Abstract:Lidocaine hydrochloride is used as an anesthetic in many clinical applications. This short communication investigates the effect of complete substitution of lidocaine hydrochloride for deionized (DI) water on the physico-chemical properties of two novel glass polyalkenoate cements. Substituting DI water with lidocaine hydrochloride resulted in cements with shorter working times but comparable setting times and mechanical properties. Fourier transform infrared spectroscopy confirmed that the setting reaction in cements containing DI water and lidocaine hydrochloride was completed within 24 h, post cement preparation and maturation. Further, it was explained that lidocaine hydrochloride binds to poly(acrylic) acid (PAA) due to electrostatic forces between the positively charged amino group of lidocaine hydrochloride and the carboxylic group of the PAA, resulting in a compact poly-complex precipitate.

Abstract:This study aimed to investigate the fluoride release properties and the effect on bond strength of two experimental adhesive cements. Synthesized particles of nano-fluorapatite (nano-FA) or nano-fluorohydroxyapatite (nano-FHA) were incorporated into a resin-modified glass ionomer cement (Fuji Ortho LC) and characterized using X-ray diffraction and scanning electron microscopy. Blocks with six different concentrations of nano-FA or nano-FHA were manufactured and their fluoride release properties evaluated by ultraviolet spectrophotometry. The unaltered glass ionomer cement Fuji Ortho LC (GC, control) and the two experimental cements with the highest fluoride release capacities (nano-FA+Fuji Ortho LC (GFA) and nano-FHA+Fuji Ortho LC (GFHA)) were used to bond composite blocks and orthodontic brackets to human enamel. After 24h water storage all specimens were debonded, measuring the micro-tensile bond strength (μTBS) and the shear bond strength (SBS), respectively. The optimal concentration of added nano-FA and nano-FHA for maximum fluoride release was 25wt.%, which nearly tripled fluoride release after 70days compared with the control group. GC exhibited a significantly higher SBS than GFHA/GFA, with GFHA and GFA not differing significantly (P>0.05). The μTBS of GC and GFA were significantly higher than that of GFHA (P⩽0.05). The results seem to indicate that the fluoride release properties of Fuji Ortho LC are improved by incorporating nano-FA or nano-FHA, simultaneously maintaining a clinically sufficient bond strength when nano-FA was added.

Abstract:Glass polyalkenoate cements (GPCs) have potential for skeletal cementation. Unfortunately, commercial GPCs all contain, and subsequently release, aluminum ions, which have been implicated in degenerative brain disease. The purpose of this research was to create a series of aluminum-free GPCs constructed from silicate (SiO₂), calcium (CaO), zinc (ZnO) and sodium (Na₂O)-containing glasses mixed with poly-acrylic acid (PAA) and to evaluate the potential of these cements for cranioplasty applications. Three glasses were formulated based on the SiO₂-CaO-ZnO-Na₂O parent glass (KBT01) with 0.03 mol % (KBT02) and 0.06 mol % (KBT03) germanium (GeO₂) substituted for ZnO. Each glass was then mixed with 50 wt % of a patented SiO₂-CaO-ZnO-strontium (SrO) glass composition and the resultant mixtures were subsequently reacted with aqueous PAA (50 wt % addition) to produce three GPCs. The incorporation of Ge in the glass phase was found to result in decreased working (142 s to 112 s) and setting (807 s to 448 s) times for the cements manufactured from them, likely due to the increase in crosslink formation between the Ge-containing glasses and the PAA. Compressive (σc) and biaxial flexural (σf) strengths of the cements were examined at 1, 7 and 30 days post mixing and were found to increase with both maturation and Ge content. The bonding strength of a titanium cylinder (Ti) attached to bone by the cements increased from 0.2 MPa, when placed, to 0.6 MPa, after 14 days maturation. The results of this research indicate that Germano-Silicate based GPCs have suitable handling and mechanical properties for cranioplasty fixation.
Evaluation of dental cements derived from mixtures of highly reactive ionomer glasses and bottle glass: Cement manipulation, mechanical, fluoride ion releasing, radiopaque and setting properties

