Abstract:Deep eutectic solvents (DESs) hold great potential in biorefining because they can efficiently deconstruct the recalcitrant structure of lignocellulose. In particular, inorganic salts with Lewis acids have been proven to be effective at cleaving lignin-carbohydrate complexes. Herein, a Zr-based DES system composed of metal chloride hydrate (ZrOCl2·8H2O) and ethylene glycol (EG) was designed and used for poplar powder pretreatment. Zr4+-based salts provide sufficient acidity for lignocellulose depolymerization. The acidity of the DES was analysed by the Kamlet-Taft solvatochromic parameter, and the results demonstrated that the acidity can be regulated by the DES composition. Under the optimum conditions (ZrOCl2·8H2O:EG molar ratio of 1:2), the DES pretreatment removes nearly 100 % hemicellulose and 94.7 % lignin. The recovered lignin exhibited a low polydispersity of 1.7. The cellulose residues deliver an efficiency of 94.4 % upon enzymatic digestion. Moreover, the DES can be easily recovered with high yield and purity, and the recycled DES still maintains high delignification and enzymatic hydrolysis efficiencies. The proposed DES pretreatment technology is promising for biomass valorization.

Abstract:Deep eutectic solvents (DESs) are a potentially high-value lignin extraction methodology. DESs prepared from choline chloride (ChCl) and three hydrogen-bond donors (HBD)-lactic acid (Lac), glycerol, and urea-were evaluated for isolation of willow (Salix matsudana cv. Zhuliu) lignin. DESs types, mole ratio of ChCl to HBD, extraction temperature, and time on the fractionated DES-lignin yield demonstrated that the optimal DES-lignin yield (91.8 wt % based on the initial lignin in willow) with high purity of 94.5% can be reached at a ChCl-to-Lac molar ratio of 1:10, extraction temperature of 120 °C, and time of 12 h. Fourier transform infrared spectroscopy (FT-IR) , 13C-NMR, and 31P-NMR showed that willow lignin extracted by ChCl-Lac was mainly composed of syringyl and guaiacyl units. Serendipitously, a majority of the glucan in willow was preserved after ChCl-Lac treatment.

Abstract:Lignin, as an abundant natural aromatic biopolymer, has a complex structure and poor solubility in most solvents, making it difficult to achieve high-value applications and becoming the largest "natural waste". In this work, two novel ternary deep eutectic solvents (DESs) consisting of choline chloride (ChCl)/lactic acid (LA) with ethylene glycol (EG) or γ-valerolactone (GVL) were designed for separating lignin from waste wheat straw. Results showed that the delignification efficiency of wheat straw treated with two ternary DESs (ChCl-LA-EG and ChCl-LA-GVL) was twice as high as that treated with ChCl-LA under mild conditions (90 °C, 4 h). The highest delignification efficiency of 83.12% was achieved by ChCl-LA-GVL treatment at 130 °C for 4 h. Moreover, ternary DESs extracted lignin exhibited lower molecular weight, better homogeneity, and remarkably higher antioxidant activities than that extracted from ChCl-LA. Furthermore, ternary DESs extracted lignin had more S units and inhibited the destruction of the lignin substructure at high temperature. Compared with ChCl-LA treatment, cellulose fiber treated with ChCl-LA-EG and ChCl-LA-GVL had less active sites, lower total surface energy, and more alkaline (electron donor) adsorption sites on cellulose fiber surface. Theory calculations further confirm that the addition of EG and GVL changes the hydrogen bond strength and electrostatic potential of the ChCl-LA system, thus promoting the separation of lignin. These results provide new insights for understanding the mechanism of lignin dissolution in DESs and for the design of advanced DESs for lignocellulosic biomass separation and valorization.

Abstract:A sustainable and renewable biorefinery will increase the economic viability of lignocellulose-derived products. In this study, a diol-based deep eutectic solvent (DES) was developed to reduce the recalcitrance of bamboo, facilitate saccharification and valorize the lignin fraction. The DES pretreatment dramatically enhanced glucan digestibility by the effective removal of lignin (as high as 85.45%) and xylan (91.12%). Notably, the recovered lignin from DES pretreatment was protected during the fractionation, showing well-preserved β-O-4 structures (31.82%–59.06%). The mechanism of lignin protection was analyzed to be accomplished by incorporating the diol hydroxyl functional groups into the α position of the lignin β-O-4 structure via etherification. This study highlighted that diol DES is a promising pretreatment solvent to valorize both cellulose and lignin fractions with high enzymatic hydrolysis yield and high-quality lignin co-product.

