Abstract:The emerging of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)caused COVID-19 pandemic.The first case of COVID-19 was reported at early December in 2019 in Wuhan City,China.To examine specific antibodies against SARS-CoV-2 in biologi-cal samples before December 2019 would give clues when the epidemic of SARS-CoV-2 might start to circulate in populations.We obtained all 88,517 plasmas from 76,844 blood donors in Wuhan between 1 September and 31 December 2019.We first evaluated the pan-immunoglobin(pan-Ig)against SARS-CoV-2 in 43,850 samples from 32,484 blood donors with suitable sample quality and enough volume.Two hundred and sixty-four samples from 213 donors were pan-Ig reactive,then further tested IgG and IgM,and validated by neutralizing antibodies against SARS-CoV-2.Two hundred and thirteen samples(from 175 donors)were only pan-Ig reactive,8(from 4 donors)were pan-Ig and IgG reactive,and 43(from 34 donors)were pan-Ig and IgM reactive.Microneutralization assay showed all negative results.In addition,213 screened reactive donors were analyzed and did not show obviously temporal or regional tendency,but the distribution of age showed a difference compared with all tested donors.Then we reviewed SARS-CoV-2 antibody results from these donors who donated several times from September 2019 to June 2020,partly tested in a previous published study,no one was found a significant increase in S/CO of antibodies against SARS-CoV-2.Our findings showed no SARS-CoV-2-specific antibodies existing among blood donors in Wuhan,China before 2020,indicating no evidence of transmission of COVID-19 before December 2019 in Wuhan,China.
Seroprevalence of SARS-CoV-2 antibodies among healthy blood donors: a systematic review and meta-analysis

Abstract:Introduction: The development of a potent immune response and antibodies against SARS-CoV-2 is important for herd immunity. The serological response may be due to a previous infection or vaccination. Healthy blood donors could represent and provide information on the immune status of the general population. Therefore, we estimated the global and regional prevalence of SARS-CoV-2 antibodies among healthy blood donors. Methods: We conducted a systematic search of PubMed, Scopus, and ProQuest from December 2019 to January 2023. After critical appraisal and quality assessment, a qualitative synthesis of the identified relevant articles was performed. The random-effects model was used to estimate the pooled prevalence of SARS-CoV-2 antibodies. Funnel plots and Egger's test were used to assess publication bias. Sensitivity analysis was performed, and heterogeneity was quantified using I2 statistics. Results: A total of 70 peer-reviewed articles were selected that together included 2,454,192 blood donors. The global estimated pooled prevalence of SARS-CoV-2 antibodies among healthy blood donors was 10.3% (95% CI: 4.6 - 18.0%, n = 70). The highest seroprevalence was observed in Asia (17.7%), followed by Africa (16.1%). The seroprevalence in studies conducted before the introduction of the vaccine was 6.1%, whereas those of studies conducted after vaccines were available was 27.6%. High seroprevalence was observed in studies that measured antibodies against the S protein of the virus (15.2%), while lower (12.5%) in those that measured antibodies against the N protein. A high seroprevalence was observed in studies that only measured IgG antibodies (16.3%), and it was 5.9% in studies that measured total antibodies. Conclusion: The prevalence of SARS-CoV-2 antibodies among healthy blood donors varies, potentially indicating geographical disparities in transmission and vaccination rates. To enhance community resilience, addressing these differences through inclusive health policies and adaptable public health measures is crucial.

Abstract:Introduction: Faith and hypnosis methods are applied in many experimental cancer treatments. Practices of religious and psychological healing have a major role in the recovery of tumors. Healing warts by prayer has been used for years, particularly in the Turkish society. The Hypothesis: This hypothesis maintains that believing is one of the key concepts in the recovery of tumors and also focuses on the effect of believing on the recovery of warts. Evaluation Hypothesis: Believing people, nerve, endocrine and immune system is activated. NK and T cells in the periphery through destruction and healing takes place in all tissues. According to our hypothesis here with us for is to believe the actual trigger. Both the prayer and the hypnosis method, in terms of cancer improvement, have been shown in many publications in the literature and have brought a new dimension to cancer recovery. Conclusion: We hypothesize that believing (prayer or hypnosis) can be effective of the healing of warts through the stimulation of the immune system.
Increased parvovirus B19 seropositivity in healthy blood donors in India

