Abstract:Iron dextran is a common anti-anemia drug, and it requires low molar mass dextran as substrate. In this work, we selected 11 amino acid residues in domain A/B of DSR-MΔ2 within a 5-angstrom distance from sucrose for site-directed mutagenesis by molecular docking. Mutation of Q634 did not affect the enzyme catalytic activity, but showed an obvious impact on the ratio of low molecular weight dextran (L-dextran, 3,000–5,000 Da) and relatively higher molecular weight dextran (H-dextran, around 10,000 Da). L-dextran was the main product synthesized by DSR-MΔ2 Q634A, and its average molecular weight was 3,951 Da with a polydispersity index <1.3. The structural characterization of this homopolysaccharide revealed that it was a dextran, with 86.0% α(1→6) and 14.0% α(1→4) glycosidic linkages. Moreover, L-dextran was oxidized with NaOH and chelated with ferric trichloride, and an OL-dextran-iron complex was synthesized with a high iron-loading potential of 33.5% (w/w). Altogether, mutation of amino acids near the sucrose binding site of dextransucrase can affect the chain elongation process, making it possible to modulate dextran size.

Abstract:Dextransucrases play a crucial role in the production of dextran from economical sucrose; therefore, there is a pressing demand to explore novel dextransucrases with better performance. This study characterized a dextransucrase enzyme, LmDexA, which was identified from the Leuconostoc mesenteroides NN710. This bacterium was isolated from the soil of growing dragon fruit in Guangxi province, China. We successfully constructed six different N-terminal truncated variants through sequential analysis. Additionally, a truncated variant, ΔN190LmDexA, was constructed by removing the 190 amino acids fragment from the N-terminal. This truncated variant was then successfully expressed heterologously in Escherichia coli and purified. The purified ΔN190LmDexA demonstrated optimal hydrolysis activity at a pH of 5.6 and a temperature of 30 °C. Its maximum specific activity was measured to be 126.13 U/mg, with a Km of 13.7 mM. Results demonstrated a significant improvement in the heterologous expression level and total enzyme activity of ΔN190LmDexA. ΔN190LmDexA exhibited both hydrolytic and transsaccharolytic enzymatic activities. When sucrose was used as the substrate, it primarily produced high-molecular-weight dextran (>400 kDa). However, upon the addition of maltose as a receptor, it resulted in the production of a significant amount of oligosaccharides. Our results can provide valuable information for enhancing the characteristics of recombinant dextransucrase and potentially converting sucrose into high-value-added dextran and oligosaccharides.
A Formal Model Conversion Approach to Developing a DEVS-Based Factory Simulator

Abstract:This article presents a model conversion approach to developing a 3D discrete event system specification (DEVS)-based factory simulator, in which the target automated manufacturing systems (AMS) are modeled by employing a job-resource relation network (JR-net), and this input JR-net model is systematically converted to a DEVS model. For this purpose, the JR-net modeling framework has been extended further to handle 3D geometric information while providing a formal specification of the JR-net. Also, a hierarchical DEVS model architecture and a systematic conversion method for converting a JR-net model into a DEVS model are proposed. Based on the proposed approach, a 3D factory simulator has been developed in C++ with OpenGL and a DEVS engine. The proposed factory simulator may be used as a virtual prototyping tool for designing new AMS. Its validity is demonstrated by constructing a virtual factory of a special type of AMS called a flexible manufacturing system.
Functional Identification of the Dextransucrase Gene of Leuconostoc mesenteroides DRP105

Abstract:Leuconostoc mesenteroides DRP105 isolated from Chinese sauerkraut juice is an intensive producer of dextran. We report the complete genome sequence of Leu. mesenteroides DRP105. This strain contains a dextransucrase gene (dsr) involved in the production of dextran, possibly composed of glucose monomers. To explore the dextran synthesis mechanism of Leu. mesenteroides DRP105, we constructed a dsr-deficient strain derived from Leu. mesenteroides DRP105 using the Cre-loxP recombination system. The secondary structure prediction results showed that Leu. mesenteroides DRP105 dextransucrase (Dsr) was coded by dsr and contained 17.07% α-helices, 29.55% β-sheets, 10.18% β-turns, and 43.20% random coils. We also analyzed the dextran yield, monosaccharide change, organic acid, and amino-acid content of Leu. mesenteroides DRP105 and Leu. mesenteroides DRP105-Δdsr. The result showed that the lack of dsr changed the Leu. mesenteroides DRP105 sugar metabolism pathway, which in turn affected the production of metabolites.

