Abstract:As efficient drug carriers, stimuli-responsive mesoporous silica nanoparticles are at the forefront of research on drug delivery systems. An acid-responsive system based on silyl ether has been applied to deliver a hybrid prodrug. Thiol-ene click chemistry has been successfully utilized for tethering this prodrug to mesoporous silica nanoparticles. Here, by altering the steric bulk of the substituent on the silicon atom, the release rate of a model drug, camptothecin, was controlled. The synthesized drug delivery system was investigated by analytical methods to confirm the functionalization and conjugation of the mesoporous silica nanoparticles. Herein, trimethyl silyl ether and triethyl silyl ether were selected to regulate the release rate. Under normal plasma conditions (pH 7.4), both types of camptothecin-loaded mesoporous silica nanoparticles (i.e., MSN-Me-CPT and MSN-Et-CPT) did not release the model drug. However, under in vitro acidic conditions (pH 4.0), based on a comparison of the release rates, camptothecin was released from MSN-Me-CPT more rapidly than from MSN-Et-CPT. To determine the biocompatibility of the modified mesoporous silica nanoparticles and the in vivo camptothecin uptake behavior, MTT assays with cancer cells and confocal microscopy observations were conducted, with positive results. These functionalized nanoparticles could be useful in clinical treatments requiring controlled drug release.
Synthesis of pH-Responsive Biodegradable Mesoporous Silica-Calcium Phosphate Hybrid Nanoparticles as a High Potential Drug Carrier.

Abstract:Biodegradability is one of the most critical issues for silica-based nanodrug delivery systems because they are crucial prerequisites for the successful translation in clinics. In this work, a novel mesoporous silica-calcium phosphate (MS-CAP) hybrid nanocarrier with a fast pH-responsive biodegradation rate was developed by a one-step method, where CAP precursors (Ca2+ and PO43-) were incorporated into silica matrix during the growth process. The morphology and structure of MS-CAP were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), N2 adsorption-desorption isotherms, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. Furthermore, the drug loading and the release behavior of MS-CAP have been tested. TEM and inductively coupled plasma-optical emission spectrometry results indicated that the pH-responsive biodegradation of MS-CAP was so fast that could be almost finished within 24 h owing to the easy dissolution of CAP embedded in the particle and the escape of Ca2+ from the structure of Si-O-Ca in acid environment. The MS-CAP exhibited a high doxorubicin (DOX) entrapment efficiency (EE) of 97.79%, which was about fourfold higher compared with that of pure mesoporous silica nanoparticles, and our density functional theory calculational results suggested that the higher drug EE of MS-CAP would originate from the strong interaction between calcium in the particle and carboxylate group of DOX. The loaded DOX was effectively released, with a cumulative release as high as 98.06% within 48 h at pH 4.5 in buffer solution, owing to the rapid degradation of MS-CAP. The obtained results indicated that the as-synthesized MS-CAP could act as a promising drug delivery system and would have a hopeful prospect in the clinical translation.

pH-sensitive CAP/SiO2 composite for efficient co-delivery of doxorubicin and siRNA to overcome multiple drug resistance.

Abstract:Long-term administration of chemotherapeutic agents often leads to multiple drug resistance (MDR), which greatly impairs the treatment outcome. To overcome this problem, a biodegradable nanocarrier based on an acid-sensitive calcium phosphate/silica dioxide (CAP/SiO2) composite was constructed for the codelivery of drug and siRNA. Anticancer drug doxorubicin (DOX) was encapsulated into the composite scaffold by interacting with the exposed Ca2+ of CAP/SiO2 to achieve high drug loading (180 μg mg-1). With further decoration of siRNA, the nanocarrier was applied to enhance the therapeutic efficacy by silencing MDR-relevant genes (P-gp) of DOX-resistance K562/ADR cancer cells. Benefiting from the intrinsic acid degradability of CAP/SiO2, the nanocomposite demonstrated pH-responsive release behavior, favoring drug/siRNA release within acidic endo-/lysosomes. Consequently, due to the drug and gene effects, this biodegradable nanomedicine demonstrated enhanced therapeutic efficiency, providing a novel strategy for cancer therapy.

