Abstract:Marek's disease virus (MDV) strain GX0101 was the first reported field strain of recombinant gallid herpesvirus type 2 (GaHV-2). However, the splenic proteome of MDV-infected chickens remains unclear. In this study, a total of 28 1-day-old SPF chickens were intraperitoneally injected with chicken embryo fibroblast (CEF) containing 2000 PFU GX0101. Additionally, a control group, consisting of four one-day-old SPF chickens, received intraperitoneal equal doses of CEF. Blood and various tissue samples were collected at different intervals (7, 14, 21, 30, 45, 60, and 90 days post-infection; dpi) for histopathological, real-time PCR, and label-free quantitative analyses. The results showed that the serum expressions of MDV-related genes, meq and gB, peaked at 45 dpi. The heart, liver, and spleen were dissected at 30 and 45 dpi, and their hematoxylin-eosin staining indicated that virus infection compromised the normal organizational structure at 45 dpi. Particularly, the spleen structure was severely damaged, and the lymphocytes in the white medulla were significantly reduced. Furthermore, liquid chromatography-mass spectrometry (LC-MS) and label-free techniques were used to analyze the difference in splenic proteome profiles of the experimental and control groups at 30 and 45 dpi. Proteomic analysis identified 1660 and 1244 differentially expressed proteins (DEPs) at 30 and 40 dpi, respectively, compared with the uninfected spleen tissues. According to GO analysis, these DEPs were involved in processes such as organelle organization, cellular component biogenesis, cellular component assembly, anion binding, small molecule binding, metal ion binding, cation binding, cytosol, nuclear part, etc. Additionally, KEGG analysis indicated that the following pathways were linked to MDV-induced inflammation, apoptosis, and tumor: Wnt, Hippo, AMPK, cAMP, Notch, TGF-β, PI3K-Akt, Rap1, Ras, Calcium, NF-κB, PPAR, cGMP-PKG, Apoptosis, VEGF, mTOR, FoxO, TNF, JAK-STAT, MAPK, Prion disease, T cell receptor, and B cell receptor. We finally screened 674 DEPs that were linked to MDV infection in spleen tissue. This study improves our understanding of the MDV response mechanism in the spleen.

Abstract:Chickens infected with Marek's disease virus (MDV) carry the virus consistently for a long time, which increases the incidence and rate of virus-induced multi-organ tumors and increases its potential for horizontal transmission. There is a positive correlation between very virulent (vv) MDV quantity and the pathology. The purpose of this study was to determine the vvMDV loads dynamics in different phases, and the correlation between the viral quantity and tumor development. We used a SYBR Green duplex real-time quantitative PCR (q-PCR) assay to detect and quantify MDV loads and distributions in different tissues, targeting the Eco-Q protein gene (meq) of the virus and the house-keeping ovotransferrin (ovo) gene of chickens. The q-PCR was performed using different tissue DNA preparations derived from chickens which were infected with 1,000 pfu of the SDWJ1302 strain and tissue samples were collected from control and MDV-infected birds on 7, 10, 15, 21, 28, 40, 60, and 90 d post-infection (DPI). The data indicated that the MDV genome was almost quantifiable in immune organs of infected chickens as early as 7 DPI, and the number of MDV genome copies in the blood and different organs peaked by 28 DPI, but then gradually decreased by 40 DPI. The levels of viral quantity in the lymphocytes, liver, and spleen were all higher than those in other organs, and that in the feather follicles was the highest among different phases of MDV infection. The vvMDV could still be detected in peripheral blood and tissues by 90 DPI, and the vast existence of virus will stimulate tissue destruction. The data provided further evidence of viral infection involving multi-organ distribution and mainly involving immune organ proliferation, resulting in immunosuppression.