Abstract:A novel ternary deep eutectic solvent (DES), consisted of choline chloride, oxalic acid and ethylene glycol, was developed as a green, low-cost and recyclable pretreatment system for multi-stage utilization of Eucommia ulmoides seed shells. Under optimum conditions, 79.7 % hemicellulose and 65.6 % lignin were quickly removed while 84.0 % cellulose was retained. After DES pretreatment, the yield and purity of gutta-percha achieved 85.1 mg/g and 96.2 %, which increased 1.4 and 1.8 folds higher than that of un-treatment ones. Meanwhile, 69.1 % enzymatic digestibility of cellulose was obtained, that was 2.3 folds higher than that of raw substrates. Moreover, 53.6 % low-condensation lignin with aromatic structures and valuable aryl-ether linkages was well collected. Importantly, the DES that has been recycled five runs can still remove 73.9 % hemicellulose and 58.0 % lignin. Overall, the DES was determined to efficiently promote the separation and conversion of high-quality gutta-percha, value-added lignin and high-yield glucose from Eucommia ulmoides seed shells.

Abstract:Abstract Stabilization of reactive intermediates is an enabling concept in biomass fractionation and depolymerization. Deep eutectic solvents (DES) are intriguing green reaction media for biomass processing; however undesired lignin condensation is a typical drawback for most acid-based DES fractionation processes. Here we describe ternary DES systems composed of choline chloride and oxalic acid, additionally incorporating ethylene glycol (or other diols) that provide the desired ‘stabilization’ function for efficient lignocellulose fractionation, preserving the quality of all lignocellulose constituents. The obtained ethylene-glycol protected lignin displays high β-O-4 content (up to 53 per 100 aromatic units) and can be readily depolymerized to distinct monophenolic products. The cellulose residues, free from condensed lignin particles, deliver up to 95.9 ± 2.12% glucose yield upon enzymatic digestion. The DES can be recovered with high yield and purity and re-used with good efficiency. Notably, we have shown that the reactivity of the β-O-4 linkage in model compounds can be steered towards either cleavage or stabilization, depending on DES composition, demonstrating the advantage of the modular DES composition.

Abstract:Current DES pretreatment is often performed under relatively severer conditions with high temperature, long time, and high DES usage. This work studied a short‐time diol DES (deep eutectic solvent) pretreatment under mild conditions to fractionate bamboo, facilitate enzymatic hydrolysis, and obtain high‐quality lignin. At an optimized condition of 130 °C for 10 min, lignin and xylan removal reached 61.34% and 84.15%, with residual glucan showing an ~90% enzymatic hydrolysis yield. Equally important, the dissolved lignin could be readily recovered with 97.51% yield, exhibiting 96.65% β‐O‐4 preservation. The fractionation and lignin protection mechanisms were unveiled by XRD, FTIR, cellulose‐DP, 2D HSQC NMR, 31P NMR and GPC analysis. This study highlighted that short‐time fractionation of bamboo can be achieved by a diol‐based DES which is an ideal strategy to upgrade the lignocellulose biomass for high enzymatic hydrolysis yields and high‐quality lignin stream.

Abstract:Lignocellulosic biomass is the most abundant natural polymer on Earth, but the efficient fractionation and refinery of all its components remain challenging. Acidic deep eutectic solvents refining is a promising method, while it is likely to cause lignin condensation and carbohydrates degradation, especially at server operation conditions. Here we propose the use of acidic deep eutectic solvent (DES), choline chloride/p-toluenesulfonic acid assisted mechanochemical pretreatment (DM) for efficient lignocellulose fractionation at mild condition. Four representative lignocellulose, wheat straw, moso bamboo, poplar wood and pine wood were selected at varied milling time (3, 6 h) to assess the fractionation ability of this strategy. This DM pretreatment demonstrated a rather high cellulose retentions (∼90 %) and extent of delignification for wheat straw and bamboo biomass, which corresponds to a high extent of enzymatic hydrolysis (∼75.5 %) for sugar platform pursuing. The extracted lignin showed rather high content of β-O-4' leakages due to the swelling effect of deep eutectic solvent and mild operation conditions. This work provided a promising strategy to fractionate lignocellulose using deep eutectic solvents with the goal of simultaneous cellulose hydrolysis and reactive lignin obtaining that is usually difficult to realize using traditional chemical fractionation approach.

Abstract:Plant-based waste biomass with lignocellulose as an important component is produced in large quantities worldwide every year. The components of lignocellulose that typically exhibit high utilization value include cellulose and hemicellulose, as well as pentoses and hexoses derived from their hydrolysis. As a pretreatment for the hydrolysis process, delignification is a pivotal step to enhance cellulose/hemicellulose accessibility and achieve high yields of fermentable sugars. Additionally, deep eutectic solvents (DESs) are the most widely used solvents for delignification during biomass fractionation due to their clean and environmentally friendly attributes. DESs dissolve lignin by inducing a large amount of β-O-4 bond cleavage and partial carbon–carbon bond cleavage, retaining cellulose in the solid residue, while most of the hemicellulose is hydrolyzed in DES pretreatment. This article provides a comprehensive review of the influence of DESs in the lignocellulose separation process. Key factors such as lignin removal rate, sugar conversion rate, and product chemical structure are critically reviewed to assess the feasibility of employing DESs for lignocellulose separation.

