Abstract:The ubiquitous occurrence of microplastics (MPs) in the marine environment is raising concern for interactions with marine organisms. These particles efficiently adsorb persistent organic pollutants from surrounding environment and, due to the small size, they are easily available for ingestion at all trophic levels. Once ingested, MPs can induce mechanical damage, sub-lethal effects, and various cellular responses, further modulated by possible release of adsorbed chemicals or additives. In this study, ecotoxicological effects of MPs and their interactions with benzo(a)pyrene (BaP), chosen as a model compound for polycyclic aromatic hydrocarbons (PAHs) were investigated in Mediterranean mussels, Mytilus galloprovincialis. Organisms were exposed for 4 weeks to 10 mg/L of low-density polyethylene (LDPE) microparticles (2.34 * 107 particles/L, size range 20–25 μm), both virgin and pre-contaminated with BaP (15 μg/g). Organisms were also exposed for comparison to BaP dosed alone at 150 ng/L, corresponding to the amount adsorbed on microplastics. Tissue localization of microplastics was histologically evaluated; chemical analyses and a wide battery of biomarkers covering molecular, biochemical and cellular levels allowed to evaluate BaP bioaccumulation, alterations of immune system, antioxidant defenses, onset of oxidative stress, peroxisomal proliferation, genotoxicity, and neurotoxicity. Obtained data were elaborated within a quantitative weight of evidence (WOE) model which, using weighted criteria, provided synthetic hazard indices, for both chemical and cellular results, before their integration in a combined index. Microplastics were localized in hemolymph, gills, and especially digestive tissues where a potential transfer of BaP from MPs was also observed. Significant alterations were measured on the immune system, while more limited effects occurred on the oxidative status, neurotoxicity, and genotoxicity, with a different susceptibility of analyzed pathways, depending on tissue, time, and typology of exposure. Molecular analyses confirmed the general lack of significant transcriptional variations of antioxidant and stress genes. The overall results suggest that microplastics induce a slight cellular toxicity under short-term (28 days) exposure conditions. However, modulation of immune responses, along with bioaccumulation of BaP, pose the still unexplored risk that these particles, under conditions of more chronic exposure (months to years) or interacting with other stressors, may provoke long-term, subtle effects on organisms' health status.

Abstract:Pharmaceuticals and microplastics constitute potential hazards in aquatic systems, but their combined effects and underlying toxicity mechanisms remain largely unknown. In this study, a simultaneous characterization of bioaccumulation, associated metabolomic alterations and potential recovery mechanisms was performed. Specifically, a bioassay on Mediterranean mussels (Mytilus galloprovincialis) was carried out with polyethylene microplastics (PE-MPLs, 1 mg/L) and citalopram or bezafibrate (500 ng/L). Single and co-exposure scenarios lasted 21 days, followed by a 7-day depuration period to assess their potential recovery. PE-MPLs delayed the bioaccumulation of citalopram (lower mean at 10 d: 447 compared to 770 ng/g dw under single exposure), although reaching similar tissue concentrations after 21 d. A more limited accumulation of bezafibrate was observed overall, regardless of PE-MPLs co-exposure (<MQL-3.2 ng/g dw). Metabolic profiles showed a strong effect of pharmaceuticals, generally independent of PE-MPLs co-exposure. Alterations of the citrate cycle (bezafibrate exposure) and steroid and prostaglandin metabolism (citalopram and bezafibrate exposures) were highlighted. PE-MPLs alone also impacted metabolic pathways, such as neurotransmitters or purine metabolism. After depuration, relevant latent or long-lasting effects were demonstrated as, for instance, the effect of citalopram on neurotransmitters metabolism. Altogether, the observed molecular-level responses to pharmaceuticals and/or PE-MPLs may lead to a dysregulation of mussels' reproduction, energy metabolism, and/or immunity.