Abstract:Objectives: To evaluate the mechanical properties, fluoride release, radiopacity, and setting characteristics of dental cements derived from highly reactive ionomer glasses and bottle glass mixtures. Methods: Two highly reactive glass series, LG99 and LG117, were synthesized, milled, sieved, and characterized using XRD and laser particle size analysis. These glasses were mixed with predetermined ratios of ground bottle glass, poly(acrylic acid), and aqueous tartaric acid to form glass ionomer cements. The cements' working time (WT), setting time (ST), fluoride release, radiopacity, compressive strength (CS), and elastic modulus (EM) were evaluated. Mean differences in CS were analyzed using multivariate ANOVA with Tukey's post hoc test at p = 0.05. Results: The WT and ST for both groups ranged from 1.5 to 2.5 min. LG99 series cements showed significantly higher CS (∼65 MPa) and EM (∼2 GPa) than LG117 series (p < 0.05). Both series showed similar fluoride release profiles, peaking at 1.2 mmol/L at 28 days. Radiopacity for LG99 ranged from 0.97 to 1.34, while LG117 ranged from 0.60 to 0.95. Solid state 27Al magic-angle spinning-nuclear magnetic resonance (MAS NMR) confirmed the presence of Al(IV) and Al(VI), indicating setting completion by one day for both series. Bottle glass showed a chemical shift at 55.8 ppm, overlapping with LG99's Al(IV) signal. The 19F MAS NMR spectra revealed Al-F and F-Sr(n) species in all glasses, with LG117 forming CaF2 after one day in deionized water. Conclusion: Mixtures of highly reactive ionomer glass and bottle glass produced cements with satisfactory properties for dental applications. Further research is needed to optimize their formulation and properties.

Abstract:The short time of action and systemic toxicity of local anesthetics limit their clinical application. The objective of this work was to develop a modified release system for the local anesthetic lidocaine (LDC), using polyvinylpyrrolidone-Tetrahydroxyborate (PVP-THB) hydrogel to improve the pharmacological properties of the drug and their anesthetic activities investigated. The mechanical properties, rheological, swelling ratio, and water content behavior of candidate formulations were investigated. An increase in THB concentrations increased the storage modulus (G') and the loss modulus (G″) but decreased the loss tangent (tan δ < 1). In vitro release of PVP and THB were assessed to evaluate potential clinical dosing of free species originating from the hydrogel structure. The profiles of PVP-THB/LDC showed that the release of associated LDC was slower than that of the free drug. Evaluating the cytotoxic effect of these hydrogels on an in vitro mouse fibroblast (L929) cells showed their good cytotoxic activity towards L929 prolonged its anesthetic action. Particles were characterized using a scanning electron microscope (SEM) and atomic force microscopy (AFM) analyses confirming the amorphous nature and the uniformity of LDC inclusion in the PVP-THB hydrogel. The continuous release of LDC from the PVP-THB hydrogel was observed for more than three days, without a remarkable initial burst, which is probably owing to the stable entrapment of LDC in the PVP-THB core of the hydrogel. We evaluated the analgesic effect of PVP-THB/LDC in the carrageenan-induced arthritis rats model. Results showed that PVP-THB/LDC has a stronger and longer analgesic effect when administered in inflamed areas. This new formulation could potentially be used in future applications involving the gradual release of local anesthetics.

Abstract:In this study, an injectable dopamine-modified alginate hydrogel combined with nano-hydroxyapatite or strontium hydroxyapatite was cross-linked via the in situ slow release of divalent ions from the hydrolysis of D-glucono-delta-lactone (GDL). The cross-linking mechanism involved the chelation of an alginate with divalent ions and the hydrogen-bonding interaction between alginate and catechol groups. As the GDL concentration at 0.5% (w/v) regulated the release of the divalent ions, which enhanced the chelation effect, it was investigated, and inorganic nanoparticles were homogeneously distributed into the hydrogel system to improve the mechanical properties and stabilize the hydrogel network. Furthermore, the effects of dopamine on the improvement of the mechanical properties were investigated. Results demonstrated that the compressive strength of this injectable hydrogel is enhanced by 4.6 times compared with that of the hydrogel without the addition of GDL, and the compressive strength is enhanced by 4.0 times compared with that of the unmodified alginate hydrogel at strain values ranging from 10% to 50%. The hydrogels combined with strontium hydroxyapatite exhibited a more compact, suitable pore size, with a three-dimensional network structure, and the particle size was 50-100 mu m; this range of values is considered as optimal for the growth of bone tissues. In addition, the gelation time can be adjusted from a few minutes to a few seconds by adjusting experiment variables. Our study provides a potential method for improving the mechanical properties of bone tissue engineering materials. Abstract:Abstract Glass ionomer cements (GIC) are used in restorative dentistry and their properties (low heat during setting, adhesion to mineralised tissue and surgical metals) make them of great interest for bone applications.However, dental GIC are based on aluminium-containing glasses, and the resulting release of aluminium ions from the cements needs to be avoided for applications as bone cements. Replacing aluminium ions in glasses for use in glass ionomer cements is challenging, as aluminium ions play a critical role in the required glass degradation by acid attack as well as in GIC mechanical stability. Magnesium ions have been used as an alternative for aluminium in the glass component, but so far no systematic study has looked into the actual role of magnesium ions. The aim of the present study is therefore the systematic comparison of the effect of magnesium ions compared to calcium ions in GIC glasses. It is shown that by partially substituting MgO for CaO in simple SiO2-CaO-CaF2 glasses, ion release from the glass and, subsequently, GIC setting behaviour can be adjusted. Magnesium ions act as typical network modifiers here but owing to their larger field strength compared to calcium ions reduce ion release from the glasses significantly. By choosing an optimum ratio of magnesium and calcium ions in the glass, GIC setting and subsequently compressive strength can be controlled.

