Abstract:Microplastics are widespread emerging contaminants that have been found globally in the marine and freshwater ecosystem, but there is limited knowledge regarding its impact on coral reef ecosystem and underpinning mechanism. In the present study, using Pocillopora damicornis as a model, we investigated cytological, physiological, and molecular responses of a scleractinian coral to acute microplastic exposure. No significant changes were observed in the density of symbiotic zooxanthellae during the entire period of microplastic exposure, while its chlorophyll content increased significantly at 12 h of microplastic exposure. We observed significant increases in the activities of antioxidant enzymes such as superoxide dismutase and catalase, significant decrease in the detoxifying enzyme glutathione S-transferase and the immune enzyme alkaline phosphatase, but no change in the other immune enzyme phenoloxidase during the whole experiment period. Transcriptomic analysis revealed 134 significantly up-regulated coral genes at 12 h after the exposure, enriched in 11 GO terms mostly related to stress response, zymogen granule, and JNK signal pathway. Meanwhile, 215 coral genes were significantly down-regulated at 12 h after exposure, enriched in 25 GO terms involved in sterol transport and EGF-ERK1/2 signal pathway. In contrast, only 12 zooxanthella genes exhibited significant up-regulation and 95 genes down-regulation at 12 h after the microplastic exposure; genes regulating synthesis and export of glucose and amino acids were not impacted. These results suggest that acute exposure of microplastics can activate the stress response of the scleractinian coral P. damicornis, and repress its detoxification and immune system through the JNK and ERK signal pathways. These demonstrate that microplastic exposure can compromise the anti-stress capacity and immune system of the scleractinian coral P. damicornis, despite the minimal impact on the abundance and major photosynthate translocation transporters of the symbiont in the short term.
Deciphering the disturbance mechanism of BaP on the symbiosis of Montipora digitata via 4D-Proteomics approach

Abstract:The coral holobiont is mainly composed of coral polyps, zooxanthellae, and coral symbiotic microorganisms, which form the basis of coral reef ecosystems. In recent years, the severe degradation of coral reefs caused by climate warming and environmental pollution has aroused widespread concern. Benzo(a)pyrene (BaP) is a widely distributed pollutant in the environment. However, the underlying mechanisms of coral symbiosis destruction due to the stress of BaP are not well understood. In this study, diaPASEF proteomics and 16S rRNA amplicon pyrosequencing technology were used to reveal the effects of 50 μg/L BaP on Montipora digitate. Data analysis was performed from the perspective of the main symbionts of M. digitata (coral polyps, zooxanthellae, and coral symbiotic microorganisms). The results showed that BaP impaired cellular antioxidant capacity by disrupting the GSH/GSSG cycle, and sustained stress causes severe impairment of energy metabolism and protein degradation in coral polyps. In zooxanthellae, BaP downregulated the protein expression of SOD2 and mtHSP70, which then resulted in oxidative free radical accumulation and apoptosis. For coral symbiotic microorganisms, BaP altered the community structure of microorganisms and decreased immunity. Coral symbiotic microorganisms adapted to the stress of BaP by adjusting energy metabolism and enhancing extracellular electron transfer. BaP adversely affected the three main symbionts of M. digitata via different mechanisms. Decreased antioxidant capacity is a common cause of damages to coral polyps and zooxanthellae, whereas coral symbiotic microorganisms are able to appropriately adapt to oxidative stress. This study assessed the effects of BaP on corals from a symbiotic perspective, which is more comprehensive and reliable. At the same time, data from the study supports new directions for coral research and coral reef protection.
Pentachlorophenol Alters the Acetate-Assimilating Microbial Community and Redox Cycling in Anoxic Soils