Abstract:tfxa (GenBank: U01242), which encoded the mature peptide of Thermomonospora fusca TF xylanase A (TfxA), was high-level expressed in Escherichia coli BL21 (DE3). The recombinant xylanase, reETfxA, was both secreted into culture medium and in the cytoplasm. reETfxA showed high xylanase activity and was purified to homogeneity by Ni-affinity resin (182.8 and 681.4 U mg−1 for crude and purified enzyme, respectively). Sodium dodecyl sulphate–polyacrylamide gel electrophoresis and Western blot analysis revealed that the molecular mass of reETfxA was approximately 42.7 kDa. The reETfxA was immobilized on a novel mesoporous SiO2 microsphere (MSM)-coated chitosan via covalent bonds, which formed from Schiff base reaction between supports and enzyme. The binding capacity of the prepared MSM-coated chitosan particles to reETfxA was approximately 272.6 mg g−1-particles. The optimum temperature values of the free and immobilized reETfxA were 65 and 70 °C, respectively. Moreover, the optimum pH values of the free and immobilized reETfxA were pH 6.0 and 5.0, respectively. The reETfxA showed relatively high thermostability. When treated at 70 °C and pH 6.0 for 15 min, the residual activities of free and immobilized reETfxA were 84.2 and 100.2%, respectively. The results obtained from the HPLC analysis showed that immobilized reETfxA released xylooligosaccharides from oat spelt, beechwood and birchwood xylans, with xylotetraose, xylotriose, and xylopentaose as the major products, respectively. Additionally, the immobilized reETfxA could directly hydrolyze the wheat bran insoluble xylan.

Abstract:Enzyme membrane reactor (EMR) provides an effective strategy for controlling molecular weight (Mw) of oligodextrans. However, the synergistic effect between dextran hydrolysis and product separation as well as continuous operation stability remains still unclear. We observed that covalently immobilized dextranase exhibited more obvious exo -hydrolysis behavior compared to free enzymes probably due to enzyme conformational alterations. Using non-covalently immobilized enzyme, suitable ultrafiltration membrane, optimal substrate/enzyme ratio and matched permeate flux, the yield of the target product reached 62.8%. However, the non-covalently immobilized dextranase exhibited reduced hydrolytic activity and more obvious membrane fouling (caused by high-Mw dextran). When free dextranase with high activity was used together with suitable permeate flux and high shear rate, the yield increased to 72.0%. The obtained EMR maintained stable for 8 h with slight fouling formation because enzymatic hydrolysis and membrane separation were well matched. The polydispersity index of the target product reached 1.3 (analytical grade) following diafiltration purification.

Abstract:Abstract Dextransucrase (DSRS) from Leuconostoc mesenteroides NRRL B-512F is a glucosyltransferase that catalyzes the synthesis of soluble dextran from sucrose or oligosaccharides when acceptor molecules, like maltose, are present. The L. mesenteroides NRRL B-512F dextransucrase-encoding gene (dsrS) was amplified by the polymerase chain reaction and cloned in an overexpression plasmid. The characteristics of DSRS were found to be similar to the characteristics of the extracellular dextransucrase produced by L. mesenteroides NRRL B-512F. The enzyme also exhibited a high homology with other glucosyltransferases. In order to identify critical amino acid residues, the DSRS sequence was aligned with glucosyltransferase sequences and four amino acid residues were selected for site- directed mutagenesis experiments: aspartic acid 511, aspartic acid 513, aspartic acid 551 and histidine 661. Asp-511, Asp-513 and Asp-551 were independently replaced with asparagine and His-661 with arginine. Mutation at Asp-511 and Asp-551 completely suppressed dextran and oligosaccharide synthesis activities, showing that at least two carboxyl groups (Asp-511 and Asp-551) are essential for the catalysis process. However, glucan-binding properties were retained, showing that DSRS has a two-domain structure like other glucosyltransferases. Mutations at Asp-513 and His-661 resulted in greatly reduced dextransucrase activity. According to amino acid sequence alignments of glucosyltransferases, α-amylases or cyclodextrin glucanotransferases, His-661 may have a hydrogen-bonding function.

