Abstract:Ricotta cheese, particularly the ovine type, is a typical Italian dairy product obtained by heat-coagulation of the proteins in whey. The aim of this work was to investigate the influence of whey protein concentration, obtained by ultrafiltration, on yield of fresh ovine ricotta cheese. Ricotta cheeses were obtained by thermocoagulation of mixtures with protein content of 1.56, 3.10, 4.16, and 7.09g/100g from the mixing of skim whey and ultrafiltered skim whey. A fat-to-protein ratio of 1.1 (wt/wt) was obtained for all mixtures by adding fresh cream. The initial mixtures, as well as the final ricotta cheeses, were analyzed for their composition and by SDS-PAGE. Protein bands were quantified by QuantityOne software (Bio-Rad, Hercules, CA) and identified by liquid chromatography-tandem mass spectrometry. Significant differences in the composition of the ricotta cheese were observed depending on protein concentration. Particularly, ricotta cheese resulting from the mixture containing 7.09g/100g of protein presented higher moisture (72.88±1.50g/100g) and protein (10.18±0.45g/100g) contents than that prepared from the mixture with 1.56g/100g of protein (69.52±1.75 and 6.70±0.85g/100g, respectively), and fat content was lower in this sample (12.20±1.60g/100g) compared with the other treatments, with mean values between 15.72 and 20.50g/100g. Each protein fraction presented a different behavior during thermocoagulation. In particular, the recovery of β-lactoglobulin and α-lactalbumin in the cheese increased as their content increased in the mixtures. It was concluded that concentrating ovine rennet whey improved the extent of heat-induced protein aggregation during the thermal coagulation process. This resulted in a better recovery of each protein fraction in the product, and in a consequent increase of ricotta cheese yield.
Validity and reliability of hand and electronic timing for 40-yd sprint in college football players

Abstract:The 40-yd sprint is the premier event for evaluating sprint speed among football players at all competitive levels. Some questions remain concerning the validity of hand timing compared with electronic timing, as well as the lack of assessment and reliability of each method. The purpose of this study was to evaluate the validity of hand timing by experienced and novice timers compared with electronic timing and to establish the reliability and smallest worthwhile difference (SWD) of each method for the 40-yd sprint. National Collegiate Athletic Association (NCAA) Division I college football players (n = 81) ran two 40-yd sprint trials, with each being timed electronically (touch pad start and infrared beam stop) and with hand-held stopwatches by 2 experienced and 4 novice timers. There was no significant difference between trials timed electronically or by experienced and novice timers. Hand timing (experienced = 4.90 ± 0.34 seconds; novice = 4.86 ± 0.33 seconds) produced a significantly faster 40-yd sprint time than electronic timing (5.12 ± 0.35 seconds) by 0.22 ± 0.07 and 0.26 ± 0.08 seconds, respectively. Relative reliability was extremely high for all comparisons with intraclass correlation coefficient >0.987. The SWD was 0.12 seconds with electronic timing and 0.14 seconds with hand timing. In conclusion, hand timing produces faster sprint times than electronic timing in college football players, independent of timer experience. Repeated 40-yd sprint trials have high relative reliability regardless of timing method. A meaningful change in 40-yd sprint performance is dependent on timing method used.

Abstract:Abstract The purpose of the study was to assess straight sprinting, change of direction ability and horizontal jump performance in youth runners according to age and gender. Two hundred and fifty-five youth runners (116 boys and 139 girls) participated in this study. The athletes were divided according to their age into five groups: under 8 yr (U8), under 10 yr (U10), under 12 yr (U12), under 14 yr (U14) and under 16 yr (U16). Significant differences (p 0.05, d = 0.29-1.17) between U14 and U16 were observed in any of the tests. With regard to age and gender, in U8 and U10 groups there were no significant differences (p > 0.05, d = 0.02-0.76) between boys and girls in any test. However, in U12 and U14 groups, significant gender differences (p < 0.05, d = 0.85-1.24) were found in the MAT. Likewise, the boys obtained better results than girls in the horizontal jump tests (p < 0.05, d = 1.01-1.26). After the classification by age, some differences were observed between both genders, depending on the fitness variable evaluated.

