Abstract:Objective To study the chemical constituents from the fruits of Lycium barbarum.Methods The compounds were isolated and purified by column chromatography and RP-HPLC.Their structures were elucidated by means of physicochemical properties and spectroscopic analyses(UV,MS,1H-NMR,and13C-NMR).Results Twelve compounds were separated and identified,including six phenolic acids: protocatechuic acid(1),dihydroisoferulic acid(2),caffeic acid(3),Z-p-hydroxy-cinnamic acid(4),E-p-hydroxy-cinnamic acid(5),and trans-cinnamic acid(6);Three coumarins: scopoletin(7),isoscopoletin(8),esculetin(9);Two pyrrole derivatives: 4-[formyl-5-(hydroxymethyl)-1H-pyrrol-1-yl] butanoic acid(10) and 4-[formyl-5-(methoxymethyl)-1H-pyrrol-1yl] butanoic acid(11);and 4′-hydroxyacetophenone(12).Conclusion Except compounds 3,5,and 7,others are isolated from this plant for the first time.Compounds 2,4,8,9,and 12 are firstly isolated from the plants in genus of Lycium L.

Abstract:A taste-sensing system was used to study dried Lycium fruits of different producing year and grades. Physical indexes were measured to classify the grade of Lycium fruits and to find correlation between grade indexes and taste characterization. The least significant difference, principal component analysis and linear regression were applied in the data processing. Significant differences were found for sourness, bitterness, astringency and aftertaste-B. Classifications according to producing years and grades were found by principle component analysis. Strong correlations between producing year, grades and taste characterization were found.Practical ApplicationsLycium barbarumL. has been used as traditional medicine in Asian and many countries. This study evaluates the taste characterization and its application in determination of producing years and grades of Lycium fruits. With multivariate analysis, it is possible to detect producing years and grades of dried Lycium fruits with taste characterization, providing evidence for fast adulteration detection of Lycium fruits.

Abstract:Lycium fruits have a high content of phenolics as bioactive constituents with various pharmacological effects, but there is a lack of comparative analysis and chemical profiling of phenolics in Lycium fruit varieties. An ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MSE) combined with chemometrics was developed to characterize the phenolics in fruits from four Lycium species, including Lycium barbarum L. (LBL), L. chinense Mill. (LCM), L. barbarum var. auranticarpum (LBA) and L. ruthenicun Murr. (LRM). 63 phenolics were identified according to reported tandem mass fragmentation patterns and the UNIFI scientific informatics platform. Nine phenolics (5, 18, 20, 29, 31, 37, 41, 43, 60) were common and predominant components among four types of Lycium fruit. The partial least squares discriminant analysis (PLS-DA) and the orthogonal partial least squares discriminant analysis (OPLS-DA) were analyzed on the basis of a matrix created from 653 sets of data, and 20 Lycium fruits were classified into four groups. Further analysis identified that phenolics profiles were characteristic for each Lycium species, and five markers (13, 29, 31, 35, 99) could be utilized for fruit identification. Subsequently, inhibitory activity against 5 alpha-reductase of phenolic extracts of Lycium fruits showed that LBL extract was the relative better effective, followed by LCM, whereas LBA and LRM showed no activity, which might be associated with the high contents of marker compounds (29, 31, 35, 43, 71, 99) in LBL. These findings will provide guidance for the development of Lycium phenolics with beneficial properties for the prevention and treatment of Benign prostatic hyperplasia (BPH). Comparative Analysis of Polyphenols in Lycium barbarum Fruits Using UPLC-IM-QTOF-MS

