Abstract:Background: Breast cancer is the most common cancer in women globally, and diagnosing it early and finding potential drug candidates against multi-drug resistant metastatic breast cancers provide the possibilities of better treatment and extending life. Methods: The current study aimed to evaluate the synergistic anti-metastatic activity of Curcumin (Cur) and Paclitaxel (Pacli) individually, the combination of Curcumin-Paclitaxel (CP), and also in conjugation with gold nanoparticles (AuNP-Curcumin (Au-C), AuNP-Paclitaxel (Au-P), and AuNP-Curcumin-Paclitaxel (Au-CP)) in various in vitro and in vivo models. Results: The results from combination treatments of CP and Au-CP demonstrated excellent synergistic cytotoxic effects in triple-negative breast cancer cell lines (MDA MB 231 and 4T1) in in vitro and in vivo mouse models. Detailed mechanistic studies were performed that reveal that the anti-cancer effects were associated with the downregulation of the expression of VEGF, CYCLIN-D1, and STAT-3 genes and upregulation of the apoptotic Caspase-9 gene. The group of mice that received CP combination therapy (with and without gold nanoparticles) showed a significant reduction in the size of tumor when compared to the Pacli alone treatment and control groups. Conclusions: Together, the results suggest that the delivery of gold conjugated Au-CP formulations may help in modulating the outcomes of chemotherapy. The present study is well supported with observations from cell-based assays, molecular and histopathological analyses.

Abstract:Phytomedicines are potential immunity-boosting components with effective anticystic properties, minimal side effects, and biomedical applications, making them valuable for combating various diseases. India is renowned globally for Ayurveda, an ancient treatment methodology known for its holistic approach in identifying the root cause of diseases. Tulsi (Ocimum sanctum) is a common household medicine in India. While essential oils from plants like Tulsi have long been recognized for their medicinal properties, there is a gap in understanding their potential in synthesizing gold nanoparticles (AuNPs) and their efficacy against breast carcinoma, particularly in the context of immunosuppressive conditions. We investigated the potential application of essential oils isolated from O. sanctum in the synthesis of AuNPs and their efficacy against MCF-7 breast carcinoma. Gas chromatography-mass spectroscopy identified compounds with potential anticancer effects against breast cancer cells. Synthesised AuNPs displayed high hemocompatibility and antimicrobial activity against nosocomial Pseudomonas aeruginosa, Escherichia coli, Vibrio cholerae, and Bacillus subtilis strains. Os-AuNPs induced chromosomal instability and mitotic arrest in the G2/M cell cycle phase. Subsequent fluorescence and cell cytometry studies demonstrated the systemic release of ROS, depolarisation of mitochondrial membrane potential, and production of apoptotic bodies. DNA damage and comet assays confirmed the anticancer potential of synthesised AuNPs. This study illuminates the potential of O. sanctum-derived AuNPs in breast carcinoma treatment, paving the way for future AuNP-based therapies in biomedicine.
Curcumin inhibits autocrine growth hormone-mediated invasion and metastasis by targeting NF-κB signaling and polyamine metabolism in breast cancer cells

Abstract:Curcumin is assumed to be a plant-derived therapeutic drug that triggers apoptotic cell death in vitro and in vivo by affecting different molecular targets such as NF-κB. Phase I/II trial of curcumin alone or with chemotherapeutic drugs has been accomplished in pancreatic, colon, prostate and breast cancer cases. Recently, autocrine growth hormone (GH) signaling-induced cell growth, metastasis and drug resistance have been demonstrated in breast cancer. In this study, our aim was to investigate the potential therapeutic effect of curcumin by evaluating the molecular machinery of curcumin-triggered apoptotic cell death via focusing on NF-κB signaling and polyamine (PA) metabolism in autocrine GH-expressing MCF-7, MDA-MB-453 and MDA-MB-231 breast cancer cells. For this purpose, a pcDNA3.1 (+) vector with a GH gene insert was transfected by a liposomal agent in all breast cancer cells and then selection was conducted in neomycin (G418) included media. Autocrine GH-induced curcumin resistance was overcome in a dose-dependent manner and curcumin inhibited cell proliferation, invasion–metastasis and phosphorylation of p65 (Ser536), and thereby partly prevented its DNA binding activity in breast cancer cells. Moreover, curcumin induced caspase-mediated apoptotic cell death by activating the PA catabolic enzyme expressions, which led to generation of toxic by-products such as H2O2 in MCF-7, MDA-MB-453 and MDA-MB-231 GH+ breast cancer cells. In addition, transient silencing of SSAT prevented curcumin-induced cell viability loss and apoptotic cell death in each breast cancer cells. In conclusion, curcumin could overcome the GH-mediated resistant phenotype via modulating cell survival, death-related signaling routes and activating PA catabolic pathway.

