Abstract:Background: Air pollution and genetic risk have been found to contribute to both onset and development of psoriasis. However, the extent to which genetic susceptibility modifies the effects of air pollutants on the risk of incident psoriasis remains unknown. Objectives: Our study aimed to assess the association between joint exposure to multiple air pollutants and the risk of psoriasis and the modification by the genetic susceptibility. Methods: This prospective study included 451,064 participants with complete air pollution data and free of psoriasis at baseline from the UK Biobank. All participants were enrolled from 2006 to 2010 and followed up to 2022. The air pollution score (APS) was calculated to assess the joint exposure to multiple air pollutants, including particulate matter (PM) with diameters ≤ 2.5 μm (PM2.5), between 2.5 and 10 μm (PM2.5-10), and ≤ 10 μm (PM10), as well as nitrogen dioxide (NO2) and nitrogen oxides (NOx). To evaluate the genetic risk, the polygenic risk score (PRS) for psoriasis was constructed. The Cox proportional hazard models were used to assess the association of air pollution and genetic susceptibility with the risk of psoriasis. Stratified analyses were conducted based on the individual characteristics. Results: During a median follow-up of 13.79 years, 4414 psoriasis events were recorded. The hazard ratios (HRs) [95% confidence intervals (CIs)] for psoriasis were 1.036 (0.936-1.147), 1.091 (0.987-1.206), 1.159 (1.048-1.283), and 1.163 (1.052-1.286) in higher quintile groups compared with the lowest quintile of APS (P trend <0.05). When considering genetic susceptibility, participants with high PRS and high APS had the greatest risk of incident psoriasis [HR (95% CI): 1.962 (1.630-2.362)] than those with low PRS and low APS. The HRs (95% CIs) for PM2.5-10, NOx, PM2.5 absorbance, PM2.5, NO2, and PM10 in the group with high exposure level and genetic risk were 1.831 (1.537-2.181), 1.722 (1.431-2.073), 1.698 (1.416-2.037), 1.619 (1.353-1.938), 1.504 (1.252-1.806), and 1.425 (1.192-1.704), respectively. Conclusions: Long-term exposure to various air pollutants is positively associated with an increased risk of incident psoriasis, particularly in individuals with high genetic risk. More comprehensive measures are needed to reduce the air pollution levels for better prevention of psoriasis.

Abstract:To investigate the association of long-term exposure to ambient air pollution with the risk of allergic rhinitis (AR), we performed a longitudinal analysis of 379,488 participants (47.4% women) free of AR at baseline in the UK Biobank. The annual average concentrations of PM2.5, PMcoarse, PM10, NO2, and NOx were estimated by land use regression models. Cox proportional hazard models were used to calculate hazard ratios (HRs) and 95% confidence intervals (CIs). A weighted polygenic risk score was constructed. During a median follow-up period of 12.5 years, 3095 AR cases were identified. We observed significant associations between the risk of AR and PM2.5 (HR: 1.51, 95% CI: 1.27-1.79, per 5 μg/m3), PMcoarse (HR: 1.28, 95% CI: 1.06-1.55, per 5 μg/m3), PM10 (HR: 1.45, 95% CI: 1.20-1.74, per 10 μg/m3), NO2 (HR: 1.14, 95% CI: 1.09-1.19, per 10 μg/m3), and NOx (HR: 1.10, 95% CI: 1.05-1.15, per 20 μg/m3). Moreover, participants with high air pollution combined with high genetic risk showed the highest risk of AR, although no multiplicative or additive interaction was observed. In conclusion, long-term exposure to air pollutants was associated with an elevated risk of AR, particularly in high-genetic-risk populations, emphasizing the urgent need to improve air quality.

