Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 10(10) probiotics cfu/kg (ProL), and birds fed with added 10(11) probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:The recent ban on the use of antibiotics as a feed additive has led to the search for alternative sources of antibiotics in the feed industry. Presently, probiotics are considered as a potential substitute for antibiotic as a live biotherapeutic agent to improve animal health and performance. Accordingly, study was focused on evaluating the effect of Saccharomyces boulardii (Sb) and Bacillus subtilis B10 (Bs) on ultrastructure modulation and mucosal immunity development in broiler chickens. A total of three hundred 1-d-old Sanhuang broilers (a Chinese cross breed) were randomized into 3 groups, each group with 5 replications (n = 20). The control group (Ctr) was fed a basal diet containing an antibiotic (virginiamycin, 20 mg/kg). Meanwhile, broilers in experimental groups received Sb and Bs (1 × 108 cfu/kg of feed) in addition to the basal diet for 72 d. The results of the experimental groups revealed a significant improvement in live BW and relative weight of bursa of Fabricius and thymus. Also, intestinal villus height, width, and number of goblet cells increased in the Sb and Bs groups. Meanwhile, modulation in the intestinal ultrastructure and increased mRNA expression levels of occluding, cloudin2, and cloudin3 (P < 0.05) were observed in the Sb and Bs groups. Moreover, IgA-positive cells significantly increased in the jejunum of Sb- and Bs-supplemented groups (P < 0.05). Intestinal cytokines interleukin-6, tumor necrosis factor-α, interleukin-10, transforming growth factor-β, and secretory IgA concentrations were (P < 0.05) improved in the probiotic groups; however, Sb induced inflammatory and antiinflammatory cytokines (P < 0.05) in comparison with the Ctr group. The present findings conclusively revealed that Sb and Bs increased IgA-positive cells in the lumen of the intestinal villus and revealed that Sb and Bs could modulate intestinal ultrastructure through increasing occluding, cloudin2, and cloudin3 mRNA expression levels in broiler intestine.

Abstract:A total of 600, 1 day old male Lingnan Yellow broiler chickens were used to investigate the effects of Clostridium butyricum (C. butyricum) on growth performance, nitrogen metabolism, intestinal morphology and cecal microflora in broiler chickens. The birds were randomly assigned into 5 treatments (6 replicate pens per treatment with 20 birds per pen) and fed the same antibiotic-free basal diets during a 42 days feeding experiment. The treatments were as follows: no addition (Control), 2.5 x 10(7) cfu C. butyricum kg(-1) of diet (CB1), 5x10(7) cfu C. butyricum kg(-1) of diet (CB2), 1x10(8) cfu. C. butyricum kg(-1) of diet (CB3) and 10 mg colistine sulfate kg(-1) of diet (Antibiotic). Compared with the control birds, birds fed either CB1 or CB2 or antibiotic diet had greater (p<0.05) Body Weight (BW) on day 21 and 42 and higher (p<0.05) Average Daily Gain (ADG) from day 1-42. Birds fed C. butyricum or antibiotic diet had lower (p<0.05) Feed-to-Gain ratio (F:G) than the control birds from day 1-42. Dietary C. butyricum decreased (p<0.05) the concentration of serum Uric Acid (UA) compared with the control diet on day 21. Supplementation with CB2 or CB3 decreased (p<0.05) serum ammonia concentration compared with the control diet on day 21 and 42. Birds fed C. butyricum diet had higher (p<0.05) ileal villus height than the control birds on day 21 and 42. Birds fed CB2 or CB3 diet had lower (p<0.05) ileal crypth depth than the control birds on day 21 and 42. Supplementation with CB1 or CB3 decreased (p<0.05) the population of cecal Escherichia coli (E. coli) compared with the control on day 21. Birds fed C. butyricum diet had higher (p<0.05) population of cecal Bifidobacterium on day 21 and birds fed CB2 diet had higher (p<0.05) cecal Bifidobacterium on day 42 compared with the control birds. Supplementation with CB1 or CB2 increased (p<0.05) the number of cecal Lactobacillus on day 21 and supplementation with CB2 increased (p<0.05) Lactobacillus on day 42 compared with the control or antibiotic groups. The results indicate that supplementation with C. butyricum promotes growth performance, modulates nitrogen metabolism improves intestinal morphology and balances cecal microflora in broiler chickens.

