Abstract:BACKGROUND:Moringa oleifera Lam (MO) is native to India and is a cash crop widely cultivated in tropical and sub-tropical areas. The health improving properties of MO has been studied from a long time ago for the numerous phenolic compounds, including vitamins, flavonoids, phenolic acids, isothiocyanates, tannins and saponins, which are present in considerable amounts in the plant. A growing spectrum of therapeutic characteristics of MO leaves has been found and used in the remission or treatment of oxidative stress, liver disease, neurological disease, hyperglycemia and cancer.HYPOTHESIS:This review focused on researches applying MO or MO leaf extract as a functional food or cure against various disease and cellular injuries. We believed it would help the discovery of therapeutic application of MO and understanding of MO phytochemistry.METHODS:The data collected in this review were extracted from researches indexed in Web of Science, google scholar, PubMed, Science Direct and Scopus to find out health benefits and biological activities of MO leaves polyphenols. The studies reporting mechanistic route of phenolic compounds of MO leaves were also considered in the present study.RESULTS:It has been reported that polyphenols of MO leaf have protective characteristics against neurodegenerative disorders through reducing DNA damage, activation of AchE activity and inhibition of caspase-3 activity. It has been reported that, they protected the kidney from damage caused by melamine through suppressed the pro-inflammatory cytokine, metallopeptidase inhibitor 1 (TIMP-1), and kidney injury molecule 1 (KIM-1). Similarly, methanol extract of MO leaves has low hypoglycemic attributes and attenuate the risk of diabetes caused by alloxan by enhancing lipid metabolism and stimulating insulin release, glucose uptake, and glycogen synthesis. In addition, MO leaves are becoming the best phytomedicine to reduce hypertension, which are naturally known as angiotensin-1converting enzyme (ACE), acetylcholinesterase, arginase and phosphodiesterase 5 (PDE5) inhibitors.CONCLUSION:MO leaves extract as a health promoting food additives for human and animals due to its great protective effect against many diseases and the widely persistent environmental toxins which disrupted cellular metabolic function. More studies are required to use the phenolic compounds of MO leaves to develop and produce drugs for controlling and treatment of various diseases.

Abstract:Moringa oleifera leaves (MOLs) have gained significant attention due to their nutritional and biological activity. Therefore, this study aimed to examine its flavor characteristics and underlying compositions. In this study, we used ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS), gas chromatography-mass spectrometry (GC-MS), electronic nose, electronic tongue, and molecular docking to comprehensively investigate the chemical properties and flavor profiles of MOLs. UPLC-Q-TOF-MS and GC-MS were instrumental in identifying the 20 non-volatile and 19 volatile constituents of MOLs, respectively. The electronic nose and electronic tongue systems provided an objective evaluation of the sweet, bitter, and spicy attributes and flavor characteristics of MOLs. Concurrently, molecular docking was employed to elucidate the material basis of flavor profiles. It revealed that glucosinolates and flavonoids are probably the key components for the bitter taste of MOLs. The sweet taste may be attributed to glucosinolates and flavonoids. The spicy scent appears to be linked to the presence of glucosinolates and alkaloids. The integration of these techniques confers a thorough understanding of the chemical composition and sensory properties of MOLs. These findings have significant implications for innovative applications in the food industry as well as pharmaceuticals and agriculture sectors; furthermore, they contribute towards enhancing the perception of Moringa oleifera as a valuable natural resource.

Abstract:Abstract Moringa oleifera is known as a drumstick tree and is cultivated in the subtropics and tropics. It exhibits antihypertensive and antidiabetic effects. An ultra-high-performance liquid chromatography method was developed for the determination of 9 phytochemicals in M. oleifera leaves and marketed products. The efficient separation was achieved within 7 min with a temperature of 45 °C by using a C-18 column as the stationary phase and water/acetonitrile with 0.05% formic acid as the mobile phase. The method was validated for linearity, repeatability, limits of detection, and limits of quantification. The limits of detections of phenolic compounds 1 – 9 were as low as 0.2 μg/mL. The photodiode array detector at 220 and 255 nm wavelengths was recruited for quantification. The key phytochemicals were detected in the range of 0.42 to 2.57 mg/100 mg sample weight in 13 dietary supplements. This study considers the quantitative analysis for lignans in M. oleifera for the first time. Isoquercitrin (5) and quercetin 3-O-(6-O-malonyl)-β−D-glucopyranoside (6) predominates the leaves of M. oleifera with inherent degradable nature detected for compound 6. Niazirin (2) was detected in amounts between 0.010 – 0.049 mg/100 mg while compound 1 was undetectable and potentially an artifact because of the fractionation process. The characterization and confirmation of components were achieved by liquid chromatography-electrospray ionization-mass spectrometry with extractive ion monitoring for the positive and negative ion modes. The developed and validated method is robust and rapid in the conclusive quantification of phytochemicals and authentication of the Moringa samples for quality assurance.