Abstract:Purpose : To compute reference centiles for 5- and 30-m sprint times relative to chronological and skeletal age in youth soccer players. Subsequently, to compare individual’s sprint performance scores derived from the chronological and skeletal age reference centiles. Methods : Sprint times were collected for a sample of male U11 to U19 soccer players (n = 1745 data points). Skeletal age data were available for a subsample (n = 776 data points). Reference centiles were fitted using generalized additive models for location, scale, and shape. Individual z scores relative to chronological and skeletal age reference centiles were computed and compared for each maturity group (late, on-time, early, and very early) using standardized mean differences (SMD). Results : Reference centiles for chronological age increased more rapidly between 10.5 and 15.5 years, while reference centiles for skeletal age increased more rapidly between 13.0 and 16.5 years. Differences in chronological and skeletal z scores for very early (SMD: −0.73 to −0.43) and late (SMD: 0.58 to 1.29) maturing players were small to large, while differences for early (SMD: −0.30 to −0.19) and on-time (SMD: 0.16 to 0.28) were trivial to small. Conclusion : Reference centiles provide a valuable tool to assist the evaluation of sprint performance in relation to chronological and skeletal age for talent identification purposes in youth soccer players.

Abstract:When portfolio managers trade the stocks of companies run by people with whom they have social connections, these trades earn better returns than trades in companies with whom they have no connections (Cohen et al., 2008). We look at the effects of social connections from the firm’s side, examining the compensation of firm executives. Executive compensation in connected firms is substantially higher than in unconnected firms. The channel through which this result occurs appears to be share voting—connected funds are more likely to vote against shareholder-initiated proposals on executive compensation, thereby protecting their cronies from the discipline of corporate governance. The evidence is consistent with higher compensation being the quid pro quo for information flow from firm to fund.

J Strength Cond Res XX(X): 000–000, 2024—The primary aim was to examine the relationship between sprint acceleration performance and the performance tests: isometric squat (ISqT), countermovement jump (CMJ), and reactive strength index (RSI). The secondary aim determined whether these tests could distinguish between sprint performance levels. Twenty-six male under-21 subjects completed the ISqT, with peak force (PF), relative PF, force at 100, 150, and 200 milliseconds (ms), and impulse (0–200 ms) measured. Jump height (JH) was calculated from the CMJ and RSI during the 10-5 repeated jump test. Subjects completed 3 × 30-m sprints with splits taken at 5, 10, 20, and 30 m. Pearson’s product moment correlations were used to assess the relationships between measures, and independent samples t tests were used to determine whether differences existed in sprint ability of those in the top and bottom quartiles for force at 100 ms, CMJ, and RSI. Significant negative moderate correlations were reported between force at 100 ms and 0–5 m and 5–10 m, significant moderate and large negative correlations between CMJ and all splits and significant large correlations between RSI and splits over 5 m. Force at 100 ms distinguished between performance levels from 0 to 5 m, CMJ from 10 to 20 m, and RSI from 20 to 30 m; faster subjects produced significantly higher force, JHs, and RSI scores. Three principal components explaining 86.1% of the variation in the data set were identified: sprint times and stretch shortening cycle capabilities (33.3%), time-specific force (29.9%), and absolute and relative strength (22.8%).

