Abstract:Purpose: Osteoarthritis (OA) is a multifaceted disease that involves pathological changes of multiple tissues in the joint. Most studies on cartilage matrix in OA have focused on changes of glycosaminoglycan and collagen content; very little is known about the alteration of collagen architecture in OA. However, the complex organization of collagen fibers in all layers of articular cartilage requires highly quantifiable imaging tools for visualization and analysis. One such tool is Second Harmonic Generation (SHG) imaging, which is a powerful two-photon confocal imaging technique ideal for imaging non-centrosymmetric molecules, such as collagen fibers. The purpose of this study is to use the SHG imaging method to evaluate collagen fiber thickness and orientation changes in early mouse experimental OA and to correlate these changes with synovial inflammation and joint destruction. Methods: OA was induced in NFκB-luciferase reporter mice (Taconic) via the destabilization of the medial meniscus (DMM) surgery. Mice were then subjected to three types of analysis between 1–10 weeks post-surgery: 1) SHG imaging. Collagen thickness and orientation were visualized using SHG imaging on cryosections. Fiber thickness was calculated as the ratio of forward to backward SHG signal (FSHG/BSHG), and directional orientation was calculated as variance of fiber orientation with respect to each other. 2) Histological analysis. Cartilage damage revealed by safranin O staining was assessed by the modified Mankin system. IHC was used to assess collagen content and macrophage presence. Synovitis in the anterior and posterior compartments of the joint were scored. 3) In vivo imaging of bioluminescence signals for NFκB activity to assess whole joint inflammation. Luciferin (25 μg) was injected intraarticularly, and mice were imaged with the IVIS Biophotonic 200 imager. As controls, sham and non-surgery mice were analyzed. Non-parametric data from Mankin scoring and synovitis analysis were evaluated via Kruskal-Wallis analysis followed by Dunn’s multiple comparison test. All other experiments were evaluated with ANOVA followed by Bonferroni’s multiple comparison test. Spearman’s correlation was performed for correlation analysis. P < 0.05: considered statistically significant. Results: Zonal differences in collagen fiber thickness and organization were observed as soon as OA initiated after DMM surgery, which persisted over time (Fig. A–C). Even at 1 week-post surgery, the DMM joint showed a decrease in collagen fiber thickness in the radial zone and an increase in collagen fiber disorganization in the superficial zone. Interestingly, in the radial zone, where the DMM joint showed decreased directional variance relative to the Sham joint, suggesting that the collagen fibers in the radial zone became more organized in OA. Bioluminescence imaging data showed that DMM and Sham control joints had a similar surge of inflammation at 1-week post-surgery, but the DMM joint exhibited a delay in resolution of inflammation in subsequent weeks. A similar trend was observed with synovitis, which we found to be mainly driven by synovial cell density and inflammatory infiltration rather than synovial thickness (data not shown). Interestingly, we found a correlation between the trend of collagen fiber thickness changes in the deep zone with that of synovitis in both the DMM and Sham knees; however, DMM surgery leads to an overall shift toward higher synovitis and thinner collagen fibers. Additionally, the trend of collagen directional variance in the superficial zone followed that of the Mankin score during OA progression (Fig. D). Conclusions: SHG imaging is a powerful approach that can be used to quantitatively analyze collagen landscape throughout articular cartilage. Combining SHG imaging with bioluminescence imaging and histological approaches, we uncovered a coordinated change between different components of cartilage matrix and inflammation in OA progression.
Mineral and cross-linking in collagen fibrils: The mechanical behavior of bone tissue at the nano-scale

Abstract:The mineralized collagen fibril is the main building block of hard tissues and it directly affects the macroscopic mechanics of biological tissues such as bone. The mechanical behavior of the fibril itself is determined by its structure: the content of collagen molecules, minerals, and cross-links, and the mechanical interactions and properties of these components. Advanced-Glycation-Endproducts (AGEs) cross-linking between tropocollagen molecules within the collagen fibril is one important factor that is believed to have a major influence on the tissue. For instance, it has been shown that brittleness in bone correlates with increased AGEs densities. However, the underlying nano-scale mechanisms within the mineralized collagen fibril remain unknown. Here, we study the effect of mineral and AGEs cross-linking on fibril deformation and fracture behavior by performing destructive tensile tests using coarse-grained molecular dynamics simulations. Our results demonstrate that after exceeding a critical content of mineral, it induces stiffening of the collagen fibril at high strain levels. We show that mineral morphology and location affect collagen fibril mechanics: The mineral content at which this stiffening occurs depends on the mineral's location and morphology. Further, both, increasing AGEs density and mineral content lead to stiffening and increased peak stresses. At low mineral contents, the mechanical response of the fibril is dominated by the AGEs, while at high mineral contents, the mineral itself determines fibril mechanics.