Abstract:A core component of every blood program is the supply of safe blood and blood products. The elevated risk of transmission through these products is due to parvovirus B19 (B19V) resistance to the virus inactivation procedures. Our study aimed to screen asymptomatic blood donors for B19V at a tertiary care hospital in Chennai, Tamil Nadu, between September 2020 and June 2021. Sera from 106 healthy blood donors who tested negative for Human immunodeficiency virus (HIV), Hepatitis B surface antigen (HBsAg), Hepatitis C virus (HCV), syphilis, and malaria were tested for anti-B19V IgM and IgG using a qualitative indirect enzyme-linked immunosorbent assay (ELISA). In the study population, 23.5% (n = 25) of donors tested IgM positive, 38.6% (n = 41) tested IgG positive, and 7.5% (n = 8) tested positive for both IgM and IgG. A proportion of 61.3% (n = 65) of the blood donors tested IgG negative, suggesting they had no past B19V infection. B19V DNA was not detected in any of the subjects. The high seroprevalence of IgM indicates that blood donors may have been recently exposed to B19V, potentially posing a risk to immunocompromised individuals and those with hematological stress. Further longitudinal studies with a larger sample size are recommended to better understand the risk of B19V transfusion transmission.

The evolution and interpretation of seroprevalence of SARS-CoV-2 antibodies among South African blood donors from the Beta to Omicron variant-driven waves

Abstract:Background and objectives: Confirmed COVID-19 diagnoses underestimate the total number of infections. Blood donors can provide representative seroprevalence estimates, which can be leveraged into reasonable estimates of total infection counts and infection fatality rate (IFR). Materials and methods: Blood donors who donated after each of three epidemic waves (Beta, Delta and first Omicron waves) were tested for anti-SARS-CoV-2 nucleocapsid antibodies using the Roche Elecsys anti-SARS-CoV-2 total immunoglobulin assay. Roche Elecsys anti-spike antibody testing was done for the post-Omicron sampling. Prevalence of antibodies was estimated by age, sex, race and province and compared to official case reporting. Province and age group-specific IFRs were estimated using external excess mortality estimates. Results: The nationally weighted anti-nucleocapsid seroprevalence estimates after the Beta, Delta and Omicron waves were 47% (46.2%-48.6%), 71% (68.8%-73.5%) and 87% (85.5%-88.4%), respectively. There was no variation by age and sex, but there were statistically and epidemiologically significant differences by province (except at the latest time point) and race. There was a 13-fold higher seroprevalence than confirmed case counts at the first time point. Age-dependent IFR roughly doubled for every 10 years of age increase over 6 decades from 0.014% in children to 6.793% in octogenarians. Conclusion: Discrepancies were found between seroprevalence and confirmed case counts. High seroprevalence rates found among Black African donors can be ascribed to historical inequities. Our IFR estimates were useful in refining previous large disagreements about the severity of the epidemic in South Africa. Blood donor-based serosurveys provided a valuable and efficient way to provide near real-time monitoring of the ongoing SARS-CoV-2 outbreak.

Abstract:Abstract In this study, we investigate the seroprevalence of SARS-CoV-2 antibodies among blood donors in the cities of Wuhan, Shenzhen, and Shijiazhuang in China. From January to April 2020, 38,144 healthy blood donors in the three cities were tested for total antibody against SARS-CoV-2 followed by pseudotype SARS-CoV-2 neutralization tests, IgG, and IgM antibody testing. Finally, a total of 398 donors were confirmed positive. The age- and sex-standardized SARS-CoV-2 seroprevalence among 18–60 year-old adults (18–65 year-old in Shenzhen) was 2.66% (95% CI: 2.24%–3.07%) in Wuhan, 0.033% (95% CI: 0.0029%–0.267%) in Shenzhen, and 0.0028% (95% CI: 0.0001%–0.158%) in Shijiazhuang, respectively. Female sex and older-age were identified to be independent risk factors for SARS-CoV-2 seropositivity among blood donors in Wuhan. As most of the population of China remained uninfected during the early wave of the COVID-19 pandemic, effective public health measures are still certainly required to block viral spread before a vaccine is widely available.