Abstract:Dextran, a microbial metabolite of diverse molecular configurations, can be biosynthesized employing selected strains of characterized species of bacteria. Dextran molecules are secreted as an extracellular polysaccharide in the culture medium of the bacterial fermentation system. This microbially produced polymer of glucose possesses multi-faceted characteristics such as its solubility in different solvents and formation of dextran solutions of needed viscosity. Several preparations can be formulated for the desired thermal and rheological properties. Due to such multifunctional characteristics, dextran with different structural specifications is a desired polysaccharide for clinical, pharmaceutical, and food industry commercial applications. Dextran and its derivative products with various molecular weights, in a range of high and low, have established their uses in drug delivery and in analytical devices using columns packed with polysaccharide gel. Therefore, being a neutral raw material, the resourcefulness of dextran preparations of different molecular weights and linkages in their polymer configuration is important. For this purpose, several studies have been performed to produce this commercially important polysaccharide under optimized bacterial cultivation processes. This article aims to overview recently published research reports on some significant applications of dextran in the pharmaceutical and food industries. Studies conducted under optimized conditions in fermentation processes for the biosynthesis of dextran of diverse molecular configurations, which are responsible for its multifunctional properties, have been summarized. Concise information has been presented in three separate tables for each group of specific bacterial species employed to obtain this extracellular microbial polysaccharide.

Optimizing Curdlan Synthesis: Engineering Agrobacterium tumefaciens ATCC31749 for Enhanced Production Using Dextrin as a Carbon Source

Abstract:A key goal in current research on industrial curdlan production is the expansion of carbon sources for fermentation. In this study, a recombinant bacterial strain, sp-AmyAXCC, capable of fermenting and synthesizing curdlan using dextrin as a carbon source, was produced via heterologous expression of IPTG-inducible α-amylase from Xanthomonas campestris NRRL B-1459 in Agrobacterium tumefaciens ATCC31749. External expression of the enzyme was confirmed by western blotting, and the expression levels of exogenous proteins during the fermentation process were monitored. Additionally, the properties of the curdlan product were characterized using attenuated total reflectance-Fourier transform infrared spectroscopy and X-ray diffraction. The recombinant strain produced curdlan at a titer of 30.40 ± 0.14 g/L, gel strength of 703.5 ± 34.2 g/cm2, and a molecular weight of 3.58 × 106 Da, which is 33% greater than the molecular weight of native curdlan (2.69 × 106 Da). In the batch fermentation of sp-AmyAXCC with 12% dextrin as a carbon source, the titer of curdlan was 66.7 g/L with a yield of 0.56 g/g, and a productivity rate of 0.62 g/L/h at 108 h. The results of this study expand the substrate spectrum for Agrobacterium fermentation in curdlan production and provides guidance for further industrialization of curdlan production.