Abstract:Proteasomes are protein degradation machines that exist in cells as heterogeneous and dynamic populations. A group of proteins function as ubiquitin receptors (UbRs) that can recognize and deliver ubiquitinated substrates to proteasome complexes for degradation. Defining composition of proteasome complexes engaged with UbRs is critical to understand proteasome function. However, because of the dynamic nature of UbR interactions with the proteasome, it remains technically challenging to capture and isolate UbR-proteasome subcomplexes using conventional purification strategies. As a result, distinguishing the molecular differences among these subcomplexes remains elusive. We have developed a novel affinity purification strategy, in vivo cross-linking (X) assisted bimolecular tandem affinity purification strategy (XBAP), to effectively isolate dynamic UbR-proteasome subcomplexes and define their subunit compositions using label-free quantitative mass spectrometry. In this work, we have analyzed seven distinctive UbR-proteasome complexes and found that all of them contain the same type of the 26S holocomplex. However, selected UbRs interact with a group of proteasome interacting proteins that may link each UbR to specific cellular pathways. The compositional similarities and differences among the seven UbR-proteasome subcomplexes have provided new insights on functional entities of proteasomal degradation machineries. The strategy described here represents a general and useful proteomic tool for isolating and studying dynamic and heterogeneous protein subcomplexes in cells that have not been fully characterized.
Temporal Dynamics of Purinergic Receptor Expression in the Lungs of Marek's Disease (MD) Virus-Infected Chickens Resistant or Susceptible to MD

Abstract:Marek's disease virus (MDV) is an economic concern for the poultry industry due to its poorly understood pathophysiology. Purinergic receptors (PRs) are potential therapeutic targets for viral infections, including herpesviruses, prompting our investigation into their role in MDV pathogenesis. The current study is part of an experimental series analyzing the expression of PRs during MDV infection. To address the early or short-acting P2 PR responses during natural MDV infection, we performed an "exposure" experiment where age-matched chickens were exposed to experimentally infected shedders to initiate natural infection. In addition, select non-PR regulatory gene responses were measured. Two groups of naïve contact chickens (n = 5/breed/time point) from MD-resistant (White Leghorns: WL) and -susceptible (Pure Columbian) chicken lines were housed separately with experimentally infected PC (×PC) and WL (×WL) chickens for 6 or 24 h. Whole lung lavage cells (WLLC) were collected, RNA was extracted, and RT-qPCR assays were used to measure specific PR responses. In addition, other potentially important markers in pathophysiology were measured. Our study revealed that WL chickens exhibited higher P1 PR expression during natural infection. WL chickens also showed higher expression of P1A3 and P2X3 at 6 and 24 h when exposed to PC-infected chickens. P2X5 and P2Y1 showed higher expression at 6 h, while P2Y5 showed higher expression at 6 and 24 h; regardless of the chicken line, PC chickens exhibited higher expression of P2X2, P2Y8, P2Y10, P2Y13, and P2Y14 when exposed to either group of infected chickens. In addition, MDV infection altered the expression of DDX5 in both WL and PC groups exposed to PC-infected birds only. However, irrespective of the source of exposure, BCL2 and ANGPTL4 showed higher expression in both WL and PC. The expression of STAT1A and STAT5A was influenced by time and breed, with major changes observed in STAT5A. CAT and SOD1 expression significantly increased in both WL and PC birds, regardless of the source of infection. GPX1 and GPX2 expression also increased in both WL and PC, although overall lower expression was observed in PC chickens at 24 h compared to 6 h. Our data suggest systemic changes in the host during early infection, indicated by the altered expression of PRs, DDX5, BCL2, ANGPTL4, and other regulatory genes during early MDV infection. The relative expression of these responses in PC and WL chickens suggests they may play a key role in their response to natural MDV infection in the lungs and long-term pathogenesis and survival.

Experimental Velogenic Viscerotropic Newcastle Disease Virus Infection in Chickens Immunologically Impaired by Treatment with Cyclophosphamide

Abstract:This study investigated whether lymphocytic depletion following chemical bursectomy influenced the severity of infection and development of lesions in chickens challenged with velogenic viscerotropic Newcastle disease virus (vvNDV). Cockerel chickens treated with cyclophosphamide on days 2, 3 and 4 post-hatch showed loss of weight, atrophy and lymphocytic depletion in the bursa of Fabricius and spleen. At 6 weeks of age, the chickens were assigned to four groups- Bursectomized intramuscularly vvNDV inoculated (BI), bursectomized uninfected (BU), non-bursectomized infected (NBI) and non-bursectomized uninfected (NBU) chickens. The BI and NBI chickens showed significant (P < .05) loss of weight than their uninfected controls. Depression, anorexia, greenish diarrhea, listlessness, tremor, and oculo-nasal discharges were observed in both infected groups, but were more severe and frequent in the NBI than in the BI chickens. Total mortalities were 100% and 95.5% for the NBI and BI chickens, respectively (P > .05). Lesions in both infected groups included atrophy of the bursa, spleen and thymus. Hemorrhages in the proventricular mucosa, intestines and cecal tonsils, as well as congestion and enlargement of the kidneys were significantly (P < .05) more severe and frequent in NBI than BI chickens. Histopathology showed necrosis and depletion of lymphocytes in the three lymphoid organs in both infected groups with more severity in the NBI than BI chickens. These results show that depletion of lymphocytes by treatment with cyclophosphamide may influence the severity of infection and development of lesions in vvNDV infection in cockerel chickens.