Abstract:Variety, geographical origin, and harvest season are important factors affecting the accumulation of polyphenols in Lycium barbarum. In this study, the effects of these factors on the polyphenolic components of this species were analyzed using ultra-performance liquid chromatography ion mobility quadrupole time-of-flight mass spectrometry. Moreover, the in vitro antioxidant activities of fruit extracts from this species were evaluated. The total polyphenolic contents of L. barbarum fruits from Jinghe County in Xinjiang and Zhongning County in Ningxia were 5.52–11.72 and 7.06–9.37 mg (gallic acid equivalent)/g dry weight, while the total flavonoid contents of L. barbarum fruits from these regions were 12.52–30.29 and 12.67–20.77 mg (rutin equivalent)/g dry weight, respectively. Overall, 39 types of polyphenols were identified in the fruit extracts, including 26 flavonoids, 10 phenolic acids, and three tannins. Of these, 11 polyphenols were quantitatively analyzed, which revealed rutin to be the most dominant polyphenolic component in fruits from Jinghe and Zhongning. There were significant differences (p < 0.05) in the polyphenolic contents and antioxidant activities of L. barbarum fruit extracts, depending on the geographical origin, variety, and harvest season. The antioxidant activity of this species was found to be significantly positively correlated with the polyphenolic contents. This study provided scientific guidance for comprehensive applications of polyphenols from different varieties of L. barbarum from separate geographical origins.

Abstract:The Lycium genus, perennial herbs of the Solanaceae family, has been an important source of medicines and nutrient supplements for thousands of years in China, where seven species and three varieties are cultivated. Among these, Lycium barbarum L. and Lycium chinense Mill., two "superfoods", together with Lycium ruthenicum Murr, have been extensively commercialized and studied for their health-related properties. The dried ripe fruits of the genus Lycium are well recognized as functional foods for the management of various ailments including waist and knee pain, tinnitus, impotence, spermatorrhea, blood deficiency and weak eyes since ancient times. Phytochemical studies have reported numerous chemical components in the Lycium genus, categorized as polysaccharides, carotenoids, polyphenols, phenolic acids, flavonoids, alkaloids and fatty acids, and its therapeutic roles in antioxidation, immunomodulation, antitumor treatment, hepatoprotection and neuroprotection have been further confirmed by modern pharmacological studies. As a multi-functional food, the quality control of Lycium fruits has also attracted attention internationally. Despite its popularity in research, limited systematic and comprehensive information has been provided on the Lycium genus. Therefore, herein, we provide an up-to-date review of the distribution, botanical features, phytochemistry, pharmacology and quality control of the Lycium genus in China, which will provide evidence for further in-depth exploration and comprehensive utilization of Lycium, especially its fruits and active ingredients in the healthcare field.
Interspecies Developmental Differences in Metabonomic Phenotypes of Lycium Ruthenicum and L-barbarum Fruits

Abstract:Fruits of Lycium ruthenicum (LR) and L. barbarum (LB) in Solanaceae family contain abundant bioactive metabolites used widely as functional food and natural medicine. To characterize the fruit developmental molecular phenotypes, we comprehensively analyzed metabolite composition of both Lycium fruits at three developmental stages using the combined NMR, liquid chromatography tandem mass spectrometry, and gas chromatography-flame ionization detector/mass spectrometry methods. The metabonomes of these fruits were dominated by over 90 metabolites including sugars, amino acids, tricarboxylic acid (TCA) cycle intermediates, fatty acids, choline metabolites, and shikimate-mediated plant secondary metabolites. Metabolic phenotypes of two species differed significantly at all three developmental stages; LB fruits contained significantly more sugars and amino acids but less TCA cycle intermediates, fatty acids, and secondary metabolites than LR. Interspecies differences for fatty acid levels were much greater after color-breaking than precolor-breaking. Furthermore, LR fruits contained more osmolytes than LB fruits indicating different osmoregulation requirements for these fruits during development. Significant differences were also present in biosynthesis of shikimate-mediated plant secondary metabolites in LR and LB. These findings provided essential metabolic information for plant physiology of these Lycium species and their utilizations demonstrating the usefulness of this metabonomic phenotyping approach for studying fundamental biochemistry of the plant development.
Metabolite Differences of Polyphenols in Different Litchi Cultivars (Litchi chinensis Sonn.) Based on Extensive Targeted Metabonomics

