Abstract:Bluetongue (BT) is a severe arboviral disease affecting sheep, cows, and other wild ruminants, caused by the Bluetongue virus (BTV). The virus has evolved into over 32 serotypes, rendering existing vaccines less effective. While the structural proteins of this virus represent promising targets for vaccine development, they unfortunately exhibit high amino acid polymorphism and are laden with numerous inhibitory epitopes. Structural proteins such as VP1 and VP7 are highly conserved and may contain epitopes capable of triggering cross-reactive cell-mediated immunity (CMI). In this study, we identified highly conserved MHC-I and -II-restricted T cell epitopes within VP1, VP5, and VP7 BTV proteins and developed an effective -immuno-informatics-based broad-spectrum BT multiepitope vaccine for bovine and laboratory mouse systems. The conserved epitopes utilized in the vaccines are highly antigenic, non-allergenic, non-toxic, and capable of inducing IFN- (only CD4+ T cell epitopes). Both mouse and bovine vaccines were tethered with Toll-like receptor (TLR)-4-agonist adjuvants, beta-defensin 2, and the 50s ribosomal unit to stimulate innate immunity for CMI development. Protein-protein docking analysis revealed strong binding affinities, while extensive 100-nanosecond molecular dynamics simulations indicated stable complexes between the vaccine structures and TLR4. Vaccination simulation studies demonstrated their ability to trigger proinflammatory responses. Therefore, these novel vaccine designs necessitate further exploration through wet lab experiments to evaluate their immunogenicity, safety, and effectiveness for practical deployment in livestock.

Abstract:The bluetongue virus (BTV), transmitted by biting midges, poses a significant threat to livestock globally. This orbivirus induces bluetongue disease, leading to substantial economic losses in the agricultural sector. The current control measures have limitations, necessitating the development of novel, efficient vaccines. In this study, an immunoinformatics approach is employed to design a multi-epitope subunit vaccine for Ovis aries targeting six BTV serotypes. Focusing on the VP2 capsid protein, the vaccine incorporates B-cell, helper-T lymphocytes (HTL), and cytotoxic T-cell lymphocytes (CTL) epitopes. Molecular docking reveals stable interactions with TLR2 and TLR4 receptors, suggesting the stability of the complex, indicating the potential viability of the multi-epitope vaccine. The computational approach offers a rapid and tailored strategy for vaccine development, highlighting potential efficacy and safety against BTV outbreaks. This work contributes to understanding BTV and presents a promising avenue for effective control.

Abstract:Abstract Bovine coronavirus (BCoV) poses a significant threat to the global cattle industry, causing both respiratory and gastrointestinal infections in cattle populations. This necessitates the development of efficacious vaccines. While several inactivated and live BCoV vaccines exist, they are predominantly limited to calves. The immunization of adult cattle is imperative for BCoV infection control, as it curtails viral transmission to calves and ameliorates the impact of enteric and respiratory ailments across all age groups within the herd. This study presents an in silico methodology for devising a multiepitope vaccine targeting BCoV. The spike glycoprotein (S) and nucleocapsid (N) proteins, which are integral elements of the BCoV structure, play pivotal roles in the viral infection cycle and immune response. We constructed a remarkably effective multiepitope vaccine candidate specifically designed to combat the BCoV population. Using immunoinformatics technology, B-cell and T-cell epitopes were predicted and linked together using linkers and adjuvants to efficiently trigger both cellular and humoral immune responses in cattle. The in silico construct was characterized, and assessment of its physicochemical properties revealed the formation of a stable vaccine construct. After 3D modeling of the vaccine construct, molecular docking revealed a stable interaction with the bovine receptor bTLR4. Moreover, the viability of the vaccine’s high expression and simple purification was demonstrated by codon optimization and in silico cloning expression into the pET28a (+) vector. By applying immunoinformatics approaches, researchers aim to better understand the immune response to bovine coronavirus, discover potential targets for intervention, and facilitate the development of diagnostic tools and vaccines to mitigate the impact of this virus on cattle health and the livestock industry. We anticipate that the design will be useful as a preventive treatment for BCoV sickness in cattle, opening the door for further laboratory studies.
In silico design of a multi-epitope vaccine against tick-borne encephalitis virus via immunoinformatic analysis