Abstract:Green and low cost deep eutectic solvents (DESs) are promising to replace the solid acids and ionic liquids in biomass fractionation process. To enhance the lignocellulose pretreatment efficiency, an acidic DES that composed of Brønsted acid (ZnCl2) as hydrogen bond acceptor and Lewis acid (lactic acid) as hydrogen bond donator was designed. This bifunctional DES was used for the extraction of lignin from poplar sawdust. Under the optimal pretreatment condition, the ZnCl2-lactic acid DES could recover 95.2 wt% of lignin with a purity of 92.1%. The recovered lignin demonstrated a low polydispersity of 1.67 and small amount of β-aryl-ethers. Moreover, the acidic DES had a good recyclability and reusability. Such performance was attributed to the presence of bifunctional acid sites, which help selectively cleave lignin-carbohydrate complex linkages. The acidity and polarity of Brønsted acid can be modulated by the Lewis acid, thus synergistically promote the lignin extraction and production.

Abstract:Novel and green cholinium amino acids-based deep eutectic solvents (DESs) and their aqueous mixtures were synthesized and employed in deconstructing poplar for hemicellulose fractionation. The effects of water content in DESs on hemicellulose dissociation and structural features were comprehensively investigated, along with the reusability of DESs for treatment. The integration of water into DESs could facilitate hemicellulose fractionation, and the cholinium lysine: urea with 5 wt% water (CL: U-5) demonstrated the best performance with a hemicellulose yield of 59.2%. Further structure analysis revealed that hemicelluloses with various branching degrees and molecular weights were obtained with varying water content of DESs. Furthermore, the CL: U-5 had recyclability and reusability with a 40.5% hemicellulose yield obtained after reused three times. The novel and eco-friendly cholinium amino acids-based DESs treatment provides an effective and sustainable strategy for hemicellulose fractionation from woody biomass.

Abstract:Deep eutectic solvents (DESs), a novel and environmentally-friendly solvent, have high potential for biomass pretreatment due to its advantages of low cost, low toxicity, strong solubility, excellent selectivity and biocompatibility. Two types of DES (binary and ternary) were synthesized and characterized, and optimized ternary DES was selected to pretreat wheat straw for enhancement of the solubility of lignocellulose. Moreover, enzymatic hydrolysis was tested to verify the performance of pretreatment. In addition, the changes in surface morphology, structure and crystallinity of wheat straw pretreated by DES were analyzed to reveal the pretreatment mechanism. Experimental results indicated that viscosity exhibited little difference in different types of DESs, and a declining trend as the temperature increases in same DES. The ternary DES pretreatment efficiently enhanced the solubility of typical lignocellulose, with the optimal removal rate of lignin at approximately 69.46%. Furthermore, the total sugar concentration of the residue was about 5.1 times more than that of untreated wheat straw after the pretreated samples were hydrolyzed by the cellulase for 24 h, indicating that DES has the unique ability to selectively extract lignin and hemicellulose from wheat straw while retaining cellulose, and thus enhanced the solubility of lignocellulose. The scanning electron microscope (SEM) observation and X-ray diffraction (XRD) determination showed that the surface of wheat straw suffered from serious erosion and the crystallinity index of wheat straw increased after DES5 pretreatment. Therefore, DES cleaves the covalent bond between lignin and cellulose and hemicellulose, and reduces the intractability of lignin resulting in the lignin dissolution. It suggests that DES can be used as a promising and biocompatible pretreatment way for the cost-effective conversion of lignocellulose biomass into biofuels.

Abstract:Deep eutectic solvent (DES) is a newly-emerged green solvent for efficient pretreatment of lignocellulosic feedstock. To improve the component fractionation performance of neutral DES, p- toluenesulfonic acid ( p- TsOH) was employed as catalyst to form a novel ternary DES with benzyltriethylammonium chloride (TEBAC) and glycerol (Gly) for pretreatment of soybean straw. Under the optimum reaction conditions (TEBAC:Gly = 1:12, 1.6 wt% p- TsOH and reacted at 90 °C for 160 min), the lignin and hemicellulose removal from soybean straw were amounted to 92.0 % and 88.2 %, respectively. The pretreated substrate showed satisfactory enzymatic hydrolysis performance, as the glucose and reducing sugar concentrations reached 37.3 g/L and 42.3 g/L, respectively, after 72 h saccharification under the action of cellulase with a relatively low enzyme loading of 10 FPU/g cellulose. This method provides an efficient and mild route for utilization of agricultural waste and production of platform monosaccharides.