Abstract:Knowledge about the modulation of gut microbiota improves our understanding of the underlying mechanism by which probiotic treatment benefits the chickens. This study examined the effects of Bacillus subtilis DSM 32315 on intestinal structure and microbial composition in broilers. Broiler chicks were fed basal diets without or with B. subtilis supplementation (1.0 × 109 spores/kg of diet). Supplemental B. subtilis increased average body weight and average daily gain, as well as elevated villus height and villus height to crypt depth ratio of ileum in broilers. Multi-dimension analysis showed a certain degree of separation between the cecal microbiota from treatment and control groups. Increased Firmicutes abundance and reduced Bacteroidetes abundance in cecum were observed responded to B. subtilis addition, which also increased the abundances of Christensenellaceae and Caulobacteraceae, and simultaneously decreased the abundances of potentially harmful bacteria such as Vampirovibrio, Escherichia/Shigella and Parabacteroides. Network analysis signified that B. subtilis addition improved the interaction pattern within cecal microbiota of broilers, however, it exerted little influence on the metabolic pathways of cecal microbiota by comparison of the functional prediction of metagenomes. In conclusion, supplemental B. subtilis DSM 32315 improved growth performance and intestinal structure of broilers, which could be at least partially responsible by the manipulation of cecal microbial composition.

Abstract:Dietary inclusion of probiotic Bacillus spp. beneficially affect the broiler chickens by balancing the properties of the indigenous microbiota causing better growth performance. The effects of three Bacillus spp. on the growth performance, intestinal morphology and the compositions of jejunal microflora were investigated in broiler chickens. A total of 480 1-day-old male Arbor Acres broilers were randomly divided into four groups. All groups had six replicates and 20 birds were included in each replicate. The control birds were fed with a corn-soybean basal diet, while three treatment diets were supplemented with Bacillus coagulans TBC169, B. subtilis PB6, and B. subtilis DSM32315 with a dosage of 1 × 109 cfu/kg, respectively. The experiment lasted for 42 days. The compositions and diversity of jejunal microflora were analyzed by MiSeq high-throughput sequencing. The B. coagulans TBC169 group showed marked improvements of growth performance, nutrient digestibility and intestinal morphology compared with the other B. subtilis treatments. B. coagulans TBC169 supplementation improved the average body weight (BW), average daily weight gain (ADG), total tract apparent digestibility of crude protein and gross energy (GE), and reduced feed conversion rate (FCR) compared with the control group (P < 0.05). The villus height to crypt depth ratio (VH/CD) of jejunum and duodenum was increased in the birds fed with B. coagulans TBC169 compared with the control group (P < 0.05). However, two B. subtilis treatments presented more positive variation of the jejunum microflora of chickens than that in the B. coagulans TBC169 group. B. subtilis PB6 and B. subtilis DSM32315 treatments improved the diversity of jejunal microbiota on day 21 compared with the control (P < 0.05), while which were decreased on day 42 (P < 0.05). The supplementation with B. coagulans TBC169 significantly improved the proportion of Firmicutes, otherwise two B. subtilis significantly improved the proportion of Proteobacteria, Bacteroidetes, Actinobacteria, and Acidobacteria at the phylum level during starter phase and decreased the proportion of Bacteroidetes during growing phase compared with the control. The supplementation with B.subtilis DSM32315 significantly improved the proportion of Clostridiales during starter phase, whereas two B. subtilis significantly improved the proportion of Pseudomonas, Burkholderia, Prevotella, DA101 during growing phase at the genus level compared with the control. In conclusion, the dietary supplementation with probiotic Bacillus spp. strains improved body weight and intestinal morphology in broiler chickens, which might be associated with the gut microbiota.

