Abstract:Several scoring systems have been used to predict mortality from major surgery. By predicting risk in this way, the score can help select patients for surgery, or identify those who need specific interventions to manage their risk [1, 2]. Hitherto, it has not been suggested that the very act of using a score either actually harms patients (akin to a curse) or protects them (like a holy incantation). However, some recent data suggests that the former may be the case: the act of performing a predictive score can be lethal, even if this score does not influence subsequent management. How can this be, and what is the evidence? Carlisle recently commented in an editorial [3] on data from de Buck van Overstraeten et al., assessing the value of using the CR-POSSUM score in a Belgian cohort [4]. The original study was well conducted with interesting conclusions, but those aspects are not our concern here. In his editorial, Carlisle statistically analysed the results of applying three other common scores (SORT [5], the UK audit model [6] and the Torbay model [7]) to the Belgian data, in addition to the CR-POSSUM score [8] used by de Buck van Overstraeten et al. Note that in this theoretical analysis, the management of patients was not dictated by the score outcome, which was something only retrospectively applied at a distance. Carlisle’s aim was to show that some predictive models (he concluded the UK audit tool and Torbay model) were more consistent with the actual mortality rates published in the Belgian study. However, the same data can be presented in different ways to address distinct issues. I was interested in a completely separate question: what can be learned from plotting the mortalities predicted by the various scoring systems?
Comparison of Angiotensin‐Converting Enzyme Inhibition with Angiotensin II Receptor Antagonism in the Human Forearm

Abstract:Summary The object of this study was to differentiate losartan, an AT1-selective angiotensin II (ANG II) receptor antagonist, from enalapril, an angiotensin-converting enzyme (ACE) inhibitor, by measuring forearm vascular responses to AI, AII, and bradykinin. Eight healthy men were studied in a randomised, 4-period crossover study in which placebo, enalapril (10 mg), losartan (20 mg) and losartan (100 mg) were given double-blind on separate occasions. Forearm blood flow was measured by venous occlusion plethysmography during sequential infusions of ANG I, ANG II, and bradykinin into the brachial artery 4–6 h after dosing. Analysis of variance for repeated measures indicated that losartan inhibited constriction to ANG I and ANG II (both p < 0.02) in a dose-dependent manner without significantly influencing vasodilator responses to bradykinin. Enalapril (10 mg) inhibited AI similarly to losartan 100 mg without significantly influencing responses to angiotensin II, and augmented vasodilator responses to bradykinin (p < 0.0001). In human forearm vasculature, oral losartan (20–100 mg) inhibits vasoconstriction to ANG I and ANG II without significantly influencing bradykinin-induced vasodilation, whereas enalapril selectively inhibits ANG I-induced vasoconstriction while potentiating the vasodilator effect of bradykinin.

Abstract:Natural compounds including bis-heterocyclic moieties displayed several applications in pharmaceutical, agrochemical, and industrial fields. Di-thiazoles are effective core of several anticancer drugs such as bleomycin. Derived from importance of di-thiazoles, herein, we synthesized novel series of di-thiazoles from condensation reaction of thiazole thiosemicarbazone derivative with α-haloketones. Physical features and chemical structures of new compounds formed were discussed and assured based on their spectral and conformational analysis. The cytotoxic potency of newly synthesized thiazole derivatives against colon (HCT-116) and hepatocellular (HepG2) carcinoma-cell lines was determined by MTT assay. Compounds 7a, 9a, 9c, and 9e revealed marked anticancer activity especially against (HCT-116). Molecular docking study was executed over chosen compounds referring to expected promising features against LDH-5, which appeared agree with biological results. This study was utilized to predict prior binding-efficiency of active thiazole compounds.