Abstract:During bone formation, collagen fibrils mineralize with carbonated hydroxyapatite, leading to a hybrid material with excellent properties. Other minerals are also known to nucleate within collagen in vitro. For a series of strontium- and calcium-based minerals, we observed that their precipitation leads to a contraction of collagen fibrils, reaching stresses as large as several megapascals. The magnitude of the stress depends on the type and amount of mineral. Using in-operando synchrotron x-ray scattering, we analyzed the kinetics of mineral deposition. Whereas no contraction occurs when the mineral deposits outside fibrils only, intrafibrillar mineralization generates fibril contraction. This chemomechanical effect occurs with collagen fully immersed in water and generates a mineral-collagen composite with tensile fibers, reminiscent of the principle of reinforced concrete.

Abstract:Progressive stiffening of collagen tissue by bioapatite mineral is important physiologically, but the details of this stiffening are uncertain. Unresolved questions about the details of the accommodation of bioapatite within and upon collagen's hierarchical structure have posed a central hurdle, but recent microscopy data resolve several major questions. These data suggest how collagen accommodates bioapatite at the lowest relevant hierarchical level (collagen fibrils), and suggest several possibilities for the progressive accommodation of bioapatite at higher hierarchical length scales (fibres and tissue). We developed approximations for the stiffening of collagen across spatial hierarchies based upon these data, and connected models across hierarchies levels to estimate mineralization-dependent tissue-level mechanics. In the five possible sequences of mineralization studied, percolation of the bioapatite phase proved to be an important determinant of the degree of stiffening by bioapatite. The models were applied to study one important instance of partially mineralized tissue, which occurs at the attachment of tendon to bone. All sequences of mineralization considered reproduced experimental observations of a region of tissue between tendon and bone that is more compliant than either tendon or bone, but the size and nature of this region depended strongly upon the sequence of mineralization. These models and observations have implications for engineered tissue scaffolds at the attachment of tendon to bone, bone development and graded biomimetic attachment of dissimilar hierarchical materials in general.
Mechanics of mineralized collagen fibrils upon transient loads

Abstract:Collagen is a key structural protein in the human body, which undergoes mineralization during the formation of hard tissues. Earlier studies have described the mechanical behavior of bone at different scales highlighting material features across hierarchical structures. Here we present a study that aims to understand the mechanical properties of mineralized collagen fibrils upon tensile/compressive transient loads, investigating how the kinetic energy propagates and it is dissipated at the molecular scale, thus filling a gap of knowledge in this area. These specific features are the mechanisms that Nature has developed to passively dissipate stress and prevent structural failures. In addition to the mechanical properties of the mineralized fibrils, we observe distinct nanomechanical behaviors for the two regions (i.e., overlap and gap) of the D-period to highlight the effect of the mineralization. We notice decreasing trends for both wave speeds and Young s moduli over input velocity with a marked strengthening effect in the gap region due to the accumulation of the hydroxyapatite. In contrast, the dissipative behavior is not affected by either loading conditions or the mineral percentage, showing a stronger dampening effect upon faster inputs compatible to the bone behavior at the macroscale. Our results improve the understanding of mineralized collagen composites unveiling the energy dissipative behavior of such materials. This impacts, besides the physiology, the design and characterization of new bioinspired composites for replacement devices (e.g., prostheses for sound transmission or conduction) and for optimized structures able to bear transient loads, e.g., impact, fatigue, in structural applications.
Spherulitic Crystal Growth Drives Mineral Deposition Patterns in Collagen‐Based Materials