Abstract:Acetate is a common electron donor that can drive microbial reductive processes in anaerobic environments. Apart from acting as a terminal electron acceptor, chlorinated organic pollutant of pentachlorophenol (PCP) has antimicrobial properties but little is known about its effect on anaerobic microbial populations during bioremediation. To elucidate the effect of PCP on the anaerobic microbial community, DNA-based stable isotope probing was performed using C-13-acetate as a substrate in two depths (0-20 cm and 80-100 cm) of mangrove soils. The addition of PCP had little influence on Fe(III) reduction, but dramatically inhibited the SO42- reduction, resulting in a high emission of CH4 under anoxic conditions. Correspondingly, PCP significantly affected the composition of prokaryotic C-13-labeled OTUs (Operational Taxonomic Units) at both soil depths. Members of Clostridiales (Firmicutes) and Burkholderiales (Betaproteobacteria) were dominant C-13-acetate utilizers and potential PCP degraders. The PCP addition decreased sulfate reduction process through inhibition of classical sulfate- and sulfur-reducing bacteria belonging to families of Desulfarculaceae, Desulfobulbaceae and Desulfobacteraceae. Our study showed that indigenous microbial communities associated with terminal electron-accepting pathways (e.g. Fe3+ and SO42- reduction) changed differently during PCP dechlorination. These findings suggest a great impact of chlorinated pollutants on the soil biogeochemical processes.

Abstract:In this letter, an ultra-wideband (UWB) bandpass filter (BPF) based on CPW-to-microstrip transition structure is proposed. To improve its selectivity, an interdigital coupled-line and split ring resonator (SRR) are used to generate two extra transmission zeros located at the lower and upper edge of the passband separately. The measured results show that the proposed filter possesses satisfactory performance such as compact size, sharp roll-off, and wide fractional bandwidth of 133 % (2.35-11.75 GHz). Moreover, a notched band with deep attenuation introduced by coupling T-shaped short stub (CTSSS) is achieved to block the ITU8.0 band with centre frequency of 8.2 GHz. And the notched band is controllable. This filter can be a good candidate for ultra-wideband applications.

Pentachlorophenol Exposure Induced Neurotoxicity by Disrupting Citrulline Metabolism in Larvae and Adult Zebrafish.

Abstract:Pentachlorophenol (PCP) is a ubiquitous environmental toxicant with various adverse effects. Although its neurotoxicity has been reported, the underlying mechanism and subsequent detoxification remain unclear. In this study, embryos and adult zebrafish were exposed to PCP to determine its potential neurotoxic mechanism and protective indicators. The survival rate, heart rate, mobility time, active status and moving distance were significantly decreased in larvae after 30 μg/L PCP exposure. Likewise, the mobile time, latency to the first movement, velocity and moving distance of adult zebrafish were significantly reduced by PCP exposure. Untargeted metabolomics analysis of larvae revealed that arginine and proline metabolism was the primary pathway affected by PCP exposure, reflected by increased proline and decreased citrulline (CIT) contents, which were confirmed by quantitative data. PCP exposure suppressed the conversion from arginine to CIT in larvae by downregulating the expression of nos1 and nos2a. Ornithine content was increased in the brains and intestines of adult zebrafish after PCP exposure, which inhibited ornithine catabolism to CIT by downregulating otc, resulting in reduced CIT. Intriguingly, CIT supplementation significantly restored the neurobehavioral defects induced by PCP in larvae and adult zebrafish. CIT supplementation upregulated the expression of ef1α and tuba1 in larvae and inhibited the downregulation of ef1α in the brains of adult zebrafish. Taken together, these results indicated that CIT supplementation could protect against PCP-induced neurotoxicity by upregulating the expression of genes involved in neuronal development and function.