Abstract:A comprehensive overview of the associations between air pollution and the risk of gastrointestinal (GI) diseases has been lacking. We aimed to examine the relationships of long-term exposure to ambient particulate matter (PM) with aerodynamic diameter <= 2.5 mu m (PM2.5), 2.5-10 mu m (PMcoarse), <= 10 mu m (PM10), nitrogen dioxide (NO2), and nitrogen oxides (NOx), with the risk of incident GI diseases, and to explore the interplay between air pollution and genetic susceptibility. A total of 465,703 participants free of GI diseases in the UK Biobank were included at baseline. Land use regression models were employed to calculate the residential air pollutants concentrations. Cox proportional hazard models were used to evaluate the associations of air pollutants with the risk of GI diseases. The dose-response relationships of air pollutants with the risk of GI diseases were evaluated by restricted cubic spline curves. We found that long-term exposure to ambient air pollutants was positively associated with the risk of peptic ulcer (PM2.5 : Q4 vs. Q1: hazard ratio (HR) 1.272, 95% confidence interval (CI) 1.179-1.372, NO2: 1.220, 1.131-1.316, and NOx: 1.277, 1.184-1.376) and chronic gastritis (PM2.5: 1.454, 1.309-1.616, PM10 : 1.232, 1.112-1.366, NO2: 1.456, 1.311-1.617, and NOx: 1.419, 1.280-1.574) after Bonferroni correction. Participants with high genetic risk and high air pollution exposure had the highest risk of peptic ulcer, compared to those with low genetic risk and low air pollution exposure (PM2.5: HR 1.558, 95%CI 1.384-1.754, NO2: 1.762, 1.395-2.227, and NOx: 1.575, 1.403-1.769). However, no significant additive or multiplicative interaction between air pollution and genetic risk was found. In conclusion, long-term exposure to ambient air pollutants was associated with increased risk of peptic ulcer and chronic gastritis.

Abstract:Abstract Air pollution (AP) is an alarming public health crisis worldwide. Common ambient pollutants include sulfur dioxide (SO2), carbon monoxide (CO), ozone (O3), coarse particulate matter (PM10), fine particulate matter (PM2.5), and nitrogen dioxide (NO2). Although previous studies have shown that AP has been associated with various cardiovascular, respiratory and inflammatory diseases, the relationship between AP and psoriasis remains unknown. This nationwide, population-based case–control study aims to explore the relationship between AP and incident psoriasis, using data from the National Health Insurance Research Database of Taiwan between 2012 and 2018. Patients with newly diagnosed psoriasis were included, which was confirmed by board-certified dermatologists or rheumatologists on at least three occasions. Exclusion criteria included patients with a diagnosis of psoriasis before 2012, patients under 20 years of age in 2012, or incomplete AP data. For each patient with psoriasis, four controls were matched considering age, sex, urbanization level, index date and comorbidities. AP data were retrieved from 73 general air quality monitoring stations operated by the Taiwan Environmental Protection Administration. Exposure to air pollutants (SO2, CO, O3, PM10, PM2.5 and NO2) was analysed in the 5 years preceding psoriasis diagnosis and in controls. The mean pollution levels were further divided into four quartiles (1 = lowest and 4 = highest) to assess the dose–response effect. A conditional logistic regression model was used to assess the association. Our study included 45 093 patients with psoriasis and 180 372 matched controls. No significant intergroup difference was found in age, sex, urbanization level or comorbidities. After adjusting for age, sex, urbanization, index date and comorbidities, the second and third concentration quartiles of CO exposure were associated with a decreased psoriasis incidence [adjusted odds ratio (aOR) 0.94, 95% confidence interval (CI) 0.91–0.97; and aOR 0.91, 95% CI 0.89–0.94, respectively), while the third concentration quartile of PM10 (aOR 1.05, 95% CI 1.02–1.08) and the second concentration quartile of PM2.5 (aOR 1.08, 95% CI 1.04–1.11) were associated with a slightly increased incidence of psoriasis. Using a model incorporating all air pollutants, we found that the levels of PM10 (quartile 4: aOR 1.09, 95% CI 1.03–1.16), PM2.5 (quartile 2: aOR 1.08, 95% CI 1.04–1.13) and NO2 (quartile 4: aOR 1.14, 95% CI 1.05–1.23) were associated with a slight increase in the incidence of psoriasis. Our study shows that although exposure to certain PM10, PM2.5 and NO2 levels was significantly associated with incident psoriasis, the increased risks were trivial, with no dose–response relationship.

