Abstract:This work reported the development, performance and microbial community of microalgal-bacterial biofilms cultivated in a continuous-flow photoreactor for municipal wastewater treatment under various conditions. Results showed that microalgal-bacterial biofilms were successfully developed at a HRT of 9 h without external aeration, with a biofilm concentration of around 4690 mg/L being achieved in the steady-state. It was found that further increase of HRT to 12 h did not improve the overall accumulation of biofilm, whereas the growth of microalgae in biofilms was faster than bacteria in the initial stage, indicated by an increased chlorophyll- a&b content in biofilms. After which, the chlorophyll- a&b content in biofilms gradually stabilized at the level comparable with the seed, suggesting that there was a balanced distribution of microalgae and bacteria in biofilms. About 90% of TOC, 71.4% of total nitrogen and 72.6% of phosphorus were removed by microalgal-bacterial biofilms mainly through assimilation in the steady-state photoreactor run at the HRT of 12 h with external aeration. The community analysis further revealed that Cyanobacteria and Chloroflexi were the main components, while Chlorophyta appeared to be the dominant eukaryotic algal community in biofilms. This study could offer new insights into the development of microalgal-bacterial biofilms in a continuous-flow photoreactor for sustainable low-carbon municipal wastewater treatment.

Abstract:Microalgal-indigenous bacterial wastewater treatment (MBWT) emerges as a promising approach for the concurrent removal of nitrogen (N) and phosphorus (P). Despite its potential, the prevalent use of MBWT in batch systems limits its broader application. Furthermore, the success of MBWT critically depends on the stable self-adaptation and synergistic interactions between microalgae and indigenous bacteria, yet the underlying biological mechanisms are not fully understood. Here we explore the viability and microbial dynamics of a continuous flow microalgae-indigenous bacteria advanced wastewater treatment system (CFMBAWTS) in processing actual secondary effluent, with a focus on varying hydraulic retention times (HRTs). The research highlights a stable, mutually beneficial relationship between indigenous bacteria and microalgae. Microalgae and indigenous bacteria can create an optimal environment for each other by providing essential cofactors (like iron, vitamins, and indole-3-acetic acid), oxygen, dissolved organic matter, and tryptophan. This collaboration leads to effective microbial growth, enhanced N and P removal, and energy generation. The study also uncovers crucial metabolic pathways, functional genes, and patterns of microbial succession. Significantly, the effluent NH 4 + -N and P levels complied with the Chinese national Class-II, Class-V, Class-IA, and Class-IB wastewater discharge standards when the HRT was reduced from 15 to 6 h. Optimal results, including the highest rates of CO 2 fixation (1.23 g L −1 ), total energy yield (32.35 kJ L −1 ), and the maximal lipid (33.91 %) and carbohydrate (41.91 %) content, were observed at an HRT of 15 h. Overall, this study not only confirms the feasibility of CFMBAWTS but also lays a crucial foundation for enhancing our understanding of this technology and propelling its practical application in wastewater treatment plants.

Abstract:The piggery digestate of high ammonia was mixed with the anoxic aerated effluent of high nitrate and phosphorus, to cultivate a microalgal-bacterial consortium for simultaneous pollution removal and resource recovery. The highest removal of total inorganic nitrogen was achieved at 324.77 mg/L in 40% piggery digestate mixed with 60% anoxic aerated effluent, along with the most microalgae biomass production. The crude protein and fatty acids of C14-C20 in microalgae cells were 21.80% and 69.78%, indicating that this mixing strategy could produce abundant microalgal biomass suitable for biofuel generation and animal feed. High-throughput sequencing showed that microbial diversity increased and Paenibacillus, Thiopseudomonas and Pseudomonas were the dominant species promoting microalgal growth. Overall, these results provided a new insight of mixing two types of wastewaters for cultivating microalgal-bacterial consortia, to remove contamination and recover nutrients simultaneously.