Abstract:Gallid alphaherpesvirus 2 (GaHV2) is an oncogenic avian herpesvirus inducing Marek’s disease (MD) and rapid-onset T-cell lymphomas. To reveal molecular events in MD pathogenesis and tumorigenesis, the dynamic splenic transcriptome of GaHV2-infected chickens during early infection and pathogenic phases has been determined utilizing RNA-seq. Based on the significant differentially expressed genes (DEGs), analysis of gene ontology, KEGG pathway and protein-protein interaction network has demonstrated that the molecular events happening during GaHV2 infection are highly relevant to the disease course. In the ‘Cornell Model’ description of MD, innate immune responses and inflammatory responses were established at early cytolytic phase but persisted until lymphoma formation. Humoral immunity in contrast began to play a role firstly in the intestinal system and started at late cytolytic phase. Neurological damage caused by GaHV2 is first seen in early cytolytic phase and is then sustained throughout the following phases over a long time period. During the proliferative phase many pathways associated with transcription and/or translation were significantly enriched, reflecting the cell transformation and lymphoma formation. Our work provides an overall view of host responses to GaHV2 infection and offers a meaningful basis for further studies of MD biology.
A Marek's Disease Virus Messenger RNA-Based Vaccine Modulates Local and Systemic Immune Responses in Chickens

Abstract:Marek's disease (MD), caused by the Marek's disease virus, is a lymphoproliferative disease in chickens that can be controlled by vaccination. However, the current vaccines can limit tumor growth and death but not virus replication and transmission. The present study aimed to evaluate host responses following intramuscular injection of an mRNA vaccine encoding gB and pp38 proteins of the MDV within the first 36 h. The vaccine was injected in low and high doses using prime and prime-boost strategies. The expression of type I and II interferons (IFNs), a panel of interferon-stimulated genes, and two key antiviral cytokines, IL-1β and IL-2, were measured in spleen and lungs after vaccination. The transcriptional analysis of the above genes showed significant increases in the expression of MDA5, Myd88, IFN-α, IFN-β, IFN-γ, IRF7, OAS, Mx1, and IL-2 in both the spleen and lungs within the first 36 h of immunization. Secondary immunization increased expression of all the above genes in the lungs. In contrast, only IFN-γ, MDA5, MyD88, Mx1, and OAS showed significant upregulation in the spleen after the secondary immunization. This study shows that two doses of the MDV mRNA vaccine encoding gB and pp38 antigens activate innate and adaptive responses and induce an antiviral state in chickens.

Sequential histopathological changes of the lower respiratory and gastrointestinal tracts in chickens infected with velogenic Newcastle disease virus

Abstract:Velogenic Newcastle disease is a highly contagious and fatal disease that affects domestic poultry, and is clinically manifested by haemorrhagic gastroenteritis and respiratory distress. This study investigated the sequential histopathogic changes in the lungs, proventriculus, and intestines of broiler chickens infected with velogenic Newcastle disease virus (vNDV). Fifty broiler chickens (6-weeks-old) were randomly assigned into two groups of 25 each: UC – unvaccinated with Newcastle disease (ND) vaccines and intranasallly inoculated with vNDV, UU – unvaccinated uninfected. At 4, 5 and 6 days post-infection (d.p.i) tissue samples were collected from birds in each group for histopathology. The lungs showed hyperaemia, haemorrhages, inflammatory oedema and cellular infiltrations in the secondary bronchi, parabronchi (atria, infundibular, air and blood capillaries of the exchange tissue), depletion of bronchial-asociated lymphoid tissues, loss of cilia and hyperplasia of surface epithelium of bronchi and parabronchi at 4 d.p.i. The lesions progressed to collapsed air capillaries with fibroplasia and fibrosis of the area at 6 d.p.i. Similar inflammatory changes were observed in the proventriculus, but accompanied by degeneration and attenuation of surface epithelium of proventiculus and proventricular glands at 4 d.p.i., and severe necrosis and disintegration of the proventricular glands at 6 d.p.i. The intestine showed severe inflammatory changes in the mucosa, submucosa and serosa and depletion of gut-associated lymphoid tissues at 4 d.p.i., and ulcerative enteritis at 5 d.p.i. The lumen was filled with necrotic debris at 6 d.p.i. This study reinforces the concept making pathologic animal experiments important tools for a complete characterization of strains in susceptible animals, as this information will be helpful in the accurate diagnosis of velogenic Newcastle disease in chickens.
Role of Macrophages in the Pathogenesis of Genotype VII Newcastle Disease Virus in Chickens