Abstract:Nonpregnant and pregnant rats were continuously exposed for 3 weeks to an aerosol containing 1, 3 and 10 mg lead/m3 air and to a combination of 3 mg Pb/m3 and 500 ppm carbon monoxide. At the two lower lead doses, fetal blood lead levels exceeded those of the mothers. Active transport mechanisms were discussed to be responsible for these differences. A decrease of the fetal blood lead level below the maternal level in the high exposure group was explained by an increasing storage capacity of the fetal livers with increasing lead doses. Lead concentrations of the maternal livers exceeded the nonpregnant values at all 3 doses, probably caused by a higher ventilation and altered pharmacokinetics of lead in pregnancy. Additional CO-inhalation lowered the storage capacity of the livers of the adult animals and raised the blood concentration. In the fetuses additional CO-inhalation raised liver lead concentrations.
Setting Characteristics and High Compressive Strength of an Anti-washout, Injectable Calcium Phosphate Cement Combined with Thermosensitive Hydrogel

Abstract:In this work, a thermosensitive poly(D,L-lactide-co-glycolide)-poly(ethylene glycol)-poly(D,L-lactide-co-glycolide) (PLGA-PEG-PLGA) hydrogel was introduced into calcium phosphate cement (CPC) to enhance the anti-washout property of CPC. The effects of the hydrogel on the setting time, injectability, anti-washout property and compressive strength of CPC were thoroughly investigated. The results showed that the hydrogel significantly increased the injectability and anti-washout property of CPC, meanwhile maintained the setting time with an acceptable range. Moreover, the hydrogel improved the initial compressive strength of CPC. The composite cement with 20% v/v hydrogel in the liquid phase showed fine crystals of hydration product, a more compact microstructure and lower porosity compared with control CPC. The analysis of X-ray diffraction (XRD), infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) indicated that suitable volume ratio of hydrogel (20% v/v) in the setting liquid of CPC could promote the formation of hydroxyapatite in the early hydration period. The degradation behavior of the cement was characterized by immersion tests in simulated body fluid. The hydrogel had no adverse effect on the degradation rate of CPC over the immersion period of 23 days. This study indicated that incorporating PLGA-PEG-PLGA hydrogel could be a promising strategy to reinforce the handing properties and initial compressive strength of calcium phosphate cement.

Abstract:Local anesthetics (LAs) are a class of drugs, which have wide applications in the treatment of post-operative pain care management. Long-acting LAs that can be given as a single dose analgesic are desperately needed. The prepared sulfobutylether β-cyclodextrin (SCD)/hyaluronic acid (HA) hydrogels were characterized by Fourier transform infrared and X-ray diffraction patterns. The as-prepared SCD/HA hydrogels greatly enhanced their swelling behavior and in vitro degradation properties. Furthermore, a scanning electron microscope reported that the surface morphology of the Lidocaine (LDC)-loaded SCD/HA hydrogels was smooth, and a significant change in porosity was observed after the addition of LDC (0.5%, 1.0%, and 1.5% w/v). The occurrence of cross-linking between the LDC and SCD/HA hydrogels was studied through rheological analysis. Approximately 94% of lidocaine (LDC) was released from the SCD/HA-LDC formulations by 24 h. The cytotoxicity of SCD/HA-LDC was studied against fibroblast (3T3) cells. SCD/HA-LDC showed a prolonged in vitro release and lower cytotoxicity when compared to free LDC. Furthermore, in vivo evaluation of the anesthetic properties in animal models showed that the SCD/HA-LDC showed a significantly prolonged analgesic effect when compared to free LDC. Moreover, the SCD/HA-LDC exhibited good biodegradability and biocompatibility in histological analyses. Overall, the findings suggest that the LDC-loaded SCD/HA hydrogels had a synergistic impact in prolonging analgesia without generating toxicity, and hence might be used as a long-acting analgesia therapeutic care.

