Abstract:Monochloramine (NH2Cl) is a physiological oxidant produced by activated neutrophils. In the present work, we studied the underlying mechanism of cytotoxic effects of NH2Cl on an undifferentiated rat pheochromocytoma PC12 cell line and the protective effects of antioxidants. The cells treated with 100μM NH2Cl exhibited signs of apoptotic cell death such as phosphatidylserine exposure and caspase activation. To understand the mechanism of NH2Cl cytotoxicity, we examined the effect of various kinds of antioxidants including α-tocopherol (α-Toc) and β-tocopherol (β-Toc). These antioxidants exerted a protective effect against NH2Cl-induced cell death, and α-Toc exhibited the most potent inhibitory effect among the antioxidants used. A loss of cellular glutathione was observed in the cells treated with 100μM NH2Cl. The formation of reactive oxygen species (ROS) was also measured using the fluorescent probe dichlorofluorescin diacetate. The fluorescence intensity increased prior to cell death and an antioxidant, such as α-Toc, suppressed the increase in ROS. Interestingly, β-Toc also exerted similar inhibitory effects on cytotoxicity and caspase activation. These results suggest that free radical mediated process is involved in NH2Cl-induced PC12 cell death and that tocopherols inhibit this cell death via antioxidative function.
Acute Effect of Omethoate on Serum Enzyme and the Protective Role of Vitamin C in Rats

Abstract:Objective To investigate the changes of serum malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px)in rats with acute omethoate intoxication and the ameliorative effects of vitamins C.Methods An experimental model of 72 Wistar rats was made by subcutaneous injection with 0 1 LD_ 50 and 0 5 LD_ 50 omethoate.Rats were equally assigned to the control group,0 1 LD_ 50 omethoate group,0 5 LD_ 50 omethoate group and 0.5 LD_ 50 omethoate +VitC(200mg/kg) group.Serum MDA,SOD and GSH-Px were determined at various intervals.Results Serum MDA level increased significantly in 0 1 LD_ 50 and 0 5 LD_ 50 omethoate group compared to the control group (P0 05),and increased significantly in 0 5 LD_ 50 omethoate group compared to 0 1 LD_ 50 omethoate group (P0 05),but decreased significantly in 0 5 LD_ 50 omethoate +VitC group (P0 05).The activity of SOD and GSH-Px decreased significantly in 0 1 LD_ 50 and 0 5 LD_ 50 omethoate group compared to the control group (P0 05),decreased significantly in 0 5 LD_ 50 omethoate group compared to 0 1 LD_ 50 omethoate group (P0 05),and increased significantly in 0 5 LD_ 50 omethoate +VitC group (P0 05).Conclusions The levels of lipid peroxidation go up and the activities of anti-oxidant enzyme go down in rats with acute omethoate intoxication,and treatment with vitamin C can reduce lipid peroxidation caused by omethoate.
[Effect of low doses of active chlorine on commensal populations of intestinal bacteria in rats by oral ingestion]

Abstract:Chlorine-containing substances are widely used as disinfectants for treating equipment surfaces and technological aids for antimicrobial treatment of a number of foodstuff in the food industry. The toxic and bactericidal effects of the active (free) chlorine they contain are well understood for the concentrations used in practice, whereas little is known about the effect of its residual (subinhibitory) amounts on the organism and on the microbiota, including the ability to induce antimicrobial resistance. The aim of the study was to investigate the effect of different doses of active chlorine at oral administration on the commensal bacteria of intestinal microbiota, body weight gain and micromorphological features of the liver in rats. Material and methods. The study was carried out on male Wistar rats, with an initial body weight of 90-100 g, which for 4 weeks received doses of active chlorine together with drinking water ad libitum, with the given concentration: subinhibitory (10 mg/L), threshold (50 mg/L) and aggravated (100 mg/L). Chloramine was used as a chlorinecontaining agent stable in aqueous solution. Body weight gain was monitored daily. After decapitation, the caecum was taken for microbiota examination as well as the liver. The phenotypic antimicrobial resistance characteristics of Enterobacteriaceae and Enterococci were studied by the disk diffusion method. Additionally, a micromorphologic study of liver slices was performed. Results. Insignificant negative deviations in the body weight gain of rats in the experimental groups receiving subinhibitory doses of active chlorine, combined with a reduced level of Enterococci and Enterobacteriaceae representatives, were revealed. No significant effect of chlorine on the levels of resistant Escherichia coli populations was found, but a tendency to exhibit antimicrobial resistance of Enterococci isolated from rats receiving low doses of active chlorine (10 and 50 mg/l) was detected. No signs of toxic effect on the liver tissue most sensitive to chlorine were detected, but some accumulations of inflammatory cells in the liver slice were revealed. Conclusion. Low doses of chlorine-containing substances at their oral consumption are not indifferent for rats' organism, causing negative phenomena in intestinal bacteria and in liver tissues at the level of tendency. It is expedient to continue studies in this direction.