Abstract:Prodrug and drug delivery systems are two effective strategies for improving the selectivity of chemotherapeutics. Molecularly imprinted polymers (MIPs) have emerged as promising carriers in targeted drug delivery for cancer treatment, but they have not yet been integrated with the prodrug strategy. Reported here is an MIP-based smart prodrug delivery system for specific targeting, prolonged retention time, and tumor microenvironment-triggered release. 5'-Deoxy-5-fluorocytidine (DFCR) and sialic acid (SA) were used as a prodrug and a marker for tumor targeting, respectively. Their co-imprinted nanoparticles were prepared as a smart carrier. Prodrug-loaded MIP specifically and sustainably accumulated at the tumor site and then gradually released. Unlike conventional prodrug designs, which often require in-liver bioconversion, this MIP-based prodrug delivery is liver-independent but tumor-dependent. Thus, this study opens new access to the development of smart prodrug delivery nanoplatforms.

Abstract:The safe and efficient delivery of drugs is regarded as a fundamental strategy for enhancing the low effectiveness of cancer treatments. Herein, a simple preparation approach for a multifunctional drug delivery system was reported. The novel structure of spiropyran (SP), 1-(6 ́-bromo-hexyl)−3,3-dimethyl indoline-6 ́ -methoxy benzopyran, was used to modify sodium carboxymethyl cellulose (NaCMC) for controlling drug release. SP has a heightened sensitivity to variations in pH, with a linear response range of .2–5.5. The elevated drug loading (31.68 %) can be attributed to the hydrophobic interaction of SP and the adsorption of NaCMC. Porous materials composed of zinc ions and dimethylimidazole (ZIF-8) as an embedded shell, which serves as a barrier that effectively reduces drug leakage from 20 % to 12 %, thus mitigating the side effects of camptothecin (CPT) in alkaline conditions. In acidic conditions, the collapse of ZIF-8 provides zinc ions that act as crosslinking agents for NaCMC, thereby improving the channel of drug release and enhancing the release amount from 51.9 % to 66.7 % compared with SP-grafted NaCMC micelles. The inherent biological adhesion of NaCMC ensures sustained drug release for a period of up to 40 h. These pH-responsive carriers hold great promise in the safe and effective treatment of cancer in the gastrointestinal tract, with CPT emerging as a highly promising candidate.

Abstract:Drug delivery system (DDS) is able to realize the treatment of cancer with lower side effects and higher efficiency comparing to conventional chemotherapy by controlling the in vivo dynamics of drugs. In recent years, there are many studies regarding to the release control of anticancer drug loaded in DDS in order to enhancing its performance. However, so far the reported controllable drug release DDSs require external stimulation such as light and cooling as triggers for drug release, which cannot reach the deep part inside the human body. To control the release in the deep part inside the human body, here, we aimed at to develop a new DDS carrier which can control drug release by applying magnetic field. Magnetic mesoporous silica (MMS) and ureido-containing poly (allylamine-co-allylurea) copolymers (PAU) were used in this study. PAU is a kind of polymer having upper critical temperature. By modifying it to the surface of magnetic mesoporous silica, it is possible to realize the control of drug release of the DDS carrier triggered by the core heat generation induced by an alternating magnetic field, thereby realizing the control of drug release in the deep part inside the human body. DDS carrier developed in this research enables highly efficient treatment that combines hyperthermia with core heat generation and drug treatment with drug release at the cancer in the body. To synthesize PAU modified MMS (MMS-PAU), magnetite was prepared by thermal decomposition method and MMSs were synthesized by a template method. PAU was prepared by reacting polyallylamine with potassium cyanate. Finally, PAU was grafted on the surface of MMS by introducing amide bonds. We confirmed that MMS-PAU has the drug release control ability and MMS-PAU is effective for cancer therapy.

Surface molecularly imprinted polymers (MIPs) are an efficient and rapidly advancing approach, increasingly being utilized in drug delivery systems (DDSs). This research involved synthesizing a core-shell structure, consisting of a metal-organic framework (MOF) core and a doxorubicin (DOX) imprinted polymethacrylamide shell, prepared via precipitation polymerization method in green media. The resulting MOF@MIP was utilized as a nanocarrier for DOX delivery, exhibiting high drug loading capacity and pH responsiveness delivery. The materials were characterized using different techniques such as X-ray diffraction (XRD), scanning electron microscopy (SEM), fourier transform infrared (FTIR), transmission electron microscopy (TEM), dynamic light scattering (DLS), thermal gravimetric analysis (TGA), and zeta potential (ZP). In vitro studies on DOX loading and release demonstrated that a higher amount of DOX was released in a sustained manner under tumor tissue conditions (pH 5, 41 °C) compared to normal physiological conditions (pH 7.4, 37 °C). Additionally, cytotoxicity evaluations revealed significant cytotoxic effects of DOX-loaded MOF@MIP on MCF-7 cells (IC50: 2 μg/mL). Based on these findings, the synthesized MOF@MIP shows promise as a potential candidate for the development of anticancer therapeutic delivery platforms. Mimosa/quince seed mucilage-co-poly (methacrylate) hydrogels for controlled delivery of capecitabine: Simulation studies, characterization and toxicological evaluation