Abstract:Litchi is an important fruit cultivated in tropical and subtropical areas with high nutritious and delicious flavor and the pulp is the main part of the fruit consumed. Previous studies found that litchi had high total phenol content and antioxidant activity, but most of them focused on the identification of single or a few phenolic components with a low throughput test, and the metabolic differences of cultivars are still unknown to a some extent. In this study we used widely targeted metabolome based on ultra-performance liquid chromatography coupled with mass spectrometry (UPLC-MS/MS) to analyze the polyphenol metabolites of five different genotypes of mature litchi fruit. A total of 126 polyphenol metabolites in eight categories were identified to reveal the composition and differences of polyphenol; 15 common differential metabolites and 20 specific differential metabolites to each cultivar were found for the first time. The results infer that flavonoids, flavonols, hydroxycinnamoyls and catechins are the main polyphenol metabolites of litchi pulp. Cluster analysis showed that there were three groups of polyphenols from high to low; early maturing Feizhixiao is a kind of high polyphenol content cultivars, especially in catechins, anthocyanins, flavonols, quinic acids and hydroxycinnamoyls. The polyphenols in the flesh of mature litchi are rich, and there are significant differences among cultivars; there was a level of correlation between the contents of phenolics and the maturity of litchi cultivars; the content of phenolics in early maturing litchi cultivars appeared higher than those of mid- to late-maturing cultivars. This experiment will provide significant reference information for cultivation, breeding, processing and consumption.

Abstract:There is much variation in the distributions of secondary metabolites, such as flavonoids, among plant tissues. The flavonoids in Chinese jujube ( Ziziphus jujuba Mill.) and sour jujube ( Ziziphus spinosa Hu.) provide various health benefits. Variation in the distributions of flavonoids among the tissues of jujube and sour jujube has not yet been characterized. A total of 101 flavonoid metabolites, including 30 flavones, 21 flavonols, 11 flavanones, 11 isoflavanones, 8 flavanonols, 7 chalcones, 4 flavanols, 3 flavone glycosides, 2 phenolic acids, 2 ×anthones and 2 unclassified flavonoids, were identified in the fruits, leaves, flowers and seeds of Z. jujuba cv . Dongzao (DZ) and Z. spinosa cv . Liyuanzhenzhu8 (LYZZ8) using UPLC-ESI MS/MS. Principal component analysis and hierarchical cluster analysis revealed that flavonoids from the same tissues of DZ and LYZZ8 were clustered, with the exception of those from the leaves. Tissue-specific flavonoid metabolites in different tissues of DZ and LYZZ8 were identified. The main flavonoids varied among tissues, the total flavonoid content and the content of most individual flavonoids in all tissues of DZ and LYZZ8 varied ( p < .05). Furthermore, 40, 50, 49 and 47 differential flavonoid metabolites were detected in the fruits, leaves, flowers and seeds of DZ and LYZZ8, respectively. Potential chemical markers for differentiating among tissues in DZ and LYZZ8 as well as among the same tissues of DZ and LYZZ8 were identified. Overall, our findings provided new insights into the flavonoid of jujube and will aid the utilization of jujube and sour jujube.

Abstract:Wolfberry (Lycium barbarum) is a traditional cash crop in China and is well-known worldwide for its outstanding nutritional and medicinal value. Lycium ruthenicum is a close relative of Lycium barbarum but differs significantly in size, color, flavor and nutritional composition. To date, the metabolic differences between the fruits of the two wolfberry varieties and the genetic basis behind them are unclear. Here, we compared metabolome and transcriptome data of two kinds of wolfberry fruits at five stages of development. Metabolome results show that amino acids, vitamins and flavonoids had the same accumulation pattern in various developmental stages of fruit but that Lycium ruthenicum accumulated more metabolites than Lycium barbarum during the same developmental stage, including L-glutamate, L-proline, L-serine, abscisic acid (ABA), sucrose, thiamine, naringenin and quercetin. Based on the metabolite and gene networks, many key genes that may be involved in the flavonoid synthesis pathway in wolfberry were identified, including PAL, C4H, 4CL, CHS, CHI, F3H, F3'H and FLS. The expression of these genes was significantly higher in Lycium ruthenicum than in Lycium barbarum, indicating that the difference in the expression of these genes was the main reason for the variation in flavonoid accumulation between Lycium barbarum and Lycium ruthenicum. Taken together, our results reveal the genetic basis of the difference in metabolomics between Lycium barbarum and Lycium ruthenicum and provide new insights into the flavonoid synthesis of wolfberry.