Abstract:Considering the vast physical and neural developments experienced throughout adolescence, the reliability of physical performance may vary in youth populations. This study aimed to examine the reliability of change of direction (COD) and agility tests in youth soccer players. Altogether, 86 youth soccer players, aged 13.6 ± 2.0 years, volunteered to participate. Data were collected from a modified 505 COD test (m505COD) and the Y-sprint drill in both pre-planned (Y-SprintPRE) and reactive (Y-SprintREACT) conditions during 2 sessions, 7 days apart. Anthropometric data including body mass, standing stature, and sitting height were also collected. COD and agility tests demonstrated good reliability (ICC = 0.81–0.91; CV = 1.2–2.0; d = 0.00–0.31; p < 0.01) for our entire sample. However, we observed a small negative relationship between age and intersession differences for the Y-SprintPRE (r = −0.28; p = 0.04), and moderate negative relationships between both age (r = −0.41; p < 0.01), and maturity offset (r = −0.39; p < 0.01) for the Y-SprintREACT. Although the COD and agility tests adopted within this study possess good intersession reliability, we observed greater intersession differences for younger and less mature individuals. We suggest that while COD and agility tests may provide meaningful objective data for monitoring the development of youth soccer players, these tests should be used with caution when evaluating younger, more immature athletes.
Reliability and validity of sprint performance using the Alex7 motorized device

Abstract:Introduction: Advancements in technology have recently made it possible to implement effective training solutions across different environmental conditions. This study evaluated the reliability and validity of measures obtained from the innovative motorized device, Alex7 (Inosportas, Lithuania), and differences in speed and kinematic characteristics between resisted and assisted sprinting in young football players. Methods: Twenty-seven male athletes (mean age: 16.5 ± 0.8 years; height: 179.5 ± 6.9 cm; body weight: 67.7 ± 8.3 kg) each performed 30-m sprints twice under three different conditions: regular, resisted, and assisted sprinting. The Alex7 device provided the assistance and resistance during sprints. Results were compared with those from Witty timing gates. Ground contact time, flight time, stride length, and pace were measured using the OptoJump system. Reliability was assessed using two-way mixed intraclass correlation coefficients (ICCs) for single measures, the standard error of the mean (SEM), and the coefficient of variation (CV). Pearson's correlation coefficient determined the associations between Alex7 and Witty timing systems. Criterion-referenced validity was based on the mean difference and CV. Systematic bias was determined by limits of agreement using Bland-Altman analysis. Results: Running times obtained using the Alex7 equipment exhibited good to excellent test-retest reliability between sessions (ICC, 0.83-0.94) and good to excellent correlation (Pearson's r = 0.88-0.98) between the Alex7 and Witty systems in both assisted and resisted running conditions. However, the Alex7 device consistently produced longer running times than the Witty device (up to 0.16 s difference, p < 0.001). The different running conditions produced substantial variations in kinematic variables, such as stride length, ground contact time, and running speed (p < 0.001 for all), but the effects on flight time and running pace were smaller. Discussion: The Alex7 device shows high reliability for creating resisted and assisted running conditions for young football players. However, it tends to overestimate running time, necessitating caution when assessing the time parameters.
Sex‐based differences in track running distances of 100, 200, 400, 800, and 1500m in the 8 and under and 9–10‐year‐old age groups

Abstract:There are contradictory claims regarding sex‐based differences in athletic performance before puberty, but there has been minimal evaluation of sex‐based differences in competitive running performance before puberty. The purpose of this project was to determine if there are prepubertal sex‐based differences in track running performance. Finalist times from the USA Track and Field National Youth Outdoor Championships and National Junior Olympic Championships during the years 2016–2023 for running distances of 100, 200, 400, 800, and 1500m in the 8 and under and 9–10‐year‐old age groups were analyzed for sex‐based differences. In the 8 and under age group, the males were, on average, faster (p = 0.000, Hedges' g = 0.632–0.834) than females by 4.0% in the distance of 100m, 4.7% in 200m, 5.3% in 400m, 6.7% in 800m, and 6.1% in 1500m. In the 9–10‐year‐old age group, the males were, on average, faster (p = 0.000, Hedges' g = 0.584–1.089) than females by 2.9% in the distance of 100m, 4.6% in 200m, 4.0% in 400m, 4.0% in 800m, and 5.9% in 1500m. In each distance and age group between 2016 and 2023, the individual fastest male was faster than the individual fastest female by 3.7 ± 2.3%. The present data indicate that, in elite competition, males in the 8 and under and 9–10‐year‐old age groups typically run faster than females of the same age by 2.9%–6.7% for running distances of 100, 200, 400, 800, and 1500m. These findings are of particular importance as government agencies and sports organizations consider policies regarding single sex sports competition for youth.