Abstract:Microplastics are ubiquitous in the world's oceans and pose serious environmental concerns, including their ingestion and the release of potentially toxic mixtures of intrinsic and extrinsic chemical compounds (i.e. leachates; MPLs). Mussels, as key intertidal bioengineers and filter-feeders are particularly susceptible to both exposure pathways. While the effects of microplastic ingestion have been widely investigated, research on the impacts of MPLs has only recently begun. This study examined the influence of MPLs derived from beached pellets collected in two separate regions, namely France and Portugal, on the respiration rates of two key ecosystem engineers, Mytilus edulis and Mytilus galloprovincialis . Possibly due to distinct mixtures of leached chemicals, unlike Portuguese-MPLs, exposure to French-MPLs significantly decreased the respiration rate of both mussel species. This research provides new insights into the physiological impacts of MPLs on bioengineer species, highlighting the importance of MP source and potential cascading effects at the ecosystem level. While we reported significant effects on mussel respiration after acute MPL exposure, future research should investigate long-term impacts and potential detoxification mechanisms to clarify the effects of MPs on mussel physiological performance and their potential consequences on specie fitness.

Abstract:Plastic pollution and invasive species are recognised as pervasive threats to marine biodiversity. However, despite the extensive on-going research on microplastics’ effects in the biota, knowledge on their combination with additional stressors is still limited. This study investigates the effects of polyamide microplastics (PA-MPs, 1 mg/L), alone and in combination with the toxic exudate from the invasive red seaweed Asparagopsis armata (2%), after a 96 h exposure, in the mussel Mytilus galloprovincialis. Biochemical responses associated with oxidative stress and damage, neurotoxicity, and energy metabolism were evaluated in different tissues (gills, digestive gland, and muscle). Byssus production and PA-MP accumulation were also assessed. Results demonstrated that PA-MPs accumulated the most in the digestive gland of mussels under PA-MP and exudate co-exposure. Furthermore, the combination of stressors also resulted in oxidative damage at the protein level in the gills as well as in a significant reduction in byssus production. Metabolic capacity increased in both PA-MP treatments, consequently affecting the energy balance in mussels under combined stress. Overall, results show a potential increase of PA-MPs toxicity in the presence of A. armata exudate, highlighting the importance of assessing the impact of microplastics in realistic scenarios, specifically in combination with co-occurring stressors, such as invasive species.

Abstract:Filter feeders are target species for microplastic (MP) pollution, as particles can accumulate in the digestive system, disturbing feeding processes and becoming internalized in tissues. MPs may also carry pathogens or pollutants present in the environment. This work assessed the influence of polystyrene (PS) MP size and concentration on accumulation and depuration time and the role of MPs as vectors for metallic (Cd) and organic (benzo(a)pyrene, BaP) pollutants. One-day exposure to pristine MPs induced a concentration-dependent accumulation in the digestive gland (in the stomach and duct lumen), and after 3-day depuration, 45 μm MPs appeared between gill filaments, while 4.5 μm MPs also occurred within gill filaments. After 3-day exposure to contaminated 4.5 μm MPs, mussels showed increased BaP levels whilst Cd accumulation did not occur. Here, PS showed higher affinity to BaP than to Cd. Three-day exposure to pristine or contaminated MPs did not provoke significant alterations in antioxidant and peroxisomal enzyme activities in the gills and digestive gland nor in lysosomal membrane stability. Exposure to dissolved contaminants and to MP-BaP caused histological alterations in the digestive gland. In conclusion, these short-term studies suggest that MPs are ingested and internalized in a size-dependent manner and act as carriers of the persistent organic pollutant BaP.

Abstract:Electrocardiograms (ECGs) are widely used by clinicians to identify the functional status of the heart. Thus, there is considerable interest in automated systems for real-time monitoring of arrhythmia. However, intra- and inter-patient variability as well as the computational limits of real-time monitoring poses significant challenges for practical implementations. The former requires that the classification model be adjusted continuously, and the latter requires a reduction in the number and types of ECG features, and thus, the computational burden, necessary to classify different arrhythmias. We propose the use of adaptive learning to automatically train the classifier on up-to-date ECG data, and employ adaptive feature selection to define unique feature subsets pertinent to different types of arrhythmia. Experimental results show that this hybrid technique outperforms conventional approaches and is therefore a promising new intelligent diagnostic tool.