Abstract:Cancer, a prevalent disease across various societies, presents a significant challenge in treatment research. Studies show that combination therapies are one of the methods that can help in the effective treatment of cancer. Chemotherapy and radiation therapy are among the main cancer treatments and in this project, for combined chemoradiotherapy treatment, carbon nanotubes were used as improved carriers of chemotherapy in tumors, as well as a substrate for the preparation of radiation sensitizers for local radiation therapy. Following the synthesis of CNT-Platinum-Curcumin nanoparticles (CNT-Pt-CUR), a series of analyses were conducted to verify the successful production of these nanoparticles. Techniques such as Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS), UV-Vis spectroscopy, Fourier Transform Infrared Spectroscopy (FTIR), and X-Ray Diffraction (XRD) were employed. The characterization data revealed a spherical shape Pt nanoparticle morphology with an 8.5 nm diameter on rod-shape CNT, as observed through TEM. Furthermore, FTIR analysis confirmed the successful loaded of the drug into the nanoparticles, highlighting the potential of this approach in cancer treatment. Then, hemolysis and (3(-4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests on normal cells were used to assess the biocompatibility of CNT-Pt-CUR nanoparticles. It also explored the anticancer efficacy of these nanoparticles at varying concentrations against cancer cells, both with and without exposure to X-rays. The research confirmed the successful synthesis of these nanoparticles and demonstrated their potential impact on cell viability. Specifically, breast cancer cells exhibited heightened susceptibility to toxicity when exposed to nanoparticles and X-rays. Further analysis revealed that the toxicity of nanoparticles is dose-dependent, and modifying the surface of carbon nanotube (CNT) nanoparticles with CUR significantly reduced blood toxicity. Interestingly, nanoparticle toxicity was significantly amplified in the presence of X-rays, suggesting mechanisms such as DNA damage and increased reactive oxygen species (ROS) levels within cells.
Risk factors for drug diversion in a pain clinic patient population.

Abstract:BACKGROUND Prescription drug abuse and drug diversion have soared at the same time prescribing practices have increased, prompting the pain medicine community to seek more accurate knowledge of the sources of prescription drugs used non medically. OBJECTIVE To determine the extent of drug diversion and its risk factors in patients treated in a pain clinic. METHODS An anonymous patient survey was developed based on factors examined for their association with diversion identified from a literature search. Patients were asked to participate as they arrived for appointments at six chronic pain management clinics. Questions ascertained whether patients had experienced drug loss, theft, or diversion and, if so, the number of episodes. Data were analyzed to determine associations between identified risk factors and the following four identified ways of diverting prescription medications: stolen medications, lost medications, sharing of medications, and selling of medications. Data were also analyzed to determine the extent of drug diversion within the studied population. RESULTS Respondents had experienced a 45 percent incidence of some form of drug diversion on at least one occasion. The most common type of drug diversion was loss due to theft, reported by 30 percent of the respondents. Findings suggest family history of drug abuse and a history of criminal behavior can increase likelihood of drug diversion. CONCLUSIONS Drug diversion in a pain clinic population was common in this brief survey. Such diversion may contribute to the problems of nonmedical use of prescription drugs.

Phytocompounds-based therapeutic approach: Investigating curcumin and green tea extracts on MCF-7 breast cancer cell line