Abstract:Tick-borne encephalitis virus (TBEV) is a serious pathogen that poses a significant threat to humans, causing encephalitis that can result in lifelong sequelae. In this study, we focused on the complete proteomes of the five current TBEV subtypes to identify dominant epitopes. Immunoinformatics tools were employed to screen for LBL, HTL, and CTL epitopes. These epitopes were then linked using various linkers and combined with adjuvants and histidine tag. The vaccine underwent a series of physicochemical property analyses, including secondary structure prediction, three-dimensional structure prediction, molecular docking, molecular dynamics simulation, immune simulation, and in silico cloning. The results indicate that the vaccine is highly conserved, strongly immunogenic, stable, non-allergenic, and non-toxic. Molecular docking and molecular dynamics simulation demonstrate that the vaccine can form a stable binding complex with TLR3. Immune simulation analysis shows that the vaccine effectively stimulates both cellular and humoral immune responses, accompanied by an increase in cytokine titers. Furthermore, through codon optimization and in silico cloning, the vaccine can be stably and effectively expressed in the Escherichia coli system. As an effective candidate for TBEV vaccination, the multi-epitope vaccine developed in this study has promising application prospects and provides a new approach for the research, development, and improvement of vaccines targeting TBEV.

Abstract:Introduction: BCoV is one of the significant causes of enteritis in young calves; it may also be responsible for many respiratory outbreaks in young calves. BCoV participates in the development of bovine respiratory disease complex in association with other bacterial pathogens. Our study aimed (1) to map the immunogenic epitopes (B and T cells) within the major BCoV structural proteins. These epitopes are believed to induce a robust immune response through the interaction with major histocompatibility complex (MHC class II) molecules (2) to design some novel BCoV multiepitope-based vaccines. Materials and methods: The goal is achieved through several integrated in silico prediction computational tools to map these epitopes within the major BCoV structural proteins. The final vaccine was constructed in conjugation with the Choleratoxin B toxin as an adjuvant. The tertiary structure of each vaccine construct was modeled through the AlphaFold2 tools. The constructed vaccine was linked to some immunostimulants such as Toll-like receptors (TLR2 and TLR4). We also predicted the affinity binding of these vaccines with this targeted protein using molecular docking. The stability and purity of each vaccine construct were assessed using the Ramachandran plot and the Z-score values. We created the in silico cloning vaccine constructs using various expression vectors through vector builder and Snap gene. Results and discussion: The average range of major BCoV structural proteins was detected within the range of 0.4 to 0.5, which confirmed their antigen and allergic properties. The binding energy values were detected between -7.9 and -9.4 eV and also confirmed their best interaction between our vaccine construct and Toll-like receptors. Our in silico cloning method expedited the creation of vaccine constructs and established a strong basis for upcoming clinical trials and experimental validations. Conclusion: Our designed multiepitope vaccine candidates per each BCoV structural protein showed high antigenicity, immunogenicity, non-allergic, non-toxic, and high-water solubility. Further studies are highly encouraged to validate the efficacy of these novel BCoV vaccines in the natural host.

Abstract:Serious outbreaks of bluetongue, an arbovirus of domestic and wild ruminants caused by bluetongue virus serotypes (BTV), have occurred around the world. More than 27 distinct serotypes are recognized throughout the world. A new virus, BTV-25 (Toggenburg orbivirus [TOV]), was first detected in Switzerland, and has not yet been found in China. VP2 is an important outer shell protein that defines BTV serotypes and is, therefore, an ideal target antigen for serotype identification. To produce a monoclonal antibody against VP2 of BTV-25, the segment 2 gene was divided into three segments, cloned into pET-28a (+) and pMAL-c5X vectors, and the protein was expressed in E. coli BL21 with different tags. Monoclonal antibodies (mAbs) were prepared by using the purified His-25A, 25B, 25C proteins as the immunogen and the purified MBP-25A, 25B, 25C proteins as the detection antigen. Twelve hybridoma cell lines stably secreting mAbs against different VP2 segments of BTV-25 were produced. The segment 2 gene was cloned into pFastBac™HT B vector and a positive recombinant plasmid pFastBac-VP2 was used to identify mAbs. The recombinant baculovirus BACV-VP2 and eukaryotic expression of protein VP2 were obtained by the recombinant bacmid BAC-VP2 transfected Sf9 insect cells; western blotting showed that only eight mAbs were reactive. Finally, we identified the epitopes of VP2 recognized by three specific mAbs (25A-2B6, 25B-2G3, 25C-4B2) using phage display technology. The linear epitopes of VP2 protein were "359LYP361", "580NT581", "620TFR622". The preparation of mAbs and identification of the epitopes provided a foundation to analyze VP2, and may assist in the serological diagnosis of BTV-25.

