Abstract:There is a growing body of evidence that suggests that both biodegradable and conventional (non-degradable) microplastics (MP) are hazardous to soil health by affecting the delivery of key ecological functions such as litter decomposition, nutrient cycling and water retention. Specifically, soil fauna may be harmed by the presence of MPs while also being involved in their disintegration, degradation, migration and transfer in soil. Therefore, a comprehensive understanding of the interactions between MPs and soil fauna is essential. Here, we conducted a 120-day soil microcosm experiment applying polyethylene (PE) and polylactic acid (PLA), in the absence/presence of the earthworm Eisenia nordenskioldi to estimate the relative singular and combined impact of MPs and earthworms on the soil bacterial community. Our findings revealed contrasting effects of PE and PLA on the composition and diversity of soil bacteria. All treatments affected the community and network structure of the soil bacterial community. Compared to the control (no MPs or earthworms), PE decreased bacterial alpha diversity, while PLA increased it. Patescibacteria were found to be significantly abundant in the PE group whereas Actinobacteria and Gemmatimonadetes were more abundant in PE, and PLA and earthworms groups. The presence of earthworms appeared to mediate the impact of PE/PLA on soil bacteria, potentially through bacterial consumption or by altering soil properties (e.g., pH, aeration, C availability). Earthworm presence also appeared to promote the chemical aging of PLA. Collectively, our results provide novel insights into the soil-fauna-driven impact of degradable/nondegradable MPs exposure on the long-term environmental risks associated with soil microorganisms.

Effect of microplastic pollution on the gut microbiome of anecic and endogeic earthworms

Abstract:Microplastic (MP) pollution constitutes an emerging type of pollution threatening both aquatic and terrestrial ecosystems. The impact on aquatic ecosystems has been extensively studied, but the effect on terrestrial ecosystems and their inhabitants is mostly underexplored. In this study, we explored the effect of MP pollution on gut bacterial microbiome of endogeic (Aporrectodea caliginosa) and anecic (Lumbricus terrestris) earthworms. The experiments were performed in sandy soil with 0.2% of low-density polyethylene MPs (LDPE MPs). We observed that the endogeic earthworms had 100% survival, while anecic earthworms survived 25 days in the control (i.e. in absence of MPs) and 21 days in the treatment with LDPE MPs. The main driver of shifts in the diversity and composition of the bacterial communities in the gut of tested earthworms was the lifestyle of the worms, followed by the presence of MPs. The bacterial microbiome diversity was significantly different among the two types of earthworms, and the highest bacterial diversity was found in the gut of the endogeic earthworms. The effect of MPs on gut bacterial microbiome was clearly observed in the changes in the relative abundance of several phyla and families of the bacterial communities in both types of earthworms, although it was most evident in the anecic earthworms. The Actinobacteriota, Proteobacteria, and Firmicutes were the main groups enhanced in the MP treatments, suggesting enrichment of the bacterial communities with potential plastic degraders.
Effects of polyethylene microplastics stress on soil physicochemical properties mediated by earthworm Eisenia fetida

Abstract:Microplastics (MPs) are widely distributed in soil environments, but their ecological risks are not fully understood. To fill this knowledge gap, incubation experiments were conducted to explore the physiological response of Eisenia foetida (E. fetida) to polyethylene MP stress and its effects on soil physicochemical properties. E. fetida was incubated in soils amended with MPs of two particle sizes (13 μm and 130 μm) at six concentrations (0, 1, 3, 6, 10 and 20 g MPs·kg-1 soil) under laboratory conditions. The toxicity of 13 μm MPs on the growth and survival of E. fetida was greater than that of 130 μm MPs. Excessive reactive oxygen species accumulation induced by high MP concentrations decreased superoxide dismutase activity and increased malondialdehyde content. Soil pH increased significantly in the 130 μm treatments. MPs increased the contents of soil organic carbon and available potassium. However, the presence of MPs did not significantly alter available phosphorus or nitrate nitrogen content. MP contamination in soil may have adverse impacts on the growth of earthworms, induce oxidative stress in earthworms, and change soil physicochemical properties. In addition, the effects of MPs are size-dependent and dose-dependent. This study provides new evidence for the ecological risks of MP pollution in the earthworm-soil systems.
[Levels of endogenous plasma cortisol in patients with rheumatoid arthritis and other diffuse connective tissue diseases].

