Abstract:Most existing studies have focused on identifying the origin of species with protected geographical indications while neglecting to determine the proximate geographical origin of different species. In this study, we investigated the feasibility of using near- and mid-infrared spectroscopy to identify the origin of 156 Polygonatum kingianum samples from six regions in Yunnan, China. In this work, spectral images of different modes reveal more information about the P. kingianum. Comparing the performance of traditional machine learning models according to single spectrum and data fusion, the middle-level data fusion-principal component model has the best performance, and its sensitivity, specificity, and accuracy are all 1, and the model has the least number of variables. The residual convolutional neural network (ResNet) model constructed in the 1050-850 cm-1 band confirms that fewer variables are beneficial in improving the accuracy of the model. In conclusion, this study verifies the feasibility of the proposed strategy and establishes a practical model to determine the source of P. kingianum.

Abstract:Polygonati rhizoma (PR) is an important and widely used product in Chinese traditional medicine and edible goods. The time-consuming nature of breaking dormancy in both rhizomes and seeds means that improving variety selection is limited to collection, identification, and selection of germplasms. In this study, we used two DNA-based molecular marker techniques—inter simple sequence repeat (ISSR) and start codon targeted (SCoT)—to assess the genetic diversity and population structure among PR source plants collected from 47 different regions and belonging to 12 populations (P1–P12). For molecular markers analysis, 15 ISSR and 10 SCoT markers were tested. Total number of 159 fragments (150–4000 bp) were amplified based on ISSR analysis with a range from 6 to 17 bands, 153 of them were polymorphic, ranging from 97.27 to 100%. For SCoT analysis, 164 polymorphic bands (150–5000 bp) were observed, varying from 14 to 19 bands for each primer. Nei’s genetic diversity analysis showed that the highest value was found in P11 for ISSR and P4 for SCoT markers. The highest Nei’s genetic diversity value was observed in P5 for combined markers and the low in P2. Nei’s dendrogram constructed with combined markers indicated a 75–89% of genetic similarity coefficient among populations. Population structure analysis revealed an optimum number of three groups, the same as their geographical distribution. This knowledge on PR genetic diversity can be used in future breeding programs, genetic improvement, product enhancement, and germplasms conservation.

Abstract:In order to quickly determine the processing degree and quality of Polygonati Rhizome (PR), the rapid qualitative and quantitative model for the whole steaming process was established in this study. Firstly, the contents and the chromaticity value of polysaccharide, saponin, 5-hydroxymethyl furfural (5-HMF), 5-hydroxymaltol, polygonatine A, fructose, glucose, sucrose, and the chromaticity value in 98 batches of PR during the whole processing were determined. The chromaticity value was used to establish the qualitative discrimination model of the whole steaming process by partial least squares-discriminant analysis (PLS-DA). Then, the near-infrared (NIR) spectra of all samples were collected to establish the NIR qualitative discriminant model. The samples of four different processing stages, raw products, less processed products, finished processed products, and over processed products, were well distinguished in two qualitative models, and the model validation results were good. Finally, the chromaticity value, the content, and NIR spectra were imported into TQ Analyst software. After screening the pretreatment methods, the optimal pretreatment method combined with partial least squares (PLS) was selected to establish the NIR quantitative model of 11 indicators. The correlation coefficient of models was above 0.9, indicating that the model had high prediction accuracy. Through the establishment of the qualitative discriminant model, the different processing degrees of PR could be accurately distinguished, and the NIR quantitative prediction model could accurately predict the content and the chromaticity value, which provided a scientific basis for the quality identification of PR in the whole process of processing.

