Abstract:Biodegradable macroporous hydrogels were prepared using a biodegradable crosslinker based on chitosan derivatives. The chitosan crosslinker was synthesized through the amidating reaction of amino groups in the chitosan oligosaccharide (CSO) chains and carboxyl groups in acrylic acid (AA) under the catalysis of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride (EDC) and N-hydroxysuccinimide (NHS). The amidating reaction is able to provide polymerizable carbon-carbon double bonds for CSO. The chemical structure of the resulting compound chitosan oligosaccharide-graft-acrylic acid (CSO-g-AA) was characterized by Fourier transform infrared spectroscopy (FTIR) and H-1 nuclear magnetic resonance (H-1 NMR) spectroscopy. The content of double bonds was measured by titration analysis. The measurement showed a grafting ratio of AA on CSO of about 3.5% (on average two acryoyl groups per CSO-g-AA chain) when the applied molar ratio of AA to CSO was 1.0. Polyacrylamide (PAM) cryogels were synthesized by the free radical polymerization of acrylamide using a modified freezing polymerization method crosslinked by CSO-g-AA. The obtained cryogels were macroporous hydrogels which could be degraded into solutions of linear polymer chains when treated with appropriate enzymes. Then we used snailase as a model enzyme to study the biodegradation process. The degradation was monitored by morphological studies using a confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM), mechanical strength, and swelling ratio. The duration of the degradation process was adjustable from one month to two months when using different concentrations of snailase solutions.
Triethyl orthoformate mediated a novel crosslinking method for the preparation of hydrogels for tissue engineering applications: characterization and in vitro cytocompatibility analysis

Abstract:This paper describes the development of a new crosslinking method for the synthesis of novel hydrogel films from chitosan and PVA for potential use in various biomedical applications. These hydrogel membranes were synthesized by blending different ratios of chitosan (CS) and poly(vinyl alcohol) (PVA) solutions and were crosslinked with 2.5% (w/v) triethyl orthoformate (TEOF) in the presence of 17% (w/v) sulfuric acid. The physical/chemical interactions and the presence of specific functional groups in the synthesized materials were evaluated by Fourier transform infrared (FT-IR) spectroscopy. The morphology, structure and pore size of the materials were investigated by scanning electron microscopy (SEM). Thermal gravimetric analysis (TGA) proved that these crosslinked hydrogel films have good thermal stability which was decreased as the CS ratio was increased. Differential scanning calorimetry (DSC) exhibited that CS and PVA were present in the amorphous form. The solution absorption properties were performed in phosphate buffer saline (PBS) solution of pH7.4. The 20% PVA–80% CS crosslinked hydrogel films showed a greater degree of solution absorption (183%) as compared to other compositions. The hydrogels with greater CS concentration (60% and 80%) demonstrated relatively more porous structure, better cell viability and proliferation and also revealed good blood clotting ability even after crosslinking. Based on the observed facts these hydrogels can be tailored for their potential utilization in wound healing and skin tissue engineering applications.

Abstract:Biocompatible hydrogels based on water-soluble chitosan-ethylene glycol acrylate methacrylate (CS-EGAMA) and polyethylene glycol diamethacrylate (PEGDMA) were synthesized by photopolymerization. Characterization of morphology, weight loss, water state of hydrogel, pH-sensitivity and cytotoxicity were investigated by scanning electron microscopy (SEM), thermal gravimetric analysis (TGA), X-ray diffraction (XRD), differential scanning calorimetry (DSC), Fourier transform infrared (FTIR), swelling test and methylthiazolydiphenyl-tetrazolium bromide (MTT) assay. The results indicated that the hydrogels were sensitive to pH of the medium, no cytotoxicity for L929 and SW1353, satisfactory for the composite to be used in bioapplications.