Abstract:Microalgae-bacteria consortia is considered to be an efficient way to improve wastewater treatment. However, the interactions between microalgae and bacteria were not clear. In this study, commercially available bacterial inoculum were used as bio-augmentation to improve the performance of microalgae-based wastewater treatment. Co-occurrence network analysis was used to investigate the interactions between microalgae and bacteria. Results showed that bio-augmentation could increase both microalgal growth and wastewater treatment efficiency. Microalgal biomass increased by 1.2 times. Removal efficiencies of total organic carbon, ammonium nitrogen and total phosphorus increased by 5.27%, 3.41%% and 5.74%, respectively. Decreasing trends for Cyanobacteria and Salinarimonas but increasing trends for Cyclobacteriaceae were observed in the microalgal culture system with bio-augmentation. Greater variations of microbial community were found in the treatment without bio-augmentation. Co-occurrence network analysis showed that the main functions of microbial community in the treatment with bio-augmentation were both nutrient removal and organic pollutant degrading. For the treatment without bio-augmentation, main function of microbial community was nutrient removal. It indicated that bio-augmentation increased the number of organic degrading bacteria, such as Bacillus and Paenibacillaceae. The interactions between these bacteria and microalgae promoted the degradation of organic pollutants in the wastewater, providing sufficient inorganic carbon sources for the photosynthesis of microalgae and thus promoting the growth of microalgae and the removal of nitrogen and phosphorus from wastewater.

Abstract:This study investigated the possibility of coupling anaerobic hydrolysis in an anaerobic membrane bioreactor (AnMBR) with mixotrophic microalgae cultivation in a membrane photobioreactor (MPBR) for the sustainable treatment of municipal wastewater. Using the hydrolyzed wastewater discharged from AnMBR, Chlorella pyrenoidosa in MPBR grew in a mixotrophic mode and realized rapid growth. During the stable operation, MPBR achieved average carbon capture rate of 42.82 mg L−1 d−1 and algal lipid production rate of 19.66 mg L−1 d−1. The average reduction in TN, TP, and TOC during stable operation was 96.7%, 98.0%, and 95.9%, respectively. Mass balance analysis showed that the overall system captured 14.76 mg of carbon from the atmosphere per liter of wastewater treated. Therefore, this AnMBR-MPBR hybrid system simultaneously realized advanced treatment of municipal wastewater, efficient production of algal lipid, and carbon capture from atmosphere, and thus has a good potential in the sustainable treatment of municipal wastewater.

Abstract:Low carbon/nitrogen (C/N) wastewater treatment is an emerging issue for conventional treatment technologies. To achieve this goal, a novel membrane photobioreactor (MPBR), integrating microalgae consortium (MC) and activated sludge (AS) phases, was investigated. The separation of the MC and AS phases could improve the removal of pollutants in low C/N wastewater, which could achieve the rapid denitrogenation through NH4 +-N assimilation in MC phase, and effectively reduce organic matter concentration in AS phase, resulting in better nutrient recovery, lower costs, and stable pH. The results in phase I showed that the pollutants, such as chemical oxygen demand (COD), ammonium (NH4 +-N), and total phosphorus (TP), met the Environmental Quality Standards for Surface Water (GB3838–2002) by MPBR. The AS phase with low DO, which could enrich heterotrophic bacteria (e.g., Bacteroidetes vadinHA17, Spirochaeta and Ahniella) and nitrogen cycling bacteria (e.g., Mycobacterium, Bacillus and Ellin6067), could promote the removal of COD and NO3 --N in the continuous effluent. Based on the co-occurrence pattern analysis, the main influencing factor of MC and AS phases was the reduction of COD, which also illustrated the synergistic removal of organic matter by the cooperative interaction of microalgae and heterotrophic bacteria. From a genetic perspective, the internal circulation mode also enhanced microbial nitrogen metabolism in the MPBRs, with higher abundances of nitrification- and denitrification-related functional genes in the MC phase, and higher abundances of nitrification, dissimilatory nitrate reduction, and denitrification genes in the AS phase. This study provided an experimental and theoretical basis for MPBR in low-C/N wastewater treatment.