Abstract:Background Air pollutants are considered as non-negligible risk factors of idiopathic pulmonary fibrosis (IPF). However, the relationship between long-term air pollution and the incidence of IPF is unknown. Our objective was to explore the associations of air pollutants with IPF risk and further assess the modification effect of genetic susceptibility. Methods We used land-use regression model estimated concentrations of nitrogen dioxide (NO2), nitrogen oxides (NOx) and particulate matter (fine particulate matter with diameter <2.5 μm (PM2.5) and particulate matter with diameter <10 μm (PM10)). The polygenic risk score (PRS) was constructed using 13 independent single nucleotide polymorphisms. Cox proportional hazard models were used to evaluate the associations of air pollutants with IPF risk and further investigate the modification effect of genetic susceptibility. Additionally, absolute risk was calculated. Results Among 433 738 participants from the UK Biobank, the incidence of IPF was 27.45 per 100 000 person-years during a median follow-up of 11.78 years. The adjusted hazard ratios of IPF for each interquartile range increase in NO2, NOx and PM2.5 were 1.11 (95% CI 1.03–1.19), 1.07 (95% CI 1.01–1.13) and 1.09 (95% CI 1.02–1.17), respectively. PM2.5 had the highest population attribution risk, followed by NOx and NO2. There were additive interactions between NO2, NOx and PM2.5 and genetic susceptibility. Participants with a high PRS and high air pollution had the highest risk of incident IPF compared with those with a low PRS and low air pollution (adjusted hazard ratio: NO2 3.94 (95% CI 2.77–5.60), NOx 3.08 (95% CI 2.21–4.27), PM2.5 3.65 (95% CI 2.60–5.13) and PM10 3.23 (95% CI 2.32–4.50)). Conclusion Long-term exposures to air pollutants may elevate the risk of incident IPF. There are additive effects of air pollutants and genetic susceptibility on IPF risk. Long-term exposures to air pollutants were significantly associated with increased risk of incident idiopathic pulmonary fibrosis (IPF). Abstract:BACKGROUND:The effect of long-term exposure to air pollution on the development of chronic obstructive pulmonary disease (COPD) is still controversial, and the role of the interactions of air pollution with genetic risk and lifestyle in COPD risk is unclear.METHODS:We included 452762 participants derived from the UK Biobank. Annual concentrations of air pollutions, including particle matter (PM2.5, PM10), nitrogen oxides (NOx), and nitrogen dioxide (NO2), were assessed using land-use regression model. We applied Cox proportional hazard model to evaluate the associations between air pollution and COPD risk. In addition, we constructed a polygenic risk score and a lifestyle score, and assessed whether genetic susceptibility and lifestyle modified the effect of air pollution on the COPD risk.FINDINGS:Each interquartile range (IQR) increase in annual concentrations of PM2.5, PM10, NOx, and NO2 was associated with 1.17 (95% CI: 1.15,1.19), 1.05 (95% CI: 1.03,1.06), 1.13 (95% CI: 1.11,1.14), and 1.19 (95% CI: 1.16,1.21) times the risk of COPD, respectively. We observed an additive interaction between PM2.5 and genetic risk (P-interact=0.095), and a negative interaction between PM2.5 and lifestyle (P-interact=0.062). The HRs for each IQR increase in PM2.5 were 1.21, (95% CI: 1.16-1.25) and 1.24, (95% CI: 1.21-1.26) in individuals with healthy and unfavourable lifestyle, respectively; and 1.16, (95% CI: 1.13-1.19) and 1.19, (95% CI: 1.16-1.22) in those with low genetic risk and high genetic risk, respectively. Participants with high air pollution exposure, high genetic risk and unfavourable lifestyle showed the highest risk of COPD.INTERPRETATION:Long-term exposure to air pollution was associated with increased risk of COPD, especially in those with high genetic risk and unfavourable lifestyle.FUNDING:None.