Abstract:Sacituzumab govitecan (IMMU-132) is an antibody-drug conjugate (ADC) made from a humanized anti-Trop-2 monoclonal antibody (hRS7) conjugated with the active metabolite of irinotecan, SN-38. In addition to its further characterization, as the clinical utility of IMMU-132 expands to an ever-widening range of Trop-2-expressing solid tumor types, its efficacy in new disease models needs to be explored in a nonclinical setting. Unlike most ADCs that use ultratoxic drugs and stable linkers, IMMU-132 uses a moderately toxic drug with a moderately stable carbonate bond between SN-38 and the linker. Flow cytometry and immunohistochemistry disclosed that Trop-2 is expressed in a wide range of tumor types, including gastric, pancreatic, triple-negative breast (TNBC), colonic, prostate, and lung. While cell-binding experiments reveal no significant differences between IMMU-132 and parental hRS7 antibody, surface plasmon resonance analysis using a Trop-2 CM5 chip shows a significant binding advantage for IMMU-132 over hRS7. The conjugate retained binding to the neonatal receptor, but it lost greater than 60% of the antibody-dependent cell-mediated cytotoxicity activity compared to that of hRS7. Exposure of tumor cells to either free SN-38 or IMMU-132 demonstrated the same signaling pathways, with pJNK1/2 and p21(WAF1/Cip1) upregulation followed by cleavage of caspases 9, 7, and 3, ultimately leading to poly-ADP-ribose polymerase cleavage and double-stranded DNA breaks. Pharmacokinetics of the intact ADC in mice reveals a mean residence time (MRT) of 15.4 h, while the carrier hRS7 antibody cleared at a similar rate as that of the unconjugated antibody (MRT ∼ 300 h). IMMU-132 treatment of mice bearing human gastric cancer xenografts (17.5 mg/kg; twice weekly × 4 weeks) resulted in significant antitumor effects compared to that of mice treated with a nonspecific control. Clinically relevant dosing schemes of IMMU-132 administered either every other week, weekly, or twice weekly in mice bearing human pancreatic or gastric cancer xenografts demonstrate similar, significant antitumor effects in both models. Current Phase I/II clinical trials ( ClinicalTrials.gov , NCT01631552) confirm anticancer activity of IMMU-132 in cancers expressing Trop-2, including gastric and pancreatic cancer patients.
In Vitro Cytotoxicity and Antibacterial Activity of Hypochlorous Acid Antimicrobial Agent