Abstract:Background Blood transfusion is an essential and lifesaving procedure for many acute and chronic diseases. Though saving millions of lives, it carries the risk of transfusion of transfusion-transmitted infections (TTIs), including hepatitis B. Detection of this infection prior to transfusion saves potentially vulnerable patients from an additional burden and prevents the further spread of disease. Aim and objectives Our present study aimed to determine the seroprevalence of hepatitis B virus (HBV) in blood donors at the Rajendra Institute of Medical Sciences (RIMS) in Jharkhand, a tribal-preponderant state of India. Materials and methods After obtaining approval from the institutional ethics committee, a retrospective observational study was conducted among the eligible blood donors visiting RIMS from April 2016 to March 2023. A total of 195,507 subjects were included in the study. All blood donation samples collected in ethylenediaminetetraacetic acid (EDTA) vials were tested for five TTIs: human immunodeficiency virus 1 and 2, HBV, hepatitis C virus (HCV), malaria and syphilis. HBV testing was conducted via chemiluminescence technique to check f or the presence of hepatitis B surface antigen (HBsAg) in plasma. Results Among the study sample of 195,507 donors, the prevalence of HBsAg positivity was 0.87%. Among all the TTIs, more than 50% (51.93%) were HBsAg positive. The positivity percentage was higher in male donors and HBsAg positivity rose with an increase in replacement donors. Conclusions HBV is a major health concern in developing countries such as India due to its high endemicity. Therefore, early detection of HBV carriers in the blood donor population helps in curbing the spread of further infection and it also helps policymakers to develop different health programs to reduce further incidence of the infection in the general population.

Abstract:Objectives: We aimed to evaluate the duration and breadth of antibodies elicited by inactivated COVID-19 vaccinations in healthy blood donors. Methods: We performed serological tests on 1,417 samples from 658 blood donors who received two (n=357), or three (n=301) doses of COVID-19 inactivated vaccine. We also accessed the change in antibody response before and after booster vaccination in 94 participants and their neutralization breadth to the current variants after the booster. Results: Following vaccination, for either the 2- or 3-dose, the neutralizing antibodies (nAbs) peaked with about 97% seropositivity approximately within one month but subsequently decreased over time. Of plasmas collected 6-8 months after the last immunization, the nAb seropositivities were 37% and 85% in populations with 2-dose and 3-dose vaccinations, respectively. The nAbs of plasma samples (collected between 2-6 weeks after the 3 rd dose) from triple-vaccinated donors (n=94) showed a geometric mean titer of 145.3 (95% CI: 117.2 to 180.1) against the ancestral B.1, slightly reduced by 1.7-fold against Delta variant, but markedly decreased by 4-6 fold in neutralizing Omicron variants, including the sub-lineages of BA.1 (5.6-fold), BA.1.1 (6.0-fold), BA.2 (4.2-fold), B.2.12.1 (6.2-fold) and BA.4/5 (6.5-fold). Conclusion: These findings suggested that the 3 rd dose of inactivated COVID-19 vaccine prolongs the antibody duration in healthy populations, but the elicited-nAbs are less efficient in neutralizing circulating Omicron variants.