Abstract:A series of 3-aryl-2-quinoxaline-carbonitrile 1,4-di-N-oxide derivatives were designed, synthesized and evaluated for hypoxic and normoxic cytotoxic activity against human SMMC-7721, K562, KB, A549 and PC-3 cell lines. Many of these new compounds displayed more potent hypoxic cytotoxic activity compared with TX-402 and TPZ in the tumor cells based evaluation, which confirmed our hypothesis that the replacement of the 3-amine with the substituted aryl ring of TX-402 increases the hypoxic anti-tumor activity. The preliminary SAR revealed that 3-chloro was a favorable substituent in the phenyl ring for hypoxic cytotoxicity and 7-methyl or 7-methoxy substituted derivatives exhibited better hypoxic selectivity against most of the tested cell lines. The most potent compound, 7-methyl-3-(3-chlorophenyl)-quinoxaline-2-carbonitrile 1,4-dioxide (9h) was selected for further anti-tumor evaluation and mechanistic study. It also exhibited significant cytotoxic activity against BEL-7402, HepG2, HL-60, NCI-H460, HCT-116 and CHP126 cell lines in hypoxia with IC50 values ranging from 0.31 to 3.16 mu M, and preliminary mechanism study revealed that 9h induced apoptosis in a caspase-dependent pathway.

Abstract:Various sets of thiazole, thiophene, and 2-pyridone ring structures containing a dimethylaniline component were synthesized. Substituted thiazoles 2 – 3 and thiophenes 5 – 7 were produced by reacting thiocarbamoyl compound 4 with α-halogenated reagents in different basic conditions. Also, a series of 2-pyridone derivatives 9a – f substituted with dimethylaniline was synthesized through Michael addition of malononitrile to α,β-unsaturated nitrile derivatives 8a – f . The synthesized products were structurally proven by spectroscopic methods such as IR, 1 H NMR, 13 C NMR, and MS data. Furthermore, the anti-cancer efficacy of the compounds was assessed using the MTT assay on two cell lines: hepatocellular carcinoma ( HepG-2 ) and breast cancer ( MDA-MB-231 ). The results showed the highest growth inhibition for derivatives 2 , 6 , 7 , and 9c , which were further examined for their IC 50 values. The IC 50 for compound 2 showed equipotent activity (IC 50 = 1.2 μM) against the HepG-2 cell line compared to Doxorubicin (IC 50 = 1.1 μM). Compounds 2 , 6 , 7 and 9c showed very good ADME assessments for further drug administration. Moreover, the PASS theoretical prediction for the compounds showed high antimitotic and antineoplastic activities for compounds 2 , 6 , 7 , and 9c , as well as potent inhibition activity for the insulysin enzyme (IDE). Molecular docking stimulations were performed on CDK1/CyclinB1/CKS2 (PDB ID: 4y72) and BPTI (PDB ID: 2ra3). When docked into (PDB ID: 4y72), all of the tested compounds showed considerable inhibition, and the 2-pyridone derivative 9d had the maximum binding affinity (− 8.1223 kcal/mol). While thiophene derivative 6 offered the maximum binding affinity (− 7.5094 kcal/mol) when docked into (PDB ID: 2ra3).

Abstract:In this study, some new pyrazoline derivatives bearing cyano or nitro groups were synthesized. The structures of the compounds were characterized by IR, 1H-NMR, 13C-NMR and elemental analysis data. The ABTS·+, DPPH·, CUPRAC and β-Carotene/linoleic acid assays were carried out to determine the antioxidant activity of the synthesized pyrazolines. Compound P14 showed higher antioxidant activity than the standard substance BHA with IC50 values of 1.71±0.31 μM and 0.29±0.04 μM in ABTS+ and β-carotene/linoleic acid assays, respectively. Compound P12 also exhibited higher antioxidant activities than BHA with an IC50 value of 0.36±0.14 μM in β-carotene/linoleic acid analysis. In activity studies of pyrazolines against cholinesterase (AChE and BChE), tyrosinase, α-amylase and α- glucosidase, compound P1 (IC50 = 39.51±3.80 μM) showed higher activity against α-amylase and compounds P5 and P12 displayed higher activity against α-glucosidase than acarbose with IC50 values of 14.09±0.62 and 83.26±2.57 μM, respectively. The drug-like properties such as Lipinski and Veber, bioavailability and toxicity risks of the synthesized compounds were also evaluated. The compounds were predicted to be compatible with Lipinski and Veber rules, have high bioavailability and low toxicity profiles. Moreover, molecular docking studies were performed to better understand the high activity of the compounds against a-amylase and a-glucosidase enzymes.