Abstract:A novel monoclonal rat IgM antibody (8F7) is described which detects the inducible 68 kDa heat shock protein (hsp68) in man, mouse, rat, pig and cattle. Hsp68 expression is analysed in various cell types and cell lines by immunoblot with antibody 8F7 and in parallel by autoradiography after metabolic labeling with [35S]methionine. The new antibody cross-reacts with proteins of 180 and 48 kDa, which are not heat shock-inducible and show a pattern of expression different than hsp68.

Abstract:In order to explore the chemical structure, immunoactivity and the structure-activity relationship of Lycium barbarum polysaccharides, 5 polysaccharide components (LBP1 - LBP5) were separated and purified from the crude Lycium barbarum polysaccharides(LBP) with the methods of chromatography, gel filtration and SDS-PAGE electrophoresis, and from LBP1, LBP3, LBP4 the glycoconjugates LbGp1, LbGp3, LbGp4 and their glycan chains LbGp1-OL, LbGp3-OL, LbGp4-OL were also obtained, the chemical characteristics, carbohydrate components, amino acid components and major structure of which were elucidated with methylation, fractional acid hydrolization and NMR techniques. The six samples have direct elevation effect on splenocyte proliferation in mice and the effects of glycan chains were stronger than that of respective conjugates. These results suggest that the immunoactive components of the fruit of Lycium barbarum L. be a kind of carbohydrate conjugates with complex structure. The glycan chains may be the important active structure of carbohydrate conjugates.

Abstract:In order to fully understand the variations of fruit quality-related phytochemical composition in Chinese bayberry (Myrica rubra Sieb. et Zucc.), mature fruit of 17 cultivars from Zhejiang and Jiangsu provinces was used for the investigation of fruit quality attributes, including fruit color, soluble sugars, organic acids, total phenolics, flavonoids, antioxidant capacity, etc. Sucrose was the main soluble sugar, while citric acid was the main organic acid in bayberry fruit. The content of total phenolics and total flavonoids were positively correlated with 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) antioxidant activity and 2,2ʹ-azino-bis(3-ethylbenzothiazoline- 6-sulphonic acid) (ABTS) radical scavenging activity. Five anthocyanidins, i.e., delphinidin–hexoside (Dp–Hex), cyanidin-3–O-galactoside (C-3–Gal), cyanidin-3–O-glucoside (C-3–Glu), pelargonidin-3–O-glucoside (Pg-3–Glu) and peonidin-3-O-glucoside (Pn-3–Glu), and seven flavonols compounds, i.e., myricetin-3-O-rhamnoside (M-3–Rha), myricetin deoxyhexoside–gallate (M-DH–G), quercetin-3-O-galactoside (Q-3–Gal), quercetin-3– O-glucoside (Q-3–Glu), quercetin-3–O-rhamnoside (Q-3–Rha), kaempferol-3–O-galactoside (K-3–Gal) and kaempferol-3–O-glucoside (K-3–Glu), were identified and characterized among the cultivars. The significant differences in phytochemical compositions among cultivars reflect the diversity in bayberry germplasm, and cultivars of good flavor and/or rich in various health-promoting phytochemicals are good candidates for future genetic breeding of bayberry fruit of high quality. In conclusion, our results may provide important information for further breeding or industrial utilization of different bayberry resources.