Abstract:This study investigated the effects of dietary supplementation with Bacillus subtilis (B. subtilis) or Bacillus licheniformis (B. licheniformis) on growth performance, immunity, antioxidant capacity, short chain fatty acid (SCFA) production, and the cecal microflora in broiler chickens. In total, 360 male, 1-day-old Cobb 500 birds were randomly divided into 3 groups: the control group was fed a basal diet; the B. subtilis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. subtilis; the B. licheniformis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. licheniformis. Results showed that chickens supplemented with either B. subtilis or B. licheniformis had comparatively higher (P < 0.05) body weight and average daily gain, whereas no difference (P > 0.05) was observed in feed efficiency. Concentrations of serum IgA, IgY, and IgM, as well as anti-inflammatory IL-10 were significantly increased (P < 0.05), and proinflammatory IL-1β and IL-6 were significantly decreased (P < 0.05) by B. subtilis or B. licheniformis supplementation. Moreover, chickens fed with diets supplemented by either B. subtilis or B. licheniformis had greater antioxidant capacity, indicated by the notable increases (P < 0.05) in glutathione peroxidase, superoxide dismutase, and catalase, along with decrease (P < 0.05) in malondialdehyde. Compared to the control group, levels of SCFA, excluding acetic and propionic acid, in cecal content had improved (P < 0.05) by adding B. licheniformis, and significant increase (P < 0.05) in acetic and butyric acid was observed with B. subtilis supplementation. Microbial analysis showed that both B. subtilis or B. licheniformis supplementation could increase butyrate-producing bacteria such as Alistipes and Butyricicoccus, and decrease pathogenic bacteria such as the Synergistetes and Gammaproteobacteria. In summary, dietary supplemented with B. subtilis or B. licheniformis improved growth performance, immune status, and antioxidant capacity, increased SCFA production, and modulated cecal microbiota in chickens. Moreover, B. licheniformis was more effective than B. subtilis with the same supplemental amount.

Abstract:To study the influence of different levels of Bacillus subtilis on growth performance, nutrition metabolism and intestinal microflora of 1 to 42 d Arbor Acres (AA) broilers, a total of 800 one-day-old healthy broilers were randomly divided into 5 groups with 4 replicates per group and 40 broilers per replicate. Broilers were fed a basic diet (group 1) which acted as the control group, and 4 other groups (2 to 5) were fed the basal diet with B. subtilis added at concentrations of 100, 150, 200 and 250 mg/kg, respectively for 42 days. The results showed as follow: the average daily gain (ADG) of group 4 was significantly higher than (P < 0.05) that of group 1, and the average daily feed intake (ADFI) of group 5 was the highest but the differences between groups were not significant (P > 0.05). The feed to gain ratio (F/G) of all the experimental groups was lower than that of the control and the difference was significant in group 4 (P < 0.05). In addition, supplementation of B. subtilis increased the apparent metabolism of crude protein (P > 0.05), crude fat (P > 0.05), dry matter (P > 0.05) and organic matter (P < 0.05). B. subtilis decreased the Escherichia coli and Salmonella populations in the cecum. This shows that adding B. subtilis to the broiler diet can improve the growth performance, increase feed efficiency, regulate serum index and reduce harmful bacteria in the intestinal tract. Based on our study, it could be recommended that addition of B. subtilis at 200 mg/kg could improve the growth performance of broilers.

Abstract:Studies have found that a dietary supplement of Bacillus amyloliquefaciens improved the growth performance, increased the nutrient digestibility of hosts and modulated the intestinal microflora. A total of 360 1-day-old Ross broilers were randomly divided into three treatments: a control group with a basal diet, an antibiotic group with a basal diet and added colistin sulphate, and a probiotics group with a basal diet and added Bacillus amyloliquefaciens. The HiSeq high-throughput sequencing analysis of 16S rRNA was used to investigate the differences in birds' caecal microflora, and metabolomics was used to analyse changes in caecal metabolites. Results showed that the supplementation of Bacillus amyloliquefaciens significantly improved the BW and ADG compared with the control birds. Results of sequencing indicated that (i) 645, 670, 596 unique operational taxonomic units (OTUs) were found in birds supplemented with Bacillus amyloliquefaciens on day 7, 21 and 42, separately, (ii) due to the diversity and relative abundance of the birds' caecal microflora, the OTUs of the caecal microflora clustered according to age and treatment, except on day 42, (iii) among the six predominate families (Ruminococcaceae, Lachnospiraceae, Enterobacteriaceae, Erysipelotrichaceae, Lactobacillaceae and Rikenellaceae), the supplementation of Bacillus amyloliquefaciens significantly increased Enterobacteriaceae on day 42, (iv) Bacillus amyloliquefaciens increased the relative abundance of Faecalibacterium and Ruminococcus on day 21, increased the Faecalibacterium and Blautia and decreased the Ruminococcus on day 42. The metabolomics of caecal metabolites showed that the dietary Bacillus amyloliquefaciens changed the caecal metabolites involved of amino acid metabolism and glyceride metabolism, and the antibiotics changed the caecal metabolites that were related to carbohydrates and amino acid metabolism on day 21.