Abstract:N'-(2-((4-(Benzo[d][1, 3]dioxol-5-yl)-3-cyano-6-(naphthalen-2-yl)-pyridin-2-yl)oxy)- acetyl)benzohydrazide (6) and hydrazide derivatives (7–10) were synthesized via reaction of 3-cyano-pyridinacetohydrazide (5) with benzoylchloride and the appropriate aldehyde, respectively. Also, 4-(Benzo[d][1, 3]dioxol-5-yl)-2-(2-(3-methyl-5-oxo-4,5-dihydro-1H-pyrazol-1-yl)-2-oxoethoxy)-6-(naphthalen-2-yl) nicotinonitrile (11) was prepared through reaction of (5) with ethylacetoacetate. In addition, the hydrazide 5 was used as a starting material for synthesizing other nicotinonitrile derivatives (12, 13) and acetohydrazides (14, 15). Moreover, new 2, 5-disubsitituted-1,3,4-oxadiazoles (16, 17) were furnished by the reaction of (5) with benzoic acid derivatives. The cytotoxicity for some new compounds was investigated against human breast cancer MCF-7 and MDA-MB-231 cell lines at concentrations (0, 10, 20, 40, 60, 80, and 100 μg/ml) using MTT assay. Compounds (4, 9, 14 and 17) were recorded the lowest half-maximal inhibitory concentrations (IC50s) against MCF-7 and MDA-MB-231 cell lines via cytotoxicity assays. These results were confirmed using molecular docking experiments. From the previous observations, we suggested that our synthesized compounds could be of great potential against breast cancer cells.Graphical

Abstract:In this paper, we present the synthesis and characterization of two known sulfonyl hydrazides (1 and 2) and their new sulfonyl hydrazone derivatives (9–20), as well as in vitro and in silico investigations of their cytotoxic properties against human lung (A549) and human breast (MCF-7) cancer cell lines. The target compounds (9–20) obtained in high yields were synthesized for the first time by a multi-step reaction, and their structures were confirmed by elemental analysis and various spectral techniques, including FT-IR, 1H-, and 13C-NMR. The antiproliferative profiles of these compounds (1, 2, and 9–20) in this study were determined at concentrations of 200, 100, 50, and 25 μM against selected cancer cell lines for 72 h using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method. Except for compounds 1 and 2, other compounds (9–20) demonstrated cytotoxic activity at concentrations lower than 200 μM. The newly synthesized compounds (9–20) demonstrated antiproliferative activities at a micromolar level, with IC50 values in the range of 29.59–176.70 μM for the A549 cell line and 27.70–170.30 μM for the MCF-7 cell line. Among these compounds, compound 15 (IC50 = 29.59 μM against A549 cell line and IC50 = 27.70 μM against MCF-7 cell line) showed the highest cytotoxic activity against these two cancer cell lines compared to the reference drug cisplatin (IC50 = 22.42 μM against A549 cell line and IC50 = 18.01 μM against MCF-7 cell line). From docking simulations, to establish a plausible binding mode of compounds, we noticed that compound 15 demonstrated the highest affinity (−6.8508 kcal/mol) for estrogen receptor-beta (ERbeta) compared to others, suggesting promising ERbeta binding potential. Most compounds followed Lipinski's rule of five, with acceptable logP values. Additionally, all had mixed gastrointestinal absorption and limited blood–brain barrier permeability. Overall, our study proposed new sulfonyl hydrazones as a potential class of anticancer agents.