Abstract:Municipal wastewater (MWW) was treated by a sequential pilot microalgal cultivation process. The cultivation was performed inside a specifically designed low-cost photobioreactor (PBR) system. A microalgal consortium 2:1 was developed using Tetraselmis indica (TS) and Picochlorum sp. (PC) in the first stage and PC:TS (2:1) in the second stage and the nutrient removal efficiency and biomass production and biomolecules production was evaluated and also compared with monoculture in a two-stage sequential cultivation system. This study also investigated the effect of seasonal variations on microalgae growth and MWW treatment. The results showed that mixed microalgal consortium (TS:PC) had higher nutrient removal efficiency, with chemical oxygen demand (COD), total phosphate (TP), and total nitrate (TN) removal efficiencies of 78.50, 84.49, and 84.20%, respectively, and produced a biomass of 2.50 g/L with lipid content of 37.36% in the first stage of cultivation under indoor conditions. In the second stage of indoor cultivation, the PC:TS consortium demonstrated maximum COD, TP, and TN removal efficiencies of 92.49, 94.24, and 94.16%, respectively. It also produced a biomass of 2.65 g/L with a lipid content of 40.67%. Among all the seasonal variations, mass flow analysis indicated that the combination of mixed consortium-based two-stage sequential process during the winter season favored maximum nutrient removal efficiency of TN i.e. 88.54% (84.12 mg/L) and TP i.e., 90.18% (43.29 mg/L), respectively. It also enhanced total biomass production of 49.10 g in 20-L medium, which includes lipid yield ∼15.68 g compared to monoculture i.e., 82.06% (78.70 mg/L) and 82.87% (40.26 mg/L) removal of TN and TP, respectively, and produced biomass 43.60 g with 11.90 g of lipids.

Abstract:Microalgal-bacterial biofilms have been increasingly considered of great potential in wastewater treatment due to the advantages of microalgal-bacterial synergistic pollutants removal/recovery, CO 2 sequestration, and cost-effective biomass-water separation. However, such advantages may vary widely among different types of microalgal-bacterial biofilms, as the biofilms could be formed on different shapes and structures of attachment substratum, generating "false hope" for certain systems in large-scale wastewater treatment if the operating conditions and pollutants removal properties are evaluated based on the general term "microalgal-bacterial biofilm". This study, therefore, classified microalgal-bacterial biofilms into biofilms formed on 2D substratum, biofilms formed on 3D substratum, and biofilms formed without substratum (i.e. microalgal-bacterial granular sludge, MBGS). Biofilms formed on 2D substratum display higher microalgae fractions and nutrients removal efficiencies, while the adopted long hydraulic retention times were unacceptable for large-scale wastewater treatment. MBGS are featured with much lower microalgae fractions, most efficient pollutants removal, and acceptable retention times for realistic application, yet the feasibility of using natural sunlight should be further explored. 3D substratum systems display wide variations in operating conditions and pollutants removal properties because of diversified substratum shapes and structures. 2D and 3D substratum biofilms share more common in eukaryotic and prokaryotic microbial community structures, while MGBS biofilms are more enriched with microorganisms favoring EPS production, biofilm formation, and denitrification. The specific roles of stratified extracellular polymeric substances (EPS) in nutrients adsorption and condensation still require in-depth exploration. Nutrients removal uncertainties caused by microalgal-bacterial synergy decoupling under insufficient illumination, limited microbial community control, and possible greenhouse gas emission exacerbation arising from microalgal N 2 O generation were also indicated. This review is helpful for revealing the true potential of applying various microalgal-bacterial biofilms in large-scale wastewater treatment, and will provoke some insights on the challenges to the ideal state of synergistic pollutants reclamation and carbon neutrality via microalgal-bacterial interactions.