Abstract:Background: In recent years, an increasing number of observational studies have reported the impact of air pollution on autoimmune diseases (ADs). However, no Mendelian randomization (MR) studies have been conducted to investigate the causal relationships. To enhance our understanding of causality, we examined the causal relationships between particulate matter (PM) and nitrogen oxides (NO x ) and ADs. Methods: We utilized genome-wide association study (GWAS) data on PM and NO x from the UK Biobank in European and East Asian populations. We also extracted integrated GWAS data from the Finnish consortium and the Japanese Biobank for two-sample MR analysis. We employed inverse variance weighted (IVW) analysis to assess the causal relationship between PM and NO x exposure and ADs. Additionally, we conducted supplementary analyses using four methods, including IVW (fixed effects), weighted median, weighted mode, and simple mode, to further investigate this relationship. Results: In the European population, the results of MR analysis suggested a statistically significant association between PM 2.5 and psoriasis only (OR = 3.86; 95% CI : 1.89–7.88; P IVW < 0.00625), while a potential association exists between PM 2.5–10 and vitiligo (OR = 7.42; 95% CI : 1.02–53.94; P IVW < 0.05), as well as between PM 2.5 and systemic lupus erythematosus (OR = 68.17; 95% CI : 2.17–2.1e+03; P IVW < 0.05). In East Asian populations, no causal relationship was found between air pollutants and the risk of systemic lupus erythematosus and rheumatoid arthritis ( P IVW > 0.025). There was no pleiotropy in the results. Conclusion: Our results suggest a causal association between PM 2.5 and psoriasis in European populations. With the help of air pollution prevention and control, the harmful progression of psoriasis may be slowed.

Abstract:Background Limited research has been conducted on the association between long-term exposure to air pollutants and the incidence of gout. Objectives This study aims to assess the individual and combined effects of prolonged exposure to five air pollutants (NO 2 , NO x , PM 10 , PM coarse and PM 2.5 1 ) on the incidence of gout among 458,884 initially gout-free participants enrolled in the UK Biobank. Methods Employing a land use regression model, we utilized an estimation method to ascertain the annual concentrations of the five air pollutants. Subsequently, we devised a weighted air pollution score to facilitate a comprehensive evaluation of exposure. The Cox proportional hazards model was utilized to investigate the association between ambient air pollution and gout risk. Interaction and stratification analyses were conducted to evaluate age, sex, BMI, and genetic predisposition as potential effect modifiers in the air pollution-gout relationship. Furthermore, mediation analyses were conducted to explore the potential involvement of biomarkers in mediating the association between air pollution and gout. Results Over a median follow-up time of 12.0 years, 7,927 cases of gout were diagnosed. Significant associations were observed between the risk of gout and a per IQR increase in NO 2 (HR 2 : 1.05, 95% CI 3 : 1.02-1.08, p =0.003), NO x (HR: 1.04, 95% CI: 1.01-1.06, p =0.003), and PM 2.5 (HR: 1.03, 95% CI: 1.00-1.06, p =0.030). Per IQR increase in the air pollution score was associated with an elevated risk of gout ( p =0.005). Stratified analysis revealed a significant correlation between the air pollution score and gout risk in participants ≥60 years (HR: 1.05, 95% CI: 1.02-1.09, p =0.005), but not in those <60 years ( p =0.793), indicating a significant interaction effect with age ( P-interaction =0.009). Mediation analyses identified five serum biomarkers (SUA:15.87%, VITD: 5.04%, LDLD: 3.34%, GGT: 1.90%, AST: 1.56% 4 ) with potential mediation effects on this association. Conclusions Long-term exposure to air pollutants, particularly among the elderly population, is associated with an increased risk of developing gout. The underlying mechanisms of these associations may involve the participation of five serum biomarkers.