Abstract:Phenothiazines ˗ Antipsychotic drugs, which have been studied from centuries and attracted researcher’s attention toward its antitumor activities apart from their fundamental neuroleptic properties. Hence, we have targeted different structures of phenothiazines to develop potent antitumor agents. In the present study, we have synthesized various heterocyclic derivatives incorporating phenothiazine compounds via one-pot multicomponent reaction by means of N-alkyl-3-formylphenothiazine under reflux/ultrasonic method and different spectral characterization techniques such as FT-IR, mass, 1H and 13C NMR spectroscopy were used to validate the molecular structures of the synthesized compounds. The antitumor properties of all the synthesized compounds were investigated using MTT and LDH assay against HeLa cell lines. The results shown that the compounds 4 b, 2a and 3a have maximum LDH release (57.69 ± 4.69, 57.7 ± 4.6 & 67.8 ± 4.9) at 1000 µg concentration.

Abstract:Aim: The extensive utilization of 2-phenylbenzothiazole due to their wide array of biological activities, particularly in cancer therapy, has caused great attention to explore more potent derivatives.Materials & methods: We report the synthesis of 2-phenylbenzothiazole tagged 1,2,3-triaozle (8) through Cu(I)-catalyzed cycloaddition of alkyne side chain with aryl-substituted azides.Results: The in vitro experiments, using MTT and 99mTc-MIBI cell uptake methods, demonstrated the remarkable anticancer activity of these compounds against A549, SKOV3 and MCF7 cell lines.Conclusion: Compounds 8b, 8f and 8i possessed high cytotoxic activity as compared with doxorubicin. Compound 8g has a similar inhibitory effect on the proliferation of breast cancer cells as doxorubicin. In silico study indicated that compound 8 would be a good lead for the development of new potent anticancer agents.

Abstract:Background The chemistry of condensed heterocyclic compounds has emerged in numerous reports for their diverse biological properties and drug discovery. Pyrimidine and triazine scaffolds have been utilized as therapeutic agents in many medicinal applications. Many research groups have designed and synthesized pyrimidine moieties as they are incorporated in nucleic acid bases. Objective In this report, we have designed and synthesized a variety of 2-mercaptothieno pyrimidine and thienotriazine derivatives and 2-mercaptothienopyrimidines conjugated with sugar moiety. The newly synthesized compounds were tested for their biological activity against breast (MCF-7), liver (HepG-2), and prostate (PC-3) cancer cell lines as well as a normal cell line (human normal melanocyte, HFB4) and were also analyzed for in silico studies to determine their potential. Materials and methods A variety of 2-mercaptothienopyrimidine and thienotriazine derivatives were prepared via cyclization of ethyl 2-amino-4,5,6,7-tetrahydrobenzo[b]thiophene-3-carboxylate (1) and 2-amino- N -phenyl-4,5,6,7-tetrahydrobenzo[ b ]thiophene-3-carboxamide (9) . Two derivatives of 2-mercaptothienopyrimidines conjugated with sugar moiety were also prepared. The products were screened for their biological activity against breast (MCF-7), liver (HepG-2), and prostate (PC-3) cancer cell lines as well as the normal cell line (human normal melanocyte, HFB4) in comparison with the known anticancer drug 5-fluorouracil using the MTT assay. Results and conclusion The results indicated that most of the tested compounds exhibited no activity against the growth of HFB4. Compounds 5, 8, 10, 12, and 14 revealed effective antiproliferative activity against MCF-7 cell lines with IC 50 of 4.6, 6.2, 5.4, 7, and 3.25 μg/ml, respectively, compared with 5-fluorouracil (IC 50 of 3.97 μg/ml). In the same sense, the evaluation of cytotoxic effect of the tested compounds against human liver HepG-2 cancer cell lines revealed that compounds 5, 8, 10, 12, and 14 showed cytotoxic activity close to that of the standard drug (IC 50 values of 5.77±0.99, 7.23±0.98, 4.42±1.32, 7.9±0.90, and 5.1±11.28 μg/ml, respectively, vs. 4.27±0.58 μg/ml for 5-fluorouracil). Free binding energy was estimated by docking and MM-GBSA calculation. Molecular dynamics simulation followed by MM-GBSA calculation was correlated to the cytotoxic effect. Compound 14 illustrated the highest MM-GBSA value (−20.38) and the best cytotoxic effect.

