Abstract:In this work, the soy protein/chitosan-based microsphere (SCM), as a stable biocompatible vehicle of oleanolic acid (OA), was constructed for the first time under acidic conditions, and OA was firstly used for quality maintenance of minimally processed products. The effect of soybean protein isolate/chitosan ratio, particle concentration and oil fraction on emulsion stability was studied. Additionally, the interfacial characteristics of SCM with and without loading OA were determined by cryo-SEM. Results demonstrated that the encapsulation efficiency of SCM embedded with 0.15% OA (SCM-OA0.15%) rated up to 87.53% +/- 2.23%, and SCM-OA showed smaller particle size, greater stability, and better liquid-state behavior, as well as the enhanced antibacterial and antioxidant properties in comparison to the control and SCM group. The minimally processed apples were used to evaluate the potential application of SCM-OA delivery system and it was evident that SCM-OA contributed to reducing weight loss, maintaining total phenol content and inhibiting color browning of minimally process apples. These findings shed new light on the emerging biocompatible vehicles of plant-derived bioactive substance.
Optimization of Enzymatic Extraction of Rosemary Acid by Response Surface Methodology and Its Antioxidant Activity

Abstract:The enzyme-assisted extraction of rosemary acid from Perilla frutescens was optimized applying one-factor-at-a-time method (OFAT) and response surface methodology (RSM) and the antioxidant activity of the rosemary acid was investigated. Preliminary investigations were conducted into the effect of enzyme dosage, hydrolysis temperature, time and pH on the extraction yield of rosemary acid. Furthermore, the optimization of process parameters for extracting rosemary acid by cellulase hydrolysis of Perilla frutescens was done by RSM using Box Behnken design. Meanwhile, the antioxidant activity was assessed by superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assays. The optimal conditions for rosemary acid extraction were determined as follows:enzyme dosage, 3%;hydrolysis temperature, 45℃;time, 12 min;and pH, 4. The maximum extraction yield of rosemary acid of 0.617%was obtained in experiments conducted under these optimal conditions, being not significantly different from the predicted value (0.621%). The rosemary acid showed strong scavenging capacity on superoxide anion and DPPH radicals, thereby having potent antioxidant activity.

Abstract:OBJECTIVE:To optimize the conditions for the extraction of panax notogingseng saponins (PNS).METHODS:After selected extraction solvent and suitable particle, we employed orthogonal experimental design to examine the conditions for the extraction by determination of PNS.RESULTS:Significant effect was observed only in extraction times.CONCLUSION:The optimum condition for extraction of PNS was to extract panax notogingseng (Burk.) F. H. Chen with 10 times 70% ethanol for 1.5 hours for 3 times.

Abstract:An activity-based approach to optimize the ultrasonic-assisted extraction of antioxidants from Pericarpium Citri Reticulatae (Chenpi in Chinese) was developed. Response surface optimization based on a quantitative composition-activity relationship model showed the relationships among product chemical composition, antioxidant activity of extract, and parameters of extraction process. Three parameters of ultrasonic-assisted extraction, including the ethanol/water ratio, Chenpi amount, and alkaline amount, were investigated to give optimum extraction conditions for antioxidants of Chenpi: ethanol/water 70:30 v/v, Chenpi amount of 10 g, and alkaline amount of 28 mg. The experimental antioxidant yield under the optimum conditions was found to be 196.5 mg/g Chenpi, and the antioxidant activity was 2023.8 μmol Trolox equivalents/g of the Chenpi powder. The results agreed well with the second-order polynomial regression model. This presented approach promised great application potentials in both food and pharmaceutical industries.

Abstract:Ultrasonic extraction technique (UET) was used to extract crude polysaccharides (OCAP) from Ornithogalum Caudatum Ait (OCA), an orthogonal experiment (L(9) (3)(4)) was applied to optimize extraction conditions. Membrane separation technology and gel filtration chromatography were used to fractionate OCAP, the antioxidant and immunomodulatory activities were evaluated by radical scavenging and spleen lymphocyte proliferation assay. The optimal conditions were determined: extraction time 60min, ultrasound power 500W, ratio of water to raw material 30ml/g and extraction number 3. Under these conditions, the yield of OCAP was 36.77±1.76%. OCAP was fractionated into three major fractions (OCAP-I, OCAP-II and OCAP-II), that all fractions possessed considerable antioxidant activity. OCAP-II and OCAP-III exhibited good immunomodulatory activities. The results indicated that UET is a very useful method for extraction bioactive polysaccharides from plant materials. OCAP could be explored as a potential antioxidant and immunostimulating agent for use in medicine or functional foods.