Abstract:In pain relief, lidocaine has gained more attention as a local anesthetic. However, there are several side effects that limit the use of local anesthetics. Therefore, it is hypothesized that a hydrogel system with facile design can be used for prolonged release of lidocaine. In this study, we developed a formulation comprises of sodium alginate (SA) and graphene oxide (GO) to prolong the release of lidocaine. The gelation was induced by physically crosslinking the alginate with Ca2+ ions. The formation of blank SA and GO-reinforced SA hydrogels was investigated with different concentration of Ca2+ ions. The controlled release of lidocaine hydrochloride (LH) on both hydrogel systems was studied in PBS solution. The GO-reinforced SA hydrogels exhibited more sustained release than SA hydrogels without GO. In vitro biocompatibility test in L929 fibroblast cells confirmed the non-toxic property of hydrogels. Furthermore, to prove the in-situ gelation and biodegradability of hydrogels the hydrogels were injected on mice model and confirmed the stable gel formation. The hydrogels implanted onto the subcutaneous tissue of hydrogels retained over one week. These results indicate that LH-loaded GO-reinforced SA hydrogel can be a potential biomaterial for controlled release of local anesthetics.

Chemical characterization of a degradable polymeric bone adhesive containing hydrolysable fillers and interpretation of anomalous mechanical properties

Abstract:An experimental, light-curable, degradable polyester-based bone adhesive reinforced with phosphate glass particles ((P(2)O(5))(0.45)(CaO)(x)(Na(2)O)(0.55-)(x), x=0.3 or 0.4mol) or calcium phosphate (monocalcium phosphate/beta-tricalcium phosphate (MCPM/beta-TCP)) has been characterized. Early water sorption (8wt.% at 1week) by the unfilled set adhesive catalysed subsequent bulk degradation (4wt.% at 2weeks) and substantial decline in both elastic and storage moduli. Addition of phosphate glass fillers substantially enhanced this water sorption, catalysed greater bulk mass loss (40-50 and 52-55wt.%, respectively) but enabled generation of a microporous scaffold within 2weeks. The high levels of acidic polymer degradation products (38-50wt.% of original polymer) were advantageously buffered by the filler, which initially released primarily sodium trimetaphosphate (P(3)O93-). Calcium phosphate addition raised polymer water sorption to a lesser extent (16wt.%) and promoted intermediate early bulk mass loss (12wt.%) but simultaneous anomalous increase in modulus. This was attributed to MCPM reacting with absorbed water and beta-TCP to form more homogeneously dispersed brushite (CaHPO(4)) throughout the polymer. Between 2 and 10weeks, linear erosion of both polymer (0.5wt.%week(-1)) and composites (0.7-1.2wt.%week(-1)) occurred, with all fillers providing long-term buffer action through calcium and orthophosphate (PO43-) release. In conclusion, both fillers can raise degradation of bone adhesives whilst simultaneously providing the buffering action and ions required for new bone formation. Through control of water sorption catalysed filler reactions, porous structures for cell support or substantially stiffer materials may be generated.

Abstract:The main aim of this study was to assess the impact of the environment on the mechanical and tribological properties of glass-carbomer cements used in dentistry. The properties of the Glass Cements Polyalkene (GCP) Glass Fill material, belonging to glass-polyalkene cements, were tested after placing it in various environments: air, distilled water, artificial saliva simulating a neutral environment (pH = 7), and simulating inflammation (pH = 4). The research material included four samples and a two-year reference material. The analysis of volumetric consumption and the assessment of the impact of solubility on the stability of glass-carbomer cements were carried out using tribological measurements and Vickers hardness measurements. In addition, microstructural characterization of the materials was performed using scanning electron microscopy (SEM). It was observed that the lowest wear (0.04%), the most stable microstructure, and the lowest average hardness (21.52 HV 0.1) were exhibited by the material stored in artificial saliva simulating a neutral environment (pH = 7). The least stable microstructure and statistically the highest hardness (77.3 HV 0.1) was observed in the test sample, which was stored in air for two years and then in distilled water. The highest consumption (0.11%) was recorded in the case of cement placed in artificial saliva simulating inflammation (pH = 4). The results obtained in this study indicate specific trends in the influence of the environment in which the tested cement is located, such as air, distilled water, air/distilled water, artificial saliva simulating a neutral environment, and simulating inflammation, on its structure, hardness, and wear.