Abstract:This research focused on developing pH-regulated intelligent networks using quince and mimosa seed mucilage through aqueous polymerization to sustain Capecitabine release while overcoming issues like short half-life, high dosing frequency, and low bioavailability. The resulting MSM/QSM-co-poly(MAA) hydrogel was evaluated for several parameters, including complex structure formation, stability, pH sensitivity, morphology, and elemental composition. FTIR, DSC, and TGA analyses confirmed the formation of a stable, complex cross-linked network, demonstrating excellent stability at elevated temperatures. SEM analysis revealed the hydrogels' smooth, fine texture with porous surfaces. PXRD and EDX results indicated the amorphous dispersion of Capecitabine within the network. The QMM9 formulation achieved an optimal Capecitabine loading of 87.17 %. The gel content of the developed formulations ranged from 65.21 % to 90.23 %. All formulations exhibited excellent swelling behavior, with ratios between 65.91 % and 91.93 % at alkaline pH. In vitro dissolution studies indicated that up to 98 % of Capecitabine was released after 24 h at pH 7.4, demonstrating the potential for sustained release. Furthermore, toxicological evaluation in healthy rabbits confirmed the system's safety, non-toxicity, and biocompatibility.

Abstract:Selective cancer cell targeting, controlled drug release, easy construction and multiple therapeutic modalities are some of the desirable characteristics of drug delivery systems. We designed and built simple capsule-like molecular imprinted polymer (MIP)-based nanoparticles for targeted and chemo-photothermal synergistic cancer therapy. Using dopamine (DA) as functional monomer, cross-linking agent as well as photo-thermal agent, ZIF-8 (zeoliticimidazolate framework-8) as drug carrier, epitope of EGFR (epidermal growth factor receptor) as template molecules, molecular imprinted polymer (MIP) drug carrier was constructed. The ability of MIP layer to bind to EGFR epitope endowed the MD () particles to recognize EGFR-overexpressing cancer cells, while the pH-responsiveness and photothermal conversion ability of PDA (polydopamine) achieved chemo-photothermal synergistic effects upon NIR irradiation. Taken together, the MD nanoparticles integrated cancer cell targeting recognition, intelligent drug release, biocompatibility and chemo-photothermal effects, and is therefore a promising tool for targeted cancer therapy with minimal toxicity to normal cells, as well as tumor imaging.

Abstract:This study aims to synthesize a core-shell gelatin-based carbon quantum dot-molecularly imprinted polymer (MIP@g-CQD) via the precipitation free-radical polymerization process using methotrexate (MTX) as a model anticancer template. To investigate the efficiency of the prepared photoluminescent MIP@g-CQD as a pH-responsive nano-carrier, MTX was loaded into MIP@g-CQD by soaking in a drug solution and the release behavior of the loaded drug was evaluated in the necessary pH values (7.4, 5). The successful synthesis of materials was characterized using PL, TEM, FE-SEM, DLS, and FT-IR analyses. Interestingly, the created cavities in the core-shell nano-carriers can interact with the MTX molecules effectively, leading to an increase in the loading capacity. According to the obtained results from Langmuir adsorption isotherms, the imprinting factor was calculated (IF = 4.91). Also, the binding kinetics of MTX revealed the creation of particular recognition sites in the core-shell polymeric network. The MTX-loaded MIP@g-CQD displayed a low rate and limited release at the simulated physiological environment (pH 7.4, 37 °C), but it is increased at tumor tissue (pH 5, 41 °C) conditions, which can lead to long-term and sustained release of MTX in the desired target. This property of MIP@g-CQD could avoid the release of MTX in normal physiological conditions, decreasing the possible side effects of MTX drug. Owing to the existence of amide functional groups in the nano-carrier structure and its negatively charged nature, the MTT assay displayed desirable cytotoxicity against the breast cancer cell line (MCF-7) for the MTX-loaded nano-carrier. According to the obtained results, the prepared safe photoluminescent MIP@g-CQD with appropriate pH-responsivity has a high ability to be applied as an anticancer and bio -detection agent.