Abstract:Microplastic (MP) contamination in the aquatic environment is a cause of concern worldwide since MP can be taken up by different organisms, altering different biological functions. In particular, evidence is accumulating that MP can affect the relationship between the host and its associated microbial communities (the microbiome), with potentially negative health consequences. Synthetic microfibers (MFs) represent one of the main MPs in the marine environment, which can be accumulated by filter-feeding invertebrates, such as bivalves, with consequent negative effects and transfer through the food chain. In the mussel Mytilus galloprovincialis, polyethylene terephthalate (PET) MFs, with a size distribution resembling that of an MF released from textile washing, have been previously shown to induce multiple stress responses. In this work, in the same experimental conditions, the effects of exposure to PET-MF (96 h, 10, and 100 μg/L) on mussel hemolymph microbiome were evaluated by 16S rRNA gene amplification and sequencing. The results show that PET-MF affects the composition of bacterial communities at the phylum, family and genus level, with stronger effects at the lowest concentration tested. The relationship between MF-induced changes in hemolymph microbial communities and responses observed at the whole organism level are discussed.

Abstract:Microplastic contamination is a growing marine environmental issue with possible consequences for seafood safety. Filter feeders are the target species for microplastic (MPs) pollution because they filter large quantities of seawater to feed. In the present study, an experimental contamination of Mytilus galloprovincialis was conducted using a mixture of the main types of MPs usually present in the seawater column (53% filaments, 30% fragments, 3% granules) in order to test the purification process as a potential method for removing these contaminants from bivalves intended for human consumption. A set of molecular biomarkers was also evaluated in order to detect any variations in the expression levels of some genes associated with biotransformation and detoxification, DNA repair, cellular response, and the immune system. Metabolic responses of the marine mussel Mytilus galloprovincialis after exposure to microplastics of different shapes and sizes

Abstract:Microplastics (MP) are ubiquitous pollutants with diverse shapes, sizes, and characteristics that pose critical risks to marine organisms and the environment. In this study, we used the Mediterranean mussel Mytilus galloprovincialis as a marine benthic organism model to investigate the metabolic consequences of exposure to different polyethylene terephthalate MP sizes and shapes: round (27-32 μm), small fibers (200-400 μm), large fibers (3000 μm), small fragments (20 μm), medium fragments (45-75 μm), and large fragments (>150 μm). After exposure to high concentrations (100 mg L-1) of MP for 14 days, round and small fiber-type MP were highly accumulated in mussels. Metabolomic analysis revealed that exposure to round and small fiber-type MP induced significant changes in 150 metabolites. Partial least squares-discriminate analysis (PLS-DA) showed that the round and small fiber MP treatment groups displayed similar cluster patterns that differed from those of the control group. In addition, only 22 annotated metabolites related to histidine, valine, leucine, and isoleucine degradation/biosynthesis and vitamin B6 and aminoacyl-tRNA biosynthesis were significantly affected by round or small fiber-type MP. Among the histidine metabolites, round and small fiber-type MP upregulated the levels of L-histidine, L-glutamate, carnosine, imidazole-4-acetaldehyde, 4-imidazolone-5-propanoate, and methylimidazole acetaldehyde and downregulated methylimidazole acetic acid and N-formimino-L-glutamate. These results suggest novel insights into the potential pathways through which MP of specific sizes and shapes affect metabolic processes in mussels.
Major characteristics of microplastics in mussels from the Portuguese coast

Abstract:The present study reports the quantity, shape, colour and chemical properties of microplastics (MP) and MP-like in whole soft tissues of the mussel Mytilus spp. collected in January and February 2019 from four natural banks in the Portuguese coast. Three sites are located in estuarine areas influenced by anthropogenic pressures and freshwater discharges, and one in the coast far from urbanised areas. An alkaline digestion (KOH) of biological tissues was used and a polymeric identification of 20% of the visually sorted particles was achieved using the Fourier-transform mid-infrared spectroscopy (FT-MIR). MP and MP-like concentrations ranged from 0.54 to 3.0 items g-1 without significant differences among the sites. Particle size varied from 36 to 4439 μm, being fibers the most abundant shape (50%) followed by films (22%) and spherules (18%). FT-MIR revealed that 69% of the analysed particles were plastic, being identified six polymers and two polymeric blends, and 32% were cellulose-based materials. Fibers identified in mussel tissues were mainly composed of cotton and viscose (77%). This study emphasizes the importance of the polymer's spectroscopic identification after microscopic observation to recognise MP.
A Comparative Assessment of the Chronic Effects of Micro- and Nano-Plastics on the Physiology of the Mediterranean Mussel Mytilus galloprovincialis