Abstract:Many analysts advocatepatient exit as a strategy for consumers who experience poor-quality care. Exit is believed to have the potential to improve patient welfare by having patients leave (or "exit") poor-performing health care providers, thus signaling their dissatisfaction with the quality of care they have received and thereby admonishing those providers to improve. However, the validity of exit as a signal of consumer dissatisfaction hinges on how closely it reflects dissatisfaction. Intergroup differences in the propensity to exit could also result in unintended consequences. This article examines the association between consumer experience and the decision to change one's usual care providers. It also investigates if there are any intergroup differences in the propensity for changing providers according to insurance status, gender, and race or ethnicity. Data come from household surveys conducted by the Center for Studying Health System Change. Results show significant intergroup differences in propensity for switching usual source of care for voluntary or involuntary reasons related to insurance, rural residency, age, income, race, and ethnicity. Policy implications of the empirical results on exit, voice, and consumerism are discussed.
The Retest Reliability of Common Tests to Assess Physical Qualities in Adolescent Rugby League Players

Abstract:Purpose: Rigorous evidence concerning the reliability of physical tests among young athletes is lacking. Therefore, this study aimed to comprehensively determine the retest reliability of the most commonly used tests to assess physical qualities in adolescent rugby league players, and to provide the standard error of measurement and minimal detectable change for each test to support objective athlete monitoring. Method: A repeated-measures design was employed with 50 adolescent, schoolboy rugby league players (16.2 [1.3] y) completing the same testing battery across 2 consecutive weeks. Within each week, stature, body mass, and ∑4 skinfold thickness assessments, as well as countermovement jump, medicine ball throw, and 1-repetition maximum bench press, back squat, and prone row tests were conducted on the first testing day. Forty-eight hours later, a second testing day contained the 10- and 20-m linear sprints, 505-Agility Test, L-run Test, and Multistage Fitness Test. Results: Nonsignificant (P > .05), trivial differences (dav = 0.00-0.16) were evident between trials in all tests. The coefficient of variation ranged from 0.05% to 4.02% across tests. The intraclass correlation ranged from .704 to .999. All tests presented relatively low standard error of measurement, with accompanying minimal detectable change provided. Conclusion: These data can help inform decision making among end-users when implementing tests in a repeated manner and interpreting testing data.

Abstract:The aims of the present study were to determine anthropometric and physical characteristics according to age and gender of young athletes, and to quantify the relationships among these characteristics. Sixty-four young high-level runners were divided into two groups according to their age: U14 = under 14 year’s athletes; U16 = under 16 year’s athletes. The U16 group presented higher values of the sprint 15 m (p < 0.01, d = 1.14) and modified agility test (p < 0.01, d = 0.66) than the U14 group. However, the boys group presented higher values in the 505 agility test (p < 0.01, d = 0.63), modified agility test (p < 0.01, d = 1.26), horizontal countermovement jump (p < 0.05, d = 0.56), vertical countermovement jump (p < 0.01, d = 0.61) and arm swim vertical countermovement jump (p < 0.01, d = 0.63) than the girls group.

