Abstract:The hydroxyl radical (. OH), one of the most reactive and deleterious reactive oxygen species (ROS), has been suggested to play an essential role in many physiological and pathological scenarios. However, a reliable and robust method to detect endogenous . OH is currently lacking owing to its extremely high reactivity and short lifetime. Herein we report a fluorescent probe HKOH-1 with superior in vitro selectivity and sensitivity towards . OH. With this probe, we have calibrated and quantified the scavenging capacities of a wide range of reported . OH scavengers. Furthermore, HKOH-1r, which was designed for better cellular uptake and retention, has performed robustly in detection of endogenous . OH generation by both confocal imaging and flow cytometry. Furthermore, this probe has been applied to monitor . OH generation in HeLa cells in response to UV light irradiation. Therefore, HKOH-1 could be used for elucidating . OH related biological functions.
Responsive Nanoprobe for Ratiometric Florescence Detection of Hydroxyl Radical in Macrophage Polarization

Abstract:Quantification of hydroxyl radical (•OH), one form of reactive oxygen species (ROS), plays critical roles in early diagnosis and treatment monitoring of various diseases. In this work, we report the development of a responsive nanoprobe for ratiometric fluorescence detection and imaging of •OH in macrophage polarization. The nanoprobe, BSA-CCA@LDH-SRB, was designed and prepared using coumarin 3-carboxylic acid (CCA) as the sensing unit for •OH, and sulforhodamine B (SRB) loaded on layered double hydroxide (LDH) served as the fluorescent reference component. The coupling of CCA to bovine serum albumin (BSA) and the loading of BSA-CCA on the surface of LDH enabled the nanoprobe for fluorescence detection of •OH with high sensitivity and minimal interference from other biomolecules, ions, and ROS. The emission of the prepared BSA-CCA@LDH-SRB at 444 nm emerged and the intensity was increased according to the concentration of •OH, while the emission at 580 nm was maintained, allowing the nanoprobe for ratiometric fluorescence (F444/580) detection of •OH. Loading of the BSA protein on LDH surface, the biocompatibility and colloidal stability of the LDH-based fluorescent nanoprobe were further improved, facilitating the detection of •OH generation in macrophage polarization stimulated by both biomolecules and physical ultrasound irradiation. This study thus offers a new nanoprobe as the tool for investigating •OH evolutions, advancing the biomedical investigations of macrophage polarization associated inflammation.

Abstract:Remarkable advances of biological understanding have been made on the complex roles of reactive oxygen/nitrogen species (ROS/RNS) both as signaling molecules and pathogenic contributors in health and disease contexts. However, due to a persistent lack of reliable molecular tools, particularly small-molecule fluorogenic probes with decisively robust properties such as selectivity, sensitivity, chemo-/photo-stability, organelle-specificity, and rapid turn-on, researchers continue to face challenges of how to establish a strong mechanistic base for ever more intricate phenomena. Here we report the design, synthesis and application of several efficient fluorescent probes (HKSOX, HKGreen/Yellow, HKOCl, HKPerox, and HKOH series) for the selective detection and molecular imaging of endogenous superoxide, peroxynitrite, hypochlorous acid, hydrogen peroxide and hydroxyl radical, respectively, with cytosol, mitochondria and lysosome targeting capacity. Those probes open up exciting opportunities for establishing new experimental paradigms and accelerating mechanistically focused biological discoveries.