Abstract:The authors tried to obtain information on levels of endogenous plasma cortisol in patients with rheumatoid arthritis and other diffuse connective tissue diseases, in particular systemic lupus erythematosus. Endogenous cortisol was assessed by radioimmunoanalysis at 8 a.m. and 4 p.m. If the sum of the morning and afternoon values reached the level of 280 nmol/l (sum of the lowest still normal values), they considered the daily cortisol secretion still normal. In a group of 30 patients who did not have glucocorticoid therapy the authors found only in one patient a lower daily endogenous cortisol secretion. In 117 patients with a relatively severe course of rheumatoid arthritis where small doses of corticoids (equivalents of 5-10 mg Prednisone) were administered for prolonged periods, a reduced daily secretion of endogenous plasma cortisol was recorded in almost 40%. Repeated examinations made in 13 subjects with rheumatoid arthritis revealed relatively frequent differences in values of endogenous plasma cortisol. In rare instances extremely low plasma levels of endogenous cortisol were found. Similar results as in rheumatoid arthritis, although assessed in a relatively small number of probands, were obtained by the authors also in patients with other diffuse connective tissue diseases, in particular those with systemic lupus erythematosus. Those patients had for prolonged periods roughly double doses of corticoids as compared with patients with rheumatoid arthritis. During repeated examinations the endogenous plasma cortisol levels varied also. The authors are aware that from the assembled results it is so far not possible to draw any definite conclusions and that further investigations are needed which will test the impact of endogenously produced corticoid.
Earthworms improve the rhizosphere micro-environment to mitigate the toxicity of microplastics to tomato (Solanum lycopersicum)

Abstract:Microplastics (MPs) are widespread in agricultural soil, potentially threatening soil environmental quality and plant growth. However, toxicological research on MPs has mainly been limited to individual components (such as plants, microbes, and animals), without considering their interactions. Here, we examined earthworm-mediated effects on tomato growth and the rhizosphere micro-environment under MPs contamination. Earthworms (Eisenia fetida) mitigated the growth-inhibiting effect of MPs on tomato plant. Particularly, when exposed to environmentally relevant concentrations (ERC, 0.02% w/w) of MPs, the addition of earthworms significantly (p < 0.05) increased shoot and root dry weight by 12-13% and 13-14%, respectively. MPs significantly reduced (p < 0.05) soil ammonium (NH4+-N) (0.55-0.69 mg/kg), nitrate nitrogen (NO3--N) (7.02-8.65 mg/kg) contents, and N cycle related enzyme activities (33.47-42.39 μg/h/g) by 37.7-50.9%, 22.6-37.2%, and 34.2-48.0%, respectively, while earthworms significantly enhanced (p < 0.05) inorganic N mineralization and bioavailability. Furthermore, earthworms increased bacterial network complexity, thereby enhancing the robustness of the bacterial system to resist soil MPs stress. Meanwhile, partial least squares modelling showed that earthworms significantly influenced (p < 0.01) soil nutrients, which in turn significantly affected (p < 0.01) plant growth. Therefore, the comprehensive consideration of soil ecological composition is important for assessing MPs ecological risk.

Abstract:Microplastics (MPs) pollution is identified as an important new global change factor potentially influencing ecosystem functions and services. MPs have been shown to strongly affect soil biota. However, whether soil biota respond differently to biodegradable and conventional MPs, and the generalizable pattern of responses of soil biota to MPs size, dose and the time of exposure to MPs remain poorly understood. Here we conducted a global-scale meta-analysis of 781 observations from 92 publications to investigate the effects of MPs on soil biota (i.e., microbiota and fauna). We also explored how soil biota respond to different experimental conditions (e.g., MPs properties and experimental duration). Results showed that MPs significantly increased soil microbial biomass, while decreased the growth and reproduction of soil fauna (nematodes, springtails and earthworms). However, MPs significantly decreased soil bacterial diversity (Shannon index) and shifted microbial community structure, and the effect of MPs on soil bacteria was more intense than that on soil fungi. The responses of soil biota were dependent on MPs types, sizes, concentrations and experimental duration. For example, biodegradable MPs had stronger impacts on soil microbial community, while had weaker negative impacts on soil fauna than conventional MPs. Likewise, smaller size of MPs appeared to have more adverse impacts on soil bacterial diversity, and the growth and reproduction of nematodes, but this trend was opposite in the reproduction of earthworms. Furthermore, the response of soil microbial community and fauna significantly correlated with the MPs concentration and experimental duration, suggesting that soil biota exposed to MPs for high concentrations and long periods were more vulnerable. Regardless of specific experimental conditions, these results highlight that prolonged MPs pollution may have considerable negative effects on the structure and function of soil biota and consequently affect the ability of soil ecosystems to provide ecosystem services and sustain human societies.