Abstract:Background The COVID‐19 pandemic has major implications on the entire blood supply system worldwide. Seroepidemiological studies are certainly necessary for better understanding the global burden that the COVID‐19 pandemic represents. Objectives In this study, we analysed the association between demographic factors, COVID‐19 severity, vaccination status and the reactivity of anti‐SARS‐CoV‐2 IgG antibodies in Serbian blood donors. Materials and Methods In a prospective study, demographic data and data related to previous SARS‐CoV‐2 infection, COVID‐19 severity and vaccination status among whole blood donors were analysed, from February 10 to August 10, 2022, at the Blood Transfusion Institute of Vojvodina, Serbia. The detection and determination of the level of anti‐SARS‐CoV‐2 IgG antibodies were performed using LIAISON® SARS‐CoV‐2 TrimericS IgG immunoassay. Results A total of 1190 blood donors were included, 24.5% were female and 75.5% were male while their average age was 41 years. Anti‐SARS‐CoV‐2 antibody values ranged from 2.40 to 3120 BAU/ml with a mean value of 1354.56 BAU/ml. Statistical analysis showed that COVID‐19 severity and vaccination status are linked with reactivity of anti‐SARS‐CoV‐2 antibodies, while gender and age of voluntary blood donors are not related to the values of anti‐SARS‐CoV‐2 antibodies. Conclusion The values of anti‐SARS‐CoV‐2 antibodies in voluntary blood donors in Serbia are kept relatively high, especially in blood donors who have overcome the severe COVID‐19, as well as in donors who have been vaccinated against COVID‐19. Further SARS‐CoV‐2 seroprevalence studies in our country are certainly still necessary so global strategies to fight against COVID‐19 would be adequately evaluated.

Abstract:CONTEXT: Alloimmunization by foreign red cell antigens is a matter of concern as it may lead to hemolysis in transfused patients as well as fetus of pregnant females. AIMS: This study aimed to perform a comparative analysis of prevalence and type of irregular antibodies in healthy donors, vis-a-vis blood transfusion recipients. SETTINGS AND DESIGN: Blood samples of 4000 individuals comprising healthy donors, exposed patients, and nonexposed patients were collected and were analyzed for irregular antibodies. MATERIALS AND METHODS: Commercially available three-cell antigen panel was used for the antibody screening. The samples positive in antibody screen were further subjected to an extended 11-cell panel for antibody identification in low-ionic strength saline with and without enzyme. STATISTICAL ANALYSIS: Statistical analysis was done using SPSS for Windows 15.0 program. Chi-square test was used for detecting statistical significance of exposure to red blood cell antigens in the formation of alloantibodies. RESULTS: Of the 4000 samples, antibodies were identified in 105 (2.6%) samples. Overall, nonexposed group showed a seropositivity of 0.36%, while the exposed group showed a seropositivity of 9.4%. Anti-D was the most common antibody found in 38 patients (33.3%). Anti-E was the most common antibody in males, while anti-D was the most common antibody in females. CONCLUSIONS: Since the risk of alloimmunization is more common in multitransfused patients, it is advisable to screen at least those cases for irregular antibodies.

Abstract:Objective: To investigate the rate of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody among unvaccinated voluntary blood donors in Chongqing, and to provide evidence for epidemic surveillance. Methods: A total of 10,208 voluntary blood donors from January 5 to January 20, 2021, in the Chongqing area were collected, and the SARS-CoV-2 immunoglobulin (Ig) G and IgM antibodies were detected by chemiluminescence, and the differences of antibody-positive rate in different gender, age, ABO blood group, and different risk areas were analyzed. Results: Among 10208 blood donors, 10 were found to be positive for SARS-COV-2 IgG antibody, giving a positivity rate of SARS-COV-2 IgG at 0.10%, and 29 were positive for SARS-CoV-2 IgM antibody, with a positivity rate of SARS-CoV-2 IgM at 0.28%. There was no statistical difference in the positive rate of antibody among different genders, ages, and ABO blood types, but it was related to the number of confirmed coronavirus disease 2019 (COVID-19) cases in each city. Conclusions: The SARS-CoV-2 seroprevalence rate in Chongqing was low and correlated with the number of confirmed COVID-19 cases.

Abstract:Blood safety is a critical aspect of healthcare systems worldwide involving rigorous screening, testing, and processing protocols to minimize the risk of transfusion-transmitted infections (TTIs). The present study offers a comprehensive assessment of the prevalence of hepatitis B virus (HBV), hepatitis C virus (HCV), human immunodeficiency virus (HIV), and syphilis among blood donors in southern Thailand. It explores the consequences of the COVID-19 pandemic on the blood transfusion service, donor characteristics, and the prevalence of TTIs. A retrospective analysis of 65,511 blood donors between 2018 and 2022 was conducted at Songklanagarind Hospital, Thailand. The socio-demographic characteristics of the donors were examined using the Chi-square test to assess the relationship between TTIs serological positivity and donor characteristics. The donors were divided into pre-COVID-19 (2018–2019) and during COVID-19 (2020–2022) groups to evaluate the impacts of COVID-19. The study found that HBV had the highest overall prevalence at 243 per hundred thousand (pht), followed by syphilis (118 pht), HCV (32 pht), and HIV (31 pht) over a five-year period of study. After COVID-19, the prevalence of HBV decreased by 21.8%; HCV decreased by 2.1%; HIV increased by 36.4%; and syphilis increased by 9.2%. The socio-demographic characteristics and TTIs prevalence were significantly altered over time. This study provides insights into blood donor characteristics and TTIs prevalence in southern Thailand, highlighting the understanding of the impact of COVID-19 on the spread of TTIs.