Abstract:Objective Air pollution has been linked to multiple psychiatric disorders, but little is known on its long-term association with schizophrenia. The interaction between air pollution and genetic susceptibility on incident schizophrenia has never been reported. We aimed to explore the associations between long-term air pollution exposure and late-onset schizophrenia and evaluate whether genetic susceptibility could modify the association. Methods This population-based prospective cohort study included 437,802 middle-aged and elderly individuals free of schizophrenia at baseline in the UK Biobank. Land use regression models were applied in the estimation of the annual average concentrations of nitrogen dioxide (NO 2 ), nitrogen oxides (NO x ), fine particulate matter (PM 2.5 ), and inhalable particulate matter (PM 10 ) at residence. The associations between air pollutants and schizophrenia were evaluated by using Cox proportional hazard models. A polygenic risk score of schizophrenia was constructed for exploring potential interaction of air pollutants with genetic susceptibility. Results An interquartile range increase in PM 2.5 , PM 10 , NO 2 , and NO x was associated with the hazard ratios (HR) for incident schizophrenia at 1.19, 1.16, 1.22, and 1.09, respectively. The exposure-response curves for the association of air pollution with incident schizophrenia were approximately linear. There are additive interactions of air pollution score (APS), PM 10 , NO 2 , and NO x with genetic risk. Specifically, compared with participants with low genetic susceptibility and low APS, the HR was 3.23 for individuals with high genetic risk and high APS, among which 0.49 excess risk could be attributed to the additive interaction, accounting for 15 % of the schizophrenia risk. Conclusion This large-scale, prospective cohort study conveys the first-hand evidence that long-term air pollution exposure could elevate schizophrenia incidence in later life, especially for individuals with higher genetic risks. The findings highlight the importance of improving air quality for preventing the late-onset schizophrenia in an aging era, especially among those with high genetic risks.

Abstract:Abstract Background Psoriasis can be provoked by both external and internal factors. The effects of environmental factors on psoriasis remain unclear. Objectives To investigate the effects of air pollution on outpatient visits for psoriasis. Methods A distributed lag nonlinear model following quasi-Poisson regression was used to evaluate the lag effects of air pollutants on psoriasis outpatient visits, adjusting for potential confounders. Stratified analyses were performed to identify potential effect modifications by sex, age and season. Results In total, 13 536 outpatient visits for psoriasis were recorded in Wuhan, China from 1 January 2015 to 31 December 2019. In the single-pollutant model, exposures to particulate matter (PM) smaller than 2.5 μm (PM2.5), PM smaller than 10 μm (PM10), NO2 and SO2 were found to be significantly associated with increased daily psoriasis outpatient visits. For the largest effects, a 10-μg m−3 increase in concentrations of PM2.5 (lag1), PM10 (lag1), NO2 (lag0) and SO2 (lag3) corresponded to 0.32% [95% confidence interval (CI) 0.01–0.63], 0.26% (95% CI 0.05–0.48), 0.98% (95% CI 0.01–1.96) and 2.73% (95% CI 1.01–4.47) increases in psoriasis outpatient visits, respectively. In the two-pollutant model, only NO2 showed significant and stable effects on the outpatient visits for psoriasis. Conclusions Ambient air pollution, especially NO2, appears to be significantly associated with an increased risk of outpatient visits for psoriasis in Wuhan, China. Air pollution control and exposure prevention could be effective measures to relieve the symptoms of psoriasis among these patients.

Abstract:Rationale: Both genetic and environmental factors contribute to lung cancer, but the degree to which air pollution modifies the impact of genetic susceptibility on lung cancer remains unknown. Measurements and Main Results: The results showed significant associations between the risk of lung cancer and PM2.5 (hazard ratio [HR], 1.63; 95% confidence interval [CI], 1.33-2.01; per 5 μg/m3), PM10 (HR, 1.53; 95% CI, 1.20-1.96; per 10 μg/m3), NO2 (HR, 1.10; 95% CI, 1.05-1.15; per 10 μg/m3), and NOx (HR, 1.13; 95% CI, 1.07-1.18; per 20 μg/m3). There were additive interactions between air pollutants and the genetic risk. Compared with participants with low genetic risk and low air pollution exposure, those with high air pollution exposure and high genetic risk had the highest risk of lung cancer (PM2.5: HR, 1.71; 95% CI, 1.45-2.02; PM10: HR, 1.77; 95% CI, 1.50-2.10; NO2: HR, 1.77; 95% CI, 1.42-2.22; NOx: HR, 1.67; 95% CI, 1.43-1.95). Conclusions: Long-term exposure to air pollution may increase the risk of lung cancer, especially in those with high genetic risk.