Abstract:The objectives of the study were as follows: (1) to develop two methods for the preparation of macroporous composite chitosan/hyaluronic acid (Ch/HA) hydrogels based on covalently cross-linked Ch and low molecular weight (Mw) HA (5 and 30 kDa); (2) to investigate some properties (swelling and in vitro degradation) and structures of the hydrogels; (3) to evaluate the hydrogels in vitro as potential biodegradable matrices for tissue engineering. Chitosan was cross-linked with either genipin (Gen) or glutaraldehyde (GA). Method 1 allowed the distribution of HA macromolecules within the hydrogel (bulk modification). In Method 2, hyaluronic acid formed a polyelectrolyte complex with Ch over the hydrogel surface (surface modification). By varying compositions of the Ch/HA hydrogels, highly porous interconnected structures (with mean pore sizes of 50–450 μm) were fabricated and studied using confocal laser scanning microscopy (CLSM). Mouse fibroblasts (L929) were cultured in the hydrogels for 7 days. Cell growth and proliferation within the hydrogel samples were studied via MTT-assay. The entrapment of low molecular weight HA was found to result in an enhancement of cell growth in the Ch/HA hydrogels compared to that in the Ch matrices. The Ch/HA hydrogels after bulk modification promoted better cell adhesion, growth and proliferation than the samples prepared by using Method 2 (surface modification).

Abstract:Background Tissue engineering, which involves the selection of scaffold materials, presents a new therapeutic strategy for damaged tissues or organs. Scaffold design based on blends of proteins and polysaccharides, as mimicry of the native extracellular matrix, has recently become a valuable strategy for tissue engineering. Objective This study aimed to construct composite hydrogels based on natural polymers for tissue engineering. Methods Composite hydrogels based on blends of gelatin with a polysaccharide component (chitosan or alginate) were produced and subsequently enzyme crosslinked. The other three hydrogels, chitosan hydrogel, sodium alginate hydrogel, and microbial transglutaminase-crosslinked gelatin (mTG/GA) hydrogel were also prepared. All hydrogels were evaluated for in vitro degradation property, swelling capacity, and mechanical property. Rat adipose-derived stromal stem cells (ADSCs) were isolated and seeded on (or embedded into) the above-mentioned hydrogels. The morphological features of ADSCs were observed and recorded. The effects of the hydrogels on ADSC survival and adhesion were investigated by immunofluorescence staining. Cell proliferation was tested by thiazolyl blue tetrazolium bromide (MTT) assay. Results Cell viability assay results showed that the five hydrogels are not cytotoxic. The mTG/GA and its composite hydrogels showed higher compressive moduli than the single-component chitosan and alginate hydrogels. MTT assay results showed that ADSCs proliferated better on the composite hydrogels than on the chitosan and alginate hydrogels. Light microscope observation and cell cytoskeleton staining showed that hydrogel strength had obvious effects on cell growth and adhesion. The ADSCs seeded on chitosan and alginate hydrogels plunged into the hydrogels and could not stretch out due to the low strength of the hydrogel, whereas cells seeded on composite hydrogels with higher elastic modulus, could spread out, and grew in size. Conclusion The gelatin-polysaccharide composite hydrogels could serve as attractive biomaterials for tissue engineering due to their easy preparation and favorable biophysical properties.

Abstract:Gelatin and chitosan (CS) are widely used natural biomaterials for tissue engineering scaffolds, but the poor mechanical properties of pure gelatin or CS hydrogels become a big obstacle that limits their use as scaffolds, especially in load-bearing tissues. This study provided a novel mechanism of forming interpenetrating network (IPN) of gelatin methacryloyl (GelMA) and CS hydrogels by covalent bonds and hydrophobic interactions through photocrosslinking and basification, respectively. By characterization of the compressive and tensile moduli, ultimate tensile stress and strain, it was found that semi-IPN and IPN structure can greatly enhance the mechanical properties of GelMA-CS hydrogels compared to the single network CS or GelMA. Moreover, the increase of either GelMA or CS concentration can strengthen the hydrogel network. Then, the swelling, enzymatic degradation, and morphology of GelMA-CS hydrogels were also systematically investigated. The excellent biocompatibility of GelMA-CS hydrogels was demonstrated by large spreading area of bone mesenchymal stem cells on hydrogel surfaces when CS concentration was <2% (w/v). According to this study, the multiple requirements of properties can be fulfilled by carefully selecting the GelMA and CS compositions for IPN hydrogels.