Abstract:Introduction Medicinal Polygonatum species is a widely used traditional Chinese medicine with high nutritional value, known for its anti‐fatigue properties, enhancement of immunity, delays aging, improves sleep, and other health benefits. However, the efficacy of different species varies, making the quality control of medicinal Polygonatum species increasingly important. Polysaccharides are important in medicinal Polygonatum species because of their potential functional properties, such as antioxidation, hypoglycemia, protection of intestinal health, and minimal toxicological effects on human health, as well as high polysaccharide levels. Objective This study developed a qualitative medicinal Polygonatum species model and a polysaccharides predictive model based on attenuated total reflection Fourier transform infrared spectroscopy (ATR‐FTIR) combined with a multivariate analysis approach. Materials and Methods ATR‐FTIR spectral information of 334 medicinal Polygonatum species samples was collected and the spectral information of different modes was analyzed. The ATR‐FTIR spectral differences of three medicinal Polygonatum species were studied by multivariate analysis combined with four spectral preprocessing and three variable selection methods. For the prediction of polysaccharides in Polygonatum kingianum Collett & Hemsl. (PK), we initially determined the actual content of 110 PK polysaccharide samples using the anthrone‐sulfuric acid method, then established partial least squares regression (PLSR) and kernel PLSR models in conjunction with attenuated total reflectance Fourier transform infrared (ATR‐FTIR) spectroscopy. Results In the visualization analysis, the orthogonal partial least squares‐discriminant analysis (OPLS‐DA) model based on second‐order derivative (SD) preprocessing was most suitable for medicinal Polygonatum species species binary classification, spectral differences between Polygonatum cyrtonema Hua (PC) and other species are evident; in the hard modeling, SD preprocessing improves the accuracy of non‐deep learning models for the classification of three medicinal Polygonatum species. In contrast, residual neural network (ResNet) models were the best choice for species identification without preprocessing and variable selection. In addition, the partial least squares regression (PLSR) model and Kernel‐PLSR model can quickly predict PK polysaccharides content, among them, the Kernel‐PLSR model with SD pretreatment has the best prediction performance, residual prediction deviation (RPD) = 7.2870, Rp = 0.9905. Conclusion In this study, we employed ATR‐FTIR spectroscopy and various treatments to discern different medicinal Polygonatum species. We also evaluated the effects of preprocessing methods and variable selection on the prediction of PK polysaccharides by PLSR and Kernel‐PLSR models. Among them, the ResNet model can achieve 100% correct classification of medicinal Polygonatum species without complex spectral preprocessing. Furthermore, the Kernel‐PLSR model based on SD‐ATR‐FTIR spectra had the best performance in polysaccharides prediction. In summary, by integrating ATR‐FTIR spectroscopy with multivariate analysis, this research accomplished the classification of medicinal Polygonatum species and the prediction of polysaccharides. The methodology offers the benefits of speed, environmental sustainability, and precision, highlighting its significant potential for practical applications. In future research, on the one hand, it can be further investigated using a portable infrared spectrometer, and on the other hand, infrared spectroscopy can also be applied to the prediction of other chemical components of medicinal Polygonatum species.

Abstract:Rhizomes of the Polygonatum species are well-known in traditional Chinese medicine. The 2020 edition of Chinese Pharmacopoeia includes three different species that possess different pharmacological effects. Due to the lack of standardized discriminant compounds there has often been inadvertently incorrect prescriptions given for these medicines, resulting in serious consequences. Therefore, it is critical to accurately distinguish these herbal Polygonatum species. For this study, UPLC-Q-TOF-MS/MS based metabolomics was employed for the first time to discriminate between three Polygonatum species. Partial least squares discriminant analysis (PLS-DA) models were utilized to select the potential candidate discriminant compounds, after which MS/MS fragmentation patterns were used to identify them. Meanwhile, metabolic correlations were identified using the R language package corrplot, and the distribution of various metabolites was analyzed by box plot and the Z-score graph. As a result, we found that adenosine, sucrose, and pyroglutamic acid were suitable for the identification of different Polygonatum species. In conclusion, this study articulates how various herbal Polygonatum species might be more accurately and efficiently distinguished.