Abstract:Construction of multifunctional stimuli-responsive nanotherapeutics enabling improved intratumoral penetration of therapeutics and reversal of multiple-drug resistance (MDR) is potent to achieve effective cancer treatment. Herein, we report a general method to synthesize pH-dissociable calcium carbonate (CaCO3) hollow nanoparticles with amorphous CaCO3 as the template, gallic acid (GA) as the organic ligand, and ferrous ions as the metallic center via a one-pot coordination reaction. The obtained GA–Fe@CaCO3 exhibits high loading efficiencies to both oxidized cisplatin prodrug and doxorubicin, yielding drug loaded GA–Fe@CaCO3 nanotherapeutics featured in pH-responsive size shrinkage, drug release, and Fenton catalytic activity. Compared to nonresponsive GA–Fe@silica nanoparticles prepared with silica nanoparticles as the template, such GA–Fe@CaCO3 confers significantly improved intratumoral penetration capacity. Moreover, both types of drug-loaded GA–Fe@CaCO3 nanotherapeutics exhibit synergistic therapeutic efficacies to corresponding MDR cancer cells because of the GA–Fe mediated intracellular oxidative stress amplification that could reduce the efflux of engulfed drugs by impairing the mitochondrial-mediated production of adenosine triphosphate (ATP). As a result, it is found that the doxorubicin loaded GA–Fe@CaCO3 exhibits superior therapeutic effect towards doxorubicin-resistant 4T1 breast tumors via combined chemodynamic and chemo-therapies. This work highlights the preparation of pH-dissociable CaCO3-based nanotherapeutics to enable effective tumor penetration for enhanced treatment of drug-resistant tumors.

Molecularly Imprinted Layer-Coated Hollow Polysaccharide Microcapsules Toward Gate-Controlled Release of Water-Soluble Drugs

Abstract:Molecularly imprinted polymers (MIPs) as drug delivery carriers have been explored, but it still remains a significant challenge to achieve a high drug loading amount and controlled drug release rate of MIPs. Here, we report a layer-by-layer self-assembly method for doxorubicin (DOX) imprinting of biocompatible microcapsules, which consist of carboxymethyl cellulose–(chitosan/alginate)5 and are able to encapsulate and deliver DOX. It has been demonstrated that the maximum loading capacity of the obtained hollow molecularly imprinted microcapsules (MIMs) to DOX was up to 155.1 μmol g−1. Uncommonly, the release process of DOX-loaded MIMs could be sustained for more than 168 h, resulting from the imprinted sites being blocked by DOX molecules, which entered the imprinted sites through electrostatic interaction. Moreover, the interaction could be weakened at lower pH due to the protonation of the carboxyl groups of O-carboxymethyl chitosan, leading to the DOX departing from the imprinted sites (namely DOX release). Meanwhile, in vitro experiments demonstrated that the hollow DOX-imprinted microcapsules had good biocompatibility, the encapsulated DOX could effectively induce the apoptosis of U373 malignant glioma cells and had better efficacy than that of free DOX in terms of tumor inhibition within 144 h. These findings indicate that the construction of the novel smart microcapsules of natural polymers plays a crucial role in the performance of controlled drug release. This facile strategy reported herein can be further expected to fabricate delivery systems of various water-soluble drugs.

Abstract:MIL-based molecularly imprinted polymer (MIP) nanocomposites were successfully synthesized through a simple and versatile stirring auxiliary encapsulation method. MIP as a carrier has been applied to the highly efficient selective recognition and sustained release of doxorubicin hydrochloride (DOX). The adsorption mechanism and release behavior of MIP@DOX in vitro were also discussed. Adsorption studies showed that MIP using DOX as template had specific selectivity to DOX, and its optimal drug loading efficiency reached 97.99%. The adsorption isotherm accorded with Freundlich models. The cumulative release curve showed that at the conditions of pH 5.5 and 7.4, the nanomaterials have a slow-release effect on the release of DOX. In addition, the cytotoxicity and bioactivity of MIP nanoparticles on HepG2 and HL-7702 cell lines measured by MTT assay also proved their low toxicity and biological activity. The cell activity of HepG2 and HL-7702 incubated with MIP for 24 h was 69.9% and 76.07%, respectively. These results collectively illustrated that the MIP nano-materials synthesized in this study can be efficiently employed to the drug delivery systems.

