Abstract:The aim of this study was to evaluate the preservation effect of chitosan (CS) and lysozyme (Lys) coating on the quality of large yellow croaker (Larimichthys crocea) via physicochemical analyses, microbiological and sensorial assessments during refrigerated storage. Results showed that CS films incorporated with 0.6 mg.mL(-1) of lysozyme exhibited the strongest inhibition effect against S. aureus, whose inhibition zone was 16.10 +/- 0.71 mm. The utilization of CS singly, or CS-lysozyme (Lys) coating significantly reduced lipid oxidation (thiobarbituric acid value, TBA) and improved the sensorial scores of fishes. Control and treated groups exceeded the maximum permissible level (30 mg N/100 g) of total volatile basic nitrogen (TVB-N) in fishery products on days 6 (control) and 9 (CS or CS-Lys treated), respectively. Fish samples reached the value of 7.0 Log CFU/g (Total Viable Count, TVC) on days 15 (control), whereas both CS and CS-Lys treated flesh samples never exceed this limit value during the refrigerated storage. Results of our study indicate CS-Lys have high efficiency in inhibiting microorganism growth and lipid oxidation and in maintaining sensorial quality, which is a promising method to maintain the quality and extend the shelf life of refrigerated fishes. Abstract:The effect of fermented soybeans from three different origin places (Hunan, Guangdong, and Sichuan in China) on protein oxidation and freshness of largemouth bass (Micropterus salmoides) was investigated during repeated freezing-thawing (FT) cycles. The Hunan fermented soybean exhibited the best antioxidant capacity according to the capacity of Fe3+ reducing power, DPPH radical scavenging activity, super oxide radical scavenging assay, hydroxyl radical scavenging assay, and reduced glutathione (GSH). As expected, 0.1% fermented soybean significantly (P < 0.05) delayed protein oxidation including the decline of reactive and total sulphydryls, Ca2+ ATPase activity, and freshness as well as the increase of total volatile base nitrogen (TVB-N) during repeated freezing-thawing cycles treatments. It was concluded that the fermented soybean was not only a seasoning, but also an antioxidant in prepared fishes.
Protein Denaturation and Oxidation in Chilled Hairtail (trichiutus Haumela) As Affected by Electrolyzed Oxidizing Water and Chitosan Treatment

Abstract:The effect of chitosan (CH) coating combined with electrolyzed oxidizing water (EOW) on microbiological and physicochemical of hairtail (Trichiutus haumela) was evaluated during 15days of storage at -3 degrees C. Hairtail samples treated by EOW combined with 1.5% (w/v) CH were compared with samples soaked in single material. For storage of 15days, the thiobarbituric acid value of hairtails increased steadily to 1.79, 1.52, 1.48mg/100g (p<0.05) in the EOW, CH, and EOW+CH group, respectively. All of these were significantly lower than that of the control group (2.01 +/- 0.04mg malonaldehyde/100g; p<0.05). Microbiological results, total volatile basic nitrogen, pH, and peroxide value also revealed that EOW and CH significantly retarded hairtail spoilage. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis pattern of sarcoplasmic proteins revealed CH+EOW could slow down the protein deterioration of fillet. In this study, CH coating combined with EOW effectively extended the shelf life of hairtail to 6-7days.

Abstract:Chitosan was applied to make fish ball from silver carp,and its correlative effects on gelling quality and lipid oxidation were evaluated by colour,texture,water retention,and TBA(2-thiobarbituric acid).The colour,texture and water retention were improved and the lipid oxidation was suppressed when 1.0%chitosan was added in gels.Adding 1.0%chitosan could be considered as a promising approach in the processing of silver carp gels to improve thermal gelling properties and delay lipid oxidation.

Abstract:Spectrophotometric- and liquid chromatography/mass spectrometry (LC/MS)-based lipidomics analyses were performed to explore the changes of lipid profiles in pike eel (Muraenesox cinereus) under stable chlorine dioxides (ClO2) and vacuum-packed treatment during chilled storage. The peroxide value (PV) and malondialdehyde (MDA) content in ClO2 treated and vacuum-packaged (VP) samples were significantly reduced compared to simple-packaged (SP) samples during whole chilled storage. The LC/MS-based lipidomics analyses identified 2182 lipid species in the pike eel muscle classified into 39 subclasses, including 712 triglycerides (TGs), 310 phosphatidylcholines (PCs), 153 phosphatidylethanolamines (PEs), and 147 diglycerides (DGs), among others. Further, in comparison with fresh pike eel (FE) muscle, 354 and 164 higher and 420 and 193 lower abundant levels of differentially abundant lipids (DALs) were identified in SP samples and VP samples, respectively. Compared with the VP batch, 396 higher and 404 lower abundant levels of DALs were identified in the SP batch. Among these, PCs, PEs, TGs, and DGs were more easily oxidized/hydrolyzed, which could be used as biomarkers to distinguish FE, SP, and VP samples. This research provides a reference for controlling lipid oxidation in fatty fish.