Abstract:Polymethyl methacrylate (PMMA) bone cement is now widely used in percutaneous vertebro plasty (PVP) and percutaneous kyphoplasty (PKP). However, studies showed that the radiopacifiers (zirconia, barium sulfate, etc.) added to PMMA will have a negative impact on its use, e.g. barium sulfate will weaken the mechanical properties of bone cement and lead to bone absorption and aseptic loosening. Iodine is an element existing in the human body and has good imaging performance. Iodine contrast agent has been used in clinic for many years and has abundant clinical data. Therefore, using iodine instead of barium sulfate may be a promising choice. In this paper, the effect of different content of diatrizoate sodium (DTA, C11H8I3N2NaO4) on the properties of PMMA was studied and compared with the traditional PMMA bone cement containing 30 wt% barium sulfate. The mechanical properties, setting properties, radiopacity, and biocompatibility of bone cement were evaluated. The compressive strength of PMMA bone cement with 20 wt% DTA can reach 76.38 MPa. DTA released from bone cement up to 14 days accounted for only 2.3% of its dosage. The water contact angle was 62.3°. The contrast of bone cement on X-ray film was comparable to that of bone cement containing 30 wt% barium. The hemolysis rate was lower than 4%, and there was no obvious hemolysis. PMMA with 20 wt% DTA can maintain the relative growth rate of MC3T3-E1 and L929 cells above 80%. The results show that adding 20 wt% DTA into PMMA can obtain good radiopacity while maintaining its mechanical properties, setting properties, and biocompatibility. DTA can be used as a promising candidate material for PMMA bone cement radiopacifier.
Molecular interactions of hydrated co-amorphous systems of prilocaine and lidocaine

Abstract:It is generally accepted that water as a plasticizer can decrease the glass transition temperatures (T g s) of amorphous drugs and drug excipient systems. However, previous studies suggest that water, as an anti-plasticizer, can increase the T g s of co-amorphous systems of prilocaine (PRL) and lidocaine (LID). In order to investigate the intermolecular interactions between water and co-amorphous PRL-LID systems, Fourier transform infrared spectroscopy (FTIR) and principal component analysis (PCA) were conducted. Water was found to bind with the carbonyl groups of PRL and LID molecularly evenly in the hydrated co-amorphous PRL-LID systems. Quantum chemical simulations visually confirmed the interactions between water and co-amorphous PRL-LID systems. Furthermore, the physical stability of hydrated co-amorphous PRL-LID systems was improved due to the anti-plasticizing effect of water, compared with the anhydrous samples. The preference of water to interact with the carbonyl groups of PRL and LID as binding sites could be associated with the anti-plasticizing effect of water on the co-amorphous PRL-LID systems.

Abstract:The principal goal of the present investigation was to enterprise new and effective drug delivery vesicle for the sustained delivery of local anesthetic lidocaine hydrochloride (LDC), using a novel combination of copolymeric hydrogel with tetrahydroxyborate (COP–THB) to improve bioactivity and therapeutic potential. To support this contention, the physical and mechanical properties, rheological characteristics, and component release of candidate formulations were investigated. An optimized formulation of COP–THB containing LDC to an upper maximum concentration of 1.5% w/w was assessed for drug crystallization. The biocompatibility of the prepared COP–THB hydrogel was exhibited strong cell survival (96%) and growth compatibility on L929 fibroblast cell lines, which was confirmed by using methods of MTT assay and microscopic observations. The COP–THB hydrogel release pattern is distinct from that of COP–THB/LDC hydrogels by the slow-release rate and the low percentage of cumulative release. In vivo evaluations were demonstrated the anesthetic effects and toxicity value of treated samples by using mice models. In addition, COP–THB/LDC hydrogels significantly inhibit in vivo tumor growth in mice model and effectively reduced it is in vivo toxicity. The pharmacological evaluation showed that encapsulation of LDC in COP–THB hydrogels prolonged its anesthetic action with favorable in vitro and in vivo compatibility. This novel design may theoretically be used in promising studies involving the controlled release of local anesthetics.Development a modified sustained release system for the local anesthetic lidocaine.PVP-THB hydrogel to improve the pharmacological properties of the drug and their anesthetic activities.Profiles of PVP-THB/LDC showed that the effective release of associated lidocaine.This new formulation could potentially be used in future local anesthetics.

