Abstract:In many bacterial species, including the aquatic bacterium and human pathogen Vibrio cholerae, the second messenger cyclic diguanylate (c-di-GMP) modulates processes such as biofilm formation, motility, and virulence factor production. By interacting with various effectors, c-di-GMP regulates gene expression or protein function. One type of c-di-GMP receptor is the class I riboswitch, representatives of which have been shown to bind c-di-GMP in vitro. Herein, we examined the in vitro and in vivo function of the putative class I riboswitch in Vibrio cholerae, Vc1, which lies upstream of the gene encoding GbpA, a colonization factor that contributes to attachment of V. cholerae to environmental and host surfaces containing N-acetylglucosamine moieties. We provide evidence that Vc1 RNA interacts directly with c-di-GMP in vitro, and that nucleotides conserved among this class of riboswitch are important for binding. Yet the mutation of these conserved residues individually in the V. cholerae chromosome inconsistently affects the expression of gbpA and production of the GbpA protein. By isolating the regulatory function of Vc1, we show that the Vc1 element positively regulates downstream gene expression in response to c-di-GMP. Together these data suggest that the Vc1 element responds to c-di-GMP in vivo. Positive regulation of gbpA expression by c-di-GMP via Vc1 may influence the ability of V. cholerae to associate with chitin in the aquatic environment and the host intestinal environment.

Expression and Purification of Two Major Outer Membrane Proteins from Vibrio Alginolyticus

Abstract:Vibrio alginolyticus , a Gram-negative bacterium, is one of Vibrio pathogens common to human and marine animals. Outer membrane proteins of bacteria play an important role during infection and induction of host immune response. In present research, two outer membrane protein genes ( OmpK and OmpW ) of V. alginolyticus were cloned and expressed. The open reading frames of OmpK and OmpW contain 846 bp and 645 bp, respectively, the mature proteins consist of 261 and 193 amino acid residues. At the signal peptides positions −3 to −1, the amino acids were V-M-A in OmpK and V-F-A in OmpW , which consistented with the observed sequence V-X-A of the signal peptides of transmembrane OMP. The alignment analysis indicated that both proteins were highly conserved, which could serve as surface antigens for vaccine candidates. SDS-PAGE indicated two genes over-expressed in E. coli BL21 (DE3). By affinity chromatography on Ni 2+ -nitriloaceate resin, the recombinant proteins were purified from inclusion bodies. Western blot analysis revealed that both proteins had immunoreactivity, which provided a base for further study on the evaluation of diagnostication and vaccine candidates.
Study on VVC recombinant protein cytolytic activity mechanism of an extracellular cytolytic toxin of Vibrio vulnificus

Abstract:Objective To investigate the cytolytic activity of extracellular cytolytic toxin rVVC of Vibrio vulnificus on the apoptosis of human ECV304 cells, and to analyze the activities of Caspase-3,-8 and -9. Methods The cytotoxic effect of refolded rVVC on the growth and apoptosis of ECV304 cells was identified by MTT, Hochest33342/PI fluorescent staining, flow cytometry and DNA agarose electrophoresis analysis, respectively. The activities of Caspase-3, -8 and -9 was measured using a colorimetric method. Results The viability of human ECV304 cells exposed to rVVC was inhibited by rVVC after 24 h. 2.0 HU/ml rVVC groups had a better cytotoxic effect to human ECV304 than that of 0.5 HU /ml rVVC groups. The apoptosis of human ECV304 cells in 2.0 HU/ml rVVC+40 μmol/L Z-VAD-FMK groups was relative reduced than that of 2.0 HU/ml of rVVC groups. After 0.5 h treatment with 2.0 HU/ml of rVVC, the Caspase-3 activity in human ECV304 cells increased gradually and reached the peak at 3 h (versus control groups, P<0.01). The activity of Caspase-8 and -9 remained unchanging. Conclusion The rVVC has cytotoxic effect on human ECV304 and the cytolysin is probably correlated with Caspase-3.
Recombination Expression and Cytolytic Bioactivity of vvhA Fusion Protein:Cytolysin of V.vulnificus