Abstract:In this study,acrylated hydroxypropyl chitosan(AHCS) with different substitution degree(SD) of acryloyl,i.e.,1.03%,3.55% and 5.21%,was prepared by chemical modification of chitosan(CS).Through free radical initiation reaction,AHCS and polyethylene glycol diacrylate(PEGDA) were crosslinked to form CS-PEG hydrogel.SEM indicated it was a porous scaffold with high connection rate.Our results suggested that the sol content(SC) and swelling ratio(SR) of the hydrogels decreased with increase of SD,and SR was higher in acid or alkaline conditions than in neutral condition.While the degradation rate(DR) of chitosan in the hydrogels also decreased with increase of SD.Cell experiment indicated that chitosan and polyethylene glycol crosslinked hydrogel had good biocompatibility.

Abstract:A new scheme was proposed for synthesizing poly(ethylene glycol)-g-chitosan (PEG-g-CS), where methoxy poly(ethylene glycol) iodide (MPEG-I) (Mn 2000) was used for N-substitution of triphenylmethyl chitosan (TPM-CS) in organic medium. The graft copolymers were obtained by subsequent removal of protecting groups with dichloroacetic acid. By varying PEG-I/TPM-CS feed ratio, the grafting levels (GL) of PEG can be adjusted. The chitosan derivatives were characterized by FTIR, 1H NMR, 13C NMR and DSC. All the copolymers were soluble in water over wide pH range. Furthermore, organosolubility of the hybrids in DMF and DMSO was also achieved when the DS value more than 24%. The lysozyme degradation rate of the copolymers in aqueous neutral medium decreased with the increase of GL value.
Nonionic polymer cross-linked chitosan hydrogel: preparation and bioevaluation.

Abstract:A straightforward method to prepare nonionic polymer (polyacrylamide, PAM) cross-linked chitosan hydrogel has been developed. The chitosan-polyacrylamide (CS-PAM) hydrogel could be quickly obtained by simply mixing of chitosan and polyacrylamide solutions under very benign condition (room temperature, <30s). The cytotoxicity and hemocompatibility of the CS-PAM hydrogel were subsequently investigated. Cells retained normal morphology even when the concentration of CS-PAM hydrogel in culture system was as high as 640gmL(-1), indicating the CS-PAM hydrogel has minimal cytotoxicity to A549 and HeLa cells. Meanwhile, no hemolysis was observed after incubating the CS-PAM hydrogel with red blood cells for 6h, further suggesting excellent biocompatibility of the hydrogel. Scanning electron microscopy images illustrated that the CS-PAM hydrogel can absorb red blood cells through the interaction between the protonated amine groups on chitosan and the negative charged residues on red blood cell membranes. Given its low cost, simple preparation, and excellent biocompatibility, this CS-PAM hydrogel might be a potential biomaterial for future healthcare and biomedical applications.
Synthesis of hydrogels from biomaterials and their potential application in tissue engineering

Abstract:In this study, a series of hydrogels were synthesized from chitosan(s) that was crosslinking with glutaraldehyde at different concentrations. Ascorbic acid in an acidic medium was used to facilitate non-covalent interactions. The chitosan(s) was obtained from shrimp cytoskeleton; while ascorbic acid was extracted from xoconostle juice. The hydrogel reaction was monitored by UV-vis spectroscopy (550 nm) to determine the reaction kinetics and reaction order at 60 °C. The hydrogels structures were characterized by NMR, FT-IR, HR-MS and SEM, while the degree of cross-linking was examined with TGA-DA. The extracellular matrices were obtained as stable hydrogels where reached maximum crosslinking was of 7 %, independent of glutaraldehyde quantity added. The rheological properties showed a behavior of weak gels and a dependence of crosslinking agent concentration on strength at different temperatures. The cytotoxicity assay showed that the gels had no adverse effects on cellular growth for all concentrations of glutaraldehyde.