Abstract:Background/purpose: Bacteria -associated oral diseases such as dental caries and periodontitis are widespread epidemics that cause oral pain and loss of function. The purpose of this study was to evaluate the in vitro cytotoxicity and antibacterial activity of different concentrations of hypochlorous acid (HOCl). Materials and methods: Five different concentrations (100, 200, 300, 400, and 500 ppm) of HOCl were evaluated for their antimicrobial efficacy against Gram-negative (A. actinomycetcmcomitans and P. gingivalis) and Gram-positive bacteria (S. mutans and S. sanguinis) after treatment for 1 and 10 min. Sodium hypochlorite (NaOCl) and chlorhexidine (CHX) were used as positive controls. In addition, HOCl was examined for L929 cytotoxicity and RAW 264.7 growth. Results: The bacteriostatic ratio of NaOCl was comparable to that of CHX and significantly (P < 0.05) higher than that of all HOCl solutions. Higher HOCl concentration had significantly (P < 0.05) higher antibacterial effect, and the bacteriostatic ratio of 10 min treatment was slightly higher than that of 1 min treatment. CHX and NaOCl seeded into L929 cells resulted in low cell viability with only 30-39%, much significantly (P < 0.05) lower than all HOCl groups (greater than 80%). All HOCl solutions met the recommendations of ISO 10993-5 and showed no cytotoxicity, although there was a concentration -dependent decrease in cell viability. All antimicrobial agents showed the same trend of response to RAW 264.7 as L929. Conclusion: Within the limit of this study, 400 ppm HOCl disinfectant may be a potential antimicrobial candidate for mouthwash, endodontic irrigants, and periodontitis treatment. IMPACT OF PARTIAL STENOSIS OF TRACHEA ON ANTIOXIDANT SYSTEM IN YOUNG RATS

Abstract:Introduction. Recently, there has been a significant rise in the attention of researchers and clinicians towards the issue of complex hypoxic trauma affecting various organs and body systems. This is particularly important when considering the cells that are most vulnerable to oxygen deprivation, including brain tissue, lungs, certain organs of the gastrointestinal tract, systemic immunity. The significance of this problem becomes even more relevant when examining the impact of hypoxia on the cells of sexually immature mammals and humans, as well as exploring potential treatments for acute hypoxic conditions including laryngomalacia and stridor observed in children under the age of one year. Moreover, there are a number of associated complex and unresolved issues in experimental clinical practice. The aim of the study is to investigate changes in the enzyme activity of the pro-oxidant-antioxidant system of the lung tissue cells and the central nervous system (cerebrocortex) during the experimental modelling of respiratory pathology (by ligating the upper trachea of rats), the features of their recovery and the dynamics of this process after the surgical removal of the tracheal ligature. Materials and methods. The studies were performed on 37 sexually immature male Wistar rats, aged 2528 days. The test animals were divided into four groups: the first group served as the control (intact) rats, the second group consisted of rats with experimentally induced tracheal stenosis, which were removed from the experiment in 7 days after the operation, the third group consisted of rats with experimentally induced tracheal stenosis who were removed from the experiment in 21 days after the operation, and the fourth group consisted of rats whose tracheal ligature was removed on the 7th day after the operation and then they were removed from the experiment on the 21st day. The following biochemical parameters were studied: malondialdehyde content, activity of catalase, glutathione peroxidase, glutathione reductase, and superoxide dismutase. Results. The findings obtained demonstrate an increase in malondialdehyde content and antioxidant enzyme activity in the lung and brain tissues of rats on the 7th and 21st days following partial tracheal stenosis. Restoring airway ventilation did not fully normalize these indicators to the control values. In conclusion, the findings indicate that the impaired pro-oxidant-antioxidant balance in lung and central nervous system cells did not completely restore even two weeks after relieving the compressive effect on the trachea and resolving the hypoxic respiratory system disorders partially. This suggests the necessity to continue therapy with pharmacological antioxidant agents following surgical intervention. However, further research is required to explore this aspect of the problem in depth.