Detection of anti-nucleocapsid antibodies associated with primary SARS-CoV-2 infection in unvaccinated and vaccinated blood donors

Abstract:Anti-nucleocapsid (N) antibody assays can be used to estimate SARS-CoV-2 infection prevalence in regions employing anti-spike based COVID-19 vaccines. However, poor sensitivity of anti-N assays in detecting infections after vaccination (VI) has been reported. To support serological monitoring of infections, including VI, in a large blood donor cohort (N=142,599), we derived a lower cutoff for identifying previous infection using the Ortho VITROS Anti-SARS-CoV-2 Total-N Antibody assay, improving sensitivity while maintaining specificity >98%. Sensitivity was validated in samples donated after self-reported infections diagnosed by a swab-based test. Sensitivity for first infections in unvaccinated donors was 98.1% (95% CI: 98.0,98.2) and for VI was 95.6% (95.6,95.7), using the standard cutoff. Regression analysis showed sensitivity was reduced in the Delta compared to Omicron period, in older donors, asymptomatic infections, ≤30 days after infection and for VI. The standard Ortho anti-N threshold demonstrated good sensitivity, which was modestly improved with the revised cutoff.

Abstract:AIM: The present study was undertaken with an aim to investigate the sero prevalence of HIV and hepatitis in blood donors. Introduction: Blood transfusion constitutes of an important part of patient care, as up to 10% of hospitalized patients are transfused. But unsafe blood transfusion have the potential to transmit a diversity of infections to blood recipients. These infections could be due to viruses, bacteria, protozoans , and/or prions . Viral agents include human immunodeciency virus (HIV) , Hepatitis B virus (HBV) and Hepatitis C virus (HCV). Material And Methods: The present study was undertaken with an aim to investigate the sero-prevalence of HIV and hepatitis in blood donors. This is a cross sectional study of 200 blood donors in of Acharya shree Chandra college of medical sciences and hospital with age ranging from 18 to 49 years from time period October 2023 to March 2024. Result: Out of 200 donors, only 18 were positive for hepatitis.( 13 male and 5 female). Also 8 out of 200 were positive for HIV ,out of which 8 were male and 1 was female. It is Conclusion: necessary to estimate the prevalence of infections in the donor community using more sensitive and specic tests than the early tests used.
Seroprevalence of transfusion-transmissible infections among family replacement donors and voluntary non-remunerated blood donors during the COVID-19 pandemic in sub Saharan Africa

Abstract:Introduction : According to WHO, regular, voluntary, unpaid blood donors are the safest group of donors, as they have the lowest prevalence of blood transmitted infections. However, family/replacement blood donors is widely used in sub Saharan Africa and this practice was exacerbated during the COVID 19 pandemic. This study aimed to compare the seroprevalence of infectious markers in family replacement blood donors and voluntary non-remunerated blood donors during the COVID 19 pandemic in a country of sub Saharan Africa. Materials and Methods Blood donors received at the National Centre of Blood Transfusion ( NBTC) of Dakar from August 1 st to October 31 th 2021, were included in this study. All donors completed a pre-donation questionnaire. Donors identity, epidemiological parameters, reasons for donation and laboratory results were collected in the Inlog ® software of the NBTC. The serological tests for HBV, HIV and HCV were performed with chemiluminescence technology. The Rapid Plasma Reagent test was used to find out treponemal antibodies. The determination of ABO and Rh blood groups was performed using monoclonal antisera following classical hemagglutination test on a plate. Results A total of 5002 donors were collected during this COVID-19 pandemic period. Blood family/replacement donors represented 54.0% and new voluntary donors represented 52.6%. Comparison of HIV, HCV and syphilis markers seroprevalence showed no statistically significant difference between new voluntary donors and new family replacement donors ( p>0.05 ). However, for HBV the seroprevalence was significantly higher in new family replacement donors ( p=0,002 ). Conclusion The proper supply of blood was impacted by the COVID-19 pandemic meanwhile replacement donations had contributed to limiting the damage observed with blood shortages. However, the significant differences noted on the seroprevalences of transfusion-transmissible infections between voluntary non-paid donors and family/replacement donors strengthens WHO recommendations for the selection of volunteer non-paid donors to lower transfusion-transmissible HBV in sub Saharan Africa.