Abstract:Long-term evolution of Newcastle disease virus (NDV) results in substantial alteration in viral pathogenesis. NDVs of genotype VII, a late genotype, show marked tropism to lymphoid tissues, especially to macrophages in chickens. However, the role of macrophages in the pathogenesis of genotype VII NDV is still unclear. Herein, NDV infectivity in macrophages and the role of macrophages in the pathogenesis of genotype VII NDV in chickens were investigated. We reported that NDV strains of genotype VII (JS5/05) and IV (Herts/33) can replicate in the adherent (predominantly macrophages) and non-adherent cells (predominantly lymphocytes) derived from chicken peripheral blood mononuclear cells (PBMCs), and significantly higher virus gene copy was detected in the adherent cells. In addition, JS5/05 had significantly higher infectivity in PBMC-derived adherent cells than Herts/33, correlating with its enhanced tropism to macrophages in the spleen of chickens. Interestingly, the depletion of 68% of macrophages exerted no significant impact on clinical signs, mortality and the systematic replication of JS5/05 in chickens, which may be associated with the contribution of non-depleted macrophages and other virus-supportive cells to virus replication. Macrophage depletion resulted in a marked exacerbation of tissue damage and apoptosis in the spleen caused by JS5/05. These findings indicated that macrophages play a critical role in alleviating tissue damage caused by genotype VII NDV in chickens. Our results unveiled new roles of macrophages in NDV pathogenesis in chickens.

Transcriptional Analysis Of Host Responses To Marek'S Disease Virus Infection In Chicken Thymus

Abstract:Marek's disease virus (MDV) is a cell-associated alpha-herpesvirus that causes T-cell lymphomas and nervous disorders in chickens. Different from other lymphoid organs, the thymus is the site of T-cell maturation and differentiation. However, the transcriptional response to MDV infection in the chicken thymus is still not known. In this study, we performed genome-wide expression analysis in thymus tissues of RB1B-infected chickens at different time points to investigate the molecular mechanisms of MDV pathogenesis. The number of differentially expressed genes with 2-fold or higher changes (>2) are as follows: 1,250 genes (7 dpi), 834 genes (14 dpi), 1,958 genes (21 dpi), and 2,306 genes (28 dpi). Gene ontology enrichment analysis revealed that the upregulated genes were involved in immune and inflammatory response at 7 dpi; angiogenesis, cytoskeleton organization, cell adhesion, and signal transduction showed different expressions at 21 and 28 dpi. The expression pattern of 18 randomly selected genes was confirmed by real-time RT-PCR. Several differently expressed host genes associated with tumor development are discussed. We identified the global host-gene expression pattern in the thymus of chickens that responded to MDV infection. Impaired Intestinal Mucosal Immunity in Specific-Pathogen-free Chickens after Infection with Very Virulent Infectious Bursal Disease Virus.

Abstract:Diarrhea is one of the most important clinical side effects that occurs in chickens after infection with bursal disease virus. However, the kinetics of the intestinal mucosal immune parameters in infectious bursal disease virus (IBDV)-infected birds remains unclear. Forty-five specific-pathogen-free chickens were randomly divided into 2 groups [30 chickens in the very virulent IBDV (vvIBDV)-infected group and 15 chickens in the control group] to determine the effect of vvIBDV infection on intestinal mucosal immunity. All chickens were raised in negative-pressure isolators and fed the same diet. Results indicate that severe histologic lesions were observed in the intestinal tract in the vvIBDV-infected group, and the villus heights of the duodenum, jejunum, and ileum were decreased after vvIBDV infection (P < 0.01 or P < 0.05). The number of intestinal intraepithelial lymphocytes, mast cells, and goblet cells decreased significantly in the vvIBDV-infected group compared with the control (P < 0.01 or P < 0.05) on d 1, 2, and 3 postchallenge with vvIBDV. Very virulent IBDV infection reduced the expression of each fragment of intestinal secretory IgA at all 3 time points. Intestinal alkaline phosphatase decreased dramatically on d 2 and 3 postchallenge with vvIBDV. Taken together, these results indicate that vvIBDV infection contributes to considerable damage to the normal structure of the intestine and intestinal mucosal immune parameters of specific-pathogen-free chickens. Our research helps to increase an understanding of the pathogenesis of diarrhea induced by vvIBDV infection. These findings provide new insights into the pathogenesis and pathophysiology of vvIBDV infection.
COMPARATIVE PATHOLOGICAL STUDY OF EXPERIMENTAL INFECTION OF NEWCASTLE DISEASE VIRUS GENOTYPES VII IN CHICKENS AND DUCKS