Abstract:Microplastics (MPs) are defined as plastic debris with particle size smaller than 5 mm, which have been frequently detected in environments. In this study, earthworms (Eisenia foetida) were exposed to three different sized polystyrene (PS; 0.1, 10 and 100 μm) at 10 and 100 mg per kg soil for 21 days. We examined the contents of metals (Cu, Zn, Ni and Pb) and gut microbial communities in earthworms exposed to MPs. Results showed that MPs reduced the accumulation of Ni and Pb in earthworms on the 21st day. The composition of gut bacterial communities was altered in earthworms exposed to MPs, especially 10 μm MPs, featuring a higher relative abundance of Proteobacteria (44.5%) and Bacteroidetes (27.1%) than the control group. Additionally, gut microorganisms including genus Paenibacillus and Achromobacter in earthworms were expected to be potential biomarkers for Cu and Zn. The microbial community networks showed that MPs affected bacterial community connections by size effect, and MPs with smaller size increased the complexity of earthworm microbial community networks. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that metabolism was the main difference in the bacterial communities in different treatments, especially carbohydrate metabolism and amino acid metabolism, which was relevant to the survival and growth of bacteria. This study provides insight into the environmental risks of MPs on terrestrial organisms.

Abstract:Global concern continues to mount regarding the accumulation of microplastics (MPs) in soil. However, little is known about how various types of MPs influence the properties of soil ecosystems. Here, we evaluated the effects of six different types of MPs, including low-density polyethylene (LDPE), polyamide (PA), polystyrene (PS), polyhydroxy-alkanoates (PHA), polybutadiene styrene (PBS), and polylactide (PLA), on soil physicochemical properties, enzyme activities, and microbial communities. At the end of a 230-day soil incubation, we observed significant changes in soil moisture content, soil organic carbon, pH, NH4+-N, NO3--N, and available phosphorus. The addition of MPs had a significant influence on the activities of soil β-glucosidase, acid phosphatase, urease, and fluorescein diacetate hydrolase, with effects varying with MP type. Results of 16S rRNA gene high throughput sequencing showed that MP exposure had little effect on soil microbial alpha diversity, but that PHA contamination significantly reduced ACE, Chao1, and Shannon index values. MP contamination also altered soil microbial community composition. In particular, the relative abundance of Firmicutes increased significantly while the relative abundance of Actinobacteriota, Proteobacteria (especially the nitrogen-fixing rhizobia), and Acidobacteriota decreased following exposure to PHA. Redundancy analysis showed that acid phosphatase and pH were the two main environmental factors affecting bacterial community structure at the phylum and order levels. Furthermore, Tax4Fun2 analysis found that MP treatment disrupted fundamental bacterial metabolic pathways. Our findings indicate that different types of MPs can affect soil fertility, bacterial community structure, and function in various ways, and highlight that biodegradable MPs may alter soil bacterial communities more than conventional MPs.

Abstract:placed intraluminally, the potential for graft dilatation may be limited by the surrounding aortic wall, but its natural history is yet to be defined. Nevertheless, transluminally placed endovascular grafts present a less invasive approach to treatment of aneurysms and therefore have tremendous intrinsic appeal. There is need for well controlled clinical trials in which the various transluminally placed endovascular graft devices are compared with standard operative treatment methods in groups of patients with aortic aneurysm morphology and similar risk factors. In particular, these studies will need to document the effective long-term treatment of patients with aneurysms having acceptable early and late patency and morbidity and mortality rates. With cautious optimism, it is conceivable that these endovascular devices may replace anywhere between 30% to 70% of current vascular prosthetic grafts, depending on how effective and safe they prove to be.