Exploratory Analysis of Sprint Force-Velocity Characteristics, Kinematics and Performance across a Periodized Training Year: A Case Study of Two National Level Sprint Athletes

Abstract:Objective: This case study aimed to explore changes to sprint force-velocity characteristics across a periodized training year (45 weeks) and the influence on sprint kinematics and performance in national level 100-meter athletes. Force-velocity characteristics have been shown to differentiate between performance levels in sprint athletes, yet limited information exists describing how characteristics change across a season and impact sprint performance, therefore warranting further research. Methods: Two male national level 100-meter athletes (Athlete 1: 22 years, 1.83 m, 81.1 kg, 100 m time: 10.47 s; Athlete 2: 19 years, 1.82 cm, 75.3 kg, 100 m time: 10.81 s) completed 12 and 11 force-velocity assessments, respectively, using electronic timing gates. Sprint mechanical characteristics were derived from 30-meter maximal sprint efforts using split times (i.e., 0-10 m, 0-20 m, 0-30 m) whereas step kinematics were established from 100-meter competition performance using video analysis. Results: Between the preparation (PREP) and competition (COMP) phase, Athlete 1 showed significantly large within-athlete effects for relative maximal power (PMAX), theoretical maximal velocity (v0), maximum ratio of force (RFMAX), maximal velocity (VMAX), and split time from 0 to 20 m and 0 to 30 m (-1.70 ≤ ES ≥ 1.92, p ≤ 0.05). Athlete 2 reported significant differences with large effects for relative maximal force (F0) and RFMAX only (ES: ≤ -1.46, p ≤ 0.04). In the PREP phase, both athletes reported almost perfect correlations between F0, PMAX and 0-20 m (r = -0.99, p ≤ 0.01), however in the COMP phase, the relationships between mechanical characteristics and split times were more individual. Competition performance in the 100-meter sprint (10.64 ± 0.24 s) showed a greater reliance on step length (r ≥ -0.72, p ≤ 0.001) than step frequency to achieve faster performances. The minimal detectable change (%) across mechanical variables ranged from 1.3 to 10.0% while spatio-temporal variables were much lower, from 0.94 to 1.48%, with Athlete 1 showing a higher 'true change' in performance across the season compared to Athlete 2. Conclusions: The estimated sprint force-velocity data collected across a training year may provide insight to practitioners about the underpinning mechanical characteristics which affect sprint performance during specific phases of training, plus how a periodized training design may enhance sprint force-velocity characteristics and performance outcomes.

Abstract:The objective of the current study was to explore the correlation between repeated sprint sets (RSS) ability and several physical attributes, including maximum sprint speed, maximal aerobic speed, maximal anaerobic speed, aerobic capacity, and explosive strength. Moreover, the aim was to assess the suitability of RSS as a comprehensive evaluation tool for physical qualities and to determine which physical field tests most accurately predict RSS in elite young male soccer players. A total of thirty-two young elite male soccer players (mean age 14.6 ± 0.3 years; predicted years from peak height velocity (PHV): − 0.4 ± 0.3; years in training: 3.7 ± 0.5) voluntarily participated in the study. The players participated in eight consecutive specific physical tests, with a minimum 72-h recovery between each session to minimize the impact of fatigue during the second trial. The participants completed the tests in the following order: RSS test, Vam-Eval test, a constant velocity test performed until exhaustion at 100% of vVO 2 max (tlim100), 20-m Multi-Stage Shuttle Run test (V MSRT ), Yo-Yo Intermittent Recovery Test level 1 (Yo-Yo IR1), Maximal Anaerobic Shuttle Running Test (V MASRT ), Maximal Sprinting Speed Test (20-m flying sprint), Countermovement Jump (CMJ), and Standing Long Jump test (SLJ). The results of the study showed that there were very large negative correlations between tlim100 and SST (sum of sprint times), and large negative correlations between Yo-Yo IR1, Vam-Eval, and SST during RSS in young elite male soccer players ( p < 0.05). Additionally, V MASRT and SLJ demonstrated a moderate negative correlation with SST ( p < 0.05). In contrast, significant positive correlations were found between 20-m flying sprint and the SST ( p < 0.05). According to the stepwise multiple linear regression analysis, the primary predictors of SST, ranked by importance, were tlim100 and Yo-Yo IR1. These two predictors collectively accounted for 72% of the variance in players' SST ( p < 0.0001). Due to the importance of aerobic capacity and short repeated accelerations/sprint sets for overall competitive performance in soccer, in conclusion, our results suggest that elite young male soccer players should perform both high intensity interval training and aeorobic capactity exercises as part of soccer training if the primary outcome is to improve repeated sprint ability performance.

