Abstract:Occupational exposure to pesticides is a known risk for disrupting cellular immune response in flower workers due to their use of multiple chemical products, poor work conditions, and inadequate protection. Recently, the analysis of pesticide use patterns has emerged as an alternative to studying exposure to mixtures of these products. This study aimed to evaluate the association between exposure to different patterns of pesticide use and the cytokine profile of flower workers in the State of Mexico and Morelos, Mexico.

Exposure of Young Children Working on Mexican Tobacco Plantations to Organophosphorous and Carbamic Pesticides, Indicated by Cholinesterase Depression

Abstract:Background Organophosphorous (OP) and carbamic pesticides are used in large quantities on tobacco plantations in Nayarit State, Mexico, where up to 3000 children and their families work. OP and carbamic pesticides are easily inhaled or absorbed through the skin and children may be particularly vulnerable to pesticides because of their smaller body mass, their height leaves.and more regular hand- mouth contact. The aim of this study is to assess the effect of pesticide exposure on acetylcholinesterase levels of very young migrant Mexican tobacco workers and younger siblings.Methods Blood samples were collected from 160 children aged 0-14 years during harvest (exposure) and from 62 children in their home communities 6-9 months after harvest (baseline). Samples were tested for cholinesterase corrected for haemoglobin and ambient temperature.Results Fifteen per cent of children had depression scores ranging from -40% to 190% of their baseline levels. Thirty-three per cent of children had depression scores of at least 15% and 86% of children were anaemic.Conclusion These results suggest that very young children are exposed to potentially harmful and toxic amounts of pesticides while working. Further research is needed to assess the actual acute and chronic health impacts of such exposures.
Association of pesticide exposure with respiratory health outcomes and rhinitis in avocado farmworkers from Michoacán, Mexico

Abstract:Background A growing literature suggests associations between occupational pesticide exposure and respiratory health. In this study, we aimed to examine the association of exposure to insecticides, fungicides, and herbicides, individually and as a mixture, with respiratory health outcomes and rhinitis in avocado farmworkers from Michoacán, Mexico. Material and methods We conducted a cross-sectional study of 105 avocado farmworkers between May and August 2021. We quantified 12 insecticide, fungicide, and herbicide metabolites in urine samples collected during two study visits (8–10 weeks apart). We collected survey data on self-reported pesticide use during the 12 months prior to the baseline survey and estimated annual exposure-intensity scores (EIS) using a semi-quantitative exposure algorithm. We also assessed respiratory symptoms, including wheezing, chest tightness, wheezing after exercise, and night cough. We used generalized linear regression models to examine associations of individual urinary metabolite concentrations and annual EIS with respiratory health outcomes and rhinitis. Mixture effects were assessed using Bayesian Weighted Quantile Sum (BWQS) regression. Results After adjusting for multiple comparisons, we observed mostly null associations of individual pesticide metabolite concentrations and annual EIS with the outcomes of interest. However, in BWQS analyses, we found evidence of a mixture association of urinary pesticide metabolites with increased odds of night cough (OR: 5.34, 95 % CrI: 1.67, 20.62). Pyrethroid metabolites 3-phenoxybenzoic acid and cis- and trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylic acid were the main contributors to this association (43 %). Conclusions Our findings indicate that exposure to a mixture of pesticides, particularly pyrethroid insecticides, may be associated with night cough in avocado farmworkers.

Abstract:In occupational settings workers are often exposed to pesticides at relatively high doses compared to environmental exposures. Long-term exposure to pesticides has been associated with numerous adverse health effects in epidemiological studies, and oxidative stress is often claimed as one of the underlying mechanisms. In fact, different pesticides have been reported to induce oxidative stress due to the generation of free radicals and/or alteration in antioxidant defense enzymes. The present study examined greenhouse workers regularly exposed to diverse pesticides under integrated production system, and a group of controls of the same geographic area without any chemical exposure. Two different periods of the same crop season were assessed, one of high exposure (with greater use of pesticides) and other of low exposure (in which a less use of these compounds was made). Non-specific biomarkers of oxidative stress, e.g. thiobarbituric acid reactive substances (TBARS), ferric reducing ability of serum (FRAS), total thiol groups (SHT), gamma-glutamyl transpeptidase (GGT) and paraoxonase-1 (PON1) were measured in serum samples from all study subjects, alongside erythrocyte acetylcholinesterase (AChE). Results are suggestive of a mild increase in oxidative stress associated with pesticide exposure, which was compensated by an adaptive response to raise the antioxidant defenses and thus counter the detrimental effects of sustained oxidative stress. This response led to significantly increased levels of FRAS, SHT and PON1 in greenhouse workers relative to controls. Furthermore, AChE was decreased likely as a result of oxidative stress as workers did not use organophosphate insecticides.