Abstract:Abstract Background Lactic acid bacteria can synthesize dextran and oligosaccharides with different functionality, depending on the strain and fermentation conditions. As natural structure-forming agent, dextran has proven useful as food additive, improving the properties of several raw materials with poor technological quality, such as cereal by-products, fiber-and protein-rich matrices, enabling their use in food applications. In this study, we assessed dextran biosynthesis in situ during fermentation of brewers ́ spent grain (BSG), the main by-product of beer brewing industry, with Leuconostoc pseudomesenteroides DSM20193 and Weissella confusa A16. The starters performance and the primary metabolites formed during 24 h of fermentation with and without 4% sucrose (w/w) were followed. Results The starters showed similar growth and acidification kinetics, but different sugar utilization, especially in presence of sucrose. Viscosity increase in fermented BSG containing sucrose occurred first after 10 h, and it kept increasing until 24 h concomitantly with dextran formation. Dextran content after 24 h was approximately 1% on the total weight of the BSG. Oligosaccharides with different degree of polymerization were formed together with dextran from 10 to 24 h. Three dextransucrase genes were identified in L. pseudomesenteroides DSM20193, one of which was significantly upregulated and remained active throughout the fermentation time. One dextransucrase gene was identified in W. confusa A16 also showing a typical induction profile, with highest upregulation at 10 h. Conclusions Selected lactic acid bacteria starters produced significant amount of dextran in brewers’ spent grain while forming oligosaccharides with different degree of polymerization. Putative dextransucrase genes identified in the starters showed a typical induction profile. Formation of dextran and oligosaccharides in BSG during lactic acid bacteria fermentation can be tailored to achieve specific technological properties of this raw material, contributing to its reintegration into the food chain.

Abstract:Dextran, an α-glucan mainly composed of (α1 → 6) linkages, has been widely applied in the food, cosmetic, and medicine industries. Dextranase can hydrolyze dextran to synthesize oligodextrans, which show prominent properties and promising applications in the food industry. Dextranases are widely distributed in bacteria, yeasts, and fungus, and classified into glycoside hydrolase (GH) 13, 15, 31, 49, and 66 families according to their sequence similarity, structural features, and reaction types. Dextranase, as a dextran-hydrolyzing enzyme, displays great application potential in the sugar-making, oral health care, medicine, and biotechnology industries. Here we mainly focused on presenting the enzymatic properties, structural features, and versatile (potential) applications of dextranase. To date, seven crystal structures of dextranases from GH 13, 15, 31, 49, and 66 families have been successfully solved. However, their molecular mechanisms for hydrolyzing dextran, especially on the size determinants of the hydrolysates, remain largely unknown. Additionally, the classification, microbial distribution, and immobilization technology of dextranase were also discussed in detail. This review discussed dextranase from different aspects with the ambition to present how they constitute the groundwork for promising future developments.
Structural characterization of the maltose acceptor-products synthesized by Leuconostoc mesenteroides NRRL B-1299 dextransucrase

Abstract:The glucooligosaccharides (GOS), produced by Leuconostoc mesenteroides NRRL B-1299 dextransucrase through an acceptor reaction with maltose and sucrose, were purified by reverse phase chromatography. Logarithmic plots of retention time vs. dp of the GOS gave three parallel lines suggesting the existence of at least three families of homologous molecules. The structure (13C and 1H NMR spectroscopy) and reactivity of the purified molecules of the three families were investigated. All the products bear a maltose residue at the reducing end. The GOS in the first family (named OD) contained additional glucosyl residues all alpha-(1-->6) linked. The smallest molecule in this first series was panose or alpha-D-glucopyranosyl-(1-->6)-D-maltose (dp 3). All the OD molecules were shown to be good acceptors for dextransucrase in the presence of sucrose. The second family, named R, was composed of linear GOS containing alpha-(1-->6)-linked glucosyl residues and a terminal alpha-(1-->2)-linked residue at the non-reducing end of the molecule; the smallest molecule in this family was alpha-D-glucopyranosyl-(1-->2)-D-panose (dp 4). The third family, R', was formed of GOS containing additional residues linked through alpha-(1-->6) linkages that constitute the linear chain, and an alpha-(1-->2)-branched residue located on the penultimate element of the chain, near the non-reducing end. The smallest molecule in this series is alpha-D-glucopyranosyl-(1-->6)-[alpha-D-glucopyranosyl-(1-->2)]-alpha-D- glucopyranosyl-(1-->6)-D-panose, dp 6. R and R' GOS are very poor acceptors for L. mesenteroides NRRL B-1299 dextransucrase. This study makes it possible to suggest a rather simple reaction scheme, where molecules Ri, R'i and ODi of the same dp all result from the glucosylation of the same GOS: ODi-l.
Microbial production of dextran using pineapple waste extract: a two-step statistical optimization of submerged fermentation conditions and structural characterization

