Abstract:Observations were made on the behavior and fertility of 24 female ra ts during 13 weeks of administration of plant estrogen prepared from silage fodder containing red clover. Later an extract of pure clover was used. Hydrolysed isoflavones were extracted with alcohol. Dosage was equivalent to 3 gm of dry fodder orally per day. Extracts were given absorbed in oatflakes and moistened with milk. Control groups received the same feedings except for the extracts. During the first 2 weeks rats of the test group were more inert than the controls. Their feet became bluish and ear tips of some were necrotic. Later when other fodder was increased they became more lively and aggressive. Vaginal openings occurred earlier in the treated groups. No vaginal plugs were found in the test group. 1 treated rat had given birth and 4 others were found to be 1 to 3 days pregnant when sacrificed at the end of the experiment. In the control group there were litters in all but 1 rat with 5 to 10 young per litter for a total of 58 offspring. The 1 rat in the test group which gave birth had 6 offspring while the 4 pregnant rats had an average of 8.6 fetuses per rat. The complete absence of vaginal plugs in the test group contrasted with the control group in which they were found in 5 out of 7 mated rats with due delivery 3 weeks later. From these studies it seems that plant estrogens in the amounts employed will suffice to provoke malfunctions of fertility. Consequently the quantity of plant estrogens may be a component factor in the fertility of rodents in nature. Large amounts are destructive to the number of progeny but small amounts increase fertility.
Preparation, Characterization, and Foaming Properties of Soy Protein Nanoparticles by the Cross-Linking Reaction Induced by Microbial Transglutaminase

Abstract:Native soy proteins have limited surface properties, but fortunately, it is expected that their foaming behaviors can be improved through the enzymatic cross-linking method. In this paper, by using the cross-linking method with microbial transglutaminase (mTG) as the catalyst, soy protein isolate (SPI), soy β-conglycinin (7S), and soy glycinin (11S) nanoparticles (NPs) were prepared in the molten state of the protein induced by dithiothreitol (DTT). We compared the structural characteristics of these nanoparticles, including particle sizes, morphologies, zeta potentials, surface hydrophobicities, changes of spatial structure, and cross-linking degrees. In addition, their foaming properties, including foamabilities, foam stabilities, and bubble diameters, were also tested. The results show that there are no significant differences in the foamabilities between the native soy proteins and their nanoparticles with similar initial foam heights. However, it is found that 11S NPs have enhanced foam stabilities with longer half-lives and smaller changes in bubble size, which is closely related to their high wettability and cross-linking degree. This study may provide references for the application of soy protein nanoparticles as new foam stabilizers to replace the traditional foaming agents in the food industry.
Optimization of medium compositions and X-ray irradiation to enhance monacolin K production by Monascus purpureus in submerged fermentation

Abstract:The secondary metabolite Monacolin K (MK), derived from Monascus, exhibits potential lipid-lowering and anti-tumor effects. In this study, a mutant strain M. purpureus M400–6 was generated through X-ray treatment, resulting in an average MK production of 181.97 mg/L (a 54.20% increase compared to the parent strain M. purpureus M1) over five successive generations. Furthermore, optimization of medium compositions using response surface methodology led to a further enhancement in MK production by the mutant strain to 266.63 mg/L. Moreover, based on the outcomes of single-factor experiments, the addition of L-citrulline demonstrated a significantly higher level of enhancement in MK production. Therefore, a comparative transcriptome analysis was conducted to unveil the impact of L-citrulline on MK biosynthesis in M. purpureus. The data revealed a significant upregulation of glycolysis pathways, while oxidative phosphorylation including the TCA cycle and the electron transport chain, exhibited significant downregulation. Furthermore, the metabolisms of serine, glycine, threonine, alanine, aspartic acid and glutamate were found to be significantly disturbed. Additionally, the expressions of MPs and MK biosynthetic gene cluster showed a significant downregulation and upregulation respectively. Overall, these findings provide insights into enhancing MK production in submerged fermentation and elucidate the impact of L-citrulline on secondary metabolism.