Abstract:Comparative hemagglutination-enhancement (HE) tests demonstrated diversified patterns of antigenic specificities both in the fiber and vertex capsomer part of pentons of human adenovirus types 3, 11 (subgroup I), 9, 15 (II), 1, 2, 4, 5, 6 (III), and 12. All fibers contained a type-specific antigen. Subgroup II and III fibers, in addition, contained specificities both unique for each subgroup and also common to the two subgroups. Fibers of serotypes 4 and 12 displayed a somewhat deviating behavior. All vertex capsomers tested shared a group-specific part. This was the only antigenic specificity demonstrable for serotype 12. Maximal penton HE titers of all sera were reached in tests with incomplete hemagglutinin of type 11. In addition, maximal HE activity of sera against individual serotypes also was recorded against pentons of other members of the same subgroup. Antigen characteristics of vertex capsomers of type 4 indicated a closer relationship to subgroup I than to subgroup III. The toxin activity of pentons was more sensitive to trypsin treatment than their capacity to function as incomplete hemagglutinin. Homotypic antipenton sera, unabsorbed or absorbed with homotypic fibers to remove antibodies against this component, and, to a varying extent, also heterotypic antipenton sera could neutralize toxin activity. Antifiber sera could neutralize toxin activity of pentons carrying short fibers (10 nm, type 3) but not of those carrying long fibers (28 to 31 nm, type 2). It is concluded that toxin activity is carried by a specific part of vertex capsomers and that cell detachment can be brought about via a direct contact between this component and cell membranes. Fiber-mediated attachment does not seem to be necessary for this biological activity to become expressed.

Abstract:As Bacille Calmette-Guérin (BCG) vaccine's effectiveness is limited to only children, the development of new tuberculosis (TB) vaccines is being studied using several platforms, and a novel TB vaccine that overcomes this limitation is required. In this study, we designed an effective multi-epitope vaccine against Mycobacterium tuberculosis using immunoinformatic analysis. First, we selected 11 highly antigenic proteins based on previous research: Ag85A, Ag85B, Ag85C, ESAT-6, MPT64, Rv2660c, TB10.4, HspX, GlfT2, Fas, and IniB. Among these antigens, 10 linear B-cell epitopes, 9 helper T-cell epitopes, and 16 cytotoxic T-cell epitopes were predicted to design the multi-epitope vaccine. To improve the immunogenicity of the candidate vaccine, three different adjuvants, griselimycin, human beta-defensin 3 (HBD3), and 50s ribosomal protein (50sRP), were attached with linker sequences to the vaccine model. The immunogenic, antigenic, allergenic, and physicochemical properties of the resulting designed multi-epitope vaccines were predicted in silico. Moreover, 3D structural modeling, refinement, and validation were used to select a model for further evaluation. Molecular docking analysis revealed a consistent and significant binding affinity of the candidate vaccine for toll-like receptors (TLRs), TLR-2, -3, and -4. Immune simulation performed using C-ImmSim demonstrated that three rounds of immunization with multi-epitope vaccines induced a high production of cytokines and immunoglobulins related with both cellular and humoral immune response. Moreover, we constructed vaccine candidate composed of 50sRP and evaluated its immunogenicity in a mouse model. Consequently, this in silico-engineered multi-epitope structure can elicit adaptive immune responses and represents a promising novel candidate for TB vaccine development.