Abstract:A wastewater-born and settleable algal-bacterial culture, cultivated in a stirred tank photobioreactor under lab conditions, was used to remove the carbon and nutrients in municipal wastewater and accumulate biomass simultaneously. The algal-bacterial culture showed good settleable property and could totally settle down over 20 min, resulting in a reduction of total suspended solids from an initial 1.84 to 0.016 g/l. The average removal efficiencies of chemical oxygen demand, total kjeldahl nitrogen and phosphate were 98.2 ± 1.3%, 88.3 ± 1.6% and 64.8 ± 1.0% within 8 days, respectively, while the average biomass productivity was 10.9 ± 1.1 g/m(2) · d. Accumulation into biomass, identified as the main nitrogen and phosphorus removal mechanism, accounted for 44.9 ± 0.4% and 61.6 ± 0.5% of total inlet nitrogen and phosphorus, respectively. Microscopic analysis showed the main algae species in the bioreactor were filamentous blue-green algae. Furthermore, denaturing gradient gel electrophoresis and 16S rDNA gene sequencing revealed that the main bacteria present in the photobioreactor were consortia with sequences similar to those of Flavobacteria, Gammaproteobacteria, Bacteroidia and Betaproteobacteria. This study explores a better understanding of an algae-bacteria system and offers new information on further usage of biomass accumulated during treatment.

Abstract:The use of microalgal-bacterial consortiums has attracted interest owing to their potential in sewage treatment. However, the performance of sequencing batch reactor (SBR) coupled with microalgal-bacterial consortiums has not been investigated extensively. The present study investigates two SBRs namely R1 and R2, operated for 60 d; R1 was supplemented with microalgal-bacterial consortium, while R2 contained conventional activated sludge. The average removal efficiency for chemical oxygen demand, ammonia nitrogen, total nitrogen, and total phosphorus in R1 were 91.7%, 98.8%, 69.9%, and 64.3%, which improved by 4.9%, 3.4%, 10.1%, and 8.6% compared to R2, respectively. The maximum chlorophyll content was 345.77 mg L−1 and the maximum lipid content was 340 mg L−1 in R1 which indicates the potential of algal biomass production. Microbial analysis showed the community characteristics and the population dynamics in R1 and R2 at different stages, and revealed the influence of the microalgal-bacterial consortium on microbial community evolution. These results suggest that microalgal-bacterial consortium-based systems can be used to treat wastewater and efficiently utilize sewage as a source of high-value compounds.

Abstract:A novel vertical rotating microalgae-bacteria symbiotic biofilm reactor was built to treat the mixed wastewater containing municipal and soybean soaking wastewater. The reactor was operated in both sequential batch and semi-continuous modes. Under the sequential batch operation mode, the maximum removal rates for Chemical Oxygen Demand (COD), Total Nitrogen (TN), Total Phosphorus (TP), and Ammonia Nitrogen (NH 4 + -N) of the mixed wastewater were 95.6 %, 96.1 %, 97.6 %, and 100 %, respectively. During the semi-continuous operation, the water discharge indices decreased gradually and eventually stabilized. At stabilization, the removal rates of COD, TN, and NH 4 + -N achieved 98 %, 95 %, and 99.9 %, respectively. The maximum biomass productivity of the biofilm was 2.69 g·m −2 ·d -1 . Additionally, the carbohydrate, protein and lipid comprised approximately 22 %, 51 % and 10 % of the dry weight of Chlorella . This study demonstrates the great potential of the microalgae-bacteria symbiotic biofilm system to treat food and domestic wastewater while harvesting microalgal biomass.

Intensified microalgae production and development of microbial communities on suspended carriers and municipal wastewater