Abstract:Background: Breast cancer (BC) has transcended lung cancer as the most common cancer in the world. Due to the disease's aggressiveness, rapid growth, and heterogeneity, it is crucial to investigate different therapeutic approaches for treatment. According to the World Health Organization (WHO), Plant-based therapeutics continue to be utilized as safe/non-toxic complementary or alternative treatments for cancer, even in developed countries, regardless of how cutting-edge conventional therapies are. Despite their low bioavailability, curcumin (CUR) and green tea (GT) represent safer therapeutic options. Due to their potent molecular-modulating properties on various cancer-related molecules and signaling pathways, they are considered gold-standard therapeutic agents and have been incorporated into the development of one or more therapeutic strategies of BC treatment. Methods: We investigated the modulatory role of CUR and GT extracts on significant multi molecular targets in MCF-7 BC cell line to assess their potential as BC multi-targeting agents. We analyzed the phytocompounds in GT leaves using High-performance liquid chromatography (HPLC) and Gas chromatography-mass spectrometry (GC-MS) techniques. The mRNA expression levels of Raf-1, Telomerase, Tumor necrosis factor alpha (TNF-α) and Interleukin-8 (IL-8) genes in MCF-7 cells were quantified using quantitative real-time PCR (qRT-PCR). The cytotoxicity of the extracts was assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and the released Lactate dehydrogenase (LDH), a valuable marker for identifying the programmed necrosis (necroptosis). Additionally, the concentrations of the necroptosis-related proinflammatory cytokines (TNF-α and IL-8) were measured using enzyme-linked immunosorbent assay (ELISA). Results: In contrast to the GT, the results showed the anticancer and cytotoxic properties of CUR against MCF-7 cells, with a relatively higher level of released LDH. The CUR extract downregulated the oncogenic Raf-1, suppressed the Telomerase and upregulated the TNF-α and IL-8 genes. Results from the ELISA showed a notable increase in IL-8 and TNF-α cytokines levels after CUR treatment, which culminated after 72 h. Conclusions: Among both extracts, only CUR effectively modulated the understudy molecular targets, achieving multi-targeting anticancer activity against MCF-7 cells. Moreover, the applied dosage significantly increased levels of the proinflammatory cytokines, which represent a component of the cytokines-targeting-based therapeutic strategy. However, further investigations are recommended to validate this therapeutic approach.

Abstract:This abstract explores the potential of curcuma as both a drug delivery method and an anticancer treatment. Curcuma, a natural compound derived from turmeric, has gained attention for its bioactive properties. As a drug delivery method, curcuma’s lipophilic nature allows it to encapsulate various therapeutic agents, enhancing their solubility and bioavailability. Moreover, curcuma’s inherent anti-inflammatory and antioxidant characteristics contribute to its potential as an effective anticancer treatment. This abstract reviews recent research on curcuma-based drug delivery systems and its mechanisms of action against cancer cells, highlighting its promising role in advancing drug delivery strategies and cancer therapies. Certainly, in recent years, researchers have focused on harnessing the unique properties of curcuma for drug delivery purposes. Its ability to form stable complexes with hydrophobic drugs, along with its biocompatibility, makes it an attractive candidate for improving the delivery of poorly water-soluble drugs. By encapsulating these drugs within curcuma-based nanoparticles or micelles, researchers aim to enhance drug stability and release kinetics, ultimately leading to more effective treatments with reduced side effects. Additionally, curcuma’s potential as an anticancer treatment has been extensively investigated. Its active compound, curcumin, exhibits multifaceted effects on various cancer cell types. Curcumin has been shown to modulate signaling pathways involved in cell proliferation, apoptosis, and angiogenesis. Moreover, curcumin’s ability to target tumor cells while sparing normal cells makes it a promising candidate for combination therapies that enhance the effectiveness of traditional chemotherapy agents.Clinical studies have explored curcuma’s role in reducing cancer growth, metastasis, and even improving patients’ response to chemotherapy. However, challenges such as low bioavailability and rapid metabolism of curcumin have prompted innovative approaches, including nanoparticle formulations and co-administration with absorption-enhancing agents .In conclusion, the combined potential of curcuma as a drug delivery method and an anticancer treatment opens up exciting possibilities for the future of pharmaceutical research. By addressing its limitations and further exploring its mechanisms of action, curcuma-based therapies could revolutionize drug delivery strategies and contribute to more targeted and effective cancer treatments.