Abstract:The gut microbiota plays important roles in animal overall health and productiveness. Balancing host gut microbiota by probiotics has been documented. Our previous study showed that Saccharomyces boulardii (Sb) and Bacillus subtilis B10 (Bs) significantly improve growth performance and modulate the intestinal histomorphology in broilers. To increase the knowledge regarding Sb and Bs, this study investigated the effects of these 2 probiotic strains on the gut microbiota in broilers. Three hundred 1-day-old Sanhuang broilers (Chinese cross breed) were randomly divided into 3 groups, each group with 5 replications (n = 20). The control group (CK) was fed a basal diet containing an antibiotic (virginiamycin, 20 mg/kg) and the other 2 groups received Sb and Bs (1 × 108 cfu/kg of feed) in addition to the basal diet. After 72 d of treatment, pyrosequencing revealed that the bacterial communities varied along the section of intestinal tract in the control and Bs groups, but not in the Sb group. No difference in microbial diversity was observed among 3 groups. The major phyla observed along the GI tract of broilers (particularly in the duodenum and cecum) were Firmicutes, Bacteroidetes, Proteobacteria, and Verrucomicrobia, which were considered potentially growth performance-related. Bacteroidetes, Proteobacteria, and Verrucomicrobia were observed at a much higher abundance in the jejunums and ileums of the Sb group (P < 0.05). In addition, the jejunal microbial communities formed 3 different clusters at either the genus level or the category of metabolism among the groups, based on the principal component analyses. These data indicated that Sb and Bs can modulate the microbial ecosystem, and subsequently enhance the health status of broilers.
Benefit of Dietary Supplementation with Bacillus subtilis BYS2 on Growth Performance, Immune Response, and Disease Resistance of Broilers

Abstract:A strain of Bacillus subtilis (B. subtilis) BYS2 was previously isolated from Mount Tai, which is located in Tai'an City in the Shandong Province of China. The strain was then stored in the Environmental Microbiology Laboratory at Shandong Agricultural University. To evaluate the effect of the bacterium preparation in broiler production, we fed the bacterium (106 CFU/g) to 1-day-old broilers and continued this feeding for 6 weeks to analyze its effect on growth and immune performance. We found that the average weight of the bacterium-fed group increased by 17.19% at weeks 5 compared to the control group (P < 0.05). The height of the villi in the duodenum and jejunum and the ratio of villi to crypt were significantly increased in the bacterium-fed group at weeks 5 (P < 0.05). Also, the IgG in the serum of broilers in the experimental group increased by 31.60% (P < 0.05) and IgM 30.52% (P < 0.05) compared with those in the control group. The expressions of the major pattern recognition receptors (PRRs), antiviral proteins, pro-inflammatory cytokines, and β-defensins were significantly higher than those in the control group (P < 0.05). Meanwhile, the bursa immune organ indices of broilers in the experimental group were significantly higher than those in the control group (P < 0.05). Also, after 5 weeks of continuous feeding, when infected with Escherichia coli (E. coli) O1K1 and Newcastle disease virus (NDV) F48E8, the content of bacteria and virus in tissues and organs of the experimental group decreased significantly, and the survival rate of infected chickens increased by 31.1% and 17.7%, respectively (P < 0.05). These results show that the anti-infective B. subtilis BYS2 could, to some extent, replace antibiotics to promote growth, improve innate immunity, and enhance disease resistance in broilers.
Effects of Dietary Bacillus subtilis HC6 on Growth Performance, Antioxidant Capacity, Immunity, and Intestinal Health in Broilers

