Abstract:A major problem related to the fabrication of drug-loaded fibers by wet spinning is the significant loss of load when the drugs and spinning dopes are directly mixed together. We report a novel method to fabricate drug-loaded alginate fibers by spinning an aqueous mixture containing alginate and alginate nanocapsules incorporated with drugs. The alginate fibers exhibit spotty features of nanocapsules located close to the surface of the fibers. Adsorption experiments demonstrate the superabsorbent properties of the fibers, as well as the dependence of adsorption profiles on salt ionic strength. The adsorption processes of all the fibers are well-described by the pseudo-second-order kinetics model. Drug release tests show that the cumulative release amount increased with an increase in the proportion of nanocapsules present in the fiber. In addition, the burst release behaviors of the fibers incorporated with the nanocapsules were also diminished dramatically. This work suggests that adding nanocapsules containing drugs into the spinning dopes is a promising strategy to fabricate novel drug-loaded fibers for immediate drug delivery for wound dressing.
[Research of diclofenac sodium-loaded gelatin scaffold with anti-inflammatory activity for promoting in vivo cartilage regeneration]

Abstract:Objective: To develop a diclofenac sodium-loaded gelatin scaffold with anti-inflammatory activity and provide a new avenue for alleviating the inflammatory response and enhancing cartilage regeneration in vivo. Methods: Diclofenac sodium was homogeneously mixed with gelatin to prepare a diclofenac sodium-loaded porous gelatin scaffold by freeze-drying method as the experimental group, and a pristine porous gelatin scaffold was served as a control group. The general morphology of the scaffold was observed, the pore size of the scaffold was measured by scanning electron microscopy, the porosity of the scaffold was calculated by drainage method, the loading of diclofenac sodium into the gelatin scaffold was detected by fourier transform infrared spectrometer and X-ray diffraction examinations, and the release kinetics of diclofenac sodium from gelatin scaffold was tested using an in vitro release assay. The two scaffolds were co-cultured with lipopolysaccharide-predisposed RAW264.7 in vitro, and the expressions of interleukin 1β (IL-1β) and tumor necrosis factor α (TNF-α) were detected by reverse transcription polymerase chain reaction (RT-PCR), enzyme-linked immuno sorbent assay, and Western blot, to detect the in vitro anti-inflammatory effect of the drug-loaded scaffold. Thereafter, the second generation chondrocytes of New Zealand white rabbits were inoculated on the two groups of scaffolds for in vitro culture, and the cytocompatibility of the scaffold was tested by live/dead staining and cell counting kit 8 assay, the feasibility of in vitro cartilage regeneration of the scaffold was evaluated via gross observation, HE staining, Safranin-O staining, and immunohistochemical collagen type Ⅱ staining, as well as biochemical quantitative analyses. Finally, the two groups of chondrocyte-scaffolds were implanted subcutaneously into New Zealand white rabbits, and after 4 weeks, the general observation, HE staining, safranin O staining, immunohistochemical collagen type Ⅱ staining, and biochemical quantitative analyses were performed to verify the cartilage regeneration in vivo, and the expression of inflammation-related genes CD3 and CD68 was detected by RT-PCR to comprehensively evaluate the anti-inflammatory performance of the scaffolds in vivo. Results: The two scaffolds exhibited similar gross, microporous structure, pore size, and porosity, showing no significant difference (P>0.05). Diclofenac sodium was successfully loaded into gelatin scaffold. Data from in vitro anti-inflammatory assay suggested that diclofenac sodium-loaded gelatin scaffold showed alleviated gene and protein expressions of IL-1β and TNF-α when compared with gelatin scaffold (P<0.05). The evaluation of cartilage regeneration in vitro showed that the number of living cells increased significantly with the extension of culture time, and there was no significant difference between the two groups at each time point (P>0.05). White cartilage-like tissue was regenerated from the scaffolds in both groups, histological observation showed typical cartilage lacuna structure and specific cartilage extracellular matrix secretion. There was no significant difference in the content of cartilage-specific glycosaminoglycan (GAG) and collagen type Ⅱ between the two groups (P>0.05). In vivo experiments showed that the samples in the experimental group had porcelain white cartilage like morphology, histologic staining showed obvious cartilage lacuna structure and cartilage specific extracellular matrix, the contents of GAG and collagen type Ⅱ were significantly higher than those in the control group, and the protein and mRNA expressions of CD3 and CD68 were significantly lower than those in the control group, with significant differences (P<0.05). Conclusion: The diclofenac sodium-loaded gelatin scaffold presents suitable pore size, porosity, and cytocompatibility, as well as exhibited satisfactory anti-inflammatory ability, providing a reliable scheme for alleviating the inflammatory reaction of regenerated cartilage tissue after in vivo implantation and promoting cartilage regeneration in vivo.
Evaluating Antibacterial Efficacy and Biocompatibility of PAN Nanofibers Loaded with Diclofenac Sodium Salt