Abstract:Introduction: Borna disease virus 1 (BoDV-1) is an emerging zoonotic RNA virus that can cause severe acute encephalitis with high mortality. Currently, there are no effective countermeasures, and the potential risk of a future outbreak requires urgent attention. To address this challenge, the complete genome sequence of BoDV-1 was utilized, and immunoinformatics was applied to identify antigenic peptides suitable for vaccine development. Methods: Immunoinformatics and antigenicity-focused protein screening were employed to predict B-cell linear epitopes, B-cell conformational epitopes, and cytotoxic T lymphocyte (CTL) epitopes. Only overlapping epitopes with antigenicity greater than 1 and non-toxic, non-allergenic properties were selected for subsequent vaccine construction. The epitopes were linked using GPGPG linkers, incorporating β-defensins at the N-terminus to enhance immune response, and incorporating Hit-6 at the C-terminus to improve protein solubility and aid in protein purification. Computational tools were used to predict the immunogenicity, physicochemical properties, and structural stability of the vaccine. Molecular docking was performed to predict the stability and dynamics of the vaccine in complex with Toll-like receptor 4 (TLR-4) and major histocompatibility complex I (MHC I) receptors. The vaccine construct was cloned through in silico restriction to create a plasmid for expression in a suitable host. Results: Among the six BoDV-1 proteins analyzed, five exhibited high antigenicity scores. From these, eight non-toxic, non-allergenic overlapping epitopes with antigenicity scores greater than 1 were selected for vaccine development. Computational predictions indicated favorable immunogenicity, physicochemical properties, and structural stability. Molecular docking analysis showed that the vaccine remained stable in complex with TLR-4 and MHC I receptors, suggesting strong potential for immune recognition. A plasmid construct was successfully generated, providing a foundation for the experimental validation of vaccines in future pandemic scenarios. Discussion: These findings demonstrate the potential of the immunoinformatics-designed multi-epitope vaccines for the prevention and treatment of BoDV-1. Relevant preparations were made in advance for possible future outbreaks and could be quickly utilized for experimental verification.

Abstract:Infectious bursal disease virus is the causative agent of infectious bursal disease (Gumboro disease), a highly contagious immunosuppressive disease of chicken with a substantial economic impact on small- and large-scale poultry industries worldwide. Currently, live attenuated vaccines are widely used to control the disease in chickens despite their issues with safety (immunosuppression and bursal atrophy) and efficiency (breaking through the maternally-derived antibody titer). To overcome the drawbacks, the current study has, for the first time, attempted to construct a computational model of a multiepitope based vaccine candidate against infectious bursal disease virus, which has the potential to overcome the safety and protection issues found in the existing live-attenuated vaccines. The current study used a reverse vaccinology based immunoinformatics approach to construct the vaccine candidate using major and minor capsid proteins of the virus, VP2 and VP3, respectively. The vaccine construct was composed of four CD8 + epitopes, seven CD4 + T-cell epitopes, 11 B-cell epitopes and a Cholera Toxin B adjuvant, connected using appropriate flexible peptide linkers. The vaccine construct was evaluated as antigenic with VaxiJen Score of 0.6781, immunogenic with IEDB score of 2.89887 and non-allergenic. The 55.64 kDa construct was further evaluated for its physicochemical characteristics, which revealed that it was stable with an instability index of 16.24, basic with theoretical pI of 9.24, thermostable with aliphatic index of 86.72 and hydrophilic with GRAVY score of −0.256. The docking and molecular dynamics simulation studies of the vaccine construct with Toll-like receptor-3 revealed fair structural interaction (binding affinity of −295.94 kcal/mol) and complex stability. Further, the predicted induction of antibodies and cytokines by the vaccine construct indicated the possible elicitation of the host's immune response against the virus. The work is a significant attempt to develop next-generation vaccines against the infectious bursal disease virus though further experimental studies are required to assess the efficacy and protectivity of the proposed vaccine candidate in vivo .

Novel multi-epitope vaccine against bovine brucellosis: approach from immunoinformatics to expression