Abstract:The aim of the study is to investigate cytolytic bioactivity of vvhA fusion protein (rVvhA): cytolysin of V.vulnificus on the apoptosis of human ECV304 cells. rVvhA was expressed highly in E.coli BL21 (DE3) bearing recombinant plasmid pET-28a(+)vvhA induced by 0.5 mmol/L IPTG. The expressed product was denatured and then purified by metal affinity chromatography using Ni2+-NTA resin. Finally, rVvhA was refolded using dilution and step dialysis. The bioactivity of rVvhA was confirmed by cytolysis against sheep erythrocytes and evaluated by MTT, Hoechst33342/PI fluorescent staining and flow cytometry. The results showed that rVvhA was purified to a purity of 88.6%. The fusion protein had a cytotoxic effect against sheep erythrocytes in time- and dose- dependent manner and reduced the viability of human ECV304 cells in a dose-dependent manner; The caspase pan inhibitor, Z-VAD-FMK, can partially inhibit rVvhA induced apoptosis. rVvhA has cytotoxic effect on human ECV304 cells and this process is probably correlated with the activation of caspase.

Abstract:The Vibrio vulnificus (V. vulnificus) hemolysin (VVH) is a pore-forming cholesterol-dependent cytolysin (CDC). Although there has been some debate surrounding the in vivo virulence effects of the VVH, it is becoming increasingly clear that it drives different cellular outcomes and is involved in the pathogenesis of V. vulnificus. This minireview outlines recent advances in our understanding of the regulation of vvhA gene expression, the biological activity of the VVH and its role in pathogenesis. An in-depth examination of the role of the VVH in V. vulnificus pathogenesis will help reveal the potential targets for therapeutic and preventive interventions to treat fatal V. vulnificus septicemia in humans. Future directions in VVH research will also be discussed.

Abstract:SummaryVibrio cholerae, the causative agent of the severe diarrheal disease cholera, has evolved signal transduction systems to control co‐ordinately the expression of virulence determinants. It was previously shown that the presence of the bile salts glycocholate and taurocholate in the small intestine causes dimerization of the transmembrane transcription factor TcpP by inducing intermolecular disulphide bonds in the TcpP periplasmic domain. In this study, they further investigated the mechanism of how taurocholate affects V. cholerae virulence determinants. In vitro assay of TcpP oxidation by VcDsbA showed that VcDsbA induced TcpP dimerization in the presence of taurocholate. Taurocholate bound to VcDsbA with a KD of 40 ± 2.5 μM, and also bound other Dsb proteins, including EcDsbA, EcDsbC and VcDsbC. Taurocholate inhibited VcDsbA reductase activity without affecting VcDsbA secondary structure or thermostability. VcDsbA and its substrates were more extensively reduced in the presence of taurocholate, as compared with their redox state in the absence of taurocholate. The data presented here not only provide new insights into the mechanism by which bile salts induce V. cholerae virulence but also suggest a means by which to develop inhibitors against DsbA.
Expression of the Cytolysin Gene from Vibrio Vulnificus in E.coli and Identification of Its Hemolytic Activity

Abstract:To express the cytolysin gene from Vibrio vulnificus in E.coli and to evaluate its hemolytic activity in order to provide basis for the study on immunologic activities and for the preparation of kits for monoclonal antibodies,the expression plasmid pET28a(+)—vvhA was constructed,the recombinant protein was purified by Ni-affinity chromatography after the inclusion bodies were washed by three step washings,and activity of the purified protein was vertified by hemolysis test.It was found that a highly expressed and highly purified(purity90%)recombinant protein VVC was thus obtained and as demonstrated by the rabbit red cell hemolysis test,its hemolytic activity was also proved to be quite high with a activity of 0.2 μg/HU.These results indicate that the cytolysin gene from V.vulnificus successfully express in the E.coli system.