Abstract:Side effects of the drugs’ oral administration led us to examine the possibility of using diclofenac sodium (DLF) in a polymeric drug delivery system based on electrospun polyacrylonitrile (PAN) nanofibers, which can be produced cost-effectively and with good applicability for transdermal treatments. The inclusion of DLF in PAN nanofibers increased the nanofiber diameter from ~200 nm to ~500 nm. This increase can be attributed to the increase in the spinning solution viscosity. FTIR spectra confirm the entrapment of the DLF into the PAN nanofibers. X-ray diffraction pattern showed that the inclusion of the DLF in the PAN nanofibers had caused the misalignment in the polymeric chains of the PAN, thus resulting in a decrease of the peak intensity at 2θ = 17o. The DLF loaded PAN nanofibers were efficient against the gram-positive Staphylococcus aureus (S. aureus) and gram-negative Escherichia coli (E. coli), with maximum inhibition zone of 16 ± 0.46 mm for E. coli and 15.5 ± 0.28 mm for S. aureus. Good cell viability ~95% for L929 cells in more extended incubation periods was reported. A gradual release of DLF from the PAN nanofiber was observed and can be attributed to the stability of Pan in PBS medium. Cell adhesion micrographs show that cell-cell interaction is stronger than the cell-material interaction. This type of weak cell interaction with the wound dressing is particularly advantageous, as this will not disturb the wound surface during the nursing of the wound.

Abstract:Nanofiber scaffolds have been used widely for wound healing applications. These scaffolds are commonly produced from different biopolymers, but usually a combination of synthetic and natural biopolymers is preferred because they exhibit both physio-mechanical stability and an improved environment for cell growth. In this article, polycaprolactone–gelatin (PCL–gel) hybrid nanofibers were produced by two nozzle electrospinning. Cellulose nanocrystals (CNC) were synthesized and incorporated into the gel and PCL nanofibers to improve their properties. SEM images of nanofibers showed two diameter distributions with diameter averages of 140 nm and 215 nm, corresponding to gel and PCL, respectively. Although XRD patterns showed a decrease in crystallinity, the crystal sizes increased from 16.1 and 11.1 to 19.4 and 12.4 nm respectively for the (110) and (200) crystalline planes when CNC was present in the nanofibers. Mechanical studies revealed an 80% and 60% increase in modulus and tensile strength, respectively, when CNC was incorporated. In vitro studies showed that the CNC-incorporated scaffold degraded 25% more rapidly. However, the MTT assay, cell morphology, and fluorescence staining experiments showed that CNC did not affect nanofiber biocompatibility, and cells could grow, differentiate and cover the scaffold surface. Scaffolds with and without CNC both promoted robust wound healing in Balb/c mice.

Abstract:Controlled drug release systems are the subject of many investigations to achieve the therapeutic effect of drugs. They have numerous advantages, such as localized effects, lower side effects, and less onset of action. Among drug-delivery systems, electrospinning is a versatile and cost-effective method for biomedical applications. Furthermore, electrospun nanofibers are promising as drug carrier candidates due to their properties that mimic the extracellular matrix. In this work, electrospun fibers were made of Poly-L-lactic acid (PLA), one of the most widely tested materials, which has excellent biocompatible and biodegradable properties. A curcuminoid, bisdemethoxycurcumin (BDMC) was added in order to complete the drug delivery system. The PLA/BDMC membranes were characterized, and biological characteristics were examined in vitro. The results show that the average fiber diameter was reduced with the drug, which was mainly released during the first 24 h by a diffusion mechanism. It was seen that the use of our membranes loaded with BDMC enhanced the rate of proliferation in Schwann cells, the main peripheral neuroglial cells, and modulated inflammation by reducing NLRP3 inflammasome activation. Considering the results, the prepared PLA/BDMC membranes hold great potential for being used in tissue engineering applications.