Abstract:Mineral propagates following a spherulitic growth process, following the protein scaffold provided by collagen fibrils. These spherulites exhibit a curved‐layered structure that contains a multitude of mineral platelets. Nanosized spaces remain unmineralized within the fibril, likely determined by the presence of macromolecular complexes, giving rise to the characteristic lacy pattern observed in transversal cross‐section. The formation of the hard tissues that provide support and mobility to organisms is achieved through the interplay of inorganic crystals and an organic framework composed of collagen and a small percentage of non‐collagenous proteins. Despite their clinical relevance, the mechanisms governing mineralization of the extracellular matrix are still poorly understood. By using 3D electron tomography and high‐resolution electron microscopy imaging and spectroscopy, it has been demonstrated that mineralization proceeds through a spherulitic‐like crystal growth process. First, aggregates of disordered crystals form in the interfibrillar spaces, which lead to the mineralization of adjacent fibrils. Mineral propagates steadily through the inter‐ and intrafibrillar spaces of the collagen structure forming layered spherulites that grow to confluence. The structure of the collagen fibrils serves as a protein scaffold to guide the formation of a myriad of platelet‐shaped crystallites that make up each of these spherulites. At their periphery, nanosized unmineralized areas remain, leading to the formation of the characteristic lacy pattern observed in the transversal cross‐section of mature calcified tissues. This study provides fundamental insights into the bone formation process and represents a potential strategy for complex materials design.

Abstract:Bone is mineralized tissue constituting the skeletal system, supporting and protecting body organs and tissues. At the molecular level, mineralized collagen fibril is the basic building block of bone tissue, and hence, understanding bone properties down to fundamental tissue structures enables to better identify the mechanisms of structural failures and damages. While efforts have focused on the study of the micro- and macro-scale viscoelasticity related to bone damage and healing based on creep, mineralized collagen has not been explored on a molecular level. We report a study that aims at systematically exploring the viscoelasticity of collagenous fibrils with different mineralization levels. We investigate the dynamic mechanical response upon cyclic and impulsive loads to observe the viscoelastic phenomena from either shear or extensional strains via molecular dynamics. We perform a sensitivity analysis with several key benchmarks: intrafibrillar mineralization percentage, hydration state, and external load amplitude. Our results show a growth of the dynamic moduli with an increase of mineral percentage, pronounced at low strains. When intrafibrillar water is present, the material softens the elastic component but considerably increases its viscosity, especially at high frequencies. This behaviour is confirmed from the material response upon impulsive loads, in which water drastically reduces the relaxation times throughout the input velocity range by one order of magnitude, with respect to the dehydrated counterparts. We find that upon transient loads, water has a major impact on the mechanics of mineralized fibrillar collagen, being able to improve the capability of the tissue to passively and effectively dissipate energy, especially after fast and high-amplitude external loads.

Abstract:Bone, with excellent mechanical properties, can adapt to a variety of external forces through adjust its complex hierarchical structure. The basis of the hierarchical structure is mineralized collagen fibers, which consist of mineral crystals and collagen fibers. To better understand the mechanical properties of mineralized collagen fibers, we establish a series of representative volume element models of mineralized collagen fibers based on the different arrangement of mineral crystals in the collagen fibers. The effects of crystal aspect ratio, volume fraction, and crystal defect on the effective elastic modulus and Poisson's ratio of mineralized collagen fibers are calculated by combining the homogenization theory with the finite element method. Results show that the elastic modulus of mineralized collagen fibers can be regulated effectively by adjusting the aspect ratio and volume fraction of crystals. This research explains the mechanical advantages of mineralized collagen fibers in the structure, provides a basis for the calculation of high level structure of bone and offers insight into the design of bionic composite materials.

Abstract:Despite significant efforts utilizing advanced technologies, the contentious debate surrounding the intricate mechanism underlying collagen fibril mineralization, particularly with regard to amorphous precursor infiltration and phase transformation, persists. This work proposes an amorphous calcium phosphate (ACP)‐mediated pathway for collagen fibril mineralization and utilizing stochastic optical reconstruction microscopy technology, and has experimentally confirmed for the first time that the ACP nanoparticles can infiltrate inside collagen fibrils. Subsequently, the ACP‐mediated phase transformation occurs within collagen fibrils to form HAP crystallites, and significantly enhances the mechanical properties of the mineralized collagen fibrils compared to those achieved by the calcium phosphate ion (CPI)‐mediated mineralization and resembles the natural counterpart. Furthermore, demineralized dentin can be effectively remineralized through ACP‐mediated mineralization, leading to complete restoration of its mechanical properties. This work provides a new paradigm of collagen mineralization via particle‐mediated phase transformation, deepens the understanding of the mechanism behind the mineralization of collagen fibrils, and offers a new strategy for hard tissue repair. Nonlinear Stress‐Induced Transformations in Collagen Fibrillar Organization, Disorder and Strain Mechanisms in the Bone‐Cartilage Unit