Abstract:The extraction procession of total saponins from Cortex Albizziae was optimized by single factor experiment and orthogonal design with saponin yield as the detective marker. The optimum extraction condition was extracting with 14 folds of 70% alcohol for 2 times under reflux, 2 h at every time. The optimized extraction condition is reasonable and scientific, and it could be used in future large-scaled preparation.

Abstract:A Box–Behnken design (BBD) of response surface methodology (RSM) was employed to further optimize ultrasonic‐assisted extraction conditions for Ophiopogon japonicus polysaccharides. The results showed that optimized extraction conditions were liquid‐solid ratio of 22:1 mL/g, ultrasonic time of 27 min, and ultrasonic power of 200 W. Under these conditions, the mean experimental value of extraction yield (39.49%) was achieved, which was 1.33 times that of distilled water leaching extraction (29.68%). In addition, SPs‐1 had high α‐glucosidase and α‐amylase inhibitory activities as well as DPPH· and ABTS·+ scavenging activities with an IC50 value of 0.060, 0.021, 0.50 and 0.10 mg/mL. Therefore, SPs‐1 from Ophiopogon japonicus could be considered as multifunctional biological ingredients to be used in anti‐hyperglycemic pharmaceutical formulation and antioxidant in functional foods. Abstract:Oat bran is a traditional agricultural byproduct and rarely used in edible oil processing. In this paper, oat bran oil (OBO) was firstly extracted by subcritical butane extraction (SBE) and the extraction process was optimized using response surface methodology. Three variables involving liquid-to-solid ratio, extraction time and extraction temperature were studied. The optimum conditions for extraction of OBO were obtained as follows: liquid-to-solid ratio 4.30, extraction time 48.15 min, and extraction temperature 46.52 °C. Based on this, an alternative method (SBE-e) for cosolvent (ethanol) was proposed to improve SBE method. Compared to conventional hexane extraction (CHE), the SBE-e had significant effect on yield, bioactive compounds (phytosterols and phenols) and antioxidant capacity (AC) in the extracted OBO. The results indicated that the proposed methods were appropriate for OBO extraction. Additionally, OBO had the potential to be an acceptable substitute for edible oil, owing to its desirable physicochemical characteristics, a balanced fatty acids composition and high antioxidant capacity.

Abstract:This study investigated the effects of ultrasound-assisted fermentation (UAF) on the preparation of antioxidant peptides (UAFP) from okara and examined their content, chemical structures, and antioxidant activity. After the optimal ultrasonic processing (time, 20 min; frequency, 45 KHz; power, 120 W/L), the peptide content yield reached the maximum of 12.36 ± 0.02 mg/mL, and their DPPH free radical scavenging rate was 65.15 ± 0.32 %. UAF increased the number of globular aggregates with deeper gullies, a looser structure, and higher porosity. The experiments conducted using the oxidative stress injury model of HepG2 cells showed that okara UAFP promoted cell growth and exerted a protective effect. Moreover, ultrasonic treatment remarkably improved the environmental stability (NaCl, glucose, sodium benzoate, temperature, pH, metal ions) and antioxidant activity of UAFP. Concisely, optimal ultrasonic processing can aid the fermentation of agroindustrial by-products to prepare antioxidant peptides, such as natural food antioxidant peptides from soybean waste.