Abstract:Due to the world-wide concern relating to herb quality and safety, there is a momentum to authenticate the geographical origin of herb with multi-platform techniques. This study attempted to assess multi-platform information as a practical strategy for the geographical traceability of the fruits of Amomum tsao-ko. To this aim, one hundred and eighty dried fruits of A. tsao-ko from five geographical regions were analyzed by near infrared (NIR) and ultraviolet visible (UV-Vis) spectroscopy. On this basis, two variable dimension reduction strategies, including principal component analysis (PCA) and sequential and orthogonalized partial-least squares (SO-PLS), and two variables selection strategies, including variable importance in projection (VIP) and sequential and orthogonalized covariance selection (SO-CovSel), were performed to extract the feature information in the two blocks. Partial least squares discriminant analysis (PLS-DA) classification algorithm combined with fused matrices was used to identify the geographical origins. The results of PLS-DA models indicated that SO-PLS and SO-CovSel, taking advantage of the sequential modeling coupled to orthogonalization, could not only identify the common information presented in the two blocks but also provide more concise methods without any loss of classification ability, which could be employed in authenticating the geographical regions of the fruits of A. tsao-ko, effectively.

Abstract:Panax notoginseng is an important functional health product, and has been used worldwide because of a wide range of pharmacological activities, of which the taproot is the main edible or medicinal part. However, the technologies for origin discrimination still need to be further studied. In this study, an ICP-MS/MS method for the accurate determination of 49 elements was established, whereby the instrumental detection limits (LODs) were between 0.0003 and 7.716 mg/kg, whereas the quantification limits (LOQs) were between 0.0011 and 25.7202 mg/kg, recovery of the method was in the range of 85.82% to 104.98%, and the relative standard deviations (RSDs) were lower than 10%. Based on the content of multi-element in P. notoginseng (total of 89 mixed samples), the discriminant models of origins and cultivation models were accurately determined by the neural networks (prediction accuracy was 0.9259 and area under ROC curve was 0.9750) and the support vector machine algorithm (both 1.0000), respectively. The discriminant models established in this study could be used to support transparency and traceability of supply chains of P. notoginseng and thus avoid the fraud of geographic identification.

Abstract:Polygonatum species have great potential in fighting chronic and hidden hunger. In this study, five Polygonatum species collected from different populations were cultivated in a common garden for 4 years. The species mainly differed in yield, saponin and polysaccharide contents, stem diameter, leaf width, inflorescence length, and floret inflorescence length. P. cyrtonema (PC) provides high-quality yield when planted in Zhejiang, with output as high as 7.5 tons per hectare and a promising breeding potential. Moreover, stem diameter can be used as an indicator of the harvest in the screening of varieties. In addition, the formation of plant genetic traits from different provenances is affected by the climatic factors of the origin. Furthermore, near-infrared spectroscopy combined with chemometrics for polysaccharide and saponin quantitation provides a rapid assessment of PC quality. Our findings provide a scientific basis for the development and sustainable utilization of PC as a high-yielding and high-quality forest crop.

Abstract:Rhubarb, a widely used traditional Chinese medicine (TCM), is primarily used for purging in practice. It is derived from the dried roots and rhizomes of R. tanguticum Maxim. ex Balf. (RT), Rheum officinale Baill. (RO) and R. palmatum L. (RP). To date, although the three varieties of rhubarb have been used as the same medicine in clinical, studies have found that they have different chemical compositions and pharmacological effects. To ensure the stability of rhubarb for clinical use, a simple and effective method should be built to compare and discriminate three varieties of rhubarb. Here, ultra-performance liquid chromatography-diode array detection (UPLC-DAD) fingerprints combined with chemometric methods were developed to evaluate and discriminate 29 batches of rhubarb. Similarity evaluation, hierarchical cluster analysis (HCA) and principal component analysis (PCA) showed that the chemical constituents of the three varieties of rhubarb were significantly different, and the three varieties could be effectively distinguished. Finally, all the 14 common peaks were identified by ultra-performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS). In this research, the developed UPLC fingerprints offer a simple, reliable and specific approach for distinguishing different varieties of rhubarb. This research aims to promote the scientific and appropriate clinical application of rhubarb from three varieties.