Abstract:Triple-negative breast cancer (TNBC) accounts for 15% of all breast cancers and is highly aggressive. Despite an initial positive response to chemotherapy, most patients experience rapid disease progression leading to relapse and metastasis. This is attributed to the presence of breast cancer stem cells (BCSCs) within the tumor, which are characterized by self-renewal, pluripotency, and resistance mechanisms. Targeting BCSCs has become critical as conventional therapies fail to eradicate them due to a lack of specific targets. Curcumin, a polyphenol derived from turmeric (Curcuma longa), exhibits anticancer effects against breast cancer cells and BCSCs. The use of curcumin derivatives has been suggested as an approach to overcome the bioavailability and solubility problems of curcumin in humans, thereby increasing its anticancer effects. The aim of this study was to evaluate the cellular and molecular effects of six synthetic compounds derived from the natural polyphenol epigallocatechin gallate (EGCG) (TL1, TL2) and curcumin derivatives (TL3, TL4, TL5, and TL6) on a TNBC mesenchymal stem-like cell line. The activity of the compounds against BCSCs was also determined by a mammosphere inhibition assay and studying different BCSC markers by Western blotting. Finally, a drug combination assay was performed with the most promising compounds to evaluate their potential synergistic effects with the chemotherapeutic agents doxorubicin, cisplatin, and paclitaxel. The results showed that compounds exhibited specific cytotoxicity against the TNBC cell line and BCSCs. Interestingly, the combination of the curcumin derivative TL3 with doxorubicin and cisplatin displayed a synergistic effect in TNBC cells.

Abstract:Binding sites for angiotensin II (Ang II) and angiotensin converting enzyme (ACE) were studied by autoradiography in human, rat and bovine adrenals and human pheochromocytomas. In human and rat adrenals, binding sites for both Ang II and ACE were found in the medulla and zona glomerulosa. In bovine adrenals, ACE binding sites were not detectable, while Ang II binding was higher in the zona glomerulosa, lower in the rest of the cortex and undetectable in the medulla. In pheochromocytomas, ACE binding was homogeneously distributed, but no Ang II binding was detected. These results suggest that circulating and/or locally formed Ang II could regulate adrenomedullary and zona glomerulosa functions in man, and show that chromaffin cells in tumors retain ACE binding sites but lose Ang II binding sites. Thus, there are alterations in the regulation of the renin-angiotensin system in chromaffin tumors.

Abstract:Pancreatic cancer is characterized by desmoplasia; crosstalk between pancreatic cancer cells (PCCs) and pancreatic stellate cells (PSCs) leads to the deposition of extracellular matrix proteins in the tumor environment resulting in poor vascularity. Targeting either PCCs or PSCs individually has produced mixed results, and there is currently no effective strategy to target both cell types simultaneously. Previously, we demonstrated, through in vitro cell culture experiments, that a specific gold nanoparticle-based nanoformulation containing the anti-EGFR antibody cetuximab (C225) as a targeting agent and gemcitabine as a chemotherapeutic agent effectively targets both PCCs and PSCs simultaneously. Herein, we extend our studies to test the ability of these in vitro tested nano formulations to inhibit tumor growth in an orthotopic co-implantation model of pancreatic cancer in vivo . Orthotopic tumors were established by co-implantation of equal numbers of PCCs and PSCs in the mouse pancreas. Among the various formulations tested, 5 nm gold nanoparticles coated with gemcitabine, cetuximab and poly-ethylene glycol (PEG) of molecular weight 1000 Da, which we named ACGP441000, demonstrated optimal efficacy in inhibiting tumor growth. The current study reveals an opportunity to target PCCs and PSCs simultaneously, by exploiting their overexpression of EGFR as a target, in order to inhibit pancreatic cancer growth.

Abstract:This paper presents the first phase shifter based on sol-gel Barium Calcium Zirconate Titanate (BCTZ) thin ferroelectric film. It is a coplanar waveguide distributed phase shifter with BCTZ varactors used as tuning elements. It presents a continuous phase shift of 285° at 12 GHz (180° at 8 GHz) with a 20 V bias voltage. Its figure of merit is 26°/dB and its return loss is better than 12 dB for the 8 to 12 GHz frequency bandwidth. The presented phase shifter is the only phase shifter based on low cost sol-gel deposition to compete with the best reported X band ferroelectric phase shifters.