Abstract:Background: Much increasing evidence has suggested that long-term complications post vaccination of SARS-CoV-2 experience a wide range of complication including diabetes. The risk and burden of type 1 diabetes is extensively reported, but type 2 diabetes mellitus (T2D) has yet to be characterized. To address this gap, we aimed to examine trends of long-term complications post SARS-CoV-2 infection and vaccination in diabetes incidence among the Saudi population. Methods: In this cross-sectional hospital-based study, we analyzed the blood profile of first-time blood donors from the University Hospital of King Abdulaziz University, Jeddah. Saudi Arabia. Various blood parameters, HbA1c was measured in the month of May 2023. All the donors were non-diabetic and were never diagnosed with T2D before the current blood donation. 203 healthy subjects donated their blood, out of which 104 had abnormally high HbA1c tending towards diagnosis of T2D and 99 had with blood profiles. The study followed the STROBE reporting guidelines. Results: Out of 203 donors 104 (male 50(48.1%), female 54(51.9%)) were diagnosed with increased HbA1c (8.24 in males) compared to 7.61 of HbA1c in females. 35.6% were above ˃65 years, with 52.9% with O+ from the ABO blood group. Liver functions indicated significant p˂0.05, 0.04, increased amount of GGT (46.47 U/L), Alkaline phosphatase (99.93 ±64.26 uL) respectively in HbA1c elevated donors KFT represented significant p˂0.05, 0.02 elevated levels of urea (6.73 ±5.51 mmol/L), creatinine (129.97 ±195.17 umol/L) respectively along with elevated values of Lactate dehydrogenase (LDH) (263.72± 196.70 uL) and triglycerides (1.66 ±0.74mmol/L) when compared to normal value of HbA1c donors. Discussion: In the present cross-sectional study, significant increase in HbA1c, trending towards increased cases of T2D post SARS-CoV-2 infection and vaccination. Males are much affected compared to females. Further maximum number of cases were from donors above the age of 65 years with altered partial LFT (GGT, Alkaline phosphatase), KFT (urea, creatinine), lipid profile (TG) and LDH in post SARS-CoV-2 and vaccination blood donors. Conclusion: Increase in HbA1c in 50% of donors, irrespective of gender, is an alarming figure for health authorities, with altered LFT, KFT and LDH tests and, in the near future, may increase the incidence of T2D. Large-scale population-based studies are required to prevent future incidences of T2D in young children who will be vaccinated.

Abstract:Background: As per the recommendation of the United States Food and Drug Administration, more research is needed to determine the antibody titer against COVID-19 vaccination. background: As per the recommendation of the United States Food and Drug Administration, more research is needed to determine the antibody titer against COVID-19 vaccination. Objective: The study aimed to understand the relationship between the antibody titer to the demographics, infection severity, and cycle threshold (CT) values of confirmed COVID-19 patients. Methods: Initially, we obtained consent from 185 populations and included sixty RT-PCRpositive COVID-19 patients from Kamrup District in the Northeast State of Assam, India. The vaccination status was recorded and tested for the level of serum immunoglobulin (IgG). The CT values, gender, and clinical symptoms-based scoring (CSBS) correlated with their IgG value. Results: Around 48% of participants gained an antibody titer more than the threshold value and showed CT values between 18-25. Moreover, the maximum distributed score above the average was found between the CT values 18-25.