Abstract:Action mechanism of Monascus pigments (MPs) production promoted by glycerol was investigated based on physiological and transcriptome analyses. Monascus pilosus MS-1 was cultured in synthetic media with 0, 40 or 160 g/L glycerol which marked as CK, G40 and G160, respectively. Monascus growth and MPs production were significantly enhanced by glycerol. The numbers of differential expression genes (DEGs) belonging to CK vs G40, CK vs G160 and G40 vs G160 groups were 955, 1847 and 537, respectively. DEGs found in CK vs G40 group involving in oxidation-reduction and glycolytic processes were up-regulated while that of RNA-DNA hybrid ribonuclease activity and RNA-dependent DNA biosynthetic process were down-regulated. The DEGs significantly enriched GO terms belonging to groups related to G160, nucleolus, 90S preribosome and small and large subunit precursors were down-regulated. KEGG analysis indicated that most of the DEGs belonging to CK vs G40 and CK vs G160 groups related to glycolysis and carbon metabolism were up-regulated. However, the DEGs belonging in groups related to G160 involved in ribosome biogenesis in eukaryotes and pyrimidine metabolism were down-regulated. Therefore, it proposed that MPs production promoted by glycerol mainly attribute to more substrates for MPs biosynthesis generated from glycolysis and carbon metabolism.

Mannitol improves Monascus pigment biosynthesis with rice bran as a substrate in Monascus purpureus

Abstract:To reduce the production cost of Monascus pigments (MPs), the utilization of rice bran (RB), an agricultural waste product, as a substrate in submerged fermentation was conducted in this study. To improve MP production, different nutritional ingredients including mannitol (Man), NH4NO3 (AN), ZnSO4 (Zn), and optimization (Opti), which was a synthesis of the three above ones, were added in rice bran (RB) medium. The yields of MPs, pigment constituents, and growth and development of Monascus purpureus M9 were investigated in this study. Man had the maximum color value of 3,532 U/g, which was 18.69 times more than that of RB and reached up to 76.65% of the value of rice (Rice) fermentation. Man significantly increased the production of two orange pigments, monascorubrin and rubropunctatin, of which the yields were 69.49 and 95.36% of the counterpart of Rice. The biomass and colony diameter of Opti presented the maximum value among different groups. AN and RB induced more asexual spore formation, whereas Opti and Man promoted sexual spore production. Comparative transcriptomic analysis showed that different nutritional ingredients led to changes in pigment production, promoting the growth and development of M. purpureus M9 through the regulation of related gene expression. Man and Opti improved MP production by regulating the primary metabolism, including the Embden-Meyerhof pathway (EMP), the pentose phosphate (PP) pathway, the tricarboxylic (TCA) cycle, fatty acid degradation (FAD), fatty acid biosynthesis (FAB), amino acid metabolism (AAM), and fructose and mannose metabolism (FMM), to provide the precursors (acetyl-CoA and malonyl-CoA) for MP biosynthesis. This study presents a low-cost method for increasing MP production and explains the molecular mechanisms of different nutritional ingredients for enhancing MP biosynthesis.
Effective enhancement of the ability of Monascus pilosus to produce lipid-lowering compound Monacolin K via perturbation of metabolic flux and histone acetylation modification

Abstract:Monacolin K (MK), also known as lovastatin, is a polyketide compound with the ability to reduce plasma cholesterol levels and many other bio-activities. Red yeast rice (also named Hongqu) rich in MK derived from Monascus fermentation has attracted widespread attention due to its excellent performance in reducing blood lipids. However, industrial Monascus fermentation suffers from the limitations such as low yield of MK, long fermentation period, and susceptibility to contamination. In this study, we firstly blocked the competitive pathway of MK biosynthesis to create polyketide synthase gene pigA (the key gene responsible for the biosynthesis of Monascus azaphilone pigments) deficient strain A1. Then, based on the strategies to increase precursor supply for MK biosynthesis, acetyl-CoA carboxylase gene acc overexpression strains C1 and C2 were constructed with WT and A1 as the parent, respectively. Finally, histone deacetylase gene hos2 overexpression strain H1 was constructed by perturbation of histone acetylation modification. HPLC detection revealed all these four strains significantly increased their abilities to produce MK. After 14 days of solid-state fermentation, the MK yields of strains A1, C1, C2, and H1 reached 2.03 g/100 g, 1.81 g/100 g, 2.45 g/100 g and 2.52 g/100 g, which increased by 28.5 %, 14.7 %, 43.9 % and 36.1 % compared to WT, respectively. RT-qPCR results showed that overexpression of hos2 significantly increased the expression level of almost all genes responsible for MK biosynthesis after 5-day growth. Overall, the abilities of these strains to produce MK has been greatly improved, and MK production period has been shortened to 14 days from 20 days, providing new approaches for efficient production of Hongqu rich in MK.
Enhancing extracellular monascus pigment production in submerged fermentation with engineered microbial consortia