Abstract:Chronic inflammation plays a side effect on tissue regeneration, greatly inhibiting the repair or regeneration of tissues. Conventional local delivery of anti-inflammation drugs through physical encapsulation into carriers face the challenges of uncontrolled release. The construction of an inflammation-responsive prodrug to release anti-inflammation drugs depending on the occurrence of inflammation to regulate chronic inflammation is of high need. Here, we construct nanofiber-based scaffolds to regulate the inflammation response of chronic inflammation during tissue regeneration. An inflammation-sensitive prodrug is synthesized by free radical polymerization of the indomethacin-containing precursor, which is prepared by the esterification of N-(2-hydroxyethyl) acrylamide with the anti-inflammation drug indomethacin. Then, anti-inflammation scaffolds are constructed by loading the prodrug in poly(ε-caprolactone)/gelatin electrospun nanofibers. Cholesterol esterase, mimicking the inflammation environment, is adopted to catalyze the hydrolysis of the ester bonds, both in the prodrug and the nanofibers matrix, leading to the generation of indomethacin and the subsequent release to the surrounding. In contrast, only a minor amount of the drug is released from the scaffold, just based on the mechanism of hydrolysis in the absence of cholesterol esterase. Furthermore, the inflammation-responsive nanofiber scaffold can effectively inhibit the cytokines secreted from RAW264.7 macrophage cells induced by lipopolysaccharide in vitro studies, highlighting the great potential of these electrospun nanofiber scaffolds to be applied for regulating the chronic inflammation in tissue regeneration.
Enhanced Anti‐inflammatory Effects of Diclofenac Delivered Orally via Polyvinylpyrrolidone K30/Silk Fibroin Nanoparticles in a Murine Model of Carrageenan‐Induced Paw Edema

Abstract:Diclofenac has a relatively low oral bioavailability (50‐60%) and is quickly metabolized with a half‐life of less than 1 h. Therefore, the oral therapeutic effect of diclofenac is not optimal. This research developed polyvinylpyrrolidone K30‐functionalized silk fibroin nanoparticles as an effective delivery system for diclofenac (FNPs‐PVP‐DC). The FNPs‐DC and FNPs‐PVP‐DC were formulated by two methods of adsorption and solvent exchange. Depending on the formulation factors, the obtained particles exhibited different properties of nano‐scale sizes (400‐800 nm), narrow size distribution, negatively charged surfaces (‐17 to ‐19 mV), high PVP K30 incorporation (23%‐50%), pHpzc of ~6.6, and appropriate chemical interactions. Interestingly, particles formulated by the adsorption method showed low drug encapsulation efficiencies of < 15%, whereas the solvent exchange method yielded moderate results of ~40%. The FNPs‐DC possessed aggregated patterns, while the FNPs‐PVP‐DC were more uniformly distributed. All formulations limited diclofenac release (< 20%) under gastric conditions and sustained its release in the intestinal environment. In in‐vivo carrageenan‐induced paw edema mice model, the FNPs‐PVP‐DC demonstrated a 20‐30% higher anti‐inflammatory effect and a faster onset of action (within 1 h) compared to pure diclofenac at the same dose (5 mg/kg). These findings suggest that FNPs‐PVP‐DC have promising potential as novel oral anti‐inflammatory products.

Fabrication of dual drug-loaded polycaprolactone-gelatin composite nanofibers for full thickness diabetic wound healing

Abstract:Aim: Design of moxifloxacin and ornidazole co-loaded polycaprolactone and gelatin nanofiber dressing for diabetic wounds. Materials & methods: The composite nanofibers were prepared using electrospinning technique and characterized for in vitro drug release, antibacterial activity, laser doppler and in vivo wound healing. Results: The optimized nanofiber demonstrated an interconnected bead free nanofiber with average diameter <200 nm. The in vitro drug release & antimicrobial studies revealed that optimized nanofiber provided drug release for >120 h, thereby inhibiting growth of Escherichia coli and Stapyhlococcus aureus. An in vivo wound closure study on diabetic rats found that optimized nanofiber group had a significantly higher wound closure rate than marketed formulation. Conclusion: The nanofiber provided prolonged drug release and accelerated wound healing, making it a promising candidate for diabetic wound care.