Abstract:Abstract The use of pesticides has increased during the past decades, also increasing the risk of exposure to toxic pesticides that can cause detrimental health effects in the future. This is of special concern among farmers in low-to-middle-income countries that may lack proper training in the safe use of these chemicals. To assess the situation in Bolivia a cross-sectional study in three agricultural communities was performed ( n = 297). Handling, use of personal protective equipment (PPE) and pesticide exposure were assessed by a questionnaire and measurements of urinary pesticide metabolites (UPMs). Results showed that methamidophos (65%) and paraquat (52%) were the most commonly used pesticides and that 75% of the farmers combined several pesticides while spraying. Notably, only 17% of the farmers used recommended PPEs while 84% reported to have experienced symptoms of acute pesticide poisoning after spraying. UPM measurements indicated high levels of exposure to chlorpyrifos, pyrethroids and 2,4D and that men generally were more highly exposed compared to women. Our study demonstrates that farmers who are better at following recommendations for pesticide handling and use of PPE had a significantly lower risk of having high UPM levels of most measured pesticides. Our results thus confirm the need of proper training of farmers in low-to-middle-income countries in proper protection and pesticide handling in order to reduce exposure levels and health problems.

Abstract:Organophosphate pesticides (OPs) have greatly facilitated food production worldwide, and their use is not limited to agriculture and the control of pests and disease vectors. However, these substances can directly affect the immune response of non-target organisms. In this sense, exposure to OPs can have negative effects on innate and adaptive immunity, promoting deregulation in humoral and cellular processes such as phagocytosis, cytokine expression, antibody production, cell proliferation, and differentiation, which are crucial mechanisms for host defense against external agents. This review focuses on the scientific evidence of exposure to OPs and their toxic effects on the immune system of non-target organisms (invertebrates and vertebrates) from a descriptive perspective of the immuno-toxic mechanisms associated with susceptibility to the development of bacterial, viral, and fungal infectious diseases. During the exhaustive review, we found that there is an important gap in the study of non-target organisms, examples of which are echinoderms and chondrichthyans. It is therefore important to increase the number of studies on other species directly or indirectly affected by Ops, to assess the degree of impact at the individual level and how this affects higher levels, such as populations and ecosystems.

Abstract:AIMS: To identify neurobehavioural deficits among workers exposed to organophosphorous (OP) pesticides in their occupation.METHODS: This study was conducted during the period when pesticides were applied to cotton crops in the fields in Menoufiya Governorate, Egypt. Fifty two occupationally exposed male workers were compared to 50 unexposed male controls who were similar in age, socioeconomic class, and years of education (> or =12 years). All participants completed a questionnaire (assessing personal, occupational, and medical histories), general and neurological clinical examination, neurobehavioural test battery (including tests for verbal abstraction, problem solving, attention, memory, and visuomotor speed), personality assessment, and serological analysis for serum acetylcholinesterase.RESULTS: After correcting for confounders of age and education, the exposed participants exhibited significantly lower performance than controls on six neurobehavioural tests (Similarities, Digit Symbol, Trailmaking part A and B, Letter Cancellation, Digit Span, and Benton Visual Retention). A longer duration of work with pesticides was associated with lower performance on most neurobehavioural tests after adjusting for multiple comparisons. Although serum acetylcholinesterase was significantly lower in the exposed than the control participants, it was not significantly correlated with either neurobehavioural performance or neurological abnormalities.CONCLUSIONS: Occupational exposure to OP pesticides was associated with deficits in a wider array of neurobehavioural functions than previously reported, perhaps because of higher exposure in this population. Moderate chronic OP exposure may not only affect visuomotor speed as reported previously, but also verbal abstraction, attention, and memory.