Abstract:In this study, we develop and validate a simultaneous quantification of polyphenols method based on an ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI-MS/MS) to adequately understand how different habitats influence the quality and profile of Moringa oleifera polyphenol. Furthermore, principal component analysis (PCA) and hierarchical cluster analysis (HCA) were used to compare and discriminate 25 samples collected from different areas. A significant correlation was found between the polyphenol profile and the collection area. Significant differences in the polyphenol content of Moringa oleifera from different regions indicate that the genetic diversity of Moringa oleifera was relatively rich, possibly due to differences in cultivation conditions, climate, or soil environment resulting in the accumulation of different polyphenols. These observations provide a theoretical basis for subsequent Moringa oleifera germplasm selection and development research. Furthermore, the quantitative analysis methodology used to characterize the polyphenols may be used toward developing quality assessment and future pharmacodynamic investigations of Moringa oleifera.

Abstract:The metabolites of Moringa oleifera Lam. ( M. oleifera ) leaves are involved in their biological function. However, a thorough examination of metabolites has not been possible due to the complex nature of metabolite distribution in M. oleifera leaves from different regions. The purpose of this study was to investigate regional differences in M. oleifera leaf metabolite profiles. Leaves from the Dehong, Baoshan, and Yuanmou regions of Yunnan Province were studied using ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-Q ExactiveTM HF-X MS)-based untargeted metabolomics. A total of 2084 electrospray ionization-positive compounds and 1294 electrospray ionization-negative compounds were identified. The three sample groups were found to be in different quadrants of the principal component analysis (PCA) map; hierarchical cluster analysis (HCA) indicated that variations in habitat conditions were the cause of the unstable quality of M. oleifera leaves; and Kyoto Encyclopedia of Genes and Genomes (KEGG) and orthogonal partial least squares-discrimination analysis (OPLS-DA) showed regional differences in metabolite numbers and associated pathways. These were the observed variations in the chemical compositions of M. oleifera leaves. Flavonoid and phenylpropanoid biosyntheses were the main focus in pathways related to the biosynthesis of other secondary metabolites. Phenolic substances played a significant role as either leaders or connectors in metabolic pathways. The results provide a deeper understanding of phytochemical variations in M. oleifera leaves from different geographical regions which has potential application in the quality control of raw materials.

Abstract:Moringa oleifera Lam. is a functional tree that is known to produce a variety of metabolites with purported pharmacological activities. It is frequently called the "miracle tree" due to its utilization in numerous nutraceutical and pharmacological contexts. This study was aimed at studying the chemical space of M. oleifera leaf extracts through molecular networking (MN), a tool that identifies metabolites by classifying them based on their MS-based fragmentation pattern similarities and signals. In this case, a special emphasis was placed on the flavonoid composition. The MN unraveled different molecular families such as flavonoids, carboxylic acids and derivatives, lignin glycosides, fatty acyls, and macrolactams that are found within the plant. In silico annotation tools such as network annotation propagation (NAP) and DEREPLICATOR, an unsupervised substructure identification tool (MS2LDA), and MolNet enhancer were also explored to further compliment the classic molecular networking output within the Global Natural Product Social (GNPS) site. In this study, common flavonoids found within Moringa oleifera were further annotated using MS2LDA. Utilizing computational tools allowed for the discovery of a wide range of structurally diverse flavonoid molecules within M. oleifera leaf extracts. The expansion of the flavonoid chemical repertoire in this plant arises from intricate glycosylation modifications, leading to the creation of structural isomers that manifest as isobaric ions during mass spectrometry (MS) analyses.