Abstract:Introduction The potentialities of hyaluronic acid (HyA) and chitosan (CS) (Fig. 1) as biomaterials have been thoroughly investigated (1, 2). HyA is a mucopolysaccharide of the vertebrate extracellular tissue matrix. Its high viscosity in dilute solution collapses when subjected to a shearing stress; therefore, it can be injected through fine needles (3). Recent biomedical HyA applications included ophthalmic surgery, arthritis treatment, and scaffolds for wound healing, tissue engineering and the use as a component in implant materials (4). CS, obtained from partial chitin deacetylation, has gained increasing importance in the pharmaceutical field owing to its good biocompatibility, non-toxicity and biodegradability (5). It is also used in the food industry and in cosmetics. Having cationic characteristics, CS can interact with the anionic polymer HyA to form a composite material with unique properties (4, 6). Based on this we considered obtaining an injectable, slow-degradable biomaterial for damaged soft tissue regeneration consisting of CS/HyA (7). In particular, this study aimed to prepare and characterize porous CS beads, suitable to inglobing HyA chains, and to study their interactions with other molecules in solution.

Abstract:Hydrogels are fascinating biomaterials that can act as a support for cells, i.e., a scaffold, in which they can organize themselves spatially in a similar way to what occurs in vivo. Hydrogel use is therefore essential for the development of 3D systems and allows to recreate the cellular microenvironment in physiological and pathological conditions. This makes them ideal candidates for biological tissue analogues for application in the field of both tissue engineering and 3D in vitro models, as they have the ability to closely mimic the extracellular matrix (ECM) of a specific organ or tissue. Polysaccharide-based hydrogels, because of their remarkable biocompatibility related to their polymeric constituents, have the ability to interact beneficially with the cellular components. Although the growing interest in the use of polysaccharide-based hydrogels in the biomedical field is evidenced by a conspicuous number of reviews on the topic, none of them have focused on the combined use of two important polysaccharides, chitosan and pectin. Therefore, the present review will discuss the biomedical applications of polysaccharide-based hydrogels containing the two aforementioned natural polymers, chitosan and pectin, in the fields of tissue engineering and 3D in vitro modeling.

Abstract:Chitosan, a cationic polysaccharide, exhibits promising potential for tissue engineering applications. However, the poor mechanical properties and rapid biodegradation have been the major limitations for its applications. In this work, an effective strategy was proposed to optimize the mechanical performance and degradation rate of chitosan gel scaffolds by regulating the water content. Physical chitosan hydrogel (HG, with 93.57 % water) was prepared by temperature-controlled cross-linking, followed by dehydration to obtain xerogel (XG, with 2.84 % water) and rehydration to produce wet gel (WG, with 56.06 % water). During this process, changes of water content significantly influenced the water existence state, hydrogen bonding, and the chain entanglements of chitosan in the gel network. The mechanical compression results showed that the chitosan gel scaffolds exhibited tunable compressive strength (0.3128–139 MPa) and compressive modulus (0.2408–1094 MPa). XG could support weights exceeding 65,000 times its own mass while maintaining structural stability. Furthermore, in vitro and in vivo experiments demonstrated that XG and WG exhibited better biocompatibility and resistance to biodegradation compared with HG. Overall, this work contributes to the design and optimization of chitosan scaffolds without additional chemical crosslinkers, which has potential in tissue engineering and further clinical translation.