Influence of Water on Amorphous Lidocaine

Abstract:Water is generally regarded as a universal plasticizer of amorphous drugs or amorphous drug-containing systems. A decrease in glass-transition temperature (Tg) is considered the general result of this plasticizing effect. A recent study exhibits that water can increase the Tg of amorphous prilocaine (PRL) and thus shows an anti-plasticizing effect. The structurally similar drug lidocaine (LID) might show similar interactions with water, and thus an anti-plasticizing effect of water is hypothesized to also occur in amorphous LID. However, the influence of water on the Tg of LID cannot be determined directly due to the very low stability of LID in the amorphous form. It is possible to predict the Tg of LID from a co-amorphous system of PRL-LID using the Gordon-Taylor equation. Interactions were observed between PRL and LID based on the deviations between the experimental Tgs and the Tgs calculated by the conventional use of the Gordon-Taylor equation. A modified use of the Gordon-Taylor equation was applied using the optimal co-amorphous system as a separate component and the excess drug as the other component. The predicted Tg of fully hydrated LID could thus be determined and was found to be increased by 0.9 ± 0.7 K compared with the Tg of water-free amorphous LID. It could be shown that water exhibited a small anti-plasticizing effect on LID.
Surface modification of a novel glass to optimise strength and deliverability of an injectable alginate composite

Abstract:It is estimated that 1–6% of the adult population have an intracranial aneurysm. Aneurysm coiling is the current preferred treatment method; however, over 20% of coiled aneurysms recur. A novel glass–alginate composite hydrogel has been developed to treat aneurysms, which is designed to completely fill the aneurysm space and prevent aneurysm recurrence. This hydrogel is composed of a polymeric alginate, a novel bioactive glass and glucono-delta-lactone. This novel injectable hydrogel exhibits characteristics suitable for the treatment of cerebral aneurysms. However, poor hydrophilicity of the glass phase results in inhomogeneity and agglomerate formation within the composite, resulting in suboptimal deliverability and strength. This study examines the effect of surface modification of the glass particles using an acid washing technique, designed to increase glass surface hydrophilicity resulting in a homogeneous sample. This study found that acid washing of the glass not only decreased agglomeration and inhomogeneity but also lengthened working times and increased strength of the resultant hydrogel. This lengthened working time, allowed for an increased glass content and, as a result, further increased compressive strength and radiopacity of the resultant hydrogel. Glass particle size analysis revealed that the relative quantity of fine particles was reduced. Surface analysis of the glass particles revealed an increase in hydrophilic silanol groups and increased surface network connectivity. These factors, combined with a decreased surface calcium and an increased surface gallium content, are postulated as the likely reasons for the observed increased strength, working time and hardening time.

Development of novel strontium containing bioactive glass based calcium phosphate cement

Abstract:Objective: The aim of this study was to investigate the effect on properties of increasing strontium substitution for calcium in bioactive glasses used as precursors for novel calcium phosphate cements. Methods: Glasses were produced by progressively substituting strontium for calcium. Cements were prepared by mixing the glass powder with Ca(H2PO4)2 powder with a 2.5% solution of Na2HPO4. Setting times and compressive strength were measured after 1h, 1 day, 7 days and 28 days immersion in Tris buffer solution. X-ray diffraction (XRD), Fourier transform infrared spectroscopy and radiopacity were measured and crystal morphology was assessed using scanning electron microscopy. Results: A correlation between the phases formed, morphology of the crystallites, setting time and compressive strength were analyzed. Setting time increased proportionally with strontium substitution in the glass up to 25%, whereas for higher substitutions it decreased. Compressive strength showed a maximum value of 12.5MPa and was strongly influenced by the interlocking of the crystals and their morphology. XRD showed that the presence of strontium influenced the crystal phases formed. Octacalcium phosphate (Ca8H2(PO4)6·5H2O, OCP) was the main phase present after 1h and 1 day whereas after 28 days OCP was completely transformed to strontium-containing hydroxyapatite (SrxCa(10-x)(PO4)6(OH)2, SrHA). Radiopacity increased proportionally to strontium substitution in the glass. Significance: A novel method to develop a bone substitute forming in vitro SrHA as a final product by using a bioactive glass as a precursor was shown. These novel injectable bioactive glass cements are promising materials for dental and orthopedic applications. Further in vivo characterizations are being conducted.