Auxotrophy for uridine increases the sensitivity of Aspergillus niger to weak-acid preservatives.

Abstract:Weak-acid preservatives such as sorbic acid are added to foods to prevent fungal spoilage. The modes of action of weak-acid preservatives are only partially understood and, in this paper, further insight is presented into the mechanisms by which weak acids inhibit the growth of fungi. Uridine-requiring strains of Aspergillus niger were shown to be more sensitive to weak acids (including sorbic, acetic and benzoic acids) than wild-type (WT) strains. In contrast, sensitivity to other, non-acidic, antifungal substances was similar in mutant and WT strains. By complementing a pyrG(-) strain of A. niger with an intact pyrG gene, WT-like resistance to weak-acid preservatives was restored. Using (14)C-labelled uridine, sorbic acid was shown to completely inhibit uridine uptake in germinating conidia in a non-competitive manner. It is therefore proposed that the additional weak-acid sensitivity of the pyrG(-) strains was caused by weak-acid inhibition of uridine uptake. Several other auxotrophic strains of A. niger were screened for sensitivity to acetic, sorbic and decanoic acids. Strains auxotrophic for either adenine or uridine were found to have enhanced sensitivity but, in contrast, amino acid auxotrophs showed resistance comparable to that of the WT. Uridine auxotrophs of Saccharomyces cerevisiae were not more sensitive to weak acids compared to WT strains. In conclusion, this study describes a previously unknown mechanism of action of weak acids against the filamentous fungus A. niger, which may fundamentally affect our understanding of the preservation of food against spoilage fungi.
Associations between Urinary Pesticide Metabolites and Serum Inflammatory Biomarkers in Adolescents Living in an Agricultural Region

Abstract:Background In-vitro and in-vivo studies have shown evidence for the immuno-modulatory properties of different pesticides. However, few epidemiological studies on inflammation and pesticide exposure exist, with none in children and adolescents. Associations between pesticide metabolites in urine and inflammatory biomarkers in serum were evaluated among children and adolescents (n=512) of rural Ecuador as part of the ESPINA study Methods Seventeen urinary biomarkers of insecticide, herbicide and N, N-diethyl-meta-toluamide (DEET) insect repellent exposure were measured. Among them, acetamiprid-N-desmethyl [AND], 3,5,6-Trichloro-2-pyridinol [TCPy], para-nitrophenol [PNP], malathion dicarboxylic acid [MDA], 3-phenoxybenzoic acid [3-PBA], 2,4-Dichlorophenoxyacetic acid [2,4-D], glyphosate and 3-(diethylcarbamoyl) benzoic acid [DCBA] were included in analyses as they were detected in >30% of participants. Serum analysis included c-reactive protein (CRP), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), soluble intercellular adhesion molecule-1 (sICAM-1), soluble vascular cell adhesion molecule-1 (sVCAM-1), serum amyloid A (SAA), and soluble CD14 (sCD14). Associations were evaluated by generalized estimating equations (GEE) and partial least squares (PLS) regression, adjusting for demographic, anthropometric and socioeconomic variables. Results Positive quadratic associations were found between 2,4-D and CRP (β2=0.13, [0.00, 0.27]), IL-6 (β2=0.10, [0.04, 0.15]), SAA (β2=0.13, [0.01, 0.30]), sICAM-1 (β2=53.25, [27.26, 79.24]) and sVCAM-1 (β2=61.11, [30.52, 91.70]). The pesticide metabolites PLS composite variable was positively associated with IL-6 (β=0.09, [0.01. 0.17]), SAA (β=0.43, [0.13, 0.17]), sICAM-1 (β=63.52, [9.92, 117.13]), sVCAM-1 (β=69.03, [8.29, 129.76]) and TNF-α (β=0.08, [0.00, 0.16]), and negatively associated with CRP (β=-0.28, [-0.49, -0.08], Figure 1). Conclusions Our findings demonstrate a novel pesticide/herbicide-inflammation link in adolescents, which may be an underlying mechanism of the health impacts of pesticides/herbicides.