Abstract:In this study, we investigated the impact of microbial interactions on Monascus pigment (MP) production. We established diverse microbial consortia involving Monascus purpureus and Lactobacillus fermentum. The addition of Lactobacillus fermentum (4% at 48 h) to the submerged fermentation of M. purpureus resulted in a significantly higher MP production compared to that achieved using the single-fermentation system. Co-cultivation with immobilized L. fermentum led to a remarkable increase of 59.18% in extracellular MP production, while mixed fermentation with free L. fermentum caused a significant decrease of 66.93% in intracellular MPs, contrasting with a marginal increase of 4.52% observed during co-cultivation with immobilized L. fermentum and the control group respectively. The findings indicate an evident enhancement in cell membrane permeability of M. purpureus when co-cultivated with immobilized L. fementum. Moreover, integrated transcriptomic and metabolomic analyses were conducted to elucidate the regulatory mechanisms underlying MP biosynthesis and secretion following inoculation with immobilized L. fementum, with specific emphasis on glycolysis, steroid biosynthesis, fatty acid biosynthesis, and energy metabolism.
Comparative Transcriptomic and Metabolomic Analyses Reveal the Regulatory Effect and Mechanism of Tea Extracts on the Biosynthesis of Monascus Pigments

Abstract:Monascus pigments (MPs) are natural edible pigments with high safety and strong function, which have been widely used in food and health products. In this study, different types of tea extracts (rich in polyphenols) were used to regulate the biosynthesis of MPs. The results showed that 15% ethanol extract of pu-erh tea (T11) could significantly increase MPs production in liquid fermentation of Monaco's purpureus M3. Comparative transcriptomic and metabolomic analyses combined with reverse transcription-quantitative polymerase chain reaction (RT-qPCR) were used to further explore the regulatory mechanism of T11 on the biosynthesis of MPs. Comparative transcriptomic analysis showed that there were 1503 differentially expressed genes (DEGs) between the Con group and the T11 group, which were mainly distributed in carbohydrate metabolism, amino acid metabolism, energy metabolism, lipid metabolism, metabolism of terpenoids and polyketides, etc. A total of 115 differential metabolites (DMs) identified by metabolomics between the Con and T11 groups were mainly enriched in glutathione metabolism, starch and sucrose metabolism, alanine, aspartic acid and glutamate metabolism and glycine, serine and threonine metabolism, etc. The results of metabolomics were basically consistent with those of gene transcriptomics, indicating that the regulatory effect of T11 on the biosynthesis of MPs is mainly achieved through affecting the primary metabolic pathway, providing sufficient energy and more biosynthetic precursors for secondary metabolism. In this study, tea extracts with low economic value and easy access were used as promoters of MPs biosynthesis, which may be conducive to the application of MPs in large-scale industrial production. At the same time, a more systematic understanding of the molecular regulatory mechanism of Monascus metabolism was obtained through multi-omics analysis.

Abstract:The study aimed to maximize the production of natural pigments from Monascus spp. using Saccharina japonica as a solid state fermentation substrate at optimized physical and nutritional conditions. The yield of yellow, orange, and red pigments of Monascus purpureus were 83.01, 80.07, and 79.87 AU gdfs−1 and Monascus kaoliang were 83.23, 77.22, and 75.09 AU gdfs−1, respectively, when providing external 0.8% glucose and 0.4% peptone at incubation temperature 30 °C for 20 days. Medium inoculum size of 3 mL and moisture content of 50% for M. purpureus and 60% for M. kaoliang showed the highest pigment production. Scanning electron microscopy showed better mycelial growth of M. purpureus than M. kaoliang on S. japonica. Monascus spp. fermented seaweed was highly bioactive showing DNA protection and antioxidant activity because of increasing phenolic and flavonoid contents during fermentation period. The highest reducing sugar content was found at 15 days and glucanase enzyme activity was found at 20 days of fermentation for both of the species. The pigments were separated by thin layer of chromatography and the Rf values were obtained 0.91 and 0.84 for yellow and orange pigments and two spots of 0.69 and 0.41 for red pigments. At different physicochemical stress, red pigment of M. purpureus showed more stability than that of M. kaoliang. So, it can be concluded that S. japonica was a very suitable substrate at optimum conditions for Monascus spp. natural pigment production.