Abstract:Sprint performance plays a crucial role in various sports. Short sprints vary in court size and competitive characteristics but are common in many sports. Although the relationship between age and muscle strength has been explored in short sprints, there is limited understanding of how various physical factors interact, particularly concerning differences in the acceleration phase. This study examined the relationship between sprint times at 0 to 2.5 m, 2.5 to 5 m, and 5 to 10 m intervals and various factors in junior athletes. Results indicated that sprint times increased with age, and is correlated with muscle strength and flexibility. Partial correlation analysis showed that faster times in the 0 to 2.5 m interval were associated with higher hip flexibility; in the 2.5 to 5 m interval, faster times were associated with higher core flexibility; and in the 5 to 10 m interval, a relationship with standing long jump performance was confirmed. Furthermore, lower fat free body weight translated to higher performance. In the acceleration phase of 10 m, flexibility immediately after the start and the subsequent horizontal propulsive force is important factors that are strongly related to performance change in each interval. The study results illuminate the mechanism behind short sprints in junior athletes and emphasize the importance of daily training, considering the sprint distance required for each sport.

Neonatal rat cardiac fibroblasts express three types of voltage-gated K+ channels: regulation of a transient outward current by protein kinase C.

Abstract:Cardiac fibroblasts regulate myocardial development via mechanical, chemical, and electrical interactions with associated cardiomyocytes. The goal of this study was to identify and characterize voltage-gated K(+) (Kv) channels in neonatal rat ventricular fibroblasts. With the use of the whole cell arrangement of the patch-clamp technique, three types of voltage-gated, outward K(+) currents were measured in the cultured fibroblasts. The majority of cells expressed a transient outward K(+) current (I(to)) that activated at potentials positive to -40 mV and partially inactivated during depolarizing voltage steps. I(to) was inhibited by the antiarrhythmic agent flecainide (100 microM) and BaCl(2) (1 mM) but was unaffected by 4-aminopyridine (4-AP; 0.5 and 1 mM). A smaller number of cells expressed one of two types of kinetically distinct, delayed-rectifier K(+) currents [I(K) fast (I(Kf)) and I(K) slow (I(Ks))] that were strongly blocked by 4-AP. Application of phorbol 12-myristate 13-acetate, to stimulate protein kinase C (PKC), inhibited I(to) but had no effect on I(Kf) and I(Ks). Immunoblot analysis revealed the presence of Kv1.4, Kv1.2, Kv1.5, and Kv2.1 alpha-subunits but not Kv4.2 or Kv1.6 alpha-subunits in the fibroblasts. Finally, pretreatment of the cells with 4-AP inhibited angiotensin II-induced intracellular Ca(2+) mobilization. Thus neonatal cardiac fibroblasts express at least three different Kv channels that may contribute to electrical/chemical signaling in these cells.
Small-sided game-related physical performance is not influenced by the sprint and power performance of youth male soccer players

Abstract:The aim of this study was to analyse the effects of sprint and power performance on physical fitness and small-sided game-related physical performance in youth male soccer players, using a median split analysis to separate faster and slower players, and powerful and weaker ones. Thirty youth male soccer players (age = 16.9 ± 1.4 years; height = 174.1 ± 7.1 cm; body mass = 63.1 ± 7.9 kg; % body fat = 15.5 ± 3.2) completed the following physical tests: 40 m linear sprint test, repeated sprint ability (RSA) test, countermovement jump (CMJ), horizontal jump (HJ) and a half-squat (HS) power test. In addition, players completed a 4 vs. 4 with goalkeepers small-sided game while external loads were recorded. According to their sprint and HS power performance, players were divided into fast and slow, and powerful and weak. Faster players performed better in 5 m sprint (p = 0.004, Effect Size (ES) = −1.158), 10 m sprint (p 0.05) were observed between powerful and weaker players in the fitness tests and SSG-related physical performance. These results highlight the importance of grouping youth soccer players by their sprinting capacity to design specific and individualized training strategies and suggest that SSG-related physical performance is not influenced by their sprint or power performance.