Abstract:A two-step optimization protocol was attempted to optimize the condition for dextran production from pineapple waste using Leuconostoc mesenteroides NCIM-2198. Response surface methodology, in combination with numerical optimization technique, was explored for this purpose. In the first step, the initial medium pH (6.5–7.5), incubation temperature (25–35 °C), and time (12–60 h) were optimized from 45 full factorial runs. The maximum dextran yield was estimated as 1.46 g·[100 mL] −1 while incubated at 29.2 °C for 57.2 h at an initial pH of 7.15. Further, sucrose concentration (2–10 g·[100 mL] −1 ) and culture volume (3–7 mL·[100 mL] −1 ) were optimized from 15 experimental runs. The maximum dextran yield (1.47 g·[100 mL] −1 ) was obtained at 7.6 g·[100 mL] −1 of sucrose with 3 mL·[100 mL] −1 of culture volume at the previously optimized fermented broth. The response surface models were validated to explain the interaction between factors affecting dextran yield. The structural characteristics of the exopolysaccharide were analyzed. Fourier-transform infrared spectra showed that the exopolysaccharide contains similar spectral peaks as that of standard dextran. Nuclear magnetic resonance spectroscopy confirms the exopolysaccharide was dextran with mainly α-1-6 glycosidic bonds. Scanning electron microscopy explained its porous structure, which would be useful in retaining water and thus giving texturizing and viscosifying properties.

In situ production and characterization of cloud forming dextrans in fruit-juices

Abstract:Turbidity in beverages is typically achieved by addition of emulsion based cloud systems. Their intrinsic instability necessitates the widespread use of technological measures and use of food additives to prevent emulsion decay. In this work, we explored the possibility to establish a new generation of natural, stable clouding systems based on bacterial dextrans. Lactobacillus hordei TMW 1.1907 originating from water kefir was used to produce dextrans in sucrose supplemented apple or grape juices. By varying the fermentation conditions, two distinct types of dextran molecules could be produced at yields ranging from 2.5 to 8.5 g/L. The dextran-containing fermentates showed an unchanged turbidity after pasteurization at acidic pH and subsequent storage for three months. No sedimentation of particles occurred upon storage. Neutralization of the acidic fruit juices to pH 7 prior to fermentation significantly increased the dextran yields. The molecular weight, rms radii and turbidity of dextrans produced at 20 °C were higher than those produced at 30 °C. Characterization of the isolated dextrans by asymmetric flow field-flow fractionation coupled to multi-angle laser light scattering revealed a random-coil like structure and rms radii ranging from 66.0 to 87.4 nm. The averaged molar masses of the cloud forming dextrans were in the approximate range of 103.1 to 141.6 MDa. In conclusion, our results demonstrate the possibility to ferment fruit juices for in situ production of dextrans exhibiting novel techno-functional properties beyond gelling and thickening.

Abstract:Alpha-amylase was widely used in food industries, textile technology, paper manufacturing and so on. In this paper, the inducing mechanism of corn dextrins with different DE values (dextrose equivalent value) on production of alpha-amylase by Bacillus subtilis (B. subtilis) ZJF-1A5 was investigated. The results showed that the yield of alpha-amylase by B. subtilis ZJF-1A5 was increased by using dextrin with a certain DE value range as carbon source, which could be attributed to the presence of oligosaccharide in dextrins. By ordinary fermentation with oligosaccharide as carbon source, it was found that the inducing activity of maltopentaose was the strongest. It could be confirmed that the dextrins played important roles during the process of production of alpha-amylase by B. subtilis ZJF-1A5.