Abstract:The ultrastructural mechanisms enabling physiological compression in cartilage/bone interfaces in joints are explored using X‐ray nanomechanical imaging and 3D modeling of the hydrated collagen fibril/proteoglycan nanostructure. The approach reveals synergistic and microscale spatially‐graded stress‐induced interactions between hidden modes like fibrillar compaction, fluid flow, crystallinity transitions, and reorientation. These molecular‐to‐microscale biophysical mechanisms advance fundamental understandings of joint function and pathological degeneration. By developing a 3D X‐ray modeling and spatially correlative imaging method for fibrous collagenous tissues, this study provides a comprehensive mapping of nanoscale deformation in the collagen fibril network across the intact bone‐cartilage unit (BCU), whose healthy functioning is critical for joint function and preventing degeneration. Extracting the 3D fibril structure from 2D small‐angle X‐ray scattering before and during physiological compression reveals of dominant deformation modes, including crystallinity transitions, lateral fibril compression, and reorientation, which vary in a coupled, nonlinear, and correlated manner across the cartilage‐bone interface. A distinct intermolecular arrangement of collagen molecules, and enhanced molecular‐level disorder, is found in the cartilage (sliding) surface region. Just below, fibrils accommodate tissue strain by reorientation, which transitions molecular‐level kinking or loss of crystallinity in the deep zone. Crystalline fibrils laterally shrink far more (20×) than they contract, possibly by water loss from between tropocollagen molecules. With the calcified plate acting as an anchor for surrounding tissue, a qualitative switch occurs in fibrillar deformation between the articular cartilage and calcified regions. These findings significantly advance this understanding of the complex, nonlinear ultrastructural dynamics at this critical interface, and opens avenues for developing targeted diagnostic and therapeutic strategies for musculoskeletal disorders.

Murine Metatarsus Bone and Joint Collagen-I Fiber Morphologies and Networks Studied With SHG Multiphoton Imaging

Abstract:Chronic inflammatory disease of bones and joints (e.g., rheumatoid arthritis, gout, etc.), but also acute bone injury and healing, or degenerative resorptive processes inducing osteoporosis, are associated with structural remodeling that ultimately have impact on function. For instance, bone stability is predominantly orchestrated by the structural arrangement of extracellular matrix fibrillar networks, i.e., collagen-I, -IV, elastin, and other proteins. These components may undergo distinct network density and orientation alterations that may be causative for decreased toughness, resilience and load bearing capacity or even increased brittleness. Diagnostic approaches are usually confined to coarse imaging modalities of X-ray or computer tomography that only provide limited optical resolution and lack specificity to visualize the fibrillary collagen network. However, studying collagen structure at the microscopic scale is of considerable interest to understand the mechanisms of tissue pathologies. Multiphoton Second Harmonic Generation (SHG) microscopy, is able to visualize the sterical topology of the collagen-I fibrillar network in 3D, in a minimally invasive and label-free manner. Penetration depths exceed those of conventional visible light imaging and can be further optimized through employing decalcification or optical clearing processing ex vivo. The goal of this proof-of-concept study was to use SHG and two-photon excited fluorescence (2-PEF) imaging to mainly characterize the fibrillary collagen organization within ex vivo decalcified normal mouse metatarsus bone and joint. The results show that the technique resolved the fibrillar collagen network of complete bones and joints with almost no artifacts and enabled to study the complex collagen-I networks with various fiber types (straight, crimped) and network arrangements of mature and woven bone with high degree of detail. Our imaging approach enabled to identify cavities within both cortical and trabecular bone architecture as well as interfaces with sharply changing fiber morphology and network structure both within bone, in tendon and ligament and within joint areas. These possibilities are highly advantageous since the technology can easily be applied to animal models, e.g., of rheumatoid arthritis to study structural effects of chronic joint inflammation, and to many others and to compare to the structure of human bone.
Effect of nanostructure of mineralized collagen scaffolds on their physical properties and osteogenic potential.