Abstract:The effects on the quality of frozen red sea bream, which were pretreated by soaking in solutions containing trehalose, carboxymethyl chitosan magnetic nanoparticles (CCMN), and glycerin or 0.1% herring antifreeze protein (hAFP), were investigated. In this study, the viscoelastic properties of protein studied with the dynamic rheology, Raman and intrinsic fluorescence spectrometry were used to explore the conformation changes of MFP. Surface hydrophobicity, particle size, and zeta potential were carried out to analyze protein aggregation. Solubility, the content of total sulfhydryl/disulfide bond/carbonyl/dityrosine, and the Ca<sup>2+</sup>-ATPase activity were measured to explore the degree of protein oxidation. SDS-PAGE was conducted to analyze the protein denaturation. Results showed that the pretreatment of red sea bream with soak solutions could improve the viscoelasticity of fillets protein, stabilize the secondary and tertiary conformation of the MFP, inhibit the protein aggregation and oxidation, and decrease the degree of protein denaturation compared with the control (4 °C thawing), especially the soak solutions containing hAFP could minimize the freeze-thaw damage. To summarize, the hAFP helped to retain the above characteristics of frozen fillets much better than that of conventional cryoprotectants (glycerin) to improve the quality of the frozen product after thawing.

Effects of ε-polylysine and chitooligosaccharide Maillard reaction products on quality of refrigerated sea bass fillets

Abstract:Background: The Maillard reaction is a promising and safe method for obtaining chitooligosaccharide conjugates with proteins or peptides as food preservatives. This study aims to investigate the moisture state, physicochemical properties, and shelf-life of sea bass fillets treated with ε-polylysine (ε-PL) and chitooligosaccharides (COS), which are Maillard reaction products (LC-MRPs), during refrigerated storage. Results: The results of microbiological analysis and confocal laser scanning microscope (CLSM) revealed that LC-MRPs could retard microbial growth effectively. Compared with control, other treated groups could strongly retard the increase in the thiobarbituric acid (TBA) value, the K-value and the total volatile basic nitrogen (TVB-N) value, and also inhibited the softening of texture and the accumulation of biogenic amines in fish. The results of low-field nuclear magnetic resonance (LF-NMR) and magnetic resonance imaging (MRI) indicate that LC-MRPs could delay the water migration of fillets and increase water holding capacity (WHC). Through sensory evaluation, the application of LC-MRPs increased the shelf-life of refrigerated sea bass fillets for another 9 days. Conclusion: Maillard reaction products derived from chitooligosaccharides and ε-polylysine have strong potential for preserving sea bass. © 2022 Society of Chemical Industry.
Salt reduction in myofibrillar protein gel via inhomogeneous distribution of sodium-containing encapsulated fish oil coacervate: Mucopenetration ability of sodium carboxymethyl cellulose

Abstract:The potential application of fish oil microcapsules as salt reduction strategies in low-salt myofibrillar protein (MP) gel was investigated by employing soy protein isolates/carboxymethyl cellulose sodium (SPI-CMC) coacervates enriched with 25 mM sodium chloride and exploring their rheological characteristics, taste perception, and microstructure. The results revealed that the SPI-CMC coacervate phase exhibited the highest sodium content under 25 mM sodium level, albeit with uneven distribution. Notably, the hydrophilic and adhesive properties of CMC to sodium facilitated the in vitro release of sodium during oral digestion, as evidenced by the excellent wettability and mucopenetration ability of CMC. Remarkably, the fish oil microcapsules incorporating SPI-CMC as the wall material, prepared at pH 3.5 with a core-to-wall ratio of 1:1, demonstrated the highest encapsulation efficiency, which was supported by the strong hydrogen bonding. Interestingly, the presence of SPI-CMC coacervates and fish oil microcapsules enhanced the interaction between MPs and strengthened the low-salt MP gel network. Coupled with electronic tongue analysis, the incorporation of fish oil microcapsules slightly exacerbated the non-uniformity of sodium distribution. This ultimately contributed to an enhanced perception of saltiness, richness, and aftertaste in low-salt protein gels. Overall, the incorporation of fish oil microcapsules emerged as an effective salt reduction strategy in low-salt MP gel.