Abstract:As microplastics (MP) become ubiquitous, their interactions with heavy metals threatens the coral ecosystem. This study aimed to assess the combined toxicity of MP and copper (Cu) in the environment of coral. Goniopora columna was exposed to polyethylene microplastics (PE-MP) combined with Cu2+ at 10, 20, 50, 100, and 300 μg/L for 7 days. Polyp length and adaptability were recorded daily, and coral samples were collected at 1, 3, 5, and 7 days to analyse zooxanthellae density and antioxidant activity. Tissue observations and the analysis of MP and Cu2+ accumulation were conducted on the 7th day. After 1 day of exposure, PE-MP combined with different concentrations of Cu2+ significantly decreased polyp length and adaptability compared with PE-MP alone. Simultaneously, a significant increase in malondialdehyde (MDA) content, lead to coral oxidative stress, which was a combined effect with PE-MP. After 3 days of exposure, PE-MP combined with Cu2+ at >50 μg/L significantly reduced zooxanthellae density, damaging the coral's symbiotic relationship. In antioxidant enzyme activity, superoxide dismutase (SOD) activity decreased significantly after 1 day of exposure. After 3 days of exposure, glutathione peroxidase (GPx) activity significantly increased with Cu2+ at >20 μg/L. After 5 days of exposure, PE-MP combined with different concentrations of Cu2+ significantly reduced catalase (CAT), glutathione (GSH), and glutathione transferase (GST) activity, disrupting the antioxidant enzyme system, and acting antagonistically to PE-MP alone. Tissue observations revealed that the PE-MP combined with Cu2+ at >50 μg/L caused severe mesenteric atrophy, vacuolar, and Cu2+ accumulation in the coral mesenteric compared with PE-MP alone. The results suggest that combined exposure of PE-MP and copper leads to more severe oxidative stress, disruption antioxidant enzyme system, tissue damage, and Cu2+ accumulation, resulting in a significant maladaptation of corals to the environment.
Physiological Responses of Pocillopora acuta and Porites lutea Under Plastic and Fishing Net Stress

Abstract:Marine debris has become a global problem affecting coral health around the globe. However, the photophysiological responses of corals to marine debris stress remain unclear. Therefore, this study firstly investigated transparent and opaque plastic bag shading and fishing nets directly contacting the coral. Photosynthetic performance, pigment content, symbiont density, and calcification rate of a branching coral Pocillopora acuta and a massive coral Porites lutea were investigated after 4 weeks of exposure to marine debris. The results show that the maximum quantum yield of PSII significantly decreased in P. lutea with all treatments, while P. acuta showed no effect on the maximum quantum yield of PSII from any treatments. Transparent plastic bag shading does not affect P. acuta , but significantly affected the maximum photochemical efficiency of P. lutea . Photoacclimation of cellular pigment content was also observed under opaque plastic bag shading for both species at week 2. Fishing nets had the strongest effect and resulted in P. acuta bleaching and P. lutea partial mortality as well as a decline in zooxanthellae density. Calcification rate of P. acuta significantly decreased with treatments using opaque plastic bag and fishing net, but for P. lutea only the treatment with fishing net gave any observable effects. This study suggests that the sensitivities of corals to marine debris differ strongly by species and morphology of the coral.

Abstract:Pollution of marine environments with microplastic particles has increased rapidly during the last few decades and its impact on marine lives have recently gained attention in both public and scientific community. Scleractinian corals are the foundation species of coral reef ecosystems that are greatly affected by the microplastics (MPs), yet little is known about the effects of microplastics on the coral azooxanthellate. In the present study, effects of the exposure and ingestion of polyvinyl chloride (PVC), polyethylene (PE), polyethylene terephthalate (PET), and polyamide 66 (PA66) were studied on the physiological responses of Tubastrea aurea. Our results shows that coral ingested microplastics in four treatment groups and the exposure of microplastics inhibited the antioxidant capacity, immune system, calcification and energy metabolism of the coral Tubastrea aurea. Superoxide dismutase (SOD), catalase (CAT), alkaline phosphatase (AKP), and total antioxidant capacity (TAC) were reduced by 29.4%, 35.5%, 73.9%, and 52.2% in the corals exposed to PVC, respectively. PET microplastics impacted more severely on pyruvate kinase (PK), Na, K-ATPase (Na, K-ATP), Ca-ATPase (Ca-ATP), Mg-ATPase (Mg-ATP), Ca-Mg-ATPase (Ca, Mg-ATP), and glutathione (GSH). Activity of these enzymes decreases to 89.6%, 66.7%, 63.6%, 60.4%, 48.4%, and 50.5% respectively. We anticipate that this work will provide important preliminary data for better understanding the effects of MPs on stony corals azooxanthellate.