Abstract:Tissue engineering has enabled development of nanostructured collagen scaffolds to meet current challenges in regeneration of lost bone. In this study, extrafibrillarly-mineralized and intrafibrillarly-mineralized collagen scaffolds were fabricated separately by a conventional crystallization method and a biomimetic, bottom-up crystallization method. Atomic force microscopy (AFM) was employed to examine the nanotopography and nanomechanics of the mineralized collagen scaffolds. The in vitro cell responses to the surface of the mineralized collagen scaffolds were analyzed by laser scanning microscope and field emission scanning electron microscopy. AFM imaging showed that these two mineralized collagen scaffolds exhibited different nanostructure, including the size, morphology and location of the apatites in collagen fibrils. The nanomechanical testing demonstrated that the intrafibrillarly-mineralized collagen scaffold, with bone-like hierarchy, featured a significantly increased Young's modulus compared with the extrafibrillarly-mineralized collagen scaffold in both dry and wet conditions. However, these two mineralized collagen scaffolds had a similar thermal behavior. From the cell culture experiments, the intrafibrillarly-mineralized collagen scaffold showed higher cell proliferation and alkaline phosphatase activity than the extrafibrillarly-mineralized collagen scaffold. The utmost significance of this study is that the nanostructure of the mineralized collagen scaffolds can affect the initial cell adhesion, morphology and further osteogenic potential. The present study will help us to fabricate novel biomaterials for bone grafting and tissue engineering applications.

Abstract:Numerous studies of associations between dampness and respiratory diseases have been conducted, but their implications remain inconclusive. In this study of 13,335 parent-reported questionnaires (response rate: 85.3%), we analyzed associations between home dampness and asthma and related symptoms in 4- to 6-year-old children in a cross-sectional study of Shanghai. Indicators of home dampness were strongly and significantly associated with dry cough, wheeze, and rhinitis symptoms. In the current residence, children with visible mold spots (VMS) exposure had 32% higher risk of asthma (adjusted OR, 95% CI: 1.32, 1.07-1.64); damp clothing and/or bedding (frequently) was strongly associated with dry cough (1.78, 1.37-2.30); condensation on windows was strongly associated with hay fever (1.60, 1.27-2.01). In the early-life residence, VMS or damp stains (frequently) were strongly associated with dry cough (2.20, 1.55-3.11) and rhinitis ever (1.57, 1.11-2.21). Associations between dampness and diseases among children with or without family history of atopy were similar. The total number of dampness indicators had strong dose-response relationships with investigated health outcomes. Actions, including opening windows of the child's room at night and cleaning the child's room frequently, could potentially mitigate 25% of home VMS, thereby preventing more than 1.5% of attributable risk of the studied symptoms.
Physically based 3D finite element model of a single mineralized collagen microfibril

Abstract:Mineralized collagen microfibrils in human bone provide its mechanical properties (stiffness, elasticity, ductility, energy dissipation and strength). However, detailed 3D finite element models describing the mechanical behavior of the mineralized collagen microfibrils are still lacking. In the current work, we developed a 3D finite element model of the mineralized collagen microfibril that incorporates the physical 3D structural details. The model components consist of five tropocollagen molecules, mineral hydroxyapatite and intermolecular cross-links joining primarily the ends of the tropocollagen molecules. Dimension, arrangement and mechanical behavior of the constituents are based on previously published experimental and theoretical data. Tensile and compressive loads were applied to the microfibril under different conditions (hydrated and dehydrated collagen) to investigate the relationship between the structure and the mechanical behavior of the mineralized collagen microfibril (stress-strain curve and elastic modulus). The computational results match the experimental available data well, and provide insight into the role of the phases and morphology on the microfibril behavior. Our predicted results show that the mechanical properties of collagen microfibrils arise due to their structure and properties. The proposed 3D finite element model of mineralized collagen microfibril contributes toward the investigation of the bottom-up structure-property relationships in human bone.