Abstract:The DEX gene encoding an extracellular dextranase from Lipomyces starkeyi was cloned into vector pPIC9k-His6 and was expressed in Pichia pastoris GS115 strain under the control of AOX1 promoter. After 107 h of the 5 L-scaled fermentation, wet cells weight of the recombinant P. pastoris Mut(+) strain reached to 588.6 g/L, and the concentration of dextranase and enzyme activity in the supernatant were 0.46 g/L and 83900 U/L, respectively. The activity of dextranase was improved 17.56-fold by cation-exchange chromatography only with a final yield of 71.61% and the specific activity of the purified enzyme was 181.96 U/mg. The purified dextranase, analyzed by SDS-PAGE and Western blotting, showed only one homogeneous band. Then the factors affecting the dextranase activity were evaluated. The optimal temperature and pH was 30 degrees C and pH 4.5, respectively. Metal ions Al3+, Cu2+, Fe3+, and SDS could completely inhibit the enzyme activity, whereas Mg2+ enhanced 145% of the enzyme activity. These characters are much different from what was previously reported for the L. starkeyi dextranase that was either expressed in S. cerevisiae or purified from natural L. starkeyi. Abstract:As a soluble food raw material with a low calorie content, resistant dextrin (RD) has broad application prospects in the food industry. Branching enzymes (BEs), as a key enzyme for RD preparation, can break the α-1,4 glycosidic bonds of donor chains and reconstruct the cleaved chains to acceptor chains through the α-1,6 glycosidic bonds. BEs with high transglucosidic activity toward amylopectin and short-chain substrates are urgently needed to increase the quality of RD. Herein, BE derived from Thermuobifida fusca (TfBE) was mined and characterized. The optimal temperature and pH of the TfBE were 40 °C and 6.5, respectively. A total of 1500 U/gsubstrate TfBE reacted with 20% (w/v) pyrodextrin for 12 h, the ratio of α-1,4 to α-1,6 glycosidic bonds was changed from 3.52:1 to 2.33:1, and the content of enzyme-resistant components notably increased from 44.0% to 53.8%. Furthermore, to make full use of receptor chains and small molecular sugars in the reaction system, a multienzyme complex of TfBE with T. fusca α-cyclodextrin glucosyltransferase (TfCGTase), TfBE with TfCGTase and Aspergillus nidulans α-glucosidase (AnGS) was used to further increase the enzyme resistance of RD from 44.0% to 65.3% and 70.6%, respectively. The developed multienzyme complex method could effectively contribute to improving the production quality and efficiency of RD preparation.

Acceptor reactions of maltodextrins with Leuconostoc mesenteroides B-512FM dextransucrase

Abstract:The acceptor products of maltose with Leuconostoc mesenteroides B-512FM dextransucrase are panose (6(2)-alpha-D-glucopyranosyl maltose) and a homologous series of 6(2)-isomaltodextrinosyl maltoses. The structures of the acceptor products of dextransucrase with other maltodextrins, maltotriose to maltooctaose (G3-G8), were determined by using the known specificities of alpha-glucosidase and porcine pancreatic alpha-amylase, and by methylation analysis. It has been found that dextransucrase transfers a D-glucopyranosyl residue to C-6 of either the nonreducing end or the reducing end residues of the maltodextrins, G3-G8, forming an alpha(1----6) linkage. When a D-glucose was transferred to the nonreducing residue, the first product was also an acceptor to give the second product, which served as an acceptor to give the third product, etc. to give a homologous series. When D-glucose was transferred to the reducing residue, the first product did not readily serve as an acceptor to give products or it served only as a very poor acceptor to give a small amount of the next homologue. The effectiveness of maltodextrins as acceptors decreased as the size of the maltodextrin chain increased. Maltotriose was 40% as effective as maltose and maltooctaose was only 6% as effective.