Abstract:Microplastic pollution in the ocean is a major problem, as its pervasiveness elicits concerns the health impacts microplastics may have on marine life (such as reef-building corals). As a primary endpoint, the organismal lipidome can define the weakening of fitness and reveal the physiological context of adverse health effects in organisms. To gain insight into the effects of microplastics on coral health, lipid profiling was performed via an untargeted lipidomic approach on the coral Turbinaria mesenterina exposed to ~10 μm polystyrene microparticles for 10 days. Considerable microplastic accumulation and obvious effects relating with immune activation were observed in the coral treated with a near environmentally relevant concentration of microplastics (10 μg/L); however, these effects were not evident in the high level (100 μg/L) treatment group. In particular, increased levels of membrane lipids with 20:4 and 22:6 fatty acid chains reallocated from the triacylglycerol pool were observed in coral host cells and symbiotic algae, respectively, which could upregulate immune activity and realign symbiotic communication in coral. High levels of polyunsaturation can sensitize the coral cell membrane to lipid peroxidation and increase cell death, which is of greater concern; additionally, the photoprotective capacity of symbiotic algae was compromised. As a result, coral physiological functions were altered. These results show that, realistic levels of microplastic pollution can affect coral health and should be a concern.

Abstract:Shoulder pain may be caused by a variety of abnormalities including glenohumeral instability, rotator cuff disease, cervical radiculopathy, nerve entrapment syndromes, neoplasm, tendinitis, adhesive capsulitis, trauma, and degenerative disease of any one of the joints about the shoulder.1, 13, 16 Patients experiencing entrapment neuropathy of the suprascapular nerve usually complain of shoulder pain.21 Symptoms involve the infraspinatus muscle if the entrapment takes place at the spinoglenoid notch, or the supraspinatus and infraspinatus muscles if the lesion is at the suprascapular notch. 3,5,22,24,30 Mass lesions such as ganglion cysts are a common cause of suprascapular nerve entrapment. 8,12,17,18,25 Glenoid labral abnormalities, including acute tears and degenerative changes, are associated with glenohumeral instability.4,9 There is evidence that periarticular abnormalities in other areas of the body may lead to the leakage of synovial fluid and the formation of so-called ganglion cystS.7,1l,15,20 We report a case of ganglion cyst of the shoulder associated with a posterior glenoid labral tear and glenohumeral instability.
Differential bioaccumulation and tolerances of massive and branching scleractinian corals to polycyclic aromatic hydrocarbons in situ

Abstract:Scleractinian corals are capable of accumulating polycyclic aromatic hydrocarbons (PAHs) in reef environments; however, the mechanism behind their PAHs tolerance is unknown. This study investigated the occurrence and bioaccumulation of PAHs in coral reef ecosystems and examined the physiological responses induced by PAHs in coral hosts and their algal symbionts, the massive coral Galaxea fascicularis and branching coral Pocillopora damicornis . G. fascicularis had a higher PAHs accumulation capacity than P. damicornis . Both the coral hosts and algal symbionts preferentially accumulated acenaphthene, dibenzo( a , h )anthracene, and benzo( a )pyrene. The accumulated PAHs by G. fascicularis and P. damicornis hosts was accompanied by a reduction in detoxification ability. The accumulated PAHs could induce oxidative stress in P. damicorni hosts, thus G. fascicularis demonstrated a greater tolerance to PAHs compared to P. damicornis . Meanwhile, their algal symbionts had fewer physiological responses to accumulated PAHs than the coral hosts. Negative effects were not observed with benzo( a )pyrene. Taken together, these results suggest massive and branching scleractinian corals have different PAHs bioaccumulation and tolerance mechanisms, and indicate that long-term PAHs pollution could cause significant alterations of community structures in coral reef ecosystems.