Abstract:This study aimed to investigate the impact of Bacillus subtilis HC6 on the growth performance, immunity, antioxidant capacity, and intestinal health of broilers. A total of 180 one-day-old white feather broilers were randomly divided into two experimental groups, each comprising six replicates of fifteen chicks from 1 to 50 d of age. The groups were either fed a basal diet (CON) or the same diet supplemented with 5 × 108 cfu/kg of Bacillus subtilis HC6 (BS). Our results indicated that compared with the CON, dietary supplementation with BS increased feed efficiency during d 21–50 and d 1–50 (p < 0.05). Moreover, BS supplementation enhanced antioxidant capacity in the serum and liver, and also decreased the activity of diamine oxidase and the level of endotoxins (p < 0.05). Additionally, BS treatment increased the villi height in the jejunum and ileum, increased the ratio of villus height/crypt depth in the ileum, upregulated the expression of tight junction proteins in the jejunal mucosa, and downregulated the levels of IL-22 and IFN-γ on day 50 (p < 0.05). Principal coordinates analysis yielded clear clustering of two groups; dietary BS increased the relative abundance of Bacteroidales_unclassified (genus) and Olsenella (genus), and decreased the abundance of genera Alistipes on day 50, which identified a strong correlation with FCR, serum differential metabolites, or differential gene expression in the jejunal mucosa by spearman correlation analysis. The PICRUSt2 analysis revealed that supplementation with BS enriched the pathways related to xenobiotics biodegradation and metabolism, carbohydrate metabolism, energy metabolism, signaling molecules and interaction, the digestive system, and transport and catabolism. These results demonstrated that dietary BS increased feed efficiency, antioxidant capacity, and the mRNA expression of pro-inflammatory cytokines in the jejunal mucosa; and decreased the activity of diamine oxidase in serum, which might be attributed to the modulation of community composition and the functions of cecal microbiota in white-feathered broilers.

Abstract:This study was conducted to evaluate the effects of three Bacillus strains on growth performance, digestive enzyme activities, antioxidative capacity, serum immunity, and biochemical parameters in broilers. A total of 360 one-day-old Ross 308 chicks were randomly allocated into four groups with three replicates per group (n = 30). The control group was fed a basal diet, whereas the other groups fed basal diet supplemented with either Bacillus subtilis natto or Bacillus licheniformis or Bacillus cereus (10(8) cfu/kg) for 42 days, respectively. The results revealed that the probiotic-treated groups markedly improved final body weight, daily weight gain, and the activities of trypsin, amylase, lipase and total protease (p < 0.05). Moreover, chicks fed probiotics had higher serum glutathione peroxidase activity and O-2(-) level, as well as hepatic catalase and superoxide dismutase activities, whereas malondialdehyde levels in serum and liver were reduced (p < 0.05). The significant increased IgA (p < 0.05) was observed in the probiotics groups as compared to the control group. In addition, dietary administration of probiotic strain markedly reduced the levels of serum ammonia, uric acid, total cholesterol, and triglyceride. Taken together, these three probiotic Bacillus showed beneficial effects on chickens with minor strain specificity.

Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 1010 probiotics cfu/kg (ProL), and birds fed with added 1011 probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:The objective of this study was to investigate the effects of Bacillus subtilis-based probiotic supplementation in broiler chicken diets on growth performance, feed efficiency, intestinal cytokine, and tight junction (TJ) protein mRNA expression. Zero-day-old broiler chicks (n = 140) were randomly assigned to one of five dietary treatments: basal diet (CON); basal diet supplemented with either antibiotic bacitracin methylene disalicylate (BMD); or probiotics, namely, B. subtilis strain 1781 (PB1), a combination of B. subtilis strain 1104 + strain 747 (PB2), or B. subtilis strain 1781 + strain 747 (PB3). Body weight and feed intake were measured at 14 days of age, and the feed conversion ratio (FCR) was calculated. At 14 days of age, ileal samples were collected and used for intestinal cytokine, TJ protein, and mucin gene expression analysis using qRT-PCR. The chickens supplemented with antibiotic (BMD) and B. subtilis strain 1781 alone (PB1) had significantly higher body weights compared to controls of the same age. Dietary supplementation with antibiotic (BMD) or probiotics (PB1, PB2, PB3) significantly improved the feed efficiency as evidenced by decreased FCR compared to controls. No differences were observed in the expression of IL1β, IL17F, IFNγ, and MUC2 gene among the different treatment groups. However, elevated expression of IL6 (BMD, PB1, PB2), IL8 (PB2), and TNFSF15 (PB1, PB2, PB3) compared to controls was observed in the ileum. IL2 and IL10 expression was upregulated in chicks in the PB2 and PB3 groups, and IL4 was elevated in the PB1 group. IL13 was elevated in all probiotic-fed groups (PB1, PB2, PB3). Probiotic supplementation was also shown to significantly increase the expression of TJ proteins JAM2, ZO1 (PB2, PB3), and occludin (PB1, PB2). Taken together, B. subtilis supplementation altered intestinal immune activity and influenced gut barrier integrity through increased tight junction gene expression.