Abstract:Newcastle disease virus (NDV) is a highly contagious virus infects a wide range of avian species including chicken and ducks. The disease can be fatal for many types of them. It was believed that ducks and geese act as natural reservoirs of NDV because of their natural resistance. Recently, NDV isolates were pathogenic for ducks. Therefore, our study aimed to investigate the pathogenicity of highly pathogenic NDV genotype VII in white Ross commercial broiler chickens and Mallard ducks on the basis of clinical, histological observation and immunohistochemistry findings of virus antigen through studying the sequential pathological lesions of both species at 2nd, 5th and 10th dpi. Both species were inoculated via intranasal route by NDV genotype VII (accession number MT887290). Clinical lesions and mortalities were evident mostly in chickens rather than in ducks. Respiratory, digestive and nervous signs were clearly manifested in chickens. Ducks showed low level of mortalities and the nervous signs were the most obvious symptoms. Microscopic findings of chickens showed severe pancreatitis, pneumonia, enteritis as well as pericarditis and brain malacia. Similar findings were noticed in ducks but with lower degree of severity. Brain lesions in ducks included encephalitis and perivascular reaction mostly within the cerebellar tissue. Immunohistochemistry reaction against NDV antigen showed marked immunostaining within epithelial cells, purkinje cells and inflammatory cells mostly the macrophages. The immunohistochemistry reaction was correlated with the pathological findings in both species. In conclusion, the mortality and lesions of NDV in both species were correlated with systemic alterations associated with pancreatitis, pneumonia and encephalopathy. Moreover, the obtained data revealed possible susceptibility of ducks to NDV genotype VII, which might be need forthcoming viral vaccination strategy for ducks against NDV.

Abstract:Abstract Marek’s disease virus (MDV) is a highly oncogenic alphaherpesvirus that causes deadly T-cell lymphomas and serves as a natural virus-induced tumor model in chickens. Although Marek’s disease (MD) is well controlled by current vaccines, the evolution of MDV field viruses towards increasing virulence is concerning as a better vaccine to combat very virulent plus MDV is still lacking. Our understanding of molecular and cellular immunity to MDV and its immunopathogenesis has significantly improved, but those findings about cellular immunity to MDV are largely out-of-date, hampering the development of more effective vaccines against MD. T-cell-mediated cellular immunity was thought to be of paramount importance against MDV. However, MDV also infects macrophages, B cells and T cells, leading to immunosuppression and T-cell lymphoma. Additionally, there is limited information about how uninfected immune cells respond to MDV infection or vaccination, specifically, the mechanisms by which T cells are activated and recognize MDV antigens and how the function and properties of activated T cells correlate with immune protection against MDV or MD tumor. The current review revisits the roles of each immune cell subset and its effector mechanisms in the host immune response to MDV infection or vaccination from the point of view of comparative immunology. Anim. Res., 27: 1–5. In this experiment, 414 chickens were reared in a house in which the previous flock had experienced Marek's disease. Each bird was given 10 doses of infectious bursal disease vaccine on day 11 and on day 19 half of the chickens in every other pen were vaccinated with 2 doses of chicken anemia virus vaccine. All birds were bled on day 23 for MHC haplotype stereotyping. Clostridium perfringens, Clostridium septicum and Staphylococcus aureus were used to challenge the birds on day 29 which were then killed one week after challenge. Statistical analysis showed the percentage of cellulitis in the CAV vaccinated group and NON-CAV vaccinated group was of the same level, while the percentage of myositis occurring in the CAV vaccinated group was significantly higher than that which occurred in the chickens of NON-CAV vaccinated group. Chickens with MHC haplotype of BA9/BA9 experienced significantly lower levels of cellulitis than did chickens of B21/B21 haplotype. For both the MHC haplotypes, chickens in the CAV vaccinated groups were more resistant than the chickens of NON-CAV vaccinated group to the development of cellulitis.