Abstract:In this study, we prepared pH-sensitive hybrid nanospheres through the implementation of a facile molecularly imprinted polymer (MIP) technique combined with a UV-initiated precipitation polymerization method using vancomycin (VA) for the templates. During the course of this investigation, both 2-hydroxyethyl methacrylate (HEMA) and 2-(diethylamino) ethyl methacrylate (DEAEMA) were utilized as the functional monomers, while ethylene glycol dimethacrylate (EGDMA) was used as a cross-linker. The obtained MIP nanospheres exhibited well-controlled particle size, with a drug loading capacity of about 17%, much higher than that of the non-imprinted polymer (NIP) nanospheres (5%). In addition, the VA loading quantity was closely correlated with the dosage of the cross-linking agent, and the MIP nanospheres exhibited a slower and more controlled VA release rate than the NIP nanospheres. Moreover, these MIP nanospheres were sensitive to pH values, and consequently showed an increasing release rate of VA as the pH level was decreased. The VA-loaded MIP nanospheres showed the higher antibacterial ratio of over 92% against Staphylococcus aureus (S. aureus) while the NIP nanospheres were friendly to S. aureus. These MIP nanospheres can be promising for targeting drug delivery system to achieve specific therapies such as preventing bacterial infections and killing cancer cells without damaging health cells and tissues.

Abstract:Molecularly imprinted polymers (MIPs) have garnered the interest of researchers in the drug delivery due to their advantages, such as exceptional durability, stability, and selectivity. In this study, a biocompatible MIP drug adsorption and delivery system with high loading capacity and controlled release, was prepared based on chitosan (CS) and graphene quantum dots (GQDs) as the matrix, and the anticancer drug oxaliplatin (OXAL) as the template. Additionally, samples without the drug (non-imprinted polymers, NIPs) were created for comparison. GQDs were produced using the hydrothermal method, and samples underwent characterization through FTIR, XRD, FESEM, and TGA. Various experiments were conducted to determine the optimal pH for drug adsorption, along with kinetic and isotherm studies, selectivity assessments, in vitro drug release and kinetic evaluations. The highest drug binding capacity was observed at pH 6.5. The results indicated the Lagergren-first-order kinetic model (with rate constant of 0.038 min −1 ) and the Langmuir isotherm (with maximum adsorption capacity of 17.15 mg g −1 ) exhibited better alignment with the experimental data. The developed MIPs displayed significant selectivity towards OXAL, by an imprinting factor of 2.88, in comparison to two similar drugs (cisplatin and carboplatin). Furthermore, the analysis of the drug release profile showed a burst release for CS-Drug (87% within 3 h) at pH 7.4, where the release from the CS-GQD-Drug did not occur at pH 7.4 and 10; instead, the release was observed at pH 1.2 in a controlled manner (100% within 28 h). Consequently, this specific OXAL adsorption and delivery system holds promise for cancer treatment.

Abstract:To address the undesirable reactions associated with matrine (MAT) injection in clinical settings, a high-loading drug delivery system (DDS) based on pH-sensitive molecularly imprinted polymer (MAT@MIPs) was prepared for the first time. The imprinted materials containing recognition sites for the matrine were formed by using carboxyl-functionalized multiwalled carbon nanotubes as a supportive matrix and dopamine as a cross-linker due to its exceptional biocompatibility. Subsequently, the optimal reaction conditions and adsorption performance of MAT@MIPs were systematically investigated. The obtained polymers were characterized and evaluated by Fourier transform infrared spectrometry, scanning electron microscopy, elemental analysis, and thermogravimetric analysis. Results indicated that the MIPs demonstrated a favorable imprinting factor (2.36) and a high binding capacity (21.48 mg·g −1 ) for matrine. In vitro studies, we performed cell counting kit-8 assays in HepG2 cells, then the drug delivery capabilities of MAT-loaded MIPs were validated through light microscopy analyses, and the matrine content in culture medium was quantified using ultra high-performance liquid chromatography-mass spectrum synchronously. The facile fabrication of MAT@MIPs presents a viable solution for designing high-loading and pH-responsive DDS, which can offer a novel administration approach for drugs requiring injection in clinical applications.