Abstract:The hard tissue(tooth and bone) in vertebrates is formed by the biomineralization processes, which are regulated and controlled by bio-systems. Among them, the mineralized collagen fibril is an essential building block for the construction of biominerals. The mineralized collagen is a hybrid material of nano inorganic mineral (calcium phosphate) and assembled organic molecules(collagen). The advanced nano hierarchical and ordered structure of the mineralized collagen fibrils will endow biological hard tissue with excellent mechanical properties (such as hardness and toughness). The structure feature and mineralization process may inspire new materials fabrications aimed at hard tissue repair. It is important but challenging to reconstruct the ordered organic/inorganic hybrid structures via intrafibrillar mineralization of collagen, and that is the key to the tissue repair. In this review, the same basic questions are discussed including hierarchical structures of bones, the assembled structure of collagen fibrils and the nano-hybrid structure of mineralized collagen fibrils, the interactions between the organic(collagen and non-collagenous proteins) and inorganic counterparts and their interface, the modification of mineralizing interface by the functional molecules, and the control of collagen mineralization and their applications in tissue repair. Some unresolved but urgent issues for collagen mineralization are also discussed, such as the regulation of amorphous precursors for the infiltration into the nano-compartment of collagen fibrils, the control of the mineralization rate and degree, as well as the large scale fabrication of ordered collagen fibrils and their mineralization.

Spectroscopy and spectral analysis

Abstract:By comparing IR spectra of collagen, collagen/CaCl2 and collagen/calcium phosphate, it was found that intensities of amide I, II and III bands were significantly decreased after mineralization. Band amide I shifted to wards lower wave number. This shift indicated chemical interaction between carboxyl groups and Ca ions formed in tie mineralization. The result indicated that there was another nucleation site, i.e. carbonyl (dropO-O) on collagen, besides the previous reported nucleation site of carboxyl (-OOOH) on collagen. The decrease of amide I peak intensity was mainly due to blockage of C-O stretch. Comparing the IR spectra of demineralized collagen and pure collagen, amide I, II and III peaks intensity and position were almost the same. When the mineral dissolved, amide peaks rebounded, which indicated that the crystals nucleated on these sites enwrapped the groups and blocked the groups vibration. The decreases of peak intensity of amide I, II and III were different due to their different vibration modes. The result confirmed that the carbonyl group was one kind of the nucleation site during collagen mineralization. The relationship between the process of collagen mineralization and variety of amide bonds was also studied.

Abstract:The absence of a conducive bone formation microenvironment between fractured ends poses a significant challenge in repairing large bone defects. A promising solution is to construct a bone formation microenvironment that mimics natural bone tissue. Biomimetic mineralized collagen possesses a chemical composition and microstructure highly similar to the natural bone matrix, making it an ideal biomimetic bone substitute material. The microstructure of biomimetic mineralized collagen is influenced by various factors, and its biomineralization and microstructure, in turn, affect its physicochemical properties and biological activity. We aimed to utilize mineralization time and solution concentration as variables and employed the polymer-induced liquid precursor strategy to fabricate mineralized collagen with diverse microstructures, to shed light on how mineralization parameters impact the material microstructure and physicochemical properties. We also investigated the influence of microstructure and physicochemical properties on cell biocompatibility and the bone-forming microenvironment. Through comprehensive characterization, we examined the physical and chemical properties of I-EMC under various mineralization conditions and assessed the in vitro and in vivo biocompatibility and osteogenic performance. By investigating the relationship between mineralization parameters, material physicochemical properties, and osteogenic performance, we revealed how microstructures influence cellular behaviors like biocompatibility and osteogenic microenvironment. Encouragingly, mineralization solutions with varying concentrations, stabilized by polyacrylic acid, successfully produced intrafibrillar and extrafibrillar mineralized collagen. Compared to non-mineralized collagen, all mineralized samples demonstrated improved bone-forming performance. Notably, samples prepared with a 1× mineralization solution exhibited relatively smooth surfaces with even mineralization. Extending the mineralization time enhanced the degree of mineralization and osteogenic performance. Conversely, samples prepared with a 2× mineralization solution had rough surfaces with large calcium phosphate particles, indicating non-uniform mineralization. Overall, our research advances the potential for commercial production of mineralized collagen protein products, characterized by dual biomimetic properties, and their application in treating various types of bone defects.