Abstract:Plant Based Natural Products (PBNPs) have been subject of interest since ancient time due to their use in food, industrial and biomedical applications. Research attention has further augmented to explore their phytochemical composition, properties, and potential application in the post-COVID era. In the present study phytochemical screening has been carried out with Methanolic Leaf Extracts of Moringa oleifera (MLEMO) followed by Gas Chromatography-Mass Spectrometry (GCMS) analysis. Phytochemical analysis of MLEMO revealed the presence of Alkaloids, Carbohydrates, Coumarins, Flavonoids, Glycosides, Phenol, Proteins, Quinones, Saponins, Steroids, Tannins and Terpenoids. Further, GCMS analysis revealed the presence of 41 compounds of which Dihydroxyacetone; Monomethyl malonate; 4H-Pyran-4-one,2,3-dihydro-3,5-dihydroxy-6-methyl; 1,3-Propanediol, 2-ethyl-2-(hydroxymethyl); Propanoic acid, 2-methyl-, octyl ester; 3-Deoxy-d-mannoic lactone; Sorbitol; Inositol; Cyclohexanemethanol, alpha-methyl-4-(1-methylethyl), Hexadecanoic acid, Methyl palmitate; n-Hexadecanoic acid (Palmitic acid); 9-Octadecenoic acid, methyl ester; Phytol; 9,12,15-Octadecatrienoic acid; Octadecanoic acid; 9-Octadecenamide were prominent. Most of the compounds in the list are bioactive and possess medicinal properties that are expected to serve as a baseline lead for the development of therapeutic agents. Abstract:In this study, an ultrasonic-assisted extraction (UAE) procedure with selected deep eutectic solvents (DES) as solvent was first designed to simultaneously optimize the total phenolic/flavonoid content (TPC/TFC) and antioxidant activities of Moringa oleifera L. leaves (MOLs) by using response surface methodology (RSM). The key factors for RSM were selected based on the design of the experimental results along with a three-factors-five-level, central composite design (CCD), including 20 experimental runs. The analysis of variance (ANOVA) results revealed that the water content in DES had a significant influence on all responses, while the ultrasonic time and the ratio of liquid to solid had no statistically significant effects on the total phenolic content. The optimal conditions of the combination of TPC/TFC and antioxidant activities were obtained as follows: 37% water content in DES, 144 W ultrasonic power, and 40°C ultrasonic temperature. The measured parameters corresponded with the predicted results. Moreover, a comparative study confirmed that the optimized DES-based UAE yielded further higher TPC, TFC, and antioxidant activities than other extraction methods. The results of HPLC analysis in optimized conditions verified that the MOLs extracts with DES-based UAE included 14 phenolic compounds with high concentrations of vicenin-2 (17.6 mg/g) and orientin (23.6 mg/g). The present study supplied a green and high-efficient method for extracting high levels of anti-oxidative phenolic compounds from MOLs.

Abstract:Moringa oleifera Lam. has been used as a traditional medicine for the treatment of numerous diseases. A simultaneous high-performance liquid chromatography (HPLC) analysis was developed and validated for the determination of the contents of crypto-chlorogenic acid, isoquercetin and astragalin, the primary antioxidative compounds, in M. oleifera leaves. HPLC analysis was successfully conducted by using a Hypersil BDS C18 column, eluted with a gradient of methanol-1% acetic acid with a flow rate of 1 mL/min, and detected at 334 nm. Parameters for the validation included linearity, precision, accuracy and limits of detection and quantitation. The developed HPLC method was precise, with relative standard deviation < 2%. The recovery values of crypto-chlorogenic acid, isoquercetin and astragalin in M. oleifera leaf extracts were 98.50, 98.47 and 98.59%, respectively. The average contents of these compounds in the dried ethanolic extracts of the leaves of M. oleifera collected from different regions of Thailand were 0.081, 0.120 and 0.153% (w/w), respectively. The developed HPLC method was appropriate and practical for the simultaneous analysis of crypto-chlorogenic acid, isoquercetin and astragalin in the leaf extract of M. oleifera. This work is valuable as guidance for the standardization of the leaf extracts and pharmaceutical products of M. oleifera.