Abstract:Pentachlorophenol (PCP) represents a critical concern worldwide due to its toxicity and recalcitrance to degradation. The capacity of Mucor plumbeus to transform PCP into several detoxification metabolites, including tetrachlorohydroquinone and several phase II conjugates, was observed by LC-HRMS. The data obtained support the degradation pathway proposed previously. PCP effects in M. plumbeus, an unsequenced species, were investigated using a proteomics approach (bidimensional gel electrophoresis followed by MALDI TOF/TOF analyses). The mycelial proteins identified in the differentially accumulated spots allowed the identification of PCP responsive proteins. The presence of PCP increased the energy demand, altered the cell wall architecture and cytoskeleton and induced a significant stress response. The latter was emphasised by the up-accumulation of protein species associated with defence mechanisms (e.g. HSP70 and cytochrome c peroxidase). Overall the data produced corroborate the capability of PCP to uncouple oxidative-phosphorylation in mitochondria. Importantly, one of the identified mycelial protein species, a NAD- and Zn-dependent ADH, is likely to be involved in PCP degradation. Amongst the fungal secretome analysed, no putative PCP degradative enzymes were detected. This work constitutes the first toxicoproteomic study involving a Zygomycota fungus and the very first concerning the effect of PCP in a fungal proteome.

Abstract:SUMMARY Disease epizootics are increasing with climatic shifts, yet within each system only a subset of species are identified as the most vulnerable. Understanding ecological immunology patterns as well as environmental influences on immune defenses will provide insight into the persistence of a functional system through adverse conditions. Amongst the most threatened ecosystems are coral reefs, with coral disease epizootics and thermal stress jeopardizing their survival. Immune defenses were investigated within three Caribbean corals, Montastraea faveolata, Stephanocoenia intersepta and Porites astreoides, which represent a range of disease and bleaching susceptibilities. Levels of several immune parameters were measured in response to elevated water temperature and the presence of a commercial pathogen-associated molecular pattern (PAMP) – lipopolysaccharide (LPS) – as an elicitor of the innate immune response. Immune parameters included prophenoloxidase (PPO) activity, melanin concentration, bactericidal activity, the antioxidants peroxidase and catalase, and fluorescent protein (FP) concentration. LPS induced an immune response in all three corals, although each species responded differently to the experimental treatments. For example, M. faveolata, a disease-susceptible species, experienced significant decreases in bactericidal activity and melanin concentration after exposure to LPS and elevated temperature alone. Porites astreoides, a disease-resistant species, showed increased levels of enzymatic antioxidants upon exposure to LPS independently and increased PPO activity in response to the combination of LPS and elevated water temperature. This study demonstrates the ability of reef-building corals to induce immune responses in the presence of PAMPs, indicating activation of PAMP receptors and the transduction of appropriate signals leading to immune effector responses. Furthermore, these data address the emerging field of ecological immunology by highlighting interspecific differences in immunity and immunocompetences among Caribbean corals, which are reflected in their life-history characteristics, disease susceptibilities and bleaching-induced mortality.

Organic ultraviolet filter mixture promotes bleaching of reef corals upon the threat of elevated seawater temperature

Abstract:Global reef degradation is a critical environmental health issue that has triggered intensive research on ocean warming, but the implications of emerging contaminants in coral habitats are largely overlooked. Laboratory experiments assessing organic ultraviolet (UV) filter exposure have shown that these chemicals negatively affect coral health; their ubiquitous occurrence in association with ocean warming may pose great challenges to coral health. We investigated both short- (10-day) and long-term (60-day) single and co-exposures of coral nubbins to environmentally relevant organic UV filter mixtures (200 ng/L of 12 compounds) and elevated water temperatures (30 °C) to investigate their effects and potential mechanisms of action. The initial 10-day exposure of Seriatopora caliendrum resulted in bleaching only under co-exposure conditions (compounds + temperature). The 60-day mesocosm study entailed the same exposure settings with nubbins of three species ( S. caliendrum , Pocillopora acuta and Montipora aequituberculata ). Bleaching (37.5 %) and mortality (12.5 %) of S. caliendrum were observed under UV filter mixture exposure. In the co-exposure treatment, 100 % S. caliendrum and P. acuta bleached associating with 100 % and 50 % mortality, respectively, and significant increase of catalase activities in P. acuta and M. aequituberculata nubbins were found. Biochemical and molecular analyses indicated significant alteration of oxidative stress and metabolic enzymes. The results suggest that upon the adverse effects of thermal stress, organic UV filter mixture at environmental concentrations can cause bleaching in corals by inducing a significant oxidative stress and detoxification burden, suggesting that emerging contaminants may play a unique role in global reef degradation.