Development of SSR markers for genetic diversity analysis and species identification in Polygonatum odoratum (Mill.) Druce based on transcriptome sequences

Abstract:Polygonatum odoratum (Mill.) Druce is a well-known traditional Chinese herb belonging to the Polygonatum. However, the understanding of the genetic diversity of this species at the molecular level is limited due to the lack of transcriptomic and genomic information. In this study, 37,387 unigenes were assembled based on the transcriptome sequencing of the rhizome of Polygonatum odoratum (Mill.) Druce., and 11,021 single- sequence repeats (SSR) motifs, mainly consisting of single-nucleotide repeats (44.44%), dinucleotides (31.06%), and trinucleotides (22.59%), were identified. Based on these SSR motifs, 9,987 primer pairs of SSR markers were designed and 68 SSR markers were randomly selected for verification, of which 21 SSR markers showed polymorphisms among the 24 Polygonatum odoratum germplasms. Ninety-four alleles were detected: the observed alleles ranged from 2 to 11, the effective alleles varied from 1.086 8 to 4.916 8, the Shannon diversity index was 0.173 2~1.749 7, and the polymorphism information content PIC ranged from 0.076 7 to 0.803 9. Based on our analysis of genetic diversity (SSR genotypes) and population structure, we divided the 24 germplasm resources into two groups, indicating that the germplasm with similar geographical origins can be grouped together. In addition, the primers 'YZ14' and 'YZ47' could effectively distinguished the related species: Polygonatum kingianum Coll.et Hemsl., Polygonatum sibiricum Red., Polygonatum cyrtonema Hua, Polygonatum zanlanscianense Pamp. and Polygonatum odoratum (Mill.) Druce. This is the first study in which a dataset of expressed sequence tag (EST)-SSR markers is constructed for the Polygonatum odoratum (Mill.) Druce, and these newly developed EST-SSR markers provided a very efficient tool for genetic relationship analysis, species identification and marker-assisted selection breeding of Polygonatum odoratum (Mill.) Druce.
Identification of Radix Bupleuri From Different Geographic Origins Using Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry and Support Vector Machine Algorithm

Abstract:Background: The geographic origin of Radix bupleuri is an important factor affecting its efficacy, which needs to be effectively identified. Objective: The goal is to enrich and develop the intelligent recognition technology applicable to the identification of the origin of traditional Chinese medicine. Method: This article establishes an identification method of Radix bupleuri geographic origin based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and support vector machine (SVM) algorithm. The Euclidean distance method is used to measure the similarity between Radix bupleuri samples, and the quality control chart method is applied to quantitatively describe their quality fluctuation. Results: It is found that the samples from the same origin are relatively similar and mainly fluctuate within the control limit, but the fluctuation range is large, and it is impossible to distinguish the samples from different origins. The SVM algorithm can effectively eliminate the impact of intensity fluctuations and huge data dimensions by combining the normalization of MALDI-TOF MS data and the dimensionality reduction of principal components, and finally achieve efficient identification of the origin of Radix bupleuri, with an average recognition rate of 98.5%. Conclusions: This newly established approach for identification of the geographic origin of Radix bupleuri has been realized, and it has the advantages of objectivity and intelligence, which can be used as a reference for other medical and food-related research. Highlights: A new intelligent recognition method of medicinal material origin based on MALDI-TOF MS and SVM has been established.