Abstract:Interest in phenolic compounds has recently increased due to their potential as antioxidant agents. Moringa leaves (Moringa oleifera) have a high content of phenolic compounds, which presents them as a promising source for the extraction of these compounds. However, it is necessary to use analytical techniques to identify and quantify their actual compound content to determine the quality of the raw material and the potential of the final product. Ultrasound assisted extraction is a green, rapid and environmentally friendly extraction technique that produces high quality extracts from natural products. Hence, the aim of this research is to optimize the variables of the ultrasound-assisted extraction (UAE) process for the extraction of moringa leaves using response surface methodology (RSM) to extract the main flavonoids using methanol as the extraction solvent. A Box–Behnken design (BBD) has been used to evaluate the effect of the ultrasound extraction process variables. Finally, the optimal extraction time in the range between 2 and 30 min was determined. The results revealed that the methanol concentration was the most influential variable. The optimal extraction time was established at 15 min. Six of the main flavonoids were quantified by UHPLC-DAD and identified by UHPLC-QToF-MS. The results confirmed that UAE is an efficient method for the extraction and subsequent analysis and quality control of the flavonoids that can be found in moringa leaves.

Abstract:Moringa oleifera Lam. ( M. oleifera ) is a fast-growing and esculent plant with high nutritional and medicinal values. Traditionally, M. oleifera has been applied to treat anaemia, asthma, pain in joints, headaches, diarrhoea and so forth. Phytochemical studies have manifested that more than 300 components, including flavonoids, phenolic acids, organic acids, alkaloids, lignans, glucosinolates, carotenoids, proteins, fats, minerals, and so on, are present in different parts of M. oleifera . Simultaneously, M. oleifera has exhibited extensive notable pharmacological activities, such as antioxidant, anti-inflammatory, antimicrobial, anti-cancer, hypoglycemic, hypolipidemic, hypotensive, hepatoprotective, etc. Consequently, on the one hand, this work aims to comprehensively and holistically review the nutritional components, chemical constituents and pharmacological activities on various parts of M. oleifera , as well as its applications in functional food products and medicines. On the other hand, this work also looks forward to the new prospects and the future developments in the research of M. oleifera , and thus provides valuable insights for the follow-up and in-depth studies of this multifunctional tree.

Moringa oleifera Lam. Leaf improves constipation of rats induced by low-fiber-diet: A proteomics study

Abstract:Ethnopharmacological relevance: The leaf of Moringa oleifera Lam., a medicinal and edible herb for thousands of years in Ayurveda, is used as Pancha (na) karma (purgative) during and after the body cleansing process, to treat constipation, reduce cholesterol and body weight. Aim of study: The aim was to investigate the diarrhea effects and possible mechanism of M. oleifera leaves in constipation rats. Materials and methods: The hot water extract of M. oleifera leaves (WEMOL) was prepared and analyzed using LC-20AT HPLC system. The constipated rat model was induced by feeding with low fiber diet for 21 days. After oral administration of WEMOL for 7 days, the excretion parameter analysis, gastro-intestinal propulsion, histological analysis by HE and Alcian blue staining, and gastrointestinal hormone in rat's digestive tract through ELISA were used to evaluate the laxative effect of WEMOL. Label-free quantitation (LFQ) with LC-MS/MS, bioinformatics and Western blot were used to discover and verify the signal pathways and key proteins of WEMOL related to diarrhea. Results: The contents of isoquercitrin and astragalin were 2.7 mg/g and 1.7 mg/g, respectively in WEMOL. The stool number, weight, and water content of constipation rats were significantly reduced, indicating model had been established successfully. WEMOL (1.25 and 2.5 g/kg) increased water content of feces and the levels of Gas, MTL, NPY in gastric antrum and VIP, SP in colon of constipation rats, improved the muscle layer thickness and mucin secretion of colon. The proteomics revealed a total of 1731 differential proteins and 9 signaling pathways, WEMOL increased the expression of Vamp2, Gnai3, and Prkacb. Conclusions: The laxative mechanism of WEMOL maybe modulate the signaling pathways mediated by 5-HT and Ach receptors, related to gastrointestinal motility and intestinal fluid secretion. It can be considered as the scientific connotation of Pancha (na) karma of M. oleifera leaves in ayurveda.
Metabolite profiling of different solvent extracts of Moringa oleifera seeds and correlation with DPPH radical scavenging activity via 1H NMR-based metabolomics