Abstract:The widespread use of small molecules having heterocyclic rings to design anticancer agents, and the antioxidant potential of their phenolic conjugates led us to investigate phenol-based heterocyclic compounds' anticancer, antioxidant activities, and drug-likeness properties. While compounds were evaluated in regards to cancer and non-cancer cell lines by the MTT method, antioxidant effects were carried out using the DPPH method. Drug-likeness properties were also evaluated using SwissADME software. The mean CC 50 values were in the range 18.7 – >400 μM toward carcinoma cell lines. Compound 2-(benzo[ d ]thiazol-2-ylthio)-1-(4-hydroxyphenyl)ethanone showed the highest selectivity cytotoxicity and it showed antioxidant effect with IC 50 = 0.036 ± 0.004 mg/mL. All compounds generally showed promising pharmacokinetic behaviors by obeying the various drug-likeness approaches.

Abstract:A series of novel thiazolyl-pyrazoline derivatives containing benzodioxole (C1–C20) have been designed and synthesized. Among of the synthesized compounds, 2-(5-(benzo[d][1,3]dioxol-5-yl)-3-(4-bromophenyl)-4,5-dihydro-1H-pyrazol-1-yl)-4-(4-bromophenyl)thiazole (C6) displayed the most potent inhibitory activity for HER-2 (IC50=0.18μM for HER-2). Antiproliferative assay results indicated that compound C6 owned high antiproliferative activity against MCF-7 and B16-F10 in vitro, with IC50 value of 0.09 and 0.12μM, respectively, being comparable with the positive control Erlotinib. Docking simulation was further performed to determine the probable binding model. Based on the preliminary results, compound C6 with potent inhibitory activity in tumor growth would be a potential anticancer agent.

Abstract:New sets of functionalized thiazolidinone and thiadiazole derivatives were synthesized, and their cytotoxicity was evaluated on HepG2, MCF‐7, HTC‐116, and WI38 cells. The synthetic approach is based on the preparation of 4‐(4‐acetamidophenyl)thiosemicarbazide (4) and their thiosemicarbazones 5a‐e, which are converted to the corresponding thiazoldin‐4‐one compounds 6a‐e upon cyclization with ethyl bromoacetate. The thiadiazole compounds 9 and 12 were obtained by reacting 4‐(4‐acetamidophenyl)thiosemicarbazide with isothiocyanates and/or ethyl 2‐cyano‐3,3‐bis(methylthio)acrylate, respectively. The thiazolidinone compounds 6c and 6e exhibited strong cytotoxicity against breast cancer cells, with an IC50 (6.70±0.5 μM) and IC50 (7.51±0.8 μM), respectively, very close to that of doxorubicin (IC50: 4.17±0.2 μM). In addition, the anti‐cancer properties of the tested thiazolidinone and thiadiazole scaffolds were further explored by the molecular docking program (MOE)‐(PDB Code‐1DLS). Compounds 5d, 5e, 6d, 6e, and 7 have the best binding affinity, ranging from ‐8.5386 kcal.mol‐1 to ‐8.2830 kcal.mol‐1.

In Vitro Genotoxic and Antigenotoxic Effects of Ten Novel Synthesized 4-Thiazolidinone Derivatives

Abstract:Heterocyclic compounds are found in a variety of drug molecules, and bioactive natural products. 4-Thiazolidinones (4-TZDs), which represent an important class of heterocyclic compounds, are of great interest today with their diverse bioactivities. In this study, ten novel 4-TZD derivatives (C1-C10) were synthesized, characterized by spectroscopic techniques, and their genotoxic, and antigenotoxic properties were investigated in vitro using the Ames Salmonella/microsome mutagenicity assay in the concentration range of 0.2-1.0 mM/plate. The results revealed that none of the compounds were mutagenic on the three different Salmonella typhimurium strains up to the highest concentration tested. Furthermore, in our study, C1, C4, C6, and C9 showed significant, ranging from moderate to strong, antigenotoxic effects against mutagen-induced DNA damage at relatively higher doses. Among these, C4 had the best potential to inhibit the number of revertant colonies induced by 9-aminoacridine (9-AA), with a maximum inhibition rate of 47.9 % for 1.0 mM/plate. As a result, preliminary knowledge about the safety of the use of ten novel synthesized 4-TZD compounds likely to exhibit many bioactivities was obtained in this study. In addition, the significant in vitro antimutagenic activity of some derivatives increases the importance of studies for the development of new pharmacological agents for cancer prevention.
Potential Antioxidant Activity of New Tetracyclic and Pentacyclic Nonlinear Phenothiazine Derivatives