Abstract:Objective: The purpose of this study is to explore the influence of ozone repeated short exposures on airway/lung inflammation, airway hyperresponsiveness (AHR) and airway hypersecretion in ovalbumin (OVA) sensitized/challenged asthmatic mouse model. Methods: OVA sensitization was performing by intraperitoneal injection. Ozone exposures (3ppm for 3hours) were given one hour after aerosolized OVA challenges (once every other day, 4 times totally). Methacholine (MCH) bronchial provocation tests, Liu's staining of BALF cell smears, hematoxylin-eosin (HE) staining and Periodic Acid-Schiff (PAS) staining of lung tissue were performed. Interleukins (ILs; IL-4, IL-13, IL-1 beta, and IL-18) protein (ELISA) and mRNA expression levels (RT-qPCR) in murine lung, 8-hydroxy-2'-deoxyguanosine (8-OHdG, ELISA), malondialdehyde (MDA, thiobarbituric acid assay), reduced glutathione (GSH, spectrophotometric method) in bronchoalveolar lavage fluid (BALF), and GSH1 mRNA relative expression levels (RT-qPCR) in lung tissue were analyzed. Result: Repeated ozone exposures down-regulated the AHR to MCH in mice undergoing OVA sensitization and challenge, however not all parameters associated with asthma were decreased since obvious mucus hypersecretion was induced and airway inflammation increased slightly, especially around small airways. Following ozone co-exposure, the increase of IL-4 and IL-13 levels in murine lung caused by OVA sensitization/challenge were reversed. Instead, levels of IL-1 beta in BALF remained, higher than negative control group. Ozone repeated short exposures also induced significant increase of 8-OHdG in BALF in OVA sensitized and challenged mice. Conclusion: For asthmatic mice undergoing ozone exposures, AHR is not an accurate indicator of the severity of asthma. Repeated short ozone exposures increase mucus hypersecretion, possibly via an increase in oxidative stress and immune dysregulation.
Chlorine disinfection reduces the exposure risks of inhaled reclaimed water

Abstract:The increasing application of reclaimed water in urban areas is a rising health concern. For instance, the inhalation of reclaimed water induces polymorphonuclear cell-dominant inflammation in the lung, and prefibrosis can develop after subchronic exposure. The major toxic source, free endotoxins, could be inhibited by chlorination, yet whether the chlorination can increase or reduce the inhalation toxicity of reclaimed water is still unclear. Here, we treated secondary effluent samples with different doses of chlorine, and then mice were exposed to these samples via inhalation. We studied inflammatory markers such as polymorphonuclear cell, proinflammatory cytokines in lung lavage, induced bronchus-associated lymphoid tissue, and immunohistochemical analysis of fibrotic markers. Results show that endotoxin activities increased slightly from 951 endotoxin units per milliliter to the highest activity of 1133 endotoxin units per milliliter after disinfection. However, pulmonary inflammation was reduced after 5 and 10 mg/L chlorine treatment. The aggregate size of endotoxin displayed a decline after chlorination. Furthermore, the potential cytotoxicity of the disinfection byproducts after chlorination was not observed in the lungs after subchronic exposure. This study demonstrates that sufficient chlorination can prevent acute and subchronic pulmonary damage caused by the reclaimed water. In addition, endotoxin activity should not be used as an index for acute and subchronic toxicity control.
Inhibitory effect of hydrogen sulfide on ozone-induced airway inflammation, oxidative stress, and bronchial hyperresponsiveness.

Abstract:Exposure to ozone has been associated with airway inflammation, oxidative stress, and bronchial hyperresponsiveness. The goal of this study was to examine whether these adverse effects of ozone could be prevented or reversed by hydrogen sulfide (H2S) as a reducing agent. The H2S donor sodium (NaHS) (2 mg/kg) or vehicle (PBS) was intraperitoneally injected into mice 1 hour before and after 3-hour ozone (2.5 ppm) or air exposure, and the mice were studied 24 hours later. Preventive and therapeutic treatment with NaHS reduced the ozone-induced increases in the total cells, including neutrophils and macrophages; this treatment also reduced levels of cytokines, including TN-alpha, chemokine (C-X-C motif) ligand 1, IL-6, and IL-1 beta levels in bronchial alveolar lavage fluid; inhibited bronchial hyperresponsiveness; and attenuated ozone-induced increases in total malondialdehyde in bronchoalveolar lavage fluid and decreases in the ratio of reduced glutathione/oxidized glutathione in the lung. Ozone exposure led to decreases in the H2S production rate and in mRNA and protein levels of cystathionine-beta-synthetase and cystathionine-gamma-lyase in the lung. These effects were prevented and reversed by NaHS treatment. Furthermore, NaHS prevented and reversed the phosphorylation of p38 mitogen-activated protein kinase and heat shock protein 27. H2S may have preventive and therapeutic value in the treatment of airway diseases that have an oxidative stress basis.
Augmenting effect of acetic acid for acidification on bactericidal activity of hypochlorite solution