Here, we investigated the impact of occupational pesticide exposure on systemic cortisol levels in female rural workers diagnosed with BC. Occupational exposure was assessed by interviews with a standardized questionnaire. Plasma samples (112 from pesticide-exposed women and 77 from unexposed women) were collected in the afternoon, outside the physiological cortisol peak, and analyzed by a chemiluminescent paramagnetic immunoassay for the quantitative determination of cortisol levels in serum and plasma. The results from both groups were categorized according to patients' clinicopathological and exposure data. BC pesticide-exposed women presented higher levels of cortisol than the unexposed. Higher cortisol levels were also detected in the exposed group with more aggressive disease (triple-negative BC), with tumors over 2 cm, with lymph node metastases, and with high risk of disease recurrence and death. These findings demonstrated that there is an association between pesticide exposure and BC that affected cortisol levels and correlated to poor disease prognosis.

Determinants of organophosphorus pesticide urinary metabolite levels in pregnant women from the CHAMACOS cohort

Abstract:Organophosphosphorus pesticides (OPs) are widely used as insecticides in agriculture. Human exposure to OPs has been linked to adverse effects including poorer child neurodevelopment, reduced birth weight, altered serum hormone levels, and reduced semen quality. We measured six OP dialkyl phosphate (DAP) metabolites [three dimethyl alkylphosphates (DMs) and three diethyl alkylphosphates (DEs)] in urine samples collected two times during pregnancy (~13 and ~26 weeks gestation) from 594 women participating in the CHAMACOS birth cohort study and resided in an agricultural community in the United States (U.S.) between 1999 and 2000. Previous studies have shown these women have higher OP exposures compared with the general U.S. population. We examined bivariate associations between prenatal DAP metabolite levels and exposure determinants such as age, season, years living in the US, housing characteristics, fruit and vegetable consumption, occupation and residential proximity to agricultural fields. Final multivariable models indicated that season of urine collection was significantly associated (p < 0.01) with specific gravity-adjusted DM, DE and total DAP metabolites; samples collected in fall and winter had higher concentrations than those collected in spring-summer. Specific gravity-adjusted levels of DM and total DAP metabolites were significantly higher in women who had resided in the U.S. for 5 years or less (p < 0.05). Levels of DM metabolites also increased with daily fruit and vegetable servings (p < 0.01), and levels of DE metabolites were higher in residences with poorer housekeeping quality (p < 0.01) and in mothers that worked in agriculture (p < 0.05). These findings suggest that there are multiple determinants of OP exposure in pregnant women.
Urinary Dialkylphosphate Metabolites Concentrations Of Organophosphorous Pesticides Among Occupationally Exposed Workers And General Population In Shanghai Of China

Abstract:The determination of organophosphorus (OP) pesticide metabolites in human biological fluids represents an important biomarker of exposure in the general population and exposed workers. We measured the urinary excretion of organophosphorus pesticide metabolites to evaluate occupational and non-occupational exposure to pesticides in Shanghai (Eastern China) area. The urine samples were collected from 30 exposed packing workers(20 males, 10 females, mean age 40 years) in dimethoate workshop and 60 healthy adults (37 males, 23 females, mean age 23 years) without any report of occupational exposure. Six alkylphosphates, specific metabolites of organophosphorus pesticides, dimethylphosphate (DMP), dimethylthiophosphate (DMTP), dimethyldithiophosphate (DMDTP), diethylphosphate (DEP), diethylthiophosphate (DEDP) and diethyldithiophosphate (DEDTP) were measured. Dimethylphosphates were found in detectable concentrations in the majority of samples (100.0%, 100.0% and above 90.0% for DMP, DMTP and DMDTP among exposed group, 85.0%, 48.0% and 52.0% for DMP, DMTP and DMDTP among control group, respectively). Urinary diethylphosphates (DEP, DETP) concentrations were above the detection limit in 40.0%, 20.0% of samples for exposed group and 30.0%, 18.0% of samples for control group, respectively. DEDTP was detected below the limit of detection except post-shift exposed worker with the detection frequency of 6.7%. Excretion levels were the highest for DMTP in all subjects. In a word, a very high percentage of urine samples positive for several pesticide metabolites emerged, with higher levels of urinary excretion in general population and exposed workers in Chinese subjects.
Organophosphate pesticide exposure among farm women and children: Status of micronutrients, acetylcholinesterase activity, and oxidative stress