Abstract:Objective: The aim of this study was to prepare different hydrogels using chitosan and PVA polymers and to evaluate the cytotoxicity of hydrogels through cell culture studies. In addition, FTIR spectrum measurements were aimed to determine the specific chemical groups and bonds in the structures of hydrogels and materials. Method: In this study, different hydrogels were prepared and 3- (4,5-dimethyl-2-thiazolyl) -2,5-diphenyl tetrazolium bromide (MTT) test was performed to evaluate the toxicity of hydrogels on mouse fibroblast cell line (L-929). In addition, specific chemical groups of materials and hydrogels used in hydrogels were determined via FTIR spectrum study. Results: According to the MTT cytotoxicity results B1 (94.20±1.30 %) and D2 (82.45±1.05 %) formulations showed the greatest and lowest L-929 cell viability respectively. According to the results of the FTIR absorption spectrum, the aliphatic C = O tension band of rh-EGF was observed at 1634 cm-1. Conclusions: Results showed that chitosan-PVA hydrogels containing r-EGF types were observed to increase mouse fibroblast cell viability. In the FTIR spectrum results, spectral bands showing specific chemical groups of chitosan, PVA, PEGylation agents, rh-EGF and C2 formulation were obtained.
Thixotropic chitosan hydrogels for biomedical applications: Unravelling the effect of chitosan concentration on the mechanical behaviour

Abstract:Chitosan hydrogels are highly sought after in biomedical applications due to their biocompatibility, non-toxicity, and tunable mechanical properties. Nevertheless, achieving stable hydrogels with the required mechanical strength remains a notable obstacle. In this study, we prepared chitosan hydrogels with enhanced stability using minimal concentrations of glutaraldehyde to mitigate toxicity risks. The resulting hydrogel system is characterized by strong covalent bonds formed through imine and hydrogen bonding, facilitated by the aldehyde groups of glutaraldehyde. Rheological analyses reveal the viscoelastic properties of the hydrogels, showcasing stable gel-like structures with shear-thinning behavior, which are further enhanced with increasing chitosan concentration. Our observation demonstrates that the complex shear moduli of 0.5 - 1.5 w/v % chitosan hydrogel ranged between 3.08 kPa and 8.5 kPa, closely matching the shear moduli of the brain and connective tissues. The shear moduli obtained for the 2w/v% (10.2 kPa) and 2.5w/v% hydrogels (11.34 kPa) conform to the shear moduli of liver, fat, relaxed muscle, and breast gland tissue. Thixotropic studies reveal good structural recovery behaviour in the hydrogels. The swellability of 2w/v% and 2.5w/v% hydrogels (50-70%) suggests their suitability for bone tissue engineering, with moderate porosity required for bone growth (60-75%). Moreover, the yield stress values of 0.5 - 1.5 w/v % hydrogels satisfy the requirements for soft tissue engineering, while those of 2 and 2.5 w/v% match the requirements for bone tissue engineering. Our findings suggest that chitosan hydrogels of varying concentrations (0.5–2.5% w/v) hold promise for a range of biomedical applications, including tissue engineering for brain, nerve, soft, and bone tissues, and wound dressing. This research addresses the pressing need for stable chitosan hydrogels with tailored mechanical properties, potentially offering solutions to challenges in biomedical materials design.

Abstract:Fabricating robust and multi-functional hydrogels is of great importance and challenge. In this work, chitosan (CS) and polyvinyl alcohol (PVA) were used to design antiseptic, conductive and robust hydrogels by a two-step method. Chemical structures of gels, the degree of crystallinity, the state of water in hydrogels, as well as their microstructures were characterized via a combination of FT-IR, XRD, DSC and SEM. Segment lengths of cross-linking points were calculated from elastic rubber theory. Their mechanical properties were evaluated on the electronic testing machine. It was shown that the tensile strength and elongation at break of single PVA hydrogel were only 200 kPa and 135%, respectively, due to the heterogenous structure with pore sizes between 1.5 ~ 8.2 μm. By introducing CS into PVA matrix followed with soaking in a saturated NaCl solution, the network became homogeneous with a pore size of 0.5 ~ 1.1 μm. Moreover, free water changed to bond water, and frictions between polymer chains increased because of hydrophobic associations and entanglements of CS segments. As a result, the tensile stress and strain increased to 3800 kPa and 270%, respectively. The gel also exhibited antiseptic property, electrical conductivity and swelling-resistant properties. The strength after reaching swell equilibrium was 3400 kPa, much higher than most gels at swollen states. This gel might find applications in bionic cartilage, sensors, food preservation and wearable devices.