Abstract:Wastewater represents an alternative source of nutrients in which to grow microalgae, whilst improving the quality of the wastewater, and reducing the downstream treatment required. However, commercialisation of microalgal cultures for such duties faces a number of challenges, predominantly high cost and low productivity. Suspended-solid reactors (ssPBR) can reduce the operational costs, while promoting attached and suspended microalgae growth. In the present study, a novel approach was developed by integrating microalgal wastewater treatment with carrier systems to favour the growth of both attached and suspended cells of T. obliquus. This study found that T. obliquus was able to uptake nutrients from municipal wastewater, achieving removals of 99.3-99.9 % NH3-N, 54.5-88.5 % PO43- and 92.8-94.5 % DTC. The addition of a 12.5 % volumetric fill ratio of carriers in ssPBRs produced higher microalgal cell productivity (1.2·106 ± 2.5·105 cell ml-1 d-1) than the control (4.3·105 ± 2.8·105 cell ml-1 d-1). MinION nanopore sequencing was conducted to assess the impact of microalgal and carrier treatment on wastewater bacterial communities. It was found not only that bacterial communities had changed after the treatment but also the ones attached differed from the ones suspended. Untreated wastewater was characterised by the abundance of sewer bacteria genera such as Aliarcobacter and Arcobacter, whilst, after treatment, microbial communities were characterised by the presence of photosynthetic freshwater (Limnococcus, Stanieria) and bioremediation-like bacteria genera (Pseudomonas, Rheinheimera). In conclusion, the addition of 12.5 % fill carrier ratio increased microalgal productivity, while stimulating changes in the algal microbiome, and creating distinctly different populations in the free and attached environments.

Abstract:The rise in living standards has generated a demand for higher aquatic environmental quality. The microalgal community and the surrounding organic molecules, environmental factors and microorganisms, such as bacteria, are together defined as the phycosphere. The bacteria in the phycosphere can form consortia with microalgae through various forms of interaction. The study of the species in these consortia and their relative proportions is of great significance in determining the species and strains of stable algae that can be used in sewage treatment. This article summarizes the following topics: the interactions between microalgae and bacteria that are required to establish consortia; how symbiosis between algae and bacteria is established; microalgal competition with bacteria through inhibition and anti‐inhibition strategies; the influence of environmental factors on microalgal‐bacterial aggregates, such as illumination conditions, pH, dissolved oxygen, temperature and nutrient levels; the application of algal‐bacterial aggregates to enhance biomass production and nutrient reuse; and techniques for studying the community structure and interactions of algal‐bacterial consortia, such as microscopy, flow cytometry, and omics.

Abstract:Microalgae have been developed as promising candidates for bioenergy production, coupled with answering challenges related to water pollution and global warming. When combined with wastewater, microalgal biomass production could be freed from a strong dependence on freshwater and chemical nutrients, as well as achieve the additional advantage of wastewater reclamation. However, despite those dual benefits, certain limitations on the growing of algae in wastewater remain to be resolved, including the inevitable presence of bacteria in wastewater, which influences biomass productivity and quality in various ways. Pointing at microalgal-bacterial relationships, this study provides an updated review of the application of microalgal-bacterial consortia (MABC) to benefit biomass yield and harvest, and in wastewater remediation, focusing on the main interactions established between the microorganisms integrated within MABC and the factors influencing the behaviours of MABC. The challenges faced by the MABC biotechnology are also discussed, which are primarily rooted in undesirable bacteria that parasitically eat microalgal products and inhibit algal growth through nutrient competition, lysate exudation, or reducing the algal resistance to biotic stress. However, there is a lack of systematic studies on maintaining stable and effective operation of MABCs for wastewater cultivation and high-value bioproduct generation. Knowledge gaps are identified as including systematic information about the responses of MABC to culture conditions and wastewater-borne bacterial communities, and the metabolic mechanisms underpinning the interactions between algae and bacteria in wastewater. Further research focuses and methodologies are proposed in this review, making full use of the advent of omics and computational technology.

Abstract:The treatment of micro-polluted water via a constructed wetland (CW), such as fishpond aquaculture wastewater, encounters problems such as low effectiveness and poor stability. In this study, an emerging algae-bacteria microbial fuel cell (AB-MFC) was coupled with CW to strengthen the treatment of fishpond aquaculture wastewater. Compared with those in the control group (without AB-MFC), the average removal efficiencies of chemical oxygen demand, total phosphorus, total nitrogen, NH 4 + -N, NO 3 — N, and NO 2 — N in AB-MFC-CW were enhanced by 23.84%, 21.44%, 15.07%, 16.91%, 15.02%, and 9.83%, respectively. High-throughput sequencing revealed that Acinetobacter , Cyanobium, Pseudomonas , and Ottowia were relatively abundant on the electroactive AB biofilm. Geobacter , Aminicenantales and Clostridium grew well in the MFC Anode. The economic evaluation indicated that energy consumption per ton of sewage treatment was 0.0045 kWh/m 3 . These results confirm that the AB-MFC-CW is feasible for enhancing the treatment of aquaculture wastewater at both technical and economic levels through pilot-scale verification.