Abstract:Background: Evidence linking air pollutants and the risk of schizophrenia remains limited and inconsistent, and no studies have investigated the joint effect of air pollutant exposure and genetic factors on schizophrenia risk. Aims: To investigate how exposure to air pollution affects schizophrenia risk and the potential effect modification of genetic susceptibility. Method: Our study was conducted using data on 485 288 participants from the UK Biobank. Cox proportional hazards models were used to estimate the schizophrenia risk as a function of long-term air pollution exposure presented as a time-varying variable. We also derived the schizophrenia polygenic risk score (PRS) utilising data provided by the UK Biobank, and investigated the modification effect of genetic susceptibility. Results: During a median follow-up period of 11.9 years, 417 individuals developed schizophrenia (mean age 55.57 years, s.d. = 8.68; 45.6% female). Significant correlations were observed between long-term exposure to four air pollutants (PM2.5; PM10; nitrogen oxides, NOx; nitrogen dioxide, NO2) and the schizophrenia risk in each genetic risk group. Interactions between genetic factors and the pollutants NO2 and NOx had an effect on schizophrenia events. Compared with those with low PRS and low air pollution, participants with high PRS and high air pollution had the highest risk of incident schizophrenia (PM2.5: hazard ratio = 6.25 (95% CI 5.03-7.76); PM10: hazard ratio = 7.38 (95% CI 5.86-9.29); NO2: hazard ratio = 6.31 (95% CI 5.02-7.93); NOx: hazard ratio = 6.62 (95% CI 5.24-8.37)). Conclusions: Long-term exposure to air pollutants was positively related to the schizophrenia risk. Furthermore, high genetic susceptibility could increase the effect of NO2 and NOx on schizophrenia risk.

Abstract:Background Air pollution has been related to incidence of type 2 diabetes (T2D). We assessed the joint association of various air pollutants with the risk of T2D and examined potential modification by obesity status and genetic susceptibility on the relationship. Methods and findings A total of 449,006 participants from UK Biobank free of T2D at baseline were included. Of all the study population, 90.9% were white and 45.7% were male. The participants had a mean age of 56.6 (SD 8.1) years old and a mean body mass index (BMI) of 27.4 (SD 4.8) kg/m 2 . Ambient air pollutants, including particulate matter (PM) with diameters ≤2.5 μm (PM 2.5 ), between 2.5 μm and 10 μm (PM 2.5–10 ), nitrogen dioxide (NO 2 ), and nitric oxide (NO) were measured. An air pollution score was created to assess the joint exposure to the 4 air pollutants. During a median of 11 years follow-up, we documented 18,239 incident T2D cases. The air pollution score was significantly associated with a higher risk of T2D. Compared to the lowest quintile of air pollution score, the hazard ratio (HR) (95% confidence interval [CI]) for T2D was 1.05 (0.99 to 1.10, p = 0.11), 1.06 (1.00 to 1.11, p = 0.051), 1.09 (1.03 to 1.15, p = 0.002), and 1.12 (1.06 to 1.19, p < 0.001) for the second to fifth quintile, respectively, after adjustment for sociodemographic characteristics, lifestyle factors, genetic factors, and other covariates. In addition, we found a significant interaction between the air pollution score and obesity status on the risk of T2D ( p -interaction < 0.001). The observed association was more pronounced among overweight and obese participants than in the normal-weight people. Genetic risk score (GRS) for T2D or obesity did not modify the relationship between air pollution and risk of T2D. Key study limitations include unavailable data on other potential T2D-related air pollutants and single-time measurement on air pollutants. Conclusions We found that various air pollutants PM 2.5 , PM 2.5–10 , NO 2, and NO, individually or jointly, were associated with an increased risk of T2D in the population. The stratified analyses indicate that such associations were more strongly associated with T2D risk among those with higher adiposity.