Abstract:A novel bilayer structure composed of poly (Ɛ-caprolactone) (PCL) nanofibers deposited on a hydrogel layer of gelatin (GE) and hyaluronic acid (HA) was prepared via needleless electrospinning with anticipated drug delivery properties. Ibuprofen (IBU) was encapsulated into PCL nanofibers in two different concentrations, i.e., 5 wt% and 7 wt%. Quantification of incorporated IBU of drug-loaded nanofibers was performed by thermogravimetric analysis that proved its successful loading in the nanofibrous structure. The values of Young's modulus of GH hydrogel films showed appreciable mechanical strength. GH film exhibited a smaller contact angle than PCL nanofibers which advocated its hydrophilicity. Drug release profiles of IBU-loaded bilayer films were studied by UV–vis spectroscopy and a controlled release of IBU up to 45% during 48 h was recorded. The antibacterial analysis of these bilayer samples showed mild inhibition against E. coli bacterial strain in case of IBU-loaded nanofibers, whereas no inhibition against S. epidermidis was observed. Hence, all these findings supported the fact that the IBU-loaded bilayer samples with controlled drug release system and hydrophilicity could be a potential candidate for an effective wound dressing.
Preparation and characterization of diclofenac transdermal patch using ethyl cellulose and polyvinyl alcohol by electrospinning

Abstract:In recent years, the adverse effects linked to the oral administration of diclofenac, a non-steroidal anti-inflammatory drug (NSAID), have urged interest in exploring alternative administration routes, specifically transdermal drug delivery (TDD). Among the techniques emerging for creating nanoscale fibres suitable for TDD systems, electrospinning has attracted attention. The primary objective of this study was to formulate and evaluate nanofibers loaded with diclofenac diethylamine using ethylcellulose (EC) and polyvinyl alcohol (PVA) polymers. The influence of penetration enhancers, namely farnesol and nerolidol, on the properties of these fibres was investigated. Employing the electrospinning technique, nanofibers composed of EC or PVA polymers, with or without penetration enhancers, were fabricated, all maintaining a constant drug-to-polymer ratio. Mechanical strength and release kinetics from a cellophane membrane for the obtained nanofibers were investigated. A Franz diffusion cell was employed to determine the drug penetration rates through rat skin for selected fibre formulations. To gain insights into potential interactions and changes in the drug's solid state, FTIR spectroscopy, DSC, and XRD analyses were conducted. Microscopic examination unveiled that EC-based fibres exhibited homogeneity and lacked bead formations, albeit with larger diameters than other formulations. In contrast, both EC and PVA-based fibres displayed bead formations and nanometer-scale widths. The incorporation of penetration enhancers led to the production of uniform, bead-free nanofibers, with minor impacts on fiber size. EC fibres exhibited the lowest tensile strength and modulus of elasticity, but maximum flexibility. The addition of PVA and penetration enhancers contributed to enhanced fibre toughness and resistance to deformation. These alterations also positively influenced drug release rates, as observed in experiments conducted on cellophane membranes. Notably, when comparing drug release on rat skin to cellophane membranes, a slower initial release followed by a steeper slope was noted, attributed to the beneficial role of penetration enhancers on skin permeation. Analyzing results from DSC revealed the transition of the crystalline form of diclofenac diethylamine to an amorphous state within the nanofiber matrix which was also supported by XRD data. In light of these findings, electrospun nanofibers comprising a combination of EC and PVA polymers, alongside a penetration enhancer and diclofenac diethylamine, exhibit considerable promise for utilization as transdermal patches. Fibrous Guided Tissue Regeneration Membrane Loaded with Anti-Inflammatory Agent Prepared by Coaxial Electrospinning for the Purpose of Controlled Release