Abstract:Atractylodis rhizoma is a common bulk medicinal material with multiple species. Although different varieties of atractylodis rhizoma exhibit variations in their chemical constituents and pharmacological activities, they have not been adequately distinguished due to their similar morphological features. Hence, the purpose of this research is to analyze and characterize the volatile organic compounds (VOCs) in samples of atractylodis rhizoma using multiple techniques and to identify the key differential VOCs among different varieties of atractylodis rhizoma for effective discrimination. The identification of VOCs was carried out using headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) and headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS), resulting in the identification of 60 and 53 VOCs, respectively. The orthogonal partial least squares discriminant analysis (OPLS-DA) model was employed to screen potential biomarkers and based on the variable importance in projection (VIP ≥ 1.2), 24 VOCs were identified as critical differential compounds. Random forest (RF), K-nearest neighbor (KNN) and back propagation neural network based on genetic algorithm (GA-BPNN) models based on potential volatile markers realized the greater than 90 % discriminant accuracies, which indicates that the obtained key differential VOCs are reliable. At the same time, the aroma characteristics of atractylodis rhizoma were also analyzed by ultra-fast gas chromatography electronic nose (Ultra-fast GC E-nose). This study indicated that the integration of HS-SPME-GC-MS, HS-GC-IMS and ultra-fast GC E-nose with chemometrics can comprehensively reflect the differences of VOCs in atractylodis rhizoma samples from different varieties, which will be a prospective tool for variety discrimination of atractylodis rhizoma.
Rapid Quality Assessment of Polygoni Multiflori Radix Based on Near-Infrared Spectroscopy

Abstract:The precise and prompt determination of quality control indicators such as moisture, stilbene glycosides, and anthraquinone glycosides is crucial in assessing the quality of Polygoni Multiflori Radix. Near-infrared spectroscopy is a nondestructive analytical technique that offers a more desirable approach than traditional methods for assessing content levels. In this study, various spectral preprocessing techniques were used to preprocess the raw spectral data. The spectral data were correlated with the determination of three-component contents using the partial least squares regression (PLSR) method. Then different algorithms, such as competitive adaptive weighted sampling (CARS), Monte Carlo uninformative variable elimination (MCUVE), and random frog hopping (RF), were used for model simplification and feature selection. The data suggest that the first-order deconvolution derivative (1st Dev.) processing of the spectral data is superior to other methods in all three model evaluation metrics. The PLSR model for moisture, stilbene glycosides, and anthraquinone glycosides produced the calibration coefficient of determination (R2C) of 0.82, 0.52, and 0.58, the root mean square error of cross validation (RMSECV) of 0.91%, 0.77%, and 0.69%, the prediction coefficient of determination (R2P) of 0.72, 0.28, and 0.54, the root mean square error of prediction (RMSEP) of 0.65%, 0.81%, and 0.75%, and relative percentage differences (RPDs) of 1.7, 1.0, and 0.8. After optimizing the model using CARS, R2C increased by 0.15%, 0.41%, and 0.34%, RMSECV decreased by 0.53%, 0.32%, and 0.24%, R2P increased by 0.21%, 0.63%, and 0.35%, RMSEP decreased by 0.36%, 0.41%, and 0.31%, and RPD increased by 1.1, 0.9, and 0.6, significantly improving the predictive capacity of the model. This research provides a feasible method for rapid compliance testing of Polygoni Multiflori Radix. To further improve the model's performance and applicability, it is necessary to continuously expand the sample set with different varieties and locations for wide variation.

Combing machine learning and elemental profiling for geographical authentication of Chinese Geographical Indication (GI) rice

Abstract:Abstract Identification of geographical origin is of great importance for protecting the authenticity of valuable agri-food products with designated origins. In this study, a robust and accurate analytical method that could authenticate the geographical origin of Geographical Indication (GI) products was developed. The method was based on elemental profiling using inductively coupled plasma mass spectrometry (ICP-MS) in combination with machine learning techniques for model building and feature selection. The method successfully predicted and classified six varieties of Chinese GI rice. The elemental profiles of 131 rice samples were determined, and two machine learning algorithms were implemented, support vector machines (SVM) and random forest (RF), together with the feature selection algorithm Relief. Prediction accuracy of 100% was achieved by both Relief-SVM and Relief-RF models, using only four elements (Al, B, Rb, and Na). The methodology and knowledge from this study could be used to develop reliable methods for tracing geographical origins and controlling fraudulent labeling of diverse high-value agri-food products.