Abstract:In the present study, profiling of the Moringa oleifera seeds metabolome was carried out by employing proton nuclear magnetic resonance (1H NMR) spectroscopy combined with multivariate data analysis (MVDA) of 3 different solvent extracts. The principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA) score plot reveal that methanol extract was discriminated from ethyl acetate and hexane extracts by PC1 while ethyl acetate and hexane extracts were well separated from methanol extract by PC2. The PLS-DA loading plot highlighted the potential metabolites, which are responsible for the group separation observed in the score plot. Further detailed examination of the loading plot shows that methanol extract contains significantly higher amount of vitamins, sterols, amino acids and fatty acids compared to the other extracts. A total of 37 compounds were detected from the 3 different solvents upon which the methanolic extract was identified to contain more metabolites and in a wider range than the other organic solvent extracts. Based on PLS analysis, ergosterol, oleic acid, isoleucine, riboflavin, cholesterol, leucine, ascorbic acid, stigmasterol, tryptophan, choline, histidine and cysteine displayed strong correlation to 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity. Hence, this extract would be more appropriate in metabolite extraction for analysis and for therapeutical benefits. Therefore, NMR spectroscopy combined with MVDA in compliment with the right choice of solvent for extraction could be utilized by applicable industries to obtain maximum valued metabolites within a short period of time. Besides having high diversity of metabolites, M. oleifera seeds can serve as potential nutritional source to develop new functional foods, and even as a source of biodiesel. Abstract:Moringa Oleifera is a fast growing deciduous tree with its different plant parts commonly used in herbal medicine, also with good socio-economic and industrial values. The ethanolic extract of M.Oleifera root bark is shown to be rich in nutrients, owing to the presence of essential phytochemicals in the root bark. The phytochemicals or secondary metabolites of the ethanolic extract of the root bark were screened and identified. Also, acute toxicity study of the ethanolic extract of the root bark was carriedout in Albino Rats according to Institutional Animal Care and Use (IACU) guidelines. The fresh M.Oleifera root was harvested at the inception of the rainy season at Pil Gani District of Langtang North LGA and excess soil washed off as the root bark was properly peeled, air dried, pulverized to powdered form and stored for subsequent use. Serial extractions using water, acetone, ethanol, and ethyl acetate were carriedout by Simple Maceration. The ethanolic extract was seen to have the best yield of the phytochemicals qualitatively identified as tannins, saponins, carbohydrates, flavonoids, alkaloids, steroids, terpenes, cardiac glycosides and anthraquinones. Acute toxicity test was determined by Lorke’s method and the LD50 was shown to be at the highest limit dose of 5000mg/kg, which signifies that the extract is non toxic at such single dosage. The ethanolic M.Oleifera root bark extract contains nutritive phytochemicals with vast medicinal uses. No sign of acute toxicity was observed on oral administration as the median lethal dose was shown to be safe at the highest limit single dose of 5000mg/kg.

Abstract:Background: Moringa oleifera is a tree that grows in tropical and subtropical areas around the world. Its leaves, seeds, bark, roots, and flowers are used as ingredients in meals and medicinal applications. Moringa leaf extracts have been studied to contain antioxidant compounds such as phenolic molecules. Recent extraction techniques such as ultrasound and microwaves are alternatives to increase the extraction performance of phenolic compounds while preserving their antioxidant activity.Objective: The approach consisted of optimizing, using a Composite Central Design of Response Surface Methodology (CCD-RMS), the process conditions, for ultrasound-assisted extraction (UAE) of antioxidant phenolic compounds from Moringa oleifera leaves. Methods: A 33 CCD-RMS was used; three independent variables were studied: ethanol concentration (25%, 50%, 75%), time (10, 20, and 30 min), and temperature (25, 37.5, and 50°C). The results showed that the optimal UAE conditions were an ethanol concentration of 52.4%, extraction time of 3.2 min, and temperature of 58.5°C; under these conditions the phenolic content was 13.92±0.21 mg GAE/g sample the total flavonoid content was 10.60±0.06 mg QE/g sample, and 52.22±2.01% and 72.81±1.58% of DPPH• and ABTS•+ radicals were inhibited, respectively.Conclusion: This optimization will make it possible to assess better use of M. oleifera leaves as an antioxidant ingredient in functional food development.Keywords: Moringa oleifera, composite central design, optimization, flavonoids, phenols, antioxidant activity.
Simultaneous UPLC-TQ-MS/MS determination of six active components in rat plasma: application in the pharmacokinetic study of Cyclocarya paliurus leaves