Abstract:Aims: Bactericidal activity of chlorine solution is enhanced by weak acidification. We compared the effects of various acids on the bactericidal activity of hypochlorite solution to establish a method for safe and effective use of an acidic hypochlorite solution. Methods and results: The bactericidal activities of acidic hypochlorite solutions that had been adjusted to pH 5.0 with hydrochloric acid, acetic acid, citric acid, lactic acid, formic acid, phosphoric acid or sulphuric acid against Bacillus subtilis spores were compared. The acidic solutions prepared with hydrochloric acid and acetic acid showed the highest bactericidal activity, and all of the spores (5 x 106 cfu ml(-1)) were killed within 10 min. On the other hand, the solutions prepared with citric acid and lactic acid showed no bactericidal activity against any bacterial strains tested in this study despite the low pH. The amount of chlorine gas produced by the preparation using acetic acid was sixfold less than that produced from the preparation using hydrochloric acid. Conclusions: Acetic acid is the most suitable and safe acid for the preparation of an acidic hypochlorite solution. Significance and impact of the study: The results of this study provide useful information for establishing a method for safe and effective use of an acidic hypochlorite solution.
Uses of gaseous hypochlorous acid for controlling microorganisms in indoor spaces

Abstract:Hypochlorous acid (HOCl) is an active species in the chlorination process. Hypochlorite salts that release hypochlorite ion (OCl-) have been used for more than 200 years as disinfecting, cleaning, deodorizing, and decolorizing agents in various technological fields. In the food industry, sodium hypochlorite is the most widely used among chlorine compounds. The antimicrobial activity of a dilute hypochlorite solution is attributed largely to HOCl because of its cell membrane permeability. OCl- exhibits an excellent cleaning action for organic soils on solid surfaces. HOCl has been used as an aqueous solution, and its objects to be treated are things. In hypochlorite solution, HOCl is volatile and easily volatilized by stirring, bubbling, atomizing, or forced-air vaporization. On the other hand, OCl- is non-volatile and stays in the solution. Recently, the scope of objects to be treated with hypochlorite solution has been expanded to indoor spaces, and the use of gaseous hypochlorous acid (HOCl（g) ) has been studied intensively. This review describes the mechanisms of actions of hypochlorous acid as liquid-based and gaseous disinfectants and provides the evidence for the safety and effectiveness of HOCl（g) for controlling microorganisms in indoor spaces.
[Effects of Hyperoxia on Inflammatory Response in Lung of Infantile Rats].

Abstract:OBJECTIVE:To investigate the effects of hyperoxia on inflammatory response in lung of infantile rats.METHODS:Forty 21-day-old male Sprague-Dawley (SD) rats were randomly divided into five groups: room-air control group and 12, 24, 48, 72 hours hyperoxia groups, with the rats continuously exposed to room-air and oxygen (92%-94%) respectively. The rats were sacrificed by depletion method, and lung tissue was obtained for bronchoalveolar lavage. The contents of malondialdehyde (MDA) and the activities of myeloperoxidase (MPO) in lung tissue were assayed by thiobarbituric acid or chromometry, and the concentrations of tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6) and IL-10 in bronchoalveolar lavage fluid (BALF) were measured by enzyme linked immunosorbent assay (ELISA). The lung pathology was examined, and lung injury score was assessed.RESULTS:Compared with the room-air control group, the contents of MDA [(2.24+/-0.43) mmol/g vs. (1.57+/-0.31) mmol/g] and the activities of MPO [(1.24+/-0.25) U/g vs. (0.69+/-0.22) U/g] from lung tissue were elevated at 12 hours and 24 hours of hyperoxia exposure, respectively, and they further increased with prolongation of hyperoxia exposure (P<0.05 or P<0.01). The values of TNF-alpha [(135.2+/-44.0) ng/L vs. (94.5+/-22.3) ng/L], IL-6 [(73.1+/-14.2) ng/L vs. (55.7+/-17.3) ng/L] and IL-10 [(67.9+/-21.7) ng/L vs. (48.2+/-7.6) ng/L] in BALF were all higher at 24 hours of hyperoxia exposure than those of the room-air control group (P<0.05 or P<0.01), but decreased at 48 hours of hyperoxia exposure compared with those of 24-hour hyperoxia exposure group [TNF-alpha: (105.4+/-17.0) ng/L, IL-6: (54.3+/-17.4) ng/L, IL-10: (50.9+/-6.9) ng/L, all P<0.05]. Lung injury scores were higher at 12 hours of hyperoxia exposure as compared with those of the room-air control group (4.5+/-1.4 vs. 1.3+/-0.5), and it further increased with prolongation of hyperoxia exposure (P<0.05 or P<0.01).CONCLUSION:Hyperoxia can lead to lung inflammatory injury in infantile rats, and the expressions of TNF-alpha, IL-6 and IL-10 in BALF may reach the peak at 24 hours of hyperoxia exposure.