Abstract:Soils always receive microplastics (MPs) from plastic mulching, compost, and sewage irrigation, but the effects of MPs on soil environment remain largely unexplored. The objectives of this study were to investigate the effects of three MPs (membranous polyethylene (PE), fibrous polypropylene (PP), and microsphere PP) on enzyme activities and microbial community structure in one loamy and sandy soil. The concentration of microsphere PP (2 mg g‐1) was one‐tenth of those of the other two MPs (20 mg g‐1). The results showed that the effects of three MPs on urease, dehydrogenase and alkaline phosphatase activities followed the order: fibrous PP > membranous PE > microsphere PP, membranous PE > microsphere PP > fibrous PP and fibrous PP > microsphere PP > membranous PE, respectively. Results from high‐throughput sequencing of 16S rRNA revealed that the membranous PE and fibrous PP raised the alpha‐diversities of the soil microbiota, whereas the diversity indexes of microbiota on MPs surfaces were significantly lower than those in the amended soils. MPs significantly altered the microbial community structure, especially for the enrichment of Acidobacteria and Bacteroidetes, the depletion of Deinococcus‐Thermus and Chloroflexi. Aeromicrobium, Streptomyces, Mycobacterium, Janibacter, Nocardia, Arthrobacter were prone to inhabit on the MPs surfaces.

Abstract:Microplastics (MPs), including conventional hard-to-biodegrade petroleum-based and faster biodegradable plant-based ones, impact soil structure and microbiota in turn affecting the biodiversity and functions of terrestrial ecosystems. Herein, we investigated the effects of conventional and biodegradable MPs on aggregate distribution and microbial community composition in microhabitats at the aggregate scale. Two MP types (polyethylene (PE) and polylactic acid (PLA) with increasing size (50, 150, and 300 μm)) were mixed with a silty loam soil (0-20 cm) at a ratio of 0.5 % (w/w) in a rice-wheat rotation system in a greenhouse under 25 °C for one year. The effects on aggregation, bacterial communities and their co-occurrence networks were investigated as a function of MP aggregate size. Conventional and biodegradable MPs generally had similar effects on soil aggregation and bacterial communities. They increased the proportion of microaggregates from 17 % to 32 %, while reducing the macroaggregates from 84 % to 68 %. The aggregate stability decreased from 1.4 mm to 1.0-1.1 mm independently of MP size due to the decline in the binding agents gluing soil particles (e.g., microbial byproducts and proteinaceous substances). MP type and amount strongly affected the bacterial community structure, accounting for 54 % of the variance. Due to less bioavailable organics, bacterial community composition within microaggregates was more sensitive to MPs addition compared to macroaggregates. Co-occurrence network analysis revealed that MPs exacerbated competition among bacteria and increased the complexity of bacterial networks. Such effects were stronger for PE than PLA MPs due to the higher persistence of PE in soils. Proteobacteria, Bacteroidetes, Chloroflexi, Actinobacteria, and Gemmatimonadetes were the keystone taxa in macroaggregates, while Actinobacteria and Chloroflexi were the keystone taxa in microaggregates. Proteobacteria, Actinobacteria, and Chloroflexi were the most sensitive bacteria to MPs addition. Overall, both conventional and biodegradable MPs reduced the portion of large and stable aggregates, altering bacterial community structures and keystone taxa, and consequently, the functions.

Abstract:Soils are both a sink and a pathway of plastic wastes, but there is a great lack of knowledge regarding their impacts on soil biota. To tackle the mechanisms of toxicity of these contaminants to soil invertebrates, earthworms (Eisenia fetida Savigny, 1826) were exposed during 28 days to different concentrations of low-density polyethylene microplastics (62, 125, 250, 500 and 1000 mg MPs kg−1 soildw) with sizes ranging between 250 and 1000 μm, in an artificial soil. The ecotoxicological responses were evaluated by analysing various oxidative stress biomarkers (catalase, glutathione S-transferase and thiobarbituric acid reactive substances), a biomarker of energy metabolism (lactate dehydrogenase) and overall organism molecular changes by Fourier transform infrared spectrometry (FTIR) and nuclear magnetic resonance (NMR) analyses. Significant effects resulting from an unbalanced oxidative stress system, expressed in terms of thiobarbituric acid reactive substances levels were recorded on earthworms exposed at the three highest concentrations tested. Despite that, no significant changes were recorded on the molecular profiles of earthworms by FTIR-ATR. NMR analysis pointed out for differences from the control, only for earthworms exposed to the lowest concentration of MPs. Considering that stress responses are complex, and involve multiple mechanisms, a cluster analysis taking into account all the parameters assessed, clearly identified two groups of earthworms separated by the concentration of 250 mg MPs kg−1 soildw, above each meaningful effects were recorded.