Abstract:The proximal promoter of the C/EBPβ gene possesses dual cis regulatory elements (TGA1 and TGA2), both of which contain core CREB binding sites. Comparison of the activities of C/EBPβ promoter-reporter genes with 5′-truncations or site-directed mutations in the TGA elements showed that both are required for maximal promoter function. Electrophoretic mobility shift and chromatin immunoprecipitation (ChIP) analyses with antibodies specific to CREB and ATF1 showed that these CREB family members associate with the proximal promoter both in vitro and ex vivo. Immunoblotting and ChIP analysis revealed that other CREB family members, CREM and ATF1, are up-regulated and associate with the proximal C/EBPβ promoter in mouse embryonic fibroblasts (MEFs) from CREB(-/-) mice. ChIP analysis of wild-type MEFs and 3T3-L1 preadipocytes revealed that interaction of phospho-CREB, the active form of CREB, with the C/EBPβ gene promoter occurs only after induction of differentiation of 3T3-L1 preadipocytes and MEFs. Consistent with the interaction of CREB and ATF1 at the TGA regulatory elements, expression of constitutively active CREB strongly activated C/EBPβ promoter-reporter genes, induced expression of endogenous C/EBPβ, and caused adipogenesis in the absence of the hormonal inducers normally required. Conversely, expression of a dominant-negative CREB blocked promoter-reporter activity, expression of C/EBPβ, and adipogenesis. When subjected to the standard adipocyte differentiation protocol, wild-type MEFs differentiate into adipocytes at high frequency, whereas CREB(-/-) MEFs exhibit greatly reduced expression of C/EBPβ and differentiation. The low level of expression of C/EBPβ and differentiation in CREB(-/-) MEFs appears to be due to up-regulation of other CREB protein family members, i.e. ATF1 and CREM.

Abstract:The use of green marine seaweed Ulva spp. as foods, feed supplements, and functional ingredients has gained increasing interest. Microwave-assisted extraction technology was employed to improve the extraction yield and composition of Ulva pertusa polysaccharides. The antioxidant activity of ulvan was also evaluated. The impacts of four independent variables, i.e., extraction time (X1, 30 to 60 min), power (X2, 500 to 700 W), water-to-raw-material ratio (X3, 40 to 70), and pH (X4, 5 to 7) were evaluated. The chemical structure of different polysaccharides fractions was investigated via FT-IR and the determination of their antioxidant activities. A response surface methodology based on a Box–Behnken design (BBD) was used to optimize the extraction conditions as follows: extraction time of 43.63 min, power level of 600 W, water-to-raw-material ratio of 55.45, pH of 6.57, and maximum yield of 41.91%, with a desired value of 0.381. Ulvan exerted a strong antioxidant effect against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and showed reducing power in vitro. Ulvan protected RAW 264.7 cells against H2O2-induced oxidative stress by upregulating the expression and enhancing the activity of oxidative enzymes such as superoxide dismutase (SOD) and superoxide dismutase (CAT). The results suggest that the polysaccharides from U. pertusa might be promising bioactive compounds for commercial use.

Abstract:The aim of this study was to investigate the effect of low-frequency ultrasound on the enzymolysis of corn protein. A L 9 (3 4 ) orthogonal design was used to optimize ultrasound pretreatment conditions. Degree hydrolysis (DH), conversion rate of protein (CR), and DPPH IC 50 were selected as analytical indicators. Under the optimal ultrasound conditions (5 W/L power, 2 s/2 s on/off time, 50°C temperature, and 25 min time), the DH, CR, and radical ( D P P H ∙ , O H ∙ ) scavenging capacities were significantly increased. Molecular weight distribution and amino acid profile analysis showed that ultrasound pretreatment enhanced the formation of short-chain peptides with molecular weight of 200–3000 Da, especially the peptides containing hydrophobic amino acids. Moreover, 40 potential antioxidant peptides were purified by C18 semipreparative column and identified by UPLC-ESI-MS. The results suggest that the optimal ultrasonic-assisted enzymolysis technology could be useful for preparation of antioxidant peptides from corn.