Abstract:Airway inflammatory diseases, such as asthma, are a global public health concern owing to their chronic inflammatory effects on the respiratory mucosa. Molecular hydrogen (H2) has recently been recognized for its antioxidant and anti-inflammatory properties. In this study, we examined the therapeutic potential of H2 in airway inflammation using an ovalbumin (OVA)-induced BALB/c mouse model of allergic asthma. Female BALB/c mice were sensitized and challenged with OVA to induce airway inflammation, and 30 mice were randomly divided into five groups: NT (non-treatment), HTC (3% H2 treatment only), NC (negative control, OVA only), PC (positive control, OVA + intranasal 1 mg/mL salbutamol 50 μL), and HT (H2 treatment, OVA + inhaled 3% H2). Various inflammatory and oxidative stress (OS)-induced markers such as white blood cells (WBCs) and their differential counts, lung histology, cytokine levels such as interleukin (IL)-4, (IL)-5, (IL)-13, interferon-gamma (IFN-γ), tumor necrosis factor-alpha (TNF-α), granulocyte-macrophage colony-stimulating factor (GM-CSF), (IL)-10, reactive oxygen species (ROS), nitric oxide (NO), glutathione peroxidase (GPx), and catalase (CAT), and total immunoglobulin E (IgE) levels were investigated. Our results showed that inhaled H2 significantly reduced inflammatory cell infiltration, OS markers, and pro-inflammatory cytokine expression while upregulating antioxidant enzyme activity. Furthermore, H2 also significantly decreased serum IgE levels, a marker of allergic inflammation. Collectively, our findings suggest that H2 inhalation is a promising treatment option for airway inflammation, offering a novel approach with potential clinical applications.

Abstract:Chlorine-containing disinfectants are widely used in hospitals to prevent hospital-acquired severe acute respiratory syndrome coronavirus 2 infection. Meanwhile, ventilation is a simple but effective means to maintain clean air. It is essential to explore the exposure level and health effects of coronavirus disease 2019 patients' inhalation exposure to by-products of chloride-containing disinfectants under frequent surface disinfection and understand the role of ventilation in mitigating subsequent airway damage. We determined ventilation dilution performance and indoor air quality of two intensive care unit wards of the largest temporary hospital constructed in China, Leishenshan Hospital. The chloride inhalation exposure levels, and health risks indicated by interleukin-6 and D-dimer test results of 32 patients were analysed. The mean ± standard deviation values of the outdoor air change rate in the two intensive care unit wards were 8.8 ± 1.5 h −1 (Intensive care unit 1) and 4.1 ± 1.4 h −1 (Intensive care unit 2). The median carbon dioxide and fine particulate matter concentrations were 480 ppm and 19 μg/m 3 for intensive care unit 1, and 567 ppm and 21 μg/m 3 for intensive care unit 2, all of which were around the average levels of those in permanent hospitals (579 ppm and 21 μg/m 3 ). Of these patients, the median (lower quartile, upper quartile) chloride exposure time and calculated dose were 26.66 (2.89, 57.21) h and 0.357 (0.008, 1.317) mg, respectively. A statistically significant positive correlation was observed between interleukin-6 and D-dimer concentrations. To conclude, ventilation helped maintain ward air cleanliness and health risks were not observed.