Abstract:Microplastics derived from polyethylene (PE) mulch films are widely found in farmland soils and present considerable potential threats to agricultural soil ecosystems. However, the influence of microplastics derived from PE mulch films, especially those derived from farmland residual PE mulch films, on soil ecosystems remains unclear. In this study, we analyzed the bacterial communities attached to farmland residual transparent PE mulch film (FRMF) collected from peanut fields and the different ecological effects of unused PE mulch film-derived microplastics (MPs) and FRMF-derived microplastics (MPs-aged) on the soil and earthworm Metaphire guillelmi gut microbiota, functional traits, and co-occurrence patterns. The results showed that the assembly and functional patterns of the bacterial communities attached to the FRMF were clearly distinct from those in the surrounding farmland soil, and the FRMF enriched some potential plastic-degrading and pathogenic bacteria, such as Nocardioidaceae, Clostridiaceae, Micrococcaceae, and Mycobacteriaceae. MPs substantially influenced the assembly and functional traits of soil bacterial communities; however, they only significantly changed the functional traits of earthworm gut bacterial communities. MPs-aged considerably affected the assembly and functional traits of both soil and earthworm gut bacterial communities. Notably, MPs had a more remarkable effect on nitrogen-related functions than the MPs-aged in numbers for both soil and earthworm gut samples. Co-occurrence network analysis revealed that both MPs and MPs-aged enhanced the synergistic interactions among operational taxonomic units (OTUs) of the composition networks for all samples. For community functional networks, MPs and MPs-aged enhanced the antagonistic interactions for soil samples; however, they exhibited contrasting effects for earthworm gut samples, as MPs enhanced the synergistic interactions among the functional contents. These findings broaden and deepen our understanding of the effects of FRMF-derived microplastics on soil ecosystems, suggesting that the harmful effects of aged plastics on the ecological environment should be considered.

Abstract:While much is known about microplastics in aquatic environments, only few studies have focused on how microplastics interact with terrestrial organisms. The objective of our study was to investigate the interactions of earthworms with microplastics in a natural environment with environmentally realistic plastic concentrations. We investigated whether earthworms would ingest microplastics and incorporate them into their bodies and cast, and whether microplastics would alter the intestinal and soil microbiome. Lumbricus terrestris was exposed to two types of microplastics, biodegradable polybutylene adipate terephthalate (PBAT) and low-density polyethylene (LDPE) in mesocosoms filled with natural soil. Microplastics were mixed with poplar leaves, and earthworms were allowed to forage for food for 20 days. Surface and bulk soil, earthworm’s cast, and earthworms themselves were sampled and analyzed for plastic content and microbial communities. Earthworms did not show macroscopic health deterioration (weight loss, vitality). We observed microplastics particles in earthworm casts and guts. Raman spectroscopy indicated that PBAT in guts and cast had degraded to some extent; however, LDPE remained unchanged among the different samples. Microbial analysis showed that soil and casts has similar microbial communities; however, they were significantly different from the gut samples. Microplastic treatments did not result in a statistically significant change in bacterial richness, diversity, or community composition for soil, casts, or guts compared to controls. Taken together, our results suggest that, at environmentally realistic concentrations and short exposure times, PBAT and LDPE microplastics do not have adverse effects on Lumbricus terrestris earthworms.
Effect thresholds for the earthworm Eisenia fetida: Toxicity comparison between conventional and biodegradable microplastics

Abstract:Biodegradable plastics have been developed to eliminate the progressive accumulation and ever-growing threat posed by conventional fossil fuel-derived plastics. The impact of these bioplastics, particularly in an agricultural context (e.g. biopolymer mulch films), however, remains poorly understood. In this study, we compared the biotoxicity of biodegradable (polylactic acid, PLA; polypropylene carbonate, PPC) and non-degradable (polyethylene, PE) microplastics using a series of standardized bioassays using the earthworm Eisenia fetida. The responses studied included: avoidance behavior, mortality, biomass, and reproduction responses. We incubated earthworms in artificial soils amended with different concentrations of microplastic (0, 0.125, 1.25, 12.5, 125, 250, and 500 g kg−1) under laboratory conditions. This wide range allowed linear regression modeling and estimation of microplastic effect thresholds. Our results showed that microplastic concentration rather than plastic type was more important in regulating earthworm responses to soil contamination. The critical threshold for microplastic contamination was 40 g kg−1, after which earthworms exhibit microplastic avoidance behavior. A significant reduction (EC10) in number of cocoons and juvenile earthworms occurred at a concentration of 53 g kg−1 and 97 g kg−1, respectively; while no significant effect was found for survival of earthworm until levels of 500 g kg−1. Overall, the two biodegradable materials (PLA and PPC), appeared to be no more biofriendly than PE. Based on reported levels of plastic contamination in soil of up to 67 g kg−1, we conclude that microplastics are now starting to pose a threat to earthworm population. To better evaluate the risk posed by biodegradable and nondegradable plastics, further mechanistic studies on how microplastics affect earthworm behavior and the potential long-term impacts of this on soil functioning are required.