Abstract:OBJECTIVE To explore the efficacy and safety of megestrol acetate silicone vaginal ring. METHODS A total of 165 healthy child-bearing age women were recruited to examine the efficacy, safety and satisfaction rate of megestrol acetate silicone vaginal ring. RESULTS The cumulative pregnancy rate of megestrol acetate silicone vaginal ring was 3.93%. And its discontinuation rate was 2.54%, the incidence of adverse events 11.52% and the incidence of adverse effects 1.81% while the symptoms were all slight. The bleeding pattern was similar to that of normal menstruation. Compared with baseline, the elevated level of triglycerides and the reduced level of low-density lipoprotein (LDL) were significantly different while no significant difference existed in high-density lipoprotein (HDL), cholesterol and hemoglobin, etc. The satisfaction rate of megestrol acetate silicone vaginal ring was 94.6% and increased with time. CONCLUSION Megestrol acetate silicone vaginal ring is an effective and safe option of contraceptives.
Associations of genetic risk factors and air pollution with incident hypertension among participants in the UK Biobank study

Abstract:The purposes of this study were to quantify the association of the combination of air pollution and genetic risk factors with hypertension and explore the interactions between air pollution and genetic risk. This study included 391,366 participants of European ancestry initially free from pre-existing hypertension in the UK Biobank. Exposure to ambient air pollutants, including particulate matter (PM2.5 PM2.5-10, and PM10), nitrogen dioxide (NO2) and nitrogen oxides (NOX), was estimated through land use regression modelling, and the associations between air pollutants and the incidence of hypertension were investigated using a Cox proportional hazards model adjusted for covariates. Furthermore, we established a polygenic risk score for hypertension and assessed the combined effect of genetic susceptibility and air pollution on incident hypertension. The results showed significant associations between the risk of hypertension and exposure to PM2.5 (hazard ratio [HR]: 1.41, 95% confidence interval [CI]: 1.29–1.53; per 10 μg/m3), PM10 (1.05, 1.00–1.09; per 10 μg/m3), and NOX (1.01, 1.01–1.02 per 10 μg/m3). Additive effects of PM2.5 and NOX exposure and genetic risk were observed. Compared to individuals with a low genetic risk and low air pollution exposure, participants with high air pollution exposure and a high genetic risk had a significantly increased risk of hypertension (PM2.5: 71% (66%–76%), PM10: 59% (55%–64%), NOX: 65% (60%–70%)). Our results indicate that long-term exposure to air pollution is associated with an increased risk of hypertension, especially in individuals with a high genetic risk.

Abstract:Abstract Inconsistent findings exist regarding the potential association between polluted air and Parkinson’s disease (PD), with unclear insights into the role of inherited sensitivity. This study sought to explore the potential link between various air pollutants and PD risk, investigating whether genetic susceptibility modulates these associations. The population-based study involved 312,009 initially PD-free participants with complete genotyping data. Annual mean concentrations of PM 2.5 , PM 10 , NO 2 , and NO x were estimated, and a polygenic risk score (PRS) was computed to assess individual genetic risks for PD. Cox proportional risk models were employed to calculate hazard ratios (HR) and 95% confidence intervals (CI) for the associations between ambient air pollutants, genetic risk, and incident PD. Over a median 12.07-year follow-up, 2356 PD cases (0.76%) were observed. Compared to the lowest quartile of air pollution, the highest quartiles of NO 2 and PM 10 pollution showed HRs and 95% CIs of 1.247 (1.089–1.427) and 1.201 (1.052–1.373) for PD incidence, respectively. Each 10 μg/m 3 increase in NO 2 and PM 10 yielded elevated HRs and 95% CIs for PD of 1.089 (1.026–1.155) and 1.363 (1.043–1.782), respectively. Individuals with significant genetic and PM 10 exposure risks had the highest PD development risk (HR: 2.748, 95% CI: 2.145–3.520). Similarly, those with substantial genetic and NO 2 exposure risks were over twice as likely to develop PD compared to minimal-risk counterparts (HR: 2.414, 95% CI: 1.912–3.048). Findings suggest that exposure to air contaminants heightens PD risk, particularly in individuals genetically predisposed to high susceptibility.