Abstract:The ocean contamination caused by micro- and nano-sized plastics is a matter of increasing concern regarding their potential effects on marine organisms. This study compared the effects of a 21-day exposure to 1.5, 15, and 150 ng/L of polystyrene microplastics (PS-MP, 3-μm) and nanoplastics (PS-NP, 50-nm) on a suite of biomarkers measured in the Mediterranean mussel Mytilus galloprovincialis. Endpoints encompassed immunological/lysosomal responses, oxidative stress/detoxification parameters, and neurotoxicological markers. Compared to PS-MP, PS-NP induced higher effects on lysosomal parameters of general stress. Exposures to both particle sizes increased lipid peroxidation and catalase activity in gills; PS-NP elicited greater effects on the phase-II metabolism enzyme glutathione S-transferase and on lysozyme activity, while only PS-MP inhibited the hemocyte phagocytosis, suggesting a major role of PS particle size in modulating immunological/detoxification pathways. A decreased acetylcholinesterase activity was induced by PS-NP, indicating their potential to impair neurological functions in mussels. Biomarker data integration in the Mussel Expert System identified an overall greater health status alteration in mussels exposed to PS-NP compared to PS-MP. This study shows that increasing concentrations of nanoplastics may induce higher effects than microplastics on the mussel’s lysosomal, metabolic, and neurological functions, eventually resulting in a greater impact on their overall fitness.

Abstract:Microplastics (MPs) constitute the predominant plastic type in marine environments. Since they occupy the same size fraction of sediment particles and planktonic organisms they are potentially bioavailable to a broad scope of organisms, such as filter feeders, which are particularly vulnerable to MP ingestion. To understand the potential impact of MPs in filter feeders it is essential to clarify the uptake, accumulation patterns and elimination rates with time of MPs. The aim of this study was to determine the depuration dynamics and accumulation in tissues of mussels Mytilus galloprovincialis exposed during 24 h to different size polystyrene MPs (1 μm and 10 μm), and depurated for a maximum of 7 days (T = 24 h, T = 48 h and T = 7 d). Mussels were chemically digested with KOH 10% and filtered to quantify the number of MP ingested, and they were cryostat sliced for MP localization in tissues. Both MP sizes were quantified in all depuration times, but mussels accumulated significantly higher quantities of 10 μm MPs throughout depuration compared to 1 μm MPs. A significant decrease was observed after 7 d depuration in mussels exposed to 10 μm. Mussels removed the same amount of 1 and 10 μm MPs after 7 days depuration. However, the depuration dynamics differed for each size-MPs and showed to be size-dependent. Most of both size MPs were eliminated in the first 24 h, but 1 μm MPs showed to pass faster through the digestive tract than 10 μm MPs. MPs of 1 μm and 10 μm were localized mainly in the lumen and a few in the epithelium of the digestive tract (stomach, intestine and digestive gland) during the depuration and in the gills after the exposure; as confirmed by Raman spectroscopy. The usage of chemical digestion and histological analysis as complementary techniques show to be suitable to infer the depuration dynamics of MPs in mussels.

Abstract:This study investigates the potential of MPs as carriers of pollutants as they can strengthen bioaccumulation of toxic metals on marine organisms. For the first time, the interaction of the metal palladium (Pd) with the widespread MPs, both with increasing concentrations in water environments from anthropogenic sources, was tested. Mytilus galloprovincialis , an important seafood product, was exposed to Pd (24 h) in two ways: water-dissolved and MPs-adsorbed, with depuration followed for 144 h. Quantification of Pd in tissues shown an accumulation 2–3 times higher (59 % of initial Pd) for mussels exposed to MPs-adsorbed Pd and higher in digestive gland than when exposed to water-dissolved Pd (25 %; higher in gills). Additionally, it was demonstrated that Pd induced oxidative stress and altered the feeding behavior of mussels. Therefore, this work support MPs as being vectors of metals (i.e. Pd) to enhance their bioaccumulation on marine organisms which highlights ecological risk of these emerging pollutants.