Abstract:The molecular weight, the triple-helix conformation, the monosaccharide content, the manner of glycosidic linkages, and the polysaccharide conjugates of polysaccharides all affect bioactivity. The purpose of this study was to determine how different molecular weights affected the bioactivity of the Lycium barbarum polysaccharides (LBPs). By ethanol-graded precipitation and ultrafiltration membrane separation, one oligosaccharide (LBPs-1, 1.912 kDa) and two polysaccharides (LBPs-2, 7.481 kDa; LBPs-3, 46.239 kDa) were obtained from Lycium barbarum. While the major component of LBPs-1 and LBPs-2 was glucose, the main constituents of LBPs-3 were arabinose, galactose, and glucose. LBPs-2 and LBPs-3 exhibited triple-helix conformations, as evidenced by the Congo red experiment and AFM data. Sugar residues of LBPs-2 and LBPs-3 were elucidated by NMR spectra. The polysaccharides (LBPs-2 and LBPs-3) exhibited much higher antioxidant capacities than oligosaccharide (LBPs-1). LBPs-3 showed higher oxygen radical absorbance capacity (ORAC) and superoxide dismutase (SOD) activity than LBPs-2, but a lower capability for scavenging ABTS+ radicals. In zebrafish, LBPs-2 and LBPs-3 boosted the growth of T-lymphocytes and macrophages, enhanced the immunological response, and mitigated the immune damage generated by VTI. Abstract:Lonicerae japonicae flos (LJF), Lonicerae japonicae caulis (LJC), Lonicerae folium (LF) and Lonicerae fructus (LFR) are derived from Lonicera japonica Thunb., which are formed due to different medicinal parts. The efficacy of the 4 medicinal materials has similarities and differences. However, little attention has been paid to illustrate the differences in efficacy from the perspective of phytochemistry. In this study, ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry (UPLC-Q-Exactive-Orbitrap-MS) was used to qualitatively analyze the ingredients in 4 herbs. A total of 86 compounds were plausibly or unambiguously identified, there were 54 common components among the 4 medicinal materials, and each kind of medicinal materials had its own unique components. On the basis of qualitative analysis, ultra-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UPLC-QQQ-MS/MS) was used to quantitatively analyze 31 components contained in 4 medicinal materials, and principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA) and other multivariate statistical analysis were furtherly performed for comparing the component contents. The results showed that the samples from the same parts were clustered into one group, and the samples from different medicinal parts were significantly different. The analysis of variable importance projection (VIP) value of the OPLS-DA model showed that 10 components including chlorogenic acid, secologanic acid, isochlorogenic acid A, loganin, lonicerin, loganic acid, secoxyloganin, sweroside, luteolin and rhoifolin were the main difference components among the 4 medicinal materials. The study not only lays a solid foundation for the intrinsic quality control of 4 medicinal materials and the study of different effects of the 4 medicinal materials at the phytochemical level, but also provides a basis for more rational utilization of various parts of L. japonica and expansion of medicinal resources.
Comparison of Bioactive Compounds and Antioxidant Activities in Differentially Pigmented Cerasus humilis Fruits

Abstract:Chinese dwarf cherry (Cerasus humilis) is a wild fruit tree and medicinal plant endemic to China. Its fruits are rich in various bioactive compounds, such as flavonoids and carotenoids, which contribute greatly to their high antioxidant capacity. In this study, the contents of bioactive substances (chlorophyll, carotenoids, ascorbic acid, anthocyanin, total flavonoids, and total phenols), antioxidant capacities, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonicacid) (ABTS+) scavenging ability, and ferric-reducing antioxidant power (FRAP)) in differentially pigmented C. humilis fruits of four varieties were determined and compared. The results revealed that anthocyanin, total flavonoids and total phenols were the three main components responsible for the antioxidant activity of C. humilis fruits. 'Jinou No.1' fruits with dark red peel and red flesh had the highest contents of anthocyanin, total flavonoids, and total phenols, as well as the highest antioxidant capacities; 'Nongda No.5' fruits with yellow-green peel and yellow flesh had the highest contents of carotenoids and chlorophyll, while 'Nongda No.6' fruit had the highest ascorbic acid content. To further reveal the molecular mechanism underlying differences in the accumulation of carotenoids and flavonoids among differentially pigmented C. humilis fruits, the expression patterns of structural genes involved in the biosynthesis of the two compounds were investigated. Correlation analysis results revealed that the content of carotenoids in C. humilis fruits was very significantly positively correlated with the expression of the ChCHYB, ChZEP, ChVDE, ChNSY, ChCCD1, ChCCD4, ChNCED1, and ChNCED5 genes (p < 0.01) and significantly negatively correlated with the expression of ChZDS (p < 0.05). The anthocyanin content was very significantly positively correlated with ChCHS, ChFLS, and ChUFGT expression (p < 0.01). The total flavonoid content was very significantly positively correlated with the expression of ChCHS, ChUFGT, and ChC4H (p < 0.01) and significantly positively correlated with ChFLS expression (p < 0.05). This study can provide a basis for understanding the differences in the accumulation of bioactive substances, and is helpful for clarifying the mechanisms underlying the accumulation of various carotenoids and flavonoids among differentially pigmented C. humilis fruits.