Abstract:We prepared novel DOX-loaded HMSNs coated with PDA and PEGMA.High loading capacity and entrapment efficiency were due to incorporation of HMSNs.The material exhibited pH and GSH mediated DOX controlled release profiles.HMSNs function as drug storage platforms and PDA and PEGMA act as gatekeepers.The drug carriers have great potentials to mediate drug release for cancer therapy.The development of multifunctional nano-drug carriers has become one of the most effective and prevailing approaches to overcome the non-selectivity and low cell uptake efficiency of drugs, as well as various side effects of traditional chemotherapy drugs. Herein, we reported novel hollow mesoporous silica nanoparticles (HMSNs) coated with polydopamine (PDA) and poly(poly(ethylene glycol) methacrylate) (PPEGMA) (PPEGMA) for the treatment of cancer. The HMSNs mainly served as a warehouse loading anticancer drug doxorubicin (DOX); the covering of PDA acted as pH-sensitive gatekeepers to control the diffusion and/or release of DOX from HMSNs in different pH stimuli environments, whilst the grafting of PPEGMA on the surface of by disulfide bonds and atom transfer radical polymerization (ATRP) increased the stability under physiological conditions and showed reductant sensitive responsiveness. The PPEGMA had a high loading capacity and entrapment efficiency of drugs through loading twice but low cytotoxicity. The in vitro release results indicated that the PPEGMA exhibited a high sensitivity to pH and glutathione (GSH) due to the PDA coating and disulfide bonds, which could control DOX release as intelligent "switches". The drug-loaded formulations could better be taken up by the cells, formed a higher concentration of DOX in the cells, and produced more remarkable apoptosis, thus achieving better treatment of tumors with high therapy efficacy. Therefore, the developed multifunctional materials could be used as a promising nano-drug carrier for cancer treatment.

Abstract:In this study, carboxymethyl chitosan (CMCS) with excellent biocompatibility was used as the "gatekeeper" to design and fabricate a pH-responsive drug delivery system (CMCS-DFNS) as paclitaxel carriers. Characterization results showed that CMCS-DFNS was successfully prepared and the nanocarriers displayed excellent drug loading efficiency of 19.8 %, and the results of the adsorption mechanism revealed that the adsorption of PTX was consistent with the Freundlich isotherm and pseudo-second-order kinetic model. Furthermore, the pH-responsive controlled release behavior at different pH (pH = 7.4, 6.5, and 5.0) was evaluated, and the results demonstrated that the cumulative release at pH 5.0 was 58.8 %, which was 2.7 times higher than that at pH 7.4, suggesting that the carrier exhibited a good pH sensitivity. The results of in vitro cellular experiments further indicated that CMCS-DFNS significantly improved the drug uptake efficiency in breast cancer MCF-7 cells. Importantly, the results of in vivo and cellular pharmacokinetic revealed that CMCS-DFNS can improve the circulation time and enhance the relative bioavailability of paclitaxel. Therefore, the fabricated pH-responsive drug delivery system has potential applications in the delivery of anti-tumor drugs, and provides a new delivery pathway for other compounds with low bioavailability.

Abstract:Therapeutic platforms such as chemotherapy that respond to physical and biological stimuli are highly desirable for effective cancer therapy. In this study, pH-responsive charge-reversal, polymer-coated mesoporous silica nanoparticles [PAH-cit/APTES-MSNs; PAH-cit refers to poly(allylamine)-citraconic anhydride; APTES refers to (3-aminopropyl)triethoxysilane] were synthesized for application as drug-delivery systems for the treatment of malignant cells. Confocal laser scanning microscopy (CLSM) revealed that the PAH-cit/APTES-MSNs nanocomposite effectively delivered and released doxorubicin hydrochloride to the nucleus of HeLa (human cervical carcinoma) cells. Additionally, the real-time dynamic drug-release process was monitored by CLSM. The current pH-controlled-smart-release platform holds promise in drug-delivery and cancer therapy-related applications.

Abstract:Molecularly imprinted hydrogel (MIH) as drug delivery system has been studied. It still remains a challenge to construct the stimuli-responsive MIH. Here, we report a coordinate bond strategy for imprinting doxorubicin (Dox) in hydrogel capable of pH-responsive and sustained drug delivery. The imprinting condition such as template–monomer interactions induced by metal ion was carefully investigated by spectroscopic methods. The obtained Dox–MIH was evaluated by absorption and in vitro release experiments. It has been demonstrated that the cupric ion mediated interaction between Dox and 4-vinyl pyridine via coordination and the optimal coordinate ratio of Dox/Cu2+ was 2:1. The rebinding amount of MIH to Dox was 2.7-fold that of nonimprinted hydrogel and the Dox-loaded MIH showed a pH-responsive release property. Not more than 10% of loaded drug was released from Dox–MIH at pH 7.2 during a time course of 7 days. However, near to 60% of loaded drug was sustainedly released at pH 5.0 during the same period. These results indicated that Dox–MIH with pH-responsive behavior possessed great promising as sustained-release delivery system of anticancer drug.