Abstract:Background: The development of the global economy has led to changes in air pollution patterns. The haze phenomenon characterized by high concentrations of particulate matter 2.5 (PM2.5) has changed to complex pollution, and photochemical pollution characterized by ozone (O3) has become increasingly prominent. Ozone pollution and its impact on human health has become an important topic that needs to be studied urgently. Objective: To investigate the effects of ozone on oxidative stress and inflammation in the nasal mucosa of a rat model. Methods: Thirty-two healthy female Sprague–Dawley rats, eight in each group, were divided into four groups using the randomized numeric table method: normal control group (NC group), normal rats with a low level of ozone inhalation exposure (NEL group, 0.5 ppm), medium ozone inhalation exposure (NEM group, 1 ppm), and high ozone inhalation exposure (NEH group, 2 ppm). The ozone inhalation exposure groups were placed in the ozone inhalation exposure system and exposed to different concentrations of ozone for 2 h each day for 6 weeks. Nasal secretion was measured, and nasal lavage and nasal mucosa were collected. Malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) activities were measured by colorimetric assay, and the nasal mucosa was analyzed by Western blot. Western blot (WB) was used to detect the expression of NF-κB p65 nuclear protein in nasal mucosa. The mRNA expression of NF-κB target genes IL-6 and IL-8 and tumor necrosis factor-α (TNF-α) was detected by real-time quantitative PCR (qRT-PCR), and the protein content of pro-inflammatory factors IL-6, IL-8, and TNF-α was detected by ELISA in serum and nasal lavage fluid. The nasal mucosa of rats was stained with hematoxylin-eosin (HE) to observe the pathological changes in the nasal mucosa. The data were analyzed by SPSS 20.0 software. Results: The amount of nasal secretion increased significantly in all groups after ozone exposure compared with that in the NC group. The MDA content of the nasal mucosa was significantly increased in the ozone-exposed group compared with the NC group, and the activity levels of SOD and GSH-Px in the nasal mucosa were lower in the ozone-exposed group than in the NC group. The mRNA expression of IL-6, IL-8, and TNF-α in the nasal mucosa of the ozone-exposed group was elevated, and the protein content of TNF-α, IL-6, and IL-8 in the nasal lavage fluid was elevated, and the content increased with the increase in ozone concentration. The expression of NF-κB p65 intracellular protein in the nasal mucosa of each ozone-exposed group was higher than that of the normal group, and the content increased with the increase in ozone concentration. Conclusions: Ozone inhalation exposure promotes oxidative stress and the release of inflammatory factors TNF-α, IL-6, and IL-8, leading to pathological damage of the nasal mucosa, the degree of which increases with increasing concentration. This pathological process may be related to the activation of the transcription factor NF-κB by ozone in the nasal mucosa of rats, which increases the expression of its target genes.

Abstract:Hydrogen has been shown to exhibit selective antioxidant properties against hydroxyl radicals, and exerts antioxidant and anti-inflammatory effects. The monocrotaline-induced model of pulmonary hypertension is suitable for studying substances with antioxidant activity because oxidative stress is induced by monocrotaline. On day 1, male Wistar rats were subcutaneously injected with a water–alcohol solution of monocrotaline or a control with an only water–alcohol solution. One group of monocrotaline-injected animals was placed in a plastic box that was constantly ventilated with atmospheric air containing 4% of molecular hydrogen, and the two groups of rats, injected with monocrotaline or vehicle, were placed in boxes ventilated with atmospheric air. After 21 days, hemodynamic parameters were measured under urethane narcosis. The results showed that, although hydrogen inhalation had no effect on the main markers of pulmonary hypertension induced by monocrotaline injection, there was a reduction in systemic blood pressure due to its systolic component, and a decrease in TGF-β expression, as well as a reduction in tryptase-containing mast cells.