Abstract:Here, with the aim of inhibiting inflammation during guided tissue regeneration membrane (GTRM) implant surgery, coaxial electro spinning was used to fabricate drug-loaded core/sheath nanofiber GTRMs capable of controlled drug release. Various amounts of the anti-inflammatory agent metronidazole (MNA) were encapsulated into the core/sheath nanofibers (where PCL was the core, gelatin the sheath, and the gelatin shell was crosslinked with genipin) in order to establish the minimal drug content necessary to achieve the appropriate anti-inflammatory effect. By using TEM and SEM, the core/sheath structure was confirmed. In vitro drug disolution results showed that the core/sheath nanofibers exhibited sustained release profiles that were superior to those nanofibers produced by blending electrospinning. Additionally, the membrane significantly inhibited the colonization of anaerobic bacteria. Furthermore, with gelatin as a shell, the core/shell nanofiber membranes showed improved hydrophilicity, which resulted in better cell adhesion and proliferation without cytotoxicity. Therefore, in this study, a simple and effective coaxial electro spinning approach was demonstrated for the fabrication of anti-inflammatory GTRMs capable of providing controlled drug release. Abstract:The current study focuses on the development of gelatin-coated polycaprolactone (PCL) nanofibers co-loaded with luliconazole and naringenin for accelerated healing of infected diabetic wounds. Inherently, PCL nanofibers have excellent biocompatibility and biodegradation profiles but lack bioadhesion characteristics, which limits their use as dressing materials. So, coating them with a biocompatible and hydrophilic material like gelatin can improve bioadhesion. The preparation of nanofibers was done with the electrospinning technique. The solid state characterization and in-vitro performance assessment of nanofibers indicate the formation of uniformly interconnected nanofibers of 200–400 nm in diameter with smooth surface topography, excellent drug entrapment, and a surface pH of 5.6–6.8. The antifungal study showed that the nanofiber matrix exhibits excellent biofilm inhibition activity against several strains of Candida . Further, in-vivo assessment of nanofiber performance on C. albicans infected wounds in diabetic rats indicated accelerated wound healing efficacy in comparison to gauge-treated groups. Additionally, a higher blood flow and rapid re-epithelialization of wound tissue in the treatment group corroborated with the results obtained in the wound closure study. Overall, the developed dual-drug-loaded electrospun nanofiber mats have good compatibility, surface properties, and excellent wound healing potential, which can provide an extra edge in the management of complex diabetic wounds.

Cross-Linked Electrospun pH-Sensitive Nanofibers Adsorbed with Temporin-Ra for Promoting Wound Healing

Abstract:Bioresponsive nanodrug delivery systems have excellent potential in tissue engineering applications. Poly-anionic and poly-cationic biopolymers have provided a superior platform for designing pH-sensitive drug delivery systems. In this regard, hyaluronic acid-chitosan-polyvinyl alcohol complex nanofibers with high quality and reproducibility were produced by optimizing the solution preparation process. In addition, the synthesized composite nanofiber, with 66.82 kN/mm toughness, 200% swelling ratio, and 60% porosity, exhibited excellent properties to meet the requirements of the ideal wound dressing. Green cross-linking with citric acid prevented the destruction of the nanofiber even after prolonged immersion in biological solutions. ζ potential studies demonstrated that the synthesized nanofiber has a negative surface charge (∼-30) at physiological pH. The pKa of the temporin-Ra peptide is about 10, and as a result the peptide molecules have a net positive charge in physiological conditions. Therefore, peptide molecules immobilized on the synthesized scaffold based on surface adsorption. In vivo evaluation has proven that the wound bed has an alkaline environment, facilitating peptide release from the nanofiber scaffold. Electrospun nanofibers can imitate the architecture of the extracellular matrix for accelerating wound healing. In vitro investigation showed better adhesion, proliferation, migration, and fibroblast cell growth on peptide-loaded nanofiber samples than other groups. In vivo studies on full-thickness wounds in the mouse model indicated that the designed nanofiber was gradually absorbed without causing dryness or infection. On day 6, the peptide-loaded nanofiber revealed 60% wound closure compared to the control group (17%). In addition, based on histological studies, the composite nanofiber demonstrated excellent tissue repair ability, hence these active nanofiber mats can be a good alternative to existing wound dressings. Gene expression studies show that the antimicrobial peptide promotes the inflammatory phase of wound healing in a shorter time frame by accelerating the tumor necrosis factor-α cytokine response.