Rapid discriminant analysis for the origin of specialty yam based on multispectral data fusion strategies

Abstract:In order to achieve rapid and effective identification of Hebei yam, a qualitative discrimination model was constructed based on near infrared (NIR), mid infrared (MIR), and microscopic Raman spectra in combination with individual spectra and multispectral data fusion strategies. The results showed that the gray wolf optimizer-support vector machine (GWO-SVM) model constructed by mid-level fusion using the three feature spectra performed the best in distinguishing the geographic origin of the yam, with a prediction accuracy of 100.00% in both the training set and the test set, and an F1 score of 1.00. The results indicated that due to spectral complementarity, NIR, MIR and Raman combined with feature-level fusion can be used as a powerful, non-destructive, fast and feasible tool for geographic origin classification and brand protection of Hebei yam. This work is expected to be a potential method for origin identification analysis and quality monitoring in the food and pharmaceutical industries.

Abstract:Polygonati rhizoma ( Huangjing in Chinese) is a traditional and classic dual-purpose material used in food and medicine. Herbalists in China and Japan have noticed several different rhizome types in Huangjing with different qualities. Rhizome of Polygonatum cyrtonema Hua and P. sibiricum Red. is divided into five types: “ Jitou -type” Polygonati rhizoma (JTPR), atypical “ Jitou -type” Polygonati rhizoma (AJTPR), “ Jiang -type” Polygonati rhizoma (JPR), “Cylinder-type” Polygonati rhizoma (CPR), and “ Baiji -type” Polygonati rhizoma (BJPR). This study observed the microstructure and histochemical localization of polysaccharides, saponins, and proteins in Huangjing . Nutritional and medicinal component data and antioxidant capacity (DPPH and ABTS) were analyzed to evaluate the quality of different types of Huangjing . The results showed that the comprehensive quality of the rhizomes, BJPR and JTPR, was better, regardless of their nutritional or medicinal values. Altogether, these results could recommend future breeding efforts to produce Huangjing with improved nutritional and medicinal qualities.

Fuzzy identification of bioactive components for different efficacies of rhubarb by the back propagation neural network association analysis of UPLC-Q-TOF/MSE and integrated effects

Abstract:Background: Rhei Radix et Rhizoma (rhubarb), as one of the typical representatives of multi-effect traditional Chinese medicines (TCMs), has been utilized in the treatment of various diseases due to its multicomponent nature. However, there are few systematic investigations for the corresponding effect of individual components in rhubarb. Hence, we aimed to develop a novel strategy to fuzzily identify bioactive components for different efficacies of rhubarb by the back propagation (BP) neural network association analysis of ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry for every data (UPLC-Q-TOF/MSE) and integrated effects. Methods: Through applying the fuzzy chemical identification, most components of rhubarb were classified into different chemical groups. Meanwhile the integration effect values of different efficacies can be determined by animal experiment evaluation and multi-attribute comprehensive indexes. Then the BP neural network was employed for association analysis of components and different efficacies by correlating the component contents determined from UPLC-Q-TOF/MSE profiling and the integration effect values. Finally, the effect contribution of one type of components may be totaled to demonstrate the universal and individual characters for different efficacies of rhubarb. Results: It suggested that combined anthraquinones, flavanols and their polymers may be the universal character to the multi-functional properties of rhubarb. Other components contributed to the individuality of rhubarb efficacies, including stilbene glycosides, anthranones and their dimers, free anthraquinones, chromones, gallic acid and gallotannins, butyrylbenzenes and their glycosides. Conclusions: Our findings demonstrated that the bioactive components for different efficacies of rhubarb were not exactly the same and can be systematically differentiated by the network-oriented strategy. These efforts will advance our knowledge and understanding of the bioactive components in rhubarb and provide scientific evidence to support the expansion of its use in clinical applications and the further development of some products based on this medicinal herb.
Application of Convolutional Neural Network-Based Feature Extraction and Data Fusion for Geographical Origin Identification of Radix Astragali by Visible/Short-Wave Near-Infrared and Near Infrared Hyperspectral Imaging