Abstract:Background: Cyclocarya paliurus (Batal.) Ijinskaja (CP) is a monotypic genus plant, also called sweet tea tree that belongs to the Juglandaceae family, which is mainly distributed in the subtropical highlands in China. Our previous work has verified that CP leaves exhibit a potent hyperglycemic effect by inhibiting pancreatic β cell apoptosis through the regulation of MPAK and Akt signaling pathways. However, the components that contribute to this potential health benefit remain undiscovered. Method: A sensitive, reliable, and validated ultra-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UPLC-TQ-MS/MS) method was developed to simultaneously determine the presence of six active components (neochlorogenic acid, chlorogenic acid, quercetin-3-O-glucuronide, kaempferol-3-O-rhamnoside, quercetin, and kaempferol) in rat plasma after a single oral administration (in a dosage of 10.5 g/kg) of an extract of CP leaves to rats. The separation was performed on a Waters ACQUITY BEH C18 column (50 mm × 2.1 mm, 1.7 μm). The detection was conducted by multiple reaction monitoring (MRM) in negative ionization mode. The two highest abundant MRM transitions without interference were optimized for each analyte. Acetonitrile and formic acid aqueous solution (0.1%) was used as the mobile phase at a flow rate of 0.3 ml/min. Result: The precision, accuracy, and recovery all satisfied the criteria of international guidance (Bioanalytical Method Validation Guidance for Industry, Food and Drug Administration), and the analytes were stable in plasma for all tested conditions. The main pharmacokinetic parameters were calculated by plasma concentration versus time profiles using the pharmacokinetics program. Conclusion: The pharmacokinetic parameters of each compound can facilitate future clinical studies.
Response Surface Methodology Optimization for Extraction of Antioxidants From Moringa Stenopetala Leaf, Study of Its Phytochemical Screening, and DPPH Antioxidant Assay

Abstract:Moringa stenopetala is a medicinal tree that originated in Africa and Asia. It has been used as a traditional medicine over centuries to treat diseases. Recent studies have shown that Moringa stenopetala is a rich source of antioxidants and other phytochemicals. This study investigated the extraction, phytochemical screening, and DPPH antioxidant assay of Moringa stenopetala leaf (MSL) extract. The study optimizes the process conditions to extract phytochemicals from the MSL. Using Design‐Expert software, the experiments were designed and optimized the extract from MSL using the response surface methodology approach. The best optimal parameters for extracting MSL extract were established as the concentration of ethanol: 50%, time: 48 h, and pH 6.5, which resulted in the highest responses: the MSL extract yield (67.95%) and the total phenol content (4.47 mg GAE/mL) with a higher desirability value of 1.0000. The crude extract was then subjected to phytochemical screening to identify the presence of various secondary metabolites. The DPPH antioxidant assay was used to estimate the antioxidant activity of MSL extract. Phytochemical screening results proved that MSL extract contains flavonoids, phenols, saponins, and tannins. DPPH antioxidant assay showed that the 0.15 mg/L of MSL crude extract had a robust antioxidant activity with a maximum percent inhibition of 85.08 ± 0.49. Thus, the results of this study suggested that MSL extract is a better source of antioxidants to use as natural preservatives in food and pharmaceutical industries.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3><p>Moringa oleifera Lam. leaf (MOL), a rich source of protein and phenolics, was traditionally used to treat various diseases including headaches, fevers, sore throat and dyslipidemia. Recently, MOL was reported to possess antioxidant, anti-dyslipidemia and hepato-renal protective activities, indicating that MOL could become a potential agent to improve metabolic disorders associated with hyperuricemia. The antihyperuricemic effect of MOL hydrolysate (MOLH) with high contents of phenolics and peptides remains unknown.</p><p><em>Aim of the study</em>: The aim of this study is to investigate xanthine oxidase (XO) inhibitory activity of MOLH, to clarify phenolic and peptide profiles of MOLH, and to evaluate possible mechanism underlying the antihyperuricemic effect of MOLH.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3><p>MOLH was prepared by enzymatic hydrolysis using commercial trypsin. XO inhibitory activity was determined by XO reaction-UPLC-MS coupling method. The chemical profiles of the phenolic and peptide fractions of MOLH were determined by UPLC-QTOF-MS/MS. The antihyperuricemic effect of MOLH was evaluated in a potassium oxonate-induced hyperuricemic rat model at doses of 200 and 500 mg/kg. Serum uric acid (UA), urea nitrogen, creatinine (CRE), triglyceride (TG), total cholesterol, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol levels, serum XO activity, liver malondialdehyde (MDA) equivalent level, renal tumor necrosis factor-α and interleukin-1β levels, and protein expression of renal urate-anion transporter 1, glucose transporter 9 and ATP-binding cassette transporter G2 were determined.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The phenolic and peptide fractions played key roles in inhibiting XO activity and blocking uric acid production. Five flavonoids and sixteen polypeptides were identified in the phenolic and peptide fractions of MOLH, respectively. MOLH (200 and 500 mg/kg) could effectively reduce the serum UA level of hyperuricemic rats (<em>p</em><0.001) by regulation of serum XO activity (<em>p</em><0.05 at 200 mg/kg, <em>p</em><0.01 at 500 mg/kg) and renal urate transporters. Besides, MOLH could improve metabolic disorders associated with hyperuricemia by its multiple actions on liver MDA (<em>p</em><0.001), serum CRE (<em>p</em><0.05 at 500 mg/kg) and serum TG (<em>p</em><0.001).</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>The results provided scientific evidence that MOLH rich in phenolics and peptides ameliorated hyperuricemia and metabolic disorders. This study validated the potential use of MOLH for regulation of hyperuricemia.</p>