Abstract:Plastic contamination in agricultural soils has become increasingly evident. Plastic mulching films are widely used in agricultural practices. However, the increased use of biodegradable plastics has, to some extent, replaced their non-degradable counterparts. The fragmentation of plastics generates microplastics (MPs), posing risk to soil functions and organisms. In this study the effects of low-density polyethylene microplastics (PE-MP) and polybutylene adipate terephthalate biodegradable microplastics (PBAT-BD-MP) originating from mulching films on the earthworm Eisenia andrei were studied. The earthworms were exposed to seven concentrations (0, 0.005, 0.05, 0.1, 0.5, 1, and 5 % w/w) based on environmentally relevant levels and worst-case scenarios on soil contamination. Survival, growth, reproduction, and biomarkers for oxidative stress [superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), glutathione S-transferase (GST), glutathione (GSH), and lipid peroxidation (LPO)] were analysed. Additionally, the Integrated Biomarker Response Index (IBR) was calculated to assess the overall oxidative stress status of the earthworms. Results showed that PE-MP exposure slightly decreased the biomass of the earthworms towards higher concentrations, whereas PBAT-BD-MPs induced growth at lower concentrations. MPs did not have a significant effect on Eisenia andrei reproduction; however, a slight negative trend was observed in juvenile production with increasing PE-MP concentrations. Both PE-MP and PBAT-BD-MP affected antioxidant system, PE-MPs with changes in CAT and GR levels and PBAT-BD-MPs inducing effects on SOD and LPO levels. Additionally, both MPs exhibited effects on soil parameters, resulting in increased soil pH and water-holding capacity at 5 % concentration. Changes in soil parameters can further affect soil organisms such as earthworms. This study provides understanding of the ecotoxicological effects of conventional and biodegradable microplastics on the earthworm Eisenia andrei. It also shows that MP particles of both conventional and biodegradable mulching films induce oxidative stress, considered as an early-warning indicator for adverse ecological effects, in environmentally relevant concentrations.
Microplastics and Tcep Pollutants in Agricultural Soil: How Do They Disrupt Earthworm Homeostasis by Interfering with Their Metabolites and Gut Microbiota?

Abstract:Agricultural soil contamination with plastic film has become a critical global environmental problem, requiring greater research on the possible co-exposure effects of microplastics (MPs) and their additives in soil ecosystems. The presence of MPs and tris (2-chloroethyl) phosphate (Tcep) in agricultural soil was investigated at nine sites in this study. Polyethylene MPs (PE-MPs) and Tcep were found at all nine sites. To study co-exposure effects on soil microbiota and earthworms, and to model a more likely exposure situation, 0.05% (w/w) PE-MPs with three particle size ranges were combined with 1.0 mg/kg Tcep. After 28 days of exposure, there was no indication that added PE-MPs and/or Tcep significantly impacted the soil microbial community structure. In earthworm, size-selective intake, digestion, and egestion of PE-MPs may occur, and Tcep co-exposure affects the residual Tcep concentration in earthworm intestines (3.52–9.31 μg/g dw). Long-term earthworm PE-MPs intake caused intestinal damage, and Tcep co-exposure increased oxidative stress, thereby influencing their feeding behavior and growth, resulting in weight loss (3.42%–14.96%), especially for the most common PE-MPs sizes (0–300 μm). An LC-MS-based metabolomic analysis revealed significantly up-regulated citrate ( p < 0.001) and down-regulated L-glutamate ( p < 0.05) in co-exposure groups altered most of the metabolic pathways, thereby impairing nervous, digestive, and excretory systems, with an associated decrease in amino acid metabolism and changes in tricarboxylic acid (TCA) cycle intermediates. The importance of gut microbiota, such as Proteobacteria ( Verminephrobacter and Bradyrhizobium ) and Firmicutes ( Bacillus ), in maintaining earthworm metabolic homeostasis, particularly for the TCA cycle and amino acid metabolism, cannot be understated. Overall, MPs and Tcep co-exposure in agricultural soil enhanced their earthworm toxicity, and may potentially endanger the development of agricultural sustainability.