Abstract:In this paper,the probiotic preparation of Lactobacillus plantarum,Saccharomyces cerevisiae an d Bacillus subtilis were used in combination with digestion and anti-viral Chinese medicine feed additives.To study the effect on growth performance and immune organs of AA broilers.A total of 180 broilers were randomly divided into 4 groups with 3 replicates of 15 each.Control group(CK)feed basal diet,Ⅱ,Ⅲ and Ⅳ group feed the basal diet with 0.05% Chinese medicine preparation,0.05% probiotic preparation, 0.05% probiotics and 0.05% Chinese medicine preparation.Feeding period was 35 d.The results showed that the addition of probiotics and Chinese herbal medicine to the diet increased the daily gain of AA broilers at various stages,and the feed utilization rate was improved obviously.The daily gain of group Ⅳ was increased by 9.08%(P < 0.05),and the feed conversion ratio was decreased by 6.62% (P < 0.05)compared with CK group at 1 ~ 21 days.The daily gain of group Ⅳ was increased by 9.69% (P < 0.05),and the feed conversion ratio was decreased by 8.96% (P < 0.05)at 21 ~ 35 day.The daily gain of group Ⅳ was increased by 12.97% (P < 0.05)compared with CK group,and the feed conversion ratio was decreased by 9.17% (P < 0.05)at 21 ~ 35 days.In addition, continuous feeding probiotics and traditional Chinese medicine compatibility preparation 35 d could improve AA broiler thymus, spleen and french capsule index. The spleen index in group Ⅱ,Ⅲ,Ⅳwere increased by 13.81%(P > 0.05),14.90%(P < 0.05),30.21%(P <0.05).The thymus index in group Ⅱ,Ⅲ,Ⅳwere increased by 4.11%(P > 0.05),3.89%(P > 0.05),8.80% (P < 0.05).The french capsule index in group Ⅱ, Ⅲ, Ⅳwere inareased by 13.06%,13.86%,28.20%(P <0.05).The results showed that the combination of proboscis and traditional Chinese medicine could significantly improve the growth performance immune organs of AA broilers in 1 ~ 35 days.Both of them had synergistic effect,which could be used in the breeding of meat and poultry part of the effect of antibiotics.

Abstract:In this study, we investigated the effects of dietary supplementation with Bacillus subtilis (QST713) on the performance and intestinal health of yellow feather broilers under Coccidia and Clostridium perfringens (CP) challenge or CP alone. One-day-old yellow-feathered broiler roosters (n = 600) were randomly assigned to 5 groups (6 replicates with 20 roosters per replicate): the Con blank group, the CIC.p group (d24 Coccidia+d28-30 of CP challenge), the CIC.p + BS group (CIC.p +100 mg/kg B. subtilis), the C.p group (d 28-34 of CP challenge), and the C.p +BS group (C.p +100 mg/kg B. subtilis). The experiment lasted 80 d. The birds were evaluated for parameters such as average daily gain (ADG), average daily feed intake (ADFI), feed efficiency (F/G), intestinal lesion score, villus histomorphometry, intestinal tight junctions, inflammatory factors, and cecal microorganisms. The results revealed that 1) C.p. increased the F/G of broilers from 22 to 42 d (P < 0.05), whereas CIC.p. significantly decreased the 42 d and 80 d body weights (BW) and 22-42 d and 1-80 d ADG (P < 0.05) and significantly increased the 22 to 42 d and 1 to 80 d F/G (P < 0.05). The number of intestinal lesions significantly increased at 35 d and 42 d (P < 0.05). CIC.p significantly decreased the jejunum and ileum villus height (VH) and the ileum villus height/crypt depth (P < 0.05) at 35 d. The challenge significantly upregulated the expression of Claudin-1 and IL-4 mRNAs in the jejunum at 35 d and significantly downregulated the expression of IL-10 mRNA in the ileum at 35 d (P < 0.05); the number of unique OTUs in the challenge group decreased significantly after challenge treatment, and the relative abundances of Romboutsia at 35 d and Cladomyces and Lactobacillus at 42 d decreased significantly (P < 0.05). 2) Compared with the challenge groups, the addition of BS decreased the F/G of broilers from 22 to 42 d. Compared with the CIC group, the addition of BS significantly increased the F/G of broilers from 22 to 42 d. Compared with that in the CIC.p group, the addition of BS significantly increased the VH in the jejunum and ileum at 35 d (P < 0.05). Compared with the challenge groups, the BS groups presented significantly lower mRNA expression levels of Claudin-1 (P < 0.05) in the jejunum at 35 d. The Shannon and Chao indices suggested that BS increased the alpha diversity of cecum microorganisms in broilers. Dietary supplementation with B. subtilis can alleviate the damage to intestinal morphology and intestinal barrier function, as well as the altered cecal flora structure in broilers caused by Coccidia and C. perfringens infections.