Abstract:Brucellosis is a zoonotic caused by the Brucella which is a well-known infectious disease agent in domestic animals and if transmitted, it can cause infection in humans. Because brucellosis is contagious, its control depends on the eradication of the animal disease in farms. There are two vaccines based on the killed and/or weakened bacteria against B. melitensis and B. abortus, but no recombinant vaccine is available for preventing the disease. The present study was designed to develop a multi-epitope vaccine against of B. melitensis and B. abortus using virB10, Omp31 and Omp16 antigens by the prediction of T lymphocytes, T cell cytotoxicity and IFN-γ epitopes. 50S L7/L12 Ribosomal protein from Mycobacterium tuberculosis was used as a bovine TLR4 and TLR9 agonist. GPGPG, AAY and KK linkers were used as a linker. Brucella construct was well-integrated in the pET-32a Shuttle vector with BamHI and HindIII restriction enzymes. The final construct contained 769 amino acids, that it was soluble protein of about ∼82 kDa after expression in the Escherichia coli SHuffle host. Modeled protein analysis based on the tertiary structure validation, molecular docking studies, molecular dynamics simulations results like RMSD, Gyration and RMSF as well as MM/PBSA analysis showed that this protein has a stable construct and is capable being in interaction with bovine TLR4 and TLR9. Analysis of the data obtained suggests that the proposed vaccine can induce the immune response by stimulating T- and B-cells, and may be used for prevention and remedial purposes, against B. melitensis and B. abortus.Communicated by Ramaswamy H. Abstract:Bluetongue (BT) is a non-contact infectious disease caused by Bluetongue virus (BTV), which can be transmitted by vector insects such as Culicoides and Aedes mosquitoes. The BTV VP2 protein encoded by the L2 gene is located at the outermost layer of the virus particle, plays a key role on mediating the adsorption and entry of virus, and it is also a main antigenic protein widely used for vaccine development. In this study, the BTV1 VP2 gene was cloned into pFastBac™Dual vector, and expressed in insect Sf21 cells. Immunized mice with purified recombinant VP2 protein can induce higher levels of antibodies. Three anti BTV1 VP2 monoclonal antibodies (mAbs) were generated (17E9C6, 17E9C8, 17E9H12), and showed high specific reactivity with recombinant VP2 protein and inactivated BTV1 virus. Finally, a novel linear B-cell epitope 296−KEPAD−300 on recombinant VP2 protein was identified by using three mAbs react with a series of continue-truncated peptides. The results of this study may provide new information on the structure and function of BTV1 VP2 protein and lay a foundation for the development of BTV1 diagnostic and prophylactic methods.

Abstract:Tick-borne viruses are a major risk from tick bites, which could result in viral infectious diseases among animals and humans. Bunyavirus causes severe fever with thrombocytopenia syndrome (SFTS), with signs and symptoms including high fever, vomiting, diarrhea, thrombocytopenia (low platelet count), leukopenia (low white blood cell count), elevated liver enzyme levels, multiple organ failure, and has a 6%–30% case-fatality rate. To date no effective drug or vaccines are available thus need urgent research for therapeutics formulation. Hence, in this study, the computational meta-analysis approach was implemented that incorporates immunoinformatics to find potential B-cell, HTL (helper T lymphocytes) and T-cell epitopes derived from antigenic SFTS proteins to design multi-epitopes vaccines for the treatment of SFTS. The predicted T cell, B cell and HTL epitopes were shortlisted and checked for antigenic properties and allergenic features. The best epitopes were then joined together to model of multi-epitopes vaccines for specific proteins (replicase and glycoprotein) and proteome wide. The constructed models were validated using in silico molecular docking approach to evaluate binding potential of the designed best constructs with TLR3 (toll like receptor 3). Following the MEVC (multi-epitopes vaccine construct) injection, the response of the immune system was significantly stimulated, and anti-toxicity of induced antibodies was tremendously enhanced. Before being neutralized, the antigen titers remained high 5–10 days after injection of replicase, glycoprotein and proteome wide constructed vaccines. For each antigenic vaccine, a significant antibody response induction was observed. Further, in vivo trials are required to affirm the effectiveness of the constructed vaccine against SFTS.