Abstract:Microalgae consortium is a promising technology for achieving low-carbon and resource utilization goals in municipal wastewater treatment. However, little is known about how the consortium affects the treatment performance in the startup stage of co-cultivation. Herein, photobioreactors were constructed with different contents of microalgae and activated sludge (AS) (wt. microalgae : wt. AS ≥ 50 %). The results showed that the concentration of microalgae increased by more than 20% with AS, and the effluents were close or lower than Chinese discharge standards within HRT 24 h (NH4±-N, TP, and COD ≤ 5.0, 0.5, and 50 mg L -1 ). Furthermore, the co-occurrence pattern of microbial populations experienced inhibition-reconstruction and reconstruction-inhibition processes, respectively, and the inter-species relationship was directly related to the effluent quality. Microalgal concentration and temperature were the key factors to the microbial community profiling. And potential microorganisms in AS promoted the microalgal growth. Bacteria and fungi formed co-metabolism through functional complementation.

Abstract:High-saline wastewater treatment processes have a high energy demand, while microalgae-driven wastewater biotechnology is both economical and environmentally friendly, and the use of microalgae to treat high-saline wastewater can effectively reduce energy consumption. However, it is unclear whether the performance of microalgae reactor can be stable for a long time. In this study, an open continuous-flow biofilm photobioreactor using Dunaliella salina was innovatively constructed, and operated in combination with a membrane bioreactor (MBR) for high-saline wastewater treatment. The purpose of this study was to investigate the long-term performance of MBR, MBPBR alone and their combined operation for the removal of organic matter, phosphorus and nitrogen, explore the pathway and mechanism of nitrogen and phosphorus removal by MBPBR, and evaluate the feasibility of MBPBR-MBR integrated system for the treatment of saline wastewater. The MBR effectively removed most of the organic matter and converted ammonia nitrogen into nitrate nitrogen, which is a preferred nitrogen source for the growth of Dunaliella salina , thereby providing favorable water quality for Dunaliella salina in the subsequent MBPBR. The MBPBR, with a hydraulic retention time (HRT) of 72 h, achieved a NO 3 - -N removal load of 74.9 g N/(m 3 ∙d) and a removal efficiency of approximately 60 %, and the PO 4 3- -P removal load of 8.33 g P/(m 3 ∙d) and removal efficiency of over 80 %. The MBR-MBPBR system demonstrated long-term stability, achieving removal efficiencies of 84.3 %, 81.6 %, and 91.0 % for COD, TN and PO 4 3- -P, respectively. Within the MBPBR, the growth of algae and their assimilation contributed 44.9 % and 38.7 % of the nitrogen and phosphorus removal, respectively. The abundance of Dunaliella salina in the MBPBR remained at approximately 90 % for over 40 days of operation, providing an opportunity for Dunaliella salina recovery. These findings provide valuable insights into the utilization of microalgae for the treatment of high-salinity wastewater, while also supporting the practical implementation of microalgae-based treatment methods for such wastewater.

Abstract:Microalgal-bacterial symbiotic wastewater treatment systems (MBSWTSs) have received widespread attention due to their capacity to achieve high pollutant removal efficiency during wastewater treatment, with low energy consumption requirements, efficient carbon sequestration, and wastewater resource utilization. This paper provides an overview of the treatment performance and current research status of MBSWTSs, including a detailed summary of the mechanisms of nitrogen, phosphorus, and carbon removal by MBSWTSs and the interactions between bacteria and microalgae. In particular, this review focuses on the influence of operational parameters on the regulation of the symbiotic system, such as the microalgal:bacterial ratio, N:P ratio, external carbon source, dissolved oxygen concentration, aeration mode, and light availability. Among these factors, the microalgal:bacterial ratio, carbon source, and light availability have an important influence on microalgal-bacterial competition, as well as the trophic mode of the system, biomass production, and the capacity for the process to be practically applied on a large scale. MBSWTSs still have some challenging aspects that have hindered their development and application, such as the unknown mechanism of microalgal-bacterial co-metabolism, limited previous practical applications, algal contamination, and harvesting difficulties. To overcome these challenges, future research requires a multidisciplinary approach, incorporating life sciences, material science, and other disciplines. Comprehensive research should be conducted on the metabolic mechanisms of MBSWTSs, the optimization of process performance and waste resource utilization, providing a theoretical and practical foundation for the practical application of MBSWTSs.