Abstract:The purpose of this study was to assess validity and reliability of sprint performance outcomes measured with an iPhone application (named: MySprint) and existing field methods (i.e. timing photocells and radar gun). To do this, 12 highly trained male sprinters performed 6 maximal 40-m sprints during a single session which were simultaneously timed using 7 pairs of timing photocells, a radar gun and a newly developed iPhone app based on high-speed video recording. Several split times as well as mechanical outputs computed from the model proposed by Samozino et al. [(2015). A simple method for measuring power, force, velocity properties, and mechanical effectiveness in sprint running. Scandinavian Journal of Medicine & Science in Sports. https://doi.org/10.1111/sms.12490] were then measured by each system, and values were compared for validity and reliability purposes. First, there was an almost perfect correlation between the values of time for each split of the 40-m sprint measured with MySprint and the timing photocells (r = 0.989-0.999, standard error of estimate = 0.007-0.015 s, intraclass correlation coefficient (ICC) = 1.0). Second, almost perfect associations were observed for the maximal theoretical horizontal force (F0), the maximal theoretical velocity (V0), the maximal power (Pmax) and the mechanical effectiveness (DRF - decrease in the ratio of force over acceleration) measured with the app and the radar gun (r = 0.974-0.999, ICC = 0.987-1.00). Finally, when analysing the performance outputs of the six different sprints of each athlete, almost identical levels of reliability were observed as revealed by the coefficient of variation (MySprint: CV = 0.027-0.14%; reference systems: CV = 0.028-0.11%). Results on the present study showed that sprint performance can be evaluated in a valid and reliable way using a novel iPhone app.

Abstract:Castrate male sheep (wethers, average liveweight 38 +/- 0.6 kg) were given one of the following diets for 10 weeks followed by euthanasia (n = 8/group): high-energy high-protein providing 1-5 times the energy required to maintain liveweight (maintenance) (group 1.5 M), low-energy low-protein at 0.5 maintenance (0.5 M), or low-energy high-protein at 0.5 maintenance (0.5 M + P). 1.5 M wethers gained 22% liveweight whereas 0.5 M and 0.5 M + P wethers lost 18 and 13% liveweight respectively. Relative to the 1.5 M group, the 0.5 M and 0.5 M + P groups had similar plasma concentrations of glucose and cortisol throughout, but elevated non-esterified fatty acids (P < 0.001) and reduced IGF-I and insulin (P < 0.05, 0.01 or 0.001) from 1 week onwards. Each week blood samples were taken every 12 min for 4 h and plasma assayed for LH. Mean concentration over 4 h, LH pulse frequency and LH pulse amplitude showed no progressive change in 1.5 M sheep. However, in both 0.5 M and 0.5 M + P groups mean LH increased (P < 0.001 and P < 0.01 respectively), pulse frequency decreased (P < 0.01 and P < 0.01) and pulse amplitude increased (P < 0.001 and P < 0.01) over the 10-week period. Anterior pituitary. LH content was greater in 0.5 M (P < 0.01) and 0.5 M + P (P < 0.05) than in 1.5 M sheep. Coronal sections (20 microns) of hypothalamic brain tissue were subjected to in situ hybridisation to determine gene expression for neuropeptide Y (NPY). NPY mRNA was concentrated in the arcuate nucleus and median eminence, with total amounts greater in both 0.5 M (310%, P < 0.001) and 0.5 M + P (333%, P < 0.01) groups than in 1.5 M sheep (100%). These data reveal that chronic low dietary energy intake by long-term castrates, with high or low protein intake, reduces LH pulse frequency but increases the circulating levels of LH by virtue of an increase in pulse amplitude, and concomitantly increases hypothalamic NPY gene expression.