Abstract:Despite being a medically significant tick-borne virus, little research has been done on the Colorado tick fever virus. This virus is a member of the Spinareoviridae family and is mainly spread in the western parts of the United States and southwest Canada. It is classified as a Coltivirus. The main vector of the virus, Dermacentor andersoni , a wood tick found in the Rocky Mountains, determines patterns of viral spread. Furthermore, no effective antiviral treatment or vaccine has yet been created to guard against viral infections. An alternate method of preventing viral infections is vaccination. To stop a potential pandemic in the future, a thermodynamically stable and economical vaccination should be discovered. The goal of this study was to develop a potential vaccine against the Colorado tick fever virus. We seek to identify epitopes within two critical proteins, the Colorado tick fever virus's structural protein VP9 and the RNA-directed RNA polymerase VP1, to predict the precise locations that B and T cells will recognize by using a variety of reverse vaccinology and Computational techniques. Through modelling, the epitopes were deliberately engineered to connect with their corresponding HLA molecules, serving as the foundation for a multi-epitope subunit vaccination. This meticulously crafted vaccination candidate was created by joining B and T cell epitopes that had been extensively examined and linked with the appropriate linkers. Numerous physiological characteristics, including molecular weight, instability index, and aliphatic index, were assessed for the vaccine model, it was 3D modelled to dock with TLR-4. Previous research suggested that adding human beta-defensin as an adjuvant to the vaccine sequence at the N-terminus might improve immunogenicity. Additionally, the C-IMMSIM server ran immunological simulations to confirm the final vaccine design. The immunological simulation analysis anticipated a natural immune response. E. coli was used as the host for in-silico cloning, and the 0.96 CAI value obtained indicates that the vaccine construct will express itself to its fullest extent in the E. coli host. The binding mode of vaccine construct has been investigated via molecular docking approach against TLR4 human receptor protein. Vaccine created in this study shown efficacious and immunogenic response during immune simulation. Furthermore, this was more accurately validated through molecular dynamic simulations where the receptor molecule strongly bonds to the active residues of the epitope during 200 ns simulation time intervals. No certain changes were inferred among both receptor molecules and epitope during this time interval and remains static. Thus, these investigations result in determining the effectiveness in treating infections caused by the Colorado tick fever virus, which will allow experimentalists for its further validation to tackle this disease.

Abstract:Abstract Tuberculosis (TB) kills more individuals in the world than any other disease, and a threat made direr by the coverage of drug-resistant strains of Mycobacterium tuberculosis (Mtb). Bacillus Calmette–Guérin (BCG) is the single TB vaccine licensed for use in human beings and effectively protects infants and children against severe military and meningeal TB. We applied advanced computational techniques to develop a universal TB vaccine. In the current study, we select the very conserved, experimentally confirmed Mtb antigens, including Rv2608, Rv2684, Rv3804c (Ag85A), and Rv0125 (Mtb32A) to design a novel multi-epitope subunit vaccine. By using the Immune Epitopes Database (IEDB), we predicted different B-cell and T-cell epitopes. An adjuvant (Griselimycin) was also added to vaccine construct to improve its immunogenicity. Bioinformatics tools were used to predict, refined, and validate the 3D structure and then docked with toll-like-receptor (TLR-3) using different servers. The constructed vaccine was used for further processing based on allergenicity, antigenicity, solubility, different physiochemical properties, and molecular docking scores. The in silico immune simulation results showed significant response for immune cells. For successful expression of the vaccine in E. coli , in-silico cloning and codon optimization were performed. This research also sets out a good signal for the design of a peptide-based tuberculosis vaccine. In conclusion, our findings show that the known multi-epitope vaccine may activate humoral and cellular immune responses and maybe a possible tuberculosis vaccine candidate. Therefore, more experimental validations should be exposed to it.

Soluble Expression and Purification of Bluetongue Virus Type 1 (BTV1) Structure Protein VP2 in Escherichia Coli and Its Immunogenicity in Mice

Abstract:Background The VP2 on the surface of the virus particle is the main structural protein of BTV, which can induce the host to produce neutralizing antibodies and play an important role in the antiviral immunity process. This study aimed to obtain the soluble VP2 and analyze its immunogenicity. Methods The gene encoding the full-length VP2 of BTV1 was amplified by PCR. The products from restriction enzyme digestion and ligase reaction between VP2 and vector pET-28a were transformed into E.coli DH5α. After PCR and sequencing detection, the positive plasmid PET28a-VP2 was transformed into E.coli BL21(DE3) and Rosetta(DE3) competent cells, expression induced by IPTG. The fusion protein was expressed in the optimized conditions with the induction of IPTG, purified by affinity chromatography and identified by SDS-PAGE and Western blotting. A total of 5 Balb/c mice aged 6–8 weeks were immunized with the fusion protein at a dose of 30 µg per mouse. Each mouse was immunized three times at an interval of 3 weeks. Results The recombinant plasmid PET28a-VP2 was successfully constructed. The expression strains were induced by 0.4 mmol/L IPTG at 16 °C for 10 h, and BTV1 VP2 was expressed in a soluble form. The purity of the recombinant VP2 protein (∼109 kDa) was about 90% in the concentration at 0.2 mg/ml afterpurification. The purified VP2 had good immunoreactivity with BTV1 positive serum. Taken together, thisstudy offered a route for producing soluble BTV VP2, which retains activity and immunogenicity, to bebeneficial to the research on developing BTV vaccine, and lay the foundation for further research on BTV.
Designing and development of efficient multi-epitope-based peptide vaccine candidate against emerging avian rotavirus strains: A vaccinomic approach