Abstract:A synergistic pretreatment that realizing effective fractionation and targeted valorization can guarantee the implementability to future biorefinery scenario. In the present study, a stepwise approach using hydrothermal and deep eutectic solvents (DES) pretreatment was developed to preferentially dissociate hemicelluloses and further remove lignin from poplar, while retaining a cellulose-rich substrate that can be easily digested via enzymatic saccharification to obtain glucose. Results showed that the hydrothermal filtrate is mainly composed of xylooligosaccharide (XOS), monosaccharides, byproducts, and xylan-type hemicelluloses, which have homogenous structures and uniform molecular weights distribution as well as excellent antioxidant activity. Subsequent DES pretreatment further removed the lignin barriers, leading to a remarkable increase in the saccharification efficiency from 15.72% to 96.33% under optimum conditions for enzymatic hydrolysis. In short, the integrated pretreatment is effective for dissociating and chemical conversion of poplar wood, which was reasonable to promote the frontier of highly available biorefinery.

Abstract:Refining lignocellulose into fermentable monosaccharides remains a challenging endeavor. In this study, we employed a novel deep eutectic solvent (DES) composed of 1-tetradecyl-3-carboxymethylimidazolium bromide (C 14 CIMBr) and lactic acid (LA) for the pretreatment of Moringa oleifera Lam. husk (M. oleifera husk), aiming to enhance lignocellulose fractionation. The optimal conditions identified were 165 °C, 43.9 min, and a molar ratio of C 14 CIMBr to LA at 1:4.8, resulting in the removal of 93.8% of lignin and 96.8% of hemicellulose. Following enzymatic hydrolysis of the pretreated M. oleifera husk, yields of glucose and xylose reached 94.3% and 86.1%, respectively, representing significant increases by factors of 7.0-fold and 10.3-fold compared to untreated M. oleifera husk. Molecular simulations indicated that the novel DES effectively disrupts the original internal hydrogen bonding network while reconstructing new DES-lignocellulosic hydrogen bonds, it also demonstrates excellent solubilization capabilities for lignin cleavage products. Finally, the yield of regenerated lignin reached 81.3%, exhibiting notable bactericidal activity under blue light irradiation.

Efficient extraction of lignin from moso bamboo by microwave-assisted ternary deep eutectic solvent pretreatment for enhanced enzymatic hydrolysis

Abstract:Applications of deep eutectic solvent (DES) systems to separate lignocellulosic components are of interest to develop environmentally friendly processes and achieve efficient utilization of biomass. To enhance the performance of a binary neutral DES (glycerol:guanidine hydrochloride), various Lewis acids (e.g., AlCl3·6H2O, FeCl3·6H2O, etc.) were introduced to synthesize a series of ternary DES systems; these were coupled with microwave heating and applied to moso bamboo. Among the ternary DES systems evaluated, the FeCl3-based DES effectively removed lignin (81.17%) and xylan (85.42%), significantly improving enzymatic digestibility of the residual glucan and xylan (90.15% and 99.51%, respectively). Furthermore, 50.74% of the lignin, with high purity and a well-preserved structure, was recovered. A recyclability experiment showed that the pretreatment performance of the FeCl3-based DES was still basically maintained after five cycles. Overall, the microwave-assisted ternary DES pretreatment approach proposed in this study appears to be a promising option for sustainable biorefinery operations.
Tandem Character of Liquid Hot Water and Deep Eutectic Solvent to Enhance Lignocellulose Deconstruction

Abstract:Pretreatment with efficient fractionation, eco‐friendliness, and low‐cost brings high‐security to future biorefinery systems. Synergistic pretreatment is a compelling blueprint to tackle the compact structure of lignocellulose towards a high‐level valorization. We designed a stepwise approach, using hydrothermal and deep eutectic solvent (DES) pretreatments, to hierarchically extract hemicelluloses and lignin from poplar, while delivering a cellulose‐rich substrate that can be easily enzymatic hydrolysis to obtain fermentable glucose and residual lignin. The lifetime of novel DES showed that the pretreatment efficiency was still largely maintained after the fourth recycling. An enhancement of enzymatic digestibility from 13.9 to 90.4% was initiated by the deconstruction of amorphous portions and robust cell wall. An ally of 23.7% xylooligosaccharides (degree of polymerization, DP 2‒6), 47.5% DES‐isolated lignin and 19.2% cellulose enzymatic lignin was harvested via this coupled process. It was reasonable to push forward the precise design of sustainable tandem pretreatment that can boost the frontier of highly available biorefinery.