Abstract:The use of injectable materials is an attractive alternative for medical treatments because it does not require surgical intervention serving as a matrix that supports and entraps drugs, and the rheological characterization is necessary to ensure safe conditions of its application in an organism. Simvastatin (SIM) was chosen to compose the formulations due to its osteopromotive properties. Therefore, this study aimed to choose two formulations based on Pluronic F127, that is thermosensitive in aqueous solutions and SIM, one containing hyaluronic acid (HA) and the other sodium alginate (SA), and to verify the influence of different SIM concentrations on the rheology of hydrogels and their release behavior in the presence of these natural polymers. The methodology was based on rheological tests to modulate gelation, thermal analysis, hydrogel characterizations and in vitro release tests. Analysis of gelation temperatures revealed that adding SIM up to 5.0 wt % minimally affect the viscosity of the hydrogels, whereas by adding natural polymers increased the viscosity at 20 °C without affecting the viscosity at 37 °C. The compositions 18%F127/5%SIM/0.25%HA and 18%F127/5%SIM/0.5%SA were chosen as optimal. The drug release results showed that the addition of natural polymers delayed SIM delivery, with better results for the HA hydrogel. After 240 h, the HA hydrogel released 64% of SIM, whereas the SA hydrogel released 85%. The results showed that the presence of SIM does not interfere with the rheology of the hydrogels and that they exhibit modified drug-release kinetics, making them promising formulations for use as injectables. In response, the significance of combinational cancer therapy has been highlighted. Traditional combinational schemes usually utilize "free" drug for multi drug administration, independently. The diverse pharmacokinetics and biodistribution greatly hinder the antitumor effects and cause systematic toxicity. To tackle the hinderance, various nanoparticulate drug delivery systems (Nano-DDSs) have been developed. However, conventional Nano-DDSs encapsulate drugs into carrier materials through noncovalent interactions, resulting in low drug loading, fixed multi drug encapsulation ratio, chemical instability and carrier-associated toxicity. Recently, carrier-free nanocombos based on self-assembling small-molecule prodrugs (SPNCs) have emerged as a versatile Nano-DDSs for multiple drug delivery. Benefited by the self-assembly capability, SPNCs could be facilely fabricated with distinct merits of ultra-high drug loading, adjustable drug ratio and negligible carrier-associated toxicity. Herein, we summarize the latest trends of SPNCs. First, a basic review on self-assembling small-molecule prodrugs is presented. Additionally, facile techniques to prepare SPNCs are introduced. Furthermore, advanced combinational therapies based on SPNCs are spotlighted with special emphasis on synergistic mechanisms. Finally, future prospects and challenges are discussed. Wound healing is a common issue in our day to day life. Our immune system repairs the damaged tissue by itself and its a time-consuming process. The GO/CuO nanocomposite (NC) was synthesized through the sol-gel method. XRD, FT-IR, Raman, and TEM analysis were used to analysis the physico-chemical properties of the sample. The GO/CuO patches were prepared using chitosan (Cs)/poly vinyl alcohol (PVA) due to its biocompatibility and biodegradable nature. The obtained patches showed better antimicrobial and wound healing property than recently reported materials. The GO/CuO NC plays a major part in angiogenesis process and in the synthesis, stabilization of extracellular matrix skin proteins. Thus, GO/CuO NC enhance the wound healing mechanism by increasing cell proliferation, antimicrobial property and rapid initiation of inflammatory. Moreover, the antimicrobial activity of CuO, GO, GO/CuO and GO/CuO patch were tested against bacteria causing wound infections. Cs/PVA patch and Cs/PVA/GO/CuO patch were analyzed for swelling, evaporation and degradation behavior. Improved Analgesic and Anti-Inflammatory Effect of Diclofenac Sodium by Topical Nanoemulgel: Formulation Development—In Vitro and In Vivo Studies

Abstract:The present study aimed to develop diclofenac sodium nanoemulgel for managing pain and inflammation using the low-energy emulsification technique. Nanoemulsion of diclofenac was formulated using clove oil with adequate amount of surfactants and cosurfactants, and it was converted to hydrogel form using Carbopol 980 as the gelling agent. The droplet size of the oil globules in the nanoemulsion was found to be 64.07 ± 2.65 nm with a low polydispersity index (0.238 ± 0.02) along with high negative zeta potential (−39.06 mV). The developed nanoemulgel exhibited non-Newtonian and pseudoplastic behavior. The in vitro release profile of the developed nanoemulgel was higher as compared to marketed and conventional gel. The carrageenan-induced paw edema test was performed in rats to evaluate the anti-inflammatory activity of developed nanoemulgel. The developed nanoemulgel showed significantly higher ( ) effect in reducing pain and inflammation symptoms as compared to marketed as well as conventional gel of diclofenac. The overall findings of the study suggest that the developed nanoemulgel formulation of diclofenac can be used as a potential approach for the management of pain and inflammation.