Abstract:[Objective] To elucidate differences of biological characteristics and cytopathogenicities between T3SS 1 and T3SS2 in Vibrio parahaemolyticus.[Methods] With the main structural protein genes vcrD1 and vcrD2 as the research object,we used the homologous recombination technique to construct a series of deletion mutants ⊿vcrD1,⊿vcrD2,⊿vcrDl-vcrD2,and complementary strains C⊿vcrD1,C⊿vcrD2 of V.parahaemolyticus SH112 strain,then analyzed the differences of growth,biofilm,motility,cytotoxicity to RAW264.7 macrophages and Caco-2 cells,and transcriptional regulation of pro-inflammatory cytokine IL-1β,IL-6 in RAW264.7 macrophages between mutant strain and wild strain.[Results] Compared to the wild strain,the mutant strains had no difference on growth rate,however,the vcrD1 mutant obviously weakened biofilm formation,motility,cytotoxicity of V.parahaemolyticus;the vcrD2 mutant positively upregulated the transcriptional levels of pro-inflammatory cytokines,and significantly attenuated cytotoxicity to cells.The ⊿vcrD1-vcrD2 double deletion mutant showed weaker biofilm,motility,cell toxicity than vcrD1 mutant strain,and as similar to the vcrD1 mutant,in the mRNA levels of inflammatory cytokines when compared with the wild strain.[Conclusion] Contributions of two T3SSs to biological characteristics and cytopathogenicities of V.parahaemolyticus are different.The T3SS1 was mainly involved in biofilm formation and motility of V.parahaemolyticus,but also has a significant cytotoxic effect;T3SS2 did not affect bacterial biofilm formation and motility,but played a negative regulation on cell inflammatory reaction,potentially contributing to bacterial immune evasion in host.T3SS1 contributed to bacterial survival in the environment;T3SS2 may help V.parahaemolyticus to evade the host immune response.Moreover,T3SS1 and T3 SS2 may play related functions on biological characteristics and cytopathogenicities of V.parahaemolyticus,but the specific mechanism still needs further study.
[VPA1045 and VPA1049 of Vibrio Parahaemolyticus Regulate Translocation of Hcp2].

Abstract:OBJECTIVE:Vibrio parahaemolyticus causes acute gastroenteritis by secreting a number of virulence factors including two sets of type VI secretion systems (T6SS1 and T6SS2). However, it remains unknown how T6SS is regulated in the bacterium and what kinds of effects the systems have on adhesion onto and invasion into the host cells. This study was attempted to examine if VPA1045 and VPA1049 regulate T6SS2.METHODS:Vpa1045 and Vpa1049 deletion mutants were constructed by homologous recombination. Real-time PCR was performed to examine the transcription level of the translocon protein gene hcp2 in V. parahaemolyticus clinical strain HZ and its mutants. Western blot was used to analyze the levels of expression and translocation of Hcp2.RESULTS:Both VPA1045 and VPA1049 contain the Che-Y domain, and belong to the two-component regulators. Deletion of either Vpa1045 or Vpa1049 did not affect expression and transcription of Hcp2, but decreased translocation of Hcp2 in the supernatant of V. parahaemolycus. Bacterial adherence to the HeLa monolayers was also significantly reduced as compared with their parent strains.CONCLUSION:The two-component regulators VPA1045 and VPA1049 regulate T6SS2 of V. parahaemolyticus post-translationally by up-regulating Hcp2 translocation.

Abstract:Background Vibrio vulnificus is a Gram-negative pathogen that infects humans through foodborne or wound infections. Victims of V. vulnificus infections face significant health risks, including cellulitis and septicemia, which have rapid disease progression and high mortality rates. Diguanylate cyclase is responsible for producing the secondary messenger cyclic di-GMP. It plays a crucial role in regulating various bacterial physiological processes, such as motility, toxicity, and pathogenicity, through transcriptional regulation and affecting cyclic di-GMP levels. However, the DGC-mediated pathogenicity regulation in V. vulnificus is still unclear. Methods The vdcR gene in V. vulnificus was studied using a deletion strain (ΔVdcR) and an overexpression strain (oeVdcR) to understand its role in regulating the bacterium's pathogenicity. The electrophoretic mobility shift assay and RT-qPCR confirmed VdcR's impact on phosphodiesterase gene expression. To investigate how VdcR affects pathogenicity, V. vulnificus variant strains were assays for hemolysis, metalloprotease activity, cytotoxicity, resistance to phagocytosis, and lethality assays of the nematode Caenorhabditis elegans after infection. Results This study discovered a virulence-associated diguanylate cyclase, VdcR, which serves as a transcriptional regulator to induce phosphodiesterases and reduce the accumulation of cyclic di-GMP. VdcR expression resulted in low hemolysis, metalloprotease, and cytotoxicity activity. It also improved the cell adhesion ability and anti-phagocytosis activity to infect the host cell and escape the macrophage phagocytosis. The constitutively expressed VdcR in V. vulnificus caused low mortality rates in Caenorhabditis elegans survival assays. Conclusion The above evidence demonstrated that VdcR suppresses the pathogenicity in V. vulnificus YJ016.

