, glucose-6-phosphatase, and glycogen synthase activities were significantly increased by 46.29%, 99.33%, 32.54%, and 26.89%, respectively (p < 0.05). The study showed that the biofloc model of culturing largemouth bass can not only enhance digestive enzyme activities, antioxidant capacity, and immune response but can also promote the process of glucose metabolism and reduce feeding costs. This study provides data support for healthy culturing of largemouth bass in future production, provides a theoretical reference for optimizing the biofloc technology culture model, and is crucial for promoting the healthy and green development of aquaculture.

Abstract:In order to investigate the effect of bacterium Bacillus amyloliquefaciens on growth performance,digestive enzymes activities,serum proteins levels and liver superoxide dismutase( SOD) activity in juvenile black seabream( Sparus macrocephalus),this paper reared the fish with 7 g body weight in a small cement pool of an indoor microwater circulation system and fed diets containing Bacillus amyloliquefaciens at a dose of 0,0. 025%,0. 05%,and 0.1%,respectively. After 56 d,the results showed that the bacterium led to improve weight gain rate( WGR) and specific growth rate( SGR) in juvenile black seabream. The WGR and SGR were significantly higher in juvenile black seabream fed by diet supplemented with 0.05% of Bacillus amyloliquefaciens than that of the fish fed by control diet.The intestinal protease and lipase activities in the fish fed by diet supplemented with Bacillus amyloliquefaciens were significantly higher than that of the control group. The total serum protein levels of juveniles fed with 0.025% Bacillus dietary supplement were significantly higher than that of the control group,among them the globulin and albumin levels were extremely increased. In addition,the intestinal amylase activity and hepatic SOD activity were also obviously higher in the 0. 1% Bacillus amyloliquefaciens dietary supplement group than that of the control. These results indicated that optimal dose of Bacillus amyloliquefaciens administration could increase the intestinal digestive enzyme activity,improve antioxidant ability in juvenile black seabream,and promote their growth performance.