Abstract:In the food processing field, it is very often that fish proteins are denatured affecting the nutritional value of the product which is vital to be solved. By using appropriate sugar donors for glycosylation with protein, improving the stability and emulsification properties of fish proteins can be achieved. This research looks into the impacts of enzymatic chitosan oligosaccharide (CO) at various concentration (0.15%, 0.30%, 0.45%, 0.60%, w/v) upon the molecular makeup and function of silver carp myofibrillar protein (MP) in an attempt to comprehend the impact of electrostatic binding among MP as well as CO on protein conformation. Analysis was done on the impact of various CO concentrations upon MP's secondary structure, conformational changes, and functional characteristics. Twelve sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) assays were implemented to monitor MP; Fourier transform infrared spectroscopy, endogenous fluorescence spectroscopy, and UV absorption spectra were carried out to investigate the influence of CO on MP; Particle size distribution, emulsifying activity index (EAI), solubility, turbidity, sulfhydryl content, carbonyl content, foaming capacity, surface hydrophobicity, emulsifying stability index (ESI), and foam persistence were all investigated. In addition, we used dynamic light scattering, scanning electron microscope, and atomic force microscope to analyze myosin (MO) and 0.60% CO-MO complex. The results demonstrated that CO and MP form complexes through hydrogen bonding and electrostatic interactions. CO modification not only delayed the oxidation of MP but also promoted MP to show better solubility, foaming, and foaming stability. In addition, CO modified myosin particle size decreased, reducing myosin's roughness and making myosin's structure more compact. To sum up, molecular interaction could change functional characteristics, and products with special properties could be developed after modification with chitosan oligosaccharide.
Effects of antioxidant types on the stabilization and in vitro digestion behaviors of silver carp scale gelatin-stabilized fish oil-loaded emulsions

Abstract:Lipid oxidation is a major challenge in the development and storage of lipid-containing food products. In this work, we extracted aquatic gelatin from silver carp scales and studied the effects of antioxidant types (water-soluble and lipid-soluble types) on the stabilization, lipid oxidation, and in vitro digestion behaviors of silver carp scale gelatin-stabilized fish oil-loaded emulsions. Vitamin C (VC), a water-soluble antioxidant, and vitamin E (VE), a lipid-soluble antioxidant, had no obvious effects on the appearance, droplet size distribution, and droplet stability of the emulsion. VC slowed the liquid-gel transition of the emulsions at room temperature. The emulsion creaming stability decreased with the increase of VC concentration, whereas it increased with the increase of VE concentration. Lipid oxidation hierarchy of emulsion groups at room temperature were VC<VE<control<pure oil. Free fatty acids were mainly released from the silver carp scale gelatin-stabilized emulsions in the simulated intestinal fluid. Moreover, compared with the control group, VC increased the free fatty acid release percentages, whereas VE decreased them. This work provided useful information for developing antioxidants in the field of food science and in value-added utilization research of aquatic by-products.

Abstract:The aim of this study was to investigate the effects of chitosan-grafted-phenolic acid (CS-g-phenolic acid) on moisture state and protein properties of vacuum packaged sea bass (Lateolabrax japonicus) fillets during refrigerated storage. Results showed that CS-g-phenolic acid could significantly lower drip loss and cooking loss, and efficiently delay the changes of water activity (Aw) and water migration. TCA-soluble peptides, protein solubility, myofibril fragmentation index (MFI), surface hydrophobicity and SDS-PAGE results revealed that graft copolymer treatment could reduce the protein oxidation and degradation of sea bass during refrigerated storage. Meanwhile, the results of intrinsic fluorescence intensity (IFI) and scanning electron microscope (SEM) showed that the graft copolymer could maintain the structure of fish. Therefore, the graft copolymer coatings combined with vacuum packaging could effectively inhibit the water loss and protein degradation of sea bass fillet during refrigerated storage.
Development and characterization of fish myofibrillar protein/chitosan/rosemary extract composite edible films and the improvement of lipid oxidation stability during the grass carp fillets storage