Abstract:Ulcerative colitis (UC), as the main inflammatory disease in the colon, is regarded as one of the most refractory diseases by World Health Organization. Hydrogen peroxide (H2O2) is a kind of reactive oxygen species (ROS) and is closely associated with the development of UC. Unfortunately, there is no an excellent fluorescence probe was designed for monitoring H2O2 in UC mice. Herein, a near-infrared (NIR) fluorescence probe (DCX-B) with large stokes shift (158 nm) is designed and synthesized, which displays good sensitivity and selectivity toward H2O2. The probe itself is non-fluorescent because intramolecular charge transfer (ICT) effect is blocked by the borate ester group. After DCX-B reacting with H2O2, the ICT process is restored and a strong fluorescence signal is released. Meanwhile, the possible reaction mechanism of DCX-B towards H2O2 is studied and proved by HPLC, MS and DFT calculation. Due to the low cytotoxicity of the probe, it is successfully applied in the image of H2O2 in two cell lines (HCT116 and HepG2 cells). Furthermore, enlightened by the properties of NIR emission and large stokes shift, DCX-B has been applied in monitoring the dynamic change of H2O2 in the diagnosis and treatment of UC mice.
Lighting Up NIR-II Fluorescence in Vivo: An Activable Probe for Noninvasive Hydroxyl Radical Imaging

Abstract:The detection of hydroxyl radical (·OH) in vivo faces challenges as ·OH has short lifetime and low concentration in the body. Fluorescence imaging within the second near-infrared window (NIR-II, 1000-1700 nm) is a promising approach to in vivo organ and tissue imaging, but ·OH fluorescent probes emitting at this region have not been reported up to now because of the difficulty of probe design. Herein, we report the strategy to fabricate the first NIR-II probe for ·OH by directly breaking/recovering the conjugated system and rigid planar structure of an organic fluorophore, which could regulate the fluorescence intensity regardless of emission wavelength. This activable probe, Hydro-1080, emitted in 1000-1400 nm after responding to ·OH. Hydro-1080 exhibited excellent sensitivity (LOD = 0.5 nM) and selectivity to ·OH. It was able to track subtle variation of [·OH] in liver induced by external stimuli and offered clear images with high contrast. This work also indicates that this simple and straightforward strategy can be extended to develop NIR-II fluorescent probes efficiently.

Abstract:The hydroxyl radical (·OH), one of the reactive oxygen species (ROS) in biosystems, is found to be involved in many physiological and pathological processes. However, specifically detecting endogenous ·OH remains an outstanding challenge owing to the high reactivity and short lifetime of this radical. Herein, inspired by the scavenging mechanism of a neuroprotective drug edaravone toward ·OH, we developed a new ·OH-specific fluorescent probe RH-EDA. RH-EDA is a hybrid of rhodamine and edaravone and exploits a ·OH-specific 3-methyl-pyrazolone moiety to control its fluorescence behavior. RH-EDA itself is almost nonfluorescent in physiological conditions, which was attributed to the formation of a twisted intramolecular charge transfer (TICT) state upon photoexcitation and the acylation of its rhodamine nitrogen at the 3′ position. However, upon a treatment with ·OH, its edaravone subunit was converted to the corresponding 2-oxo-3-(phenylhydrazono)-butanoic acid (OPB) derivative (to afford RH-OPB), thus leading to a significant fluorescence increase (ca. 195-fold). RH-EDA shows a high sensitivity and selectivity to ·OH without interference from other ROS. RH-EDA has been utilized for imaging endogenous ·OH production in living cells and zebrafishes under different stimuli. Moreover, RH-EDA allows a high-contrast discrimination of cancer cells from normal ones by monitoring their different ·OH levels upon stimulation with β-Lapachone (β-Lap), an effective ROS-generating anticancer therapeutic agent. Abstract:Hydroxyl radical (•OH), a highly reactive oxygen species (ROS), is assumed as one of the most aggressive free radicals. This radical has a detrimental impact on cells as it can react with different biological substrates leading to pathophysiological disorders, including inflammation, mitochondrion dysfunction, and cancer. Quantification of this free radical in‐situ plays critical roles in early diagnosis and treatment monitoring of various disorders, like macrophage polarization and tumor cell development. Luminescence analysis using responsive probes has been an emerging and reliable technique for in‐situ detection of various cellular ROS, and some recently developed •OH responsive nanoprobes have confirmed the association with cancer development. This paper aims to summarize the recent advances in the characterization of •OH in living organisms using responsive nanoprobes, covering the production, the sources of •OH, and biological function, especially in the development of related diseases followed by the discussion of luminescence nanoprobes for •OH detection.