Abstract:This study was conducted to examine the effects of B.subtilis on growth performance,antioxidant and immunity of broilers.A total of 216 day-old Ross 308 broilers were divided into the control group(basal diet) and the treatment group(basal diet supplemented with 105 CFU/g B.subtilis).Each group had three replicates and each replicate included 36 broilers(half male and half female).The experiment lasted for 42 d.The results showed that ADG,T-AOC and the activity of GSH-Px of the treatment group were improved significantly compared to the control group,while the level of serum MDA,NO and MDA in liver were decreased significantly.The B.subtilis could improve thymus index,bursa index and the level of serum IgG,but did not affect spleen index,serum lysozyme,IL-2 and TNF-α significantly.These results indicated that growth performance of broilers fed diet with B.subtilis was improved by increase of antioxidant and immunity.

Abstract:Probiotics are non-pathogenic microorganisms that are potentially important non-antibiotic alternatives. This study aimed to compare novel multi-strain and single-strain Bacillus probiotics and their respective influences on broiler chickens' performance, gut health, litter quality, immune response, and NBN and TLR gene expression. A total of 1200 Arbor-Acres 1-day-old broiler chicks were randomly allocated into three treatments (T1 was a control, T2 was supplemented with a combined Bacillus coagulans (2 × 10 9 cfu/g) and Bacillus licheniformis (8 × 10 9 cfu/g) probiotic strains (0.2 kg/ton of feed), and T3 was supplemented with Bacillus licheniformis (3.2 × 10 9 cfu/g) probiotic (0.5 kg/ton of feed) with eight replicas of each. Supplementing the broiler diet with either the single-strain (T3) or the multi-strain (T2) Bacillus -based probiotic raised the overall birds' body weight, body weight gain, feed conversion ratio, and European production efficiency factor compared to the control (T1), with a significant enhancement achieved by the multi-strain Bacillus product ( P = 0.005). T2 and T3 exhibited significantly improved cholesterol, Alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase, and alkaline phosphatase levels than the control ( P ≤ 0.05). The transcript levels of both NBN and TLR genes were upregulated in the liver in the T2 and T3 groups. The T2 group experienced significant reductions in gut bacterial counts, especially for Clostridia , and recorded the lowest litter moisture and nitrogen. In conclusion, supplementing broiler diets with probiotics of multiple Bacillus strains increased production profitability by promoting bird growth, improving feed intake, enhancing gut mucosa and immune organs, and upregulating genes responsible for immunity. All these inhibit the overgrowth of enteric pathogens and sustain litter quality.

Abstract:This trial was conducted to investigate the adhesion of Bacillus amyloliquefaciens to Caco-2 cells, the effects of Bacillus amyloliquefaciens on the mucosal immunity, cecal volatile fatty acids and microbial diversity in broiler chickens. Three hundred and sixty 1-d-old Ross 308 chicks were randomly divided into 3 dietary treatments groups, which birds fed with basal diet, basal diet supplemented with colistine sulfate, and basal diet supplemented with Bacillus amyloliquefaciens. Polymerase chain reaction denaturing gradient gel electrophoresis was used to analyze the change of cecal microflora, and Gas Chromatography was used to analyze the cecal volatile fatty acids. Data showed that: 1) Bacillus amyloliquefaciens had a good adhesion ability to epithelial cells; 2) the supplementation of Bacillus amyloliquefaciens significantly increased the concentration of ileal mucosal secretory IgA and interleukin 6, decreased (P Lass Than 0.05) the concentration of tumor necrosis factor-a; 3) resulted in the change of cecal microbial community, higher levels of acetic acid, methylacetic and isovaleric acid in the birds. Thus, we considered that Bacillus amyloliquefaciens enhanced the mucosal immunity, increased the cecal concentration of major volatile fatty acids and the diversity microflora community in broilers.