Abstract:Biofilm composition from fish myofibrillar protein (FMP) and chitosan solution (CS) incorporated with rosemary extract (RE) was developed and applied to monitor the freshness of fish fillets. The effects of different concentrations of RE as well as physical, mechanical, structural and functional properties of FMP/CS films were investigated. Films containing RE showed reduced water solubility and water vapor permeability and enhanced tensile strength and elongation at break. Results also showed good compatibility of the components and good dispersion of RE in the matrix. However, the content of RE (0.2%, v/v) added in the composite films produced aggregations and had negative effects on their film-forming properties. The antioxidant capacity of composite films was related to the level of RE and demonstrated by the DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging assay. Chilled grass carp fillets wrapped with different films to evaluate the preservative effect. Results of thiobarbituric acid reactive substances, pH value, Free amino acid and total volatile basic nitrogen indicated that FMP/CS/RE composite film could protect the fish fillet well and inhibit the lipid oxidation. The developed FMP/CS/RE composite films possess the potential to be applied as edible films in the food packaging industry and food cold chain transportation.
Effect of oxidation modification induced by peroxyl radicals on the physicochemical and gel characteristics of grass carp myofibrillar protein

Abstract:Protein oxidation of fish products during storage and processing is of interest recently. In this study, the effect of peroxyl radicals generated from 2,2′-azobis (2-amidinopropane) dihydrochloride (AAPH) on the physicochemical and gel properties of myofibrillar protein (MP) from grass carp was investigated. After incubation with AAPH (0.04, 0.2, 1, 5, 25 mM), the carbonyl and dityrosine content, and surface hydrophobicity index of MP were higher than that of the non-oxidation control. Conversely, this treatment decreased the free ammonia, total and free sulfhydryl content of MP progressively. The transformation from α-helix to β-sheet and β-turn in MP treated with AAPH was found. The fluorescence quenching of tryptophan occurred in the 5 and 25 mM AAPH groups. When the AAPH concentration reached to 25 mM, the protein solubility decreased to 58%, compared to the control group. Meanwhile, this AAPH treatment damaged the elastic feature of MP and also decreased the water holding capacity, gel strength and texture of gel, which were closely depended on the oxidation degree. The poor and rough microstructure provided solid and visible evidence for the loss of gel properties in oxidation treatment groups. In short, these results gave a deeper understanding for the changes of physicochemical and gel properties of fish MP subjected to oxidation environment.

Abstract:The research evaluated the effects of locust bean gum (LBG) and sodium alginate (SA) active coatings containing 0.15, 0.30 or 0.60% lemon verbena (Lippa citriodora Kunth.) essential oil (LVEO) on the bacterial diversity and myofibrillar proteins (MPs) of large yellow croaker during refrigerated storage at 4 °C for 18 days. Variability in the dominant bacterial community in different samples on the 0, 9th and 18th day was observed. Pseudomonas and Shewanella were the two major genera identified during refrigerated storage. At the beginning, the richness of Pseudomonas was about 37.31% and increased for control (CK) samples during refrigerated storage, however, the LVEO-treated samples increased sharply from day 0 to the 9th day and then decreased. LBG-SA coatings containing LVEO treatments significantly delayed MPs oxidation by retarding the formation of free carbonyl compounds and maintaining higher sulfhydryl content, higher Ca2+-ATPase activity, better organized secondary (higher contents of α-helix and β-sheet) and tertiary structures during refrigerated storage. The transmission electron microscope (TEM) images showed that the integrity of the sarcomere was damaged; the boundaries of the H-, A-, and I-bands, Z-disk, and M-line were fuzzy in the CK samples at the end of storage. However, the LVEO-treated samples were still regular in appearance with distinct dark A-bands, light I-bands, and Z-disk. In brief, LBG-SA active coatings containing LVEO treatments suggested a feasible method for protecting the MPs of large yellow croaker during refrigerated storage.

Abstract:Surimi processing industry generates huge quantities of underutilized surimi processing by-products (SPB). This study aimed to investigate the potential antioxidant-cryoprotective and gelation-enhancing effects of SPB hydrolysates on silver carp (Hypophthalmichthys molitrix) surimi based on protein oxidation, protein degradation, and gel-forming ability analysis. Results showed that untreated surimi was particularly susceptible to frozen-induced protein degradation and oxidation as well as rapid deterioration in gelation properties. Compared with 4% sucrose-added surimi, partial replacement of sucrose with 2% trypsin- and alcalase-treated SPB hydrolysates effectively delayed the oxidation of cysteine, carbonylation of amino acids, loss of Ca2+-ATPase activity, and the destroy of structural integrity of myofibrillar protein in surimi. Moreover, addition of SPB hydrolysates significantly (P < 0.05) increased the initial gelation properties and reduced the loss in gelation and water-holding capacity in surimi with prolonged frozen storage. Therefore, SPB hydrolysates could be used as natural antioxidant-cryoprotectant and gel texture enhancer in freshwater fish surimi.