Ibuprofen-Loaded Electrospun Fibrous Scaffold Doped with Sodium Bicarbonate for Responsively Inhibiting Inflammation and Promoting Muscle Wound Healing in Vivo

Abstract:Infection or inflammation can cause the in vivo microenvironment to become acidic, and an inappropriately excessive inflammatory response can significantly delay the healing process. A feasible acid-responsive ibuprofen-loaded poly(L-lactide) (PLLA) fibrous scaffold with doped sodium bicarbonate has been designed to prevent an excessive inflammatory response and promote regeneration. The results of the in vitro drug release study show that the acid-responsive fibrous scaffold exhibited a quick drug releasing response at pH 5.0 and a slow drug releasing response at pH 7.4. The in vivo rat muscle wound model study shows that the acid-responsive ibuprofen-loaded PLLA fibrous scaffold caused slighter inflammation and an earlier reparation. The immunohistochemical staining and quantitative real-time polymerase chain reaction analysis further showed that the scaffold caused lower levels of inflammatory factors and a higher expression of repair factors during the whole observation. Taken together, through the muscle wound healing process, the results demonstrate that the acid-responsive ibuprofen-loaded PLLA fibrous scaffold shows a better performance in preventing excessive inflammation than scaffolds that do not contain ibuprofen or are not acid-responsive. Additionally, the acid-responsive ibuprofen-loaded PLLA fibrous scaffold also has a faster healing process than others.
Curcumin‐loaded Poly(ε‐caprolactone) Nanofibres: Diabetic Wound Dressing with Anti‐oxidant and Anti‐inflammatory Properties

Abstract:1. Curcumin is a naturally occurring poly-phenolic compound with a broad range of favourable biological functions, including anti-cancer, anti-oxidant and anti-inflammatory activities. The low bioavailability and in vivo stability of curcumin require the development of suitable carrier vehicles to deliver the molecule in a sustained manner at therapeutic levels. 2. In the present study, we investigated the feasibility and potential of poly(caprolactone) (PCL) nanofibres as a delivery vehicle for curcumin for wound healing applications. By optimizing the electrospinning parameters, bead-free curcumin-loaded PCL nanofibres were developed. 3. The fibres showed sustained release of curcumin for 72 h and could be made to deliver a dose much lower than the reported cytotoxic concentration while remaining bioactive. Human foreskin fibroblast cells (HFF-1) showed more than 70% viability on curcumin-loaded nanofibres. 4. The anti-oxidant activity of curcumin-loaded nanofibres was demonstrated using an oxygen radical absorbance capacity (ORAC) assay and by the ability of the fibres to maintain the viability of HFF-1 cells under conditions of oxidative stress. 5. The curcumin-loaded nanofibres also reduced inflammatory induction, as evidenced by low levels of interleukin-6 release from mouse monocyte-macrophages seeded onto the fibres following stimulation by Escherichia coli-derived lipopolysaccharide. 6. The in vivo wound healing capability of the curcumin loaded PCL nanofibres was demonstrated by an increased rate of wound closure in a streptozotocin-induced diabetic mice model. 7. These results demonstrate that the curcumin-loaded PCL nanofibre matrix is bioactive and has potential as a wound dressing with anti-oxidant and anti-inflammatory properties.