Abstract:The global increase in oxidative stress related diseases such as cancer, cardiovascular, and inflammatory diseases caused by overwhelming level of free radicals in the body has encouraged the search for new antioxidant agents. Based on the ability of newly synthesized phenothiazine derivatives (6-chloro-11-azabenzo[a]phenothiazine-5-one and 6-[4-bromophenyl]-10-methyl-11-azabenzo[a]phenothiazine-5-one) to oxidize H 2 O 2 , a known free radical to sulfoxide, this study assessed the in vitro and in vivo antioxidant activity. The synthesized phenothiazine derivatives exhibited reducing power potential to convert Fe 3+ to Fe 2+ and high ability to scavenge H 2 O 2 free radical in vitro. These activities were comparable to ascorbic acid, a standard antioxidant. The catalase activity significantly increased ( p < 0.05 ) in groups 1 and 2 animals that received the phenothiazine derivatives compared to the controls (groups 3 and 4) suggesting the ability of the phenothiazine derivatives to scavenge H 2 O 2 in vivo. The malondialdehyde level in groups 1 and 2 animals was lower than that in group 3 that received the reference compound (ascorbic acid) and group 4 that received the solvent suggesting the ability of the phenothiazine derivatives to prevent lipid membrane damage. AST and bilirubin levels were higher in group 2 animals which received 6-[4-bromophenyl]-10-methyl-11-azabenzo[a]phenothiazine-5-one compared to group 3, the positive control. The results suggest that phenothiazine derivatives, especially 6-chloro-11-azabenzo[a]phenothiazine-5-one, possess antioxidant activity though 6-[4-bromophenyl]-10-methyl-11-azabenzo[a]phenothiazine-5-one was slightly toxic. This activity may be due to the presence of electron donors such as sulfur as well as the richness of hydrogen in the additional benzene rings for substitution. Further study is needed to identify tolerable doses for possible therapeutic purposes.

Abstract:The present article demonstrates the regioselective synthesis, characterization, and biological evaluation of eighteen novel pyrazolo‐benzothiazole hybrid molecules 5a‐5r. We have utilized β,β‐ditosyloxy ketones protocol to synthesize these hybrid molecules. The synthesized compounds were tested for their in‐vitro antiproliferative activities using MTT assay against breast cancer (MCF‐7), cervical cancer (HeLa), and Lung cancer (A549) cell lines. Hybrid molecules 5a, 5m, 5n, and 5o with IC50 values of 0.359 mM, 0.051 mM, 0.079 mM, and 0.259 mM respectively exhibited admirable growth inhibitory activities against MCF‐7 cancer cells even better than reference carboplatin drug having IC50 (0.439 mM). Compound 5k with IC50 value of 0.765 mM was found to be the most potent antiproliferative agent for the HeLa cancer cells. Moreover, hybrid molecule 5f with IC50 value of 0.706 mM exhibited better inhibitory activity against A549 cancer cells in comparison to the reference carboplatin drug having IC50 (0.805 mM). The mechanism of cellular toxicity was studied using Annexin V‐FITC/PI double staining method and cell cycle assay. Molecular docking studies for all the synthesized compounds have been performed in the binding pocket of VEGFR‐2 sites (PBD code: 4ASD). Finally, the ADMET profile of the potent molecules was also investigated to predict their drug‐likeness behaviour.