Abstract:Bacterial ribonuclease E (RNase E) is vital for posttranscriptional regulation by degrading and processing RNA. The RraA protein inhibits RNase E activity through protein-protein interactions, exerting a global regulatory effect on gene expression. However, the specific role of RraA remains unclear. In this study, we investigated rraA expression in Vibrio alginolyticus ZJ-T and identified three promoters responsible for its expression, resulting in transcripts with varying 5 '-UTR lengths. During the stationary phase, rraA was significantly posttranscriptionally inhibited. Deletion of rraA had no impact on bacterial growth in rich medium Luria-Bertani broth with salt (LBS) but resulted in decreased biofilm formation and increased resistance to polymyxin B. Transcriptome analysis revealed 350 differentially expressed genes (DEGs) between the wild type and the rraA mutant, while proteome analysis identified 267 differentially expressed proteins (DEPs). Integrative analysis identified 55 genes common to both DEGs and DEPs, suggesting that RraA primarily affects gene expression at the posttranscriptional level. KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis demonstrated that RraA facilitates the conversion of fatty acids, propionic acid, and branched-chain amino acids to acetyl-CoA while enhancing amino acid and peptide uptake. Notably, RraA positively regulates the expression of virulence-associated genes, including those involved in biofilm formation and the type VI secretion system. This study expands the understanding of the regulatory network of RraA through transcriptome analysis, emphasizing the importance of proteomic analysis in investigating posttranscriptional regulation. IMPORTANCE RraA is an inhibitor protein of ribonuclease E that interacts with and suppresses its endonucleolytic activity, thereby playing a widespread regulatory role in the degradation and maturation of diverse mRNAs and noncoding small RNAs. However, the physiological functions and associated regulon of RraA in Vibrio alginolyticus have not been fully elucidated. Here, we report that RraA impacts virulence-associated physiological processes, namely, antibiotic resistance and biofilm formation, in V. alginolyticus. By conducting an integrative analysis of both the transcriptome and proteome, we revealed the involvement of RraA in carbon metabolism, amino acid catabolism, and transport, as well as in the type VI secretion system. Collectively, these findings elucidate the regulatory influence of RraA on multiple pathways associated with metabolism and pathogenesis in V. alginolyticus.
Differential processing of VesB by two rhomboid proteases in Vibrio cholerae

Abstract:Rhomboid proteases are universally conserved and facilitate the proteolysis of peptide bonds within or adjacent to cell membranes. While eukaryotic rhomboid proteases have been demonstrated to harbor unique cellular roles, prokaryotic members have been far less characterized. For the first time, we demonstrate that Vibrio cholerae expresses two active rhomboid proteases that cleave a shared substrate at distinct sites, resulting in differential localization of the processed protein. The rhomboid protease rhombosortase (RssP) was previously found to process a novel C-terminal domain called GlyGly-CTERM, as demonstrated by its effect on the extracellular serine protease VesB during its transport through the V. cholerae cell envelope. Here, we characterize the substrate specificity of RssP and GlpG, the universally conserved bacterial rhomboid proteases. We show that RssP has distinct cleavage specificity from GlpG, and specific residues within the GlyGly-CTERM of VesB target it to RssP over GlpG, allowing for efficient proteolysis. RssP cleaves VesB within its transmembrane domain, whereas GlpG cleaves outside the membrane in a disordered loop that precedes the GlyGly-CTERM. Cleavage of VesB by RssP initially targets VesB to the bacterial cell surface and, subsequently, to outer membrane vesicles, while GlpG cleavage results in secreted, fully soluble VesB. Collectively, this work builds on the molecular understanding of rhomboid proteolysis and provides the basis for additional rhomboid substrate recognition while also demonstrating a unique role of RssP in the maturation of proteins containing a GlyGly-CTERM. Importance: Despite a great deal of insight into the eukaryotic homologs, bacterial rhomboid proteases have been relatively understudied. Our research aims to understand the function of two rhomboid proteases in Vibrio cholerae. This work is significant because it will help us better understand the catalytic mechanism of rhomboid proteases as a whole and assign a specific role to a unique subfamily whose function is to process a subset of effector molecules secreted by V. cholerae and other pathogenic bacteria.
From motor protein to toxin: Mutations in the zonula occludens toxin (Zot) of Vibrio cholerae phage CTXϕ suggest a loss of phage assembly function