Extraction Optimization of Polysaccharides From Corn Silk and Their Antioxidant Activities in vitro and in vivo

Abstract:Response surface technique was employed for improving the extraction of corn silk polysaccharides (CSP). Temperature, liquid-to-solid ratio, and per extraction time were all examined as separate factors. The optimal extraction parameters were determined by fitting experimental data to a second-order polynomial; a liquid-to-solid ratio of 21.5 ml/g, temperature equivalent to 88°C, and extraction time of 1.87 h. The experimental yield of the extracted polysaccharides following the application of these conditions was 4.33 ± 0.08% (dry weight), which fit quite well with the predicted value. CSP’s strong scavenging capabilities against hydroxyls, 1,1-diphenyl-2-picrylhydrazyl radicals, and superoxide anions along with its excellent reducing potential, were demonstrated in an in vitro antioxidant experiment. Meanwhile, in vivo testing revealed that CSP substantially enhanced glutathione peroxidase and superoxide dismutase activities. The Malondialdehyde levels in the liver and serum of aged mice also underwent a decrease. This study found that CSP has a substantial antioxidant potential in vitro and in vivo , suggesting that it might be used as an antioxidant in food and medicine.

Abstract:Introduction: In this study, we optimized the extraction process, analyzed the structure and assessed the antioxidant activity of Sanghuangporus baumii polysaccharide (SBP). The present results provide important information for the Sanghuangporus baumii polysaccharides in potential natural antioxidant effects. Background: The extraction and structural analysis of polysaccharides from Sanghuangporus has gained significant attention in the fields of chemistry, medicine, and life sciences. There is great significance in maximizing the extraction of polysaccharides from Sanghuangporus and developing their potential products in a scientific and rational manner. Objective: The study was designed to establish an efficient and practical extraction process for SBP, and then investigated the structure and the antioxidant activity. Methods: The Response Surface Methodology (RSM) based on Box-Behnken design was used to explore the ultrasound-assisted extraction of SBP, and the structure of SBP was studied by ultraviolet spectroscopy, infrared spectroscopy and other instrumental analysis methods. The total antioxidant capacity of SBP was studied by the Ferric Reducing Ability of Plasma (FRAP) method, and the scavenging capacity of ABTS + ·, DPPH· and OH· was carried out as the index to investigate its antioxidant activity in vitro . Results: The statistical analysis results showed that the optimal conditions for extracting SBP were an ultrasound time of 20.74 min, ultrasound power of 268.40 W and material-liquid ratio of 1:25.71. Under optimal conditions, the experimental yield of SBP was 3.36 ± 0.01%. The RSM optimization process was applied to the experiment of complex enzyme extraction of SBP, and the yield increased to 4.72 ± 0.03%. Structural analysis showed that SBP mainly consisted of glucose, a small amount of mannose and galactose, and the molecular weight distribution was uneven, mainly concentrating in the three parts of 24.5,6.4,2.5 kDa. Moreover, SBP exhibited dose-dependent and strong reducing power and radical scavenging activity. For DPPH·, ABTS + · and OH· radical scavenging assays, IC 50 values were 1.505 ± 0.050 mg/ml, 0.065 ± 0.036 mg/ml and 0.442 ± 0.007 mg/ml, respectively. Conclusion: In the present study, a β-linked heteroglucan (SBP) was extracted using the optimized process combining enzymes from the fruiting bodies of Sanghuangporus baumii. SBP exhibited effective and dose-dependent antioxidant activities. Our findings were of great value in terms of developing polysaccharides with potential natural antioxidants.

Optimization of extraction of crude polysaccharide from Cassia seed via ultrasonic assisted cellulase and study on its antioxidant activity