Abstract:The microplastic "vector effect" has received increasing attention. The aim of this study was to investigate the influence of polyethylene microplastic beads (PE MP) on accumulation and associated oxidative stress responses attributed to fluoranthene (Flu) in blue mussels, Mytilus edulis. Blue mussels were exposed for 96 h to four treatment groups: Flu-only, MP-only, Flu and MP coexposure, and Flu-incubated MP. Treatments were conducted at a low and high concentration (50 μg/L and 100 Flu μg/L and 100, and 1000 MP/mL). Results demonstrated that in both the gill and digestive gland, coexposure did not markedly affect Flu uptake, but this treatment significantly decreased tissue Flu concentrations. Antioxidant responses including activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidases (GPx), and levels of total glutathione (GSH) in both gills and digestive glands were significantly altered suggesting a perturbation of redox state induced by the exposure conditions. Although individual biomarkers varied, the biomarker profile enabled certain generalizations to be made. Antioxidant responses occurred more likely in gill tissue than in digestive gland. Individual contaminant exposures to Flu or MP led to varying responses, but coexposures and incubated exposures did not result in additive or synergistic effects. Exposure concentrations (i.e., low or high treatments) were not a consistent a predictor of response; and the internal Flu dose did not consistently predict outcome of various biomarkers. Importantly, MP-only exposure appeared to be capable of eliciting direct effects on the oxidative stress system as demonstrated by the activities of CAT and GPx. These findings warrant further investigation.

Abstract:Microplastics (MPs) disturb the normal activity of aquatic organisms at different levels, causing physiological stress and altering feeding, growth, and reproduction. Alterations of epigenetic patterns due to exposure to MPs have scarcely been studied in invertebrates. In this study, Mytilus galloprovincialis mussels (N = 61) were intermittently exposed to different concentrations of pure polystyrene microbeads for three weeks. The concentrations used in this research were similar to those currently found in certain polluted environments (E1), as well as higher doses to which mussels could be further exposed (E2 and E3). After exposure period, the global methylation patterns were investigated using Amplified Fragment Length Polymorphism (AFLPs). Significantly lower methylation was found in exposed groups compared to the control group. The level of hypomethylation increased with the concentration of microbeads. Similar results were found from field samples inhabiting two sites differentially MPs-polluted. The implications of this discovery were analysed and discussed, noting the already known effects of MPs on metabolism and cell division. Further studies on this and other sentinel organisms are recommended to understand the response of the aquatic species to the currently increasing MPs pollution.

Abstract:Results of this research focuses on microplastic contents (levels, type, size, colour) in maricultured and natural mussels (Mytilus galloprovincialis) from different Italian stocks. No significant differences were found among maricultured and natural stocks. All recovered MPs are filaments ranging within 750-6000 μm of maximum length (average values 1150-2290 μm). Feeding raw mussel could produce median MP intakes of 6.2-7.2 items/g w.w. Concerning human exposure by diet, both raw and cooked values are important. Some preliminary tests performed in this study evidenced that the cooking process determined lower MPs levels (-14%) in cooked tissues compared to raw ones, MPs were recorded in cooking water and were characterized by a lower size than in raw mussels. Results obtained by this study represent an important baseline on MPs level to evaluate environmental and human exposure risks by diet.

Abstract:Lately, the role of microplastics (MP) as vectors for dissolved contaminants and as vehicle for their transfer to aquatic organisms has received attention. Similarly to MP, other inorganic and organic particles may act as passive samplers. However, limited comparative knowledge exists at this respect. In the present study we have comparatively investigated the risk for mussel of MP and the pesticide chlorpyrifos (CPF) alone and in combination with MP and phytoplankton particles of microalgae (MP-CPF and MA-CPF, respectively). We selected MP and microalgae of similar size to expose mussel to the same volume of particles (≈1.5 mm 3 L −1 ≈ equivalent to 1.5 mg MP L −1 ) and the same concentration of contaminant (CPF, 7.6 μg L −1 ). MP were virgin HDPE microparticles (≤10 μm) while the microalgae species was Isochrisis galbana (4-8 μm). Mussels were exposed for 21 days to MP, CPF, MP-CPF and MA-CPF. Then, a suite of neurotoxicity, oxidative stress and oxidative damage biomarkers were measured in samples collected at day 7 and 21. Additionally, these biochemical markers were assessed in an integrated manner with others measured at physiological, immune and cell component level in the same organisms, previously published. Overall, MP did not elicit significant alterations on the majority of parameters measured. In contrast, mussels exposed to CPF, MA-CPF and MP-CPF showed evidence of neurotoxicity and oxidant imbalance at day 7, added to a detrimental physiological condition and immune imbalance at day 21. At the latter time MP-CPF mussels showed greater alterations than CPF or MA-CPF mussels. This suggested a synergistic toxicity of MP combined with CPF greater than that produced by the contaminants alone (MP or CPF) or by MA combined with CPF.