[Decocting kinetics of Moringa oleifera leaves: based on correlation of decocting factors and multiple components]

Abstract:Content of multiple components (neochlorogenic acid,L-tryptophan,vicenin-2,isoquercitrin,and astragalin) in Moringa oleifera leaves was determined by high-performance liquid chromatography (HPLC),and the absolute content-time curves were plotted.Based on Fick's law of diffusion and Higbie's penetration theory,the parameters of the equations were calculated,and the measured results were substituted into the mathematical model to fit the equations.The n and a obtained from the equations on the decocting time factor and the solvent volume were close to each other.The dynamic models of the five components are as follows:■.The variation of the content of multiple components in M.oleifera leaves with time and solvent volume was explored.It was found that the content of the components was the highest when the leaves were decocted for 30 min with solvent volume 12 folds of the medicinal material.The dissolution and destruction of components and the diffusion movement of components are the main causes of the content change of M.oleifera leaves at different time and with different solvent volumes.The R~2of the linear equations on the content and the equations on the decocting process (5-30min and solvent volume 12-20 folds of the medicinal materials) was≥0.999 8 and≥0.9,respectively.Thus,the content determination and the decocting kinetic model had high accuracy,which can reflect the change law of the content of key components in M.oleifera leaves during the decoction.This study is expected to serve as a reference for optimizing the decocting technology.

Abstract:Magnolol, an essential bioactive natural lignans isolated from Magnolia Officinalis, has various pharmacological activities, such as antibacterial, anti-inflammatory, antioxidation and anti-tumor. In this study, a practical strategy based on ultra-high-performance liquid chromatography coupled with linear ion trap-Orbitrap mass spectrometry (UHPLC-LTQ-Orbitrap MS) was established to reveal the metabolic fate of magnolol in rat plasma and urine. First of all, in the data-dependent scanning (DDS) acquisition mode, the ESI-MS<em><sup>n</sup></em> data sets of biological samples and reference standard were obtained by using a high-quality online data analysis method. Subsequently, off-line data-mining techniques including multiple mass defect filters (MMDFs) and high-resolution extracted ion chromatograms (HREICs) were applied to screen major-to-trace metabolites of magnolol at the full-scan ESI-MS<sup>1</sup> level. Finally, a total of 177 metabolites (prototype compound included) were preliminarily observed and characterized according to accurate mass measurement, the characteristic fragmentation patterns, chromato-graphic behaviors, and corresponding Clog <em>P</em> values. The results showed that magnolol experienced a variety of biotransformation reactions in rats, including hydroxylation, methoxylation, dehydrogenation, carboxylation, sulfonation, glucuronide conjugation, glucose conjugation, N-acetylcysteine (NAC) conjugation, glutathione conjugation and their composite reactions. In conclusion, this study not only greatly expanded the understanding of the therapeutic material basis and pharmacological mechanism of magnolol, but also provided new ideas for its toxicity evaluation, safety monitoring and drug delivery forms design.