Abstract:Monascus pigments (MPs) have been used in the food industry for more than 2,000 years and are known for their safety, bold coloring, and physiological activity. MPs are mainly yellow (YMPs), orange (OMPs), and red (RMPs). In this study, a mutant strain Monascus purpureus H14 with high production of water-soluble YMPs (WSYMPs, λ max at 370 nm) was generated instead of primary YMPs ( λ max at 420 nm), OMPs ( λ max at 470 nm), and RMPs ( λ max at 510 nm) produced by the parent strain M. purpureus LQ-6 through dual mutagenesis of atmospheric and room-temperature plasma and heavy ion beam irradiation (HIBI), producing 22.68 U/ml extracellular YMPs and 10.67 U/ml intracellular YMPs. WSYMP production was increased by 289.51% in optimal conditions after response surface methodology was applied in submerged fermentation. Application of combined immobilized fermentation and extractive fermentation improved productivity to 16.89 U/ml/day, 6.70 times greater than with conservative submerged fermentation. The produced WSYMPs exhibited good tone stability to environmental factors, but their pigment values were unstable to pH, light, and high concentrations of Ca 2+ , Zn 2+ , Fe 2+ , Cu 2+ , and Mg 2+ . Furtherly, the produced exYMPs were identified as two yellow monascus pigment components (monascusone B and C 21 H 27 NO 7 S) by UHPLC-ESI-MS. This strategy may be extended to industrial production of premium WSYMPs using Monascus .

Abstract:Extractive fermentation has been proven to be efficient in enhancing the secretion and production of secondary metabolites in submerged fermentation by Monascus spp., owing to increased cell membrane permeability and resolved product inhibition. In this study, we investigated the regulation effect of Triton X-100 on cell growth and secondary metabolite biosynthesis in submerged fermentation of M. purpureus DK. The results show that the maximum monascus pigments (MPs), citrinin (CIT) production, and specific growth rate are 136.86 U/mL, 4.57 mg/L, and 0.04 h-1, respectively, when 3 g/L of Triton X-100 is supplemented after fermentation for 10 d, and the extracellular MPs and CIT increased by 127.48% and 288.57%, respectively. RT-qPCR shows that the expression levels of MPs and CIT biosynthesis gene clusters are significantly upregulated, whereas those of glycolysis, tricarboxylic acid cycle, respiratory chains, and ATP synthase are downregulated. This study provides a vital strategy for extractive fermentation under extreme environmental conditions for further enhancing MP production.

Abstract:Monascus, a key player in fermented food production, is known for generating Monascus pigments (MPs) and monacolin K (MK), possessing bioactive properties. However, the limited stability of MPs and mycotoxin citrinin (CTN) constrain the Monascus industry. Extremolytes like ectoine, derived from bacteria, exhibit cytoprotective potential. Here, we investigated the impact of ectoine on Monascus purpureus ATCC 16365, emphasizing development and secondary metabolism. Exogenous 5 mM ectoine supplementation substantially increased the yields of MPs and MK (105%–150%) and reduced CTN production. Ectoine influenced mycelial growth, spore development, and gene expression in Monascus. Remarkably, ectoine biosynthesis was achieved in Monascus, showing comparable effects to exogenous addition. Notably, endogenous ectoine effectively enhanced the stability of MPs under diverse stress conditions. Our findings propose an innovative strategy for augmenting the production and stability of bioactive compounds while reducing CTN levels, advancing the Monascus industry.

Abstract:The chemical and enzymatic properties of 26 analogues of riboflavin are presented. These analogues include both endo- and exocyclically substituted isoalloxazines with redox potentials from -370 to -128 mV. Physical and chemical data such as the electronic absorption spectra, pKas, and redox potentials of the analogues are presented and are discussed with respect to preferred tautomeric and resonance forms. Like riboflavin, most of the analogues are shown to be catalytic oxidants of dihydro-5-deazaflavins. Analogue binding to egg white binding apoprotein has been quantitated and serves to determine the origins of binding site specificity for this protein. Nearly all of the analogues that possess D-ribityl groups are found to be processed to the FAD level by the flavokinase/FAD synthetase system of Brevibacterium ammoniagenes. Most extensively studied are the reactivities of the analogues with the NAD(P)H:flavin oxidoreductase of Beneckea harveyi. Many of the analogues are substrates in this enzymatic redox reaction, and a linear free energy-rate relation (log Vmax vs. E0' of the analogue) is seen that parallels similar relationships in the nonenzymatic oxidation of dihydro-5-deazaflavins. This suggests a common mechanism for the reactions of such diverse flavins as riboflavin, 5-deazariboflavin, and 1-deazariboflavin.
The effects of environmental factors on the synthesis of water-soluble Monascus red pigments via submerged fermentation: a review