Abstract:Radix Astragali is a prized traditional Chinese functional food that is used for both medicine and food purposes, with various benefits such as immunomodulation, anti-tumor, and anti-oxidation. The geographical origin of Radix Astragali has a significant impact on its quality attributes. Determining the geographical origins of Radix Astragali is essential for quality evaluation. Hyperspectral imaging covering the visible/short-wave near-infrared range (Vis-NIR, 380–1030 nm) and near-infrared range (NIR, 874–1734 nm) were applied to identify Radix Astragali from five different geographical origins. Principal component analysis (PCA) was utilized to form score images to achieve preliminary qualitative identification. PCA and convolutional neural network (CNN) were used for feature extraction. Measurement-level fusion and feature-level fusion were performed on the original spectra at different spectral ranges and the corresponding features. Support vector machine (SVM), logistic regression (LR), and CNN models based on full wavelengths, extracted features, and fusion datasets were established with excellent results; all the models obtained an accuracy of over 98% for different datasets. The results illustrate that hyperspectral imaging combined with CNN and fusion strategy could be an effective method for origin identification of Radix Astragali.

Abstract:Sixty peony root training samples of the same age were collected from various regions in Korea and China, and their genetic diversity was investigated for 23 chloroplast intergenic space regions. All samples were genetically indistinguishable, indicating that the DNA-based techniques employed were not appropriate for determining the samples' regions of origin. In contrast, (1)H-nuclear magnetic resonance ((1)H-NMR) spectroscopy-based metabolomics coupled with multivariate statistical analysis revealed a clear difference between the metabolic profiles of the Korean and Chinese samples. Orthogonal projections on the latent structure-discrimination analysis allowed the identification of potential metabolite markers, including γ-aminobutyric acid, arginine, alanine, paeoniflorin, and albiflorin, that could be useful for classifying the samples' regions of origin. The validity of the discrimination model was tested using the response permutation test and blind prediction test for internal and external validations, respectively. Metabolomic data of 21 blended samples consisting of Korean and Chinese samples mixed at various proportions were also acquired by (1)H-NMR analysis. After data preprocessing which was designed to eliminate uncontrolled deviations in the spectral data between the testing and training sets, a new statistical procedure for estimating the mixing proportions of blended samples was established using the constrained least squares method for the first time. The predictive procedure exhibited relatively good predictability (adjusted R (2) = 0.7669), and thus has the potential to be used in the quality control of peony root by providing correct indications for a sample's geographical origins.

Abstract:Introduction: Identifying the geographical origin of Gastrodia elata Blume contributes to the scientific and rational utilization of medicinal materials. In this study, infrared spectroscopy was combined with machine learning algorithms to distinguish the origin of G. elata BI. Objective: Realization of rapid and accurate identification of the origin of G. elata BI. Materials and methods: Attenuated total reflection Fourier transform infrared (ATR-FTIR) spectra and Fourier transform near-infrared (FT-NIR) spectra were collected for 306 samples of G. elata BI. Samples: Firstly, a support vector machine (SVM) model was established based on the single-spectrum and the full-spectrum fusion data. To investigate whether feature-level fusion strategy can enhance the model's performance, the sequential and orthogonalized partial least squares discriminant analysis (SO-PLS-DA) model was established to extract and combine two types of spectral features. Next, six algorithms were employed to extract feature variables, SVM model was established based on the feature-level fusion data. To avoid complicated preprocessing and feature extraction processes, a residual convolutional neural network (ResNet) model was established after converting the raw spectral data into spectral images. Results: The accuracy of the feature-level fusion model is better as compared to the single-spectrum model and the fusion model with full-spectrum, and SO-PLS-DA is simpler than feature-level fusion based on the SVM model. The ResNet model performs well in classification but requires more data to enhance its generalization capability and training effectiveness. Conclusion: Sequential and orthogonalized data fusion approaches and ResNet models are powerful solutions for identifying the geographic origin of G. elata BI.