Abstract:BACKGROUND: Elderly-onset atopic dermatitis (AD) is a remarkable subtype and has been put on the agenda owing to its difficulty to control. Understanding the influence of genetic and environmental exposures is crucial to preventing elderly-onset AD.OBJECTIVES: To explore the association between genes and air pollution on incident elderly-onset AD.MATERIAL AND METHODS: This study was based on UK Biobank that recruited over 500,000 participants. The genetic risks were categorized into low, intermediate, and high groups according to tertiles of polygenic risk scores. Mixed exposure to various air pollutants was assessed using the weighted quantile sum (WQS) and also categorized based on tertiles. Within each genetic risk group, whether air pollutant mixture was associated with incident elderly-onset AD was estimated.RESULTS: 337,910 participants were included in the final analysis, and the mean age was 57.1. The median years for follow-up were 12.0, and the incident cases of AD were 2545. The medium and high air pollution mixture was significantly associated with incident AD compared with the low pollution group, with HRs of 1.182 (P = 0.003) and 1.359 (P < 0.001), respectively. In contrast, HR for medium and high genetic susceptibility was only 1.065 (P = 0.249) and 1.153 (P = 0.008). The population-attributable fraction of air pollution and genetic risk was 15.5 % (P < 0.001) and 6.4 % (P = 0.004). Additionally, compared with low genetic risk and low air pollution, high genetic risk and high air pollution was significantly associated with the incidence of elderly-onset AD with a HR of up to 1.523 (P < 0.001). There were no interactive effects between each group of genetic risks and air pollution. When grouped by sex, females could observe a stronger effect by genetic and air pollutant mixture exposure.CONCLUSION: Air pollution and genetics both independently enhance the risk of newly developed AD, and the effect of air pollutants is stronger than the investigated genes.

Abstract:Introduction: Psoriasis is a common skin disease that seriously affects patients' quality of life. The association of air pollutants with psoriasis, and the extent of their effects remains unclear. Methods: Based on a distributed lag non-linear model, this study explored the short-term effects of air pollutants on outpatients with psoriasis in Hefei, China, between 2015 and 2019 by analyzing the exposure–lag–response relationship, after controlling for confounding influences such as meteorological factors, long-term trends, day of the week, and holidays. Stratified analyses were performed for patients of different ages and genders. Results: The maximum relative risks of psoriasis outpatients' exposure to SO 2 , NO 2 , and O 3 were 1.023 (95% confidence intervals (CI): 1.004–1.043), 1.170 (95% CI: 1.046–1.307), and 1.059 (95% CI: 1.030–1.090), respectively. An increase of 10 μ g / m 3 of NO 2 was associated with a 2.1% (95% CI: 0.7–3.5%) increase in outpatients with psoriasis, and a decrease of 10 μ g / m 3 of O 3 was associated with an 0.8% (95% CI: 0.4–1.2%) increase in outpatients with psoriasis. Stratified analyses showed that male subjects were more sensitive to a change in meteorological factors, while female subjects and outpatients with psoriasis aged 0–17 years old were more sensitive to a change in air pollutants. Discussion: Short-term air pollutant exposures were associated with outpatients having psoriasis, suggesting that patients and high-risk people with psoriasis should reduce their time spent outside and improve their skin protection gear when air quality is poor.

Abstract:BackgroundAmbient fine particulate matter (PM2.5) adversely affects human health and has been linked to a variety of skin disorders. However, little is known about the effects of PM2.5 on psoriasis. MethodsThe Beijing Medical Claim Data for Employees database recorded 500,266 outpatient visits for psoriasis during 2010-2017. A generalized additive quasi-Poisson model was used to examine the relationship between daily PM2.5 concentrations and outpatient visits for psoriasis with stratification by sex, age, and season. ResultsShort-term exposure to PM2.5 was associated with outpatient visits for psoriasis-related health concerns. A same-day increase of 10 mu g/m(3) in PM2.5 concentrations was associated with a 0.29% (95% confidence interval: 0.26-0.32%) increase in daily outpatient visits for psoriasis. Female and older patients appeared to be more sensitive to the effects of PM2.5 (P < 0.05). ConclusionsShort-term elevations in PM2.5 concentrations may be associated with exacerbations in psoriasis. Further work is warranted to confirm the findings and elucidate the underlying biological mechanisms.