Abstract:<p>Among various reactive oxygen species (ROS), hydroxyl radical (·OH) has the strongest oxidizability, which causes damage to a wide array of biomacromolecules in the cells, including DNA, lipid, and proteins. Therefore, the over-produced ·OH is verified to be implicated in various oxidative-stress related pathophysiological processes, such as inflammation, cancer, and cardiovascular disorder. However, the precise pathogenic roles played by this substance have been far from being clearly understood due to its high reactivity, short lifetime, and low concentration in biosystems. Thus, developing highly sensitive and selective detecting tools for monitoring ·OH is of significant importance. This review summarizes the advances that have been made in the last decade in the development of fluorescent sensors for ·OH detection based on small molecules and nanoparticles as well as their diverse biological applications.</p>

Abstract:Diabetes is one of the metabolic diseases marked by hyperglycemia and is often accompanied by the occurrence of some complications. As a biomarker of oxidative stress, hydrogen peroxide (H2O2) has close association with the occurrence and development of diabetes and its complications. Unfortunately, there is no fluorescent probe reported for imaging H2O2 in diabetic mice. Here, a novel near-infrared (NIR) fluorescent probe named QX-B was designed and synthesized to detect H2O2. For the probe, the quinolinium-xanthene dye is used as the fluorophore and borate ester is chosen as the response group. After the addition of H2O2, a strong MR fluorescence signal at 772 nm is observed. The probe not only shows high sensitivity with 10-fold enhancement but also displays excellent selectivity to H2O2 over other possible interfering species. In the meantime, the possible response mechanism of QX-B toward H2O2 was proposed and verified by the high-performance liquid chromatography (HPLC) experiment, mass spectra (MS) experiment, and density functional theory (DFT) calculation. Furthermore, based on the low cell cytotoxicity of QX-B, it has been applied in imaging exogenous and endogenous H2O2 in HeLa cells, HCT116 cells, 4T1 cells, and zebrafish successfully. More importantly, inspired by the performance of NIR fluorescence, QX-B has been used in monitoring H2O2 in diabetic mice for the first time. This provides very important information for the diagnosis and treatment of diabetes and its complications.

Abstract:Hydrogen peroxide(H2O2), as a reliable signaling biomolecule for oxidative stress, its accurate detection during agent-stimulated oxidative stress plays a vital role in pathological and physiological mechanism exploration for disease theranostics. It's necessary to develop an efficient method for their detection. In view of the advantages of fluorescent probes, we rationally constructed a novel fluorescent probe Compound 2 based on 4-(Bromomethyl)benzeneboronic acid pinacol ester_Herein, a small molecule fluorescent probe was fabricated using isoflore nitrile as fluorescent group, phenylboronic acid pinacol ester as the response group, to detect H2O2. The probe Compound 2 has a strong fluorescence intensity at 575 nm, indicating that the structure of the probe molecule is reasonably designed, and the Stokes shift is up to 172 nm. While the detection time is as low as 30 s and the LOD of the probe for H2O2 is as low as 3.7 μmol/L,the quantum yield is Φ = 40.31 %. It has been successfully used for imaging detection of H2O2 in HepG2 cells and zebrafish for its low toxicity. It can be found that this small molecule fluorescent probe can identify H2O2 in tumor cells significantly and efficiently, which would realize the early diagnosis of tumor.

Abstract:As one of endogenous reactive oxygen species (ROS), peroxynitrite (ONOO-) performs various functions in both pathological and physiological mechanisms. In this work, an optical and near-infrared (NIR) fluorescent probe (NX), which based on 3-dihydro-1H-xanthene and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) group was designed and prepared to detect ONOO-. This probe revealed an obvious optical and a fluorescent response when ONOO- was present and it exhibited higher selectivity over other ROS. Especially, the dual NIR fluorescence changes at 660 and 800 nm allowed quantitative detection of ONOO- in the range of 15-40 mu M, and the detection limit was 82 nM. Finally, the probe was effectively employed to visualize exogenous and endogenous ONOO- in HepG2 cells and zebrafish, respectively. All the results indicated the dual NIRchannel probe could serve as a potent detecting tools in studying ONOO- in vitro and in vivo.