Abstract:Background: Enteric avian rotavirus (ARV) is the etiological agent of several health problems that pose a global threat to commercial chickens. Therefore, to avoid these widespread epidemics and high mortality rates, only vaccine and strict biosecurity are required. Method: The present study employs computational techniques to design a unique multi-epitope-based vaccine candidate that successfully activates immune cells against the ARV by combining adjuvant, linker, and B and T-cell epitopes. Starting, homologous sequences in the various ARV serotypes were revealed in the NCBI BLAST database, and then the two surface proteins (VP4 and VP7) of the ARV were retrieved from the UniprotKB database. The Clustal Omega server was then used to identify the conserved regions among the homologous sequences, and the B and T-cell epitopes were predicted using IEDB servers. Then, superior epitopes-2 MHC-1 epitopes, 2 MHC-2 epitopes, and 3B-cell epitopes-were combined with various adjuvants to create a total of four unique vaccine candidates. Afterward, the designed vaccine candidates underwent computational validation to assess their antigenicity, allergenicity, and stability. The vaccine candidate (V2) that demonstrated non-antigenicity, a high VaxiJen score, and non-allergenicity was ultimately chosen for molecular docking and dynamic simulation. Results: Although the V2 and V4 vaccine candidates were highly immunogenic, V2 had a higher solubility rate. The predicted values of the aliphatic index and GRAVY value were 30.4 and 0.417, respectively. In terms of binding energy, V2 outperformed V4. Being successfully docked with TLRs, V2 was praised as the finest. After adaptation, the sequence's 50.73 % GC content outside of the BglII or ApaI restriction sites indicated that it was equivalently safe to clone. The chosen sequence was then inserted into the pET28a(+) vector within the BglII and ApaI restriction sites. This resulted in a final clone that was 4914 base pairs long, with the inserted sequence accounting for 478 bp and the vector accounting for the remainder. Conclusions: The immune-mediated simulation results for the selected vaccine construct showed significant response; thus, the study confirmed that the selected V2 vaccine candidate could enhance the immune response against ARV.

Abstract:Many countries around the world are facing severe challenges due to the recently emerging variants of SARS-CoV-2. Over the last few months, scientists have been developing treatments, drugs, and vaccines to subdue the virus and prevent its transmission. In this context, a peptide-based vaccine construct containing pathogenic proteins of the virus known to elicit an immune response was constructed. An analysis of the spike protein-based epitopes allowed us to design an "epitope-based subunit vaccine" against coronavirus using the approaches of "reverse vaccinology" and "immunoinformatics." Computational experimentation and a systematic, comprehensive protocol were followed with an aim to develop and design a multi-epitope-based peptide (MEBP) vaccine candidate. Our study attempted to predict an MEBP vaccine by introducing mutations of SARS-CoV-2 (Delta, Lambda, Iota, Omicron, and Kappa) in Spike glycoprotein and predicting dual-purpose epitopes (B-cell and T-cell). This was followed by screening the selected epitopes based on antigenicity, allergenicity, and population coverage and constructing them into a vaccine by using linkers and adjuvants. The vaccine construct was analyzed for its physicochemical properties and secondary structure prediction, and a 3D structure was built, refined, and validated. Furthermore, the peptide-protein interaction of the vaccine construct with Toll-like receptor (TLR) molecules was performed. Immune profiling was performed to check the immune response. Codon optimization of the vaccine construct was performed to obtain the GC content before cloning it into the E. coli genome, facilitating its progression it into a vector. Finally, an in-silico simulation of the vaccine–protein complex was performed to comprehend its stability and conformational behavior.