Fully Attenuated Meq and Pp38 Double Gene Deletion Mutant Virus Confers Superior Immunological Protection Against Highly Virulent Marek’s Disease Virus Infection

Abstract:Marek's disease virus (MDV) induces immunosuppression and neoplastic disease in chickens. The virus is controllable via an attenuated meq deletion mutant virus, which has the disadvantage of retaining the ability to induce lymphoid organ atrophy. To overcome this deficiency and produce more vaccine candidates, a recombinant MDV was generated from the highly virulent Md5BAC strain, in which both meq and a cytolytic replication-related gene, pp38, were deleted. Replication of the double deletion virus, Md5BAC ΔmeqΔpp38, was comparable with that of the parental virus in vitro. The double deletion virus was shown to be fully attenuated and to reduce lymphoid organ atrophy in vivo. Crucially, Md5BAC ΔmeqΔpp38 confers superior protection against highly virulent virus compared with a commercial vaccine strain, CVI988/Rispens. Transcriptomic profiling indicated that Md5BAC ΔmeqΔpp38 induced a different host immune response from CVI988/Rispens. In summary, a novel, effective, and safe vaccine candidate for prevention and control of MD caused by highly virulent MDV is reported. IMPORTANCE MDV is a highly contagious immunosuppressive and neoplastic pathogen. The virus can be controlled through vaccination via an attenuated meq deletion mutant virus that retains the ability to induce lymphoid organ atrophy. In this study, we overcame the deficiency by generating meq and pp38 double deletion mutant virus. Indeed, the successfully generated meq and pp38 double deletion mutant virus had significantly reduced replication capacity in vivo but not in vitro. It was fully attenuated and conferred superior protection efficacy against very virulent MDV challenge. In addition, the possible immunological protective mechanism of the double deletion mutant virus was shown to be different from that of the gold standard MDV vaccine, CVI988/Rispens. Overall, we successfully generated an attenuated meq deletion mutant virus and widened the range of potential vaccine candidates. Importantly, this study provides for the first time the theoretical basis of vaccination induced by fully attenuated gene-deletion mutant virus.

Abstract:Recent studies here have demonstrated that increased mast cell populations and tryptase activity contribute to lesion formation in regions of immune organs in special-pathogen-free chickens after infection with very virulent infectious bursal disease virus (vvIBDV). Mast cells and their mediators have been implicated in acute inflammatory injury after vvIBDV infection, but their precise role in this process remains elusive. In this study, the role of mast cells in the vvIBDV infection process was examined using ketotifen, a mast cell membrane stabilizer. On days 1, 2, and 3 postinfection, the bursa of Fabricius (BFs) were collected to quantify mast cells, tryptase and histamine contents by cytochemistry, immunohistochemistry and fluorospectrophotometry analyses, respectively. The results showed that the mast cell populations, tryptase expression, and histamine released increased significantly in the BFs (p<0.01) of infected birds compared to controls, and acute inflammatory responses were observed in the former. In contrast, in infected chickens pretreated with ketotifen, mast cells, tryptase, and histamine were markedly decreased (p<0.01) and probably as a result, the BFs remitted significantly. The overall results suggest that mast cells are positively involved in BF injury induced by vvIBDV infection. Inhibition of mast cell degranulation and concurrent mediator release may represent a novel strategy to modulate this process. This study, thus, advances the understanding of the acute inflammatory injury mechanisms triggered by vvIBDV infection and the contribution of mast cell activity in this process.