Abstract:In order to improve the current low extraction rate of cassia polysaccharide and improve its extraction efficiency, the ultrasonic wave assisted cellulase extraction method of cassia polysaccharide is first proposed and optimized. Orthogonal test is conducted to analyze the influence of six factors related to cellulase and ultrasonic wave on the extraction of cassia polysaccharide, and at the same time to explore the antioxidant activity of the extracted polysaccharide in vitro. The results show that the main and secondary factors affecting the extraction rate of cassia polysaccharide are pH > ultrasonic power > enzyme dosage > enzymatic hydrolysis temperature > enzymatic hydrolysis time > ultrasonic time. The optimal conditions are: enzyme dosage: 1.6%, enzymatic hydrolysis time: 80 min, enzymatic hydrolysis temperature: 52 °C, enzymatic hydrolysis pH: 6.4, ultrasonic time: 30 min, ultrasonic power: 276 W, the polysaccharide yield under the optimal conditions is 12.29%, which is 5.31% and 14.43% higher than the single enzyme extraction method and ultrasonic extraction method reported in the literature, respectively, indicating that the composite extraction method is superior to the single extraction method. The antioxidant activity analysis shows that the half inhibitory concentrations of cassia polysaccharide against DPPH·, ABTS·+, ·OH, ·O2- free radicals are 0.894 mg/mL, 1.175 mg/mL, 1.908 mg/mL and 7.601 mg/mL, respectively, which shows good antioxidant activity in vitro. Therefore, ultrasonic assisted cellulase extraction of crude polysaccharide from cassia seed is a new efficient and rapid extraction method.
Extraction, purification of saponins components from Xanthoceras sorbifolium Bunge leaves: potential additives in the food industry

Abstract:Xanthoceras sorbifolium Bunge leaves (XLs) are the agricultural by-products of Xanthoceras sorbifolium Bunge, which are widely cultivated in the north of China. For recycling XLs, in this study, the extraction optimization, construction of purification models, identification of saponins components and the antimicrobial and antioxidant activities were firstly investigated. Ultrasonic-assisted extraction of saponins from XLs using Box–Behnken design (BBD) method of response surface methodology (RSM) was investigated. The optimum conditions were as follows: the temperature 60.79 °C, ethanol concentration 71.56%, liquid/solid ratio 30.67 (v/m). Under these conditions, the optimization of the extraction with maximize yield of 7.36 ± 0.078% was obtained. Corresponding adsorption models were established to analyze the maximum yield of saponins during the adsorption process, and AB-8 resin was used for the purification of saponins using static adsorption/desorption with final purity was 38.48 ± 2.51%. Then, ten saponins, including ginsenoside-Rg1, ginsenoside-F, ginsenoside-Rg2, alpha-hederin, hederagenin, licoricesaponin G2, betulinic acid, ursolic acid, betulonic acid, and dehydro (11,12) ursolic acid lactone, were identified using ultra performance liquid chromatography-mass spectrometry (UPLC-MS). Biological evaluations showed that XLs saponins possessed good scavenging activity on DPPH and ABTS, and exerted obvious inhibitory effect on E. coli (Gram-negative bacteria) and S. areus (Gram-positive bacteria). Results indicated that the saponins extracted from XLs can be used as a natural antimicrobial and antioxidant agent to the food products. This study provided a theoretical basis for the development and utilization of the by-product saponins from the bio-products resources XLs.

Abstract:Abstract Structural and functional changes in ultrasonicated oyster protein isolates (OPI) were investigated. Ultrasound treatments were carried out with probe (20 kHz) at 200, 400 and 600 W for 15 and 30 min. The results showed that functional properties of OPI significantly improved after sonication. Absolute zeta potential and protein solubility increased by 18.40 mV and 82.5 % at 600 W for 15 min. Oil holding capacity, emulsifying activity index, emulsion stability index, foaming ability and foaming stability increased by 300 %, 15.23 m 2 /g, 9.24 min, 23.9 % and 14.8 % at 600 W for 30 min. However, ultrasound treatment significantly ( P < 0.05) decreased particle size and water holding capacity. The conformation of OPI became stretched and unfolded after sonication. Functional improvements resulted from stretched and unfolded conformation and reduction of particle size. Controlled condition of ultrasound can produce OPI with distinct structural and functional properties, which could meet the complex needs of manufactured food products in food industry, but further study is needed to understand the specific mechanism.