Abstract:Serum T(3) (3,5,3' triiodothyronine) and serum T(4) (thyroxine) concentrations were repetitively assayed by radioimmunoassay over a three-year period in two male and two female immature captive whitetip reef sharks, Triaenodon obesus. These sharks were maintained at the Waikiki Aquarium, Honolulu, Hawaii, in an open system holding pool receiving 568 liters per minute of water from a saltwater well with an iodide concentration of 0.076 mg/liter. No significant male-female difference was observed for either serum T(3) or serum T(4). No seasonal pattern of serum T(3) was detected (P = 0.07). Serum T(3) concentrations ranged (mean +/- SEM) from 0. 52 to 0.83 ng/mL (0.67 +/- 0.01; n = 64). A significant seasonal difference was observed for serum T(4) (P < 0.001). Serum T(4) concentration was higher in winter (October-January) with a mean (range +/- SEM) of 6.58 ng/mL (1.48-8.77 +/- 0.35; n = 24) and lower in summer (May-August) with a mean of 3.62 ng/mL (1.34-5.71 +/- 0. 22; n = 24). Abstract:Pregabalin (PG) and diclofenac diethylamine (DEE) are anti-inflammatory molecules that are effective in relieving inflammation and pain associated with musculoskeletal disorders, arthritis, and post-traumatic pain, among others. Intravenous and oral delivery of these two molecules has their limitations. However, the transdermal route is believed to be an alternate viable option for the delivery of therapeutic molecules with desired physicochemical properties. To this end, it is vital to understand the physicochemical properties of these drugs, dosage, and strategies to enhance permeation, thereby surmounting the associated constraints and concurrently attaining a sustained release of these therapeutic molecules when administered in combination. The present work hypothesizes the enhanced permeation and sustained release of pregabalin and diclofenac diethylamine across the skin, entrapped in the adhesive nano-organogel formulation, including permeation enhancers. The solubility studies of pregabalin and diclofenac diethylamine in combination were performed in different permeation enhancers. Oleic acid was optimized as the best permeation enhancer based on in vitro studies. Pluronic organogel containing pregabalin and diclofenac diethylamine with oleic acid was fabricated. Duro-Tak® (87-2196) was added to the organogel formulation as a pressure-sensitive adhesive to sustain the release profile of these two therapeutic molecules. The adhesive organogel was characterized for particle size, scanning electron microscopy, and contact angle measurement. The HPLC method developed for the quantification of the dual drug showed a retention time of 3.84 minutes and 9.69 minutes for pregabalin and diclofenac, respectively. The fabricated nanogel adhesive formulation showed the desired results with particle size and contact angle of 282 ± 57 nm and ≥1200, respectively. In vitro studies showed the percentage cumulative release of 24.90 ± 4.65% and 33.29 ± 4.81% for pregabalin and diclofenac, respectively. In order to accomplish transdermal permeation, the suggested hypothesis of fabricating PG and DEE nano-organogel in combination with permeation enhancers will be a viable drug delivery method. In comparison to a traditional gel formulation, oleic acid as a permeation enhancer increased the penetration of both PG and DEE from the organogel formulation. Notably, the studies showed that the use of pressure-sensitive adhesives enabled the sustained release of both PG and DEE.Therefore, the results anticipated the hypothesis that the transdermal delivery of adhesive PG and DEEbased nanogel across the human skin can be achieved to inhibit inflammation and pain.

Abstract:The aim of the current study was to fabricate naturally derived polymer based hydrogels for controlled release of diclofenac sodium (DS) for a long duration of time. In this research work, sodium alginate-co-poly(2-acrylamido-2-methyl propane sulphonic acid) (SA-co-poly(AMPS)) hydrogels were prepared by the free radical polymerization technique, where sodium alginate (SA) and 2-acrylamido-2-methyl propane sulphonic acid (AMPS) were used as the polymer and monomer while ammonium peroxodisulfate (APS) and N,N′-Methylene bisacrylamide (MBA) were used as the initiator and cross-linker, respectively. A swelling study was performed to determine the swelling index of developed hydrogels in both acidic (pH 1.2) and basic (pH 7.4) media and pH-independent swelling was observed due to the presence of AMPS. An in vitro release study was conducted to evaluate the percentage of drug released, and a high release of the drug was found at the higher pH of 7.4. Sol–gel analysis was performed to analyze the crosslinked and uncrosslinked part of the hydrogels, and results showed a rise in gel fraction as the composition of SA, AMPS and MBA increased while the sol fraction decreased and vice versa. This work demonstrated a potential for sustained delivery of diclofenac sodium by employing various concentration of SA, AMPS and MBA.