Abstract:Marek's disease virus (MDV) is a highly contagious pathogen that causes neoplastic disease, named as Marek's disease (MD). Although it was first described as polyneuritis disease, MD also induces severe immunosuppression and tumors in infected chickens. Based on biological characteristics, MDV is classified into three serotypes. MDV-1 belonging to serotype 1, includes pathogenic strains and their derivatives. Serotype 2 (MDV-2) and 3 (MDV-2) are non-pathogenic strains, usually used as vaccines. Pathogenic MDV-1 is categorized into four pathotypes: Mild virulent (m), virulent (v), very virulent (vv), and very virulent plus (vv+). MD is the first neoplastic disease that has been successfully controlled by vaccine. By using vaccines, the morbidities and mortalities of chickens were significantly reduced in the farm. However, MDV continues to evolve, and more virulent strains have gradually been seen to break through vaccinal immunity, which results in outbreak of disease. In recent years, highly virulent strains have been isolated in the world, becoming a new threat for the control of MD.The natural route of MDV infection is through inhalation of airborne cell-free virus particles from the contaminated dust and dander. Once the virus is inhaled, lung epithelial cells and macrophages are speculated to be the primary target cells for MDV infection. Then the virus infects and amplifies in B cells of lymphoid organs (early cytolytic infection). Finally, T cells, mainly activated CD4(+) T cells are recruited to the lymphoid organs and become latently infected (latency, followed by late cytolytic infection) and therefore transformed (transformation). Reactivation (late cytolytic infection) describes the recurrence of cytolytic replication in response to intra- and/or extracellular stimuli. MDV encodes more than 100 genes. Among them, meq is an oncogenic gene that is unique to MDV-1 and consistently expressed in all MD lymphomas and cell lines. Deletion of the meq gene completely abrogates the tumorigenesis induced by virulent strains, however, still maintains the ability to induce lymphoid organ atrophy. The meq-deleted viral strain could not be used as vaccine even though possessing superior protection advantages. To overcome the disadvantage and improve the safety and usefulness of meqdeleted vaccine strain, in an earlier study, we generated a meq and lorf9 deletion mutant virus as a vaccine candidate. It is important to know if the double-gene deletion vaccine strain still maintains the ability of reactivation and amplification in late cytolytic infectious stage, which may cause more viruses shedding into the environment. To test this hypothesis, Realtime PCR technology was used for quantitative analysis, by detecting MDV-specific ICP4 and chicken OVO genes. Viral content was measured from splenocytes and feather follicle of 63-days old infected chickens. Standard curves were established in the first step, and then total DNA was extracted. CT values were obtained both for ICP4 and OVO genes. Viral copy number per lymphocyte was calculated using the standard curve. The results indicate that the double-gene deletion mutant virus showed significantly reduced replication during latent and reactivation stages. This will be the theoretical basis for further analysis of immunological mechanism induced by the double-gene deletion mutant virus.

Abstract:The potential role of the mast cells in the invasion of very virulent infectious bursal disease virus (vvIBDV) is unknown. We evaluated mast cell activity and tryptase production after vvIBDV infection in special pathogen-free (SPF) chickens using cytochemistry and immunohistochemistry analyses. The results were as follows: (1) severe histologic lesions were observed in the thymus, spleen, cloacal bursa, liver, kidney and other tissues. vvIBDV viral antigens were detected and presented extensively in the parenchymatous organs, in particular, the cloacal bursa, liver, kidney, thymus, spleen and pancreas. (2) In the vvIBDV-infected group, the mast cell population increased markedly in the liver, kidney, thymus, glandular stomach, spleen and cloacal bursa on days 1, 2 and 3 after vvIBDV infection (p<0.05). However, very few mast cells were observed in those same tissues in the controls, especially in the bursa of Fabricius. (3) Tryptase, a marker for activated mast cells, has a positive correlation with mast cell distribution. The mast cells identified in the tissues were likely to be activated since they were associated with cell degranulation and the presence of tryptase. Furthermore, the co-localization of mast cells, and presence of vvIBDV antigens suggests that the mast cells were activated by vvIBDV infection. Our results also suggest that tryptase may contribute to the inflammation of acute IBD induced by vvIBDV infection. Our research contributes to the further understanding of inflammatory response mechanisms and the contribution of mast cell activity to this process.

Abstract:By planned immunization of a volunteer, two stable (≥6 months), specific, alloreactive cytolytic T‐cell clones have been established from his peripheral blood lymphocytes. One clone reacts with all serologically defined HLA‐Cw3 cells from our panel, whereas the other defines a split within the serological HLA‐B40 specificity. The two cytotoxic clones are SmIg‐negative, E‐rosette positive, EA and EAC rosette‐negative. HLA‐A, ‐B and ‐C‐positive, and also HLA‐DR‐ or ‘Ia like‐positive. In addition, they present very similar patterns of iodinated cell surface molecules as analysed by sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS‐PAGE), contrasting with that of an EBV cell line derived from the same donor.