Abstract:This research aimed to elucidate how Toona sinensis seed polyphenols (TSSs) inhibited pork myofibrillar protein (MP) oxidation and modified physicochemical and gel properties in the Fenton oxidation system. Our result displayed that TSSs had a lower carbonyl (from 1.38 to 0.59 nmol/mg) and dityrosine contents (from 47.22 to 25.07), and higher free amino content (from 386.99 to 485.00 nmol/mg), followed by ellagic acid (EA) and quercetin-3-O-rhamnoside (QC). Meanwhile, the incorporation of phenolic compounds changed secondary and tertiary structure of proteins and then increased solubility and absolute value of zeta potential, as well as decreased turbidity and average particle size. Molecular docking indicated that MP interacted with EA primarily via hydrogen bonds and hydrophobic interaction, and with QC mostly through hydrogen bonds and electrostatic interaction. Otherwise, the incorporation of EA promoted MP gel to form a honeycomb-like microstructure after oxidation. Therefore, TSSs, EA, and QC could significantly inhibit MP oxidation as well as modify its physicochemical properties, but only EA enhanced its gel properties after oxidation. The results could be useful to improve the comprehensive utilization of plant by-products and provide theoretical references for the role of TSSs in the improvement of meat products.

Abstract:Tea saponins have high surface-active and biological activities and are widely used in chemicals, food, pharmaceuticals, and pesticides. Tea saponins are usually extracted using ethanol or water, but both methods have their disadvantages, including a negative impact on the environment, high energy consumption, and low purity. In this study, we explored an effective process for extracting tea saponins from tea meal using deep eutectic solvents combined with ultrasonic extraction and enzymatic techniques. The experimental results showed that a high extraction efficiency of 20.93 ± 0.48% could be achieved in 20 min using an ultrasonic power of 40% and a binary DES consisting of betaine and ethylene glycol (with a molar ratio of 1:3) at a material–liquid ratio of 1:35 and that the purity of the tea saponins after purification by a large-pore adsorption resin reached 95.94%, which was higher than that of commercially available standard tea saponin samples. In addition, the extracted tea saponins were evaluated for their antioxidant and bacteriostatic activities using chemical and biological methods; the results showed that the tea saponins extracted using these methods possessed antioxidant properties and displayed significant antibacterial activity. Therefore, the present study developed a method for using deep eutectic solvents as an environmentally friendly technological solution for obtaining high-purity tea saponins from tea meal oil. This is expected to replace the current organic solvent and water extraction process and has great potential for industrial development and a number of possible applications.

Optimization of ultrasound-assisted aqueous two-phase extraction of polysaccharides from seabuckthorn fruits using response methodology, physicochemical characterization and bioactivities

Abstract:Background: Seabuckthorn fruits contains many active subtances, among them, the seabuckthorn polysaccharide is one of the main active ingredients, and exhibits diverse bioactivities. The extraction of polysaccharides from seabuckthorn fruits is the most important step for their wide applications. Ultrasound-assisted aqueous two-phase extraction (UA-ATPE) is a promising green method for extracting polysaccharides. Additionally, physicochemical characterization and antioxidant activities can evaluate the potential functions and applications in the food and medicine industries. Results: Based on the single-factor experiments, 20.70% (w/w) ammonium sulfate ((NH4 )2 SO4 ) and 27.56% (w/w) ethanol were determined as the suitable composition for aqueous two-phase. The optimum conditions of UA-ATPE obtained by response surface methodology were as follows: ultrasonic power (390 W), extraction time (41 min), liquid-to-material ratio (72: 1 mL/g), and the total yield of the polysaccharides reached 34.14 ± 0.10%, The molecular weights of the purified upper-phase seabuckthorn polysaccharide (PUSP) and the purified lower-phase seabuckthorn polysaccharide (PLSP) were 65 525 and 26 776 Da, respectively. PUSP and PLSP contained the same six monosaccharides (galacturonic acid, rhamnose, xylose, mannose, glucose and galactose), but with different molar ratios. Furthermore, PUSP and PLSP displayed certain viscoelastic property, had no triple helical structure, possessed different thermal stability, surface morphology and conformation in aqueous solution. PUSP and PLSP displayed strong antioxidant properties by the assays of scavenging ability of ABTS+ ·, the protection of DNA damage and erythrocyte hemolysis. Conclusion: UA-ATPE significantly increased the yield of seabuckthorn polysaccharides. PUSP and PLSP were different in many aspects, such as molar ratio, surface shape and antioxidant activities. Seabuckthornpolysaccharides possess great potential in medicine and functional foods. © 2022 Society of Chemical Industry.