Abstract:Monascus pigments (MPs) have been used as natural food pigments for many years. There is a high demand for Monascus red pigments (MRPs) to enhance color and for antibacterial and cancer prevention therapies in food and medicine. Most MRPs are not water soluble, and the yield of water-soluble MRPs is naturally low. On the other hand, water-soluble MRP is more cost effective for application in industrial mass production. Therefore, it is important to improve the yield of water-soluble MRPs. Environmental factors have a significant influence on the synthesis of water-soluble MRPs, which is crucial for the development of industrial production of water-soluble MRPs. This review introduces the biosynthetic pathways of water-soluble MRPs and summarizes the effects of environmental factors on the yield of water-soluble MRPs. Acetyl coenzyme A (acetyl-CoA) is a precursor for MPs synthesis. Carbon and nitrogen sources and the carbon/nitrogen ratio can impact MP production by regulating the metabolic pathway of acetyl-CoA. Optimization of fermentation conditions to change the morphology of Monascus can stimulate the synthesis of MPs. The appropriate choice of nitrogen sources and pH values can promote the synthesis of MRPs from MPs. Additives such as metal ions and non-ionic surfactants can affect the fluidity of Monascus cell membrane and promote the transformation of MRPs into water-soluble MRPs. This review will lay the foundation for the industrial production of water-soluble MRPs. © 2024 Society of Chemical Industry.
Phospholipid biosynthesis regulation for improving pigment production by Monascus in response to ammonium chloride stress

Abstract:In the actual industrial production process, the efficient biosynthesis and secretion of Monascus pigments (MPs) tend to take place under abiotic stresses, which often result in an imbalance of cell homeostasis. The present study aimed to thoroughly describe the changes in lipid profiles in Monascus purpureus by absolute quantitative lipidomics and tandem mass tag-based quantitative proteomics. The results showed that ammonium chloride stress (15 g/L) increased MP production while inhibiting ergosterol biosynthesis, leading to an imbalance in membrane lipid homeostasis in Monascus. In response to the imbalance of lipid homeostasis, the regulation mechanism of phospholipids in Monascus was implemented, including the inhibition of lysophospholipids production, maintenance of the ratio of PC/PE, and improvement of the biosynthesis of phosphatidylglycerol, phosphatidylserine, and cardiolipin with high saturated and long carbon chain fatty acids through the CDP-DG pathway rather than the Kennedy pathway. The inhibition of lysophospholipid biosynthesis was attributed to the upregulated expression of protein and its gene related to lysophospholipase NTE1, while maintenance of the PC/PE ratio was achieved by the upregulated expression of protein and its gene related to CTP: phosphoethanolamine cytidylyltransferase and phosphatidylethanolamine N-methyltransferase in the Kennedy pathway. These findings provide insights into the regulation mechanism of MP biosynthesis from new perspectives.IMPORTANCEMonascus is important in food microbiology as it produces natural colorants known as Monascus pigments (MPs). The industrial production of MPs has been achieved by liquid fermentation, in which the nitrogen source (especially ammonium chloride) is a key nutritional parameter. Previous studies have investigated the regulatory mechanisms of substance and energy metabolism, as well as the cross-protective mechanisms in Monascus in response to ammonium chloride stress. Our research in this work demonstrated that ammonium chloride stress also caused an imbalance of membrane lipid homeostasis in Monascus due to the inhibition of ergosterol biosynthesis. We found that the regulation mechanism of phospholipids in Monascus was implemented, including inhibition of lysophospholipids production, maintenance of the ratio of PC/PE, and improvement of biosynthesis of phosphatidylglycerol, phosphatidylserine, and cardiolipin with high saturated and long carbon chain fatty acids through the CDP-DG pathway. These findings further refine the regulatory mechanisms of MP production and secretion.

Abstract:Natural water-soluble Monascus pigments (WSMPs) have been in increasing demand but have not been able to achieve industrial production due to the low production rate. This study aimed to improve the biosynthesis and secretion of extracellular yellow pigments (EYPs) through submerged fermentation with Monascus ruber CGMCC 10,910 supplemented with sodium starch octenyl succinate (OSA-SNa). The results demonstrated that the yield was 69.68% and 48.89% higher than that without OSA-SNa in conventional fermentation (CF) and extractive fermentation (EF), respectively. The mainly increased EYP components were Y3 and Y4 in CF, but they were mainly Y1 and Y2 as well as secreted intracellular pigments, including Y5, Y6, O1, and O2, in EF. Scanning electron microscopy analysis revealed that the mycelium presented an uneven surface profile with obvious wrinkles and small fragments with OSA-SNa. It was found that a higher unsaturated/saturated fatty acids ratio in the cell membrane resulted in increased permeability and facilitated the export of intracellular yellow pigments into the broth with OSA-SNa treatment. In addition, a higher NAD+/NADH ratio and glucose-6-phosphate dehydrogenase activity provided a reducing condition for yellow pigment biosynthesis. Gene expression analysis showed that the expression levels of the key genes for yellow pigment biosynthesis were significantly upregulated by OSA-SNa. This study provides an effective strategy to promote the production of WSMPs by microparticle-enhanced cultivation using OSA-SNa.