Abstract:Gold nanoparticles (GNPs) have promising properties such as photothermal effects and could be useful for imaging and as multifunctional nanocarriers for various drugs. In this study, we synthesized polyethyleneglycol (PEG)-grafted GNPs and conjugated them with cyclodextrin (CD) to incorporate curcumin. Curcumin has anticancer effects but its therapeutic application is limited due to poor water solubility. Three types of CDs (α-, β-, and γ-CDs) were conjugated with PEGylated GNPs and the curcumin-containing CD/PEG-conjugated GNPs (cur-CD-GNPs) were characterized. Transmission electron microscopy and dynamic light scattering results showed that these cur-CD-GNPs have a small gold nanocore (approximately 5 nm) and the average size of the three cur-CD-GNPs was approximately 25-35 nm. Curcumin was efficiently incorporated into the β-CD solution and the loading efficiency of curcumin in β-CD-GNPs was the highest of the three types of CD-GNPs prepared. The cytotoxic effect of cur-CD-GNPs was investigated using a human lung cancer cell line. All cur-CD-GNPs exhibited cytotoxic effects comparable to that of curcumin solution and CD-GNPs without curcumin were not cytotoxic. These results suggest that cur-CD-GNPs may be a useful multifunctional nanomedicine, although in vivo investigations are required.

Abstract:A 70-year-old Caucasian patient is described, who presented with a bullous dermatosis on an amputation stump. The bullous lesions had developed after practice with a Kondylen-Bettung-Münster (KBM) prosthesis. The lesions disappeared completely after prednisone therapy and replacement of the prosthesis with a Thomas splint. The patient has remained free of symptoms ever since, even though prednisone was completely withdrawn after 9 months. According to skin immunofluorescence criteria, the patient suffered from pemphigoid, rather than epidermolysis bullosa acquisita as strongly suggested by the anamnesis. Synthesis, characterization and in vitro assessment of colloidal gold nanoparticles of Gemcitabine with natural polysaccharides for treatment of breast cancer

Abstract:Targeted drug delivery plays an essential role in the treatment of breast cancer because it provides therapeutic responses with low side effects for an extended period. In the present study, gemcitabine hydrochloride (GEM)-loaded colloidal gold nanoparticles have been successfully developed using Gum acacia (GA), (GEM-GA-AuNPs) as a polysaccharides based system. The gold nanoparticles were optimized using concentration of GA from (0.25%–3% w/v) containing 10 mM of Gold (III) chloride trihydrate (HAuCl4·3H2O) with optimum temperature of 80 ± 1 °C and reaction time of 30 min. The ruby-red (1% w/v) GA-AuNP was obtained and further characterized by Dynamic Light Scattering (DLS), Field Emission Scanning Electron Microscopy (FE-SEM), High-resolution transmission electron microscopy (HR-TEM), Selected area (electron) diffraction (SAED), Ultraviolet–visible spectroscopy, Fourier-transform infrared spectroscopy (FT-IR), powder X-ray diffraction (P-XRD), Differential scanning calorimetry (DSC) and Thermogravimetric Analysis (TGA). The blank GA -AuNPs (1% w/v) demonstrated the hydrodynamic diameter of 78.15 ± 2.12 nm with a zeta potential of -23.6 ± 1.21 mV. In cytotoxicity studies, compared with free GEM, GEM-GA -AuNPs displayed more cytotoxicity in human breast adenocarcinoma (MDA-MB-231) cell line. Cellular Effect of Curcumin and Citral Combination on Breast Cancer Cells: Induction of Apoptosis and Cell Cycle Arrest

Abstract:PURPOSE: The unmanageable side effects caused by current chemotherapy regimens to treat cancer are an unresolved problem. Although many phytonutrients are useful as chemoprevention without side effects, their effects are slower and smaller than conventional chemotherapy. In the present work, we examined the cumulative effect of two phytonutrients, curcumin and citral, on breast cancer cell lines and compared their effect with the known chemotherapy regimen of cyclophosphamide, methotrexate, and 5-fluorouracil.METHODS: Using cultured breast cancer and normal epithelial cells, the cytotoxic and apoptotic effect of curcumin and citral was evaluated in vitro. The synergistic effect of curcumin and citral was calculated by a combination index study using the method by Chou and Talalay. Cell death pathways and mechanisms were analyzed by measuring intracellular reactive oxygen species (ROS) and apoptotic protein levels.RESULTS: Curcumin and citral caused dose and time dependent cell death and showed a synergistic effect at effective concentration EC50 and above concentrations in breast cancer cells without disturbing normal breast epithelial cells. With combination curcumin and citral treatment, apoptosis induction and cell cycle arrest at G0/G1 phase in breast cancer cells were observed. Curcumin and citral generated ROS and activated p53 and poly (ADP-ribose) polymerase-1 mediated apoptotic pathways.CONCLUSION: The results of this study suggest that curcumin and citral in combination may be a useful therapeutic intervention for breast cancer.