Abstract:The quality of freshwater fish is easy to deteriorate during postmortem storage, so it is necessary to find an effective method to extend the shelf life of fish fillets. The aim of this study is mainly to develop a novel bilayer active film with curcumin/nisin-loaded pectin nanoparticles (CZ-NPs) and evaluate its effect on the quality of fish fillets. The selected sensory, physicochemical (pH, color qualities, TVB-N, TBARS, biogenic amines, moisture state) and microbial properties (microbial enumeration and high-throughput sequencing) were analyzed to confirm the preservative mechanism of films on the fillets. During storage at 4 °C, better sensory evaluation feedback and lower total volatile base nitrogen in the film treated samples was observed when compared to the control samples. Additionally, CZ-NPs film effectively inhibited oxidation, with the final reductions of 1.04 mg MDA/kg flesh when compared to control. As suggested by low-field NMR and magnetic resonance imaging, film treated fillets presented higher water holding capacity by controlling transformation from immobilized to free water. Meanwhile, lower microbial enumerations were also recorded in the CZ-NPs film treated group, which was reduced by a range of 1.8–2.3 log CFU/g on day 10 when compared to control. High-throughput sequencing results showed that the Pseudomonas constituted the main spoilage bacteria of fillets, and CZ-NPs film contributed to the notably suppressed growth of Pseudomonas . As a result, CZ-NPs film showed a desirable application to extend the shelf life of refrigerated flesh, as an alternative for fishery product packaging and preservation.

Abstract:Changes in protein and gel properties of ultrasound‐assisted chlorogenic acid‐grafted chitosan soaked fish fillets during storage. The perishability of aquatic products is a challenge for the processing of aquatic products, so it is better to choose suitable preservatives to prevent the degradation of proteins and the fission of their gel properties. Thus, this study investigated the effects of ultrasound‐assisted chlorogenic acid grafted chitosan (GUA) soaking of sea bass (Lateolabrax japonicus) fillets on protein oxidation and gelation properties. The results showed that GUA could reduce the formation of protein carbonyls and maintain Ca2+‐ATPase activity. Intrinsic fluorescence intensity (IFI) and SDS‐PAGE results demonstrated that GUA stabilized the tertiary structure of protein and inhibited protein degradation. Interestingly, ultrasound attenuated the protein aggregation induced by the use of chitosan‐grafted‐chlorogenic acid (CS‐g‐CA), which has good particle size during storage. Meanwhile, GUA treatment endowed the gel with a dense network microstructure, increased water holding capacity (WHC), reduced water mobility, and improved MP gel color as well as textural properties.

Abstract:Protein degradation and oxidation are two major alterations during the storage of processed bighead carp fillets. This study conducted a comparative analysis of degraded and oxidized products as well as oxidation sites in fresh, frozen and brined frozen bighead carp fillets. Frozen storage played a dominant role in protein degradation and oxidation, and brining promoted these changes. In brined frozen samples, the decreased SDS-PAGE band intensities for tropomyosin, troponin, and myosin light chain were mainly due to their degradation. Myosin heavy chain fast skeletal muscle was the most oxidized and degraded protein during storage, with modifications such as monooxidation, protein-lipid peroxidation adducts, and α-aminoadipic semialdehydes formation. Amino acids in the tail portion of myosin were prone to oxidation than the head portions. Our results provided comprehensive insights into protein degradation and oxidation in bighead carp during storage, helping to assess the specific fate of oxidative products in future dietary investigations.

Abstract:<p>The aim of this study was to investigate the impact of egg white protein (EWP) and β-cyclodextrin (βCD) mixture (3:1) on the structural and functional properties of myofibrillar proteins (MP) from silver carp during 60 days of frozen storage. Different concentrations of EWP-βCD were prepared and incorporated with MP. It was analyzed that α-helix content significantly decreased (P < 0.05) during frozen storage. EWP-βCD substantially enhanced the stability of α-helix content with the minimum increase of β-turn and random coils. Furthermore, rheological studies revealed that MP with increased EWP-βCD concentrations (4 and 6%) improved the inter-molecular protein interactions and enhanced the viscoelastic properties during the gelation process. Moreover, this addition improved the textural properties compared to control. It was also observed from microstructural analysis that MP gelling properties were decreased, which showed coarser and porous structure in control MP (0%). Meanwhile, MP gel treated with EWP-βCD (6%) showed a more compact and well-established network of proteins. All experimental analysis confirmed that EWP-βCD (6%) showed better cryoprotective properties in the preservation of the functional and structural properties of silver carp MP during frozen storage, and could be effective in the industrial storage of fish and associated products.</p>