Abstract:The density and strength of chitosan hydrogel is improved by covalent crosslinking and physical crosslinking of poly(vinyl alcohol)(PVA),which forms an interpenetrating polymer network.The pH sensitive chitosan solution was neutralized to physiological pH by glycerol phosphate and remained stable at room temperature while allowing gel formation upon heating to 37 ℃.The structure and stress of the hydrogel was strengthened by PVA and covalently crosslinked to form the hydrogel matrix.Fourier transform infrared spectroscopy test shows that the chitosan is covalently crosslinked by the formation of Schiff's base.With the crosslinked PVA and the covalent bonds,the thermosensitive hydrogel has intensified structure and may be useful for in situ gelling implants.

Abstract:Biodegradable hydrogels based on synthetic PHEA and IPN type composite hydrogel with chitosan, a natural carbohydrate polymer, were prepared by crosslinking using glutaraldehyde in aqueous mixture. Relatively strong hydrogels were formed in a shaped mold. Preparation of crosslinked gels and their swelling behavior depending on the composition, medium, and pH of solution are investigated. The porous morphology of scaffolds, obtained by freeze-drying method, was observed by scanning electron microscopy. Also preliminary results on hydrolytic degradation and cytotoxicity test of the hydrogel are included.

Abstract:At present, the ocean as a treasure house of human development resources has attracted widespread attention. Chitin is the most abundant biomass resource in the ocean, and it is also a natural macromolecule second only to cellulose in nature. Its deacetylated product, chitosan, is not only easy to modify physically and chemically, but can be processed into various forms and has excellent properties. Biocompatibility, biodegradability, antibacterial, tissue adhesion and other biological activities. Derivatization reactions are often used to improve the properties of chitosan and give it new structures and functions. In this paper, a series of chitosan ether derivatives were synthesized using alkali/urea aqueous solution as the solvent and homogeneous etherification reaction medium of chitosan, and their structures and properties were characterized. Then, chitosan and quaternized chitosan are dissolved in the acrylic acid monomer aqueous solution, and the chitosan/polyacrylic acid and quaternized high-strength polyelectrolyte composite hydrogel (PEC) are obtained by in-situ polymerization. Finally, the above-mentioned composite hydrogel is treated with Ag+ to obtain the ultra-high strength of the dual interaction of static electricity and coordination bonds, and at the same time a physical cross-linked hydrogel (DPC) with anti-fatigue. The chitosan-based hydrogel with excellent mechanical properties has good application prospects in articular cartilage, wound dressings, bionic stents and the like.

Abstract:We developed and characterized a new hydrogel system based on the physical and chemical interactions of pectin partially modified with thiol groups and chitosan modified with acrylate end groups. Gelation occurred at high pectin thiol ratios, indicating that a low acrylated chitosan concentration in the hydrogel had a profound effect on the cross-linking. Turbidity, Fourier transform infrared spectroscopy, and free thiol determination analyses were performed to determine the relationships of the different bonds inside the gel. At low pH values below the pKa of chitosan, more electrostatic interactions were formed between opposite charges, but at high pH values, the Michael-type addition reaction between acrylate and thiol took place, creating harder hydrogels. Swelling experiments and Young’s modulus measurements were performed to study the structure and properties of the resultant hydrogels. The nanostructure was examined using small-angle X-ray scattering. The texture profile analysis showed a unique property of hydrogel adhesiveness. By implementing changes in the preparation procedure, we controlled the hydrogel properties. This hybrid hydrogel system can be a good candidate for a wide range of biomedical applications, such as a mucosal biomimetic surface for mucoadhesive testing.