Abstract:Hydroxyl radicals (•OH) is one of the commonly reactive oxygen species available in biosystem and plays a key role in a number of physiological and pathological processes. Based on our previous study, fluoride could serve as a moderate electron donating group in constructing fluorescence probes. However, there is seldom work on the effects of fluoride-substituted moiety towards the photophysical properties in constructing fluorescence probes. In this work, we developed a series of novel fluorescence probes based on a quinolinium-scaffold containing fluoride substituted donor. From our results, the probe FHyR-3 which has an extended conjugated system could detect hydroxyl radical with high selectivity, sensitivity, and fast response. Furthermore, the probe has the maximum emission at 508 nm with a large stoke shift (114 nm) and exhibits low toxicity, good biocompatibility and mito-targeting. We also demonstrated its capability in tracking hydroxyl radical level in living cells and zebrafish.

Abstract:As a member of the reactive oxygen species, hydrogen peroxide (H2O2) plays critical roles in oxidative stress and cell signaling. Intracellular abnormal levels of H2O2 production are closely related to many diseases. Therefore, the real-time monitoring of H2O2 in the cells is important. In this work, we designed a novel fluorescent probe (Mito-H2O2) for the specific detection of H2O2 based on the hemicyanine skeleton, with bright near-infrared fluorescence emission. Mito-H2O2 displayed fast response, excellent water-solubility and great fluorescence intensity enhancement after the addition of H2O2. Furthermore, Mito-H2O2 has been successfully applied to image both of the exogenous and endogenous H2O2 in cells and mice with negligible cytotoxity.

Abstract:As an essential reactive oxygen species (ROS), hypobromous acid (HOBr) plays a vital role in the immune process. Nevertheless, excessive HOBr can cause a series of diseases including cancers. Therefore, the development of the in situ detection methods for intracellular HOBr in biosystems is urgent and important to further uncover the detailed physiological and pathological effects of HOBr. In this work, a simple water-soluble near-infrared (NIR) fluorescence dye was reasonably designed and synthesized, which was satisfactory chosen as a highly selective and sensitive fluorescent probe for the rapid detection of HOBr based on the electrophilic bromination reaction mechanism. This probe can perform the specific detection of HOBr, which facilitates the accurate imaging of HOBr in biosystems. Finally, this probe was successfully applied to trace mitochondrial HOBr fluctuations with high resolution in living cells and zebrafish.

Abstract:Abnormal peroxynitrite (ONOO−) expression is closely associated with the development of various pathological processes. In this study, we devised a series of fluorescent probes, namely DHU-DCM-1, DHU-DCM-2, and DHU-DCM-3, tailored specifically for monitoring ONOO− through rationally molecular design. By modulating the electron-donating site within the fluorophore structure, the optimal probe DHU-DCM-3 exhibited excellent ratiometric detecting performance. DHU-DCM-3 displayed high selectivity and sensitivity towards ONOO−, effectively distinguishing it from other reactive oxygen species. Notably, DHU-DCM-3 was capable of labeling mitochondrial ONOO−, producing an enhanced fluorescence ratio (F656/F550), thereby allowing for the visualization of ONOO− fluctuations in both exogenous and endogenous contexts within living cells. The utility of DHU-DCM-3 was further demonstrated in various applications. For instance, through detecting elevated levels of ONOO−, senile C. elegans was successfully identified by utilizing DHU-DCM-3. Moreover, relying on its ratiometric fluorescence, DHU-DCM-3 was proved to be valuable in accurately diagnosing liver injury in mice with varying degrees of severity. Overall, we have constructed a ratiometric fluorescent probe for detecting and imaging ONOO− in both subcellular organelle and in vivo, highlighting its potential significance in various biomedical applications.