Abstract:Nutritional status plays a major role in determining the possible adverse health outcomes due to pesticide toxicity. The objective of the present study was to assess the organophosphorus pesticide residue levels among farm women (FW) (24-45years) and farm children (FC) (9-12 and 13-15years) belonging to the Rangareddy district (Telangana, India) along with their micronutrient status, inhibition of acetylcholinesterase activity, and oxidative stress levels. Residues of Chlorpyrifos, Diazinon, Malathion, Monocrotophos and Phosalone were found in the serum samples of FW and FC along with significantly low levels of vitamins and minerals. Inhibition of AChE activity was observed in FW and FC and altered oxidative stress parameters among FW. Correlation studies have found significant associations between the pesticide residues, micronutrients and antioxidant enzymes. The study suggests an association between pesticide exposure coupled with micronutrient deficiency, induced AChE inhibition, and oxidative stress.

Abstract:The aim of this study was to evaluate the genotoxic effect of pesticides in exfoliated buccal cells of workers occupationally exposed in Guerrero, Mexico, using the comet assay and the micronucleus test. The study compared 111 agricultural workers in three rural communities (Arcelia 62, Ajuchitlan 13, and Tlapehuala 36), with 60 non-exposed individuals. All the participants were males. The presence of DNA damage was investigated in the exfoliated buccal cells of study participants with the comet assay and the micronucleus (MN) test; comet tail length was evaluated in 100 nuclei and 3000 epithelial cells of each individual, respectively; other nuclear anomalies such as nuclear buds, karyolysis, karyorrhexis, and binucleate cells were also evaluated. Study results revealed that the tail migration of DNA and the frequency of MN increased significantly in the exposed group, which also showed nuclear anomalies associated with cytotoxic or genotoxic effect. No positive correlation was noted between exposure time and tail length and micronuclei frequencies. No significant effect on genetic damage was observed as a result of age, smoking, and alcohol consumption. The MN and comet assay in exfoliated buccal cells are useful and minimally invasive methods for monitoring genetic damage in individuals exposed to pesticides. This study provided valuable data for establishing the possible risk to human health associated with pesticide exposure.

Abstract:In this study, we aimed to determine whether TLR‐9 T‐1486C SNP was associated with susceptibility to OA in the Turkish population. The study group comprised 272 patients with Grade 2‐3‐4 knee OA according to the Kellgren‐Lawrence scoring system and the control group was formed of 296 individuals with Grade 0–1. The TLR‐9 genotype were assessed by real‐time polymerase chain reaction. An analysis of TLR‐9 promoter −1486T/C polymorphism revealed that the −1486CC genotype appeared to have a higher risk for OA and −1486TT and −1486CT genotypes have a protective effect against the development of OA (crude OR = 0.473, 95% CI = 0.297–0.754, p = 0.002, adjusted OR = 0.531, 95% CI = 0.326–0.864, p = 0.011). This study indicate that there is a correlation of TLR‐9 T‐1486C gene polymorphism with advanced knee OA in a Turkish population. Changed in TLR expression due to different allelles may cause osteoarthrith development outcome cartilage degeneration. © 2017 Orthopaedic Research Society. Published by Wiley Periodicals, Inc. Impact of chlorpyrifos exposure on lung function in Egyptian adolescent agriculture workers

Abstract:Chlorpyrifos (CPF) is a widely used organophosphate insecticide that has been linked to detrimental health effects that range from neurological impacts to respiratory disease. The objective of this study was to assess respiratory symptoms associated with CPF exposure throughout the application season. Urine samples were collected from Egyptian adolescent applicators ( n = 206) and non-applicators ( n = 72) to assess 3,5,6-trichloro-2-pyridinol (TCPy), a biomarker for CPF exposure, along with spirometry measures to determine lung ventilatory function. Samples were collected over 7 months in 2016. Logistic regression was used to model the odds of reporting wheeze symptoms based on urinary TCPy concentrations while controlling for age and smoking in the household. Ordinal multinomial logistic regression was used to model the percent reference for forced expiratory volume in one second (rFEV 1 ) based on urinary TCPy concentration (μg/g creatinine). Wheezing increased with increasing pesticide exposure (OR = 1.74 (1.32 − 2.31)). There was no statistically significant relationship between rFEV 1 and TCPy concentration. Efforts to reduce pesticide exposure should be implemented to prevent the potential onset or exacerbation of any linked respiratory complications in adolescents.