Abstract:AbstractProphages, i.e. dormant viruses residing in bacterial cells, are not just passive passengers in the bacterial host. Several prophage-encoded genes have been shown to be contributors to bacterial virulence by mediating antimicrobial resistance or by providing toxins. Other prophage genes exhibit beneficial effects on the host by modulating e.g. motility or biofilm formation. In this study, we used an in vivo phage assembly assay and tested an extensive array of single point mutations or their combinations found in Zot, the zonula occludens toxin encoded by the Vibrio cholerae phage CTXϕ. The assay makes use of the highly homologous Zot-like protein g1p of the filamentous Coliphage M13, a motor protein that mediates the trans-envelope assembly and secretion of filamentous phages. We also measured the in vitro ATP hydrolysis of purified proteins, and quantified virus production in V. cholerae mediated by Zot or the Zot-like protein of the two Vibrio phages CTXϕ and VFJϕ. In addition, we investigated sequence variations of the Walker motifs in Vibrio species using bioinformatics method, and revealed the molecular basis of ATP binding using molecular docking and molecular dynamics simulation based on the structure predicted by AlphaFold2. Our data indicates that g1p proteins in Vibrio can easily accumulate deleterious mutations and likely lose the ability to efficiently hydrolyse ATP, while the CTXϕ Zot was further exapted to now act as an auxiliary toxin during the infection by Vibrio cholerae.
Biological and transcriptional studies reveal VmeL is involved in motility, biofilm formation and virulence in Vibrio parahaemolyticus

Abstract:Vibrio parahaemolyticus is a marine pathogen thought to be the leading cause of seafood-borne gastroenteritis globally, urgently requiring efficient management methods. V. parahaemolyticus encodes 12 resistance/nodulation/division (RND) efflux systems. However, research on these systems is still in its infancy. In this study, we discovered that the inactivation of VmeL, a membrane fusion protein within the RND efflux systems, led to reduction of the ability of biofilm formation. Further results displayed that the decreased capacity of Congo red binding and the colony of Δ vmeL is more translucent compared with wild type strains, suggested reduced biofilm formation due to decreased production of biofilm exopolysaccharide upon vmeL deletion. In addition, the deletion of vmeL abolished surface swarming and swimming motility of V. parahaemolyticus . Additionally, deletion of vmeL weakened the cytotoxicity of V. parahaemolyticus towards HeLa cells, and impaired its virulence in a murine intraperitoneal infection assay. Finally, through RNA-sequencing, we ascertained that there were 716 upregulated genes and 247 downregulated genes in Δ vmeL strain. KEGG enrichment analysis revealed that quorum sensing, bacterial secretion systems, ATP-binding cassette transporters, and various amino acid metabolism pathways were altered due to the inactivation of vmeL . qRT-PCR further confirmed that genes accountable to the type III secretion system (T3SS1) and lateral flagella were negatively affected by vmeL deletion. Taken together, our results suggest that VmeL plays an important role in pathogenicity, making it a good target for managing infection with V. parahaemolyticus.

Abstract:Vibrio parahaemolyticus is the leading cause of seafood-borne gastroenteritis in humans worldwide. The major virulence factor responsible for the enteropathogenicity of this pathogen is type III secretion system 2 (T3SS2), which is encoded on the 80-kb V . parahaemolyticus pathogenicity island (Vp-PAI), the gene expression of which is governed by the OmpR-family transcriptional regulator VtrB. Here, we found a positive autoregulatory feature of vtrB transcription, which is often observed with transcriptional regulators of bacteria, but the regulation was not canonically dependent on its own promoter. Instead, this autoactivation was induced by heterogeneous transcripts derived from the VtrB-regulated operon upstream of vtrB . VtrB-activated transcription overcame the intrinsic terminator downstream of the operon, resulting in transcription read-through with read-in transcription of the vtrB gene and thus completing the autoregulatory loop for vtrB gene expression. The dampening of read-through transcription with an exogenous strong terminator reduced vtrB gene expression. Furthermore, a V . parahaemolyticus mutant with defects in the vtrB autoregulatory loop also showed compromises in T3SS2 expression and T3SS2-dependent cytotoxicity in vitro and enterotoxicity in vivo , indicating that this autoregulatory loop is essential for sustained vtrB activation and the consequent robust expression of T3SS2 genes for pathogenicity. Taken together, these findings demonstrate that the regulatory loop for vtrB gene expression based on read-through transcription from the upstream operon is a crucial pathway in T3SS2 gene regulatory network to ensure T3SS2-mediated virulence of V . parahaemolyticus . Many bacterial transcription factors undergo autoregulation, a process by which transcription factors regulate their own transcription to amplify or reduce the output responses to a changing environment. The common mode of such autoregulation is achieved by the transcription factor acting on its own promoter. In this study, we found that VtrB, a virulence regulator of the T3SS2 genes of the major food-borne pathogen Vibrio parahaemolyticus , autoregulates its own expression but independently of its own promoter. We also demonstrated how this autoregulation occurs: VtrB activates transcription of the upstream operon and transcription extends to the vtrB gene over the relatively less effective intrinsic terminator. Read-through transcription thus underlies the autoregulatory loop of vtrB expression. Moreover, this autoregulation was essential for the amplification of V . parahaemolyticus T3SS2 expression and induction of the full virulence of this pathogen. Together, our findings not only offer new insights into how V . parahaemolyticus controls its virulence gene expression to ensure pathogenicity but also provide a framework for further exploring the analogous mechanisms for the autoactivation of transcription factor gene expression in bacteria.