Abstract:Bone biomineralization is a complex process in which type I collagen and associated non-collagenous proteins (NCPs), including glycoproteins and proteoglycans, interact closely with inorganic calcium and phosphate ions to control the precipitation of nanosized, non-stoichiometric hydroxyapatite (HAP, idealized stoichiometry Ca10(PO4)6(OH)2) within the organic matrix of a tissue. The ability of certain vertebrate tissues to mineralize is critically related to several aspects of their function. The goal of this study was to identify specific NCPs in mineralizing and non-mineralizing tissues of two animal models, rat and turkey, and to determine whether some NCPs are unique to each type of tissue. The tissues investigated were rat femur (mineralizing) and tail tendon (non-mineralizing) and turkey leg tendon (having both mineralizing and non-mineralizing regions in the same individual specimen). An experimental approach ex vivo was designed for this investigation by combining sequential protein extraction with comprehensive protein mapping using proteomics and Western blotting. The extraction method enabled separation of various NCPs based on their association with either the extracellular organic collagenous matrix phases or the inorganic mineral phases of the tissues. The proteomics work generated a complete picture of NCPs in different tissues and animal species. Subsequently, Western blotting provided validation for some of the proteomics findings. The survey then yielded generalized results relevant to various protein families, rather than only individual NCPs. This study focused primarily on the NCPs belonging to the small leucine-rich proteoglycan (SLRP) family and the small integrin-binding ligand N-linked glycoproteins (SIBLINGs). SLRPs were found to be associated only with the collagenous matrix, a result suggesting that they are mainly involved in structural matrix organization and not in mineralization. SIBLINGs as well as matrix Gla (γ-carboxyglutamate) protein were strictly localized within the inorganic mineral phase of mineralizing tissues, a finding suggesting that their roles are limited to mineralization. The results from this study indicated that osteocalcin was closely involved in mineralization but did not preclude possible additional roles as a hormone. This report provides for the first time a spatial survey and comparison of NCPs from mineralizing and non-mineralizing tissues ex vivo and defines the proteome of turkey leg tendons as a model for vertebrate mineralization.

Biomechanics and Modeling in Mechanobiology

Abstract:Bone, because of its hierarchical composite structure, exhibits an excellent combination of stiffness and toughness, which is due substantially to the structural order and deformation at the smaller length scales. Here, we focus on the mineralized collagen fibril, consisting of hydroxyapatite plates with nanometric dimensions aligned within a protein matrix, and emphasize the relationship between the structure and elastic properties of a mineralized collagen fibril. We create two- and three-dimensional representative volume elements to represent the structure of the fibril and evaluate the importance of the parameters defining its structure and properties of the constituent mineral and collagen phase. Elastic stiffnesses are calculated by the finite element method and compared with experimental data obtained by synchrotron X-ray diffraction. The computational results match the experimental data well, and provide insight into the role of the phases and morphology on the elastic deformation characteristics. Also, the effects of water, imperfections in the mineral phase and mineral content outside the mineralized collagen fibril upon its elastic properties are discussed.

Abstract:The interplay between stem cells and their extracellular microenvironment is of critical importance to the stem cell-based therapeutics in regenerative medicine. Mineralized collagen is the main component of bone extracellular matrix, but the effect of interfacial properties of mineralized collagen on subsequent cellular behaviors is unclear. This study examined the role of surface chemistry of nanoscale mineralized collagen on human periodontal ligament stem cell (hPDLSC) fate decisions. The intrafibrillarly mineralized collagen (IMC), fabricated by a biomimetic bottom-up approach, showed a bonelike hierarchy with nanohydroxyapatites (HAs) periodically embedded within fibrils. The infrared spectrum of the IMC showed the presence of phosphate, carbonate, amide I and II bands; and infrared mapping displayed uniform and higher spatial distribution of mineralization in the IMC. However, the distribution of the phosphate group differed far from that of the amide I group in the extrafibrillarly mineralized collagen (EMC), in which flowerlike HA clusters randomly depositing around the surface of the fibrils. Moreover, a large quantity of extrafibrillar HAs covered up the C═O stretch and N-H in-plane bend, resulting in substantial reduction of amide I and II bands. Cell experiments demonstrated that the hPDLSCs seeded on the IMC exhibited a highly branched, osteoblast-like polygonal shape with extended pseudopodia and thick stress fiber formation; while cells on the EMC displayed a spindle shape with less branch points and thin actin fibril formation. Furthermore, the biocompatibility of EMC was much lower than that of IMC. Interestingly, even without osteogenic induction, mRNA levels of major osteogenic differentiation genes were highly expressed in the IMC during cultivation time. These data suggest that the IMC with a similar nanotopography and surface chemistry to natural mineralized collagen directs hPDLSCs toward osteoblast differentiation, providing a promising scaffold in bone tissue regeneration.