Urinary biomarkers of exposure to organophosphate, pyrethroid, neonicotinoid insecticides and oxidative stress: A repeated measurement analysis among pregnant women

Abstract:Exposure to insecticides may be associated with increased oxidative stress (OS), but few studies have assessed the associations of OS biomarkers (OSBs) with exposure to multiple insecticides and their mixture, especially in pregnant women who are a vulnerable population. In the present study, 1,094 Chinese pregnant women were recruited and a total of 3,282 urine samples were collected at their three trimesters to measure eight metabolites of organophosphates, three metabolites of pyrethroids , nine typical neonicotinoids/their metabolites, and three OSBs of DNA damage (8-OHdG), RNA damage (8-OHG), and lipid peroxidation (HNE-MA). Among the twenty target insecticide metabolites, sixteen of them were frequently detected; thirteen of them were detected in over 86% of all the urine samples except for imidacloprid (IMI, detection frequency: 72.9%), desnitro-imidacloprid (DN-IMI, 70.0%), and clothianidin (CLO, 79.6%). The reproducibility of their concentrations across the three trimesters was poor to fair (intraclass correlation coefficients <0.50). Multiparity and warm season were related to higher urinary levels of some insecticide metabolites, while higher education level and inadequate weight gain during pregnancy were significantly associated with lower concentrations of certain insecticide metabolites. Linear mixed model analyses suggested that almost all the frequently detected insecticide metabolites [other than 3-phenoxybenzoic acid (3-PBA)] were significantly associated with elevated levels of the three OSBs (8-OHdG, 8-OHG, and HNE-MA), where the percent change (Δ%) ranged 8.10–36.0% for 8-OHdG, 8.49–34.7% for 8-OHG, and 5.92–182% for HNE-MA, respectively, with each interquartile ratio (IQR)-fold increase in the concentrations of the individual exposure biomarkers. Weighted quantile sum models demonstrated that the insecticide metabolite mixture was positively associated with the three OSBs. Overall, urinary desmethyl-clothianidin (DM-CLO) and 3,5,6-trichloro-2-pyridinol (TCPy) were the top insecticide exposure biomarkers contributing to the association with 8-OHdG and 8-OHG levels, while PNP contributed the most to the association with HNE-MA levels. These findings suggested that gestational exposure to organophosphates, pyrethroids, neonicotinoids, their transformation products, and their mixture may increase oxidative damage to lipids, RNA, and DNA during pregnancy.

Abstract:BACKGROUND: Organophosphorus pesticide (OP) exposure is known to have adverse effects on the nervous system. Children from agricultural communities are at risk of exposure to these chemicals from their indoor environments that can lead to neurological and developmental problems, including changes in behavior.OBJECTIVE: The aim of this study is to evaluate whether the take-home pathway exposure is associated with behavioral and emotional problems in Latino Orchid Community children.METHOD: The study was implemented over a period of two years (2008-2010) in an orchard farming community with a total of 324 parents who had children between the ages of 5-12 years old. Mothers of the children were asked to complete the Child Behavior Checklist (CBCL) and dust from their carpets was collected. Emotional and behavioral deficits were assessed based on the CBCL and house dust was assessed for OP concentrations. In this study, correlations between OPs in house dust and CBCL subscales were estimated using linear regression models with total OP concentrations classified by tertiles. This study also facilitated the comparison between the agricultural and non-agricultural families in terms of behavioral deficits and house dust concentrations of pesticides.RESULTS: The data from the study shows that there was a positive association between the concentration of OP residues in house dust and internalizing behavior (β=2.06, p=0.05) whereas the association with externalizing behavior was not significant after accounting for sociocultural covariates. Significant positive associations of OP residues with somatic problems (p=0.02) and thought problems (p=0.05) were also found.CONCLUSION: The data support a potential role of OP exposure in childhood development, with a specific focus on internalizing behavior. Future work focused on longitudinal studies may uncover the long-term consequences of OP exposure and behavior.