Abstract:IN 1950, Rees and Gray suggested that balanced surgical anaesthesia should consist of the triad of relaxation, narcosis and analgesia. With the increasing use of muscle relaxants for major surgery, this concept received widespread acceptance (Gray and Rees, 1952) and is the basis for the techniques employed by many anaesthetists today. Although Woodbridge (1957) and others have cast doubt on the accuracy of the use of the word "analgesia", which it has been suggested should be divided into sensory and reflex depression, the necessity for analgesia in obstetrics minor body surface operations, treatment of burns, etc., is well established. The demonstration of the anti-analgesic action of small doses of thiopentone by Clutton-Brock (1960) has raised many questions as to the choice of drugs for non-relaxant anaesthetic techniques. Clutton-Brock's findings have been verified by Dundee (1960) and this work prompted us to study the analgesic action of sub-anaesthetic concentrations of inhalation agents. In order to avoid any possible confusion in terminology, it should be pointed out that the authors refer to analgesia as a state in which there is decreased appreciation of pain. The conscious subject can indicate the presence of this state on request, while in the unconscious subject (where muscle relaxants have not been used) it can be demonstrated by absence or decreased response to a painful stimulus.
A putative Vibrio cholerae two-component system controls a conserved periplasmic protein in response to the antimicrobial peptide polymyxin B

Abstract:The epidemic pathogen Vibrio cholerae senses and responds to different external stresses it encounters in the aquatic environment and in the human host. One stress that V. cholerae encounters in the host is exposure to antimicrobial peptides on mucosal surfaces. We used massively parallel cDNA sequencing (RNA-Seq) to quantitatively identify the transcriptome of V. cholerae grown in the presence and absence of sub-lethal concentrations of the antimicrobial peptide polymyxin B. We evaluated the transcriptome of both wild type V. cholerae and a mutant carrying a deletion of vc1639, a putative sensor kinase of an uncharacterized two-component system, under these conditions. In addition to many previously uncharacterized pathways responding with elevated transcript levels to polymyxin B exposure, we confirmed the predicted elevated transcript levels of a previously described LPS modification system in response to polymyxin B exposure. Additionally, we identified the V. cholerae homologue of visP (ygiW) as a regulatory target of VC1639. VisP is a conserved periplasmic protein implicated in lipid A modification in Salmonellae. This study provides the first systematic analysis of the transcriptional response of Vibrio cholerae to polymyxin B, raising important questions for further study regarding mechanisms used by V. cholerae to sense and respond to envelope stress.

Abstract:Vibrio alginolyticus is a widely distributed marine bacterium that is a threat to the aquaculture industry as well as human health. Evidence has revealed critical roles for small RNAs (sRNAs) in bacterial physiology and cellular processes by modulating gene expression post-transcriptionally. GcvB is one of the most conserved sRNAs that is regarded as the master regulator of amino acid uptake and metabolism in a wide range of Gram-negative bacteria. However, little information about GcvB-mediated regulation in V. alginolyticus is available. Here we first characterized GcvB in V. alginolyticus ZJ-T and determined its regulon by integrated transcriptome and quantitative proteome analysis. Transcriptome analysis revealed 40 genes differentially expressed (DEGs) between wild-type ZJ-T and gcvB mutant ZJ-T-ΔgcvB, while proteome analysis identified 50 differentially expressed proteins (DEPs) between them, but only 4 of them displayed transcriptional differences, indicating that most DEPs are the result of post-transcriptional regulation of gcvB. Among the differently expressed proteins, 21 are supposed to be involved in amino acid biosynthesis and transport, and 11 are associated with type three secretion system (T3SS), suggesting that GcvB may play a role in the virulence besides amino acid metabolism. RNA-EMSA showed that Hfq binds to GcvB, which promotes its stability.