Abstract:Traditional activated sludge-based technologies have significant drawbacks, including high energy requirements and greenhouse gas emissions. Microalgae-based processes offer a promising, low-cost, and environmentally friendly alternative. However, the knowledge of treatment systems based on microalgae-bacteria consortia is limited, and even more so is their microbial composition and its relationship with operational parameters. Thus, this study explores the dynamics of microalgae-bacteria consortia in a long-term operated membrane-coupled high-rate algal pond (MHRAP) for wastewater treatment. For this, a pilot-scale MHRAP plant, located in a wastewater treatment plant in Valencia (Spain), was monitored under various hydraulic retention times (HRT) and wastewater influents: i) effluent from a primary settler and ii) effluent form pre-treatment. The biomass retention time was kept constant at 6 days. The composition of the bacterial community was studied through 16S rDNA sequencing, while 18S rDNA sequencing was used to study the microalgae. The results indicate that shorter HRTs significantly increased bacterial diversity, but not eukarya. Principal Co-ordinates Analysis (PCoA) revealed that the HRT and the incoming wastewater quality control the type of the bacterial populations. However, this effect was not observed in eukaryotic organisms. The dominant microalgae genera identified were Desmodesmus and Coelastrella , with Coelastrella becoming more prevalent at shorter HRTs. For bacteria, Verrumicrobiota dominated (18–56%) at high HRT while Proteobacteria was dominant (28–44%) at HRTs below 6 days. The changes observed in the bacterial composition, including the ammonia oxidizing bacteria (AOB) community (mainly Nitrosomonas), suggest that photo-inhibition could be taking place. The nitrite oxidizing bacteria (NOB) community was dominated by Nitrospira and Candidatus Nitrotoga . Operational parameters such as light intensity, pH, and nitrite concentration were found to significantly influence the microbial community structure. Higher light intensity and alkaline pH favored the growth of Desmodesmus , while Coelastrella thrived under lower HRTs. Bacterial diversity plays a crucial role in the treatment process, while microalgae primarily support aerobic bacterial processes by providing oxygen. These findings contribute to a deeper understanding of the complex biological processes in microalgae-bacteria consortia and offer insights into improving wastewater treatment technologies.

Abstract:The rapid expansion of global urbanization and industrialization has significantly increased the discharge of municipal wastewater, leading to issues of carbon emissions and energy consumption when using traditional biological treatment processes. This study proposes an innovative process that couples iron coagulation with microalgal–bacterial granular sludge (MBGS), with optimization and regulation based on operational conditions. The study found that the coagulation performance achieved optimal levels at an iron concentration of 25 mg/L and an anionic polyacrylamide concentration of 1 mg/L, which could remove approximately 61% of the organics and over 90% of phosphorus from raw wastewater. By relying on heterotrophic microorganisms, such as Proteobacteria, Bacteroidota, and Chloroflexi, along with the synergistic interaction between algae and bacteria, the subsequent MBGS process could further effectively remove organics over the day-night cycles. Moreover, the addition of inorganic carbon sources of NaHCO3 increased the abundance of denitrification-related genes, reduced the accumulation of nitrite within MBGS, and led to effective total nitrogen removal. These results indicate that the iron coagulation–MBGS coupling process can efficiently treat municipal wastewater, offering potential for environment-sustainable pollutant removal with reduced energy consumption. These findings provide valuable insights for the practical engineering application of MBGS in wastewater treatment systems aiming for carbon-neutral wastewater treatment.
